Abstract:We present a re-usable enzyme catalyst system via direct encapsulation of cellobiase in nonsurfactant templated sol–gel mesoporous silica host material with d-fructose as the template. The pore diameter and porosity of the silica host material, controlled by the fructose content, controlled the diffusion of substrate to the enzyme. This in situ immobilized cellobiase showed little or no leakage while could be repeatedly used as biocatalyst with little or no loss of activity after at least 9 cycles.

Abstract:Biocatalytic reactions in living cells involve complex transformations in the spatially confined microenvironments. Inspired by biological transformation processes, we demonstrate effective biocatalytic cascade driven photodynamic therapy in tumor-bearing mice by the integration of an artificial enzyme (ultrasmall Au nanoparticles) with upconversion nanoparticles (NaYF4@NaYb0.92F4:Er0.08@NaYF4)zirconium/iron porphyrin metal-organic framework core-shell nanoparticles (UMOF NPs) which act as biocatalysts and nanoreactors. The construction of core-shell UMOF NPs are realized by using a unique "solvent-assisted self-assembly" method. The integration of ultrasmall AuNPs on the UMOFs matrix leads to glucose depletion, providing Au-mediated cancer therapy via glucose oxidase like catalytic activity. Meanwhile, the UMOF matrix acts as a near-infrared (NIR) light photon-activated singlet oxygen generator through a continuous supply of oxygen via hydrogen peroxide decomposition upon irradiation. Such kinds of biocatalysts offer exciting opportunities for biomedical, catalytical ,and energy applications.

Abstract:Biocatalytic transformation in nature is inherently dynamic, spontaneous, and adaptive, enabling complex chemical synthesis and metabolism. These processes often involve supramolecular recognition among cells, enzymes, and biomacromolecules, far surpassing the capabilities of isolated cells and enzymes used in industrial synthesis. Inspired by nature, here we design a supramolecular approach to equip living cells with these capacities, enabling recyclable, efficient cascade reactions. Our two-step "plug-and-play" methodology begins by coating Escherichia coli cells with guest-containing polymers (SupraBAC) via supramolecular charge interactions, followed by the introduction of β-cyclodextrin-functionalized host enzymes through host-guest chemistry, creating a robust cell-enzyme complex. This supramolecular coating not only protects cells from various stresses, such as UV radiation, heat, and organic solvents, but also facilitates the overexpression of intracellular enzymes and the attachment of extracellular enzymes within and on SupraBAC. This combination results in efficient multienzyme cascade synthesis, enabling two- and three-step reactions in one pot. Importantly, the multienzyme system can be recycled up to five times without significant loss of activity. Our findings introduce a versatile, adaptive supramolecular coating for whole-cell catalysts, offering a sustainable and efficient solution for complex synthesis in both chemistry and industrial biotechnology.

Abstract:The development of programmable microscale materials with cell-like functions, dynamics and collective behaviour is an important milestone in systems chemistry, soft matter bioengineering and synthetic protobiology. Here, polymer/nucleotide coacervate micro-droplets are reconfigured into membrane-bounded polyoxometalate coacervate vesicles (PCVs) in the presence of a bio-inspired Ru-based polyoxometalate catalyst to produce synzyme protocells (Ru4PCVs) with catalase-like activity. We exploit the synthetic protocells for the implementation of multi-compartmentalized cell-like models capable of collective synzyme-mediated buoyancy, parallel catalytic processing in individual horseradish peroxidase-containing Ru4PCVs, and chemical signalling in distributed or encapsulated multi-catalytic protocell communities. Our results highlight a new type of catalytic micro-compartment with multi-functional activity and provide a step towards the development of protocell reaction networks.

