Abstract:Anaerobic ammonium oxidation (anammox) is a biological process by which ammonium is oxidized to dinitrogen gas by using nitrite as the electrons acceptor. Anaerobic ammonium-oxidizing bacteria play an important role in nitrogen removal from wastewater and global N-cycle. The study of metabolism of anammox bacteria will help us understand the anammox mechanism and develop anammox biotechnology. Anammox bacteria are chemoautotrophic bacteria that use CO, or HCO3- as carbon source and obtain their energy from the conversion of ammonium and nitrite into dinitrogen gas. Hydrazine has been detected as an intermediate in the anammox pathway, while hydroxylamine and nitric oxide have not been detected yet. The genomic data indicate that anammox bacteria fix carbon dioxide through acetyl-CoA pathway. The proposed anammox pathway is consistent with the available experimental data, thermodynamical calculation and biochemical determination and as well as the Ockham's razor principal.

Abstract:Anaerobic Ammonium Oxidation ("anammox") is a bacterial process in which nitrite and ammonium are converted into nitrogen gas and water, yielding energy for the cell. Anammox is an important branch of the global biological nitrogen cycle, being responsible for up to 50% of the yearly nitrogen removal from the oceans. Strikingly, the anammox process uniquely relies on the extremely reactive and toxic compound hydrazine as a free intermediate. Given its global importance and biochemical novelty, there is considerable interest in the enzymes at the heart of the anammox pathway. Unfortunately, obtaining these enzymes in sufficiently large amounts for biochemical and structural studies is problematic, given the slow growth of pure cultures of anammox bacteria when high cell densities are required. However, the anammox process is being applied in wastewater treatment to remove nitrogenous waste in processes like DEamMONification (DEMON). In plants using such processes, which rely on a combination of aerobic ammonia‐oxidizers and anammox organisms, kilogram amounts of anammox bacteria‐containing sludge are readily available. Here, we report a protein isolation protocol starting from anammox cells present in DEMON sludge from a wastewater treatment plan that readily yields pure preparations of key anammox proteins in the tens of milligrams, including hydrazine synthase HZS and hydrazine dehydrogenase (HDH), as well as hydroxylamine oxidoreductase (HAO). HDH and HAO were active and of sufficient quality for biochemical studies and for HAO, the crystal structure could be determined. The method presented here provides a viable way to obtain materials for the study of proteins not only from the central anammox metabolism but also for the study of other exciting aspects of anammox bacteria, such as for example, their unusual ladderane lipids.

Metagenomic and metatranscriptomic insights into the complex nitrogen metabolic pathways in a single-stage bioreactor coupling partial denitrification with anammox

Abstract:Combined partial-denitrification and anammox process has received growing interest for treating wastewaters with high nitrate concentrations, but the underlying molecular mechanisms are currently poorly understood due to the complexity in bacterial community composition and functionality of the coupled system. This study applied metagenomic and metatranscriptomic approaches to gain comprehensive and deep insights into the nitrogen metabolic pathways in a lab-scale partial-denitrification/anammox bioreactor showing stable nitrogen removal. Stoichiometric analysis showed that most of nitrogen was removed by the coupled system (similar to 92.1%), but dissimilatory denitrification also contributed to nitrogen gas production. The latter was further evidenced by nosZ gene transcripts in the metatranscriptome. Metagenomic and metatranscriptomic analyses identified Candidatus Jettenia caeni as the most abundant and active anammox bacteria with relative abundance and mRNA expression of over 5% and 27%, respectively, while Thauera appeared as the major denitrifying bacteria in the system (relative abundance and mRNA expression exceeded 8% and 10%, respectively). mRNA levels of hdh/hzo and hzsA/B/C genes in Candidatus Jettenia caeni and Candidatus Brocadia sinica were much higher than those of other functional genes, confirming the major roles of the anammox bacteria in nitrogen metabolism. In addition, nrfA/B/C/D genes were mainly expressed in anammox bacteria, implying their involvement in that dissimilatory nitrite reduction to ammonia (especially for species Candidatus Brocadia sinica). Nitrate reduction process in Thauera was mainly achieved by the activation of the napA/B/C genes instead of narG/H genes. This study extablished the nitrogen metabolic network based on the comprehensive analyses of the abundance and mRNA expression of functional genes, which might improve our understanding of the nitrogen metabolic mechanisms of the combined partial-denitrification and anammox process.
Efficient nitrogen removal from municipal wastewater by an autotrophic-heterotrophic coupled anammox system: The up-regulation of key functional genes

