Abstract:Globally, metabolic dysfunction-associated steatotic liver disease (MASLD), previously termed nonalcoholic fatty liver disease (NAFLD), is one of the most common liver disorders and is strongly associated with copper deficiency. To explore the potential effects and mechanisms of Lactiplantibacillus plantarum LPJZ-658, copper deficiency combined with a high-sugar diet-induced MASLD mouse model was utilized in this study. We fed 40-week-old (middle-aged) male C57BL/6 mice a copper-deficient and high-sugar diet for 16 weeks (CuDS), with supplementary LPJZ-658 for the last 6 weeks (CuDS + LPJZ-658). In this study, we measured body weight, liver weight, and serum biochemical markers. Lipid accumulation, histology, lipidomics, and sphingolipid metabolism-related enzyme expression were investigated to analyze liver function. Untargeted metabolomics was used to analyze the serum and the composition and abundance of intestinal flora. In addition, the correlation between differential liver lipid profiles, serum metabolites, and gut flora at the genus level was measured. The results show that LPJZ-658 significantly improves abnormal liver function and hepatic steatosis. The lipidomics analyses and metabolic pathway analysis identified sphingolipid, retinol, and glycerophospholipid metabolism as the most relevant metabolic pathways that characterized liver lipid dysregulation in the CuDS group. Consistently, RT-qPCR analyses revealed that the enzymes catalyzing sphingolipid metabolism that were significantly upregulated in the CuDS group were downregulated by the LPJZ-658 treatment. In addition, the serum metabolomics results indicated that the linoleic acid, taurine and hypotaurine, and ascorbate and aldarate metabolism pathways were associated with CuDS-induced MASLD. Notably, we found that treatment with LPJZ-658 partially reversed the changes in the differential serum metabolites. Finally, LPJZ-658 effectively regulated intestinal flora abnormalities and was significantly correlated with differential hepatic lipid species and serum metabolites. In conclusion, we elucidated the function and potential mechanisms of LPJZ-658 in alleviating copper deficiency combined with sugar-induced middle-aged MASLD and hope this will provide possible treatment strategies for improving MASLD.

Associations of Cumulative Exposure to Metal Mixtures with Metabolic Dysfunction-Associated Fatty Liver Disease in a Chinese Aged Population

Abstract:BACKGROUND AND AIM: Few studies have demonstrated the potential effects of metal mixtures on metabolic dysfunction-associated fatty liver disease (MAFLD). Here, we examined the associations between whole blood metal mixtures and prevalence of MAFLD among an aged population in China. METHODS: a cross-sectional study of community-dwelling elderly adults were conducted in China. Whole blood concentrations of fourteen metals were measured by inductively coupled plasma mass spectrometry (ICP-MS). MAFLD was ascertained based on ultrasonographic evidence of hepatic steatosis and the presence of overweight/obesity, type 2 diabetes mellitus, or metabolic dysregulation. We performed logistic regression and restricted cubic spline (RCS) models to independently estimate the associations between metal concentrations and MAFLD prevalence. Environmental Risk Score (ERS) constructed by adaptive elastic net (AENET) and Bayesian kernel machine regression (BKMR) analysis were utilized to examine joint effects of metal mixtures and metal interactions on MAFLD. RESULTS: In the models adjusting for multiple covariates, lower blood copper and higher selenium, iron and mercury were significantly associated with the elevated prevalence of MAFLD. Non-linear effects of copper, iron, magnesium, manganese and selenium on MAFLD were observed in RCS models. Similar metals (copper, selenium, zinc, iron, cadmium and mercury) were selected as crucial predictors for MAFLD prevalence by AENET and BKMR models. The association between ERS and MAFLD was estimated and the ERS was significantly associated with the elevated OR for MAFLD. CONCLUSIONS: Our findings provide evidence that exposure to blood copper, iron, selenium and mercury are associated with the prevalence of MAFLD. Multiple metals exposure may adversely affect MAFLD, with copper, selenium, zinc, iron, cadmium and mercury being the major contributors. Further researches are needed to clarify the cumulative effects of metal mixture in a larger population. High dietary Fructose Drives Metabolic Dysfunction-Associated Steatotic Liver Disease via Activating ubiquitin-specific peptidase 2/11β-hydroxysteroid dehydrogenase type 1 Pathway in Mice

