Abstract:Marrubium friwaldskyanum Boiss (Lamiaceae) is a Bulgarian endemic species. Overall, the essential oil (EO) composition of M. friwaldskyanum was different from that of the other Marrubium species reported in the literature. The main EO constituents of M. friwaldskyanum were (E)-caryophyllene, germacrene D, and caryophyllene oxide. The effect of the harvest stage was significant only on α-copaene, (E)-caryophyllene, caryophyllene oxide, and τ-muurolol. The concentration of α-copaene (1.26–1.83% range of the total oil), (E)-caryophyllene (31–41%), caryophyllene oxide (6.4–11.8%), and τ-muurolol (1.3–2.8%) were the highest at 2–3 pair of leaves or before flowering and lower at flowering. The harvest stage did not significantly affect the concentrations of the other six identified EO compounds β-bourbonene (1.1%), α-humulene (2.8%), germacrene D (23.3%), bicyclogermacrene (2.85%), δ-cadinene (1.1%), and spathulenol (2.8%). In a separate experiment, grinding of the biomass prior to EO extraction had a significant effect only on the concentrations of D-limonene (0.24–3.3%) and bicyclogermacrene (3.6–9.1%). Grinding in water or without water, maceration, and addition of Tween®20 had rather small effects on the EO profile. The identified EO constituents and their mean concentrations in this experiment were (E)-caryophyllene (25.4%), germacrene D (17.6%), caryophyllene oxide (9.1%), spathulenol (6.5%), τ-muurolol (5.0%), carvacrol (3.9%), α-copaene (2.5%), β-bourbonene (2.5%), δ-cadinene (2.4%), α-humulene (1.8%), and Z-β-farnesene (1.3%). Embryological studies observed anther and the development of the male gametophyte and ovule and development of the female gametophyte of M. friwaldskyanum. Furthermore, pollen and seed viability assays were conducted, and mass spectrometry-based metabolomics analysis of an extract from shoots revealed the presence of 45 natural products, identified as flavonoids, phenolic acids, and (tri)terpenoids. Overall, the phytochemistry and some of the microscopic analyses distinguished this endemic species from other species in Marrubium.

Abstract:The present study was conducted to evaluate the antioxidant and antibacterial activity of the extract of leaves and flowering of Marrubium vulgare L. The quantification of total phenols was determined using the Folin-Ciocalteu reagent, it is 2.23 mg EAG/g DM. The identification of flavonoids by the Alcl3 method, revealed the presence of anthocyanins, C-glycosides and aglycones, whose contents are 1.266mg/g, 0.385mg/g and 0.0331 mg/g, respectively. The antioxidant activity of the compounds in the methanolic extract was evaluated in vitro through two tests. In the case of DPPH test, the IC50 are 0.55, 0.97, and 1.2, 0.033 mg/ml for anthocyanins, c-glycosides, polyphenols and aglycones respectively. For FRAP assay, at a concentration of 1 mg/ml, the reducing power of iron manifested ODs of 1.389 nm; 0.752 nm, 0.496 and 0.114 nm for anthocyanins, C-glycosides, total polyphenols and aglycones respectively. The results show an excellent correlation between the two tests used, and the contents of anthocyanins and c-glycosides. The antimicrobial activity was evaluated by the agar diffusion method against (ATCC 25923), (ATCC 25922). The antibiogram shows that anthocyanins have a very Staphylococcus aureus Escherichia coli important antibacterial activity on with an inhibition zone of 16.5 mm and a diameter of 13.5 for Anthocyanins show S. aureus Escherichia coli. the lowest MIC values against the two strains and (MIC=1/16 and 1/8 respectively). C-glycosides also Staphylococcus aureus Escherichia coli have an inhibitory effect on the growth of and particularly on (MIC= 1/32 and 1/8 respectively) which is Escherichia coli Staphylococcus aureus a sensitive bacterium (gram+). Anthocyanins have marked a strong sensitivity towards the two strains. In conclusion, this work presents the compounds of L as novel antimicrobial agents and as a potential source for combating bacteria, which are increasingly Marrubium vulgare resistant to antibiotics

