Abstract:The bio-enzyme degradation technology is a promising approach to sustainably remove pollution in the water and laccase is one of the most widely used enzymes in this area. Nevertheless, the further industrial application of laccase is limited by low stability, short service, low reusability and high price. The immobilization technology can significantly improve the stability and reusability of enzymes and thus promoting their industrial applications. Nanocomposite materials have been developed and applied in the efficient immobilization of laccase due to their superior physical, chemical, and biological performance. This paper presents a comprehensive review of various nanocomposite immobilization methods for laccase and the consequent changes in enzymatic properties post-immobilization. Additionally, a comprehensive analysis is conducted on the factors that impact laccase immobilization and its water removal efficiency. Furthermore, this review examines the effectiveness of common contaminants' removal mechanisms while summarizing and discussing issues related to laccase immobilization on nanocomposite carriers. This review aims to provide valuable guidance for enhancing laccase immobilization efficiency and enzymatic water pollutant removal.

Abstract:Laccase, as an important biocatalyst, has aroused wide public interests in the field of environment and catalysis. However, the application of laccase in large-scale is still limited due to its high solubility and poor stability. In order to improve the reusability of laccase, immobilization with functional materials has attracted increasing attention. Metal-organic frameworks (MOFs) stand out as a promising carrier for its high adsorption capacity and flexible structure. The present paper reviews the current status and remaining challenges in the immobilization of laccase with MOFs. Furthermore, three methods are proposed to further improve the immobilization of laccase with MOFs, including co-immobilization of laccase and mediator, co-immobilization of laccase and other enzymes, and coupling of biocatalysis with photochemical reactions. This work, as the first review article on the immobilization of laccase with MOFs, will provide a navigation map for the subsequent selection and design of MOFs for more targeted immobilization of laccase.

Abstract:Laccase, as an environmentally friendly biocatalyst with high catalytic efficiency and less by-product, has huge potential in the field of biodegradation and attracted extensive attention. However, the substrates range of laccase was limited by its low redox potential. The involvement of redox mediators which are called the laccase-mediator system (LMS) can expand the substrate range of laccase to non-phenolic substances and improve its degradation efficiency. Herein, a co-immobilized method with practical application possibilities of laccase and 2,2-Azino-bis-3-ethylbenzothiazoline-6-sulfonic Acid (ABTS) was developed for the treatment of dyes that could not be directly degraded by laccase in wastewater. An amino-functionalized metal–organic framework (MOF), UiO-66(Zr)–NH2, was grown in situ on polyethylene terephthalate (PET) using a hydrothermal method, and the obtained composites were acted as the carrier of laccase. Laccase was covalently immobilized on the PET/UiO-66(Zr)–NH2, and then ABTS was fixed to the pores of the UiO-66(Zr)–NH2 by adsorption, realizing the co-immobilization of ABTS and laccase, named PET/ABTS@UiO-66(Zr)–NH2/Lac. PET/UiO-66(Zr)–NH2/Lac enhanced the pH stability of free laccase under acidic conditions, and also has a higher affinity for substrate and great recyclability. The resultant lamellar biocatalyst (PET/ABTS@UiO-66(Zr)–NH2/Lac) was used in the degradation of crystal violet (CV), and the CV removal was increased from 8.61% (free laccase) to 58.80% in 24 h. The removal of malachite green (MG), alizarin green (AG), and methyl orange (MO) was up to 64.82%, 61.61%, and 21.12%, respectively. The laccase and ABTS could be recycled synchronously, which avoided the loss of activity of the system. This work provides a navigation reference for the co-immobilization of LMS by MOFs.

