Abstract:Aims Radial oxygen loss (ROL) for macrophytes is intimately involved in their survival and growth, thus detailed characterizations of ROL and its implication for geochemical processes are of particular interest. We experimentally investigated ROL patterns from the submerged macrophyte species Potamogeton crispus and determined how light, temperature and O 2 content in overlying water regulate O 2 micro-distributions in the rhizosphere. Method Planar optodes were firstly used for non-destructive imaging of the O 2 micro-distributions around single roots of P. crispus planted in rhizotrons containing sediment. The dynamic changes in below-ground O 2 concentrations and oxygenation expansion at different light intensities (0–216 μmol photons m −2 ), temperatures (14 and 25 °C) and O 2 content in overlying waters (0–256 μmoL −1 ) were quantified. Results P. crispus -mediated ROL is predominantly localized to the root apices with an average rate of 168.1 ± 21.4 nmol m −2 s −1 under nearly natural conditions (light, saturated overlying water at 14 °C), maintaining a visible oxygenated rhizosphere zone with a radius of 1.33 ± 0.21 mm. ROL is closely correlated with light intensity, suggesting photosynthetic O 2 evolution. In darkness, the rhizosphere O 2 availability is significantly reduced and strongly affected by the O 2 content in overlying water, which passively diffuses into the plant leaves. Elevated temperatures lead to diminished ROL as a result of increased O 2 demand of the surrounding sediment. Conclusion A high O 2 microheterogeneity around P. crispus root apices is firstly demonstrated, and we provide direct empirical evidences that ROL are regulated dynamically by the shifting light, temperature and O 2 content in overlying water.
Concurrent Measurement of O2 Production and Isoprene Emission During Photosynthesis: Pros, Cons and Metabolic Implications of Responses to Light, CO2 and Temperature.

Abstract:Traditional leaf gas exchange experiments have focused on net CO2 exchange (Anet). Here, using California poplar (Populus trichocarpa), we coupled measurements of net oxygen production (NOP), isoprene emissions and δ18O in O2 to traditional CO2/H2O gas exchange with chlorophyll fluorescence, and measured light, CO2 and temperature response curves. This allowed us to obtain a comprehensive picture of the photosynthetic redox budget including electron transport rate (ETR) and estimates of the mean assimilatory quotient (AQ = Anet/NOP). We found that Anet and NOP were linearly correlated across environmental gradients with similar observed AQ values during light (1.25 ± 0.05) and CO2 responses (1.23 ± 0.07). In contrast, AQ was suppressed during leaf temperature responses in the light (0.87 ± 0.28), potentially due to the acceleration of alternative ETR sinks like lipid synthesis. Anet and NOP had an optimum temperature (Topt) of 31°C, while ETR and δ18O in O2 (35°C) and isoprene emissions (39°C) had distinctly higher Topt. The results confirm a tight connection between water oxidation and ETR and support a view of light-dependent lipid synthesis primarily driven by photosynthetic ATP/NADPH not consumed by the Calvin-Benson cycle, as an important thermotolerance mechanism linked with high rates of (photo)respiration and CO2/O2 recycling.

Measurement of O2 Uptake and Evolution in Leaves In Vivo Using Stable Isotopes and Membrane Inlet Mass Spectrometry

Abstract:Oxygen is both product and substrate of photosynthesis and metabolism in plants, by oxygen evolution through water splitting and uptake by photorespiration and respiration. It is important to investigate these processes simultaneously in leaves, especially in response to environmental variables, such as light and temperature. To distinguish between processes that evolve or take up O2 in leaves in the light, in vivo gas exchange of stable isotopes of oxygen and membrane inlet mass spectrometry is used. A closed-cuvette system for gas exchange of leaf discs is described, using the stable isotopes 16O2 and 18O2, with a semi-permeable membrane gas inlet and isotope mass separation and detection by mass spectrometry. Measurement of evolution and uptake, as well as CO2 uptake, at a range of light levels allows composition of a light response curve, here described for French bean (Phaseolus vulgaris) and maize (Zea mays) leaf discs.
A combined application of tunable diode laser absorption spectroscopy and isothermal micro-calorimetry for calorespirometric analysis