Abstract:Marrubium vulgare L. has a long history of use in traditional herbal medicine for the treatment of respiratory tract infections, inflammatory conditions, and pain. This study aimed to investigate the chemical composition, acute toxicity, and antinociceptive effects of the aqueous extract from M. vulgare leaves (AEMV). Antioxidant activity was evaluated using DPPH and reducing power assays. The chemical composition of AEMV was determined through LC‐MS/MS, and the levels of total phenolics, flavonoids, and condensed tannins were quantified. Acute oral toxicity was assessed in male Swiss mice with a single oral dose of AEMV (1, 2, 5 g/kg). The analgesic impact was examined through writhing, hot plate, and formalin tests. Our findings not only confirmed the safety of the extract in animal models but also revealed significant antioxidant activity in AEMV. High‐performance liquid chromatography (HPLC) analysis identified important bioactive compounds, with marrubiin being a major component. Furthermore, AEMV demonstrated robust antinociceptive properties in all conducted tests, highlighting its potential as a valuable natural source of bioactive compounds suitable for a wide range of therapeutic applications.

A seroepizootiologic study of five viruses in a swine-evaluation station.

Abstract:This serologic study was done to gain information on the spread, maintenance, and effect upon performance of five porcine viruses. Blood samples were taken from two groups of 8- to 11-week-old pigs from a large number of Indiana swine herds in a performance-testing station 1 week after entry, 7 weeks after entry (one group only), and at slaughter. The sera were tested by indirect fluorescent antibody tests for antibodies to transmissible gastroenteritis virus (TGEV), swine influenza virus (SIV), hemagglutinating encephalomyelitis virus (HEV), porcine adenovirus (PAV), and pseudorabies virus (PRV). Seroconversions to TGEV, HEV, and PAV occurred in a group of pigs entered in May and slaughtered in August (group 1). In the group that was entered in October and slaughtered in January (group 2), pigs developed antibodies to SIV, HEV, and PAV, but not to TGEV. Only 1 of the 434 pigs tested had antibodies to PRV, and there were no seroconversions to this virus. The only statistically valid effect of infection on performance was found in group 1 pigs, which had seroconverted to TGEV during the first 7 weeks of their stay. These pigs gained 0.077 kg less per day than pigs that did not develop antibodies to TGEV during that period. The pattern of serologic reactions was indicative of a relatively slow spread of these viruses in the groups. We interpret this as supporting the concept that a relatively slow spread of these viruses through large groups of pigs kept under conditions that are less than optimum for virus spread may be an important means of their interepizootic survival.

Abstract:Extracts from medicinal plants are widely used in the treatment and prevention of different diseases. Micromeria frivaldszkyana is a Balkan endemic species with reported antioxidant and antimicrobial characteristics; however, its phytochemical composition is not well defined. Here, we examined the metabolome of M. frivaldszkyana by chromatography–mass spectrometry (GC-MS), ultra-performance liquid chromatography–mass spectrometry (UPLC-MS-MS), and inductively coupled plasma mass spectrometry (ICP-MS). Amino acids, organic acids, sugars, and sugar alcohols were the primary metabolites with the highest levels in the plant extract. Detailed analysis of the sugar content identified high levels of sucrose, glucose, mannose, and fructose. Lipids are primary plant metabolites, and the analysis revealed triacylglycerols as the most abundant lipid group. Potassium (K), magnesium (Mg), zinc (Zn), and calcium (Ca) were the elements with the highest content. The results showed linarin, 3-caffeoil-quinic acid, and rosmarinic acid, as well as a number of polyphenols, as the most abundant secondary metabolites. Among the flavonoids and polyphenols with a high presence were eupatorin, kaempferol, and apigenin—compounds widely known for their bioactive properties. Further, the acute toxicity and potential anti-inflammatory activity of the methanolic extract were evaluated in Wistar rats. No toxic effects were registered after a single oral application of the extract in doses of between 200 and 5000 mg/kg bw. A fourteen-day pre-treatment with methanolic extract of M. frivaldszkyana in doses of 250, 400, and 500 mg/kg bw induced anti-inflammatory activity in the 1st, 2nd, and 3rd hours after carrageenan injection in a model of rat paw edema. This effect was also present in the 4th hour only in the group treated with a dose of 500 mg/kg. In conclusion, M. frivaldszkyana extract is particularly rich in linarin, rosmarinic acid, and flavonoids (eupatorin, kaempferol, and apigenin). Its methanolic extract induced no toxicity in male Wistar rats after oral application in doses of up to 5000 mg/kg bw. Additionally, treatment with the methanolic extract for 14 days revealed anti-inflammatory potential in a model of rat paw edema on the 1st, 2nd, and 3rd hours after the carrageenan injection. These results show the anti-inflammatory potential of the plant, which might be considered for further exploration and eventual application as a phytotherapeutic agent.