Abstract:Phenolic compounds are present in different concentrations in the effluent from numerous industrial and agricultural activities. These compounds are harmful to living organisms due to their high toxicity, hence indicating a serious environmental concern. Although conventional methods such as chemical, physical, and physicochemical procedures have been widely used for treating phenol-contaminated wastewaters, they are not useful owing to some short-comings. Compared to conventional procedures, much attention has been recently devoted to enzymatic methods because of high catalytic efficiency, mild operating conditions and environmentally friendly feature. Among various enzymes, laccases have demonstrated a superior potential for removing phenolic contaminants. Thus, this review summarizes the up-to-date literature on the use of free and immobilized laccases from different microbial source in the degradation and remediation of phenolic pollutants in batch processes and continuous reactors. In general, examples through the review approve that free laccases as well as immobilized laccases onto inorganic, organic (natural or synthetic) and hybrid supports show excellent performance in the remediation of phenolic compounds from wastewater. In contrast to immobilized laccases, free laccases suffer from high prices, low operative stability, and inability to recover and reuse in their native forms. Moreover, the possible mechanisms associated with oxidation of phenolic compounds by the laccase-catalyzed systems are assessed.

Abstract:In the past decade, we and others have consistently reported linkage to a schizophrenia (SZ) susceptibility region on chromosome 8p21. Most recently, in the largest SZ linkage sample to date, a multi‐site international collaboration performed a SNP‐based linkage scan (∼6,000 SNPs; 831 pedigrees; 121 from Johns Hopkins (JHU)), that showed the strongest evidence for linkage in a 1 Mb region of chr 8p21 from rs1561817 to rs9797 (Zmax = 3.22, P = 0.0004) [Holmans et al. 2009. Mol Psychiatry]. We have investigated this 8p21 peak region further in two ways: first by linkage and family‐based association in 106 8p‐linked European‐Caucasian (EUC) JHU pedigrees using 1,402 SNPs across a 4.4 Mb region surrounding the peak; second, by an independent case‐control association study in the genetically more homogeneous Ashkenazim (AJ) (709 cases, 1,547 controls) using 970 SNPs in a further narrowed 2.8 Mb region. Family‐based association analyses in EUC pedigrees and case‐control analyses in AJ samples reveal significant associations for SNPs in and around DPYSL2 and ADRA1A, candidate genes previously associated with SZ in our work and others. Further, several independent gene expression studies have shown that DPYSL2 is differentially expressed in SZ brains [Beasley et al. 2006. Proteomics 6(11):3414–3425; Edgar et al. 2000. Mol Psychiatry 5(1):85–90; Johnston‐Wilson et al. 2000. Mol Psychiatry 5(2):142–149] or in response to psychosis‐inducing pharmaceuticals [Iwazaki et al. 2007. Proteomics 7(7):1131–1139; Paulson et al. 2004. Proteomics 4(3):819–825]. Taken together, this work further supports DPYSL2 and the surrounding genomic region as a susceptibility locus for SZ. © 2010 Wiley‐Liss, Inc.

Abstract:Laccase is a multicopper oxidoreductase enzyme that can oxidize organics such as phenolic compounds. Laccases appear to be unstable at room temperature, and their conformation often changes in a strongly acidic or alkaline environment, making them less effective. Therefore, rationally linking enzymes with supports can effectively improve the stability and reusability of native enzymes and add important industrial value. However, in the process of immobilization, many factors may lead to a decrease in enzymatic activity. Therefore, the selection of a suitable support can ensure the activity and economic utilization of immobilized catalysts. Metal-organic frameworks (MOFs) are porous and simple hybrid support materials. Moreover, the characteristics of the metal ion ligand of MOFs can enable a potential synergistic effect with the metal ions of the active center of metalloenzymes, enhancing the catalytic activity of such enzymes. Therefore, in addition to summarizing the biological characteristics and enzymatic properties of laccase, this article reviews laccase immobilization using MOF supports, as well as the application prospects of immobilized laccase in many fields.

