Abstract:The application of high-performance liquid chromatography-mass spectrometry (HPLC-MS/MS) revealed the origin and evolution of antioxidants during the brewing process of hopped and unhopped reference beer. As tachioside (3-methoxy-4-hydroxyphenyl-β-d-glucopyranoside), arbutin (4-hydroxyphenyl-β-d-glucopyranoside), and hordatines clearly increased during the fermentation step, the raw material barley was investigated as a source of the corresponding precursors. Therefore, 4-hydroxyphenyl-β-d-glucopyranosyl-(1 → 6)-β-d-glucopyranosyl-(1 → 6)-β-d-glucopyranoside, 4-hydroxy-3-methoxyphenyl-β-d-glucopyranosyl-(1 → 6)-β-d-glucopyranoside, 4-hydroxy-3-methoxyphenyl-β-d-glucopyranosyl-(1 → 6)-β-d-glucopyranosyl-(1 → 6)-β-d-glucopyranoside, and 4-hydroxy-2-methoxyphenyl-β-d-glucopyranosyl-(1 → 6)-β-d-glucopyranosyl-(1 → 6)-β-d-glucopyranoside were isolated from barley for the first time, and identified using liquid chromatography-mass spectrometry (LC-MS) and one-dimensional/two-dimensional-nuclear magnetic resonance (1D/2D-NMR) experiments. Moreover, hordatine glucosides A, B, and C were isolated and identified from barley, and hordatine C glucoside was characterized for the first time. A fermentation model followed by HPLC-MS/MS analysis substantiated the release of tachioside from 4-hydroxy-3-methoxyphenyl-β-d-glucopyranosyl-(1 → 6)-β-d-glucopyranoside by Saccharomyces cerevisiae. Quantitation experiments monitoring the content in wheat, barley, and different barley malt types demonstrated a wide range of concentrations, providing a basis for further comprehensive investigations to optimize the antioxidant yield in beer to contribute to improved flavor stability.
Evaluation of the antioxidant characteristics of craft beer with green tea

Abstract:The addition of green tea as antioxidants to beer can improve the beer's flavor stability by protecting against staling during storage. To analyze the effect of different green teas on the flavor stability of beer, we developed an approach to rapidly evaluate their antioxidant activity. Ten types of craft beer were produced by adding different kinds of green tea during brewing, and their antioxidant activity and phenolic profiles were evaluated. The results showed remarkable variations in antioxidant activity and antioxidative compound contents, which were considerably higher in green tea beers than in non-tea beer (p < 0.05). A comprehensive evaluation function was developed to evaluate the total antioxidant activity of beers using principal component analysis. The highest total antioxidant activity was observed in Taiping Houkui beer, with a comprehensive evaluation score of 2.53. Pearson correlation analysis suggested that (-)-epigallocatechin gallate, (-)-epicatechin gallate, and (-)-epigallocatechin were strongly correlated with the total antioxidant activity of green tea beers (p < 0.01). The summation of their contents represented more than 60% of the total phenolic content of the teas, which can be used to predict the total antioxidant activity of green tea beers. PRACTICAL APPLICATION: Flavor stability is of prime concern for brewers, and flavor aging is increasingly becoming the limiting factor in beer shelf life. The application of green tea as antioxidants in beer can improve the flavor stability by protecting against beer staling during storage. The analytical method developed in this study will contribute to the rapid comparison of the effect of different green teas on the flavor stability of beer. Furthermore, the research findings demonstrate the potential benefits of green teas to beer flavor stability, which is of considerable importance in promoting the development and consumption of green teas.

Abstract:Xanthohumol (XN), possessing potent physiological activity, is exclusively derived from hops (Humulus lupulus L.) and exhibits high instability. However, its inherent instability often results in degradation during storage, leading to a decline in its activity due to the formation of various products. This study aimed to explore the stability of XN in beer under different storage conditions, establish or clarify degradation kinetic models, and understand the degradation mechanism. Our findings revealed that XN would degrade rapidly when exposed to high temperature and light. Its degradation followed a first-order kinetic model, involving reactions such as isomerization, hydration and ortho-position cyclization, resulting in the formation of five products. These insights shed light on the factors and mechanisms underlying the instability and degradability of XN, serving as a foundation for the development of a stable beer product enriched with functional XN.
Phenolics Profile and Antioxidant Activity of Special Beers