Abstract:Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most common cause of chronic liver-related morbidity and mortality. Though high fructose intake is acknowledged as a metabolic hazard, its role in the etiology of MASLD requires further clarification. Here, we demonstrated that high dietary fructose drives MASLD development and promotes MASLD progression in mice, and identified Usp2 as a fructose-responsive gene in the liver. Elevated USP2 levels were detected in the hepatocytes of MASLD mice; a similar increase was observed following fructose exposure in primary hepatocytes and mouse AML12 cells. Notably, hepatocytes overexpressing USP2 presented with exaggerated lipid accumulation and metabolic inflammation when exposed to fructose. Conversely, USP2 knockdown mitigated these fructose-induced changes. Furthermore, USP2 was found to activate the C/EBPα/11β-HSD1 signaling, which further impacted the equilibrium of cortisol and cortisone in the circulation of mice. Collectively, our findings revealed the role of dietary fructose in MASLD pathogenesis and identified the USP2-mediated C/EBPα/ 11β-HSD1 signaling as a potential target for the management of MASLD.

Abstract:Metabolites produced as intermediaries or end-products of microbial metabolism provide crucial signals for health and diseases, such as metabolic dysfunction-associated steatotic liver disease (MASLD). These metabolites include products of the bacterial metabolism of dietary substrates, modification of host molecules [such as bile acids (BAs), trimethylamine-N-oxide, and short-chain fatty acids], or products directly derived from bacteria. Recent studies have provided new insights into the association between MASLD and the risk of developing chronic kidney disease (CKD). Furthermore, alterations in microbiota composition and metabolite profiles, notably altered BAs, have been described in studies investigating the association between MASLD and the risk of CKD. This narrative review discusses alterations of specific classes of metabolites, BAs, fructose, vitamin D, and microbiota composition that may be implicated in the link between MASLD and CKD.

Abstract:Objectives: Over consumption of dietary fructose contributes to the development of metabolic dysfunction associated-steatotic liver disease (MASLD), but the role of fructose transporter in the intestine, glucose transporter 5 (GLUT5), is not clear. We aimed to investigate the role of GLUT5 as a novel therapeutic target for MASLD. Methods: Adult male C57BL/6J mice were fed by different doses of fructose in drinking water for 8 weeks from 4%-25%. The intestinal expression of GLUT5, intestinal and liver histology were measured while gut metagenome and metabolome were evaluated. The intestinal-specific Glut5 knock out mice Glut5flox/flox Vil1-Cre ki/wt (Glut5-/-) were built and fed by high fructose alone (HF) or combined with high fat diet (HFF) for 8 weeks to confirm the role of Glut5. Furthermore, we fed 5,6,7-Trimethoxyflavone (TMF), a natural product identified as GLUT5 transporter inhibitor, to wide type (WT) mice to validate its prevention effect on high fructose-induced liver injury. Results: The intestinal GLUT5 expression was significantly increased in WT mice fed by HF for 8 weeks, which developed gut dysbiosis, impaired liver function, and metabolic abnormalities. Compared with WT mice, after feeding HF or HFF diet for 8 weeks, the intestinal and liver damage were alleviated in Glut5-/- mice. Moreover, we confirmed that the natural product, TMF, could effectively inhibited the intestinal expression of GLUT5 and improved the development of MASLD in WT mice. Conclusions: The intestinal GLUT5 is a potential therapeutic target to alleviate the high fructose-induced MASLD and the natural product of GLUT5 inhibitor-TMF can be a promising candidate for MASLD prevention and intervention. Abstract:Iron deficiency is the number one nutritional problem worldwide. Iron uptake is regulated at the intestine and is highly influenced by the gut microbiome. Blood from the intestines drains directly into the liver, informing iron status and gut microbiota status. Changes in either iron or the microbiome are tightly correlated with the development of metabolic dysfunction-associated steatotic liver disease (MASLD). To investigate the underlying mechanisms of the development of MASLD that connect altered iron metabolism and gut microbiota, we compared specific pathogen free (SPF) or germ-free (GF) mice, fed a normal or low-iron diet. SPF mice on a low-iron diet showed reduced serum triglycerides and MASLD. In contrast, GF low-iron diet-fed mice showed increased serum triglycerides and did not develop hepatic steatosis. SPF mice showed significant changes in liver lipid metabolism and increased insulin resistance that was dependent upon the presence of the gut microbiota. We report that total body loss of mitochondrial iron importer Mitoferrin2 (Mfrn2−/−) exacerbated the development of MASLD on a low-iron diet with significant lipid metabolism alterations. Our study demonstrates a clear contribution of the gut microbiome, dietary iron, and Mfrn2 in the development of MASLD and metabolic syndrome.