Abstract:Consider the medicinal usefulness and ethnomedicinal uses referring to Marrubium vulgare; the CH 3 OH extract of whole plant was investigated for the antiproliferative, effect and toxicological screening. Its antiproliferative effect was evaluated against A549 (lung), HepG2 (Liver), MDA-MB-231 (breast) and MCF-7 cancer cell-lines. Doxorubicin was used as a positive control. Toxicological evaluation of the Marrubium vulgare methanol extract (MVM) was carried out in zebrafish embryos. The MVM showed more effective inhibition against MCF-7and MDA-MB-231 breast-cancer cells. The IC 50 value ranged from 43.5 to 223.5 μg/mL. In term of IC 50 MVM showed a strong antiproliferative effect averse to MDA-MB-231 and MCF-7 in terms of IC 50 values (43.5 and 46.5 μg/mL respectively). Lethal concentration of MVM in zebrafish embryos remained 5 μg/ml. The results obtained from zebrafish embryos tests indicates that the extract is toxic at some level and should be used carefully or avoid it during the pregnancy.

Exploring the Antibacterial Potential of Lamiaceae Plant Extracts: Inhibition of Bacterial Growth, Adhesion, Invasion, and Biofilm Formation and Degradation in Pseudomonas aeruginosa PAO1

Abstract:In response to the global rise in antibiotic resistance and the prevalence of bacterial biofilm-related infections, the antibacterial efficacy of methanolic, ethanolic, and aqueous extracts of 18 Lamiaceae plants from Serbia was evaluated. The total coumarins and triterpenes were detected spectrophotometrically, while a microdilution assay measured their effects on bacterial growth. Additionally, the impact of these extracts was assessed on Pseudomonas aeruginosa PAO1 adhesion and invasion in human fibroblasts and biofilm formation and degradation. The alcoholic extracts had the highest phytochemical content, with Teucrium montanum and Lavandula angustifolia being the richest in coumarins and triterpenes, respectively. Gram-positive bacteria, particularly Bacillus subtilis, were more susceptible to the extracts. Hyssopus officinalis ethanolic and Sideritis scardica methanolic extracts inhibited bacterial growth the most efficiently. Although the extracts did not inhibit bacterial adhesion, most ethanolic extracts significantly reduced bacterial invasion. Origanum vulgare and H. officinalis ethanolic extracts significantly inhibited biofilm formation, while Teucrium chamaedrys extract was the most active in biofilm degradation. This study significantly contributes to the literature by examining the antibacterial activity of Lamiaceae extracts, addressing major literature gaps, and underscoring their antibacterial potential, particularly Satureja montana and O. vulgare ethanolic extracts, linking their efficacy to coumarins and triterpenes.

