Developing photo-activable ruthenium (II) complexes for PDT: Synthesis, characterization, photophysical and biological studies

Abstract:The easy tunable photophysical properties of Ru(II) polypyridyl complexes, together with their water solubility, photochemical stability and high biocompatibility make them suitable as agents for photodynamic therapy (PDT). The development of complexes containing new ligands is a step forward in the improvement and application of these compounds as photosensitizer and photocytotoxic agents. We report the synthesis of a set of Ru(II)-polypyridyl complexes with different electronic properties and delocalized π systems: one homoleptic complex [Ru II (dpbpy) 3 ](PF 6 ) 2 ( 1 ), and three heteroleptic complexes [Ru II (dpbpy) 2 (phen)](PF 6 ) 2 ( 2 ), [Ru II (dpbpy)(phen) 2 ](PF 6 ) 2 ( 3 ) and [Ru II (dpbpy) 2 (CN–Me)](PF 6 ) 2 ( 4 ). All of them contain 4,4′-diphenyl-2,2′-bipyridine ( dpbpy ) and 1,10-phenantroline ( phen ) or N-methyl-N′-2-pyridilimidazolium (CN–Me) ligands. The complexes have been characterized by spectroscopic, structural and electrochemical methods. UV–vis absorption in solution show the red shift for the metal to ligand charge transfer (MLCT) transitions after changing phen by dpbpy and carbene ligands. The photoluminescence emission spectra of 1 – 4 supported by theoretical calculations using time-dependent density-functional theory (TDDFT) suggest that the lowest energy excited state is mainly 3 MLCT. Complex 2 exhibits the highest phosphorescence emission with a quantum yield (Φ P ) in CH 2 Cl 2 of 44.1 %, followed by complex 1 (Φ P = 40.0 %), whereas complex 4 shows the lowest quantum efficiency (Φ P = 16.0 %), what suggest that the introduction of the CN–Me carbene ligand produces a significant quenching of the phosphorescence. Phosphorescence triplet state lifetimes between 0.96 and 1.74 μs were shown for 1 – 4 in degassed CH 2 Cl 2 . All complexes are proven to be efficient singlet oxygen photosensitizers. The trend in the redox potentials becomes more positive by increasing the number of phen ligands in the complexes, in full agreement with the HOMO's energy level and the blue shift of the MLCT transitions in the absorption spectra. Complexes internalization was analysed in tumorigenic mammary epithelial SKBR-3 cells, being complexes 1 and 2 the most well internalized. The effect of the photodynamic treatment using light-activated complexes 1 and 2 for 10 min demonstrated to increase cell death, being the homoleptic complex 1 an outstanding candidate as potential theranostic agent for bioimaging and PDT.

Abstract:In this work we disclose a new family of biscyclometallated Ir( III ) complexes of the general formula [Ir(C^N) 2 (N^N)]Cl ( IrL1–IrL5 ), where HC^N is 1-phenyl-β-carboline and N^N ligands ( L1–L5 ) are different diimine ligands that differ from each other in the number of aromatic rings fused to the bipyridine scaffold. The photophysical properties of IrL1–IrL5 were thoroughly studied, and theoretical calculations were performed for a deeper comprehension of the respective variations along the series. All complexes exhibited high photostability under blue light irradiation. An increase in the number of aromatic rings led to a reduction in the HOMO–LUMO band gap causing a red-shift in the absorbance bands. Although all the complexes generated singlet oxygen ( 1 O 2 ) in aerated aqueous solutions through a photocatalytic process, IrL5 was by far the most efficient photosensitizer. Consequently, IrL5 was highly active in the photocatalytic oxidation of NADH. The formation of aggregates in DMSO at a high concentration (25 mM) was confirmed using different techniques, but was proved to be negligible in the concentration range of biological experiments. Moreover, ICP-MS studies proved that the cellular uptake of IrL2 and IrL3 is much better relative to that of IrL1 , IrL4 and IrL5 . The antiproliferative activity of IrL1–IrL5 was investigated in the dark and under blue light irradiation against different cancer cell lines. Complexes IrL1–IrL4 were found to be cytotoxic under dark conditions, while IrL5 turned out to be weakly cytotoxic. Despite the low cellular uptake of IrL5 , this derivative exhibited a high increase of cytotoxicity upon blue light irradiation resulting in photocytotoxicity indexes (PI) up to 38. IrL1–IrL4 showed lower photocytotoxicity indexes ranging from 1.3 to 17.0. Haemolytic experiments corroborated the compatibility of our complexes with red blood cells. Confocal microscopy studies proved their accumulation in mitochondria, leading to mitochondrial membrane depolarization, and ruled out their localization in lysosomes. Overall, the mitochondria-targeted activity of IrL5 , which inhibits considerably the viability of cancer cells upon blue light irradiation, allows us to outline this PS as a new alternative to traditional chemotherapeutic agents.

