Abstract:Essential oils of Magnolia biondii flower buds from Sichuan, Anhui and Henan provinces were extracted by hydrodistillation. The differences in aroma, flavour components and antibacterial and antioxidant activity were revealed. Magnolia biondii flower buds are a well‐known medicinal plant in China, and it is important to determine the differences in aroma, flavour components, and biological activity of the essential oils of M. biondii flower buds (EOM) from different origins. In this study, the essential oils of M. biondii flower buds from Sichuan (EOM‐SC), Anhui (EOM‐AH) and Henan (EOM‐HN) provinces were extracted by hydrodistillation. The condition of the extraction process was optimized. The differences in the flavour compounds were further confirmed by GC–MS and relative odour activity value (ROAV) analyses. In addition, the antibacterial and antioxidant activity was also evaluated. The results showed that the optimized condition for steam distillation was water: flower powder ratio = 10:1 with extraction time for more than 3.0 h. Interestingly, special floral, fruity and freshness flavours were found for EOM‐SC, and the medicinal herb and sweetness odours of EOM‐HN were more prominent. The numbers and contents of flavour compounds in EOM‐SC and EOM‐HN were higher than those of EOM‐AH. Eucalyptol, β‐pinene, linalool, α‐terpineol and sabinene could be proposed as the key mutual flavour compounds for three essential oils. In addition, the strong floral, fruity and freshness aroma of EOM‐SC could be associated with the high ROAVs of α‐terpineol, linalool and sabinene. Moreover, EOM‐SC exhibited better antibacterial and antioxidant activity than those of the other two essential oils.

Abstract:ETHNOPHARMACOLOGICAL RELEVANCE: Essential oils (EOs) are important active ingredients in Chinese herbal medicine. Cymbopogon citratus (DC.) Stapf, Cinnamomum cassia (L.) Presl, Salvia japonica Thunb., and Rosa rugosa Thunb. are used in local and traditional medicine to treat inflammation-related diseases. While the constituents and bioactivities of EOs of the above four Chinese herbals have been reported previously, but their anti-inflammatory and antioxidant activities have not been systematically evaluated.AIM OF THE STUDY: The purpose of this study was to research the chemical constituents, anti-inflammatory activities, and antioxidant activities of EOs of four Chinese herbal medicines.MATERIALS AND METHODS: Fresh plant materials were collected both in August 2018. The compositions of EOs were determined by the gas chromatography-mass spectrometry (GC-MS) method. TPA (12-O-tetradecylphophenol-13-acetate)-induced mouse ear model was evaluated the anti-inflammatory activities of EOs. The reduction of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), cyclooxygenase-2 (COX-2) and RelA protein of the nuclear factor kappa-B (NF-κB) in the signal pathway (P65) were measured. The antioxidant activities of EOs from four plants were determined by using 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging method.RESULTS: A total of 217 compounds were separated and identified from four EOs, mainly including trans-cinnamylaldehyde (68.75%), citronellal (38.16%), linalool (1.02-33.73%), geraniol (19.39%) and citronellol (17.18%). All four EOs reduced the damage of the ear tissue and had certain anti-inflammatory effects, and the EOs of C. citratus (CyCEOs) and S. japonica (SJEOs) had the best anti-inflammatory activities among others, even better than ibuprofen. The four EOs exhibited different DPPH radical-scavenging activities (IC50, 0.101-1.017%), where most of them were much lower than that of EOs of C. citratus (CiCEOs) (IC50, 0.101%) and EOs of R. rugosa (RREOs) (IC50, 0.142%).CONCLUSIONS: The main components determine diverse propertise of these four EOs, our results suggested that four EOs presented anti-inflammatory and antioxidant effect via reducing the expression of TNF-a, IL-6, COX-2, and NF-κB p65, which may provide a new approach for development of new anti-inflammatory drugs.

