Abstract:Previous studies have evaluated pork quality by omics methods. However, proteomics coupled with metabolomics to investigate pork freshness by using pork exudates has not been reported. This study determined the changes in the profiles of peptides and metabolites in exudates from pork stored at different temperatures (25, 10, 4, and −2 °C). Multivariate statistical analysis revealed similar changes in profiles in exudates collected from pork stored at −2 and 4 °C, and additional changes following storage at higher temperatures. We identified peptides from 7 proteins and 30 metabolites differing in abundance between fresh and spoiled pork. Significant correlations between pork quality and most of the peptides from these 7 proteins and 30 metabolites were found. The present study provides insight into changes in the peptide and metabolite profiles of exudates from pork during storage at different temperatures, and our analysis suggests that such changes can be used as markers of pork spoilage.

Unraveling the spoilage characteristics of refrigerated pork using high-throughput sequencing coupled with UHPLC-MS/MS-based non-targeted metabolomics

Abstract:The spoilage of refrigerated pork involves nutrient depletion and the production of spoilage metabolites by spoilage bacteria, yet the microbe-metabolite interactions during this process remain unclear. This study employed 16S rRNA high-throughput sequencing and non-targeted metabolomics based on ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) to reveal the core microbiota and metabolite profiles of pork during refrigeration. A total of 45 potential biomarkers were screened through random forest model analysis. Metabolic pathway analysis indicated that eleven pathways, including biogenic amine metabolism, pentose metabolism, purine metabolism, pyrimidine metabolism, phospholipid metabolism, and fatty acid degradation, were potential mechanisms of pork spoilage. Correlation analysis revealed nine metabolites-histamine, tyramine, tryptamine, D-gluconic acid, UDP-d-glucose, xanthine, glutamine, phosphatidylcholine, and hexadecanoic acid-as spoilage biomarkers, with Pseudomonas, Serratia, and Photobacterium playing significant roles. This study provides new insights into the changes in microbial and metabolic characteristics during the spoilage of refrigerated pork.
Characterization and interactions of spoilage of Pseudomonas fragi C6 and Brochothrix thermosphacta S5 in chilled pork based on LC-MS/MS and screening of potential spoilage biomarkers

Abstract:Pseudomonas and Brochothrix are the main spoilage organisms in pork, and each of these plays an essential role in the spoilage process. However, the effect of co-contamination of these two organisms in pork has not been elucidated. The changing bacterial communities during spontaneous spoilage of pork at 4 °C were evaluated using high-throughput sequencing. The dominant spoilage bacteria were isolated and these were identified as Pseudomonas fragi C6 and Brochothrix thermosphacta S5. Chilled pork was then experimentally contaminated with these strains, individually and in combination, and the progression of spoilage was assessed by analyzing various physicochemical indicators. These included total viable counts (TVC), pH, color, total volatile basic nitrogen (TVB-N), and detection of microbial metabolites. After 7 days of chilled storage, co-contaminated pork produced higher TVC and TVB-N values than mono-contaminated samples. Metabolomic analysis identified a total of 8,084 metabolites in all three groups combined. Differential metabolites were identified, which were involved in 38 metabolic pathways. Among these pathways, the biosynthesis of alkaloids derived from purine and histidine was identified as an important pathway related to spoilage. Specifically, histidine, histamine, AMP, IMP, GMP, succinic acid, and oxoglutaric acid were identified as potential spoilage biomarkers. The study showed that the combined presence of P. fragi C6 and B. thermosphacta S5 bacteria makes chilled pork more prone to spoilage, compared to their individual presence. This study provides insights that can assist in applying appropriate techniques to maintain quality and safety changes in meat during storage and further the assessment of freshness.

Abstract:Both lean meat and fat together contribute to the consumer impression of traditional Chinese bacon. The quality of this meat product is mainly dependent on numerous biochemical reactions and microbes. For the first time, this study reveals the microbial community succession and metabolic characteristics of the fat portion during the processing of traditional Chinese bacon by high-throughput sequencing and untargeted metabolomics (ultra-high performance liquid chromatography, UHPLC). The results showed that Proteobacteria and Firmicutes were the predominant bacterial phyla in traditional Chinese bacon. At the genus level, 15 core genera (relative abundance >1%) were identified, including Salinivibrio, Staphylococcus, Brochothrix, Vibrio, Psychrobacter, Acinetobacter, Carnobacterium, Aeromonas, Pseudomona, Cobetia, Macrococcus, Enterobacter, Kocuria, unclassified-f-Vibrionaceae, and Empedobacter. Distinct separation between different groups were identified by principal component analysis, revealing significant differences in the metabolic characteristics of different samples. A total of 262 metabolites were identified, and most of the metabolites belonged to lipids and lipid-like molecules, of which 60 were identified as differential metabolites. Results of correlation analysis indicated that some differential metabolites were significantly positively correlated with dominant bacteria, such as Salinivibrio, Vibrio, Cobetia and Staphylococcus, which reinforced the fact that microorganisms have an indispensable role in the quality formation of traditional Chinese bacon.