Abstract:The antioxidant activity and polyphenols content of beer associated with its low alcohol content are relevant factors for an evaluation of the nutritional quality of beer. To investigate the effect of adding foods on the nutritional quality of beer, seven special beers that were commercially available and produced adding natural foods (walnut, chestnut, cocoa, honey, green tea, coffee, and licorice) during the fermentation process were analyzed for their polyphenols and flavonoids contents, phenolics profile, and antioxidant activity. The results obtained showed that most of the special beers under study possessed antioxidant activity, as well as total polyphenols and flavonoids contents notably higher as compared with the five conventional beers analyzed. The highest polyphenols and flavonoids contents were exhibited in cocoa, walnut, chestnut, and licorice beers, followed by coffee, honey, and green tea beers. Antioxidant activity decreased in the order walnut, cocoa, chestnut, licorice, coffee, honey, and green tea. Most special beers were enriched in catechin, epicatechin, rutin, myricetin, quercetin, and resveratrol. The content of phenolic acids, especially ferulic, p-coumaric, syringic, and sinapic acids was generally higher in special beers as compared with conventional beers. Our findings showed that the addition of natural foods during the fermentation process remarkably increased antioxidant activity of beer and qualitatively and quantitatively improved its phenolics profile.
THERMODYNAMICS OF THE ANTIOXIDANT ACTIVITY OF HUMULONES AND OTHER ANTIOXIDANTS FROM BEER – A MOLECULAR MODELING APPROACH

Abstract:Recent experimental study identified eight potent antioxidants in German beers, including isoxanthohumol, (R)- and (S)-adhumulone, cis– and trans-iso-adhumulone, cis– and trans-iso- n-humulone, and desdimetyhyl-octahydro-iso-cohumulone. To provide insights into the structural basis of their radical scavenging activity, we calculated the thermodynamic feasibility of two common antioxidant mechanisms, hydrogen atom transfer (HAT) and single electron transfer followed by proton transfer (SET-PT), using the density functional theory (DFT) with B3LYP/6-311g++(2d,2p) method in the gas phase and implicit solvation model of water. The calculated bond dissociation enthalpies (BDEs) and ionization potential (IP) of all compounds were compared with the corresponding values for resveratrol, a highly potent antioxidant found in red wine. The fully reduced humulone isomer, desdimetyhyl-octahydro-iso-cohumulone, could scavenge free radicals via HAT as revealed by BDEs 5.1 and 23.9 kJ/mol lower than the values for resveratrol in gas phase and water, respectively. Furthermore, the enolic –OH group was identified as the pharmacophoric hotspot for the interaction of humulones with the reactive free radicals. The HAT potency of this group is significantly reduced through the formation of strong intramolecular hydrogen bond (IHB) with the β-keto group. Moreover, the SET-PT mechanism was thermodynamically favorable for isoxanthohumol. These results strongly suggest higher antioxidant activity of beers with the increased content of the reduced forms of humulones and their isomers.

Abstract:Herbs are rich in polyphenols compounds that are often added to beer to boost its antioxidant ability. To prove herbs can efficiently enhance the antioxidant ability of beer, the total polyphenols and flavonoids content, the distribution of phenolics profile, and antioxidant ability in commercial herbal beers were analyzed in this work. The results showed that most herbal beers had stronger antioxidant ability than conventional beers. The total polyphenols content in most herbal beers (250.8–1063.5 mg/L) was higher than that in conventional beers (179.2–308.4 mg/L), especially free phenolic acids (3.151–26.469 mg/L) such as gallic acid (0.248–2.853 mg/L), chlorogenic acid (0.422–6.420 mg/L), vanillic acid (0.567–10.256 mg/L), and caffeic acid (0.422–10.093 mg/L), which can explain the strong antioxidant ability of herbal beers to a certain extent. Among them, roselle beer (ROL) and jasmine beer (JAS) performed the strongest antioxidant ability. Furthermore, according to the distribution of phenolics profile, it was found that ROL and JAS had highest content of flavonoids including catechol, epicatechin, and quercetin compared with all beers. This was an important reason why ROL and JAS had the strongest antioxidant ability. In a word, the addition of herbs in beer brewing can effectively improve the content of polyphenols and antioxidant ability of beer, especially the herbs rich in flavonoids have the most significant contribution to the antioxidant ability of beer, which is helpful to improve beer quality.