Abstract:The 2-substituted quinazoline moiety is the core structure of many fine chemicals, pharmaceutical agents and functional molecules. The effective synthetic strategies for the 2-substituted quinazolines are highly significant. Herein, we presented a new strategy to prepare various SACs (Mn-SA@YBC, Ni-SA@YBC, Cu-SA@YBC and Co-SA@YBC) supported on yeast-derived biochar materials for the first time. These SACs remained the original morphology of yeasts. The studies of catalytic performance of SACs revealed that Mn-SA@YBC exhibited excellent catalytic performance than the Mn-nano@YBC in the dehydrogenation of 2-aminobenzyl alcohol and aromatic nitriles forming 2-substituted quinazolines. The Mn-SA@YBC exhibited good stability, and still showed high catalytic activity without any significant loss after recycled six times. The atomically dispersed Mn atoms were observed in the aberration-corrected high-angle annular-dark-field scanning transmission electron microscopy (HAADF-STEM) images of the freshly prepared and recycled Mn-SA@YBC which proved the good stability and reusability of the Mn-SA@YBC. XANES and XPS analysis indicated that the Mn atom of Mn-SA@YBC had the Mn-N3P coordination structure which was different from previous reports. A reasonable reaction mechanism had been proposed. It was believed that this work paved a new way for the design and preparation of active single atomic site catalysts on biochar materials.

Abstract:Biological catalysis and chemical catalysis are generally considered as two separate research areas, each with its unique challenges and opportunities.[1] However, the boundary between these two areas has been becoming increasingly blurred due to rapid advances in synthetic biology and development of new water‐compatible chemical catalysts. In this talk, I will discuss our recent efforts in exploring the synergy between biological catalysis and chemical catalysis and highlight three examples. The first example involves the design and development of a tandem catalytic process consisting of an olefin metathesis catalyst and a cytochrome P450 enzyme in a biphasic system, which converts a mixture of alkenes to a single epoxide.[2] The metathesis catalyst leads to an equilibrating mixture of alkenes in the organic phase, and the P450 enzyme converts one member of the mixture of alkenes to the corresponding epoxide in the aqueous phase. The second example involves the design and development of a tandem catalytic process consisting of a thermophilic glucose isomerase enzyme for glucose isomerization to fructose and a solid acid catalyst for fructose dehydration to 5‐hydroxymethylfurfural.[3] The third example involves the metabolic engineering of a recombinant E. coli strain which converts D‐galacturonic acid released from sugar beet residues by enzymatic hydrolysis to mucic acid. Abstract:Tissue-derived cultured cells exhibit a remarkable range of morphological features in vitro, depending on phenotypic expression and environmental interactions. Translation of these cellular architectures into inorganic materials would provide routes to generate hierarchical nanomaterials with stabilized structures and functions. Here, we describe the fabrication of cell/silica composites (CSCs) and their conversion to silica replicas using mammalian cells as scaffolds to direct complex structure formation. Under mildly acidic solution conditions, silica deposition is restricted to the molecularly crowded cellular template. Inter- and intracellular heterogeneity from the nano- to macroscale is captured and dimensionally preserved in CSCs following drying and subjection to extreme temperatures allowing, for instance, size and shape preserving pyrolysis of cellular architectures to form conductive carbon replicas. The structural and behavioral malleability of the starting material (cultured cells) provides opportunities to develop robust and economical biocomposites with programmed structures and functions.

Abstract:The cell membrane, as the supporting structure of cells, maintains the stability of the internal microenvironment of cells for whole-cell catalysis, but is also a barrier in transmembrane transport of substrate and product. However, methods for improving cell membrane permeability are usually accompanied with reduced membrane structural strength. Here, we show that a self-assembled biocomposite formed by Mg2+ ions and sodium deoxycholate (NaDC) can form sea urchin-shaped 3D complexes, incorporating living microbial cells. Cell membrane permeability was significantly improved by a fused structure between cell and Mg(DC)2 nanoparticles without membrane damage. Due to with hydrophobic mesopores, hydrophobic core and disseminated structure, the biocomposite greatly improved catalytic activity (up to 427.3%) of an environment-sensitive P450 enzyme expressed in Escherichia coli, which catalyzed hydroxylation with aromatic hydrocarbon. We also show that the biocomposite successfully improved the membrane permeability of live Bacillus cereus and Saccharomyces cerevisiae, making it a promising application platform for improving membrane permeability in whole-cell catalysis.
Screening of Protein-Based Ultrasmall Nanozymes for Building Cell-Mimicking Catalytic Vesicles