Abstract:To identify the microbial community and origin of the spoilage flora of bacon, the changes in microbial population numbers and community structure were followed along the processing line, using culture-independent and culture-dependent methods. 16S rRNA gene amplicon sequencing (16S-seq) analysis showed that community complexity and structure significantly differed at different processing stages. Some 428 bacterial groups were ascertained at genus level, and Acinetobacter, Pseudomonas, Psychrobacter, and Brochothrix were the predominant bacteria on raw meats. After curing specimens dominated by Psychrobacter, Weissella, Vibrio, Leuconostoc, Myroides, Acinetobacter, and Lactobacillus, a total of 33 species were identified by traditional microbiological analyses and direct sequence determination methods. Our results indicated that curing should be considered one of the primary factors during various processing steps, presumably contaminating the products directly or indirectly.
Effect of Superchilling Combined with Modified Atmosphere Packaging on Pork of Lanxi Pig

Abstract:Superchilling and modified atmosphere packaging(MAP) are two important preservation technologies of meat. The research combined the two kinds of technologies. It took Lanxi pigs as experimental material. The indexes included p H,water holding capacity,total bacterial count and sensory score. Firstly,the effects of pork were compared under conditions of 4℃ cold storage,-2℃ superchilling storage and-18℃ frozen storage.Then,the influence of CO2 and O2on quality of pork was researched partially under-2℃ superchilling. At last,the best combination of CO2 and O2was determined. The result showed that pork under-2℃ superchilling storage could keep normal quality in 15 days. CO2 could lower p H of pork and inhibits the growth of microorganism in it,while O2 could keep good color but promote the growth of microorganism. Finally,the experiment of CO2/O2 Combination shows that 40% CO2+60% O2 was the best MAP composition under-2℃superchilling storage. The fresh period was as long as 24 days.
Pseudomonas fragi and Pseudomonas lundensis drove the co-spoilage in chilled pork: Insights from the metabolome

Abstract:Pseudomonas fragi and Pseudomonas lundensis are key spoilage microorganisms in aerobically stored chilled meat, which had co-spoilage effect on chilled pork. However, the mechanism of co-spoilage interaction of P. fragi and P. lundensis has not been elucidated. The metabolism of nutrients in pork by mixed bacteria may differ from that by single strains. This study applied metabolomics and in vitro metabolite utilization experiment to assess the mechanism of that co-metabolize raw pork during storage. Results showed that co-spoilage group had 104 differential metabolites (histamine, N-methylhydantoin and D-gluconic acid, etc.) and 78 differential metabolites (putrescine, uracil acid and uracil, etc.) compared to P. fragi and P. lundensis group, respectively. These differential metabolites were mainly related to histidine metabolism, arginine biosynthesis, and purine. The co-spoilage effect of P. fragi and P. lundensis was related to the promotion of deamination and decarboxylation of histidine to produce histamine, the promotion of α-ketoglutarate and glutamate-mediated transamination, and the full utilization of the arginine deiminase pathway by the co-culture bacteria in the degradation of arginine to produce putrescine. In vitro experiment, co-cultures of these strains resulted in greater consumption of glucose, higher utilization activity, and promoted deamination and decarboxylation of amino acids. These findings reveal the complex and competitive interactions of co-culture of P. fragi and P. lundensis, providing insight into microbial spoilage mechanisms in chilled pork.
Pelvic sonography to help determine the appropriate therapy for luteal phase defects.

Abstract:In some series the most appropriate therapy for luteal phase defects is supplemental progesterone in the luteal phase. Clomiphene's efficacy is more controversial since in one series only 8% achieved a successful pregnancy versus 45% in another study. Pelvic sonography was used to evaluate follicular development and release of the ovum in 50 infertile women with luteal phase defects. The results showed that only 40% had "pure" luteal phase defects whereas 52% had immature follicles and 8% had unruptured follicles. Sixty-two percent of the patients had previous therapy for a luteal phase defect and failed to conceive. Sixty-eight percent of this group did conceive when ultrasound was used to determine the appropriate therapy. Thus ultrasound can be employed to determine if the women with a luteal phase defect should be treated with a fertility drug, e.g., clomiphene or just with luteal phase progesterone support. Supplemental progesterone might still be needed with clomiphene based on repeat endometrial biopsy results.

