Abstract:Prostate cancer is the second most frequent cancer diagnosed in men in the world today. Almost all prostate cancers are adenocarcinomas and develop from gland cells. We used the PC3 prostate cancer cell line, which is well studied and derived from a bone metastasis of a grade IV prostatic adenocarcinoma. Cannabidiol (CBD), a major non-psychoactive constituent of cannabis, is a cannabinoid with anti-tumor properties but its effects on prostate cancer cells are not studied in detail. Here, we found cannabidiol decreased prostate cancer cell (PC3) viability up to 37.25% and induced apoptotic cell death in a time and dose-dependent manner. We found that CBD activated the caspases 3/7 pathways and increased DNA fragmentation. Furthermore, we observed an increase of pro-apoptotic genes Bax, an increased level of reactive oxygen species, lower reduced glutathione level, and altered mitochondrial potential in response to CBD treatment leading to lower cellular ATP. Overall, our results suggest that CBD may be effective against prostate cancer cells.
Oxidative Phosphorylation Inhibition in Different Prostate Cancer Models and the Interplay with Androgen Receptor Signaling

Abstract:Introduction: In prostate cancer (PCa), androgen receptor signaling stimulates both glycolysis and oxidative phosphorylation (OxPhos). Early-stage prostate cancer is particularly reliant on OxPhos for its bioenergetic needs. OxPhos inhibitors have entered clinical trials. Here we investigated their interplay with androgens in different PCa cell lines. Methods: We investigated the effects on PCa cell viability of an ATPase inhibitor (oligomycin) and a complex 1 inhibitor (IACS-010759) in the presence or absence of low testosterone concentrations in vitro. Both androgen-sensitive and insensitive PCa cell lines were used. The effects were assessed using MTT assay, flow cytometry and cell morphology. Results: Treatment with oligomycin resulted in massive apoptotic death of VCAP cells in castrate conditions within 48 hours, but the simultaneous addition of low testosterone levels restored VCAP cell viability. However, complex 1 inhibition with IACS-010759 increased cell viability, which was further promoted in the presence of testosterone. Both oligomycin and IACS-010759 dramatically decreased viability in LNCaP cells, while testosterone had a small but statistically non-significant effect. The antitumor effect of OxPhos inhibitors was smaller in LNCaP-C4-2B compared to LNCaP cells. OxPhos inhibitors slightly decreased proliferation rates in androgen-independent PC3 cells and HEK293 cells. Oligomycin on LNCaP-C4-2B and PC3 cells resulted in an increased number of cells in G0-G1 phase and decreased in S-phase and G2M phase, rather than massive apoptosis. Conclusion: There is an interplay between androgen signaling and OxPhos in androgen-dependent PCa cells. Complex 1 inhibitors should be used with caution, given potential pro-tumorigenic effects in subsets of PCa cells.

Anti-Cancer Agents in Medicinal Chemistry

Abstract:Introduction: Many bioactive phytochemicals have essential significance for handling various diseases and developing new drugs. The aim was to investigate the anti-tumor activity and the underlying mechanisms of pistachio pericarp extract (PPE) and pistachio kernel extract (PKE) alone and combined with cisplatin (CP) in the treatment of prostate cancer. Methods: The effects of the PPE, PKE, and CP alone and PPE and PKE in combination with CP (PPE+CP and PKE+CP) on the proliferation of PC-3 cells were determined using the MTT assay. The fold changes of BAX, BCL-2, P53, KLK2, TNF, TGF , and NANOG expression against β-actin were determined by real-time technique. Data were analyzed by two-way ANOVA and repeated measure tests. Results: These research results indicated that a greater anti-proliferative effect of the PPE and PKE was shown in combination with CP compared with treatments using the PPE and PKE or CP alone. The extracts and Cisplatin in vitro had good synergistic effects on the inhibition of the proliferation of PC-3 cells. The IC 50 values of PKE+CP were 4.141, 2.140, and 0.884 ug/mL, and PPE+CP were 2.754, 2.061, and 0.753 ug/mL after 24 h, 48 h, and 72h treatment, respectively. Also, this result presented that the mRNA expression of BAX and P53 increased, and BCL-2, KLK2, TNF, TGF , and NANOG decreased in PC-3 cells. Conclusions: The finding of this research showed for the first time the anti-carcinogenesis effects of separately and in the combination of PPE, PKE, and CP on the PC-3 prostate cancer cells via modulating some genes and that it may be nominated for the herbal anti-cancer medications.