Abstract:Calorespirometry is the simultaneous analysis of the rate of heat emission (Rq), O2 consumption (RO2) and CO2 production (RCO2) by living systems such as tissues or organism cultures. The analysis provides useful knowledge about thermodynamic parameters relevant for e.g. biotechnology where parameter based yield maximization (fermentation) is relevant. The determination of metabolism related heat emission is easy and normally done by a calorimeter. However, measuring the amount of consumed O2 and produced CO2 can be more challenging, as additional preparation or instrumentation might be needed. Therefore, tunable diode laser absorption spectroscopy (TDLAS) was investigated as an alternative approach for respirometric analysis in order to facilitate the data collection procedure. The method determines by a spectroscopic laser non-invasively CO2 and O2 gas concentration changes in the respective vial headspaces. The gathered growth data from Pseudomonas aeruginosa cultured in two different scarce media was used to compute respiratory quotient (RQ) and calorespirometric ratios (CRCO2 [Rq/RCO2], CRO2 [Rq/RO2]). A comparison of the computed (experimental) values (for RQ, CRCO2 and CRO2) with values reported in the literature confirmed the appropriateness of TDLAS in calorespirometric studies. Thus, it could be demonstrated that TDLAS is a well-performing and convenient way to evaluate non-invasively respiratory rates during calorespirometric studies. Therefore, the technique is definitively worth to be investigated further for its potential use in research and in diverse productive environments.
Net O2 exchange rates under dark and light conditions across different stem compartments

Abstract:Woody plants display some photosynthetic activity in stems, but the biological role of stem photosynthesis and the specific contributions of bark and wood to carbon uptake and oxygen evolution remain poorly understood. We aimed to elucidate the functional characteristics of chloroplasts in stems of different ages in Fraxinus ornus. Our investigation employed diverse experimental approaches, including microsensor technology to assess oxygen production rates in whole stem, bark, and wood separately. Additionally, we utilized fluorescence lifetime imaging microscopy (FLIM) to characterize the relative abundance of photosystems I and II (PSI : PSII chlorophyll ratio) in bark and wood. Our findings revealed light-induced increases in O2 production in whole stem, bark, and wood. We present the radial profile of O2 production in F. ornus stems, demonstrating the capability of stem chloroplasts to perform light-dependent electron transport. Younger stems exhibited higher light-induced O2 production and dark respiration rates than older ones. While bark emerged as the primary contributor to net O2 production under light conditions, our data underscored that wood chloroplasts are also photosynthetically active. The FLIM analysis unveiled a lower PSI abundance in wood than in bark, suggesting stem chloroplasts are not only active but also acclimate to the spectral composition of light reaching inner compartments.
A Compensating Method for Measuring Carbon Dioxide Exchange, Transpiration, and Diffusive Resistance of Plants under Controlled Environmental Conditions

Abstract:A semi—closed compensating system is described for measuring CO2 exchange and transpiration simultaneously under controlled environmental conditions. The compensating feature permits conditions in the assimilation chamber to be kept uniform and within narrow limits. Temperature, relative humidity, carbon dioxide concentration, wind speed, and light intensity may be varied within the system. Since transpiration and photosynthesis can be measured simultaneously, diffusive resistances to carbon dioxide and water transport may also be calculated from the data. The performance of the apparatus was checked with Ambrosia trifida L. Net photosynthesis occurred at a rate of 32 mg hr—1 dm—2 of leaf area and 30°C under saturating light intensities, and transpiration took place at 5.3 gm hr—1 dm—2 under the same conditions. Both processes were sensitive to CO2 concentrations from 100 to 400 ppm and the temperatures between 15° and 30°C. Respiration was relatively insensitive to temperature below 20°C. Although the response time of the instrument was only a few seconds, net photosynthesis and dark respiration did not reach steady rates until 45—60 min after the lights were turned on or off. Diffusive resistance to CO2 transport were 0.08, 0.8, and 5.2 sec cm—1 for the boundary layer of the leaf, internal gas phase of the leaf, and the mesophyll cells, respectively. Diffuse resistances to water transport were 0.05 and 0.5 sec cm—1 for the boundary layer and internal gas phase of the leaf, respectively.

Abstract:Conventional methods to measure oxygen consumption, such as Clark-type electrodes, have limitations such as requiring a large amount of starting material. Moreover, commercially available kits for high-throughput methods are usually optimized for animal cells and mitochondria. Here, we present a novel method to measure the oxygen consumption rate using a high-throughput assay in isolated mitochondria of European beech seeds. To perform the measurements, we adapted the Agilent Seahorse XF Cell Mito Stress Test Kit protocol for measurements on plant mitochondria.