Abstract:This study evaluated the in vitro antineoplastic and antiviral potential and in vivo toxicity of twelve extracts with different polarity obtained from the herbaceous perennial plant Geum urbanum L. (Rosaceae). In vitro cytotoxicity was determined by ISO 10993-5/2009 on bladder cancer, (T-24 and BC-3C), liver carcinoma (HEP-G2) and normal embryonic kidney (HEK-293) cell lines. The antineoplastic activity was elucidated through assays of cell clonogenicity, apoptosis induction, nuclear factor kappa B p65 (NFκB p65) activation and total glutathione levels. Neutral red uptake study was applied for antiviral activity. The most promising G. urbanum extract was analyzed by UHPLC-HRMS. The acute in vivo toxicity analysis was carried out following OEDC 423. The ethyl acetate extract of aerial parts (EtOAc-AP) exhibited the strongest antineoplastic activity on bladder cancer cell lines (IC50 = 21.33-25.28 µg/mL) by inducing apoptosis and inhibiting NFκB p65 and cell clonogenicity. EtOAc and n-butanol extracts showed moderate antiviral activity against human adenovirus type 5 and human simplex virus type I. Seventy four secondary metabolites (gallic and ellagic acid derivatives, phenolic acids, flavonoids, etc.) were identified in EtOAc-AP by UHPLC-HRMS. This extract induced no signs of acute toxicity in liver and kidney specimens of H-albino mice in doses up to 210 mg/kg. In conclusion, our study contributes substantially to the detailed pharmacological characterization of G. urbanum, thus helping the development of health-promoting phytopreparations.
The Action Mechanisms, Anti-Cancer and Antibiotic-Modulation Potential of Vaccinium myrtillus L. Extract

Abstract:Background: Herbal medicinal products containing Vaccinium myrtillus L. (bilberry) fruits and fruit extracts are widely available in the market. Although bilberry leaves and stems are considered as bio-waste, they contain much higher levels of phenolic compounds than fruits. The study aimed to investigate the antimicrobial and anticancer potential of aerial part extracts from Vaccinium myrtillus L. (V. myrtillus, VM) plants harvested at high altitudes in Armenian landscape and characterize the bioactive phytochemicals. Material and methods: For evaluation of antioxidant properties, chemical-based tests (total phenolic and flavonoid content, and antiradical activity in 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) tests) and cellular antioxidant activity (CAA) assay were applied. Genotoxicity and anticancer properties of the extract alone and in combination with fluorouracil were explored in human cancer and normal cell lines. Antibacterial properties of V. myrtillus extract alone and in combination with antibiotics, as well as their effect on proton-flux rate through cell membrane were explored on bacterial strains. The characterization of active phytochemicals was done using Liquid Chromatography-Quadrupole-Orbitrap High-Resolution Mass Spectrometry (LC-Q-Orbitrap HRMS). Results: The V. myrtillus aerial part extract demonstrated promising antioxidant properties in all tests. The selective cytotoxic activity was documented against various cancer cell lines (human colon adenocarcinoma (HT29), human breast cancer (MCF-7) and human cervical carcinoma (HeLa)), while it did not inhibit the growth of tested human normal primary renal mixed epithelial cells (HREC) even at 10-fold higher concentrations. The extract did not have genotoxic properties in comet assay making it a potential source for the development of anticancer preparations. The investigated extract did not directly inhibit the growth of Escherichia coli (E. coli) and Salmonella typhimurium (S. typhimurium) strains at up to 1 mg/mL concentration. However, V. myrtillus extract enhanced the kanamycin intake and increased its efficiency against E. coli strain. The phytochemical characterization of the extract showed the presence of different groups of phenolics. Conclusions: Based on obtained data, we suggest the aerial parts of the V. myrtillus plant as an alternative source of bioactive natural products for food supplements, nutraceuticals, functional foods and medicine.

Abstract:Boerhavia diffusa is a great tropical plant and is widely used for various traditional purposes. In the present study, we examined the influence of solvents (dichloromethane, ethyl acetate, methanol and infusion (water)) on chemical composition and biological capabilities of B. diffusa. An UHPLC-HRMS method was used to determine the chemical characterization. The biological ability was examined for antioxidant, enzyme inhibitory and anti-cancer effects. To evaluate antioxidant effects, different chemical methods (ABTS, DPPH, CUPRAC, FRAP, metal chelating and phosphomolybdenum) were applied. With regard to enzyme inhibitory properties, cholinesterases, amylase, glucosidase and tyrosinase were used. The MDA-MB-231 breast cancer cell line was chosen to determine anticancer activity. Based on the UHPLC-HRMS analysis, 37 specialized metabolites were dereplicated and identified in the studied extracts. Results revealed the presence of 15 hydroxybenzoic, hydroxycinnamic, acylquinic acids, and their glycosides, one rotenoid, seven flavonoids, 12 fatty acids and two other glycosides. Among the tested extracts, the methanol extract showed a stronger antioxidant ability compared with other extracts. The methanol extract also showed the best inhibitory effects on tyrosinase and glucosidase. In the anti-cancer evaluation, the methanol extract showed stronger anticancer effects compared with water extract. In summary, our observations can contribute to the establishment of B. diffusa as a potential candidate for functional applications in the preparation.

