Abstract:The effects of extraction time, temperature, pressure and different concentration of ethanol and their interactions on the yields of amentoflavone, quercetin and ginkgetin extracted from Taxus chinensis by supercritical CO2 were investigated by using a central composite design (CCD). An CCD experimental design with four factors and five levels was used to optimize the extraction parameters. Ultra performance liquid chromatography (UPLC) was used to analyze the content of the tree components in the extracts. Experimental results show that the main effects of factors and their interactions are significant on the yields (p < 0.05). The optimal extraction conditions were established for the three compounds: yield of 4.47 mg/g for amentoflavone at 48 °C, 25 MPa, 2.02 h and 78.5% ethanol, 3.73 mg/g for quercetin at 46 °C, 24 MPa, 2.3 h, 82% ethanol and 3.47 mg/g for ginkgetin at 48 °C, 20 MPa, 2.38 h, 82% ethanol, respectively.

Abstract:Supercritical carbon dioxide (SC-CO2) extraction of bioactive compounds including flavonoids and phenolics from Ampelopsis grossedentata stems was carried out. Extraction parameters such as pressure, temperature, dynamic time and modifier, were optimized using an orthogonal array design of L9 (34), and antioxidant activities of the extracts were evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assay and ferrous ion chelating (FIC) assay. The best conditions obtained for SC-CO2 extraction of flavonoids was 250 bar, 40 °C, 50 min, and with a modifier of methanol/ethanol (1:3, v/v), and that for phenolics extraction was 250 bar, 40 °C, 50 min, and with a modifier of methanol/ethanol (1:1, v/v). Meantime, flavonoids and phenolics were found to be mainly responsible for the DPPH scavenging activity of the extracts, but not for the chelating activity on ferrous ion according to Pearson correlation analysis. Furthermore, several unreported flavonoids such as apigenin, vitexin, luteolin, etc., have been detected in the extracts from A. grossedentata stems.

Abstract:In this study, an eco-friendly supercritical carbon dioxide (SC-CO 2 ) extraction of polyphenolic compounds from Hippophae salicifolia leaf was optimized to achieve the highest extraction yield with maximum total phenolic content (TPC) and minimum IC50. The central composite design was used to establish an experimental design for RSM. The effect of the pressure, temperature, carbon dioxide flow rate, and co-solvent amount was scrutinized using variance analysis (ANOVA). Under optimized condition (25.13 MPa, 47.53 °C, 14.47 g/min, and 2.43%), the experimental data (yield of extraction: 4.38%, TPC: 84.31 mg GAE/g, and IC50: 41.94 μg/mL) showed good agreement with the predicted values (yield of extraction: 4.53%, TPC: 83.37 mg GAE/g, and IC50: 40.2 μg/mL). Nine polyphenolic compounds: gallic acid, caffeic acid, ferulic acid, vanillic acid, p-coumaric acid, quercetin, myricetin, kaempferol, and rutin were analyzed in SC-CO 2 extract using HPLC. SC-CO 2 extraction was more selective for ferulic acid, myricetin, and quercetin extraction. The study results revealed that SC-CO 2 extract had significant antibacterial activity against eight bacterial strains.

Abstract:Supercritical carbon dioxide extraction of oil from Torreya grandis seeds was optimized using response surface methodology. Its physicochemical properties, fatty acid composition, total phenolic content and antioxidant activities from two varieties namely T. grandis var. merrillii and T. grandis var. jiulongshanensis were investigated. As results, the maximum oil yield was predicted to be 35.43 g/100 g dry seed using an extraction pressure of 24.98 MPa, temperature of 51.39 °C, extraction duration of 4 h and CO2 flow rate of 15.42 L/h. T. grandis var. merrillii seed oil has better overall quality. Its acid, peroxide, iodine and saponification value were 0.63 mg KOH/g oil, 3.37 m EqO2/Kg, 124.33 mg/100 g oil, 122.86 mg KOH/g oil, respectively. Large proportions of PUFA (59.99%) were found in the oil with linoleic acid (37.81 g/100 g oil) as the main fatty acid. The oil was also found present high content of phenolic compounds (16.9 mg GAE/g) and antioxidant activity in DPPH radical scavenging assay and β-carotene bleaching test (IC50 values of 123.76 ± 0.13 μg/mL and 194.65 ± 0.09 μg/mL, respectively). Our results indicate that T. grandis var. merrillii seeds oil is a good candidate for use as nutrient rich edible oil or ingredients for functional foods.