Abstract:The metabolic pathways based on key functional genes were innovatively revealed in the autotrophic-heterotrophic coupled anammox system for real municipal wastewater treatment. The nitrogen removal performance of the system was stabilized at 88.40 ± 3.39 % during the treatment of real municipal wastewater. The relative abundances of the nitrification functional genes ammonia oxidase ( amoA/B/C ), hydroxylamine oxidoreductase ( hao ), and nitrite oxidoreductases ( nxrA/B ) were increased by 1.2–2.4 times, and these three nitrification functional genes were mostly contributed by Nitrospira that dominated the efficient nitrification of the system. The relative abundance of anammox bacteria Candidatus Brocadia augmented from 0.35 % to 0.75 %, accompanied with the increased expression of hydrazine synthase ( hzs ) and hydrazine dehydrogenase ( hdh ), resulting in the major role of anammox (81.24 %) for nitrogen removal. The expression enhancement of the functional genes nitrite reductase ( narG/H , napA/B ) that promoted partial denitrification (PD) of the system weakened the adverse effects of the sharp decline in the population of PD microbe Thauera (from 5.7 % to 2.2 %). The metabolic module analysis indicated that the carbon metabolism pathways of the system mainly included CO 2 fixation and organic carbon metabolism, and the stable enrichment of autotrophic bacteria ensured stable CO 2 fixation. Furthermore, the enhanced expression of the glucokinases ( glk , GCK , HK , ppgk ) and the abundant pyruvate kinase ( PK ) achieved stable hydrolysis ability of organic carbon metabolism function of the system. This study offers research basics to practical application of the mainstream anammox process.

Abstract:Nitrate production during anammox can decrease total nitrogen removal efficiency, which will negatively impact its usefulness for the removal of nitrogen from waste streams. However, neither the performance characteristics nor physiological shifts associated with nitrate accumulation in anammox reactors under different nitrogen loading rates (NLRs) is well understood. Consequently, these parameters were studied in a lower NLR anammox reactor, termed R1, producing higher than expected levels of nitrate and compared with a higher NLR reactor, termed R2, showing no excess nitrate production. While both reactors showed high NH4+-N removal efficiencies (>90%), the total nitrogen removal efficiency (<60%) was much lower in R1 due to higher nitrate production. Metagenomic analysis found that the number of reads derived from anammox bacteria were significantly higher in R2. Another notable trend in reads occurrence was the relatively higher levels of reads from genes predicted to be nitrite oxidoreductases (nxr) in R1. Binning yielded 27 high quality draft genomes from the two reactors. Analysis of bin occurrence found that R1 showing both a decrease in anammox bacteria and an unexpected increase in nxr. In-situ assays confirmed that R1 had higher rates of nitrite oxidation to nitrate and suggested that it was not solely due to obligate NOB, but other nxr-containing bacteria are important contributors as well. Our results demonstrate that nitrate accumulation can be a serious operational concern for the application of anammox technology to low-strength wastewater treatment and provide insight into the community changes leading to this outcome.

Abstract:To achieve high-rate nitrogen removal in municipal wastewater treatment through anaerobic ammonia oxidation (Anammox), the nitrification, partial denitrification, and Anammox processes were integrated by a step-feed strategy. An exceptional nitrogen removal load of 0.224 kg N/(m 3 ·d) was achieved by gradient-reducing the hydraulic retention time (HRT) to 5 h. Metagenomic analysis demonstrated that Nitrosospira could express all genes encoding ammonia oxidation under low nitrogen and dissolved oxygen conditions (less than 0.5 mg/L), enabling complete nitrification. With the short of HRT, the relative abundance of Thauera increased from 2.8 % to 6.4 %. Frequent substrate exchanges at such extremely short HRT facilitated enhanced synergistic interactions among Nitrosospira , Thauera , and Candidatus Brocadia . These findings provide a comprehensive understanding of the utilization of Anammox combined processes for high-speed nitrogen removal in municipal wastewater treatment and the microbial interactions involved.