Abstract:Enzymes are an important component for bottom-up building of synthetic/artificial cells. Nanozymes are nanomaterials with intrinsic enzyme-like properties, however, the construction of synthetic cells using nanozymes is difficult owing to their high surface energy or large size. Herein, the authors show a protein-based general platform that biomimetically integrates various ultrasmall metal nanozymes into protein shells. Specifically, eight metal-based ultrasmall nano-particles/clusters are in situ incorporated into ferritin nanocages that are self-assembled by 24 subunits of ferritin heavy chain. As a nanozyme generator, such a platform is suitable for screening the desired enzyme-like activities, including peroxidase (POD), oxidase (OXD), catalase (CAT) and superoxide dismutase (SOD). After screening, it is found that Ru intrinsically possesses the highest POD-like and CAT-like activities, while Mn and Pt show the highest OXD-like and SOD-like activities, respectively. Additionally, the inducers/inhibitors of various nanozymes are screened from more than 50 compounds to improve or inhibit their enzyme-like activities. Based on the screened nanozymes and their inhibitors, a proof-of-conceptually constructs cell-mimicking catalytic vesicles to mimic or modulate the events of redox homeostasis in living cells. This study offers a type of artificial metalloenzyme based on nanotechnology and shows a choice for bottom-up enzyme-based synthetic cell systems in a fully synthetic manner.
Rational design of artificial biofilms as sustainable supports for whole‐cell catalysis through integrating extra‐ and intracellular catalysis

Abstract:Biofilms are promising candidates for sustainable bioprocessing applications. This work presents a rational design of biofilm catalysts by integrating extra‐ and intracellular catalysis systems with optimized substrate channeling to realize efficient multistep biosynthesis. An assembly of four enzymes in a "three‐in‐one" structure is achieved by rationally placing the enzymes on curli nanofibers, the cell surface, and inside cells. The catalytic efficiency of the biofilm catalysts is over 2.8 folds higher than that of the control whole‐cell catalysis when the substrate benzaldehyde is fed at 100 mM. The highest yield of D‐phenyllactic acid catalyzed by biofilm catalysts under optimized conditions is 102.19 mM, also much higher than that of the control catalysis test (52.29 mM). The results demonstrate that engineered biofilms are greatly promising in integrating extra‐ and intracellular catalysis, illustrating great potentials of rational design in constructing biofilm catalysts as sustainable supports for whole‐cell catalysis.

Abstract:Enzymatic cascades catalysis engenders complex and selective intracellular transformations due to the organization of the enzymes in confined cellular environments. In this work, a biomimetic strategy is employed to develop a synthetic cell analogue for operating multi-enzymatic cascade catalysis in vitro . The glucose peroxidase (GOx) and chloroperoxidase (CPO) are co-encapsulated in the channel of dendritic mesoporous silica nanoparticles (DMSP) modified by phase-transitioned lysozyme (&). The cascade efficiency is substantially enhanced compared with the diffusion mixtures of the free enzymes in bulk solution due to some nanoscale construction, such as confinement effect, proximity effect and special distribution mode of the enzymes. The high specific surface area and high concentration of mesoporous cages in DMSP is beneficial for encapsulation of enzyme. The cross-linked cavities ensure directional transfer of the intermediate from GOx to CPO, known as "substrate channel". PTL plays multiple functions including inhibiting leaking of enzyme from DMSP, concentrating intermediate H 2 O 2 at DMSP domains and keeping this biocomposite stable in aqueous environment by improving its dispersibility. The prepared biocomposite & is very stable at harsh conditions. It can retain 42% of original catalytic activity at 90°C with incubation for 3 h while the retained activity of free enzymes is only 5% under the same conditions. Meanwhile, it can keep 92.4% of its original catalytic activity after 10 reuses. These characteristics ensure practical application of this biocatalytic material. The active HClO generated by GOx-CPO cascade has an efficient anti-tumor effect and is effectively applied to preserve fruit.

