Abstract:Significance: Functional near-infrared spectroscopy (fNIRS) presents an opportunity to study human brains in everyday activities and environments. However, achieving robust measurements under such dynamic condition remains a significant challenge. Aim: The modular optical brain imaging (MOBI) system is designed to enhance optode-to-scalp coupling and provide real-time probe 3-D shape estimation to improve the use of fNIRS in everyday conditions. Approach: The MOBI system utilizes a bendable and lightweight modular circuit-board design to enhance probe conformity to head surfaces and comfort for long-term wearability. Combined with automatic module connection recognition, the built-in orientation sensors on each module can be used to estimate optode 3-D positions in real-time to enable advanced tomographic data analysis and motion tracking. Results: Optical characterization of the MOBI detector reports a noise equivalence power (NEP) of 8.9 and 7.3 pW/√Hz at 735 nm and 850 nm, respectively, with a dynamic range of 88 dB. The 3-D optode shape acquisition yields an average error of 4.2 mm across 25 optodes in a phantom test compared to positions acquired from a digitizer. Results for initial in vivo validations, including a cuff occlusion and a finger-tapping test, are also provided. Conclusions: To the best of our knowledge, the MOBI system is the first modular fNIRS system featuring fully flexible circuit boards. The self-organizing module sensor network and automatic 3-D optode position acquisition, combined with lightweight modules (18 g/module) and ergonomic designs, would greatly aid emerging explorations of brain function in naturalistic settings.

Abstract:Portable neuroimaging technologies can be employed for long-term monitoring of neurophysiological and neuropathological states. Functional Near-Infrared Spectroscopy (fNIRS) and Electroencephalography (EEG) are highly suited for such a purpose. Their multimodal integration allows the evaluation of hemodynamic and electrical brain activity together with neurovascular coupling. An innovative fNIRS-EEG system is here presented. The system integrated a novel continuous-wave fNIRS component and a modified commercial EEG device. fNIRS probing relied on fiberless technology based on light emitting diodes and silicon photomultipliers (SiPMs). SiPMs are sensitive semiconductor detectors, whose large detection area maximizes photon harvesting from the scalp and overcomes limitations of fiberless technology. To optimize the signal-to-noise ratio and avoid fNIRS-EEG interference, a digital lock-in was implemented for fNIRS signal acquisition. A benchtop characterization of the fNIRS component showed its high performances with a noise equivalent power below 1 pW. Moreover, the fNIRS-EEG device was tested in vivo during tasks stimulating visual, motor and pre-frontal cortices. Finally, the capabilities to perform ecological recordings were assessed in clinical settings on one Alzheimer’s Disease patient during long-lasting cognitive tests. The system can pave the way to portable technologies for accurate evaluation of multimodal brain activity, allowing their extensive employment in ecological environments and clinical practice.

Abstract:Functional near-infrared spectroscopy (fNIRS) technology has been steadily advancing since the first measurements of human brain activity over 30 years ago. Initially, efforts were focused on increasing the channel count of fNIRS systems and then to moving from sparse to high density arrays of sources and detectors, enhancing spatial resolution through overlapping measurements. Over the last ten years, there have been rapid developments in wearable fNIRS systems that place the light sources and detectors on the head as opposed to the original approach of using fiber optics to deliver the light between the hardware and the head. The miniaturization of the electronics and increased computational power continues to permit impressive advances in wearable fNIRS systems. Here we detail our design for a wearable fNIRS system that covers the whole head of an adult human with a high-density array of 56 sources and up to 192 detectors. We provide characterization of the system showing that its performance is among the best in published systems. Additionally, we provide demonstrative images of brain activation during a ball squeezing task. We have released the hardware design to the public, with the hope that the community will build upon our foundational work and drive further advancements.