Abstract:In the present study, supercritical carbon dioxide (SC-CO(2)) extraction of seed oil from winter melon (Benincasa hispida) was investigated. The effects of process variables namely pressure (150-300 bar), temperature (40-50 °C) and dynamic extraction time (60-120 min) on crude extraction yield (CEY) were studied through response surface methodology (RSM). The SC-CO(2) extraction process was modified using ethanol (99.9%) as co-solvent. Perturbation plot revealed the significant effect of all process variables on the CEY. A central composite design (CCD) was used to optimize the process conditions to achieve maximum CEY. The optimum conditions were 244 bar pressure, 46 °C temperature and 97 min dynamic extraction time. Under these optimal conditions, the CEY was predicted to be 176.30 mg-extract/g-dried sample. The validation experiment results agreed with the predicted value. The antioxidant activity and fatty acid composition of crude oil obtained under optimized conditions were determined and compared with published results using Soxhlet extraction (SE) and ultrasound assisted extraction (UAE). It was found that the antioxidant activity of the extract obtained by SC-CO(2) extraction was strongly higher than those obtained by SE and UAE. Identification of fatty acid composition using gas chromatography (GC) showed that all the extracts were rich in unsaturated fatty acids with the most being linoleic acid. In contrast, the amount of saturated fatty acids extracted by SE was higher than that extracted under optimized SC-CO(2) extraction conditions.

Abstract:The effects of different operating conditions on four extraction techniques were studied and compared. The criteria analyzed were total phenol and flavonoid contents, individual flavonoids and antioxidant activity. The common operating conditions of extraction are ratio m/v: 5g:50ml; 80% ethanol with mechanical agitation. It appears that the highest values for the total phenol and flavonoid contents were reached when ultrasound assisted extraction (UAE) was carried out at 125W during 30min at 35°C, microwave assisted extraction (MAE) at 200W during 180s, supercritical CO2 extraction (SCE) at 80°C, 10MPa and high pressure extraction at 50MPa during 30min at 35°C. These conditions are not optimum to obtain the highest antioxidant activity for MAE (300W) and HPE (100MPa).

Abstract:Response Surface Methodology (RSM) was employed to optimize the extraction rate of phenolic and flavonoid contents from peanut skin by supercritical carbon dioxide (ScCO2) assisted by ethanol as entrainer. The studied extraction parameters were pressure (10 to 30 MPa), temperature (40 to 70 oC), and the ratio of ethanol (2.5 to 7.5%). Brunner’s and Esquivel’s models were applied to evaluate the extraction rate. The best-operating conditions, in the tested range, were 30 MPa, 40 °C, and 4.64% of ethanol ratio, with a maximum extraction rate of 0.22 mg/g.sec and 0.19 mg/g.sec of the phenolic and flavonoid content, respectively. Abstract:In this research, a supercritical CO2-ethanol extraction was optimized to obtain a green coffee oil rich in bioactive compounds. A face-centered central composite design was used to evaluate the effect of temperature (50–70 °C), extraction pressure (15.0–30.0 MPa), and cosolvent content (5–20%) on the extraction yield and total phenolic compound content of green coffee supercritical extract (GCSE). The experimental data were fitted to a second-order polynomial model. According to the statistical analyses, the lack of fit was not significant for either mathematical model. From the response surface plots, the extraction pressure and cosolvent content significantly impacted the extraction yield, while the total phenolic compound content was impacted by temperature and cosolvent content. The optimal conditions were a 20% cosolvent content, a pressure of 30 MPa, and a temperature of 62 °C, which predicted an extraction yield of 7.7% with a total phenol content of 5.4 mg gallic acid equivalent g GCSE−1. The bioactive compounds included 5-caffeoylquinic acid (11.53–17.91 mg g GCSE−1), caffeine (44.76–79.51 mg g GCSE−1), linoleic acid (41.47–41.58%), and palmitic acid (36.07–36.18%). Our results showed that GCSE has the outstanding chemical quality and antioxidant potential, suggesting that GCSE can be used as a functional ingredient.