Abstract:Umbelliferae plants, which are widely used as traditional Chinese medicine because of their characteristics of relieving rheumatism, alleviating fever, circulating blood and easing pain. This experimental study was based on ear edema model caused by 12-O-tetracycline-propylphenol-13-acetic acid (TPA) in mice and compared with the Ibuprofen (Ib) group. Gas chromatography-mass spectrometry (GC-MS) was used to analyse the composition of the essential oils from the four studied Umbelliferae plants (Angelica sinensis (Oliv.) Diels, A. dahurica (Hoffm.) Benth. & Hook.f. ex Franch. & Sav., A. pubescens Maxim and Foeniculum vulgare Mill.). Biologically active components in volatile oils from the four studied Umbelliferae plants were evaluated. The expression levels of inflammatory cytokines Tumor Necrosis Factor-α (TNF-α), Cyclooxygenase-2 (COX-2), Interleukin-6 (IL-6) and RelA (p65) in mouse skin were determined by immunohistochemical method. The refractive index of the four essential oils was calculated. A total of 239 compounds were identified by GC-MS from the four studied plants, and the main constituents were osthole (44.61%, APEOs), obepin (0.59%, APEOs & 86.58%, FVEOs), undecanol (8.58%, ADEOs), α-muurolene (7.95%, ADEOs) and cis-anethol (9.11%, ADEOs). E-ligustilide (0.14%, APEOs & 81.14%, ASEOs), (-)-spathulenol (0.08%, FVEOs & 1.21%, ASEOs), (-)-terpinen-4-ol (4.91%, FVEOs), 2-butylthiolane (5.76%, APEOs) and α-bisabolol (3.80%, APEOs). This study showed that all the essential oils from the four studied Umbelliferae plants contained various lactones, including ligustrongolactone, trans-anisol and imperatorin. According to the results of the TPA induction test in the mouse ear edema model, the essential oils of four Umbelliferae plants reduced the levels of inflammatory cytokines TNF-α, COX-2, IL-6 and p65. All of them showed extraordinary biological activity in anti-inflammatory, so they have potential application value for biomedical products, pharmaceutical preparations, natural functional nutrients and cosmetic additives.
Antimicrobial and cytotoxic activities of neolignans from Magnolia officinalis

Abstract:In the light of the steady increase of infections related to vancomycin-resistant enterococci (VRE) and methicillin-resistant Staphylococcus aureus (MRSA), the medicinal plant Magnolia officinalis was subjected to bioassay-directed fractionation, which led to the isolation of the known neolignans piperitylmagnolol (1), magnolol (2), and honokiol (3) from the MeOH extract. In broth-microdilution assays, 1-3 exhibited antibacterial activities against VRE and MRSA at minimum-inhibitory concentrations (MIC) in the range of 6.25-25 microg/ml, compound 1 being the most-potent antibiotic. The ratio of MBC/MIC (MBC = minimum bactericidal concentration) was < or = 2 for all compounds. The kinetics of the antibacterial action of 1 and 3 were studied by means of time-kill assays; both compounds were bactericidal against VRE and MRSA, their actions being time dependent, or both time and concentration dependent. Magnolol (2) was acetylated to magnolol monoacetate (4) and magnolol diacetate (5) (partial or full masking of the phenolic OH functions). The cytotoxic properties of 1-5 against human OVCAR-3 (ovarian adenocarcinoma), HepG2 (hepatocellular carcinoma), and HeLa (cervical epitheloid carcinoma) cell lines were evaluated. The CD50 values for compounds 1-3 were in the range of 3.3-13.3 microg/ml, derivatives 4 and 5 being much less potent. This study indicates that piperitylmagnolol (= 3-[(1S,6S)-6-isopropyl-3-methylcyclohex-2-enyl]-5,5'-di(prop-2-enyl)[1,1'-biphenyl]-2,2'-diol; 1) possesses both significant anti-VRE activity and moderate cytotoxicity against the above cancer cell lines.
Chemical composition and biological activities of essential oils from six lamiaceae folk medicinal plants