Abstract:Abstract Background To explore the anti-tumor effects of parthenolide in human pancreatic cancer. Methods BxPC-3 cell, a human pancreatic cancer, was treated with parthenolide at different concentrations. The MTT assay was used to analyze cell viability. Flow cytometry and DNA fragmentation analysis were applied to evaluate apoptosis after parthenolide treatment. The wound closure and cell invasion assay were also employed in the study. Western blotting was used to demonstrate Bad, Bcl-2, Bax, caspase-9 and pro-caspase-3 expression. Results The MTT assay indicated that the pancreatic cancer growth could be dose-dependently inhibited by parthenoolide. This phenomenon was confirmed by flow cytometry and DNA fragmentation analysis. The wound closure assay and cell invasion assay showed that BxPC-3 cell was significantly suppressed by parthenolide at 7.5 μM and 15 μM. Western Blotting demonstrated the Bcl-2 and pro-caspase-3 were down-regulated while the Bax and caspase-9 were up-regulated. No alteration in Bad expression was found after treatment. Conclusions The parthenolide can inhibit the cell growth, migration, and induce the apoptosis in human pancreatic cancer. These findings may provide a novel approach for pancreatic cancer treatment.

Abstract:Platinum-based chemotherapy remains the main systemic treatment of ovarian cancer (OC). However, the inevitable development of platinum and poly (adenosine diphosphate-ribose) polymerase inhibitor (PARPi) resistance is associated with poor outcomes, which becomes a major obstacle in the management of this disease. The present study developed "all-in-one" nanoparticles that contained the PARPi olaparib and gallium (Ga) (III) (olaparib-Ga) to effectively reverse PARPi resistance in platinum-resistant A2780-cis and SKOV3-cis OC cells and in SKOV3-cis tumor models. Notably, the olaparib-Ga suppressed SKOV3-cis tumor growth with negligible toxicity. Moreover, the suppression effect was more evident when combining olaparib-Ga with cisplatin or carboplatin, as evaluated in A2780-cis and SKOV3-cis cells. Mechanistically, the combined treatment induced DNA damage, which elicited the activation of ataxia telangiectasia mutated (ATM)/AMT- and Rad3-related (ATR) checkpoint kinase 1 (Chk1)/Chk2 signal transduction pathways. This led to the arrest of cell cycle progression at S and G2/M phases, which eventually resulted in apoptosis and cell death due to unrepairable DNA damage. In addition, effective therapeutic responses to olaparib-Ga and cisplatin combination or olaparib-Ga and carboplatin combination were observed in SKOV3-cis tumor-bearing animal models. Altogether, the present findings demonstrate that olaparib-Ga has therapeutic implications in platinum-resistant OC cells, and the combination of olaparib-Ga with cisplatin or carboplatin may be promising for treating patients with OC who exhibit resistance to both PARPi and platinum.
The Anti-Tumor Activity Of Zoledronic Acid And Synergistic Effect Of Zoledronic Acid Combination With Docetaxel On Prostate Cancer Cell Line Pc-3 In Vitro At Clinically Achievable Concentrations