Abstract:Beer is one of the oldest and most common beverages worldwide. The phenolic contents and antioxidant properties of beer are crucial factors in evaluating its nutritional quality. Special beers brewed with the addition of adjuncts are gaining in consumer preference, in response to demands for healthy food and new gustatory and olfactory stimuli. Many studies recently dealt with functional beers brewed with the addition of adjuncts. This review focuses on bioactive molecules, particularly the composition of phenolic compounds, and the antioxidant activity of beer. The current knowledge concerning the effect of the addition of adjuncts in the form of fruit, vegetables, herbs, and natural foods on the polyphenol content, antioxidant properties, and phenolic profile of beer is reviewed, with an outline of the emerging trends in brewing processes. Future studies need to complete the identification and characterization of the bioactive molecules in beer, as well as studying their absorption and metabolic fate in humans.

Abstract:In this work, selections of seven international hop varieties and three craft beers obtained from them were analyzed by SPME-GC/MS techniques with the aim to describe their volatile chemical profile. The brewing process was also reported. Furthermore, the hop extracts and beers were investigated to determine their flavonoid and polyphenol content and to evaluate their antioxidant power by DPPH and ABTS assays. The findings showed the presence of compounds belonging to different chemical classes such as monoterpenes, sesquiterpenes, alcohols, esters and fatty acids. In particular, sesquiterpenes were the main compounds with β-caryophyllene (from 1.7 to 16.2%) and humulene (from 10.8 to 43.9%) as the major components in all varieties of dried hop cones investigated. On the contrary, with the exception for the Pacific sample, monoterpenes were the class of compounds that were more abundant in the hop extracts and, among these, β-myrcene appeared to be the predominant constituent (from 31.8 to 71.4%). Regarding the craft beers obtained by adding these hop varieties, some differences in the qualitative and quantitative volatile composition have been found. All hop samples showed a high scavenging potential against both radicals. In the case of DPPH, the obtained IC50 values ranged from 0.027 to 0.047 mg/mL while they varied between 0.023 and 0.134 mg/mL by the ABTS assay. A positive correlation was found with polyphenol and flavonoid contents. Among beer samples, ACD was the richest one in polyphenols (292.0 mg GAE/100 mL beer) and flavonoids (5.8 mg QE/100 mL beer) and the most powerful against DPPH• and ABTS•+ radicals with IC50 values equal to 4.969 and 0.198 v/v%, respectively.

Abstract:Beer is one of the oldest and most consumed beverages worldwide, and recent trends point to increased consumption of functional beers. However, there is a lack in the scientific literature on the effects of adding functional adjuncts in distinct steps of the manufacturing process and its implications on the final physicochemical and sensorial profile. Therefore, the present review analyzes the ingredients used and their insertion stage to achieve a functional beer with bioactive compounds, higher antioxidant activity, and improved sensory characteristics. The addition of fruits, herbal extracts, plants, and mushrooms in beers was documented. Furthermore, adjuncts were successfully added in wort boiling, fermentation, maturation, and packaging. The wort boiling step stands out among these four due to the superior extraction of phenolic compounds from the added adjuncts. On the other hand, adjunct addition in the maturation step induced low increases in antioxidant and phenolic content of the respective enriched beers. Fruits represented the majority of adopted adjuncts among the studies evaluated. Furthermore, the addition of fruits represented a positive increment in the beer’s volatile profile and an increase in sensory acceptability. A gap in the literature was found regarding the analysis of phenolic compounds with appropriate techniques such as HPLC-MS. Furthermore, there is a need to study the bioavailability of the incorporated bioactive compounds to prove the health claims inferred about these beers. In conclusion, functional beers are a little-explored relevant field, with potential for new studies.

Abstract:Progressive cognitive impairment is a defining feature of the dementia of Alzheimer's disease (AD), yet disagreement exists over which abilities decline most precipitously and which cognitive tests are most sensitive. In this study, 51 AD patients in the early to middle stages of illness and 22 age-matched normal controls were administered a battery of neu ropsychological tests at 6-month intervals over a 2-year period. While the performance of the normal controls remained stable over the 2 years, the AD patients displayed progressive decline on all tests. The greatest declines occurred on tests requiring lexical/semantic processing (Boston Naming Test) and comprehension of syntactic relationships (Token Test). Performance on visuospatial tests (Wechsler Adult Intelligence Scale-Revised Block Design, Benton Visual Retention Test, Spatial Delayed Recognition Span Test) declined less rapidly. The findings support previous reports that language impairment may be central to the dementia of AD, and that confrontation naming is particularly sensitive to decline in this illness.
Target profiling of beer styles by their iso-α-acid and phenolic content using liquid chromatography-quadrupole time-of-flight-mass spectrometry