Abstract:Essential oils have attracted wide attention in recent years due to their extensive applications in natural functional ingredients, pharmaceutical preparations, biomedical products, and the cosmetics industry. In this study, the chemical compositions and biological activities of essential oils extracted from six Lamiaceae herbs, including Pogostemon cablin (Blanco) Benth. (PCEO), Perilla frutescens (L.) Britton (PFEO), Salvia japonica Thunb. (SJEO), Rosmarinus officinalis L. (ROEO), Lavandula angustifolia Mill. (LAEO), and Agastache rugosa (Fisch. & C. A. Mey.) Kuntze (AREO), were determined and analyzed. A total of 167 components were identified from the six essential oils by GC-MS analysis, with 35, 24, 47, 46, 54, and 37 components in PCEO, PFEO, SJEO, ROEO, LAEO, and AREO, respectively. Hierarchical cluster analysis of chemical compositions showed that the composition of the six essential oils was significantly different in content, and they were clearly divided into six classes. However, all of these six essential oils exhibited promising anti-inflammatory activity by inhibiting the expression of interleukin-1, interleukin-6, tumor necrosis factor-α, and cyclooxygenase-2 in rats with adjuvant arthritis, among which PFEO had the best performance. In addition, the six essential oils displayed significant cytotoxicity on B16 (IC50 = 86.91-228.91 μg/mL) and LNCaP cell lines (IC50 = 116.4-189.63 μg/mL). Meanwhile, all of them presented satisfactory antioxidant activity (IC50 = 4.88-13.89 μg/mL) compared with Trolox C (IC50 = 13.83 μg/mL), and SJEO (IC50 = 7.93 μg/mL) served as an optimal candidate natural antioxidant by DPPH assay. Taken together, these results indicate that the six Lamiaceae essential oils manifest excellent and diverse biological activities, enabling them to be used as perfect natural functional ingredients in antioxidant, antitumor, or anti-arthritic drugs. This study provides more references for pharmaphylogeny research and drug discovery from folk medicinal plants.
Comparative antioxidant, anticancer and antimicrobial activities of essential oils from Semen Platycladi by different extraction methods

Abstract:Semen Platycladi is a highly valued seed tonic from the Cupressaceae Platycladus orientalis (L.) Franco, which contains abundant essential oils exhibiting antioxidant, antimicrobial, sedative and hypnotic potency. The study was formulated to estimate the potential antioxidant, anticancer and antibacterial activities of its essential oils as well as its correlation with chemical constituents. The comparison of three isolation methods including steam distillation extraction, heating reflux extraction, and Soxhlet extraction of ethanol was carried out to isolate essential oils whose chemical composition were identified and evaluated by Gas Chromatography-Mass Spectrometer. It turned out that the yield of essential oils obtained through heating reflux extraction and Soxhlet extraction was almost 40 % higher than those extracted by steam distillation extraction in nearly one-quarter of a time used by the steam distillation extraction. A total of 76 compounds were identified, including ten alkanes, 14 alkenes, 13 alcohols, one aldehyde ketone, three ethers, one acid, 17 esters, five aromatic compounds, and 12 others. The essential oils extracted by steam distillation extraction were proved to have the best antioxidant and antimicrobial activity although with the lowest yield. Scanning electron microscopic observation displayed that the of S. aureus revealed serious disruption of cell membranes resulting in the loss of cell viability. Therefore, it's vital to adopt a proper method to obtain the target ingredients recommended as nutraceuticals and/or phytomedicines.

The Composition, Antioxidant and Antibacterial Activity of Essential Oils from Five Species of the Magnoliaceae Family

