Abstract:To improve the classification accuracy of fresh meat species using laser. induced breakdown spectroscopy (LIBS), the support vector machine (SVM) and principal component analysis (PCA) were combined to classify fresh meat species (including pork, beef, and chicken). A simple sample preparation to flatten fresh meat by glass slides was proposed. For each meat sample, 150 spectra were recorded and randomly arranged. The first 75 spectra were used to train a model while the others were used for model validation. By analyzing the 49 normalized spectral lines (K, Ca, Na, Mg, Al, H, O, etc.) in the different tissues, the classification model was built. The results showed that the dimensionality of input variables was decreased from 49 to 10 and modeling time was reduced from 89. 11 s to 55. 52 s using PCA, thus improving the modeling efficiency. The mean classification accuracy of 89. 11% was achieved. The method and reference data are provided for further study of fresh meat classification by laser-induced breakdown spectroscopy technique.

Abstract:Visible/near-infrared (Vis/NIR) spectroscopy was tested to predict the quality attributes of fresh pork (content of intramuscular fat, protein and water, pH and shear force value) on-line. Vis/NIR spectra (350–1100nm) were obtained from 211 samples using a prototype. Partial least-squares regression (PLSR) models were developed by external validation with wavelet de-noising and several pre-processing methods. The 6th order Daubechies wavelet with 6 decomposition levels (db6–6) showed high de-noising ability with good information preservation. The first derivative of db6–6 de-noised spectra combined with multiplicative scatter correction yielded the prediction models with the highest coefficient of determination (R2) for all traits in both calibration and validation periods, which were all above 0.757 except for the prediction of shear force value. The results indicate that Vis/NIR spectroscopy is a promising technique to roughly predict the quality attributes of intact fresh pork on-line.

Abstract:Rapid discrimination of pork in Halal and non-Halal Chinese ham sausages was developed by Fourier transform infrared (FTIR) spectrometry combined with chemometrics. Transmittance spectra ranging from 400 to 4000 cm⁻¹ of 73 Halal and 78 non-Halal Chinese ham sausages were measured. Sample preparation involved finely grinding of samples and formation of KBr disks (under 10 MPa for 5 min). The influence of data preprocessing methods including smoothing, taking derivatives and standard normal variate (SNV) on partial least squares discriminant analysis (PLSDA) and least squares support vector machine (LS-SVM) was investigated. The results indicate removal of spectral background and baseline plays an important role in discrimination. Taking derivatives, SNV can improve classification accuracy and reduce the complexity of PLSDA. Possibly due to the loss of detailed high-frequency spectral information, smoothing degrades the model performance. For the best models, the sensitivity and specificity was 0.913 and 0.929 for PLSDA with SNV spectra, 0.957 and 0.929 for LS-SVM with second derivative spectra, respectively.

Abstract:Fraudulent activities such as mislabeling of beef occurred frequently. In this study, a fast and reliable methodology for specific discrimination of fresh retail beef cuts (ribeye, striploin, brisket, beef shank, and fore shank) was established based on electrosurgical knife - rapid evaporative ionization mass spectrometry (iKnife-REIMS) technique integrated with machine learning (ML). REIMS lipidomic fingerprints of various beef cuts were characterized by ML models to explore meaningful information. Authenticity identification models were built by main ML algorithms including discriminant analysis (DA), support vector machines (SVM), and k-nearest neighbor (KNN) to realize the discrimination of mislabeling in beef cuts. Mislabeling discrimination rates of DA, SVM, and KNN were 95.54%, 99.91%, and 100%, respectively. Model performances were evaluated by confusion matrix, receiver operating characteristic curve, and area under the curve. The validation of the proposed REIMS method for detecting mislabeled beef cuts with an accuracy of 96.80% was performed. Results demonstrated that this artificial intelligent method coupled to ML – guided REIMS analysis was efficient for authenticity detection of mislabeled beef cuts.
Qualitative and Quantitative Detection of Minced Pork Quality by Near Infrared Reflectance Spectroscopy