Abstract:The potential of supercritical CO2 (SC-CO2) for the extraction of bioactive compounds from mango by-products was assessed. Carotenoid extraction was optimized using a design of experiments based on temperature (35, 55 and 70 °C), pressure (10 and 35 MPa) and co-solvent addition (0%, 10% and 20% of ethanol or acetone). Moreover, the co-extraction of phenolic acids, flavonoids and xanthonoids was evaluated in a subset of parameters. Finally, a comparison was made between SC-CO2 and a two-step organic solvent extraction of the bioactive compounds from the pulp and peel fractions of two Ecuadorian varieties. The optimal extraction temperature was found to be dependent on the bioactive type, with phenolics requiring higher temperature than carotenoids. The optimal overall conditions, focused on maximal carotenoids recovery, were found to be 55 °C, 35 MPa and 20% of ethanol. The main carotenoid was β-carotene, while phenolics differed among the varieties. The bioactive content of the peel was up to 4.1-fold higher than in the pulp fraction. Higher antioxidant activity was found in the extracts obtained with organic solvents. SC-CO2 is a promising technology for the isolation of valuable compounds from mango by-products.

Abstract:Supercritical fluid extraction using carbon dioxide (SFE-CO2) brings a convincing advance in the production of plant oils used in cosmetics, in fortified foods and dietary supplements, and in pharmaceuticals and medicine. The SFE-CO2-extracted, hexane-extracted, and cold-pressed plant oils of pumpkin (Cucurbita pepo L.), flax (Linum usitatissimum L.), linden (Tilia sp.), poppy (Papaver somniferum L.), apricot (Prunus armeniaca L.), and marigold (Calendula officinalis L.) seeds were investigated in terms of oil yield, fatty acid composition, unsaponifiable matter yield and composition, and the antioxidant activity of unsaponifiable matter. SFE-CO2 proved to be the preferred extraction method for four out of six plant materials, especially for seeds with lower oil content. However, for seeds with higher oil content, such as apricots, cold pressing is a viable alternative. A comparison of fatty acid composition did not reveal significant differences between extraction techniques. SFE-CO2 extraction improved the total phytosterol content of oils, especially pumpkin seed oil. A high variability in the antioxidant potential of the unsaponifiable matter studied was determined, with pumpkin seed oil showing the highest antioxidant activity. A correlation analysis was performed between unsaponifiable composition and antioxidant activity, and showed statistically significant correlations with squalene, cycloartenol, and an unidentified compound. This is the first comparison of the phytosterol compositions of linseed, apricot, linden, and marigold. Through continued optimization, SFE-CO2 has the potential to revolutionize the production of plant oils and provide a sustainable and efficient alternative.

Background The volatile compounds that comprise lavender essential oils, including linalool and linalyl acetate, have demonstrative therapeutic properties. The supercritical CO 2 extraction (scCO 2 ) has demonstrated efficiency and selectivity for the extraction of essential oils from vegetable matrices. Methods The solubility of lavender essential oil in scCO 2 was determined using a high-pressure variable-volume and modeled by semiempirical models. Supercritical extraction was carried out at a pressure of 180, 250 and 300 bar and a temperature of 40–60 °C, with and without cosolvents. kinetic curve has been modeled by broken and intact cells model developed by Sovová. The composition of the extracts was evaluated through GC–MS and their antioxidant activity by the DPPH method. Findings The highest values of oil solubility and extraction yield were obtained at 250 bar and 60 °C. Furthermore, the extraction yield increases significantly with the addition of ethanol as a co-solvent (0.2 % v/v). A complete set of equilibrium data and kinetic parameters has been reported on a large scale for the first time. The dominant components identified in the extract were linalool and linalyl acetate, and the extracts showed very satisfactory results for antioxidant capacity. Compared with traditional methods like Soxhlet extraction, the supercritical extracts were determined to be more interesting for the formulation of nutraceutical products or biomedical applications. Abstract:Supercritical fluid carbon dioxide (SF-CO2) was used to extract oil from Elaeagnus mollis Diels (E. mollis Diels) seed and its antioxidant ability was also investigated. The effect of extraction pressure (20–35 MPa), extraction temperature (35–65 C), extraction time (90–180 min) and seed particle size (40–100 mesh) on the oil yield were studied. An orthogonal experiment was conducted to determine the best operating conditions for the maximum extraction oil yield. Based on the optimum conditions, the maximum yield reached 29.35% at 30 MPa, 50 C, 150 min, 80 mesh seed particle size and 40 g/min SF-CO2 flow rate. The E. mollis Diels seed (EDS) oil obtained under optimal SF-CO2 extraction conditions had higher unsaturated fatty acid content (91.89%), higher vitamin E content (96.24 ± 3.01 mg/100 g) and higher total phytosterols content (364.34 ± 4.86 mg/100 g) than that extracted by Soxhlet extraction (SE) and cold pressing (CP) methods. The antioxidant activity of the EDS oil was measured by DPPH and hydroxyl radical scavenging test. EDS oil extracted by different methods exhibited a dose-dependent antioxidant ability, with IC50 values of no significant differences. Based on the results of correlation between bioactive compounds, lupeol and -tocopherol was the most important antioxidant in EDS oil.