Abstract:This study revealed the roles of microorganisms and endogenous enzymes in evolution of the metabolic characteristics of the lean portion of traditional Chinese bacon by exploring the changes in microbial succession, metabolic characteristics, lipid oxidation, lipid hydrolysis, lipoxygenase activity, lipohydrolase activity, protein degradation, and environmental factors during processing. The results showed that Proteobacteria and Firmicutes were the dominant phyla in the lean portion of traditional Chinese bacon, and 16 core genera (relative abundance > 1 %) were identified at the genus level. Significant differences were observed in the metabolic characteristics of different samples. A total of 23 differential metabolites were identified in positive and negative ion modes (P < 0.05, variable importance in the projection > 1.5). Peroxide value (POV), thiobarbituric acid reactive substances value (TBARS), proteolysis index (PI) and the degree of lipid hydrolysis increased during the processing. Phospholipids were the main hydrolysis substrates. Lipoxygenase activity increased first and then decreased, while lipohydrolase activity decreased gradually. The results of canonical correspondence analysis indicated that the salt content was the main environmental factor affecting the microbial composition. The results of Spearman correlation analysis and comprehensive discussion showed that the metabolic characteristics of the lean portion of traditional Chinese bacon during the curing period were mainly due to the catalysis of the endogenous enzymes in meat and the extracellular enzymes secreted by bacteria such as Brochothix, Acinetobacter, Psychrobacter, etc. The extracellular enzymes secreted by Salinivibrio, Vibrio, and Staphylococcus in lightfermented meat and final meat might have important contributions to the metabolic characteristics.
Novel insights from protein degradation: Deciphering the dynamic evolution of biogenic amines as a quality indicator in pork during storage

Abstract:Biogenic amines (BAs) have drawn great attention as important markers for monitoring food quality. However, the BAs content in protein degradation profiles during pork storage was inadequately investigated. In this work, the Longissimus lumborum and Breast and flank of pork were collected, and their peptides contents, free amino acids (FAAs) contents, BAs contents, and several characteristic physicochemical indexes were monitored during storage at 4 °C. As a result, the differences of nutritional components in the Longissimus lumborum and Breast and flank could not affect the shelf life of refrigerated pork. There are 161 small peptides in the Longissimus lumborum of pork identified by LC-MS. As verified, arginine, glutamic acid, valine, and alanine could serve as four indicative amino acids during protein degradation in pork, and the arginine degradation pathway is more complex. Redundancy analysis confirmed that putrescine and cadaverine were significantly related to the precursor FAAs content, and their sum value could be used as a novel quality indicator instead of the biogenic amine index (BAI). Finally, the above prediction was also verified by the other species (beef, mutton and chicken) to improve the index system of meat quality evaluation in cold chain logistics.

Abstract:Background: Although microorganisms are the main cause of spoilage in prepared beef steaks, very few deep spoilage mechanisms have been reported so far. Aiming to unravel the mechanisms during 12 days of storage at 4 °C affecting the quality of prepared beef steak, the present study investigated the changes in microbial dynamic community using a combined high-throughput sequencing combined and bioinformatics. In addition, gas chromatography-mass spectrometry combined with multivariate statistical analysis was utilized to identify marker candidates for prepared steaks. Furthermore, cloud platform analysis was applied to determine prepared beef steak spoilage, including the relationship between microbiological and physicochemical indicators and volatile compounds. Results: The results showed that the dominant groups of Pseudomonas, Brochothrix thermosphacta, Lactobacillus and Lactococcus caused the spoilage of prepared beef steak, which are strongly associated with significant changes in physicochemical properties and volatile organic compounds (furan-2-pentyl-, pentanal, 1-octanol, 1-nonanol and dimethyl sulfide). Metabolic pathways were proposed, among which lipid metabolism and amino acid metabolism were most abundant. Conclusion: The present study is helpful with respect to further understanding the relationship between spoilage microorganisms and the quality of prepared beef steak, and provides a reference for investigating the spoilage mechanism of dominant spoilage bacteria and how to extend the shelf life of meat products. © 2024 Society of Chemical Industry.
Utilizing Pork Exudate Metabolomics to Reveal the Impact of Aging on Meat Quality