Abstract:To overcome the significant challenges associated with nitrite supply and nitrate residues in mainstream anaerobic ammonium oxidation (anammox)-based processes, this study developed a combined solid-phase denitrification (SPD) and anammox process for low-strength nitrogen removal without the addition of nitrite. The SPD step was performed in a packed-bed reactor containing poly-3-hydroxybutyrate-co-3-hyroxyvelate (PHBV) prior to employing the anammox granular sludge reactor in the continuous-flow mode. The removal efficiency of total inorganic nitrogen reached 95.7 ± 1.2% under a nitrogen loading rate of 0.18 ± 0.01 kg N·m3·d-1, and it required 1.02 mol of nitrate to remove 1 mol of ammonium nitrogen. The PHBV particles not only served as biofilm carriers for the symbiosis of hydrolytic bacteria (HB) and denitrifying bacteria (DB), but also carbon sources that facilitated the coupling of partial denitrification and anammox in the granules. Metagenomic sequencing analysis indicated that Burkholderiales was the most abundant HB genus in SPD. The metabolic correlations between DB (Betaproteobacteria, Rhodocyclaceae, and Anaerolineae) and anammox bacteria (Candidatus Brocadiac and Kuenenia) in the granules were confirmed through microbial co-occurrence networks analysis and functional gene annotations. Additionally, the genes encoding nitrate reductase (Nap) and nitrite reductase (Nir) in DB primarily facilitated nitrate reduction, thereby supplying nitric oxide to anammox bacteria for subsequent nitrogen removal with hydrazine synthase (Hzs) and hydrazine dehydrogenase (Hdh). The findings provide insights into microbial metabolism within combined SPD and anammox processes, thus advancing the development of mainstream anammox-based processes in engineering applications.

Abstract:The present study reports kinetic characteristics of Anammox (anaerobic ammonium oxidation) EGSB (Expanded Granular Sludge Bed) reactor after feeding with strong ammonium-containing synthetic wastewater. The microbial communities were analysed based on their 16S rRNA gene sequences. The results showed that the volumetric nitrogen loading rate (NLR) and volumetric nitrogen removal rate (NRR) reached up to 22.87 kg N/(m(3)d) and 18.65 kg N/(m(3)d), respectively, when the influent nitrogen concentrations were 1429.1mg N/L. Monod and Haldane models both proved to be suitable in characterizing the kinetic behavior of the reactor. Based on Haldane model, the relationships among the ammonium, nitrite, nitrogen conversion rates and substrate concentrations were established with corresponding correlation coefficients of 0.992, 0.993 and 0.993, respectively. The maximum ammonium, nitrite and nitrogen conversion rates (q(max)) by the granular sludge were 381.2, 304.7 and 731.7 mg N/(gVSSd), half saturation constants (K(s)) were 36.75, 0.657 and 29.26 mg N/L and inhibition constants (K(i)) were 887.1, 13,942.1 and 1779.6 mg N/L, respectively. Anammox-EGSB reactor was found tolerant to substrate and capable of treating strong ammonium-containing wastewater. The dominant microbial population of the granular sludge in the reactor was Candidatus Kuenenia stuttgartiensis.
Genomic synergistic efficient carbon fixation and nitrogen removal induced by excessive inorganic carbon in the anammox-centered coupling system

Abstract:Given the significance of HCO3- for autotrophic anammox bacteria (AnAOB), excessive HCO3- was always provided in anammox-related systems and engineering applications. However, its impact mechanism on anammox process at genome-level remains unknown. This study firstly established an anammox-centered coupling system that entails heterotrophic partial denitrification (PD) and hydrolytic acidification (A-PDHA) fed mainly with inorganic carbon (high HCO3- concentration and low C/N ratio). Metagenomic binning and metatranscriptomics analyses indicated that high HCO3- concentration enhanced expression of natural most efficient phosphoenolpyruvate (PEP) carboxylase within AnAOB, by up to 30.59 folds. This further induced AnAOB to achieve high-speed carbon-fixing reaction through cross-feeding of phosphate and PEP precursors with heterotrophs. Additionally, the enhanced activity of transporters and catalytic enzymes (up to 4949-fold) induced by low C/N ratio enabled heterotrophs to eliminate extracellular accumulated energy precursors mainly derived from carbon fixation products of AnAOB. This maintained high-speed carbon-fixing reaction within AnAOB and supplemented heterotrophs with organics. Moreover, assimilated energy precursors stimulated nitrogen metabolism enzymes, especially NO2- reductase (968.14 times), in heterotrophs. This established an energy-saving PD-A process mediated by interspecies NO shuttle. These variation resulted in efficient nitrogen removal (>95 %) and reduced external organic carbon demand (67 %) in A-PDHA system. This study unveils the great potential of an anammox-centered autotrophic-heterotrophic coupling system for achieving cost-effective nitrogen removal and enhancing carbon fixation under excessive HCO3- doses.