Abstract:Beer styles show wide variation in color, flavor, and clarity, due to differences in their chemical content. Some of the major flavor compounds in beer are isomerized alpha acids and phenolic compounds. These were investigated as potentially discerning features between beer styles. A selection of 32 American beers covering five styles was analyzed using liquid chromatography quadrupole time-of-flight mass spectrometry, which resulted in high mass accuracy chromatograms of the studied analytes. Distinctions between the presence or relative concentrations of certain compounds were observed and related back to brewing ingredients and procedures. For example, vanillin was only observed in stout beers due to the use of roasted barley malts for brewing, while chlorogenic acid isomers were found in two sours at relatively high concentrations (189 and 34 mg/L) because of the fruits used to flavor the beers. Distinctions were further confirmed using multivariate analysis techniques, which separated three of the five beer styles (India pale ales, stouts, and sours).
Evaluation of non-volatile metabolites in beer stored at high temperature and utility as an accelerated method to predict flavour stability

Abstract:Flavour stability is vital to the brewing industry as beer is often stored for an extended time under variable conditions. Developing an accelerated model to evaluate brewing techniques that affect flavour stability is an important area of research. Here, we performed metabolomics on non-volatile compounds in beer stored at 37 °C between 1 and 14 days for two beer types: an amber ale and an India pale ale. The experiment determined high temperature to influence non-volatile metabolites, including the purine 5-methylthioadenosine (5-MTA). In a second experiment, three brewing techniques were evaluated for improved flavour stability: use of antioxidant crowns, chelation of pro-oxidants, and varying plant content in hops. Sensory analysis determined the hop method was associated with improved flavour stability, and this was consistent with reduced 5-MTA at both regular and high temperature storage. Future studies are warranted to understand the influence of 5-MTA on flavour and aging within different beer types.

Abstract:The data of 969 dental arches replaced by partial denture, and 1282 dental arches of the same edentulousness but unreplaced, examined in an unselected population, have been analyzed from the point of view of the occurrence frequency of the gingivitis, the pocket, the movability and the caries. It has been stated that the effect of the prosthesis of the remaining teeth may be evaluated only by comparison with a suitable control group with no dentures. Furthermore, it has been stated that the remaining teeth are not damaged by well planned partial dentures (with metal base plates) while the wrongly planned dentures (with plastic base plates) develop a significant damaging effect.
Development of a quantitative method for organic acid in wine and beer using high performance liquid chromatography

Abstract:This study was conducted to compare the organic acid content in liquors (red wine, white wine, and beer) using three different high-performance liquid chromatography analysis methods. Post-column reaction methods (method 2 and 3) were found to be more promising than UV-detection method (method 1). Using method 2 (two columns), the analyzed red wine was found to contain 2,652.4 mg/L tartaric acid and 1,392.9 mg/L lactic acid but relatively lower amounts of malic acid (271.0 mg/L). Furthermore, tartaric acid (1,160.8–2,749.1 mg/L) and malic acid (470.2–3,107.9 mg/L) were the major components in white wine. Beers were analyzed using method 3 (one column). In the analyzed foreign beers, lactic acid (95.9–226.4 mg/L), malic acid (62.2–110.5 mg/L), acetic acid (93.5–183.8 mg/L), and succinic acid (37.0–56.2 mg/L) were detected. Similar to the foreign beers, the contents of succinic acid in the domestic beers were the lowest. The proposed methods could be useful for quantitative analysis of organic acids in wine and beer.