Abstract:The leaves of Magnoliaceae family plants contain abundant essential oils (EOs), and these species can be used in many fields due to their high industrial, medicinal, and ornamental values. This study aims to identify the main compounds of the EOs from the leaves of five common Magnoliaceae species (Michelia maudiae, Michelia hedyosperma, Michelia macclurei, Manglietia lucida, Manglietia conifer) using hydrodistillation, GC–MS analysis, and in vitro tests. Additionally, the antioxidant and antibacterial activities of the EOs were also evaluated. The results show that 151 compounds were identified across five species, with sesquiterpenes being dominant. Some key compounds (such as β-caryophyllene, δ-amorphene, β-guaiene, globulol, and β-acorenol) were common among all the species, highlighting their crucial roles in plant physiology and resilience. Other compounds (like valeranone and nerolidol in M. maudiae and β-elemene in M. macclurei) were specific, indicating different functions. Among the five species, the essential oil of M. macclurei contains β-elemene and nerolidyl acetate, and it has the weakest antioxidant activity (IC50 value: 2918.61~21,341.98 μg/mL) but the strongest antibacterial activity (inhibition zone diameter: 8.55 ± 0.93~22.92 ± 0.46 mm; LC50 value: 0.02~0.78 mg/mL). Meanwhile, the EO of M. maudiae demonstrated the best antioxidant activity (the IC50 value was 1283.58~6258.32 μg/mL) and the second-best antibacterial activity (the inhibition zone diameter ranged from 7.61 ± 0.02 to 26.92 ± 0.46 mm, and the LC50 value was 0.03~2.28 mg/mL). Overall, the EO of M. maudiae had the best comprehensive performance. Therefore, the EOs of M. macclurei and M. maudiae showed different performances in biological activity categories, and they could be developed and used in different fields, with the possibility of discovering new applications. This brings inspiration to the potential commercial and industrial uses of sesquiterpenes in Magnoliaceae.

Abstract:Androgen levels in blood and the behaviour of male camels were determined during 1 year. In the months January to April large amounts of androgens were found in blood serum (approximately 30 ng/ml compared with approximately 2 ng/ml throughout the remaining months) as well as in a secretion from glands on the back of the neck (36 ng/ml). During this period when hormone levels were high the behaviour of the camels changed: they became unmanageable, blew-out a palatal flap from the mouth, vocalized, and spread urine over their backs with the aid of their tails. The changes in the sexual cycle of the male camels corresponded with the oestrous period in female camels.

Abstract:To provide scientific support for the exploration and application of the essential oil derived through leaves of Magnolia sieboldii K. Koch, this paper explores extraction technology optimization, composition analysis, and antibacterial properties for the essential oil derived through Magnolia sieboldii K. Koch leaves. Essential oil derived through Magnolia sieboldii K. Koch leaves was extracted by adopting hydrodistillation. With single-factor test and reaction surface improvement methods, the extraction technology of essential oil derived through Magnolia sieboldii K. Koch leaves was optimized. The structure of chemicals has been explored through the use of gas chromatographmass spectrometer (GC-MS) technology. By conducting Kirby-Bauer test and applying spread plate method, the antibacterial properties for essential oil has been measured. The liquid-solid ratio was 7.15 mL/g, the dilution period was 4.00 h, and the NaCl mass percentage was 2.26% as a consequence of the response surface optimization. The rate about gathered essential oil under such circumstances was 0.31537%. By performing chemical composition analysis, 43 compounds were measured from essential oil. Thel oil was sensitive to Staphylococcus aureus, Bacillus subtilis, and Escherichia coli with inhibitory area diameters of 17.27, 10.85, and 9.56 mm, respectively. The minimum inhibitory concentration (MIC) values for the three bacteria strains were 2.5 × 10 – 6 , 5 × 10 – 6 , and 10 × 10 – 6 g/mL, respectively. The optimum extraction rate of essential oil derived through leaves of Magnolia sieboldii K. Koch by hydro distillation is 0.35%, with the cancer-fighting substance β-elemene reaching up to a maximum of 42.53%. Simultaneously, the essential oil derived through leaves of Magnolia sieboldii K. Koch exhibits notable antibacterial effect and large development potential, suggesting the comprehensive development and utilization.