Abstract:Phenolic compounds such as phenol, bisphenol A, 2,4-dichlorophenol, 2,4-dinitrophenol, 4-chlorophenol and 4-nitrophenol are well known to be highly detrimental to both human and living beings. Thus, it is of critical importance that suitable remediation technologies are developed to effectively remove phenolic compounds from aqueous solutions. Biodegradation utilizing enzymatic technologies is a promising biotechnological solution to sustainably address the pollution in the aquatic environment as caused by phenolic compounds under a defined environmentally optimized strategy and thus should be investigated in great detail. This review aims to present the latest developments in the employment of immobilized laccase for the degradation of phenolic compounds in water. The review first succinctly delineates the fundamentals of biological enzyme degradation along with a critical discussion on the myriad types of laccase immobilization techniques, which include physical adsorption, ionic adsorption, covalent binding, entrapment, and self-immobilization. Then, this review presents the major properties of immobilized laccase, namely pH stability, thermal stability, reusability, and storage stability, as well as the degradation efficiencies and associated kinetic parameters. In addition, the optimization of the immobilized enzyme, specifically on laccase immobilization methods and multi-enzyme system are critically discussed. Finally, pertinent future perspectives are elucidated in order to significantly advance the developments of this research field to a higher level.

Abstract:Laccases have enormous potential as promising 'green' biocatalysts in environmental applications including wastewater treatment and polluted soil bioremediation. The catalytic oxidation reaction they perform uses only molecular oxygen without other cofactors, and the only product after the reaction is water. Co-immobilizing laccase-mediator system by in-situ synthesis of MOF in PVA hydrogels for enhanced laccase stability and dye decolorization efficiency

Abstract:The laccase mediator system (LMS) with a broad substrate range has attracted much attention as an efficient approach for water remediation . However, the practical application of LMS is limited due to their high solubility, poor stability and low reusability. Herein, the bimetallic Cu/ZIFs encapsulated laccase was in-situ grown in poly(vinyl alcohol) (PVA) polymer matrix . The PVA-Lac@Cu/ZIFs hydrogel was formed via one freeze-thawing cycle, and its catalytic stability was significantly improved. The mediator was further co-immobilized on the hydrogel, and this hierarchically co-immobilized ABTS/PVA-Lac@Cu/ZIFs hydrogel could avoid the continuous oxidation reaction between laccase and redox mediators. The co-immobilized LMS biocatalyst was used to degrade malachite green (MG), and the degradation rate was up to 100 % within 4 h. More importantly, the LMS could be recycled synchronously from the dye solutions and reused to degrade MG multiple times. The degradation rate remained above 69.4 % after five cycles. Furthermore, the intermediate products were detected via liquid chromatography-mass spectrometry, and the potential degradation pathways were proposed. This study demonstrated the significant potential of utilizing the MOF nanocrystals and hydrogel as a carrier for co-immobilized LMS, and the effective reuse of both laccase and mediator was promising for laccase application in wastewater treatment.

Abstract:Industrialization and population growth are leading to the production of significant amounts of sewage containing hazardous xenobiotic compounds. These compounds pose a threat to human and animal health, as well as the overall ecosystem. To combat this issue, chemical, physical, and biological techniques have been used to remove these contaminants from water bodies affected by human activity. Biotechnological methods have proven effective in utilizing microorganisms and enzymes, particularly laccases, to address this problem. Laccases possess versatile enzymatic characteristics and have shown promise in degrading different xenobiotic compounds found in municipal, industrial, and medical wastewater. Both free enzymes and crude enzyme extracts have demonstrated success in the biotransformation of these compounds. Despite these advancements, the widespread use of laccases for bioremediation and wastewater treatment faces challenges due to the complex composition, high salt concentration, and extreme pH often present in contaminated media. These factors negatively impact protein stability, recovery, and recycling processes, hindering their large-scale application. These issues can be addressed by focusing on large-scale production, resolving operation problems, and utilizing cutting-edge genetic and protein engineering techniques. Additionally, finding novel sources of laccases, understanding their biochemical properties, enhancing their catalytic activity and thermostability, and improving their production processes are crucial steps towards overcoming these limitations. By doing so, enzyme-based biological degradation processes can be improved, resulting in more efficient removal of xenobiotics from water systems. This review summarizes the latest research on bacterial laccases over the past decade. It covers the advancements in identifying their structures, characterizing their biochemical properties, exploring their modes of action, and discovering their potential applications in the biotransformation and bioremediation of xenobiotic pollutants commonly present in water sources.

