Abstract:Slightly acidic electrolyzed water (SAEW) with pH 5.0-6.5 is produced by electrolysis of dilute hydrochloric acid or salt solution in a chamber without a membrane. SAEW can effectively kill various pathogenic bacteria as one of the most potential green disinfectants. However, SAEW is susceptible to being exposed to time, air, and illumination etc. The influence of SAEW for inactivation ofEscherichia coli on white shrimp (Litopenaeus vannamei) under different washing conditions was investigated. At room temperature, the effectiveness of SAEW (with available chlorine concentration (ACC) of 20 mg/L, pH value of 6.1 and ORP of 812 mV) to inactivate Escherichia coli with different dipping times (2, 5 and 10 min), weight ratio of shrimp to SAEW (1:4, 1:10 and 1:20) was evaluated, and changes of ACC were also measured. For inoculation, each shrimp was determined to be intact, and then exposed to a 90°C water bath boiling for 20 min. to inactivate bacteria on the surface of shrimps. Each sample was inoculated with E. coli by spreading 0.5 mL onto each surface respectively and then placed in a clean bench for 20 min. to allow for the attached E. coli. The level of E. coli on the shrimp surface was ensured to be of approximately 6.0-7.0 lgCFU/mL, and bacterial cells on surfaces of shrimp or in residual solution after treatment were counted using a dilution plate count at 37°C for 24h onto nutrient agar, and shrimp that were not washed were used as a control. Moreover, the decay kinetics models of ACC in SAEW during disinfection were established. The effect of SAEW on meat quality was also evaluated, respectively from color, pH value, hardness, and the moisture distribution. The results showed that SAEW has a strong bactericidal activity againstE coli. on white shrimp, while distilled water has a relative weaker role. As the action time prolonged, and the amount of SAEW increased, the bactericidal effect of SAEW was intensified. Meanwhile, the residue bacteria colony in SAEW was also reduced and expenses of active chlorine was increased as the action time was prolonged, and the amount of SAEW increased during the disinfection process. The effective order of time on the bactericidal activity of SAEW was 10 min > 5 min > 2 min under the same ratio of shrimp to SAEW, dipping in SAEW (weight ratio of shrimp to SAEW was 1:4) for 10 min reducedE. coli population from 6.6 to 3.9 lgCFU/mL, which better than the control treatment. E.coli counts in a residual solution can reduce by 1-2 lgCFU/mL. Increasing the amount of SAEW enhanced the bactericidal effect obviously. The decay of ACC and bactericidal kinetic followed first-order kinetics during the disinfection process, and the model had good predicted results. In addition, there was no significant differences between the treated meat quality by SAEW and the controls, respectively from color, pH, hardness, and the moisture distribution of white shrimp meat. Therefore, SAEW has a strong bactericidal activity, and does not affect the meat quality of white shrimp, and has a great potential for application compared with the traditional thermal treatment.

Abstract:The influence of slightly acidic electrolyzed water (SAEW) for inactivation of Escherichia coli on white shrimp (Litopenaeus vannamei) under different washing conditions was investigated. At room temperature, effectiveness of SAEW (with available chlorine concentration of 20 mg/L, pH of 6.1 and ORP of 812 mv ) to inactive Escherichia coli with different dipping times (2 min, 5 min, and 10 min), weight ratio of shrimp to SAEW (1:4, 1:10, and 1:20), with or without vibration were compared. Bacterial cells were counted using a dilution plate count and each sample was inoculated with E coli. with initial concentration of 106 CFU/ml. The results showed that the effective order of time on bactericidal activity of SAEW was 10 min > 5 min > 2 min under the same ratio of shrimp to SAEW. Increasing the amount of SAEW enhanced the bactericidal effect obviously. Significant differences were observed in the bactericidal activity with or without vibration. For the ratio of 1:4, the number of E coli. on shrimp was reduced by 1.60 lgCFU/ml after treatment for 5 min without vibration, while 3.30 lgCFU/ml bacterial reduced with vibration at 2000 rpm. This study suggests that the SAEW treatment with suitable washing method could be a practicable way to effectively control Escherichia coli contamination on white shrimp.