SlipO2Chip - single-cell respiration under tuneable environments

Abstract:In disciplines like toxicology and pharmacology, oxygen (O2) respiration is a universal metric for evaluating the effects of chemicals across various model systems, including mammalian and microalgal cells. However, for these cells the common practice is to segregate populations into control and exposure groups, which assumes direct equivalence in their responses and does not take into account heterogeneity among individual cells. This lack of resolution impedes our ability to precisely investigate differences among experimental groups with small or limited sample sizes. To overcome this barrier, we introduce SlipO2Chip, an innovative glass microfluidic platform for precisely quantifying single-cell O2 respiration in the coordinated absence and presence of chemical solutes. SlipO2Chip comprises a wet-etched fused silica channel plate on the top and a dry-etched borosilicate microwell plate at the bottom. The microwells are coated with Pt(II) meso-tetra(pentafluorophenyl)porphine (PtTFPP), an O2 sensing optode material and an O2-independent reference dye. A custom 3D-printed holder facilitates the controlled horizontal movement ('slipping') of the channel plate over the microwell plate, thereby establishing or disrupting the fluid path over microwells. Collectively, these design elements enable the immobilization of single-cells in microwells, their exposure to controlled fluid flows, the coordinated opening and closing of microwells and repeated measurements of single-cell O2 respiration. Uniquely, by sequentially executing opening and closing it becomes possible to measure single-cell respiration prior to and after exposure to chemical solutes. In a proof-of-concept application, we utilized SlipO2Chip to measure the impact of increasing exposures of the marine bacterial signal 2-heptyl-4-quinolone (HHQ) on the dark respiration of the diatom Ditylum brightwellii at single-cell resolution. Results revealed a concentration-dependent decrease in per-cell O2 dark respiration, with a maximum reduction of 40.2% observed at HHQ concentrations exceeding 35.5 μM, and a half-maximal effective concentration (EC50) of 5.8 μM, consistent with that obtained via conventional bulk respiration methods. The ability of SlipO2Chip to sequentially assess the effects of chemical substances on single-cell O2 metabolism is advantageous for research where sample volumes are limited, such as clinical biopsies, studies involving rare microbial isolates, and toxicological studies aiming to address exposure effects while accounting for cell-to-cell variability.
Illuminating root-soil mechanics

Abstract:Soil compaction and increased global drought enhance soil mechanical strength and stiffness, which poses threat to soil health and ultimately adversely impacts society. Despite these restrictive conditions, some plant roots are able to grow under these harsh environments. This study was formed to develop a proof of concept measurement system that could be used to quaantify local plant biomechanical pressures during growth in in-situ assays. Using micro cone penetrometer root analogues, we tested the viability of combining X-ray computed tomography (XCT) with X-ray diffraction (XRD) measurements in order to understand whether we can characterize the mechanics during penetration associated with root growth. Cone penetrometers were used to penetrate into surrogate soil materials (e.g. gypsum), as these materials have porous and granular behaviour similar to natural soils but facilitate diffraction measurements due to their well defined atomic structures. Diffraction maps were generated across a sub region of the imaged space in order to quantify changes in the diffraction patterns. Diffraction maps are simplified representations by spectra peaks resulting from integrated Debeye-Scherrerr rings. Shifts in the spectra peaks are associated with deformations of the lattice crystals that comprise the surrogate soil material resulting from applied loads from the micro cone penetrometers. XCT images highlight compaction zones near the cone tip during penetration, with an increase in the compression zones radius as we push the cone deeper into the media. The XRD measurements peak shifts just outside of the local compacted zone of the cone. Qualitative finite element simulations illustrate that this is likely due to local inelastic deformations near the cone tip during penetration. However, the remote elastic deformations can be monitored as long as the inelastic zone remains local to the cone tip. While direct measurements near a root tip might be difficult due to the inelastic nature of the penetration process, the results show promise in terms of gaining insights into the locality of root growth biomechanical processes and the potential to elucidate root strategies under compacted soils in an in-situ test.