Abstract:The prevalence of metabolic-dysfunction-associated steatotic liver disease (MASLD) is alarmingly high; it is estimated to affect up to a quarter of the global population, making it the most common liver disorder worldwide. MASLD is characterized by excessive hepatic fat accumulation and is commonly associated with comorbidities such as obesity, dyslipidemia, and insulin resistance; however, it can also manifest in lean individuals. Therefore, it is crucial to develop effective therapies for this complex condition. Currently, there are no approved medications for MASLD treatment, so there is a pressing need to investigate alternative approaches. Extensive research has characterized MASLD as a multifaceted disease, frequently linked to metabolic disorders that stem from dietary habits. Evidence suggests that changes in the gut microbiome play a fundamental role in the development and progression of MASLD from simple steatosis to steatohepatitis and even hepatocellular carcinoma (HCC). In this review, we critically examine the literature on the emerging field of gut-microbiota-based therapies for MASLD and metabolic-dysfunction-associated steatohepatitis (MASH), including interventions such as fecal microbiota transplantation (FMT), probiotics, prebiotics, short-chain fatty acids, antibiotics, metabolic pathway targeting, and immune checkpoint kinase blockade.

Abstract:The study of faeces offers a unique opportunity to observe cooperation between the microbiome and the metabolism of mammalian hosts, an essential element in the study of the human metabolome. In the present study, a global metabolomics approach was used to identify metabolites differentially excreted in the faeces of cirrhotic patients compared to controls.Seventeen cirrhotic patients and 24 healthy individuals were recruited. Faecal metabolites were detected through non-targeted reversed-phase ultra-performance liquid chromatography coupled to electrospray ionization quadrupole time-of-flight mass spectrometry.A total of 9215 peaks were detected. Using unequal variance t-tests, 2393 peaks were observed with P≤0.05, approximately 74.0% of which were due to decreased faecal metabolite concentrations in liver cirrhosis vs. healthy controls. Integrating multivariate data analyses, we identified six major groups of metabolites. Relative levels of identified metabolites were as follows: strong increase in lysophosphatidylcholines, aromatic amino acids, fatty acids, and acylcarnitines, and a dramatic decrease in bile acids and bile pigments.With severe hepatic injury in patients with liver cirrhosis, malabsorption occurs along with disorders of fatty acid metabolism, potentially due to changes in gut microflora.

Abstract:Metabolic dysfunction-associated steatotic liver disease (MASLD), one of the leading causes of chronic liver disorders, is characterized by hepatic lipid accumulation. MASLD causes alterations in the antioxidant defense system, lipid, and drug metabolism, resulting in impaired antioxidant status, hepatic metabolic processes, and clearance of therapeutic drugs, respectively. In the MASLD pathogenesis, dysregulated epigenetic mechanisms (e.g., histone modifications, DNA methylation, microRNAs) play a substantial role. In this study, the development of MASLD was investigated in mice fed a high-fat, high-fructose, and high-cholesterol (FFC) diet from 2 months of age, mice treated neonatally with monosodium glutamate (MSG) on a standard diet (STD), and mice treated with MSG on an FFC diet at 7 months of age and compared to control mice (C) on STD. Changes in liver histology, detoxification enzymes, epigenetic regulation, and genes involved in lipid metabolism were characterized and compared. The strong liver steatosis was observed in MSG STD, C FFC, and MSG FFC, with significant fibrosis in the latter one. Moreover, substantial alterations in hepatic lipid metabolism, epigenetic regulatory factors, and expressions and activities of various detoxification enzymes (namely superoxide dismutase, catalase, and carbonyl reductase 1) were observed in MASLD mice compared to control mice. miR-200b-3p, highly significantly upregulated in both FFC groups, could be considered as a potential diagnostic marker of MASLD. The MSG mice fed FFC seem to be a suitable model of MASLD characterized by both liver steatosis and fibrosis and substantial metabolic dysregulation.