Antimycobacterial and Anticancer Properties of Myrtus communis Leaf Extract

Abstract:Background: Plant-derived products or extracts are widely used in folk/traditional medicine to treat several infections, ailments, or disorders. A well-known medicinal herb, Myrtus communis is an evergreen fragrant plant native to the Mediterranean region that has been used for ages in traditional medicine around the world. Materials and methods: The microplate alamarBlue assay and the well diffusion method were used to evaluate the zone of inhibition and MIC, respectively. The double-disc diffusion method was used to investigate the synergy between antibiotics and the extract. The crystal violet method was used to investigate biofilm development. The SulphoRhodamine-B assay and DNA flow cytometry were used to investigate the proliferation and subsequent distribution of cells among different phases of the cell cycle. The apoptotic and necrotic phases of the cancer cells were examined using flow cytometry in conjunction with Annexin V-FITC/PI labeling. Using the IBM SPSS statistical program, a one-way ANOVA with Tukey's post hoc test was employed for statistical analysis. Results: The ethanolic leaf extract of M. communis showed a strong growth inhibition effect (zone of inhibition: 20.3 ± 1.1-26.3 ± 2.5 mm, MIC: 4.88-312.5 µg/mL, and MBC: 39.07-1250 μg/mL) against several rapidly growing and slow-growing mycobacterial strains in a dose-dependent manner. Damage to the cell wall of bacterial cells was determined to be the cause of the antimycobacterial action. The extract inhibited biofilm formation (MBIC of 9.7 µg/mL) and eradicated already-formed mature and ultra-mature biofilms of M. smegmatis, with MBEC values of 78 µg/mL and 156 µg/mL, respectively. Additionally, the extract exhibited potent anticancer effects against diverse cancer cell lines of the breast (MCF-7), liver (HepG2), cervix (HeLa), and colon (HCT116) (IC50 for HCT116: 83 ± 2.5, HepG2: 53.3 ± 0.6, MCF-7: 41.5 ± 0.6, and HeLa: 33.3 ± 3.6) by apoptosis after arresting the cells in the G1 phase of the cell cycle. Conclusions: These results suggest that M. communis leaf extract is a potential source of secondary metabolites that could be further developed as potential anticancer and antimycobacterial agents to treat diverse types of cancers and mycobacterial infections.

Abstract:Marrubium rotundifolium Boiss. is an endemic plant distributed in Aegean Region of Türkiye. It's traditionally used in Anatolian medicine for treatment of cold and flu, dyspepsia, costiveness and intestinal spasms. However, phytochemical studies on the plant are limited. In the present study, five phenolic glycosides 4',5,6,7-tetramethoxy scutellarein (1) , apigenin-7- O- (3"- O-E-p -coumaroyl)-β- d -glucopyranoside (2 ), tiliroside (3) , 4-(β- d -glucopyranosyloxy) benzoic acid (4) , and astragalin (5) were isolated from the aerial parts of M. rotundifolium and their structures were elucidated on the basis of spectroscopic methods (1D and 2D NMR, and MALDI-TOF/MS). Moreover, anti-inflammatory activities of the isolated compounds were evaluated by measurements of interleukins (IL-1β, IL-6) and tumour necrosis factor-alpha (TNF-α) levels. Tiliroside exhibited the highest potency in all pathways. At concentrations of 3 and 6 μg/mL, it significantly decreased the levels of pro-inflammatory cytokines IL-1β (192.53 and 175.54 pg/mL), IL-6 (925.52 and 946.81 pg/mL), and TNF- α (6465.86 and 6267.67 pg/mL). This is the first report on phenolic profile of endemic M. rotundifolium . The anti-inflammatory and antidiabetic activity potential of the plant should be investigated in further studies.