Refined imaging of pH and O2 across the passivated soil-root micro-interfaces

Abstract:Herein two self-established planar optode methods （PO） were successfully deployed to in situ,high-resolution investigate the fine-scale and simultaneous variations of micro-environmental conditions e.g.pH and O 2 across the passivated soil-root interfaces.For the first time,our results quantitatively and directly revealed the high degrees of spatiotemporal heterogeneity of p H and O 2 throughout the rhizosphere at the sub-mm scale.Significant differences （P＜0.01） among three Cd treatments were observed.The addition of the attapulgite clays significant elevated the pH micro-environemnts,but had slight effects on the micro-distributions of O 2 within the soil profiles.There was a visible and distinct range of 2～5mm surrounding the rice roots where pH values were generally decreased by 0.31～0.87 and O 2 concentrations were increased to 58.56～82.01μmol/L.Notably higher rhizosphere acidification and O 2 leakage always occurred locally around the basal roots.The higher abilities of rhizosphere acidification and O 2 leakage within the passivated soil-root micro interfaces would potentially promote the reactivation and root uptake of passivated Cd.
A Simple Technique to Measure Stem Respiration Using a Horizontally Oriented Soil Chamber

Abstract:This paper introduces a simple technique, the horizontally oriented soil chamber (HOSC), to measure stem and branch respiration easily and accurately. We extend the function of the LI-6400-09 soil CO2 flux chamber by attaching a custom-built polyvinyl chloride (PVC) collar to the stem surface. For small trees and branches we use pipe bushings to connect the chamber to stem surface. Using this technique we measured stem respiration in a young ponderosa pine (Pinus ponderosa Dougl. ex Laws.) plantation in the Sierra Nevada Mountains near Georgetown, Calif., from June to December 1998. The diurnal and seasonal variations in respiration rate correlate well with the corresponding stem temperature variation. The Q10 values varied from 1.9 to 2.8, which are within the range of Q10 values (1.3-3.3) reported in previous studies. The range of our stem respiration results (3.5-7.2 µmol·m-2·s-1) compares favorably with previous studies on young ponderosa pine trees. This technique provides an alternative to measure stem respiration, which employs widely used, commercially available, portable respiration measurement equipment and requires almost no additional equipment, especially for current owners of LI-6400 systems. Thus, the HOSC technique is appropriate for examining spatial variation of stem respiration.
Evaluating high time-resolved changes in carbon isotope ratio of respired CO2 by a rapid in-tube incubation technique

Abstract:Recent insights into fractionation during dark respiration and rapid dynamics in isotope signatures of leaf- and ecosystem-respired CO(2) indicate the need for new methods for high time-resolved measurements of the isotopic signature of respired CO(2) (delta(13)C(res)). We present a rapid and simple method to analyse delta(13)C(res) using an in-tube incubation technique and an autosampler for small septum-capped vials. The effect of storage on the delta(18)O and delta(13)C ratios of ambient CO(2) concentrations was tested with different humidity and temperatures. delta(13)C ratios remained stable over 72 h, whereas delta(18)O ratios decreased after 24 h. Storage at 4 degrees C improved the storage time for delta(18)O. Leaves or leaf discs were incubated in the vials, flushed with CO(2)-free air and respired CO(2) was automatically sampled within 5 min on a microGas autosampler interfaced to a GV-Isoprime isotope ratio mass spectrometer. Results were validated by simultaneous on-line gas-exchange measurements of delta(13)C(res) of attached leaves. This method was used to evaluate the short-term (5-60 min) and diurnal dynamics of delta(13)C(res) in an evergreen oak (Quercus ilex) and a herb (Tolpis barbata). An immediate depletion of 2-4 per thousand from the initial delta(13)C(res) value occurred during the first 30 min of darkening. Q. ilex exhibited further a substantial diurnal enrichment in delta(13)C(res) of 8 per thousand, followed by a progressive depletion during the night. In contrast, T. barbata did not exhibit a distinct diurnal pattern. This is in accordance with recent theory on fractionation in metabolic pathways and may be related to the different utilisation of the respiratory substrate in the fast-growing herb and the evergreen oak. These data indicate substantial and rapid dynamics (within minutes to hours) in delta(13)C(res), which differed between species and probably the growth status of the plant. The in-tube incubation method enables both high time-resolved analysis and extensive sampling across different organs, species and functional types.
Assessing Soil Co2 Efflux Using Continuous Measurements of Co2 Profiles in Soils with Small Solid-State Sensors