Abstract:Background/Objectives: Metabolic dysfunction-associated steatotic liver disease (MASLD) is a growing global health problem closely linked to dietary habits, particularly high fructose consumption. This study investigates the combined effects of fructose and common food preservatives (sodium benzoate, sodium nitrite, and potassium sorbate) on the development and progression of MASLD. Methods: We utilized a human microbiota-associated mouse model, administering 10% fructose with or without preservatives for 11 weeks. Liver histology, hepatic gene expression (microarray analysis), biochemical markers, cytokine profiles, intestinal permeability, and gut microbiome composition (16S rRNA and Internal Transcribed Spacer (ITS) sequencing) were evaluated. Results: Fructose and potassium sorbate synergistically induced liver pathology characterized by increased steatosis, inflammation and fibrosis. These histological changes were associated with elevated liver function markers and altered lipid profiles. The treatments also induced significant changes in both the bacterial and fungal communities and disrupted intestinal barrier function, leading to increased pro-inflammatory responses in the mesenteric lymph nodes. Liver gene expression analysis revealed a wide range of transcriptional changes induced by fructose and modulated by the preservative. Key genes involved in lipid metabolism, oxidative stress, and inflammatory responses were affected. Conclusions: Our findings highlight the complex interactions between dietary components, gut microbiota, and host metabolism in the development of MASLD. The study identifies potential risks associated with the combined consumption of fructose and preservatives, particularly potassium sorbate. Our data reveal new mechanisms that are involved in the development of MASLD and open up a new avenue for the prevention and treatment of MASLD through dietary interventions and the modulation of the microbiome.

Comparison of the Microbiome-Metabolome Response to Copper Sulfate and Copper Glycinate in Growing Pigs

Abstract:This study aims to compare the fecal microbiome-metabolome response to copper sulfate (CuSO4) and copper glycinate (Cu-Gly) in pigs. Twelve Meishan gilts were allocated into the CuSO4 group and the Cu-Gly group (fed on a basal diet supplemented with 60 mg/kg copper from CuSO4 or Cu-Gly) paired in litter and body weight. After a two-week feeding trial, the Cu-Gly group had a higher copper digestibility, blood hemoglobin, and platelet volume and higher levels of plasma iron and insulin-like growth factor-1 than the CuSO4 group. The Cu-Gly treatment increased the abundance of the Lachnospiraceae family and the genera Lachnospiraceae XPB1014, Corprococcus_3, Anaerorhabdus_furcosa_group, Lachnospiraceae_FCS020_group, and Lachnospiraceae_NK4B4_group and decreased the abundance of the Synergistetes phylum and Peptostreptococcaceae family compared to the CuSO4 treatment. Moreover, the Cu-Gly group had a lower concentration of 20-Oxo-leukotriene E4 and higher concentrations of butyric acid, pentanoic acid, isopentanoic acid, coumarin, and Nb-p-Coumaroyl-tryptamine than the CuSO4 group. The abundance of Synergistetes was positively correlated with the fecal copper content and negatively correlated with the fecal butyric acid content. The abundance of the Lachnospiraceae_XPB1014_group genus was positively correlated with the plasma iron level and fecal contents of coumarin and butyric acid. In conclusion, Cu-Gly and CuSO4 could differentially affect fecal microbiota and metabolites, which partially contributes to the intestinal health of pigs in different manners.