Abstract:Design and preparation of photosensitizers (PSs) play an important role in photodynamic therapy (PDT). PDT mainly relies on the production of toxic reactive oxygen species (ROS) of the PSs. Conventional fluorophores, however, often suffer from aggregation caused quenching (ACQ), which limits the potential of PSs as fluorescent imaging agents. Molecules with aggregation-induced emission (AIE) properties maintain high fluorescence and dispersity in aqueous solutions, overcoming the ACQ effect. Ruthenium (II)-based AIE compounds are highly biocompatible molecules and can be used for response cell imaging. In the current study, two novel Ru(II)-based AIE compounds with main ligands 1,3-di(2H-tetrazol-5-yl)benzene (Hphbtz) by changing auxiliary ligand 2,2′-bipyridine (bipy) and 1,10-phenanthroline (phen) have been successfully synthesized and characterized, [Ru(Hphbtz)(bipy)2][PF6] (1) and [Ru(Hphbtz)(phen)2][PF6] (2). The NPs show strong intra-cellular fluorescence and also simultaneously exhibited potent cytotoxic activity. These compounds can self-assemble to form nanoparticles (NPs) by nanoprecipitation. The compounds are found to exhibit a high AIE property with emission maxima at 353 nm and 380 nm, respectively. And the compounds have the low IC50 (half maximal inhibitory concentration) of only 15 μg/mL (1.94 μM) and 13 μg/mL (1.58 μM) on HeLa cells, respectively. Meanwhile, negligible dark toxicity has been also observed for these NPs. The results show that [Ru(Hphbtz)(bipy)2][PF6] (1) and [Ru(Hphbtz)(phen)2][PF6] (2) NPs can inhibit cell proliferation in vitro, and may be potential candidates for photodynamic therapy.

Abstract:Ru(II) complexes have attracted attention as photosensitizers for their promising photodynamic properties. Herein, novel tris-bipyridine based Ru(II) complexes ( 6a-e ) were synthesized by introducing saturated heterocycles to improve photodynamic properties and lipid-water partition coefficients. Among them, 6d demonstrated significant phototoxicity towards three cancer cells, with IC 50 values of 5.66−7.17 μM, exceeding values in dark (IC 50 s > 100 μM). Under hypoxic conditions, 6d maintained excellent photodynamic activity in A549 cells, with PI values exceeding 24, highlighting its potential for highly effective type-I/-II photodynamic therapy by inducing ROS generation, oxidative stress, and mitochondrial damage. Additionally, it induced ferroptosis and immunogenic cell death of A549 cells by regulating the expression of relevant markers. Finally, 6d remarkably inhibited the growth of A549 transplanted tumor growth by 95.4%. This Ru(II) complex shows great potential for cancer treatment with its potent photodynamic activity and diverse mechanisms of tumor cell death.