Abstract:Laccases are versatile enzymes that belong to the multi-copper oxidase family. This enzyme has several biotechnological applications because of its ablilty to oxidize a wide range of phenolic and non-phenolic substrates. However, their large-scale applicability in bioremediation and water treatment is hindered by high salt content and extreme pH values of the polluted media which also affects the stability, recovery and recycling of laccase. Apart from some bacteria, laccase is abundantly present in several lignin-degrading white-rot fungi viz . Ascomycetes , Deuteromycetes , and Basidiomycetes . Recently, laccase has been employed in the development of biosensors as a medical diagnostic tool, biofuel cells, and in bioremediation purpose to remove herbicides, pesticides, and some toxic chemicals from the soil. However, most of the enzymes including laccase are normally unstable and susceptible to lose their activity over time. This might be avoided by maintaining the activity and lengthening the enzyme's lifespan through the use of appropriate immobilization procedures. The potential of laccase immobilized biocathodes for dye decolorization in microbial fuel cells has recently been studied. Immobilized laccase nanoparticles have potential uses as biocatalyst in the bioremediation of pollutants. In addition, advanced research considering microbial laccase has been conducted for its heterologous expression along with in silico protein engineering to attain maximum enzyme activity which can be potentially applied in different biotechnological sectors. Patent related to laccase also implied that this enzyme can be used as suitable catalyst for the production of promising anti-cancer drugs and even as a significant ingredient in cosmetics.
Enhanced pollutant degradation via green-synthesized core-shell mesoporous Si@Fe magnetic nanoparticles immobilized with metagenomic laccase

Abstract:With rapid industrial expansion, environmental pollution from emerging contaminants has increased, posing severe ecosystem threats. Laccases offer an eco-friendly solution for degrading hazardous substances, but their use as free-form biocatalysts face challenges. This study immobilized laccase (PersiLac1) on green-synthesized Si@Fe nanoparticles (MSFM NPs) to remove pollutants like Malachite Green-containing wastewater and degrade plastic films. Characterization techniques (FTIR, VSM, XRD, SEM, EDS, BET) confirmed the properties and structure of MSFM NPs, revealing a surface area of 31.297 m2.g-1 and a pore diameter of 12.267 nm. The immobilized PersiLac1 showed enhanced activity across various temperatures and pH levels, retaining over 82 % activity after 15 cycles at 80°C with minimal leaching. It demonstrated higher stability, half-life, and decimal reduction time than free laccase. Under 1 M NaCl, its activity was 1.8 times higher than the non-immobilized enzyme. The immobilized laccase removed 98.11 % of Malachite Green-containing wastewater and retained 82.92 % activity over twenty cycles of dye removal. Additionally, FTIR and SEM confirmed superior plastic degradation under saline conditions. These findings suggest that immobilizing PersiLac1 on magnetic nanoparticles enhances its function and potential for contaminant removal. Future research should focus on scalable, cost-effective laccase immobilization methods for large-scale environmental applications.

However, free laccase has the problems of poor stability and difficult recovery. Methods In this study, amino-modified and silanized MOF materials were synthesized by hydrothermal method, and laccase molecules were immobilized with glutaraldehyde as a cross-linking agent. Significant Findings The enzyme-loading capacity of the amino-functionalized modified Cu-MOF increased by 27.98% to 128.48┬Āmg/g compared with that of the control group. The modified Cu-MOF material provided excellent protection to laccase molecules in various environments. The enzyme activity of Lac-Cu-MOF-NH 2 reached 2.57 times that of the free enzyme at pH┬Ā3.5. Moreover, Lac-Cu-MOF-NH 2 achieved 95% removal of Congo red (CR) at pH┬Ā3.5, 50┬Ā┬░C, whereas CR degradation by free laccase was only 11.52%. The degradation rate of Lac-Cu-MOF-NH 2 still reached 84.63% after six cycles. Thus, Lac-Cu-MOF-NH 2 significantly improved CR-degradation efficiency by establishing a synergistic effect between adsorption and laccase degradation. A new strategy is proposed for the industrial application of laccase in the field of wastewater treatment. Abstract:Enzyme immobilization technique has a broad application prospect in the area of biotechnology, biochemistry, and environmental remediation. In this study, the composite hydrogel consisting of dopamine functionalized cellulose nanofiber and alginate was fabricated for immobilization of laccase. Infrared spectra, scanning electron micrographs, and energy dispersive X-ray analysis demonstrated that the laccase molecules were covalently immobilized onto the surface of composite hydrogel. The immobilized laccase exhibited the highest enzymatic activity about 462 U·g−1. Compared with free laccase, the pH, temperature, and storage stabilities of immobilized laccase were improved dramatically. Moreover, the immobilized enzyme can be easily separated from reaction system and reused. It retained 79.6% of its initial activity after 14 cycles of operation. Immobilized laccase was utilized for efficient degradation of bisphenol A (BPA) from polluted water, and the affecting factors on BPA removal efficiency were analyzed. Greater than 82% of BPA was removed under optimum conditions. A small amount of ABTS was added into BPA degradation; the removal efficiency had reached up to 98.7%. Results indicated that the immobilized laccase has great potential for industrial applications.Graphical abstract