Abstract:Significance: We present a fiberless, portable, and modular continuous wave-functional near-infrared spectroscopy system, Spotlight, consisting of multiple palm-sized modules-each containing high-density light-emitting diode and silicon photomultiplier detector arrays embedded in a flexible membrane that facilitates optode coupling to scalp curvature. Aim: Spotlight's goal is to be a more portable, accessible, and powerful functional near-infrared spectroscopy (fNIRS) device for neuroscience and brain-computer interface (BCI) applications. We hope that the Spotlight designs we share here can spur more advances in fNIRS technology and better enable future non-invasive neuroscience and BCI research. Approach: We report sensor characteristics in system validation on phantoms and motor cortical hemodynamic responses in a human finger-tapping experiment, where subjects wore custom 3D-printed caps with two sensor modules. Results: The task conditions can be decoded offline with a median accuracy of 69.6%, reaching 94.7% for the best subject, and at a comparable accuracy in real time for a subset of subjects. We quantified how well the custom caps fitted to each subject and observed that better fit leads to more observed task-dependent hemodynamic response and better decoding accuracy. Conclusions: The advances presented here should serve to make fNIRS more accessible for BCI applications.

Abstract:The interface program shower to the EGS Monte Carlo electromagnetic cascade shower simulation code system was written to facilitate the definition of complicated target and shielding geometries and to simplify the handling of input and output of data. The geometry is defined by a series of namelist commands in an input file. The input and output beam data files follow the SDDS (self-describing data set) protocol, which makes the files compatible with other physics codes that follow the same protocol. For instance, one can use the results of the cascade shower simulation as the input data for an accelerator tracking code. The shower code has also been used to calculate the bremsstrahlung component of radiation doses for possible beam loss scenarios at the Advanced Photon Source (APS) at Argonne National Laboratory.

Abstract:In physiological activities, the brain's electroencephalogram (EEG) signal and chemical concentration change are crucial for diagnosing and treating neurological disorders. Despite the advantages of flexible neural probes, such as their flexibility and biocompatibility, it remains a challenge to achieve in situ monitoring of electrophysiological and chemical signals on a small scale simultaneously. This study developed a new method to construct an efficient dual-sided multimodal integrated flexible neural probe, which combines a density electrode array for EEG recordings and an electrochemical sensor for detecting lactic acid. The EEG electrode array includes a 6-channel recording electrode array with each electrode 30 x 50 mu m in size, and the lactic acid sensor with overall contact is approximately 100 mu m wide. The EEG electrodes have an average impedance of 2.57 k Omega at 1 kHz and remained stable after immersing in NS (normal saline) for 3 months. The lactic acid sensor showed a sensitivity of 52.8 nA mM-1. The in vivo experiments demonstrated that the probe can reliably monitor electrophysiological signals. The probe is able to be implanted into the desired site with the help of a guide port. This flexible neural probe can provide more comprehensive insights into brain activity in the field of neuroscience and clinical practices.

Abstract:A brain–computer interface (BCI) allows users to control external devices through brain activity. Portable neuroimaging techniques, such as near-infrared (NIR) imaging, are suitable for this goal. NIR imaging has been used to measure rapid changes in brain optical properties associated with neuronal activation, namely fast optical signals (FOS) with good spatiotemporal resolution. However, FOS have a low signal-to-noise ratio, limiting their BCI application. Here FOS were acquired with a frequency-domain optical system from the visual cortex during visual stimulation consisting of a rotating checkerboard wedge, flickering at 5 Hz. We used measures of photon count (Direct Current, DC light intensity) and time of flight (phase) at two NIR wavelengths (690 nm and 830 nm) combined with a machine learning approach for fast estimation of visual-field quadrant stimulation. The input features of a cross-validated support vector machine classifier were computed as the average modulus of the wavelet coherence between each channel and the average response among all channels in 512 ms time windows. An above chance performance was obtained when differentiating visual stimulation quadrants (left vs. right or top vs. bottom) with the best classification accuracy of ~63% (information transfer rate of ~6 bits/min) when classifying the superior and inferior stimulation quadrants using DC at 830 nm. The method is the first attempt to provide generalizable retinotopy classification relying on FOS, paving the way for the use of FOS in real-time BCI.