Abstract:Chemical compositions, antioxidant and antimicrobial activities of the essential oils (REOs) of roses including Rosa damascene (RD), Rosa centifolia (RC), Rosa pomponia (RP) and Rosa chinensis Jacq “Crimson Glory” H. T. (CG), and their fractions obtained through molecular distillation (MD) were investigated. The major constitutes were: β-citronellol and nonadecane in RDEO, RCEO and RPEO, 1-nonadecene and nonadecane in CGEO. After MD, a larger proportion of more volatile compounds existed in the distillates relative to their original oils and residues. The REOs and fractions exhibited not only moderate antioxidant activities in the 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and reducing power assays, but also obvious broad-spectrum antimicrobial activities towards the seven tested microorganisms. Notably, the higher antioxidant and antimicrobial activities were observed for the distillates of RDEO, RCEO and RPEO. Moreover, the active antioxidant constitutes in the REOs were characterized as eugenol and methyl eugenol, and the potential antimicrobial components were linalool, phenethyl alcohol, β-citronellol, geraniol, eugenol and methyl eugenol. Results demonstrated that MD could be used to obtain fractions from REOs with higher antioxidant and antimicrobial activities, and the distillates of RDEO, RCEO and RPEO have potential to act as novel natural antioxidant and antimicrobial agents.

Abstract:Four new sesquiterpene lactones (SLs) ( 1 – 4 ), along with a biosynthetically related SL ( 5 ), have been isolated from the leaves of Magnolia grandiflora . Magrandate A ( 1 ) is notable as the first C18 homogemarane type SL, featuring a unique 1,7-dioxaspiro[4.4]nonan-6-one core. Compounds 2 and 3 , representing the first instances of chlorine-substituted gemarane-type SL analogs in natural products, were also identified. The structures of these isolates were elucidated through a combination of spectroscopic data analysis, electronic circular dichroism calculations, and X-ray single-crystal diffraction analysis. All isolates demonstrated anti-inflammatory activity in lipopolysaccharide-stimulated RAW264.7 cells. Notably, 3 – 5 showed a significant inhibitory effect on nitric oxide production, with IC 50 values ranging from 0.79 to 4.73 μmol·L −1 . Additionally, 4 and 5 exhibited moderate cytotoxic activities against three cancer cell lines, with IC 50 values between 3.09 and 11.23 μmol·L −1 .

Abstract:OBJECTIVE: Blumea balsamifera (L.) DC volatile oil (BBO) has a long history in China, which was used to relieve swelling and pain, burn healing and the treatment of fevers. Researches showed that BBO had valuable effects on anti-inflammatory activity, among which (-)-Linalool, (-)-Borneol and Trans-Caryophyllene are the main constituents of BBO and all have certain anti-inflammatory effect. Therefore, according to the content of these three compounds in BBO, we made a formula of (-)-Linalol, (-)-Borneol and trans-Caryophyllene, and studied the anti-inflammatory effect and mechanism of this formula.METHODS: MTS Assay was performed to measure cytotoxicity; ELISA analyzed the standards of PGE2, LTB4 and IL-1β; Griess assay method was used to analyze the production of NO and iNOS; Western blot measured the expression of HO-1, Nrf-2, IκB-α, COX-2and5-LOX; The phosphorylation expression of IKKα, IκB-α and P65 were measured by Western blot. The mRNA levels of IL-1β and TNF-α were detected by qPCR.RESULTS: The results revealed that Formula significantly inhibited cellular inflammation induced by LPS, and leukotriene B4(LTB4) and prolandin E2 (PGE2) were down-regulated by inhibiting the expression of cyclooxygenase-2 (COX-2) and lipoxygenase-5 (5-LOx) in RAW 264.7 cells. Meanwhile, Formula could inhibit the activation of NF-κB, thus reducing the LPS-induced secretion of nitric oxide (NO), inducible NO synthase (iNOS), interleukin(IL-1β) and tumor necrosis factor (TNF-α) and reducing apoptosis. In addition, Formula can effectively increase the expression of nuclear factor erythroid 2-related factor 2 (Nrf2) and antioxidant enzyme heme oxygenase-1 (HO-1), activate the Nrf2/HO-1 pathway to achieve anti-inflammatory effect. The formulation could inhibit the activation of NF-κB, thereby reducing the secretion of nitric oxide (NO), nitric oxide synthase (iNOS), interleukin (IL-1β) and tumor necrosis factor (TNF-α) induced by lips-induced apoptosis. In addition, this formula could effectively increase the expression of nuclear factor erythrocyte 2 related factor 2(Nrf2) and antioxidant enzyme heme oxygenase 1(HO-1), activate Nrf2/HO-1 pathway, and achieve anti-inflammatory effect.CONCLUSION: The results suggest that Formula may have great potential effects for the treatment of acute and chronic inflammation.