Abstract:Abstract Cells possess isolated compartments that spatially confine different enzymes, enabling high-efficiency enzymatic cascade reactions. Herein, we report a cell-inspired design of biomimetic cascade catalysis system by immobilizing Fe single atoms and Au nanoparticles on the inner and outer layers of three-dimensional nanocapsules, respectively. The different metal sites catalyze independently and work synergistically to enable engineered and cascade glucose detection. The biomimetic catalysis system demonstrates ~ 9.8- and 2-fold cascade activity enhancement than conventional mixing and coplanar construction systems, respectively. Furthermore, the biomimetic catalysis system is successfully demonstrated for the colorimetric glucose detection with high catalytic activity and selectivity. Also, the proposed gel-based sensor is integrated with smartphone to enable real-time and visual determination of glucose. More importantly, the gel-based sensor exhibits a high correlation with a commercial glucometer in real samples detection. These findings provide a strategy to design an efficient biomimetic catalysis system for applications in bioassays and nanobiomedicines.

Abstract:Bioorthogonal metallocatalysis has opened up a xenobiotic route to perform nonenzymatic catalytic transformations in living settings. Despite their promising features, most metals are deactivated inside cells by a myriad of reactive biomolecules, including biogenic thiols, thereby limiting the catalytic functioning of these abiotic reagents. Here we report the development of cytocompatible alloyed AuPd nanoparticles with the capacity to elicit bioorthogonal depropargylations with high efficiency in biological media. We also show that the intracellular catalytic performance of these nanoalloys is significantly enhanced by protecting them following two different encapsulation methods. Encapsulation in mesoporous silica nanorods resulted in augmented catalyst reactivity, whereas the use of a biodegradable PLGA matrix increased nanoalloy delivery across the cell membrane. The functional potential of encapsulated AuPd was demonstrated by releasing the potent chemotherapy drug paclitaxel inside cancer cells. Nanoalloy encapsulation provides a novel methodology to develop nanoreactors capable of mediating new-to-life reactions in cells.

Abstract:The development of biotechnology has made it possible to easily design and produce enzyme as needed,providing a powerful tool for the wide application of enzymatic catalysis in the fields of pharmaceutical production,bioanalysis,food manufacturing and fine chemicals industry.Enzymes with abundant catalytic functions and high catalytic activity and stereo-selectivity in the preparation of chiral compounds in the chemical industry are expected to improve the purity of the products,shorten the production process and simplify the separation of products.The cells which containing multiple enzymes inside have the ability of accurate and highly efficient synthesis.The multi-enzyme complex in cells has the characteristics of nano-scale and multi-level construction.The nano-proximity effect,compartmentalization effect and confinement effect are the important basis of efficient enzymatic catalysis in cells.However,the direct use of cells as catalysts in industry may cause a high cost,complex interference within the multiple enzymatic reactions,limited mass transfer within cell walls,difficulty in product separation due to the emulsion effect of cells and other shortcomings.On the other hand,with the rapid progress of nanotechnology,especially nanocatalysis in recent years,inorganic nanocatalysts can catalyze abundant non-natural reactions very efficiently,and can easily realize the physical and chemical processes of electron transfer and photo-electric conversion.The combination of biocatalysts and inorganic nanocatalysts opens new possibilities to mimic the complex catalysis processes in nature,and also provides a new way for the efficient utilization of energy and matter in biochemical manufacturing.It is an emerging and promising topic in the field of catalysis and is expected to provide new catalytic technologies for the green and sustainable chemical industry.Inspired by the intracellular multi-level organization and nano-effects of biocatalytic machines of cells and based on the concept of "Simply the complexity of cells,and take the essence of cells",we aim at extracting the key building blocks of biocatalytic machines of cells,replacing the limitations of biocatalytic processes by chemical catalysis,and then constructing simple,highly efficient and stable biocatalyst and nanocatalyst integrated systems.The biocatalyst and nanocatalyst integrated systems are expected to reproduce and even beyond the catalytic capability of cells,which would provide an innovative way to produce chemicals compared to traditional industrial biocatalysis.The project is based on the needs of sustainable development of chemical industry of China and focuses on the following major scientific issues:(1) the role of intracellular nano-effects in biocatalyst and nanocatalyst integrated systems,(2) the self-assembly principle,the adaptation mechanism and the realization strategy of industrial application of such biocatalyst and nanocatalyst integrated systems.Based on the research into the above key scientific problems,the project also targets to the following key technological challenges:(1) the massive preparation of biocatalyst and nanocatalyst integrated systems,(2) the large-scale catalytic process and reactor design using biocatalyst and nanocatalyst integrated systems as catalysts.
A three-tiered colloidosomal microreactor for continuous flow catalysis