Abstract:Near-Infrared Spectroscopy (NIRS) and diffuse optical imaging (DOI) are increasingly used to detect hemodynamic changes in the cerebral cortex induced by brain activity. Until recently, the small number of optodes in NIRS instruments has hampered measurement of optical signals from diverse brain regions. Our new DOI system has 32 detectors and 32 sources; by arranging them in a specific pattern, we can cover most of the adult head. With the increased number of optodes, we can collect optical data from prefrontal, sensorimotor, and visual cortices in both hemispheres simultaneously. We describe the system and report system characterization measurements on phantoms as well as on human subjects at rest and during visual, motor, and cognitive stimulation. Taking advantage of the system's larger number of sources and detectors, we explored the spatiotemporal patterns of physiological signals during rest. These physiological signals, arising from cardiac, respiratory, and blood-pressure modulations, interfere with measurement of the hemodynamic response to brain stimulation. Whole-head optical measurements, in addition to providing maps of multiple brain regions' responses to brain activation, will enable better understandings of the physiological signals, ultimately leading to better signal processing algorithms to distinguish physiological signal clutter from brain activation signals.

Brain-computer interface using a simplified functional near-infrared spectroscopy system

Abstract:A brain-computer interface (BCI) is a device that allows a user to communicate with external devices through thought processes alone. A novel signal acquisition tool for BCIs is near-infrared spectroscopy (NIRS), an optical technique to measure localized cortical brain activity. The benefits of using this non-invasive modality are safety, portability and accessibility. A number of commercial multi-channel NIRS system are available; however we have developed a straightforward custom-built system to investigate the functionality of a fNIRS-BCI system. This work describes the construction of the device, the principles of operation and the implementation of a fNIRS-BCI application, 'Mindswitch' that harnesses motor imagery for control. Analysis is performed online and feedback of performance is presented to the user. Mindswitch presents a basic 'on/off' switching option to the user, where selection of either state takes 1 min. Initial results show that fNIRS can support simple BCI functionality and shows much potential. Although performance may be currently inferior to many EEG systems, there is much scope for development particularly with more sophisticated signal processing and classification techniques. We hope that by presenting fNIRS as an accessible and affordable option, a new avenue of exploration will open within the BCI research community and stimulate further research in fNIRS-BCIs.

Abstract:Similar to functional magnetic resonance imaging (fMRI), functional near-infrared spectroscopy (fNIRS) detects the changes of hemoglobin species inside the brain, but via differences in optical absorption. Within the near-infrared spectrum, light can penetrate biological tissues and be absorbed by chromophores, such as oxyhemoglobin and deoxyhemoglobin. What makes fNIRS more advantageous is its portability and potential for long-term monitoring. This paper reviews the basic mechanisms of fNIRS and its current clinical applications, the limitations toward more widespread clinical usage of fNIRS, and current efforts to improve the temporal and spatial resolution of fNIRS toward robust clinical usage within subjects. Oligochannel fNIRS is adequate for estimating global cerebral function and it has become an important tool in the critical care setting for evaluating cerebral oxygenation and autoregulation in patients with stroke and traumatic brain injury. When it comes to a more sophisticated utilization, spatial and temporal resolution becomes critical. Multichannel NIRS has improved the spatial resolution of fNIRS for brain mapping in certain task modalities, such as language mapping. However, averaging and group analysis are currently required, limiting its clinical use for monitoring and real-time event detection in individual subjects. Advances in signal processing have moved fNIRS toward individual clinical use for detecting certain types of seizures, assessing autonomic function and cortical spreading depression. However, its lack of accuracy and precision has been the major obstacle toward more sophisticated clinical use of fNIRS. The use of high-density whole head optode arrays, precise sensor locations relative to the head, anatomical co-registration, short-distance channels, and multi-dimensional signal processing can be combined to improve the sensitivity of fNIRS and increase its use as a wide-spread clinical tool for the robust assessment of brain function.