Abstract:An amino-functionalized magnetic core-shell structure metal-organic framework (MOF), Fe3O4-NH2@MIL-100 (Fe), has been successfully constructed by a step-by-step method. The composite microspheres exhibited excellent magnetic characteristics, large surface area, and pore size, which were employed for the Trametes versicolor laccase immobilization. The immobilized laccase exhibited high activity recovery (72.05%) and significantly improved stability. The reactive activity remained 75% of its initial activity after storage for 4 weeks. Additionally, when the ambient reached 80 °C, the good stability enabled the immobilization laccase to keep 33% residual activity even after 6 h storage. Application of the immobilized laccase for environmental pollutant alkylphenol polyethoxylate (APEO) removal was also investigated. The results showed the immobilized laccase can remove 100% octylphenol polyethoxylate (OPEO) and 98.16% nonylphenol polyethoxylate (NPEO) in 200 min, and can still achieve 63.42% and 46.17% removal efficiency for OPEO and NPEO after reusing four cycles, respectively. The novel MOF based core-shell magnetic composite microspheres introduced by this study showed promising applications as laccase immobilization support, and both the immobilized laccase and its support had great potential in wastewater treatment.

Abstract:Environmental pollution caused by organic effluents emitted by industry has become a worldwide issue and poses a serious threat to the public and the ecosystem. Metal–organic frameworks (MOFs), comprising metal-containing clusters and organic bridging ligands, are porous and crystalline materials, possessing fascinating shape and size-dependent properties such as high surface area, abundant active sites, well-defined crystal morphologies, and huge potential for surface functionalization. To date, numerous well designated MOFs have emerged as critical functional materials to solve the growing challenges associated with water environmental issues. Here we present the recent progress of MOF-based materials and their applications in the treatment of organic effluents. Firstly, several traditional and emerging synthesis strategies for MOF composites are introduced. Then, the structural and functional regulations of MOF composites are presented and analyzed. Finally, typical applications of MOF-based materials in treating organic effluents, including chemical, pharmaceutical, textile, and agricultural wastewaters are summarized. Overall, this review is anticipated to tailor design and regulation of MOF-based functional materials for boosting the performance of organic effluent remediation.

Abstract:Per- and polyfluoroalkyl substances (PFAS) with stable carbon-fluorine bonds are used in a wide range of industrial and commercial applications. Due to their extreme environmental persistence, PFAS have the potential to bioaccumulate, cause adverse effects, and present challenges regarding remediation. Recently, microbial and enzymatic reactions for sustainable degradation of PFAS have gained attention from researchers, although biological decomposition of PFAS remains challenging. Surprisingly, laccases, the multi-copper oxidases produced by various organisms, show potential for PFAS degradation. Mediators play key roles in initiating laccase induced PFAS degradation and defluorination reactions. The laccase-catalyzed PFAS degradation reactions are relatively slower than normal biocatalytic reactions and the low activity of native laccases constrains their capacity to complete defluorination. With their low redox potential and narrow substrate scope, an innovative remediation strategy must be taken to accelerate this reaction. In this review we have summarized the status, challenges, and future perspectives of enzymatic PFAS degradation. The knowledge of laccase-based defluorination and the molecular basis of the reaction mechanisms overviewed in this study could inform future applications of laccases for sustainable PFAS remediation.