Abstract:Iron (Fe) and Copper (Cu) are essential trace minerals that are important in various biological processes within the body. High iron intake has been associated with perturbations in copper metabolism leading to copper deficiency in rats and mice. We hypothesized that high dietary iron blocks intestinal copper transport via the divalent metal transporter 1 (DMT1), which is essential for iron absorption but can also transport other divalent cations, including possibly copper. Prior to conducting experiments, to evaluate the effect of high iron intake on copper status, pilot studies were conducted in male and female intestine-specific DMT1 KO (DMT1 int/int ) mice and controls (DMT1 fl/fl ) to make both genotypes similarly iron deficient/anemic. At weanling, mice were divided into groups and administered IP iron dextran at doses of 0.5, 0.75, or 1.0 mg (DMT1 int/int ), or PBS (DMT1 fl/fl ), to raise the Hb levels in the DMT1 int/int mice since they are severely anemic. DMT1 int/int mice were then fed a 50-ppm control iron AIN-93G diet, while DMT1 fl/fl mice were fed a 2-3 ppm (iron-deficient) diet, for 4 weeks. Weekly hemoglobin (Hb) measurements were taken via tail vein bleeds. For copper studies, iron status in weanling mice of both genotypes and sexes was modified based on pilot studies, and subsequently, mice were switched to experimental diets containing 1x iron (50 ppm Fe) with marginal copper (2-3 ppm Cu) or 20x iron (1000 ppm Fe) with marginal copper for 3 weeks. Biomarkers of iron and copper status were assessed including hemoglobin content and hematocrit (Hct), serum, and tissue nonheme iron concentration, blood transferrin saturation, liver hepcidin and copper transporter (Ctr1) expression and serum ceruloplasmin (Cp) activity. Outcomes showed that administration of 0.75 mg iron dextran at weanling combined with dietary intervention resulted in lowered and comparable iron status in mice of both genotypes and sexes as indicated by Hb, Hct and hepatic nonheme iron and liver hepcidin expression. In dietary copper studies, male and female DMT1 fl/fl mice fed a high iron / marginal copper diet loaded iron in the liver and spleen, while DMT1 int/int mice had significantly lower liver and spleen nonheme iron when fed the same diet. Liver hepcidin expression was elevated in both genotypes with high iron / marginal copper feeding while no changes were observed in Ctr1 expression between genotypes or diets. Serum Cp activity was decreased by 48% in female DMT1 int/int mice as compared to a 23% decrease in DMT1 fl/fl mice fed the high iron / marginal copper diet. These findings suggest that high dietary iron feeding in DMT1 int/int mice does not cause severe iron loading but leads to lower copper content when compared to DMT1 fl/fl mice. Abstract:Background Clinical evidence substantiates a link between inflammatory bowel disease (IBD), particularly Crohn's disease (CD), and metabolic dysfunction associated steatotic liver disease (MASLD). This study aims to explore the underlying molecular mechanisms responsible for this association. Methods MASLD was induced by administering high-fat and western diets, while IBD was induced using dextran sulfate sodium salt (DSS) and the Il10 KO mice model. The investigation into the role of secondary bile acids (SBA) in ileitis involved employing metagenomic sequencing, conducting metabolomics detection, performing fecal bacterial transplantation, and constructing CD8 + T cell specific gene knockout mice. Results In MASLD+DSS and Il10 KO MASLD mice, we observed ileitis characterized by T cell infiltration and activation in the terminal ileum. This condition resulted in decreased bile acid levels in the portal vein and liver, inhibited hepatic Farnesol X receptor (FXR) activation, and exacerbated MASLD. Metagenomic and metabolomic analysis of ileum contents revealed increased Clostridium proliferation and elevated SBA levels in MASLD-associated ileitis. Experiments using germ-free mice and fecal microbiota transplantation suggested an association between SBA and MASLD-related ileitis. In vitro , SBA promoted CD8 + T cell activation via the TGR5, mTOR, and Oxidative Phosphorylation (OXPHOS) pathways. In vivo , TGR5 knockout in CD8 + T cells effectively alleviated ileitis and worsened the MASLD phenotype. Clinical data further supported these findings, demonstrating a positive correlation ileitis and MASLD. Conclusion MASLD-induced changes in intestinal flora result in elevated levels of SBA in the ileum. In the presence of compromised intestinal barrier, this leads to severe CD8 + T cell-mediated ileitis through the TGR5/mTOR/OXPHOS signaling pathway. Ileitis-induced tissue damage impairs enterohepatic circulation, inhibits hepatic FXR activation, and exacerbates the MASLD phenotype. Impact and implications Our study provides a comprehensive investigation of the interplay and underlying mechanisms connecting ileitis and MASLD. SBA produced by intestinal bacteria, as the critical link between MASLD and ileitis. This compound exerts its influence by disrupting liver lipid metabolism through the promotion of CD8 + T cell-mediated ileitis. In future endeavors to prevent and treat MASLD, it is essential to thoroughly account for the impact of the intestinal tract, especially the ileum, on liver function via the enterohepatic circulation.