Abstract:A new generation series of cyclometalated Ru( II ) polypyridyl complexes of the type [Ru(C^N)(N^N) 2 ] + , Ru1–Ru4 , were rationally designed and synthesized, where N^N = 2,2′-bipyridine (bpy) and dipyrido[3,2- d :2′,3′- f ]quinoxaline (dpq) and C^N = deprotonated methyl 1-butyl-2-aryl-benzimidazolecarboxylate with p -CF 3 C 6 H 4 or p -Me 2 NC 6 H 4 substituents in the R 3 position of the phenyl ring. The photophysical properties of Ru1–Ru4 revealed absorption maxima around 560 nm with an absorption up to 700 nm. The new Ru complexes were able to generate singlet oxygen ( 1 O 2 ) upon green light irradiation in acetonitrile, with complexes containing the CF 3 group, Ru1 and Ru3 , being the best performers. Furthermore, Ru1 and Ru3 were also able to photogenerate hydroxyl radicals OH ̇. By having PSs capable of undergoing both type I and type II mechanisms, a broader range of cytotoxic effects is achieved. Ru1–Ru4 accumulated in membrane-rich compartments, including the cytoplasmic membrane, mitochondria, and endoplasmic reticulum in HeLa cells. Upon irradiation of Ru1 with green light, all these compartments were damaged in treated cells. Based on in vitro experiments, we deduced that the compound Ru1 under irradiation has the capability to disrupt phospholipid membranes directly. Additionally, differential scanning calorimetry of living cells also indicated damage of cytoplasmic/membrane proteins, ultimately leading to cell death via oncosis.

Synthetic Ease and Exceptional In Vivo Performance of Pyrazole-based Cyclometallated Iridium Complexes

Abstract:Two luminescent cyclometallated Ir III complexes of the type [Ir(C^N) 2 (N^N)] x (X = 0 ( 1 ), and X = +1 ( 2 )) were prepared using methyl 2-phenylquinoline-4-carboxylate as C^N ligands and either a deprotonated or protonated 3-(2-pyridyl)pyrazole as the N^N chelate. The synthesis followed a well-established and straightforward procedure. Photophysical analysis unveiled their remarkable properties, featuring phosphorescent red and orange emissions attributed to 3 MLCT and 3 LLCT transitions, with high emissive quantum yields in degassed DMSO solutions. Importantly, these complexes exhibited dual functionality as potent chemotherapeutic agents and photosensitisers, with their effectiveness tailored to specific cancer cell lines. Phototherapeutic treatment was particularly effective against lung cancer A549 cells, while a chemotherapeutic approach yielded superior results against breast cancer 4T1-luc2 cells. Both complexes primarily targeted lysosomes, leading to cell death through apoptotic pathways, with IC50 values in the nanomolar range. Importantly, they demonstrated no cytotoxicity against lymphocytes, mimicking the behaviour observed in healthy cells. Encouragingly, 1 and 2 exhibited minimal in vivo toxicity. The most striking finding was the exceptional chemotherapeutic efficacy of complexes 1 and 2 against 4T1-luc2 cells in BALB/c mice. These complexes surpassed the performance of the clinically employed 5-fluorouracil, especially at early tumor stages, significantly retarding 4T1-luc2 proliferation. Further investigations at vascularised and organised tumor stages revealed that complex 1 could reduce tumor size by half compared to untreated mice, which was also confirmed through tumoral weight analysis. While these findings are preliminary, the outstanding performance of these complexes at early tumor stages against breast cancer 4T1-luc2 in vivo, their selective therapeutic feasibility (chemotherapy vs. PDT) tailored to specific cancerous cell lines, and their straightforward synthetic design make them highly attractive candidates for cancer treatment.