Abstract:Frequency-domain near-infrared spectroscopy (FD-NIRS) provides quantitative noninvasive measurements of tissue optical absorption and scattering, as well as a safe and accurate method for characterizing tissue composition and metabolism. However, the poor scalability and high complexity of most FD-NIRS systems assembled to date have contributed to its limited clinical impact. To address these shortcomings, we present a scalable, digital-based FD-NIRS platform capable of measuring optical properties and tissue chromophore concentrations in real-time. The system provides single-channel FD-NIRS amplitude/phase, optical property, and chromophore data at a maximum display rate of 36.6 kHz, 17.9 kHz, and 10.2 kHz, respectively, and can be scaled to multiple channels as well as integrated into a handheld format. The entire system is enabled by several innovations including an ultra-high-speed k-nearest neighbor lookup table method (maximum of 250,000 inversions/s for a large 2500x700 table of absorption and reduced scattering coefficients), embedded FPGA and CPU high-speed co-processing, and high-speed data transfer (due to on-board processing). We show that our 6-wavelength, broad modulation bandwidth (1-400 MHz) system can be used to perform 2D high-density spatial mapping of optical properties and high speed quantification of hemodynamics.

Abstract:Multimodal neuroimaging plays an important role in neuroscience research. Integrated noninvasive neuroimaging modalities, such as magnetoencephalography (MEG), electroencephalography (EEG) and functional near-infrared spectroscopy (fNIRS), allow neural activity and related physiological processes in the brain to be precisely and comprehensively depicted, providing an effective and advanced platform to study brain function. Noncryogenic optically pumped magnetometer (OPM) MEG has high signal power due to its on-scalp sensor layout and enables more flexible configurations than traditional commercial superconducting MEG. Here, we integrate OPM-MEG with EEG and fNIRS to develop a multimodal neuroimaging system that can simultaneously measure brain electrophysiology and hemodynamics. We conducted a series of experiments to demonstrate the feasibility and robustness of our MEG-EEG-fNIRS acquisition system. The complementary neural and physiological signals simultaneously collected by our multimodal imaging system provide opportunities for a wide range of potential applications in neurovascular coupling, wearable neuroimaging, hyperscanning and brain-computer interfaces.

Abstract:Functional brain imaging is considered a powerful and practical solution for understanding the brain and neurological diseases. While EEG is an established method for non-invasive electrical activity, electrical-impedance tomography (EIT) and near-infrared spectroscopy (NIRS) can additionally measure impedance changes and hemodynamic processes. To facilitate long-term multi-channel brain imaging in a wearable form factor without cabling overhead, there is a need for low-power local amplifiers [1] to support all these modalities. The main principle of optical hemodynamic measurements is to send light pulses into the tissue and measure the reflected light, which is modulated by the oxygen levels in the blood (Fig. 17.8.1). State-of-the-art NIRS ICs typically consume a few mW, primarily for the LEDs to meet the required light sensitivity at the photodiodes (PDs). Silicon photomultipliers (SiPMs) are promising alternatives because they have excellent low-light detection capabilities, speed of response and higher detection efficiency in both visible and near infrared range [2]. Hence, SiPMs allow deeper brain sensing depth and the possibility to sample consistent cerebral regions with larger inter-optode distance. This benefit would significantly reduce the number of NIRS channels and the associated power for a wearable NIRS device. Although SiPMs require a higher bias voltage (∼30V) than PDs, they achieve similar NIRS responses with a few hundred times less LED current. This results in a low-power NIRS ASIC and an overall power-efficient system. Existing optical sensing ICs are not suitable for a SiPM because of its large and variable output current. Trimming-based calibration methods [3] suffer from drift over time. Auto-zeroing by swapping an integrator capacitor [4][5] compensates ambient light at the cost of the integrator's headroom. Apart from ambient light, the dynamic range (DR) of the amplifier is also limited by a large NIRS signal, leading to a power-hungry readout.