Abstract:We argue that the history of political diversity in social psychology may be better characterized by stability than by a large shift toward liberalism. The branch of social psychology that focuses on political issues has defined social problems from a liberal perspective since at least the 1930s. Although a lack of ideological diversity within the discipline can pose many of the problems noted by Duarte et al., we suggest that these problems (a) are less apparent when the insights of social psychology are pitted against the insights from other social science disciplines, and (b) are less pressing than the need for other types of diversity in the field, especially ethnic and racial diversity.
Extraction and Biological Activity of Lignanoids from Magnolia officinalis Rehder & E.H.Wilson Residual Waste Biomass Using Deep Eutectic Solvents

Abstract:Lignanoids are an active ingredient exerting powerful antioxidant and anti-inflammatory effects in the treatment of many diseases. In order to improve the efficiency of the resource utilization of traditional Chinese medicine waste, Magnolia officinalis Rehder & E.H.Wilson residue (MOR) waste biomass was used as raw material in this study, and a series of deep eutectic solvents (ChUre, ChAce, ChPro, ChCit, ChOxa, ChMal, ChLac, ChLev, ChGly and ChEG) were selected to evaluate the extraction efficiency of lignanoids from MORs. The results showed that the best conditions for lignanoid extraction were a liquid-solid ratio of 40.50 mL/g, an HBD-HBA ratio of 2.06, a water percentage of 29.3%, an extract temperature of 337.65 K, and a time of 107 min. Under these conditions, the maximum lignanoid amount was 39.18 mg/g. In addition, the kinetics of the extraction process were investigated by mathematic modeling. In our antioxidant activity study, high antioxidant activity of the lignanoid extract was shown in scavenging four different types of free radicals (DPPH, ·OH, ABTS, and superoxide anions). At a concentration of 3 mg/mL, the total antioxidant capacity of the lignanoid extract was 1.795 U/mL, which was equal to 0.12 mg/mL of Vc solution. Furthermore, the antibacterial activity study found that the lignanoid extract exhibited good antibacterial effects against six tested pathogens. Among them, Staphylococcus aureus exerted the strongest antibacterial activity. Eventually, the correlation of the lignanoid extract with the biological activity and physicochemical properties of DESs is described using a heatmap, along with the evaluation of the in vitro hypoglycemic, in vitro hypolipidemic, immunomodulatory, and anti-inflammatory activity of the lignanoid extract. These findings can provide a theoretical foundation for the extraction of high-value components from waste biomass by deep eutectic solvents, as well as highlighting its specific significance in natural product development and utilization.

Abstract:Flos Chrysanthemi indici, an important medicinal and aromatic plant in China, is considered to have many different preservative and pharmacological properties. Considering the capability of essential oils (EOs), the present study is conducted to compare different extraction methods in order to improve yield and biological activities. Hydro-distillation (HD), steam-distillation (SD), solvent-free microwave extraction (SFME), and supercritical fluid extraction (SFE) are employed to prepare EOs from Flos Chrysanthemi indici. A total of 71 compounds are assigned by gas chromatography/mass spectrometry (GC–MS) in comparison with retention indices. These include 32 (HD), 16 (SD), 31 (SFME) and 38 (SFE) compounds. Major constituents of EOs differ according to the extraction methods were heptenol, tricosane, camphor, borneol, and eucalyptol. EOs extracted by SFME exhibit higher antioxidant activity. All EOs show varying degrees of antimicrobial activity, with minimum inhibitory concentration (MIC) ranging from 0.0625 to 0.125 mg/mL and SFME and SFE prove to be efficient extraction methods. EOs alter the hyphal morphology of Alternaria alternata, with visible bumps forming on the mycelium. Overall, these results indicate that the extraction method can significantly influence the composition and biological activity of EOs and SFME and SFE are outstanding methods to extract EOs with high yield and antimicrobial activity.