Abstract:Purpose: To compare the accuracy of a commercially available MLPA kit with a laboratory developed RT-PCR assay for the detection of SMN1 and SMN2 copy numbers in clinical samples. Methods: We developed and validated a laboratory developed real time PCR based test capable of detecting SMN1 and SMN2 copy numbers in individuals. We also validated an MLPA kit purchased from MRC Holland for the same purpose. We then analyzed a series of 1027 anonymized samples using both technologies. When discrepant results were obtained, each sample was re-analyzed at least twice using both platforms. Results: Five samples did not yield results in either assay. For SMN1 copy number analysis, 2 RT-PCR assays revealed indeterminant results and all 1020 other samples were concordant for SMN1 copy number. There were 9 discrepancies in SMN2 copy number determination mostly due to a variability in MLPA analysis. Conclusion: Both MLPA and RTPCR assays give a reliable estimate of SMN1 copy number and are therefore appropriate for population based carrier screening for Spinal Muscular Atrophy Type 1. Abstract:Anaerobic ammonium oxidation (anammox) is a key process to achieve autotrophic nitrogen removal from wastewater. Four continuous flow biological reactors were operated by feeding different concentrations of ammonia and with or without the addition of acetate. The best nitrogen removal was achieved at the condition of high ammonia concentration without organic carbon, with obtained the nitrogen removal loading rate of 2.2 g N/(L.d) and the nitrogen removal percentage of 92%. Candidatus Kuenenia was the dominant genus and was stimulated at a high ammonia concentration, while a high acetate concentration enhanced the denitrifying activity. Ca. Kuenenia, Nitrospira, Thauera and other microorganisms constituted the microbial network responsible for both nitrogen and carbon metabolisms. Ca. Kuenenia lacked synthetases of alanine, aspartate and methionine, which could be supplied by nitrifiers and denitrifiers. This cross-feeding strategy was conducive to efficient biosynthesis and degradation of amino acids. N-decanoyl homoserine lactone was detected in both biomass and extracellular polymeric substances phases. Acyl-homoserine lactones (AHLs) based quorum sensing and quorum quenching microorganisms coexisted in anammox systems, playing important roles in biosynthesis, sensing and quenching of AHLs.
Metatranscriptomics provides an in-depth perspective on the resistance and detoxification of anammox bacteria to dissolved oxygen in a pilot CANON process

Abstract:In the completely autotrophic nitrogen removal over nitrite (CANON) process, the conflicting oxygen requirements of anammox and ammonium-oxidizing bacteria often lead to retardation in anammox activity. However, our study achieved stable nitrogen removal with a maximum capacity of 1096 g-N/m3/d in a 20 m3 CANON reactor under long-term intensive aeration. The anammox bacteria unusually distributed in the outer layer of the biofilm and demonstrated remarkable oxygen tolerance. Their activity only declined by 18.5 % under 2.0 mg/L of dissolved oxygen. When anammox bacteria encountered oxygen exposure, they adopted some strategies. Metatranscriptomics revealed that Candidatus Kuenenia, the dominant anammox species in our system, downregulated its gene expressions involved in carbon metabolism and oxidative phosphorylation. This may reduce electron leakage that combines with O2, thereby minimizing the generation of reactive oxygen species (ROS). By contrast, the secretion of extracellular proteins and conversion of O2·- were upregulated to eliminate ROS promptly. This behavior endowed Ca. Kuenenia with a unique oxygen detoxification pathway: O2·- were initially converted to H2O2 by superoxide dismutase SOD2 and superoxide reductase dfx (major role), followed by reduction to H2O via non-heme chloroperoxidase cpo (a newly recognized mechanism in the oxygen detoxification of anammox) and catalase katE. These results expanded the current knowledge of anammox alleviating oxidative stress.