Abstract:To observe the effect of zoledronic acid alone or combination with docetaxel in different concentrations on prostate cancer PC-3 cells in vitro. Cell morphology, MTT, flow cytometry method was used to observe this effect. Above 200 mu g/ml zoledronic acid significantly enhanced growth suppression (suppression ratio >= 44.5%, P< 0.05), apoptosis (apoptosis ratio >= 5.93%, P< 0.05). The combination of ZOL and DOC inhibited the proliferation of PC-3 cells significantly. The effect was dose-depended by ZOL with a certain dose of DOC. When the concentration of DOC was 10 ng/ml, the inhibition rate was 49.3%, 61.4% respectively after the treat with 200 mu g/ml ZOL or 400 mu g/ml ZOL. The inhibition rate could rise to 59.7%, 74.2% with 200 mu g/ml ZOL or 400 mu g/ml ZOL exposure combined with 100 ng/ml DOC. Flow cytometry showed G2 and S phase of PC-3 cells in 400 mu g/ml zoledronic acid were inhibited significantly more than the control group (P< 0.05) and G1 phase was less inhibitid (P< 0.05). This research validates that ZOL has the effect of inhibiting proliferation and inducing apoptosis of hormone refractory prostate cancer PC-3 cells. ZOL arrests the cell cycle to S phase, G2/M phase. These effects are dose-dependent. The combination of ZOL and docetaxel inhibites proliferation more than each drug alone, the effect of which is also dose-dependent and the two drugs have synergia anti-tumor effect on prostate cancer PC-3 cells in vitro.

Abstract:In this study, we employed Ajwain seed extract to synthesize oxalipalladium (OX) NPs and systematically investigated their physicochemical properties and biological activities. Characterization studies revealed that the OX NPs exhibited an average size of approximately 31.2 nm with a stable zeta potential of –26 mV, indicating colloidal stability conducive to drug delivery applications. We confirmed the homogeneous and spherical nature of the NPs, with FTIR spectra highlighting the presence of functional groups consistent with OX and Ajwain extract. Notably, OX NPs demonstrated potent anticancer activity against HCT116 colon cancer cells, inducing dose-dependent apoptosis. Compared to free oxaliplatin, OX NPs exhibited enhanced cytotoxicity. Flow cytometry analysis further elucidated the apoptotic pathway induced by OX NPs, confirming their efficacy as anticancer agents. Additionally, investigation into the molecular interactions between human serum albumin (HSA) and NPs revealed structural alterations in the protein upon interaction with the ligand. Analysis using three-dimensional fluorescence spectroscopy revealed alterations in the surroundings of Tyr and Trp residues, suggesting the influence of nanoparticles (NPs) on the protein's structural integrity. In a nutshell, our study contributes to the advancement of natural product-based synthesis procedures and therapeutic strategies for colorectal cancer treatment.

Abstract:Objective: The platinum-based chemotherapeutic agent oxaliplatin displays a wide range of antitumor activities. To date, no detailed data are available about the effects of oxaliplatin on hepatocellular carcinoma (HCC) cells. Herein, the anti-proliferation effects of oxaliplatin on HCCLM3 and Hep3B cells in vitro and in vivo are studied. Research methods: Cell viability was assessed by an MTT assay and apoptosis by flow cytometry and transmission electron microscopy. Apoptosis-related proteins in HCCLM3 cells were evaluated by microarray analysis, quantitative reverse transcriptase-PCR assay and western blotting. The effect of oxaliplatin was also studied in vivo using a xenograft model. Results: Oxaliplatin inhibited the growth of HCCLM3 and Hep3B cells. Using flow cytometry, transmission electron microscopy and the terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling assay, we found that apoptosis was the main mechanism by which oxaliplatin inhibited tumor progression. Microarray analysis, quantitative reverse transcriptase-PCR and western blot analysis further demonstrated downregulation of the anti-apoptotic proteins Bcl-2 and Bcl-xL and upregulation of the pro-apoptotic protein Bax during oxaliplatin-induced apoptosis. Conclusions: The antiproliferation effect of oxaliplatin in HCC cells is due to induction of apoptosis. Therefore, oxaliplatin may be an effective treatment for HCC and its use merits further in-depth investigation.