Abstract:This study was performed to assess the changes in meat quality and metabolome profiles of meat exudate during postmortem aging. At 24 h postmortem, longissimus lumborum muscles were collected from 10 pork carcasses, cut into three sections, and randomly assigned to three aging period groups (2, 9, and 16 d). Meat quality and chemical analyses, along with the metabolomics of meat exudates using ultra-high-performance liquid chromatography coupled with a quadrupole time-of-flight mass spectrometer (UHPLC-QTOF-MS) platform, were conducted. Results indicated a declined (p < 0.05) display color stability, and increased (p < 0.05) purge loss, meat tenderness, and lipid oxidation as aging extended. The principal component analysis and hierarchical clustering analysis exhibited distinct clusters of the exudate metabolome of each aging treatment. A total of 39 significantly changed features were tentatively identified via matching them to METLIN database according to their MS/MS information. Some of those features are associated with adenosine triphosphate metabolism (creatine and hypoxanthine), antioxidation (oxidized glutathione and carnosine), and proteolysis (dipeptides and tripeptides). The findings provide valuable information that reflects the meat quality’s attributes and could be used as a source of potential biomarkers for predicting aging times and meat quality changes.

Optimization of PCR Assays of Major Foodborne Pathogens and Their Application in Investigation of Pathogen Contamination in Chilled Pork

Abstract:To optimize the PCR identification systems of major foodborne pathogens for investigating into their prevalence in chilled pork,we compared 5 assay formats of Salmonella spp(including S.typhimurium and S.enteritidis) and 6 assay formats of Escherichia coli O157:H7 reported in the literature.With two optimized assay formats,one for S.typhimurium and S.eneritidis and the other for E.coli,and with the PCR method set up in our laboratory for Listeria monocytogenes,we investigated the prevalence of L.monocytogenes,Salmonella spp and E.coli O157:H7 in 50 fresh pork samples and 150 chilled pork samples collected from Hangzhou markets.The results showed that the overall rate of pathogen contamination in the 200 samples was 21%.The incidence in chilled pork samples was higher than that in fresh pork samples(24.7% vs 10%).The incidence of L.monocytogenes was 2% in fresh pork samples and 14.7% in chilled pork samples.The incidence of Salmonella spp in chilled pork samples was also higher than that in fresh pork samples(11.3% vs 8.0%);No strain of E.coli O157:H7 was found.There were 4 strains of S.typhimurium,all from chilled pork samples.This research suggests the potential risk of major foodborne pathogens in chilled pork,probably due to contamination of the production line.
Characterization of a microbial community developing during refrigerated storage of vacuum packed Yao meat, a Chinese traditional food

Abstract:Chinese traditional Yao meat (salted pork in jelly) is a popular food product with a unique taste and high nourishing value. Exploration of the microbial ecosystem in vacuum packaged Yao meat during refrigeration was conducted by high-throughput sequencing of the V4-V5 region of the16S rRNA gene. A total of 673,965 sequences from 15 DNA samples were analyzed. More than 226 bacterial genera belonging to 13 phyla were identified, some of them typically associated with meat spoilage. The microbial community at the start of the experiment showed the lowest genetic diversity and was dominated by Bacillus. After one-week of refrigeration, Moraxellaceae, including Acinetobacter, Psychrobacter and other members, Enterobacteriaceae, and Pseudomonadaceae were the prevalent groups. The microbial community reached its highest diversity after two weeks in refrigerated storage. In the later stages of the storage period, an undetermined species of Enterobacteriaceae became predominant, and we suggest that this is the main contributor to Yao meat spoilage. Moreover, the community composition at the end of the experimental storage period was significantly different from that of previous time points. In conclusion, this meta-genomic analysis provides researchers and food industry with useful information regarding spoilage management of Yao meat.

Abstract:This study aims to investigate experimentally the influence of microbial bacterial proteins on microbial metabolites in the chilled storage of Tan Sheep meat based on the surface enhanced laser desorption/ionization-time of flight-mass spectrometry (SELDI-TOF-MS) and gas chromatography-time of flight-mass spectrometry (GC-TOF MS). The correlation analysis of differentially abundant proteins and differential metabolites detected simultaneously in different storage periods has been carried out by heatmap analysis. The results show that 30 differential expressed metabolites were statistically significant. These metabolites were identified as potential spoilage biomarkers. Among of them, 25 differential expressed metabolites can be matched to KEGG metabolic pathways. cyano-amino-acid pathway, pentose phosphate pathway, histidine pathway, purine metabolic pathways, lanine, aspartic acid and glutamate pathway were highly active. Therefore, the differentially abundant proteins contents of microbiology in metabolism can lead to differential effects on metabolic pathways.