Abstract:Partial denitrification-anammox (PD-anammox) is an innovative process to remove nitrate (NO 3 − −N) and ammonia (NH 4 + −N) simultaneously from wastewater. Stable operation of the PD-anammox process relies on the synergy and competition between anammox bacteria and denitrifiers. However, the mechanism of metabolic between the functional bacteria in the PD-anammox system remains unclear, especially in the treatment of high-strength wastewater. The kinetics of nitrite (NO 2 − −N) accumulation during denitrification was investigated using the Michaelis-Menten equation, and it was found that low concentrations of NO 3 − −N had a more significant effect on the accumulation of NO 2 − −N during denitrification. Organic matter was a key factor to regulate the synergy of anammox and denitrification, and altered the nitrogen removal pathways. The competition for NO 2 − −N caused by high COD concentration was a crucial factor that affecting the system stability. Illumina sequencing techniques demonstrated that excess organic matter promoted the relative abundance of the Denitratesoma genus and the nitrite reductase gene nirS , causing the denitrifying bacteria Denitratisoma to compete with Cadidatus Kuenenia for NO 2 − −N, thereby affecting the stability of the system. Synergistic carbon and nitrogen removal between partial denitrifiers and anammox bacteria can be effectively achieved by controlling the COD and COD/NO 3 − −N.

Abstract:The combination of thiosulfate-dependent denitratation and anammox in a single-stage reactor provides a feasible way to improve total nitrogen removal. The molar ratios of NH4+/NO3- and S2O32-/NO3- were confirmed to be two key factors affecting the reactor performance. The optimal total nitrogen removal efficiency of 99.4% was achieved at NH4+/NO3- of 0.75 and S2O32-/NO3- of 0.85. The multiple thiosulfate oxidation pathways contribute to electron buffers generated in the system. A novel isotope labeling method using 15N was applied to reveal N transformation pathways and a 3-step model was proposed. The nitrate was first converted to nitrite or nitric oxide (NO) by sulfur-oxidizing bacteria. In the second step, both nitrite and NO were utilized by anammox bacteria. Finally, the nitrate generated from anammox could be removed using sulfur deposits as electron donors. The findings provide a potential solution for mainstream nitrogen removal.

Abstract:<h3 class="a-plus-plus">Abstract</h3> Anammox bacteria have attracted attention due to their apparent importance in saving energy and reducing organic chemical demands. Here, we report the detection and quantification of anammox bacteria with an improved primer set in a validation wastewater treatment plant. The improved primer set was shown to detect a broad range of anammox bacteria (47.3%) facilitating more accurate analyses of nitrogen removal mechanisms. Nitrogen removal efficiency and dominant nitrogen removal mechanisms were compared in the modification-Johannesburg (Mod-JHB), modified Ludzack-Ettinger (MLE) single-feed, and anoxic-oxic-anoxic-oxic (AOAO) step-feed modes. In the Mod-JHB configuration, simultaneous nitrification and denitrification (SND) and anammox were found to be responsible for more than 80% of total inorganic nitrogen (TIN) removal (98.5 ± 0.8% of TIN removal). Decrease of anoxic SRT from 5 to 2.5 days did not have any obvious effect on nitrogen removal or the abundance of functional microorganisms. Microbial batch tests demonstrated that both partial nitrification and dissimilatory nitrate reduction to ammonium (DNRA) were responsible for maintaining the anammox process. Short SRT (2 days) in the aerobic zone may explain the presence of partial nitrification. This study provides insights to the analysis of nitrogen removal mechanisms in validation wastewater treatment plants (WWTPs) aiming for high nitrogen removal efficiency.

Abstract:Substrate concentrations greatly influence bacterial growth and metabolism. However, optimal nitrogen concentrations for anammox bacteria in nitrogen‐limited environments remain unclear. Here, we observed enhanced nitrogen metabolism and anabolism of anammox bacteria at low‐nitrogen conditions. Efficient nitrogen removal was achieved at ammonium and nitrite influent concentration of 30 mg/L under HRT of 1h, with an average nitrogen removal rate (NRR) of 0.73 kg N/(m3·d) in I‐ABR composed of four compartments. The highest anammox activity of 6.25 mmol N/ (gVSS·h) was observed in the fourth compartment (C4) with the lowest substrate levels (ammonium and nitrite of 11.6 mg/L and 7 mg/L). This could be resulted from the highest expression level of genes involved in nitrogen metabolism in C4, which was 1.49‐1.67 times higher than that in other compartments. Besides, the second compartment (C2) exhibited the most active anabolism at ammonium and nitrite of 17 mg/L and 13 mg/L, respectively, which contributed to the most active amino acid synthesis and thus the highest EPS (1.35 times higher) in C2. This enhanced amino acid auxotrophy between anammox bacteria with heterotrophs, and consequently, heterotrophs thrived and competed for nitrite. These results hint at the potential application of anammox process in micro‐polluted water.