Abstract:Ru(II) complexes have been recognized as excellent anticancer candidates. However, the poor cell uptake of these complexes has been the major obstacle in improving their anticancer efficacy. Along with the development of the knowledge on the anticancer nature of the Ru(II) complexes, several strategies have been designed to increase the cellular accumulation of these complexes. Among them, the light-triggered drug uptake approach is most promising. In this study, a bioactive Ru(II)-polypyridyl complex Ru-1 is constructed via incorporating a modified phenanthroline ligand into the enlarged conjugate structure. The modulation of the ligand makes Ru-1 powerful in generating singlet oxygen and effective in photodynamic therapeutic activity. Moreover, the cell uptake is improved during this photodynamic process and induces further chemotherapeutic effect, which act synergistically to enhance its anticancer activity. The photoinduced synergistic cytotoxicity of Ru-1 towards cancer cells offers an effective way to sensitize the antitumor activity of Ru(II) complexes.
Lysosome Targeting Bis-terpyridine Ruthenium(II) Complexes: Photophysical Properties and in Vitro Photodynamic Therapy.

Abstract:Three heteroleptic bis-terpyridine ruthenium(II) complexes (Ru1-Ru3) [Ru(tpy-R1)(tpy-R2)]2+ (tpy = 2,2':6',2″-terpyridine, R1/R2 = phenyl, 4-{2-[2-(2-methoxyethoxy)ethoxy]ethoxy}phenyl, pyren-1-yl, or 4-phenyl-BODIPY (boron dipyrromethene)) were synthesized and investigated for their potential applications as photosensitizers (PSs) for photodynamic therapy. All complexes displayed broad and intense absorption band in the green spectral regions (450-600 nm), which arose from the spin-allowed charge-transfer transitions mixed with ligand-localized 1π,π* transitions. All complexes show weak green emission at 513-549 nm and/or even weaker red emission at 646-674 nm at room temperature depending on the excitation wavelength and the solvent used. Incorporating the BODIPY motif to the 4'-position of one of the tpy ligands in Ru2 and Ru3 drastically prolonged the lifetimes of the lowest triplet excited states (T1) of Ru2 and Ru3 to tens of microseconds. This promoted the singlet oxygen formation sensitized by Ru2 and Ru3 upon green light activation, which in turn induced significant photocytotoxicity toward the A549 human lung cancer cell line with an EC50 value of 1.50 μM for Ru2 and 7.41 μM for Ru3 under 0.48 J·cm-2 500 nm light irradiation. Laser confocal scanning microscopy imaging revealed that Ru2 mainly distributed to lysosomes upon cell uptake. Upon 500 nm light activation, Ru2 induced lysosomal damage and subsequent mitochondrial membrane potential decrease. The dominant cell death pathway was apoptosis. These results demonstrated the potential utilization of [Ru(tpy-R1)(tpy-R2)]2+ complexes as PSs for PDT.

Abstract:The potential use of Ru(II) complexes as photosensitizers (PSs) in photodynamic therapy (PDT) has gained significant attention. In comparison with fluorophores with aggregation-caused quenching (ACQ), fluorophores with aggregation-induced emission (AIE) characteristics exhibit sustained fluorescence and dispersibility in aqueous solutions. PSs with AIE characteristics have received much attention in recent years. Herein, we reported two novel biotin-conjugated Ru(II) polypyridyl complexes (Ru1 and Ru2) with AIE characteristics. When exposed to 460 nm (10 mW cm-2) light, Ru1 and Ru2 exhibited outstanding photostability and photocatalytic activity. Ru1 and Ru2 could efficiently generate singlet oxygen and induce pUC19 DNA photolysis when exposed to 460 nm light. Interestingly, both Ru1 and Ru2 also functioned as catalysts for NADH oxidation when exposed to 460 nm light. The presence of biotin fragments in Ru1 and Ru2 enhanced the specific uptake of these complexes by tumor cells. Both complexes showed minimal toxicity to selected cells in the dark. Nevertheless, the phototoxicity of both complexes significantly increased upon 460 nm light irradiation for 15 min. Further experiments revealed that Ru2 primarily accumulated in mitochondria and might bind to mitochondrial DNA. Under 460 nm light irradiation, Ru2 induced the generation of reactive oxygen species (ROS) and NADH depletion disrupting intracellular redox homeostasis in A549 cells, activating the mitochondrial apoptosis pathway resulting in up-regulation of apoptotic marker caspase-3, effectively damaged A549 cell DNA and arrested A549 cell cycle in the S phase. In vivo anti-tumor experiments were conducted to assess the effects of Ru2 on tumor growth in A549 tumor-bearing mice. The results showed that Ru2 effectively inhibited tumor growth under 460 nm light irradiation conditions. These findings indicate that Ru2 has great potential as a targeted photosensitizer for mitochondrial targeting imaging and photodynamic therapy of tumors.