Abstract:The growth of pancreatic cancer has a high predominance in the world. Different therapeutic methods were unsuccessful due to tumor invasion and rapid metastasis. Plants have natural products that were used as therapeutic agents. Accordingly, the purpose of this research was to assess the cytotoxic effect of Portulaca Oleracea against PANC-1 cancer cell line. MTT technique and flow cytometry were done to evaluate the cytotoxicity of P.Oleracea extracts against PANC-1 cancer cell line. For finding the change of CDK and P53 expression levels, qPCR carries out. The findings of the MTT assay exhibited that P.Oleracea extracts had toxicity potential on PANC- one cancer cell line. Also, the results of gene expression showed the high expression of P53 and reduction of CDK gene expression following treatment of cancer cells with plant extracts in. The flow cytometry assay showed apoptosis induced after P.Oleracea extract treatment in PANC- one cancer cell line. Also, microscopic observation is in agreement with flow cytometry and MTT assay. Results of the current study indicated that P.Oleracea extracts significantly induce apoptosis by regulating P53 and CDK expression, consequently. Therefore, P.Oleracea may be considered as a novel finding for pancreatic cancer treatment consequently of its cytotoxic and apoptotic activity.
Effect of arsenic trioxide in blocking the cell cycle of human prostate carcinoma PC-3 cells

Abstract:Aim: To investigate the effects of arsenic trioxide (As 2O 3) in inhibiting the growth of human prostate cancer(PCA) PC-3 cells and blocking the cell cycle of PCA PC-3 cells. Methods: The experiment was completed in the central laboratory of Tangdu Hospital, Fourth Military Medical University of Chinese PLA from October 2003 to May 2004. The trial for the inhibition of cell growth in vitro (MTT colorimetry) was conducted to study the effect of As 2O 3 on the growth of cells. Then, flow cytometric analysis was performed to assay the distribution of cell cycle. Western blotting was performed to investigate the changes of cyclin dependent kinases(CDKs), cyclin dependent kinase inhibitors (CDKIs) and cyclin. Results: A significant dose- and time-dependent growth inhibition was observed in As 2O 3-treated cells. From the results of MTT assay, the absorbance was significantly different among the groups after treated with As 2O 3 at 1, 2 and 5 μmol/L for 1 to 6 days (F = 22.220, P < 0.001). As shown in the assessment of cell cycle by flow cytometry, the gathering cells at G1 phase were increased [(51.8 ± 2.4)% to (58.8 ± 2.3)%] with the concentration of As 2O 3 (1, 2 and 5 μmol/L) at 72 hours after treatment, and there were significant, differences among the groups(χ 2 = 15.846, P = 0.01), indicating that As 2O 3 can block the PC-3 cells at G1 phase. As 2O 3 strongly increased the expression of PC-3 CDKIs, Cip1/p21 and Kip1/p27, in a dose-dependent manner, as well as downregulated CDK2, 6 and cyclin E, A. Conclusion: As 2O 3 inhibits cell cycle progression of human PAC PC-3 cells and inhibits their growth, so it needs more study to provide a basis for the application of As 2O 3 in the treatment of PCA.

Oxaliplatin and Checkpoint Inhibitor Induces Immunogenic Cells Death and Promotes Therapeutic Efficacy in the Model of Murine Triple Positive Breast Cancer