Abstract:Foodborne disease outbreak caused by food microbiological contamination is a serious public health problem. Slightly acidic electrolyzed water (SAEW), a new ultra-high effect and wide-spectrum disinfectant that is colourless, odourless, and harmless to humans and the environment, is directly used on food surfaces in Japan and America. However, the underlying inactivation mechanism remains unknown. In this study, biochemical and cellular changes were observed to investigate the bactericidal mechanism of SAEW against Escherichia coli (E. coli). The results indicated that SAEW with a pH of 6.40, an oxidation-reduction potential (ORP) of 910 mV, an available chlorine concentration (ACC) of 60 mg/L, and a volume ratio of 20:1, produced the most effective sterilization action. A fluorescence-based live-dead assay was further used to demonstrate the sterilized effect and the cell esterase activity damage caused by SAEW. During the observation period, within 10 min, the cell morphology changed, which was characterized by cell expansion, cell elongation and increased membrane permeability. Meanwhile, reactive oxygen substances (ROS) were released in the bacterial cells. E. coli inactivation and apoptosis induced by SAEW were observed. Our findings illustrate that the bactericidal effects of SAEW against E. coli occurred through cellular and biochemical mechanisms of cell necrosis and apoptosis.

Abstract:The objective of this study was to investigate the inactivation efficacy of slightly acidic electrolyzed water (SAEW, pH 6.31-6.85) in the process of raw frozen shrimp handling and postharvest preservation. The experiment was carried out in the processing workshop of raw frozen shrimp of Zhejiang Yueteng Aquatic Food Co., Ltd., Hangzhou, China. The inactivation effect of SAEW in the process of raw frozen shrimp was considered on the side of raw shrimp, gloves as well as contact surfaces. Shrimp samples were treated with SAEW (29 or 16 mg L-1 of available chlorine) for 3 or 5 min under spraying conditions, meanwhile, gloves and contacting surfaces were treated with SAEW. The quality of raw frozen shrimp in the duration of storage was observed for 11 days. The bacteria count was reduced by 0.21 similar to 0.72 log units after the treatment of SAEW. The effect of SAEW on gloves was better than that of alcohol (P < 0.05), the number of survival bacteria was reduced by 1.92 log(10) CFU cm(-2) for 30 s. A significant difference (P < 0.05) was observed between SAEW and sodium hypochlorite solution on contact surfaces in bactericidal activity. Finally, the survival total bacteria and coliforms of shrimp were effectively controlled in storage, and there was no significant change (P > 0.05) on the pH values and residual chlorine of shrimp meat. This study suggests that SAEW treatment can be a way to effectively control bacteria on shrimp processing environment and to improve the quality of shrimp during storage.

Abstract:Mixed solution of slightly acidic electrolyzed water (SAEW) and artificial seawater was used to investigate the disinfection potential of SAEW in artificial seawater. Inoculated Vibrio parahaemolyticus (suspended in 3% sodium chloride alkaline peptone water and 0.85% sodium chloride water, respectively) was subjected to different mixed-SAEW and SAEW immersion treatments (5-20 mg/L available chlorine concentration (ACC)). In the presence of organic matter, 4.07 logCFU/mL significant reduction (p<0.05) was achieved after treating with 20 mg/L mixed-SAEW for 15 min. There was 5.13 logCFU/mL reduction after treating with 15 mg/L SAEW for 15 min. For V. parahaemolyticus suspended in 0.85% sodium chloride solution, it was undetected after 30 s SAEW treatment (5 mg/L ACC) or 120 s mixed-SAEW treatment (10 mg/L ACC). At a ratio of SAEW and artificial seawater at 1:15 (V/V), SAEW could inactivate V. parahaemolyticus to undetectable level in artificial seawater in one minute, which was comparable with UV treatment of 10 W. The results indicated high sanitization potential of SAEW against V. parahaemolyticus in aquaculture seawater. Abstract:This work investigates the efficacy of a combined sanitization approach involving pretreatment of nanoaerosolized water followed by H2O2 dip-washing against Salmonella Typhimurium LT2 on spinach. The addition of a pretreatment step utilizing nano-aerosolized water for 5 min significantly increased the sanitization efficacy: The number of surviving bacteria was 5.2 +/- 0.8 CFU/g, 4.7 +/- 0.2 CFU/g, and 1.1 +/- 1.1 CFU/g for H2O2 dip-washing only, H2O2 spraying only, and for the combined treatment, respectively. The enhanced efficacy of the combined treatment was attributed to the ability of nano-aerosolized water to reach and fill crevices that tended to form micro airpockets with the liquid (bulk) sanitizer. Overall, these findings may suggest that a short priming step involving nano-aerosolized water may be incorporated in traditional liquid sanitization approaches to enhance their effectiveness. Abstract:This study assessed the combined effect of slightly acidic electrolyzed water (SAEW) and slightly acidic electrolyzed water ice (SAEW-ice) on the quality of pomfrets over a period of 18 d of cold storage at 4 degrees C. A presoak for 5 min in SAEW solution (22 mg/L) was used before the pomfrets were placed on SAEW-ice (pH: 6.45; ORP: 803 mV; ACC: 18 mg/L); The changes in physicochemical properties (i.e., pH, thiobarbituric acid, total volatile basic nitrogen and texture profile), microbial loads and sensory characteristics were all analyzed. Compared with the tap water (TW) group, the total bacterial counts of the SAEW group significantly decreased by 1.27 log(10) CFU/g after immersion (p<0.05). The shelf life of the pomfrets was prolonged by 9 d by the combined treatment of SAEW and SAEW-ice during storage at 4 degrees C. On the 18th day, the gumminess and chewiness values of the pomfrets in the SAEW+SAEW-ice group were 195 g and 3.97 mJ, respectively, which were significantly higher than those of the other groups (p<0.05). The results suggested that SAEW+SAEW-ice treatments have great potential as a novel method to maintain the quality and extend the shelf life of pomfrets during refrigerated storage.