Abstract:BACKGROUNDTo extract natural volatile compounds from tea (Camellia sinensis) flowers without thermal degradation and residue of organic solvents, supercritical fluid extraction (SFE) using carbon dioxide was employed to prepare essential oil of tea flowers in the present study. Four important parameterspressure, temperature, static extraction time, and dynamic extraction timewere selected as independent variables in the SFE.RESULTSThe optimum extraction conditions were the pressure of 30 MPa, temperature of 50 degrees C, static time of 10 min, and dynamic time of 90 min. Based on gas chromatography-mass spectrometry analysis, 59 compounds, including alkanes (45.4%), esters (10.5%), ketones (7.1%), aldehydes (3.7%), terpenes (3.7%), acids (2.1%), alcohols (1.6%), ethers (1.3%) and others (10.3%) were identified in the essential oil of tea flowers. Moreover, the essential oil of tea flowers showed relatively stronger DPPH radical scavenging activity than essential oils of geranium and peppermint, although its antioxidative activity was weaker than those of essential oil of clove, ascorbic acid, tert-butylhydroquinone, and butylated hydroxyanisole.CONCLUSIONEssential oil of tea flowers using SFE contained many types of volatile compounds and showed considerable DPPH scavenging activity. Abstract:Phenolic compounds from mango (M. indica) seed kernels (MSK) var. Sugar were obtained using supercritical CO2 and EtOH as an extraction solvent. For this purpose, a central composite design was carried out to evaluate the effect of extraction pressure (11–21 MPa), temperature (40–60 °C), and co-solvent contribution (5–15% w/w EtOH) on (i) extraction yield, (ii) oxidative stability (OS) of sunflower edible oil (SEO) with added extract using the Rancimat method, (iii) total phenolics content, (iv) total flavonoids content, and (v) DPPH radical assay. The most influential variable of the supercritical fluid extraction (SFE) process was the concentration of the co-solvent. The best OS of SEO was reached with the extract obtained at 21.0 MPa, 60 °C and 15% EtOH. Under these conditions, the extract increased the OS of SEO by up to 6.1 ± 0.2 h (OS of SEO without antioxidant, Control, was 3.5 h). The composition of the extract influenced the oxidative stability of the sunflower edible oil. By SFE it was possible to obtain extracts from mango seed kernels (MSK) var. Sugar that transfer OS to the SEO. These promissory extracts could be applied to foods and other products.

Abstract:The evaluation of wastes from edible oil production industry has increasing importance because of resources deficiency and growing population day by day. It was aimed to investigate the recovery potential of oil and valuable components such as total phenolic contents (TPC), total flavonoid content (TFC) and antioxidant activities with conventional and green extraction methods from edible sunflower oil production wastes. Firstly, Soxhlet extraction was applied on the waste samples from filtration of the oil and then supercritical CO2 (SC- CO2) extraction was performed to compare extraction yield. The statistical evaluation of the SC- CO2 extraction experiments was conducted with the Minitab Software program. It was concluded that 37% extraction yield was obtained with the mixture of perlite and waste sample, while the highest recovery was observed as 27% for the perlite-free sample. The interaction of independent variables (temperature and pressure) on the recovery yields were determined as significant (p=0.002). Pressure was detected more effective on the valuable substance contents of the extracts than temperature. The most efficient conditions were 50 oC and 22.1 MPa for oil recovery, while 60 oC and 20 MPa for valuable substance recovery.