Optical Characteristics of Polycrystalline 3C-SiC for Harsh Environments

Abstract:Raman spectra of poly (polycrystalline) 3C-SiC thin films, which were deposited on the oxidized Si substrate by APCVD, have been measured. They were used to study the mechanical characteristics of poly 3C-SiC grown in various temperatures. TO and LO modes of 2.0 mum poly 3C-SiC grown at 1180degC occurred at 794.4 and 965.7 cm-1. Their FWHMs (full width half maximum) were used to investigate the stress and the disorder of 3C-SiC. The broad FWHM can explain that the crystallinity of 3C-SiC grown at 1180degC becomes polycrystalline instead of the disordered crystal. The ratio of intensity I(LO)/I(TO) ap 1.0 means that the crystal defect of 3C-SiC/SiO2/Si is small. The biaxial stress of poly 3C-SiC was obtained as 428 MPa. In the interface of 3C-SiC/SiO2, the phonon mode of C-O bonding appeared at 1122.6 cm-1. The phonon modes related to D and G bands of C-C bonding were measured at 1355.8 and 1596.8 cm-1 respectively.
Lysosomal SLC46A3 modulates hepatic cytosolic copper homeostasis

Abstract:Abstract The environmental contaminant 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) causes hepatic toxicity associated with prominent lipid accumulation in humans. Here, the authors report that the lysosomal copper transporter SLC46A3 is induced by TCDD and underlies the hepatic lipid accumulation in mice, potentially via effects on mitochondrial function. SLC46A3 was localized to the lysosome where it modulated intracellular copper levels. Forced expression of hepatic SLC46A3 resulted in decreased mitochondrial membrane potential and abnormal mitochondria morphology consistent with lower copper levels. SLC46A3 expression increased hepatic lipid accumulation similar to the known effects of TCDD exposure in mice and humans. The TCDD-induced hepatic triglyceride accumulation was significantly decreased in Slc46a3 −/− mice and was more pronounced when these mice were fed a high-fat diet, as compared to wild-type mice. These data are consistent with a model where lysosomal SLC46A3 induction by TCDD leads to cytosolic copper deficiency resulting in mitochondrial dysfunction leading to lower lipid catabolism, thus linking copper status to mitochondrial function, lipid metabolism and TCDD-induced liver toxicity.

Abstract:Background: High-copper diets have been widely used to promote growth performance of pigs, but excess copper supplementation can also produce negative effects on ecosystem stability and organism health. High-copper supplementation can damage the intestinal barrier and disturb the gut microbiome community. However, the specific relationship between high-copper-induced intestinal damage and gut microbiota or its metabolites is unclear. Objective: Using fecal microbiota transplantation and metagenomic sequencing, responses of colonic microbiota to a high-copper diet was profiled. In addition, via comparison of specific bacteria and its metabolites rescue, we investigated a network of bacteria-metabolite interactions involving conversion of specific metabolites as a key mechanism linked to copper-induced damage of the colon. Results: High copper induced colonic damage, Lactobacillus extinction, and reduction of SCFA (acetate and butyrate) concentrations in pigs. LefSe analysis and q-PCR results confirmed the extinction of L. johnsonii. In addition, transplanting copper-rich fecal microbiota to ABX mice reproduced the gut characteristics of the pig donors. Then, L. johnsonii rescue could restore decreased SCFAs (mainly acetate and butyrate) and colonic barrier damage including thinner mucus layer, reduced colon length, and tight junction protein dysfunction. Given that acetate and butyrate concentrations exhibited a positive correlation with L. johnsonii abundance, we investigated how L. johnsonii exerted its effects by supplementing acetate and butyrate. L. johnsonii and butyrate administration but not acetate could correct the damaged colonic barrier. Acetate administration had no effects on butyrate concentration, indicating blocked conversion from acetate to butyrate. Furthermore, L. johnsonii rescue enriched a series of genera with butyrate-producing ability, mainly Lachnospiraceae NK4A136 group. Conclusions: For the first time, we reveal the microbiota-mediated mechanism of high-copper-induced colonic damage in piglets. A high-copper diet can induce extinction of L. johnsonii which leads to colonic barrier damage and loss of SCFA production. Re-establishment of L. johnsonii normalizes the SCFA-producing pathway and restores colonic barrier function. Mechanistically, Lachnospiraceae NK4A136 group mediated conversion of acetate produced by L. johnsonii to butyrate is indispensable in the protection of colonic barrier function. Collectively, these findings provide a feasible mitigation strategy for gut damage caused by high-copper diets. Video Abstract.