Abstract:This study investigated the suppressive effect and the inactivation mechanism of slightly acidic electrolyzed water (SAEW) on Aeromonas hydrophila. The experiment compared different inhibition effects with SAEW and two antibiotics, gentamicin and norfloxacin. The conditions of pH of 6.5, oxidation-reduction potential (ORP) of 890 mV, available chlorine concentration (ACC) of 28 mg L-1 of SAEW, and volume ratios of 1: 10, 1: 15, and 1: 20 were used. Using exposure times of 5 and 10 min to analyze the inactivation mechanism of SAEW from the perspectives of cell membrane permeability, cell nuclear morphology, cell fluorescence staining, and other aspects, the anti-infection effects of SAEW were tested using tilapia (Oreochromis niloticus) culture experiments. SAEW had a stronger bacteriostasis effect than the antibiotics used. SAEW can inhibit the ability to reproduce, causing abnormal cell morphology and cell elongation. The cell volume was increased with a change in the membrane permeability. The cell nuclear morphology was changed, further inducing apoptosis. Based on in vitro study of tilapia in a control group, the organs (intestine, stomach, and liver) developed different degrees of infection. No obvious infection was observed in muscle samples. After SAEW treatment, SAEW showed a better antibacterial effect and improvement effects on the inflammation and structural damage induced by A. hydrophila, which could be helpful in aquaculture application.
Impact of slightly acidic and alkaline electrolyzed water on shelf-life of the chilled chicken fillet

Abstract:Electrolyzed Water is a potent multifunctional antibacterial agent with numerous applications in food safety via processing, storage as well as it has great importance in agriculture and medicinal applications. The current study investigate the powerful of the sanitizing effect of slightly acidic (SAcEW) as well as the detergent effect of slightly alkaline electrolyzed water (SAlEW) (individually and in combination) on chilled chicken fillets subjected to a dipping treatment for 5 min and stored refrigerated at 4±1°C. When treated were compared to untreated samples, it was turned out to be that SAlEW and SAcEW treatments increased the shelf life of chilled chicken fillet and remained fit for consumption till 7th and 9th days, recorded APC 4.89±0.01 and 4.9±0.0, psychrophilic plate count 3.8±0.02 and 2.18±0.03, E. coli count 1.97±0.03 and 1.98±0.02 and Molds count 3.84±0.02 and 3.56±0.68, respectively. Moreover, dipping of chicken fillet in SAlEW followed by SAcEW resulted in prolong the shelf-life until the 11th day of storage, recorded APC 4.94±0.01, psychrophilic plate count 4.67±0.02, E. coli count 1.98±0.03 and Molds count 3.88±0.03. All treatments were effective in reducing microbial populations throughout the storage, with combined treatment showing the strongest antimicrobial activity. So, EW can be effectively used to eliminate or at least to reduce bacterial contamination to an acceptable Food safety limits listed in national and international standards, references and recognized food constitutions that considered being safe for human consumption and subsequently, extending shelf life of chilled foods including chicken fillet during processing, distribution, storage and marketing.