Abstract:The present study is concerning qualitative and quantitative detection of minced pork quality based on FT-near infrared (FT-NIR) spectroscopy and achieving the rapid approach to detecting the minced pork quality. Firstly, FT-NIR spectroscopy combined with partial least squares (PLS) and least squares-support vector machine (LS-SVM) was used for minced pork quality prediction including discrimination of the different muscle type of pig and quantitative detection of the fat, protein and moisture content of pork. The result indicated that 100% recognition ratio for calibration and 96% recognition ratio for validation were achieved by PLSDA for 4 different muscle types of pig. These two methods for chemical composition detection both have good performances in predicting fat and moisture content, the correlation coefficient for calibration and validation was all more than 0.9, but the models for protein content prediction were of less well performances, the correlation coefficients for calibration and validation, RMSEC, RMSEP and RMSECV respectively were 0.722, 0.593, 1.595, 1.550 and 1.888, respectively. The LS-SVM method is more accurate in predicting each quality index than the PLSR method. The result shows that the prediction models for fat and moisture content based on LS-SVM have a better performance with high precision, good stability and adaptability and can be used to predict the fat and moisture content of minced pork rapidly, and provide a fast approach to discrimination of the different muscle type of pig.

Abstract:The objective of the present study was to estimate minced pork meat quality using visible and near infrared (Vis-NIR) spectroscopy. Two hundred twenty five carcasses samples from longissimus dorsi muscle were scanned over the Vis-NIR spectral range from 350 to 1 015 nm and analysed for intramuscular fat (IMF), protein and moisture according to the official methods. Wavelet transform was employed to eliminate the spectra noise. Partial least square regression (PLSR) and support vector machine (SVM) were used to develop Vis-NIR spectroscopy models for chemical composition detection. According to calibration statistics, the best model to predict intramuscular fat content was developed by SVM with the denoised spectra, the correlation coefficient was 0.889 for calibration and 0.888 for validation. For protein and moisture, the best model was achieved with the PLS method with the correlation coefficient of 0.869 and 0.881 for protein calibration and validation sets and 0.877 and 0.848 for moisture calibration and validation sets, respectively. And all the ratios of standard deviation of validation set to root mean square error of prediction (RPD) were not more than 3.0. Results indicated that it was possible to predict chemical composition in minced pork meat. As a fast predictor of meat quality using Vis-NIR spectroscopy, it is necessary to improve the precision and the robustness of the model for practice.

Abstract:Apert syndrome is a clinically distinct condition characterized by craniosynostosis and severe syndactyly of the hands and the feet. Apert syndrome results from either of two specific nucleotide substitutions, both C-to-G transversions, in the fibroblast growth factor receptor 2 (FGFR2) gene. To determine if Chinese Apert syndrome patients carry the same mutations, fifteen unrelated Apert syndrome patients and a fetus from a mother with Apert syndrome were studied by the use of restriction analysis and direct sequencing. The results demonstrated that 13 had the Ser252Trp mutation and 2 had the Pro253Arg mutation. Prenatal diagnosis of the fetus was successfully made.

Abstract:This study utilizes laser‐induced breakdown spectroscopy (LIBS) and machine learning methods to achieve remote bacterial identification. The Random Forest (RF) model demonstrated excellent performance in classification accuracy, recall, precision, and F1‐score, providing more accurate identification results and showing broad application prospects. Bacteria are the primary cause of infectious diseases, making rapid and accurate identification crucial for timely pathogen diagnosis and disease control. However, traditional identification techniques such as polymerase chain reaction and loop‐mediated isothermal amplification are complex, time‐consuming, and pose infection risks. This study explores remote (~3 m) bacterial identification using laser‐induced breakdown spectroscopy (LIBS) with a Cassegrain reflective telescope. Principal component analysis (PCA) was employed to reduce the dimensionality of the LIBS spectral data, and the accuracy of support vector machine (SVM) and Random Forest (RF) algorithms was compared. Multiple repeated experiments showed that the RF model achieved a classification accuracy, recall, precision, and F1‐score of 99.81%, 99.80%, 99.79%, and 0.9979, respectively, outperforming the SVM model and providing more accurate remote bacterial identification. The method based on laser‐induced plasma spectroscopy and machine learning has broad application prospects, supporting noncontact disease diagnosis, improving public health, and advancing medical research and technological development.

Abstract:The rapid classification of the varieties of walnut powder is important to protect the consumers against potential fraud, since different varieties of walnut powder vary dramatically in quality, taste and sale price. Laser-induced breakdown spectroscopy (LIBS) is a promising technology for on-line application, which is no or little sample preparation, fast analysis speed (usually seconds to minutes), remote detection available. In this study, laser-induced breakdown spectroscopy and multi-variable analysis methods were applied to classify varieties of walnut powder for the first time. Before LIBS analysis, a fairly flat surface was provided to reduce fluctuation, and no other sample preparation was needed. After acquiring the LIBS spectra, Principal Component Analysis (PCA) was used to make qualitative discrimination, and classification models were developed with Partial Least Squares Discrimination Analysis (PLS-DA) and Support Vector Machines (SVM). Three origins and different varieties from same origin were discriminated by LIBS. It was shown that origins as well as different varieties from the same origin could be differentiated successfully by laser-induced breakdown spectroscopy and chemometrics. Support vector machines can help to handle complex LIBS dataset, which improved the classified rate of model. The presented results provide the first proof-of-principle data for the on-line application of LIBS technology for classification of the varieties of walnut powder.