Abstract:Aberration of the "gut-liver axis" contributes to the development and progression of metabolic dysfunction-associated steatotic liver disease (MASLD). Here, we use multi-omics to analyze the gut microbiota composition and metabolic profile of patients with type-2 diabetes mellitus (T2DM). T2DM patients were screened for liver disease by blood tests, ultrasound, and liver stiffness measurements. Stool microbiota was analyzed by 16S rRNA gene sequencing; metabolomic profiling by Nuclear Magnetic Resonance spectroscopy and Ultra-High Performance-Mass Spectrometry. Microbiome and metabolic signatures were analyzed in the whole cohort and in matched subsets to identify signatures specific for steatosis (MASLD±) or fibrosis (Fibrosis±). Gut permeability was assessed in-vitro using monolayers of MDCK cells and trans-epithelial electric resistance (TEER). Cytokine profile was assessed in serum and stools. Overall, 285 patients were enrolled: 255 serum, 252 urine and 97 stool samples were analyzed. Anaeroplasma and Escherichia/Shigella ASVs were higher, while Butyricicoccus ASVs were lower in those with normal liver. In MASLD±, Butyricicoccus ASV was significantly higher in those with steatosis. In the Fibrosis±, Butyricicoccus ASV was significantly lower in those with fibrosis. Glycochenodeoxycholic acid-3-sulfate (G-UDCA-3S) appeared to be higher in MASLD with fibrosis. Fecal water from patients with MASLD and fibrosis caused the greatest drop in the TEER vs those with normal liver; this was reversed with protease inhibitors. Finally, fecal IL-13 was lower in MASLD with fibrosis. We identified microbiome signatures which were specific for steatosis and fibrosis and independent of other metabolic risk factors. Moreover, we conclude that protease-related gut permeability plays a role in those MASLD patients with fibrosis, and that disease progression is linked to a gut-liver axis which is at least partially independent of T2DM.

Abstract:Abstract The microencapsulation of the esterified krill oil (EKO), obtained from the transesterification of krill oil (KO) with 3,4-dihydroxyphenylacetic acid (DHPA), via complex coacervation and was investigated. The experimental findings showed that the DHPA and phenolic lipids (PLs) in the EKO affected the stability of the gelatine (GE)-EKO emulsion. To improve its stability, the effects of varying the pH of GE and the use of two emulsification devices, including the homogeniser and ultrasonic liquid processor were investigated, where the ultrasonic liquid processor was found to be a relatively more appropriate emulsification device. In addition, the capsules prepared using a pH of GE of 8.0 showed superior storage and had significantly (p <0.05) lower peroxide value as compared to those prepared with a pH of GE of 6.5. The microencapsulation of the EKO was effective in delaying the development of oxidation products during a period of 25 d of storage, at 25 °C.
Uncovering the Gut–Liver Axis Biomarkers for Predicting Metabolic Burden in Mice

Abstract:Western diet (WD) intake, aging, and inactivation of farnesoid X receptor (FXR) are risk factors for metabolic and chronic inflammation-related health issues ranging from metabolic dysfunction-associated steatotic liver disease (MASLD) to dementia. The progression of MASLD can be escalated when those risks are combined. Inactivation of FXR, the receptor for bile acid (BA), is cancer prone in both humans and mice. The current study used multi-omics including hepatic transcripts, liver, serum, and urine metabolites, hepatic BAs, as well as gut microbiota from mouse models to classify those risks using machine learning. A linear support vector machine with K-fold cross-validation was used for classification and feature selection. We have identified that increased urine sucrose alone achieved 91% accuracy in predicting WD intake. Hepatic lithocholic acid and serum pyruvate had 100% and 95% accuracy, respectively, to classify age. Urine metabolites (decreased creatinine and taurine as well as increased succinate) or increased gut bacteria (Dorea, Dehalobacterium, and Oscillospira) could predict FXR deactivation with greater than 90% accuracy. Human disease relevance is partly revealed using the metabolite–disease interaction network. Transcriptomics data were also compared with the human liver disease datasets. WD-reduced hepatic Cyp39a1 (cytochrome P450 family 39 subfamily a member 1) and increased Gramd1b (GRAM domain containing 1B) were also changed in human liver cancer and metabolic liver disease, respectively. Together, our data contribute to the identification of noninvasive biomarkers within the gut–liver axis to predict metabolic status.