Immobilization of laccase on mesoporous metal organic frameworks for efficient cross-coupling of ethyl ferulate

Abstract:Laccases act as green catalysts for oxidative cross-coupling of phenolic antioxidnt compounds, but low stability and non-recyclability limit its application. To address that, metal-organic frameworks Cu-BTC and Cr-MOF were synthesized as supports to immobilize the efficient laccase from Cerrena sp. HYB07. The Brunauer-Emmett-Teller surface area of Cu-BTC and Cr-MOF were 1213.2 and 907.1 m2/g, respectively. The two carriers respectively presented pore diameters of 1.2-10 nm and 1.4-12 nm as octahedron, indicating nano-scale mesoporosity. These Cu-BTC and Cr-MOF carriers could adsorb laccase with enzyme loading of 1933.2 and 1564.4 U/g carrier, respectively. The stability and organic solvent tolerance of Cu-BTC-laccase and Cr-MOF-laccase were both obviously improved compared to free laccase. Thermal inactivation kinetics showed that both the two immobilized laccases displayed lower thermal inactivation rate constants. Importantly, the Cu-BTC-laccase and Cr-MOF-laccase both showed much higher activity for cross-coupling of ethyl ferulate than free laccase, which had 2.5-fold higher cross-coupling efficiency than that by free laccase. The ethyl ferulate coupling product was also analyzed by mass spectroscopy and the synthesis pathway of ethyl ferulate dimer was proposed. The cross coupling of ethyl ferulate required the formation of radical intermediates of ethyl ferulate generated by laccase mediated oxidation. This work paved the way for MOFs immobilized laccase for cross coupling of antioxidant phenols.
Green synthesis of NiO NPs for metagenome-derived laccase stabilization: Detoxifying pollutants and wastes

Abstract:Laccases play a crucial role in neutralizing environmental pollutants, including antibiotics and phenolic compounds, by converting them into less harmful substances via a unique oxidation process. This study introduces an environmentally sustainable remediation technique, utilizing NiO nanoparticles (NPs) synthesized through green chemistry to immobilize a metagenome-derived laccase, PersiLac1, enhancing its application in pollutant detoxification. Salvadora persica leaf extract was used for the synthesis of NiO nanoparticles, utilizing its phytochemical constituents as reducing and capping agents, followed by characterization through different analyses. Characterization of NiO nanoparticles revealed distinctive FTIR absorption peaks indicating the nanoparticulate structure, while FESEM showed structured NiO with robust interconnections and dimensionality of about 50nm, confirmed by EDX analysis to have a consistent distribution of Ni and O. The immobilized PersiLac1 demonstrated enhanced thermal stability, with 85.55 % activity at 80 °C and reduced enzyme leaching, retaining 67.93 % activity across 15 biocatalytic cycles. It efficiently reduced rice straw (RS) phenol by 67.97 % within 210 min and degraded 70–78 % of tetracycline (TC) across a wide pH range (4.0–8.0), showing superior performance over the free enzyme. Immobilized laccase achieved up to 71 % TC removal at 40–80 °C, significantly outperforming the free enzyme. Notably, 54 % efficiency was achieved at 500 mg/L TC by immobilized laccase at 120 min. This research showed the potential of green-synthesized NiO nanoparticles to effectively immobilize laccase, presenting an eco-friendly approach to purify pollutants such as phenols and antibiotics. The durability and reusability of the immobilized enzyme, coupled with its ability to reduce pollutants, indicates a viable method for cleaning the environment. Nonetheless, the production costs and scalability of NiO nanoparticles for widespread industrial applications pose significant challenges. Future studies should focus on implementation at an industrial level and examine a wider range of pollutants to fully leverage the environmental clean-up capabilities of this innovative technology.