Abstract:Liver cancer is one of the leading causes of death motivating scientists worldwide to synthesize novel therapeutics. Ru(II)-polypyridyl complexes are extensively studied for possible therapeutic and cellular applications due to their tunable coordination chemistry, structural diversity, ligand-exchange kinetics, accessible redox states, and rich photophysical or photochemical properties. Herein, we have synthesized a series of Ru(II) polypyridyl complexes [RuII(N^N) 2 (ox)] ( 1 - 3 ), where ox is oxalate (C2O42-), N^N is 1,10-phenanthroline (phen) ( 1 ), dipyrido[3,2-d:2',3'-f]quinoxaline (dpq) ( 2 ), and dipyrido[3,2,-a:2',3'-c]phenazine (dppz) ( 3 ). The oxalate (ox 2- ) was opted as a bioactive dioxo ligand to prevent hydrolysis in aqueous media, thereby increasing the stability of the Ru(II)-polypyridyl complexes in physiological media. We thoroughly characterized all the complexes using ESI-MS, FT-IR, UV-vis, and 1H NMR spectroscopy and other physicochemical methods. The complexes were stable under physiological conditions and under low-energy green LED light (λirr= 530 nm). However, the photoirradiation of complexes resulted in the efficient generation of singlet oxygen ( 1 O 2 ) as a major reactive oxygen species (ROS). The role of extended aromatic conjugation of the N^N-donor ligands in the complexes was demonstrated on their binding propensities with CT-DNA and bovine serum albumin (BSA). Both DNA intercalation and groove binding are evidenced, while tryptophan (Trp) and tyrosine (Tyr) binding preferences are revealed from the synchronous fluorescence spectral (SFS) of BSA. The cytotoxic profiling of the complexes performed on hepatocellular carcinoma cells (HepG2) in the dark and in the presence of green light indicated their dose-dependent cytotoxicity. The [Ru II (N^N)2(ox)] complexes exhibit enhanced photocytotoxicity mediated by efficient generation of cytotoxic 1 O 2 and DNA interaction. All the complexes internalize to the HepG2 liver cancer cells efficiently and localize to the cytoplasm and nucleus. The complexes exhibited potent anti-proliferative, anti-clonogenic, and anti-migratory effects on the cancer cells, suggesting their potential promise for therapeutic applications.

Abstract:One approach to reduce the side effects of chemotherapy in cancer treatment is photodynamic therapy (PDT), which allows spatiotemporal control of the cytotoxicity. We have used the strategy of coordinating π-expansive ligands to increase the excited state lifetimes of Ir(III) half-sandwich complexes in order to facilitate the generation of 1O(2). Tracking the cellular uptake and phototoxicity of Ru(ii)-polypyridyl-1,8-naphthalimide Tröger's base conjugates