Abstract:Agricultural products have complex matrix prohibiting the development of LIBS (laser induced breakdown spectroscopy) in detecting its composition, especially tracing toxic heavy metals. The work was aiming at improving the sensitivity and accuracy of LIBS in predicting heavy metals in farm products. Fresh pork was chosen and dipped in Pb solution for making polluted samples. The samples were dried, ground to powder and pressed to pellets to remove the effect of water. The LIBS spectra of fresh and dried pork were collected at the same experimental conditions. The results demonstrated that the intensity of Pb was similar and obscure in fresh samples, but quite strong and clear in dried ones. The linear calibration curve between real content of Pb and intensity of Pb I 405.78 showed that the correlation coefficient was poor in fresh pork and good in dried samples and the limit of detection of dried samples is 5.13 mg.kg(-1). This experiment testified that the sensitivity and accuracy could be improved after the fresh samples were pretreated by simple physical methods, and it is potential in predicting heavy metals in farm products by LIBS. And this method can also be applied to other meat of detection by LIBS.

Abstract:Laser-induced breakdown spectroscopy (LIBS) is a spectroscopic analytic technique with great application potential because of its unique advantages for online/in-situ detection. However, due to the spatially inhomogeneity and drastically temporal varying nature of its emission source, the laser-induced plasma, it is difficult to find or hard to generate an appropriate spatiotemporal window for high repeatable signal collection with lower matrix effects. The quantification results of traditional physical principle based calibration model are unsatisfactory since these models were not able to compensate for complicate matrix effects as well as signal fluctuation. Machine learning is an emerging approach, which can intelligently correlated the complex LIBS spectral data with its qualitative or/and quantitative composition by establishing multivariate regression models with greater potential to reduce the impacts of signal fluctuation and matrix effects, therefore achieving relatively better qualitative and quantitative performance. In this review, the progress of machine learning application in LIBS is summarized from two main aspects: i) Pre-processing data for machine learning model, including spectral selection, variable reconstruction, and denoising to improve qualitative/quantitative performance; ii) Machine learning methods for better quantification performance with reduction of the impact of matrix effect as well as LIBS spectra fluctuations. The review also points out the issues that researchers need to address in their future research on improving the performance of LIBS analysis using machine learning algorithms, such as restrictions on training data, the disconnect between physical principles and algorithms, the low generalization ability and massive data processing ability of the model.

Abstract:Timely and accurate identification of harmful bacterial species in the environment is paramount for preventing the spread of diseases and ensuring food safety. In this study, laser-induced breakdown spectroscopy technology was utilized, combined with four machine learning methods - KNN, PCA-KNN, RF, and SVM, to conduct classification and identification research on 7 different types of bacteria, adhering to various substrate materials. The experimental results showed that despite the nearly identical elemental composition of these bacteria, differences in the intensity of elemental spectral lines provide crucial information for identification of bacteria. Under conditions of high-purity aluminum substrate, the identification rates of the four modeling methods reached 74.91%, 84.05%, 85.36%, and 96.07%, respectively. In contrast, under graphite substrate conditions, the corresponding identification rates reached 96.87%, 98.11%, 98.93%, and 100%. Graphite is found to be more suitable as a substrate material for bacterial classification, attributed to the fact that more characteristic spectral lines are excited in bacteria under graphite substrate conditions. Additionally, the emission spectral lines of graphite itself are relatively scarce, resulting in less interference with other elemental spectral lines of bacteria. Meanwhile, SVM exhibited the highest precision rate and recall rate, reaching up to 1, making it the most effective classification method in this experiment. This study provides a valuable approach for the rapid and accurate identification of bacterial species based on LIBS, as well as substrate selection, enhancing efficient microbial identification capabilities in fields related to social security and military applications.