Abstract:Significance: Current approaches to stimulating and recording from the brain have combined electrical or optogenetic stimulation with recording approaches, such as two-photon, electro-physiology (EP), and optical intrinsic signal imaging (OISI). However, we lack a label-free, all-optical approach with high spatial and temporal resolution.Aim: To develop a label-free, all-optical method that simultaneously manipulates and images brain function using pulsed near-infrared light (INS) and functional optical coherence tomography (fOCT), respectively.Approach: We built a coregistered INS, fOCT, and OISI system. OISI and EP recordings were employed to validate the fOCT signals.Results: The fOCT signal was reliable and regional, and the area of fOCT signal corresponded with the INS-activated region. The fOCT signal was in synchrony with the INS onset time with a delay of similar to 30 ms. The magnitude of fOCT signal exhibited a linear correlation with the INS radiant exposure. The significant correlation between the fOCT signal and INS was further supported by OISI and EP recordings.Conclusions: The proposed fiber-based, all-optical INS-fOCT method allows simultaneous stimulation and mapping without the risk of interchannel cross-talk and the requirement of contrast injection and viral transfection and offers a deep penetration depth and high resolution. Abstract:Abundant study on the hemodynamic response of a brain have brought quite a few advances in technologies of measuring it. The most benefitted is the functional near infrared spectroscope (fNIRS). A variety of devices have been developed for different applications. Because portable fNIRS systems were more competent to measure responses either of special subjects or in natural environment, several kinds of portable fNIRS systems have been reported. However, they all required a computer for receiving data. The extra computer increases the cost of a fNIRS system. What's more noticeable is the space required to locate the computer even for a portable system. It will discount the portability of the fNIRS system. So we designed a self-contained eight channel fNIRS system, which does not demand a computer to receive data and display data in a monitor. Instead, the system is centered by an ARM core CPU, which takes charge in organizing data and saving data, and then displays data on a touch screen. The system has also been validated by experiments on phantoms and on subjects in tasks.

However, the feasibility of utilizing fNIRS to detect LOC and FFA activity in adults remains to be validated as the depth of these regions may exceed the detection limit of fNIRS.Aim: This study aims to investigate the feasibility of using fNIRS to measure hemodynamic responses in the ventral visual pathway, specifically in the LOC and FFA, in adults.Approach: We recorded the hemodynamic activities of the LOC and FFA regions in 35 subjects using a portable eight-channel fNIRS instrument. A standard one-back object and face recognition task was employed to elicit selective brain responses in the LOC and FFA regions. The placement of fNIRS optodes for LOC and FFA detection was guided by our group's transcranial brain atlas (TBA).Results: Our findings revealed selective activation of the LOC target channel (CH2) in response to objects, whereas the FFA target channel (CH7) did not exhibit selective activation in response to faces.Conclusions: Our findings indicate that, although fNIRS detection has limitations in capturing FFA activity, the LOC region emerges as a viable target for fNIRS-based detection. Furthermore, our results advocate for the adoption of the TBA-based method for setting the LOC target channel, offering a promising solution for optrode placement. This feasibility study stands as the inaugural validation of fNIRS for detecting cortical activity in the ventral visual pathway, underscoring its ecological validity. We suggest that our findings establish a pivotal technical groundwork for prospective real-life applications of fNIRS-based research.

Abstract:Functional near-infrared spectroscopy (fNIRS) is an optical brain imaging technique that maps brain function in terms of changes in oxygenated and deoxygenated hemoglobin concentration (hemodynamic response) on the cortical surface of the brain. Wireless wearable fNIRS imaging is always preferable to study a brain in an unconstrained environment. However, an LED and photodetector based miniaturized fNIRS system suffers from several technical challenges to provide high fidelity fNIRS signal. This paper reports the design and development of an internet-of-things (IoT)-based wireless fNIRS headband to image hemodynamic response of the prefrontal cortex of the brain in naturalistic settings. The headband has four dual-wavelength LEDs and four silicon photodiode detectors arranged to provide 10 usable fNIRS channels. The designed IoT-based controller has dedicated digital and analog circuit and is operated by an embedded computing algorithm to facilitate source modulation, lock-in detection, onboard calibration, and basic signal processing to improve SNR of the optical measurements at 9 Hz. A graphical user interface (GUI) software on the host computer helps the users to visualize and record the fNIRS signal wirelessly. The experimental validation of the system was performed by imaging prefrontal cortex of the brain while the participants (n=16) performed cognitive tasks that induced four different levels of mental workload. The experimental results demonstrated that the fNIRS headband was capable of imaging hemodynamic activity of the brain. The experimental results also showed on average, four different stimulus-evoked hemodynamic responses due to the four difficulty levels of the tasks