Abstract:Various damage-associated molecular patterns (DAMPs) associated with immunogenic cell death (ICD) have been discovered, potentially leading to cancer cell elimination. Certain platinum-based compounds can trigger both cancer cell apoptosis and ICD. This study aims to investigate the effect of the therapy of anti- PDL1 with Oxaliplatin by increasing amount and increasing treatment duration of anti-PDL1 with Oxaliplatin in SK-Br-3, both in vitro and in vivo conditions. The study will use HER-2 (3+) breast cancer cell line, SKBr3. The cells will be treated with increasing concentrations of Oxaliplatin with anti-PDL1 for different durations. In vitro death of cancer cells will be measured by MTT assay, HMGB1 will be measured by western blot. Additionally, ATP release will be measured, mice will be injected with SK-Br-3 and treated with the combo therapy of anti-PDL1 with Oxaliplatin, and in vivo tumor growth will be recorded weekly for xenograft. The positive control for the experiments is cisplatin, and the negative control is IgG solution instead of aPDL1 and Oxaliplatin in PBS.There are three main possible results: (1) The combo therapy of Oxaliplatin with anti-PDL1 induces robust ICD in HER-2 triple positive breast cancer cells. (2) The combo therapy of Oxaliplatin with anti-PDL1 act as a stimulant for robust ICD in HER-2(3+) positive breast cancer cells. (3) The combo-therapy of Oxaliplatin with anti-PDL1 has no significant effect on inducing robust ICD in HER-2(3+) positive breast cancer cells. The result of the study will provide important insight into the preclinical effectiveness of Oxaliplatin with anti-PDL1 in treating HER-2 (3+) breast cancer, and it also sets the basis for future clinical studies of the drug. Future studies should focus on investigating the mechanism underlying Oxaliplatin with anti-PDL1 effectiveness in SK-Br-3.
Evaluation of the Cytotoxic Effect of Pd2Spm against Prostate Cancer through Vibrational Microspectroscopies

Abstract:Regarding the development of new antineoplastic agents, with a view to assess the selective antitumoral potential which aims at causing irreversible damage to cancer cells while preserving the integrity of their healthy counterparts, it is essential to evaluate the cytotoxic effects in both healthy and malignant human cell lines. In this study, a complex with two Pd(II) centers linked by the biogenic polyamine spermine (Pd2Spm) was tested on healthy (PNT-2) and cancer (LNCaP and PC-3) prostate human cell lines, using cisplatin as a reference. To understand the mechanisms of action of both cisplatin and Pd2Spm at a molecular level, Fourier Transform Infrared (FTIR) and Raman microspectroscopies were used. Principal component analysis was applied to the vibrational data, revealing the major metabolic changes caused by each drug, which were found to rely on DNA, lipids, and proteins, acting as biomarkers of drug impact. The main changes were observed between the B-DNA native conformation and either Z-DNA or A-DNA, with a higher effect on lipids having been detected in the presence of cisplatin as compared to Pd2Spm. In turn, the Pd-agent showed a more significant impact on proteins.

Abstract:Androgen deprivation therapy (ADT) is the primary treatment for advanced prostate cancer (PCa). However, prolonged ADT inevitably results in therapy resistance with the emergence of the castration-resistant PCa phenotype (CRPC). Hence, there is an urgent need to explore new treatment options capable of delaying PCa progression. Hispidin (HPD) is a natural polyketide primarily derived from plants and fungi. HPD has been shown to have a diverse pharmacological profile, exhibiting anti-inflammatory, antiviral, cardiovascular and neuro-protective activities. However, there is currently no research regarding its properties in the context of PCa treatment. This research article seeks to evaluate the anti-cancer effect of HPD and determine the underlying molecular basis in both androgen-sensitive PCa and CRPC cells. Cell growth, migration, and invasion assays were performed via the MTS method, a wound healing assay and the transwell method. To investigate if HPD affected the expression of proteins, Western blot analysis was conducted. Furthermore, apoptosis was assessed by Annexin V-FITC/PI staining and Western blot analyses. HPD exhibited a favorable pharmaceutical profile to inhibit cell growth; disrupt the cell cycle; attenuate wound healing, migration and invasion; and induce apoptosis in PCa cells in vitro. The mechanistic results demonstrated that HPD reduced AR, MMP-2 and MMP-9 expression and activated the caspase-related pathway, leading to programmed cell death in PCa cells. We showed the anti-cancer effect of HPD on PCa cells and confirmed its feasibility as a novel therapeutic agent. This study provides significant insights into the delineation of the molecular mechanism of HPD in PCa cells and the development of an effective and safe therapy using HPD to eliminate PCa progression.