Green analysis of Pb in pork by laser induced breakdown spectroscopy

Abstract:It is likely that pork is polluted by heavy metals during feeding pig and logistics of meat.On this circumstance,it is necessary to detect toxic metals in pork especially by green and sustainable method.In this work,the emerging technology,LIBS (laser induced breakdown spectroscopy),was applied to determine Pb in pork.In order to prove the feasibility of LIBS in doing this,pork was polluted by Pb solution in lab.At the same time,the samples were dried,ground and pressed to pellets to eliminate the effect of water and improve the homogeneity of samples.The spectra of pressed pellets were collected at optimized LIBS conditions and the real concentration of Pb was obtained by conventional AAS (atomic absorption spectroscopy).The results demonstrated that LIBS can detect the characteristic line Pb Ⅰ 405.78 when the content of samples was higher than 4.56 μg/g.The relationship between LIBS intensity of line PbI405.78 and content of Pb was gradient,but it was not simple linear.In addition,the predicted accuracy was higher in samples those had lower content than 10 μg/g when quadratic polynomial only with one variable was utilized.Whereas,the linear sensitivity was higher in samples with lower content especially below 10μg/g.This result indicated that it is possible there is some critical content which can decide different quantitative model.Accordingly,the accuracy of predicted content can be improved.This work displayed that LIBS is a feasible method in analyzing heavy metal Pb in pork and it is potential in further work.

Abstract:Laser-induced breakdown spectroscopy (LIBS) is an atomic-emission spectroscopy technique that employs a focused laser beam to produce microplasma. Although LIBS was designed for applications in the field of materials science, it has lately been proposed as a method for the compositional analysis of agricultural goods. We deployed commercial handheld LIBS equipment to illustrate the performance of this promising optical technology in the context of food authentication, as the growing incidence of food fraud necessitates the development of novel portable methods for detection. We focused on regional agricultural commodities such as European Alpine-style cheeses, coffee, spices, balsamic vinegar, and vanilla extracts. Liquid examples, including seven balsamic vinegar products and six representatives of vanilla extract, were measured on a nitrocellulose membrane. No sample preparation was required for solid foods, which consisted of seven brands of coffee beans, sixteen varieties of Alpine-style cheeses, and eight different spices. The pre-processed and standardized LIBS spectra were used to train and test the elastic net-regularized multinomial classifier. The performance of the portable and benchtop LIBS systems was compared and described. The results indicate that field-deployable, portable LIBS devices provide a robust, accurate, and simple-to-use platform for agricultural product verification that requires minimal sample preparation, if any.

[Invasive aspergillosis in hematooncological patients: advantages and disadvantages of various diagnostic methods, treatment options and financial costs of therapy].

Abstract:BACKGROUND Invasive aspergillosis (IA) is a leading invasive fungal infection in hematooncological patients. The aim of this study was to analyse the incidence, diagnostic procedures and treatment of IA in hematooncological department in large hospital in the Czech Republic. PATIENTS AND METHODS A retrospective analysis of medical and laboratory records from patients hospitalised in our department with proven/probable IA between January 2000 and December 2006 was performed. RESULTS 52 cases of IA in 51 patients were identified (17.3% proven IA/82.7% probable IA). Number of IA cases notably increased during study period (1 case of IA in 2000 vs 21 cases of IA in 2006) and majority of them was of nosocomial origin (61.5%). Pulmonary aspergillosis was diagnosed in 46 cases (88.5%). Patients treated for acute leukemia or undergoing allogeneic stem cell transplantation represent the group at the highest risk of IA (in total 52% of cases). Fever and signs of pulmonary involvement were the most common clinical signs of infection (presented in 92.3% and 69.2 cases respectively). Conventional diagnostic methods including autopsy were able to diagnose only 15 cases of IA (28.8%). In all other cases (71.2%) the diagnosis was done by detection of galactomannan (GM) in serum. Introduction of GM monitoring enabled erlier initiation of antifungal treatment by 4 days. Initial therapy of IA led to the treatment response (partial and complete) in 18 (34.6%) of infections--the highest percentage of response has been seen in voriconazole monotherapy group (42%) and when combination of voriconazole and caspofungin has been used (83%). Salvage therapy was initiated due to the failure of initial treatment in 21 (40.3%) of cases. Patients were treated mostly with combination ofvoriconazole and caspofungin and/or monotherapy with voriconazole has been used with treatment response 55% and 50% respectively. Introduction of new antifungal drugs together with increased number of patients with IA led to the marked increase of total costs spent on treatment of IA per year--from 11,5 thousands CZK in 2000 to 6,2 millions CZK in 2006. CONCLUSIONS IA is the most frequent cause of infection-related mortality in patients with haematological malignancies. Routine use of non-culture base methods in diagnosis of IA together with treatment using new, effective antifungals can improve prognosis of patients with this life threatening infection.