Abstract:Anammox-based nitrogen removal can advance sustainable wastewater treatment, especially for the mainstream wastewater treatment. Two lab-scale Anammox biofilm reactors fed with influent ammonium concentrations of 110 mg/L (Anammox-H) and 50 mg/L (Anammox-L) were operated, and nitrogen removal and mechanisms of quorum sensing (QS)/quorum quenching (QQ) were examined. The achieved nitrogen removal rates were 0.65 g N/L.d in Anammox-H and 0.25 g N/L.d in Anammox-L. Candidatus Kuenenia was dominant in both reactors. Higher contents of tight extracellular polymeric substance and hydrophobic amino acid confirmed that dense Anammox biofilm developed in Anammox-H. N-hexanoyl-L-homoserine lactone and N-octanoyl-L-homoserine lactone were detected in Anammox-H and Anammox-L. High concentrations of acyl-homoserine lactones (AHLs) in water and biomass phases of Anammox-H were consistent with that more active QS processes existed in Anammox-H. Functional genes including hao, pmoA-amoC, nirK and narGZHY were detected and harbored by Candidatus Kuenenia stuttgartiensis, Nitrosomonas europaea and Lautropia sp. SCN 69-89. These organisms not only involved in nitrogen metabolism, but also in pyruvate, fatty acid and S-Adenosy-L-methionine synthesis (important for AHLs). Influent ammonium concentrations affected the balance between QQ and QS activities, resulting in different Anammox activity and biofilm morphology. This study advanced the Anammox application for low-strength wastewater, especially from the aspect of QQ and QS balance.

Abstract:There is increasing interest to understand the interspecies interactions between microbes of anaerobic ammonium oxidation (anammox) consortia. However, the microbial nitrogen and carbon interactions in inorganic suspended growth anammox consortia are still elusive. Here, we operated a long-term Continuous Stirred Tank Reactor (CSTR) with synthetic inorganic wastewater, which achieved 80% of total nitrogen removal at the steady-state, representing a nitrogen removal rate (NRR) of 1.032 kg-N/(m3·d). Our meta-omics analyses yielded 16 high-quality draft genomes and reconstructed their potential metabolic pathways. Elevated expressions of gene transcripts encoding for enzymes involved in nitrogen and carbon metabolisms were detected in isolates obtained from the same anammox consortia, suggesting their potential metabolic roles and possible microbial interaction capacity in anammox consortia. The expressed genes encoding NO support, nitrite/ammonium loop, dissimilatory nitrate reduction to ammonium, and denitrification in anammox bacteria, and its symbiotic bacteria could have benefits to the bacterial survival and reactor performance. The Carbohydrate-active enzyme analysis revealed that carbohydrate cross-feeding in autotrophic anammox consortia might shift the microbial community. Proteobacteria and anammox bacteria might contribute acetate and glycogen for other symbiotic microorganisms to support their survival. Proteobacteria-affiliated bacteria encoded the potentials of biotin synthesis, hinting at the possibility of the regulation of the diversity of the microbial community. The observed microbial interactions in anammox consortia with inorganic feeding facilitate the purification of anammox bacteria and their engineering application.

Abstract:The low yield of anaerobic ammonium oxidation (anammox) biomass has attracted great attention because of its difficulty to be abundantly enriched. Patterns of substrate supply greatly influence microbial metabolism and behavior. The present study proposed that low nitrogen strength was beneficial to anammox biomass yield and nitrogen removal when comparing a membrane bioreactor (MBR) operated at low nitrogen strength with short hydraulic retention time (HRT) (R-low; influent: fixed at 100 mg-N L-1) and one operated at high nitrogen strength with long HRT (R-stepwise; influent: 100-700 mg-N L-1). Different nitrite concentrations in the two MBRs would indicate discrepant environments, and inevitably resulted in the discrepant microbial responses for anammox community. In particular, we found that at low nitrogen strength, increased activities of purine and pyrimidine metabolism pathways provided more abundant nucleic acids for bacterial proliferation. More active reaction of lipid and protein synthesis favored the synthesis of cellular structure. Importantly, the metabolism of cheaper amino acids was more active under low nitrogen strength, which was coupled with higher metabolic flux and potentially more active exchange of costly amino acids as public goods. In this way, more energy could be saved and applied to biomass yield. Higher active bacterial diversity and more positive interactions among bacterial species in R-low further favored biomass yield and nitrogen removal. The present study highlighted the significant effect of substrate supply patterns on anammox, which is meaningful to overcome the current bottleneck of deficient anammox biomass for application in wastewater treatment.