Binuclear [{Pd (dien)}2(μ‐pz)](ClO4)4 (Pd1) and [{Pd (dien)}2(μ‐pydz)](ClO4)4 (Pd2) complexes, where pz is 1,4‐diazine and pydz is 1,2‐diazine bridging ligands and dien (diethylenetriamine) is a tridentately coordinated triamine ligand, were synthesized and characterized using elemental analysis, NMR (1H and 13C) spectroscopy, and UV–Vis spectrophotometry. Electronic absorption spectrophotometry and fluorescence spectroscopy were used to study the interactions of the prepared Pd1 and Pd2 complexes with deoxyribonucleic acid (CT‐DNA) and bovine serum albumin (BSA). According to the UV–Vis spectrophotometric data, the complexes interact with CT‐DNA and bind to the BSA as a possible carrier to the target site. The antitumor properties of the tested palladium compounds were evaluated on three types of human cancer cells, namely, squamous cell carcinoma cells SCC‐4, cervical carcinoma cells HeLa and colorectal carcinoma cells HCT116, and one type of malignantly unchanged cells, MRC‐5 fibroblasts. The highest cytotoxicity was shown by the Pd1 compound against SCC‐4 cell, with an IC50 value of 5.4 ± 1.2 μM. The results showed that the cytotoxic potential against SCC‐4 cells of our new palladium complexes Pd1 and Pd2 was comparable with the cytotoxic potential of cisplatin and oxaliplatin. However, Pd1 and Pd2 displayed significantly lower cytotoxicity in comparison with cisplatin and oxaliplatin against non‐malignant cells, fibroblasts MRC5, so these results may suggest that local and systemic complications of antitumor therapy could be reduced and/or avoided. Both compounds selectively decreased the viability of SCC‐4 cells by induction of apoptosis. Also, Pd1 and Pd2 induced S‐phase cell cycle arrest in human SCC‐4 cells, so that could be another mechanism to halt the uncontrolled growth and division of cancer cells. Moreover, the precise molecular mechanism of Pd1 and Pd2 complexes‐induced apoptosis was the activation of the intrinsic signaling pathway of apoptosis, which also included activation of caspase‐3. Therefore, our new palladium (II) complexes might have a promising role as the potential future cytostatic agents.

IND-2, a Quinoline Derivative, Inhibits the Proliferation of Prostate Cancer Cells by Inducing Oxidative Stress, Apoptosis and Inhibiting Topoisomerase II

Abstract:In men, prostate cancer (PC) is the most frequently diagnosed cancer, causing an estimated 375,000 deaths globally. Currently, existing therapies for the treatment of PC, notably metastatic cases, have limited efficacy due to drug resistance and problematic adverse effects. Therefore, it is imperative to discover and develop novel drugs for treating PC that are efficacious and do not produce intolerable adverse or toxic effects. Condensed quinolines are naturally occurring anticancer compounds. In this study, we determined the in vitro efficacy of IND-2 (4-chloro-2-methylpyrimido[1″,2″:1,5]pyrazolo[3,4-b]quinolone) in the PC lines, PC-3 and DU-145. IND-2 significantly inhibited the proliferation of PC-3 and DU-145, with IC50 values of 3 µM and 3.5 µM, respectively. The incubation of PC-3 cells with 5 and 10 µM of IND-2 caused the loss of the mitochondrial membrane potential in PC-3 cells. Furthermore, IND-2, at 5 µM, increased the expression of cleaved caspase-3, cleaved caspase-7 and cleaved poly (ADP-ribose) polymerase (PARP). The incubation of PC-3 cells with 5 µM of IND-2 significantly decreased the expression of the apoptotic protein, B-cell lymphoma 2 (Bcl-2). Furthermore, 5 and 10 µM of IND-2 produced morphological changes in PC-3 cells characteristic of apoptosis. Interestingly, IND-2 (2.5, 5 and 10 µM) also induced mitotic catastrophe in PC-3 cells, characterized by the accumulation of multinuclei. The incubation of DU-145 cells with 1.25 and 5 μM of IND-2 significantly increased the levels of reactive oxygen species (ROS). Finally, IND-2, at 10 μM, inhibited the catalytic activity of topoisomerase IIα. Overall, our findings suggest that IND-2 could be a potential lead compound for the development of more efficacious compounds for the treatment of PC.
Enhancing Effects of Olaparib by Alpha- and Beta-Emitting Radionuclides, X-Rays, and Ultraviolet A Light in Combination with Ortho-IodoHoechst in a Prostate Cancer Cell Model