Abstract:Aralia chinensis L. is a traditional Miao ethnic medicine known for its pain and inflammation relief and its ability to dispel wind and dampness. This study aimed to assess its antitumour activity and identify the chemical constituents of A. chinensis essential oils. Gas chromatography–mass spectrometry was used to analyse the volatile oil composition, which identified 35, 35, and 24 constituents in essential oils from roots, stems, and leaves, respectively. Network pharmacology predicted the possible key targets of common components in breast-cancer treatment, which revealed AKT1, SRC, EGFR, STAT3, and MAPK3 as high-priority targets with high active-constituent affinity. CCK-8 assay confirmed the inhibitory effect of the essential oils on MCF-7 breast-cancer cells, with oils from Aralia rhizomes, stems, and leaves inhibiting cell viability by 77%, 64%, and 62%, respectively. The active components of Aralia essential oils show promise for breast-cancer treatment by targeting AKT1, SRC, EGFR, and other key factors.

Abstract:The present study aimed to investigate the composition and potential anticancer activities of essential oils obtained from two species, myrrh and frankincense, by hydrodistillation. Using gas chromatography-mass spectrometry (GC-MS), 76 and 99 components were identified in the myrrh and frankincense essential oils, respectively, with the most abundant components, 2-Cyclohexen-1-one, 4-ethynyl-4-hydroxy-3,5,5-trimethyl- and n-Octylacetate, accounting for 12.01 and 34.66%, respectively. The effects of the two essential oils, independently and as a mixture, on five tumor cell lines, MCF-7, HS-1, HepG2, HeLa and A549, were investigated using the MTT assay. The results indicated that the MCF-7 and HS-1 cell lines showed increased sensitivity to the myrrh and frankincense essential oils compared with the remaining cell lines. In addition, the anticancer effects of myrrh were markedly increased compared with those of frankincense, however, no significant synergistic effects were identified. The flow cytometry results indicated that apoptosis may be a major contributor to the biological efficacy of MCF-7 cells.

Abstract:Magnolia grandiflora L. (Magnoliaceae) is a plant of considerable medicinal significance; its flowers and seeds have been used in various traditional remedies. Radioligand binding assays of n-hexane seeds extract showed displacement of radioligand for cannabinoid (CB1 and CB2) and opioid δ (delta), κ (kappa), and µ (mu) receptors. Bioactivity-guided fractionation afforded 4-O-methylhonokiol (1), magnolol (2), and honokiol (3), which showed higher binding to cannabinoid rather than opioid receptors in radioligand binding assays. Compounds 1-3, together with the dihydro analog of 2 (4), displayed selective affinity towards CB2R (Ki values of 0.29, 1.4, 1.94, and 0.99 μM, respectively), compared to CB1R (Ki 3.85, 17.82, 14.55, and 19.08 μM, respectively). An equal mixture of 2 and 3 (1:1 ratio) showed additive displacement activity towards the tested receptors compared to either 2 or 3 alone, which in turn provides an explanation for the strong displacement activity of the n-hexane extract. Due to the unavailability of an NMR or X-ray crystal structure of bound neolignans with the CB1 and CB2 receptors, a docking study was performed to predict ligand-protein interactions at a molecular level and to delineate structure-activity relationships (SAR) of the neolignan analogs with the CB1 and CB2 receptors. The putative binding modes of neolignans 1-3 and previously reported related analogs (4, 4a, 5, 5a, 6, 6a, and 6b) into the active site of the CB1 and CB2 receptors were assessed for the first time via molecular docking and binding free-energy (∆G) calculations. The docking and ∆G results revealed the importance of a hydroxyl moiety in the molecules that forms strong H-bonding with Ser383 and Ser285 within CB1R and CB2R, respectively. The impact of a shift from a hydroxyl to the methoxy group on experimental binding affinity to CB1R versus CB2R was explained through ∆G data and the orientation of the alkyl chain within the CB1R. This comprehensive SAR, influenced by the computational study and the observed in vitro displacement binding affinities, has indicated the potential of magnolia neolignans for developing new CB agonists for potential use as analgesics, anti-inflammatory agents, or anxiolytics.