Abstract:This paper describes a new method to monitor continuously soil CO2 profiles using small solid-state CO2 sensors buried at different depths of the soil. Based on the measurement of soil CO2 profile and a gaseous diffusivity model, we estimated soil CO2 efflux, which was mainly from heterotrophic respiration, and its temporal variation in a dry season in a Mediterranean savanna ecosystem in California. The daily mean values of CO2 concentrations in soils had small variation, but the diurnal variation was significant and correlated well with soil temperature. The daily mean CO2 concentration remained steady at 396 μmol mol−1 at 2 cm depth during the dry summer from days 200 to 235 in 2002. Over the same period, CO2 concentration decreased from 721 to 611 μmol mol−1 at 8 cm depth, and from 1044 to 871 μmol mol−1 at 16 cm. The vertical soil CO2 concentrations changed almost linearly with depth up to 16 cm, but the gradient varied over time. Based on the soil CO2 gradient and the diffusion coefficient estimated from the Millington–Quirk model, continuous soil CO2 efflux was calculated. The daily mean values of CO2 efflux slightly decreased from 0.43 to 0.33 μmol m−2 s−1 with a mean of 0.37 μmol m−2 s−1. The mean diurnal range of CO2 efflux was greater than the range of daily mean CO2 efflux within the study period. The diurnal variation of soil CO2 efflux ranged from 0.32 to 0.45 μmol m−2 s−1 with the peak value reached between 14:30 and 16:30 h. This pattern corresponded well with the increase in soil temperatures during this time. By plotting CO2 efflux vs. soil temperature, we found that CO2 efflux correlated exponentially with soil temperature at the depth of 8 cm, with R2 of 0.86 and Q10 of 1.27 in the summer dry season. The Q10 value increased with the depth of soil temperature measurements. The high correlation between CO2 efflux and temperature explains the diurnal pattern of CO2 efflux, but moisture may become another factor driving the seasonal pattern when moisture changes over seasons. The estimated CO2 efflux using this method was very close to chamber measurements, suggesting that this method can be used for long-term continuous measurements of soil CO2 efflux.

Abstract:Root-triggered microscale variations in O2 distribution in the rhizosphere of young Phragmites australis are important for nutrient removal in sediments. In this study, the micro-scale O2 dynamics and the small-scale changes of soluble reactive phosphorus (SRP) and ammonium (NH4+) in the rhizosphere of P. australis were investigated using planar optodes and high-resolution dialysis (HR-Peeper), respectively. Results suggested that root O2 leakage has a highly variable distribution depending on the stage of root growth, the site of O2 leakage gradually shift from the entire emerging main roots to the main root tip and subsequently shifted the emerging lateral roots. The O2 concentration increased in the rhizosphere with increasing light intensity and O2 levels in the overlying water. Continuous O2 release from the lateral roots causes the formation of iron plaque on the surface of lateral roots, which reduce the mobility of P by adsorption of iron plaque in the rhizosphere. The oscillation of oxic-anoxic root zones improves nitrogen removal through the processes of anammox, heterotrophic denitrification and nitrification. This work from the micro-scale demonstrates that the O2 concentration is the spatio-temporal variations in the rhizosphere, and it presents an important role for nutrient removal in sediments.

Abstract:Plant uptake and transportation of resources, such as water, nutrients and photosynthates, are crucial for plant growth, and a main driver of ecosystem functioning and community responses to environmental changes (Evert, 2006; Ma et al., 2018; Steidinger et al., 2019). Plant roots form a highly branched system with great heterogeneity in structure and function, hierarchically organized from lower order distal roots to higher order basal roots. Abstract:Knowledge on the availability of dissolved oxygen inside microfluidic cell culture systems is vital for recreating physiological-relevant microenvironments and for providing reliable and reproducible measurement conditions. It is important to highlight that in vivo cells experience a diverse range of oxygen tensions depending on the resident tissue type, which can also be recreated in vitro using specialized cell culture instruments that regulate external oxygen concentrations. While cell-culture conditions can be readily adjusted using state-of-the-art incubators, the control of physiological-relevant microenvironments within the microfluidic chip, however, requires the integration of oxygen sensors. Although several sensing approaches have been reported to monitor oxygen levels in the presence of cell monolayers, oxygen demands of microfluidic three-dimensional (3D)-cell cultures and spatio-temporal variations of oxygen concentrations inside two-dimensional (2D) and 3D cell culture systems are still largely unknown. To gain a better understanding on available oxygen levels inside organ-on-a-chip systems, we have therefore developed two different microfluidic devices containing embedded sensor arrays to monitor local oxygen levels to investigate (i) oxygen consumption rates of 2D and 3D hydrogel-based cell cultures, (ii) the establishment of oxygen gradients within cell culture chambers, and (iii) influence of microfluidic material (e.g., gas tight vs. gas permeable), surface coatings, cell densities, and medium flow rate on the respiratory activities of four different cell types. We demonstrate how dynamic control of cyclic normoxic-hypoxic cell microenvironments can be readily accomplished using programmable flow profiles employing both gas-impermeable and gas-permeable microfluidic biochips.