Abstract:Background: New therapeutic strategies for metastatic castration-resistant prostate cancer (mCRPC) have been developed in the past to achieve the best response rates. Most recently, the use of combination therapies has been explored to optimize patient outcomes. Poly(ADP-ribose) polymerase inhibitors (PARPi) may help to treat mCRPC more effectively. Objectives: This study aimed to determine whether the combination of a PARPi with different radiation qualities results in different levels of radiosensitization of PC-3 cells. Methods: The radiosensitizing potential of Olaparib in combination with 177Lu, 223Ra, X-rays and photodynamic therapy (PDT) using the UVA light-activated photosensitizer ortho-iodoHoechst33258 (oIH) was evaluated by determining the clonogenic survival, DNA damage and cell cycle analysis. Results: Here, we show that this combination strategy differentially sensitized PC-3 cells to different radiation qualities. The combination of 177Lu with Olaparib increased the numbers of persistent double-strand breaks (DSBs) by a factor of 3.3 and cell death in PC-3 cells. Overall, the β-emitter 177Lu indicated a higher radiosensitization efficacy compared to 223Ra, with X-rays corresponding to dose modification factors (DMF) of 1.77, 1.17 and 1.16 respectively. Even in the case of the α-emitter 223Ra, the effects were much less pronounced than for 177Lu. PARPi also showed a slight potentiation of the cytotoxic effects both in co-treatment with X-rays and with PDT. Conclusions: The results of our study indicate a potential role for Olaparib in further optimizing the PSMA radioligand therapy (PRLT) outcomes. However, further evaluation of the combination of PARPi with PRLT is needed to gain more insights into improving the benefit to patients suffering from mCRPC.

Abstract:Prostate cancer (PCa) is the second leading cause of cancer death among men in the United States. Surgery, radiation therapy, chemotherapy, and androgen deprivation therapy are currently the standard treatment options for PCa. These have poor outcomes and result in the development of castration-resistant prostate cancer (CRPC), which is the foremost underlying cause of mortality associated with PCa. Taxanes, diterpene compounds approved to treat hormonal refractory PCa, show poor outcomes in CRPC. Polygodial (PG) is a natural sesquiterpene isolated from water pepper (Persicaria hydropiper), Dorrigo pepper (Tasmannia stipitata), and mountain pepper (Tasmannia lanceolata). Previous reports show that PG has an anticancer effect. Our results show that PG robustly inhibits the cell viability, colony formation, and migration of taxane-resistant CRPC cell lines and induces cell cycle arrest at the G0 phase. A toxicity investigation shows that PG is not toxic to primary human hepatocytes, 3T3-J2 fibroblast co-cultures, and non-cancerous BPH-1 cells, implicating that PG is innocuous to healthy cells. In addition, PG induces oxidative stress and activates apoptosis in drug-resistant PCa cell lines. Our mechanistic evaluation by a proteome profiler–human apoptotic array in PC3-TXR cells shows that PG induces upregulation of cytochrome c and caspase-3 and downregulation of antiapoptotic markers. Western blot analysis reveals that PG activates apoptotic and DNA damage markers in PCa cells. Our results suggest that PG exhibits its anticancer effect by promoting reactive oxygen species generation and induction of apoptosis in CRPC cells.