Abstract:Slightly acidic electrolyzed water (SAEW) has been recognized as an effective bactericidal agent with free chlorine, but its limitations include its instability and its great dependence on equipment. Newly developed circulating electrolyzed water (CEW) with a higher available chlorine concentration (ACC) could successfully overcome these limitations. In this study, SAEW (ACC of 20 mg/L), CEW1 (ACC of 200 mg/L), and CEW2 (ACC of 20 mg/L) were evaluated for changes in properties (pH, oxidization reduction potential [ORP], and ACC) during storage in open or closed glass bottles under light or dark conditions at room temperature (approximately 20 degrees C) and after washing pork and lettuce. Additionally, the washed pork and lettuce were evaluated for total viable counts, pH and general appearance. The results showed that CEW1 with a higher ACC has better stability than SAEW with a lower ACC for the storage and washing experiments, and CEW still remained stable after dilution with distilled water. The property indices of EWwere greatly affected for the pork-washing experiments compared with the lettuce-washing experiments, probably due to the existence of alkaline and organic materials on the surface of pork. Furthermore, EWs were more effective for inactivating microbes in lettuce than in pork, while there was no significant difference in tissue pH and the general appearance of pork and lettuce. These findings indicated that CEW with a higher ACC shows potential for reducing foodborne pathogens on pork and lettuce without effects on their physicochemical characteristics, and it can be applied in a diluted form.

Abstract:The present study investigates a small-scale system comprising a sequential treatment with slightly acidic electrolyzed water (SAEW) washing and UVC light-emitting diodes (LEDs) for the inactivation of Salmonella on lettuce. Lettuce was inoculated with a Salmonella cocktail to achieve a final concentration of 6.67 log(10) CFU/g. Alone treatment by SAEW washing (available chlorine concentration-ACC 20-80 ppm, 1-7 min) or UVC-LED irradiation (50-200 mu W/cm(2), 1-30 min) resulted in the reduction of 1-1.8 log(10) CFU/g Salmonella on lettuce. The sequential treatment with SAEW washing and UVC-LED significantly (p < 0.05) enhance the reduction of Salmonella on lettuce to 2.56-2.97 log(10) CFU/g. The results also revealed that the sequential treatment did not result in obvious compromise in the qualities (color, chlorophyll, and total soluble solid content) of lettuce. Therefore, the proposed sequential treatment of SAEW washing and UVC-LED in this study has the potential to assure the food safety of fresh produce.
Combined Effect of High Pressure and Slightly Acidic Electrolyzed Water on Sterilization of Fresh-cut Fruits and Vegetables

Abstract:Due to the operation of cutting,fresh-cut fruits and vegetables are susceptible to microbial infection.In order to extend the shelf life of fresh-cut fruits and vegetables,the effects of the concentration and amount of available chlorine of slightly acidic electrolyzed water (SAEW) and the pressure level and holding time of high pressure (HP) on sterilization of fresh-cut carrots were studied through single factor experiment.Consequently,the combination treatment conditions were as follows:the concentration of available chlorine of slightly acidic electrolyzed water was 30 mg/L and the amount of available chlorine of slightly acidic electrolyzed water was 200 mL;the pressure level ranged from 100 MPa to 400 MPa,holding time was 5 min and the total processing time was 15 min.Fresh-cut carrots and apples were treated by the different combined treatments,and both brain heart infusion agar (BHIA) and vilolet red bile agar (VRBA) were used for the detection of E.coli.With the aim to explore the inactivation effects of combination treatment,during the detection,the slightly acidic electrolyzed water was chosen instead of HP sterilized water to do further test.The result shows that combined treatment can improve the efficiency of inactivation,but the enhancement was not significant at lower pressure.At 400 MPa,HP treatment combined with SAEW displayed the most significant enhancement of inactivation efficiency,where E.coli could not be detected on fresh-cut carrot,but some E.coli was still detected on the fresh-cut apples in BHIA.When comparing the results in BHIA and VRBA,it can be seen that the SAEW showed significant lethal effect on E.coli,while HP showed both lethal and sublethal effect on E.coli.
Antibacterial activity and mechanism of slightly acidic electrolyzed water combined with ultraviolet light against Salmonella enteritidis