Abstract:This study aimed to gain deeper insights into the microbiota composition and population dynamics, monitor the dominant bacterial populations and identify the specific spoilage microorganisms (SSOs) of vacuum-packed bacon during refrigerated storage using both culture-independent and dependent methods. High-throughout sequencing (HTS) showed that the microbial composition changed greatly with the prolongation of storage time. The diversity of microbiota was abundant at the initial stage then experienced a continuous decrease. Lactic acid bacteria (LAB) mainly Leuconostoc and Lactobacillus dominated the microbial population after seven days of storage. A total of 26 isolates were identified from different growth media using traditional cultivation isolation and identification method. Leuconostoc mesenteroides and Leuconostoc carnosum were the most prevalent species since day 15, while Lactobacillus sakei and Lactobacillus curvatus were only found on day 45, suggesting that they could be responsible for the spoilage of bacon. Serratia, Rahnella, Fusobacterium and Lactococcus underwent a dramatic increase at some point in individual batchs which may be considered as potential contributors to the spoilage.

Abstract:Staphylococcus xylosus, Leuconostoc mesenteroides, Carnobacterium maltaromaticum , Leuconostoc gelidum , and Serratia liquefaciens were investigated for their roles in in the spoilage of sterilized smoked bacon. These five strains, individually and in combination, were applied as starters on sliced bacon at 4–5 log 10 CFU/g using a hand-operated spraying bottle and stored for 45 days at 0–4°C. Dynamics, diversity, and succession of microbial community during storage of samples were studied by high-throughput sequencing (HTS) of the V3–V4 region of the 16S rRNA gene. A total of 367 bacterial genera belonging to 21 phyla were identified. Bacterial counts in all the inoculated specimens increased significantly within the first 15 days while the microbiota developed into more similar communities with increasing storage time. At the end of the storage time, the highest abundance of Serratia (96.46%) was found in samples inoculated with S. liquefaciens . Similarly, for samples inoculated with C. maltaromaticum and L. mesenteroides , a sharp increase in Carnobacterium and Leuconostoc abundance was observed as they reached a maximum relative abundance of 97.95 and 81.6%, respectively. Hence, these species were not only the predominant ones but could also have been the more competitive ones, potentially inhibiting the growth of other microorganisms. By analyzing the bacterial load of meat products using the SSO model, the relationships between the microbial communities involved in spoilage can be understood to assist further research.

Abstract:The aim of the present study was to investigate the evolution of the volatile compounds of aerobically stored sterile pork meat as a consequence of the metabolic activities of inoculated specific spoilage microorganisms. Thus, Pseudomonas fragi, Pseudomonas putida, Lactobacillus sakei and Leuconostoc mesenteroides were inoculated in monocultures, dual cultures and a cocktail culture of all strains on sterile pork meat stored aerobically at 4 and 10 °C. Microbiological and sensory analyses, as well as pH measurements, were performed, along with headspace solid-phase microextraction gas chromatography/mass spectroscopy (headspace SPME–GC/MS) analysis. Data analytics were used to correlate the volatile compounds with the spoilage potential of each stain using multivariate data analysis. The results for the sensory discrimination showed that the volatiles that dominated in spoiled samples consisted mostly of alcohols, ketones and two esters (butyl acetate and ethyl acetate), while at fresh samples, dimethyl sulfide, furans, acetoin and ethyl lactate were detected. On the other hand, 2-butanone, diacetyl and acetaldehyde were among the volatile compounds that were mainly correlated with the inoculated meat during storage. In addition, P. fragi was positively correlated with a higher number of volatiles compared to the other strains, strengthening the hypothesis that volatile compound production is strain-dependent.

Abstract:Pseudomonas fragi is primarily responsible for the spoilage of various foods, especially meat. The aim of this study was to investigate the antibacterial mechanism of 3-carene against P. fragi. 3-Carene treatment decreased the phospholipid content and the fluidity of the cell membrane, induced reactive oxygen species (ROS) generation and affected respiratory chain dehydrogenase, oxoglutarate dehydrogenase and citrate synthase in P. fragi. Metabolomics and proteomics analyses further showed that in the presence of 3-carene, 519 proteins, 136 metabolites in positive ion mode and 100 metabolites in negative ion mode were differentially expressed. These proteins and metabolites were primarily involved in amino acid metabolism, fatty acid degradation, the tricarboxylic acid cycle (TCA cycle) and other processes. Consequently, the stimulation of 3-carene altered cell membrane properties, disturbed important amino acid and energy metabolism, and even caused oxidative stress. Additionally, the results of total viable counts and the total volatile base nitrogen indicated that 3-carene could significantly improve the preservation of refrigerated pork. This study suggested that 3-carene has promising potential to be developed as a food preservative.