In-situ measurements of dissolved gases in xylem sap as tracers in plant physiology

Abstract:Trees transport gases from below ground into the atmosphere through the process of transpiration. Tracing gases transported through this mechanism continuously and under field conditions remains an experimental challenge. Here we measured gases dissolved in tree sap in-situ and in real time, aiming to simultaneously analyse the transport of several gases (He, Ar, Kr, N2, O2, CO2) from the soil, through the trees, into the atmosphere. We constructed and inserted custom-made semi-permeable membrane probes in the xylem of a fir tree and measured gas abundances at different heights using a portable gas equilibrium membrane-inlet mass spectrometer ('miniRUEDI'). With this method we were able to continuously measure the abundances of He, Ar, Kr, N2, O2, CO2 in sap over several weeks. We observed diurnal variations of CO2 and O2 concentrations that reflected tree physiological activities. As a proof of concept that trees do uptake dissolved gases in soil water, we irrigated the tree with He-enriched water in a tracer experiment, and were able to determine upwards sap flow velocity. Measurements of inert gases together with reactive species as CO2 and O2 allows to separate physical transport and exchange of gases derived from the soil or the atmosphere from biological reactions. We discuss the opportunities that our technique provides for continuous in-situ measurements of gases in tree sap.
A New Non-invasive Technique for Measuring 3D-Oxygen Gradients in Wells During Mammalian Cell Culture

Abstract:Oxygen tension plays an important role in overall cell function and fate, regulating gene expression, and cell differentiation. Although there is extensive literature available that supports the previous statement, little information is to be found about accurate O2 measurements during culture. In fact, O2 concentration at the cell layer during culture is commonly assumed to be equal to that of the incubator atmosphere. This assumption does not consider oxygen diffusion properties, cell type, cell density, media composition, time in culture nor height of the cell culture medium column. In this study, we developed a non-invasive, optical sensor foil-based technique suitable for measuring the 3D oxygen gradient that is formed during cell culture as a result of normal cell respiration. For this propose, we created a 3D printed ramp to which surface an oxygen optode sensor foil was attached. The ramps were positioned inside the culture wells of 24 well plate prior cell seeding. This set up in conjunction with the VisiSens TD camera system allows to investigate the oxygen gradient formation during culture. Cultivation was performed with three different initial cell densities of the cell line A549 that were seeded on the plate containing the ramps with the oxygen sensors. The O2 gradient obtained after 96 h of culture showed significantly lower O2 concentrations closer to the bottom of the well in high cell density cultures compared to that of lower cell density cultures. Furthermore, it was very interesting to observe that even with low cell density culture, oxygen concentration near the cell layer was lower than that of the incubator atmosphere. The obtained oxygen gradient after 96 h was used to calculate the oxygen consumption rate (OCR) of the A549 cells, and the obtained value of ~100 fmol/h/cell matches the OCR value already reported in the literature for this cell line. Moreover, we found our set up to be unique in its ability to measure oxygen gradient formation in several wells of a cell culture plate simultaneously and in a non-invasive manner.

Simultaneous Measurement of Gross Oxygen Evolution and Underlying Photosynthetic Redox Reactions: A Case Study Using Cyanobacteria

Abstract:In phytoplankton, the intricate balance between respiration and photosynthesis is co-regulated to ensure efficient energy management and adaptation to varying environmental conditions. In cyanobacteria, both processes occur on the same membrane, sharing electron transport carriers within the same cellular compartment. By studying the interaction between photosynthesis and respiration, we can better understand how cyanobacteria balance their energetic budget for survival. In this study, we present an integrated approach that combines tracking gas exchange between cyanobacteria and their environment with analysing the redox kinetics of the underlying photosynthetic electron transport chain. This combined system allows for real-time, simultaneous acquisition of respiration and photosynthesis data. For example, it enabled us to show that the electron transport rate generated by photosystem II, translated to in-vivo oxygen concentration, equals the actual concentration of oxygen produced by water splitting plus the amount of oxygen respired. We further demonstrate that our system can accurately assess light respiration in wild-type strains of cyanobacteria, which amounts to 1/10 of their photosynthetic activity under optimal growth conditions. This level of accuracy was previously achievable only with specific cyanobacteria mutants. We envision applying this system in monitoring programs to elaborate on the role of photosynthetic light reactions within the broader context of primary productivity and to understand its dynamics in response to fluctuations in external environmental conditions.