Abstract:Ruthenium(ii) complexes are attracting significant research attention as a promising class of photosensitizers (PSs) in photodynamic therapy (PDT). Having previously reported the synthesis of two novel Ru(ii)-polypyridyl-1,8-naphthalimide Tröger's base compounds 1 and 2 with interesting photophysical properties, where the emission from either the Ru(ii) polypyridyl centres or the naphthalimide moieties could be used to monitor binding to nucleic acids, we sought to use these compounds to investigate further and in more detail their biological profiling, which included unravelling their mechanism of cellular uptake, cellular trafficking and cellular responses to photoexcitation. Here we demonstrate that these compounds undergo rapid time dependent uptake in HeLa cells that involved energy dependent, caveolae and lipid raft-dependent mediated endocytosis, as demonstrated by confocal imaging, and transmission and scanning electron microscopy. Following endocytosis, both compounds were shown to localise to mostly lysosomal and Golgi apparatus compartments with some accumulation in mitochondria but no localisation was found to the nucleus. Upon photoactivation, the compounds increased ROS production and induced ROS-dependent apoptotic cell death. The photo-activated compounds subsequently induced DNA damage and altered tubulin, but not actin structures, which was likely to be an indirect effect of ROS production and induced apoptosis. Furthermore, by changing the concentration of the compounds or the laser used to illuminate the cells, the mechanism of cell death could be changed from apoptosis to necrosis. This is the first detailed biological study of Ru(ii)-polypyridyl Tröger's bases and clearly suggests caveolae-dependent endocytosis is responsible for cell uptake - this may also explain the lack of nuclear uptake for these compounds and similar results observed for other Ru(ii)-polypyridyl complexes. These conjugates are potential candidates for further development as PDT agents and may also be useful in mechanistic studies on cell uptake and trafficking.
Dipyrrinato‐Iridium(III) Complexes for Application in Photodynamic Therapy and Antimicrobial Photodynamic Inactivation

Abstract:The generation of bio‐targetable photosensitisers is of utmost importance to the emerging field of photodynamic therapy and antimicrobial (photo‐)therapy. A synthetic strategy is presented in which chelating dipyrrin moieties are used to enhance the known photoactivity of iridium(III) metal complexes. Formed complexes can thus be functionalized in a facile manner with a range of targeting groups at their chemically active reaction sites. Dipyrrins with N‐ and O‐substituents afforded (dipy)iridium(III) complexes via complexation with the respective Cp*‐iridium(III) and ppy‐iridium(III) precursors (dipy = dipyrrinato, Cp* = pentamethyl‐η 5 ‐cyclopentadienyl, ppy = 2‐phenylpyridyl). Similarly, electron‐deficient Ir III (dipy)(ppy) 2 complexes could be used for post‐ functionalization, forming alkenyl, alkynyl and glyco‐appended iridium(III) complexes. The phototoxic activity of these complexes has been assessed in cellular and bacterial assays with and without light; the Ir III (Cl)(Cp*)(dipy) complexes and the glyco‐substituted iridium(III) complexes showing particular promise as photomedicine candidates. Representative crystal structures of the complexes are also presented.

Abstract:Photodynamic therapy (PDT) is a promising cancer treatment approach with the advantages of low toxicity and noninvasive characteristics. In this study, a series of metalloporphyrin–indomethacin conjugates tethered with poly(ethylene glycol) (PEG) chains were prepared and characterized. The singlet oxygen production of the conjugates was evaluated through 2′, 7′-dichlorofluorescin (DCFH) method. Because of the heavy atom effect, the metal porphyrin complexes exhibited the higher singlet oxygen ( 1 O 2 ) quantum yield than that of free base porphyrin. The order of 1 O 2 yield of the synthesized porphyrins was PtPor > PdPor > ZnPor > Por. The MTT assay using HeLa cells verified the low cytotoxicity of porphyrin–indomethacin conjugates in the dark. Upon irradiation, the platinated porphyrin (PtPor) showed the highest therapeutic activity among these conjugates, probably due to its high efficiency of 1 O 2 generation. The cellular uptake and subcellular localization of the conjugates were further evaluated through a confocal laser scanning microscope. The results showed that the conjugates were primarily localized in the lysosomes of HeLa cells.