Abstract:This study aimed to investigate the inactivation effect and mechanism of combined treatment with slightly acidic electrolyzed water (SAEW) and ultraviolet light (UV, 253.7 nm, 10 W) (SAEW + UV) on Salmonella enteritidis . The result showed that S. enteritidis was 100% inactivated (9.21 log CFU/mL) by SAEW + UV at an available chlorine concentration (ACC) of 50 mg/L for 60 s. At the same time, the reduction value of SAEW and UV alone treatment was 6.61 and 2.27 log CFU/mL, respectively. The constructed model (R 2 = 0.993) demonstrated that SAEW + UV is a highly effective germicidal method under organic matter interference (P < 0.05). The bactericidal effect was positively correlated with time and ACC, and there was also a significant interaction between time and ACC (P < 0.05). In addition, the cell membrane conductivity of S. enteritidis increased, intracellular proteins and nucleic acids exuded, intracellular ROS accumulated, and ATP decreased after SAEW + UV treatment for 60 s at ACC of 10 mg/L (P < 0.05). The results showed that SAEW + UV might disrupt the bacterial cell membrane and lead to the accumulation of intracellular ROS and a decrease of intracellular ATP, which resulted in bacterial death. Further studies are needed to understand the specific ROS production pathways of SAEW + UV on S. enteritidis.

A Successful Technique for the Surface Decontamination of Salmonella enterica Serovar Typhimurium Externally Contaminated Whole Shell Eggs Using Common Commercial Kitchen Equipment

Abstract:Eggs are a highly nutritious food source used in a wide range of food products. In Australia, eggs are a frequent source of foodborne salmonellosis outbreaks, associated with eggshell contamination with Salmonella enterica serovar Typhimurium (ST). Despite their potentially hazardous nature, raw eggs are often used and consumed in mayonnaise, mousse, ice cream and eggnog. The aim of this study was to develop a shell egg decontamination method that removed ST contamination from the outside of an egg without impacting its usability. The decontamination method was developed by the adaptation of a temperature-controlled water bath (commonly present in kitchens and associated with the sous-vide technique) for the surface decontamination of eggs. The outside of whole eggs was artificially inoculated with two ST strains. The eggs were decontaminated by placing in a sous-vide cooker with the water heated to 57°C. The remaining viable ST present on the whole shell egg, crushed shells, internal egg contents and sous-vide water were enumerated over time by culturing onto XLD agar. The quality of the uncontaminated heat-treated eggs was determined by measuring the Haugh unit, yolk index, albumen pH, thermocoagulation, and stability of foam. A blind control study was conducted to assess the acceptability and usability of the treated eggs by chefs and food handlers for the preparation of mayonnaise. Complete decontamination of ST was achieved by treating eggs for 9 min in the sous-vide cooker (57°C). No statistically significant difference was observed in the quality of treated eggs compared with nontreated eggs using the quality measurements and acceptability score from chefs. This method provides a simple approach that can be adopted by chefs and food handlers to obtain safe eggs before the preparation of raw egg products.

Abstract:Poultry and livestock contribute a significant amount of energy and protein to the human diet. However, a main concern is the undesirable microbial population from the poultry and livestock system that may cause diseases in both humans and animals. Electrolyzed water has shown broad-spectrum sanitization efficiency and gained considerable popularity. This review aims to provide an overview of the use of electrolyzed water under different environment conditions for sanitization in livestock and poultry systems. Electrolyzed water serves as an excellent antimicrobial and anti-biofilm agent against various pathogenic, disease-producing, and undesirable microbes in every step of the livestock and poultry sectors from farm to fork. The synergistic effects of the oxidation reduction potential, pH, and chlorine concentration of electrolyzed water enable its potential and prompt mechanism for controlling and eliminating various microorganisms. Compared to other conventional disinfectants, electrolyzed water has been shown to be more efficient and effective. As an environmentally-friendly sanitizer, electrolyzed water has been applied in livestock and poultry production and processing sectors such as animal drinking water, feed decontamination, house sanitation, transportation system, slaughterhouse sanitation, meat-processing industry decontamination, sanitization of milk-processing industries, hatchery and eggshell decontamination. This review is aimed to discuss the microbial control in livestock and poultry industry by applying electrolyzed water as a novel sanitizer.