The feasibility of rapidly identifying kudzu powder origin was investigated based on laser-induced breakdown spectroscopy (LIBS) technology combined with chemometrics methods. The discriminant models based on the full spectrum include extreme learning machine (ELM), soft independent modeling of class analogy (SIMCA), K-nearest neighbor (KNN) and random forest (RF), and the accuracy of models was more than 99.00%. The prediction results of KNN and RF models were best: the accuracy of calibration and prediction sets of kudzu powder from different producing areas both reached 100%. The characteristic wavelengths were selected using principal component analysis (PCA) loadings. The accuracy of calibration set and the prediction set of discrimination models, based on characteristic wavelengths, is all higher than 98.00%. Random forest and KNN have the same excellent identification results, and the accuracy of calibration and prediction sets of kudzu powder from different producing areas reached 100%. Compared with the full spectrum discriminant analysis model, the discriminant analysis model based on the characteristic wavelength had almost the same discriminant effects, and the input variables were reduced by 99.92%. The results of this research show that the characteristic wavelength can be used instead of the LIBS full spectrum to quickly identify kudzu powder from different producing areas, which had the advantages of reducing input, simplifying the model, increasing the speed and improving the model effect. Therefore, LIBS technology is an effective method for rapid identification of kudzu powder from different habitats. This study provides a basis for LIBS to be applied in the genuineness and authenticity identification of Chinese medicine.
Rapid Discrimination of the Categories of the Biomass Pellets Using Laser-Induced Breakdown Spectroscopy

Abstract:As a renewable energy alternative to fossil fuels, biomass pellets have attracted much attention due to its promising advantages of convenient use and easy combustion. Rapid discrimination of different biomass pellets and selecting the one with better combustion performance are of great significance to improve the energy utilization. In this study, laser-induced breakdown spectroscopy (LIBS) coupled with chemometrics methods were used to discriminate biomass pellets. Lignocellulose components were firstly determined and further analyzed according to LIBS spectra. Principal component analysis (PCA), partial least squares discrimination analysis (PLS-DA), support vector machines (SVM), radial basis function neural network (RBFNN) and extreme learning machine (ELM) were applied to quantitatively distinguish biomass pellets. The RBFNN model showed a reliable discrimination power, with 100% and 96.88% average recognition accuracy in calibration and prediction sets respectively. Visualization analysis based on the RBFNN model was applied to intuitively discriminate the biomass pellets with different colors. In addition, rice husk with relatively poor combustion performance could be accurately distinguished from wood biomass pellets by all discrimination models. The results indicated that LIBS combined with chemometrics methods could be a novel and reliable approach to discriminate biomass pellets and select the category with better combustion performance.

Abstract:Abstract In recent years, a laser-induced breakdown spectrometer (LIBS) combined with machine learning has been widely developed for steel classification. However, the much redundant information of LIBS spectra increases the computation complexity for classification. In this work, restricted Boltzmann machines (RBM) and principal component analysis (PCA) were used for dimension reduction of datasets, respectively. Then, a support vector machine (SVM) was adopted to process feature information. Two models (RBM-SVM and PCA-SVM) are compared in terms of performance. After optimization, the accuracy of the RBM-SVM model can achieve 100%, and the maximum dimension reduction time is 33.18 s, which is nearly half of that of the PCA model (53.19 s). These results preliminarily indicate that LIBS combined with RBM-SVM has great potential in the real-time classification of steel.

Abstract:Delayed graft function (DGF) is a common problem in kidney transplantation and is associated with adverse graft outcomes and increased cost of care. The purpose of this study was to determine if the duration of dialysis increases the risk of DGF. All primary deceased donor renal transplants between January 2000 and December 2003 were identified in the Organ Procurement and Transplant Network database. Two separate definitions of DGF were used: dialysis in the first week post‐transplant (DPT) and creatinine drop of < 25% in the first 24 h or slow graft function (SGF). The rate of DPT and SGF increased from 5.7% and 34.4%, respectively, for pre‐emptively transplanted patients, to 32% and 49.9% for patients who had been on dialysis for 6 or more years. When compared to pre‐emptive transplantation, increasing duration of dialysis increased the adjusted risk of both DPT and SGF (OR 6.64 (95% CI 5.49–8.03) and OR 1.76 (95% CI 1.56–2.00) for patients on dialysis for 6 or more years, for DPT and SGF, respectively. A strong association between duration of dialysis and DGF exists, and investigations into the mechanisms by which dialysis influences DGF may lead to useful interventions to limit this complication.