Abstract:<p>Optical fibers made of polymeric materials possess high flexibility that can potentially integrate with flexible electronic devices to realize complex functions in biology and neurology. Here, a multichannel flexible device based on four individually addressable optical fibers transfer‐printed with flexible electronic components and controlled by a wireless circuit is developed. The resulting device offers excellent mechanics that is compatible with soft and curvilinear tissues, and excellent diversity through switching different light sources. The combined configuration of optical fibers and flexible electronics allows optical stimulation in selective wavelengths guided by the optical fibers, while conducting distributed, high‐throughput biopotential sensing using the flexible microelectrode arrays. The device has been demonstrated in vivo with rats through optical stimulation and simultaneously monitoring of spontaneous/evoked spike signals and local field potentials using 32 microelectrodes in four brain regions. Biocompatibility of the device has been characterized by behavior and immunohistochemistry studies, demonstrating potential applications of the device in long‐term animal studies. The techniques to integrate flexible electronics with optical fibers may inspire the development of more flexible optoelectronic devices for sophisticated applications in biomedicine and biology.</p>

Abstract:Abstract Understanding the cytoarchitecture and wiring of the brain requires improved methods to record and stimulate large groups of neurons with cellular specificity. This requires miniaturized neural interfaces that integrate into brain tissue without altering its properties. Existing neural interface technologies have been shown to provide high-resolution electrophysiological recording with high signal-to-noise ratio. However, with single implantation, the physical properties of these devices limit their access to one, small brain region. To overcome this limitation, we developed a platform that provides three-dimensional coverage of brain tissue through multisite multifunctional fiber-based neural probes guided in a helical scaffold. Chronic recordings from the spatially expandable fiber probes demonstrate the ability of these fiber probes capturing brain activities with a single-unit resolution for long observation times. Furthermore, using Thy1-ChR2-YFP mice we demonstrate the application of our probes in simultaneous recording and optical/chemical modulation of brain activities across distant regions. Similarly, varying electrographic brain activities from different brain regions were detected by our customizable probes in a mouse model of epilepsy, suggesting the potential of using these probes for the investigation of brain disorders such as epilepsy. Ultimately, this technique enables three-dimensional manipulation and mapping of brain activities across distant regions in the deep brain with minimal tissue damage, which can bring new insights for deciphering complex brain functions and dynamics in the near future.

Abstract:Assessing the neural correlates of motor and cognitive processes under naturalistic experimentation is challenging due to the movement constraints of traditional brain imaging technologies. The recent advent of portable technologies that are less sensitive to motion artifacts such as Functional Near Infrared Spectroscopy (fNIRS) have been made possible the study of brain function in freely-moving participants. In this paper, we describe a series of proof-of-concept experiments examining the potential of fNIRS in assessing the neural correlates of cognitive and motor processes in unconstrained environments. We show illustrative applications for practicing a sport (i.e., table tennis), playing a musical instrument (i.e., piano and violin) alone or in duo and performing daily activities for many hours (i.e., continuous monitoring). Our results expand upon previous research on the feasibility and robustness of fNIRS to monitor brain hemodynamic changes in different real life settings. We believe that these preliminary results showing the flexibility and robustness of fNIRS measurements may contribute by inspiring future work in the field of applied neuroscience.