Abstract:Abstract Integrative colloidosomes with hierarchical structure and advanced function may serve as biomimetic microreactors to carry out catalytic reactions by compartmentalizing biological species within semipermeable membranes. Despite of recent progress in colloidosome design, integration of biological and inorganic components into tiered structures to tackle the remaining challenges of biocatalysis is highly demanded. Here, we report a rational design of three-tiered colloidosomes via the Pickering emulsion process. The microreactor consists of crosslinked amphiphilic silica-polymer hybrid nanoparticles as the semipermeable shell, an enzyme-incorporated catalytic sub-layer, and a partially-silicified adsorptive lumen. By leveraging confinement and enrichment effect, we demonstrate the acceleration of lipase-catalyzed ester hydrolysis within the microcompartment of organic-inorganic hybrid colloidosomes. The catalytic colloidosomes are further assembled into a closely packed column for enzymatic reactions in a continuous flow format with enhanced reaction rates. The three-tiered colloidosomes provide a reliable platform to integrate functional building blocks into a biomimetic compartmentalized microreactor with spatially controlled organization and high-performance functions.

A Bioinspired Photocatalysis and Electrochemiluminescence Scaffold for Simultaneous Degradation and in Situ Evaluation

Abstract:Nanostructured scaffolds that optimize self‐responsive ability, visible light harvesting, redox center distribution, and species shuttling porosity have been mimicked much less often, especially using organic crystalline substances. Inspired by a natural photosynthetic thylakoid membrane, a synthetic strategy with general applicability to construct a hollow organic crystalline nanosphere by orderly hybridizing imine‐linked covalent organic frameworks (COFs) with polymeric hollow g‐C3N4 nanosphere is proposed. As a proof‐of‐concept application, in a homemade microreaction cell, these hollow organic nanospheres are shown to function as robust scaffolds, heterojunction shells, and light‐harvesting antennas, enabling excellent visible‐light‐driven photocatalysis for the almost complete degradation of tetracycline, a gravely residual antibiotic worldwide. Significantly, a new electrochemiluminescence (ECL) technology derived from biomimetic self‐responsiveness is developed for in situ evaluation of the photodegradation process, accessing kinetics and other relevant parameters at an extremely low‐abundance target, which remains challenging to date, albeit highly desirable. This work provides a general synthetic strategy for obtaining COFs‐derived hollow crystalline organic nanospheres for promising photocatalysis applications and offers a new perspective on the development of ECL technology for simultaneous analysis and treatment of actual sewage.
Pickering-Droplet-Derived MOF Microreactors for Continuous-Flow Biocatalysis with Size Selectivity

Abstract:Enzymatic microarchitectures with spatially controlled reactivity, engineered molecular sieving ability, favorable interior environment, and industrial productivity show great potential in synthetic protocellular systems and practical biotechnology, but their construction remains a significant challenge. Here, we proposed a Pickering emulsion interface-directed synthesis method to fabricate such a microreactor, in which a robust and defect-free MOF layer was grown around silica emulsifier stabilized droplet surfaces. The compartmentalized interior droplets can provide a biomimetic microenvironment to host free enzymes, while the outer MOF layer secludes active species from the surroundings and endows the microreactor with size-selective permeability. Impressively, the thus-designed enzymatic microreactor exhibited excellent size selectivity and long-term stability, as demonstrated by a 1000 h continuous-flow reaction, while affording completely equal enantioselectivities to the free enzyme counterpart. Moreover, the catalytic efficiency of such enzymatic microreactors was conveniently regulated through engineering of the type or thickness of the outer MOF layer or interior environments for the enzymes, highlighting their superior customized specialties.