Abstract:The extraction of natural compounds requires extraction technologies that protect extract quality and environmental protection. The extraction of L. rivularis stalks was studied as a function of temperature, pressure, and specific solvent consumption, using a Box–Behnken design. The condition for the highest extraction yield was determined. The effect of temperature, pressure, and specific solvent consumption on extraction yield from Leptocarpha rivularis stalks was studied using a Box–Behnken design. The effect of pressure on extraction yield was the most important, followed by temperature and specific solvent consumption (p ≤ 0.05). The extraction yield correlated (p ≤ 0.05) positively with the CO2 density. A diffusion model adequately described the cumulative extraction curve with the highest yield at the selected extraction condition. The inhibition of linoleic acid oxidation and the inhibition of lipoxygenase enzyme were higher in the supercritical (SC) extract than in the hydroethanolic extract. SC extraction could be used to isolate compounds from L. rivularis stalks with functional value.

Abstract:Safflower seed oil extraction with supercritical CO2 at series operational parameters of pressure, temperature, flow rate and particle size was investigated in a bench scale apparatus. The results show that the extraction yields plotted as a function of time are significantly affected by the extraction pressure, flow rate and particle size, but extraction yields plotted versus CO2 used are scarcely affected by flow rate. Extraction temperature has a slight effect on the extraction curves. In order to describe the extraction process, the Sovova’s extended Lack’s Model (SLM) was used and the experimental data were well fitted by it. The extraction was scaled up to pilot plant and the computed values of SLM are in good agreement with the pilot plant data. Additionally, the quality of safflower seed oil obtained by supercritical CO2 extraction is superior to that of oil obtained by traditional methods. It is noted that a new method of changing flow rate was proposed to improve the process efficiency and proved to be valuable by experiment.

Abstract:Abstract A wide range of protein cancer biomarkers is currently recommended in international guidelines for monitoring the growth and regression of solid tumors. However, a number of these markers are also present in low concentrations in blood obtained from healthy individuals and from patients with benign diseases. In contrast, evidence has accumulated that suggests that modified methylated DNA is strongly related to the cancer phenotype. The modifications found in modified methylated DNA include a global loss of methylation in the genomes of the tumor cells as well as focal hypermethylation of gene promoters. Because tumor cells naturally secrete DNA and upon cell death leak DNA, modified methylated DNA can be detected in blood, urine, sputum and other body fluids. At present international guidelines do not include recommendations for monitoring modified methylated DNA. The low level of evidence can partly be explained by incomplete collection of serial blood samples, by analytical challenges, and by lack of knowledge of how monitoring studies should be designed and how serial marker data obtained from individual patients should be interpreted. Here, we review the clinical validity and utility of methylated DNA for monitoring the activity of malignant disease.
Supercritical carbon dioxide extracts of Schinus terebinthifolia fruits and their utilization against microbial illness, lipase, and butyrylcholinesterase activities in vitro

Abstract:The extraction methods used to obtain natural products face some problems, such as solvent toxicity, high extraction time, and low yields. Supercritical carbon dioxide fluid extraction (SFE-CO2) is an encouraging extraction system for obtaining high-yield of natural extracts. In this work, Schinus terebinthifolia fruits were extracted via SFE-CO2 using two conditions: A (static extraction) (SE) for 15 min, followed by dynamic extraction (DE) for 45 min, and B (without SE but with DE for 60 min). The extract yield was 0.205 g and 0.236 g via condition A and B, respectively. High-performance liquid chromatography assessment revealed the occurrence of several constituents with high quantities in the extract at condition B. The well diffusion test showed inhibition of 26 ± 0.1, 25 ± 0.2, 29 ± 0.1, 33 ± 0.2, 27 ± 0.1, and 8.0 ± 0.1 mm zones using the extract at condition B, while at condition A there were low inhibition zones towards Staphylococcus aureus, Pseudomonas areginosa, Bacillus subtilis, Escherichia coli, Candida albicans, and Aspergillus niger, correspondingly. Lipase (obesity stimulant) and butyrylcholinesterase (Alzheimer stimulant) were inhibited by the extract at condition B with IC50 quantities of 27.03 and 4.83 μg/mL, while it was 37.45 and 17.57 μg/mL, respectively at condition A.