Abstract:Among the animal derived food products, contamination of poultry eggs, and egg shell surface is one of the major causes for foodborne salmonellosis in the United States. As a means of reducing the pathogen transfer to the internal egg contents, polysaccharide-based coatings containing antimicrobial phytochemicals could potentially serve as a biocontrol strategy for shelled egg products. The current study investigated the efficacy of four GRAS (Generally Recognized as Safe)-status plant-derived compounds, namely, caproic acid (CAO), caprylic acid (CAY), linalool (LIN) and cuminaldehyde (CUM), as pectin-based coating treatments, individually or in combination, for reducing Salmonella Heidelberg (SH) on shell eggs. A three-strain mixture of SH (~8.0 log CFU in 50 μL inoculum) was spot-inoculated on surface sterilized white-shelled eggs. Eggs were evenly coated with either pectin-based treatments of CAO (1%), CAY (1%), LIN (1%) and CUM (1%), individually, or a combination of 4 phytochemicals (COMB- each phytochemical at 0.5% v/v level of inclusion). The treated eggs were stored at 4°C and SH counts were enumerated on days 0, 1, 3, 5, 7, 14, and 21 of storage. The study was replicated thrice, 3 eggs/treatment/day time point, and the data were analyzed using two-way ANOVA with significance tested at p < 0.05. On day 0, pectin-coated control eggs had ~7.6 log CFU of SH/egg. At the end of refrigerated storage (day 21), pectin-based coating of CAO and CAY at 1% level reduced SH by 2.0–2.5 log CFU/egg ( P < 0.05) when compared to controls. In addition, the CUM and LIN based coatings produced 3.0 log and 3.9 log reduction, respectively, in SH counts on eggs by day 21 of storage. Among the treatments with phytochemical combinations, COMB1 [pectin (2%) + Caprylic acid, caproic acid and cuminaldehyde (each at 0.5% level)] was found to be most effective, reducing SH counts to 2.5–3.3 log CFU/egg from day 0 through day 14, and by the end of storage period (day 21), a 3.5 log CFU reduction/egg ( p < 0.05) compared to untreated controls. Morphological studies of treated eggs using atomic force microscopy (AFM) have shown that the roughness of eggs can be influenced by a combination of various compounds. Results indicate the potential efficacy of the aforesaid phytochemicals in reducing SH on shell eggs; however, further studies investigating their industrial feasibility and effects on sensory attributes of eggs are warranted.

Abstract:Salmonella Typhimurium (S. Typhimurium) contamination poses a significant challenge to breeder egg hatchability and chick health, necessitating the exploration of alternative disinfection methods. This study investigates the potential of phage vB_SPuM_SP02 (SP02) as a novel disinfectant for breeder eggs contaminated with S. Typhimurium SM022. Phage SP02 was isolated from poultry farm effluent and characterized for morphology, biological properties, and genome properties. Experimental groups of specific pathogen-free (SPF) eggs were treated with Salmonella and phage SP02, and efficacy was assessed through hatching rates, chick survival, weight, Salmonella load, immune organ indices, and intestinal flora. Phage treatment effectively eradicated Salmonella contamination on eggshells within 12 h, resulting in increased hatching and survival rates compared to controls. Furthermore, phage treatment mitigated weight loss and tissue Salmonella load in chicks without causing immune organ damage while reducing Salmonella spp. abundance in the intestinal tract. This study demonstrates the potential of phage SP02 as an eco-friendly and efficient disinfectant for S. Typhimurium-contaminated breeder eggs, offering promising prospects for practical application in poultry production.