Abstract:Photodynamic therapy (PDT) represents a promising approach for cancer treatment. However, the oxygen dependency of PDT to generate reactive oxygen species (ROS) hampers its therapeutic efficacy, especially against hypoxic solid tumors. In addition, some photosensitizers (PSs) have dark toxicity and are only activatable with short wavelengths such as blue or UV light, which suffer from poor tissue penetration. Herein, we developed a novel hypoxia-active PS with operability in the near infrared (NIR) region based on the conjugation of a cyclometalated Ru(II) polypyridyl complex of the type [Ru(C^N)(N^N)2] to a NIR emitting COUPY dye. The novel Ru(II) coumarin conjugate exhibits water solubility, dark stability in biological media and high photostability along with advantageous luminescent properties that facilitate both bioimaging and phototherapy. Spectroscopic and photobiological studies revealed that this conjugate efficiently generates singlet oxygen and superoxide radical anions, thereby achieving high photoactivity toward cancer cells upon highly penetrating 740 nm light irradiation even under hypoxic environments (2% O2). The induction of ROS-mediated cancer cell death upon low energy wavelength irradiation along with the low dark toxicity exerted by this Ru(II) coumarin conjugate could circumvent tissue penetration issues while alleviating the hypoxia limitation of PDT. As such, this strategy could pave the way to the development of novel NIR and hypoxia active Ru(II)-based theragnostic PSs fuelled by the conjugation of tunable, low molecular weight COUPY fluorophores.

A Photodynamic and Photochemotherapeutic Platinum‐Iridium Charge‐Transfer Conjugate for Anticancer Therapy

Abstract:The novel hetero-dinuclear complex trans , trans , trans -[Pt IV (py) 2 (N 3 ) 2 (OH)(μ-OOCCH 2 CH 2 CONHCH 2 -bpyMe)Ir III (ppy) 2 ]Cl ( Pt-Ir ), exhibits charge transfer between the acceptor photochemotherapeutic Pt(IV) ( Pt-OH ) and donor photodynamic Ir(III) ( Ir-NH 2 ) fragments. It is stable in the dark, but undergoes photodecomposition more rapidly than the Pt(IV) parent complex ( Pt-OH ) to generate Pt(II) species, an azidyl radical and 1 O 2 . The Ir(III) * excited state, formed after irradiation, can oxidise NADH to NAD⋅ radicals and NAD + . Pt-Ir is highly photocytotoxic towards cancer cells with a high photocytotoxicity index upon irradiation with blue light (465 nm, 4.8 mW/cm 2 ), even with short light-exposure times (10–60 min). In contrast, the mononuclear Pt-OH and Ir-NH 2 subunits and their simple mixture are much less potent. Cellular Pt accumulation was higher for Pt-Ir compared to Pt-OH . Irradiation of Pt-Ir in cancer cells damages nuclei and releases chromosomes. Synchrotron-XRF revealed ca . 4× higher levels of intracellular platinum compared to iridium in Pt - Ir treated cells under dark conditions. Luminescent Pt-Ir distributes over the whole cell and generates ROS and 1 O 2 within 1 h of irradiation. Iridium localises strongly in small compartments, suggestive of complex cleavage and excretion via recycling vesicles (e.g. lysosomes). The combination of PDT and PACT motifs in one molecule, provides Pt-Ir with a novel strategy for multimodal phototherapy.

Abstract:Trinuclear ruthenium(II) polypyridyl complexes anchored to benzimidazole-triazine/ trisamine scaffolds were investigated as photosensitizers for photodynamic therapy. The trinuclear complexes were noted to produce a significant amount of singlet oxygen in both DMF and aqueous media, are photostable and show appreciable emission quantum yields (ɸ em ). In our experimental setting, despite the moderate phototoxic activity in the HeLa cervical cancer cell line, the phototoxic indices (PI) of the trinuclear complexes are superior relative to the PIs of a clinically approved photosensitizer, Photofrin®, and the pro-drug 5-aminolevulinic acid (PI: >7 relative to PI: >1 and PI: 4.4 for 5-aminolevulinic acid and Photofrin®, respectively). Furthermore, the ruthenium complexes were noted to show appreciable long-term cytotoxicity upon light irradiation in HeLa cells in a concentration-dependent manner. Consequently, this long-term activity of the ruthenium(II) polypyridyl complexes embodies their ability to reduce the probability of the recurrence of cervical cancer. Taken altogether, this presents a strong motivation for the development of polymetallic complexes as anticancer agents.