Abstract:Autoimmune New Zealand mice provide animal models of human systemic lupus erythematosus. Investigations suggest that the disease is multifactorial in origin; genetic, viral, biochemical, and hormonal factors are all capable of modifying the expression of autoimmunity. However, the immune system is the common pathway of disease expression. Early abnormal functioning of the immune system involves excessive effector function and inadequate suppressor function. Since New Zealand mice act as if they are excessively stimulated, the defect in suppressor function may be relative rather than absolute. Thus, excessive B-cell stimulation cannot be controlled by available regulatory processes. Thymic developmental abnormalities point to an early primary defect in the generation of regulatory T cells. As NZB and NZB/NZW mice pass from neonatal to young adult life, they lose suppressor cell function, probably from the loss of one of at least two cell types that interact to produce suppression. The lost cell appears to be an immature or precursor cell. Anti-T cell antibodies and immune complexes may accelerate the loss of suppressor cells and thereby lead to more rapid depletion of the suppressor cell precursors. Beneficial therapeutic efforts include administration of suppressor cells or their products, steroid hormones—glucocorticoids and androgens—immunosuppressive drugs, antiinflammatory agents, and antiviral drugs.
Complexation of transition metals by chelators added during mashing and impact on beer stability

Abstract:Beer inevitably changes due to an array of staling reactions. A major factor in beer ageing is the involvement of transition metals (iron, copper, manganese) in oxidative reactions. To tackle the flavour stability issue, metal chelation was investigated. Based on previous research, five primary chelators (tannic acid, gallic acid, EDTA, citric acid and phytic acid) were screened using experimental design for their capacity to reduce the content of wort transition metals. The chelating agents were added under varying conditions (mash out temperature, mash pH, grain bill, chelator concentration, addition time) during laboratory scale mashing to assess how they altered complexation and metal load. Fourteen alternative chelators (ferulic acid, tartaric acid, quercetin, chlorogenic acid and ten polyphenolic food extracts: green tea, pomegranate, grape seed, reishi, cinnamon, curcuma, milk thistle, ginkgo, grapefruit seed and raspberry) were also explored. Metal ions were analysed using inductively coupled plasma optical emission spectrometry and wort oxidative stability by electron spin resonance spectroscopy. Mash pH was the most decisive of all tested process variables: acidified mashing (pH 6 to 5) produced worts with more iron, manganese and zinc (230, 320 and 150%, respectively). Addition of effective chelators counteracted this undesirable effect for iron. Green tea extract, tannic acid and, particularly, pomegranate extract all resulted in lower wort iron. Conversely, addition of EDTA, caused iron, manganese and zinc to increase. Pomegranate extract (90% ellagic acid) was the best performing chelator and reduced radical generation in wort (80% reduction by 60 mg/L addition), making it a promising novel compound in the improvement of beer shelf life. © 2021 The Authors. Abstract:Hop-derived esters contribute to beer flavor and changes in the ester spectrum are believed to be an important driver in the flavor–instability of hoppy beers. To date, there exists no published method that enables reliable quantification of hop-derived esters in beer. As the availability of such a method is vital to enlarge the understanding of beer flavor stability, the current article is concerned with the development, validation, and application of a headspace solid phase microextraction gas chromatography selected ion monitoring mass spectrometry (HS-SPME-GC-SIM-MS) based methodology for quantification of 16 hop-derived esters in beer. The validation data shows that choosing suitable HS-SPME-GC-SIM-MS conditions enabled reliable quantification of esters across a range of 1–200 µg/L, with calculated limits of quantification being well below 1 µg/L. Spiking experiments using terpenes and terpenoids evidenced method robustness, most importantly when two commercially available stable isotope labeled internal standards (d6-geranyl acetate and 13C-methyl octanoate) were used. Application to beer samples indicated that beers brewed with different qualities/quantities of hops and differing in freshness could be well differentiated by HS-SPME-GC-SIM-MS analysis of their ester profile. Influence of Packaging Material on Polyphenol Content and Antioxidant Activity in Some Commercial Beers

Abstract:Using two methods (ferric reducing antioxidant power and radical scavenging activity), the total polyphenol content (Folin–Ciocalteu reagent) and polyphenol patterns (HPLC) in 10 commercial lager beer brands produced in Romania was determined. Samples bottled in glass, plastic and aluminium packages were analysed for each brand when available. Results have indicated considerable variations in the total and individual phenolic contents as well as antioxidant activity across beer brands. A statistical analysis was performed to assess the influence of packaging type on the antioxidant activity and phenolic content of the beers. Statistical differences were found between the DPPH and FRAP methods in glass, aluminium and plastic material. Moreover, the antioxidant activity based on the DPPH method is influenced by the type of packaging material, while in the case of FRAP method, no statistical difference was reported. Furthermore, the same analysis has shown that the polyphenol concentration is invariant to the type of material.