Abstract:AbstractThe Influence of water source on the production of slightly acidic electrolyzed water (SAEW) and its sanitization efficacy were investigated. Two different water sources (tap water (TW) and underground water (UGW)) were applied to produce slightly acidic electrolyzed water (SAEW) at same setting current, with similar electrolyte flow rate (EFR) and concentration. Properties of SAEW were evaluated based on pH, Available chlorine concentration (ACC) and oxidation-reduction potential (ORP). Methods for the optimization of SAEW production process was examined to obtain high ACC value by implanting different types of electrolytes. Effect of ACC and pH of SAEW were evaluated in vitro towards inactivate foodborne pathogens. The results indicated that TW with hardness of 29 ppm produced effectively SAEW than through UGW (12 ppm) using electrolytes. Likewise, low water hardness could be reinforced by combining HCL with a salt (NaCl or KCL). The optimized SAEW production system was determined at 4% HCl + 2.0 M KCL with EFR of 2 mL/min and 4% HCl + 3.0 M KCL with EFR of 2 mL/min resulting in higher ACC value of 56.5 and 65.5 ppm, respectively using TW. Pathogenic vegetative cells were completely inactivated within 1 min of treatment in SAEW with 20 ppm. Viability observations using Confocal and TEM Microscopy, Flow cytometry, and antimicrobial activity were carried out to confirm the sanitizing effect and cell membrane disruption. Based on the experimental results obtained, it provides a foundation for future advancement towards commercial application of SAEW in the food and agricultural industries.

Abstract:Various pathogenic microorganisms present in municipal solid waste (MSW) can pose significant hazards to both the environment and public health. Spraying Slightly Acidic Electrolyzed Water (SAEW) is an effective method for decontaminating microorganisms in the air and on surfaces in the waste sorting rooms. SAEW treatment, with an effective chlorine concentration (ECC) of 28.4–44.5 mg/L and a pH range of 5.0–6.5, was conducted in the waste sorting rooms of the old, high-rise, and villa residential communities in Shanghai, China. The efficacy of SAEW treatment was assessed by examining the microbial population in the air, on waste containers, on sanitation workers' hands, and the microbial community structure of MSW samples. SAEW treatment resulted in the total bacterial reduction ranging from 0.07 to 1.70 log 10 colony-forming units (CFU)/cm 2 on waste containers, with corresponding reduction efficiencies between 15.7% and 98.0%. Additionally, the positive rates of coliforms were decreased by 18.7%–95.2%. Furthermore, the application of SAEW demonstrated a significant impact on reducing airborne bacterial and fungal levels. SAEW was proven effective on object surfaces, in the air and on human skin, achieving disinfection efficiencies ranging from 22.7% to 54.3% on sanitation workers' hands. And the most significant inhibition of microbial growth was observed in the waste sorting room of the villa residential community. The microbial diversity and structure of MSW samples exhibited noticeable differences before and after the SAEW application. These findings suggest that SAEW holds significant potential as an ideal, environmentally friendly broad-spectrum disinfectant for preventing and controlling microbial contamination in the waste sorting rooms.

Abstract:In the natural environment, most microorganisms live in mixed-species biofilms, in which the metabolism and growth of organisms are different from that in single-species biofilms. Adhesive bacteria and their biofilms on the surface of food processing equipment are the sources of cross-contamination, leading to the risk for humans. Slightly acidic electrolyzed water (SAEW) has been proposed as a novel sanitizer in the food and agriculture industry. In this study, we investigated the changes in the physical properties of SAEW under different conditions and the disinfection abilities of SAEW against spore-forming and non-spore-forming pathogens. Furthermore, we examined the disinfection abilities of SAEW after 12 months of shelf life on a mixed-species biofilm of Listeria monocytogenes Scott A and Staphylococcus aureus . The results showed that SAEW at 30 and 50 ppm achieved all-kill of the spore-forming pathogen Bacillus cereus within 30 s. Changes in the ACC and pH of the produced SAEW were generally affected by the storage conditions. Both spore-forming and non-spore-forming pathogens were not detected under treatment with 50 ppm SAEW for 5 min under HDPE-closed conditions throughout the whole storage period. Moreover, 25 mg/L SAEW can inactivate L. monocytogenes Scott A and S. aureus biofilm cells in ~2.45 and 2.57 log CFU/mL in biofilms within 5-min treatment. However, the decline of the two bacteria in the mixed-species biofilm was 1.95 and 1.43 log CFU/mL, respectively. The changes in the cell membrane permeability of the mixed-species biofilm under treatment with SAEW were observed by using atomic force microscopy and confocal laser scanning microscopy. L. monocytogenes Scott A was more sensitive to SAEW in the mixed-species biofilm cells. These findings exhibited strong antibiofilm activities of SAEW in impairing biofilm cell membranes, decreasing cell density, and eliminating biofilm, which suggest that SAEW is an excellent antibacterial agent in the food processing industries.