Abstract:Marrubium astracanicum Jacq., Lamiaceae, is an herbaceous plant distributed in Iran, Turkey, and southern Caucasia. In the present study, α-glucosidase inhibitory principles of M. astracanicum aerial parts were investigated by a bioassay-guided approach. The extraction procedure from the plant aerial parts was carried out successively using petroleum ether, ethyl acetate, and methanol. The ethyl acetate extract, with the highest in vitro α-glucosidase inhibitory activity, was subjected to further chemical investigation. The structures of the isolated compounds were elucidated by 1 H-NMR and 13 C-NMR spectral analyses as apigenin-7- O -(3′′-( E ) -p -coumaroyl)-β- d -glucoside, apigenin-7- O -(6′′-( E )- p -coumaroyl)-β- d -glucoside, martynoside, apigenin-7- O -β- d -glucoside, and verbascoside. In addition, the antidiabetic potential of the isolated compounds was evaluated in an in vitro α-glucosidase inhibitory assay, and kinetic and molecular docking studies were done for the most active compound. Apigenin-7- O -(3′′-( E ) -p -coumaroyl)-β- d -glucoside and apigenin-7- O -(6′′-( E )- p -coumaroyl)-β- d -glucoside showed the most potent in vitro α-glucosidase inhibitory activity with IC 50 values of 57.4 ± 1.2 and 186.4 ± 2.5 μM, respectively, which was about 4 to 13 times lower than the IC 50 value of the reference compound acarbose (751.2 ± 0.4 μM).
Exploring the anti-cancer properties of essential oils from some Lamiaceae species against human cancer cells with multivariate analysis

Abstract:The present study was aimed to determine phytochemical characterization, anticancer and antioxidant activities of essential oils obtained from 11 Lamiaceae species, including Clinopodium serpyllifolium subsp . barbatum (P.H.Davis) Bräuchler, Lavandula angustifolia Miller, Mentha × piperita L., Mentha pulegium L. , Ocimum basilicum L., Rosmarinus officinalis L., Salvia aramiensis Rech.f., Salvia fruticosa Miller, Salvia officinalis L., Satureja thymbra L., and Thymbra spicata L. taxa from Türkiye. The chemical composition of the essential oils (EOs) was identified using GC–MS, and antioxidant activity was determined using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′- azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) free radical scavenging methods. Furthermore, the anticancer potential of the EOs was evaluated using MTT method against human cell lines, including hepatocellular carcinoma (HepG2), gastric carcinoma (NCI-N87), breast adenocarcinoma (MCF-7), human prostate carcinoma (LNCaP clone FGC-Luc2) cancer cells, and non-cancerous human umbilical vein endothelial cells (HUVECs). The antioxidant activities of the extracts varied from 31.96 to 74.96%. Consistent with antioxidant activities, the anticancer activities of the EOs of S. officinalis, L. angustifolia , and R. officinalis were significantly higher than others. However, almost all EOs were found to inhibit cell viability and induce apoptosis of cancer cells, while the EOs at different concentrations exhibited the highest anticancer activity against NCI-N87, followed by HepG2, MCF-7, and LNCaP cancer cells, with the IC 50 value ranging from 10.98 ± 0.12 to 78.08 ± 1.21 μg/mL, respectively. Variability in phytochemical components, free radical scavenging and anticancer activities of EOs was revealed by principal component analysis (PCA) and agglomerative hierarchical clustering (AHC). The major components of the EOs distilled from Lamiaceae species were found as eucalyptol, thujone, linalool, linalyl acetate, pulegone, carvone and carvacrol at different concentrations in the EOs. Overall, it can be clearly concluded that the EO samples distilled from some medicinal plants belonging to the Lamiaceae family contain valuable phytochemical compounds and accordingly exhibit remarkable biological activities.

Abstract:The objective of this study was to assess the biological activities of Anthemis cotula L. and explore its potential therapeutic effects on A549 lung cancer cells. Within this particular framework, the aerial components of the plant were collected and subjected to extraction using a Soxhlet apparatus to obtain extracts in the form of water, ethanol, and methanol. The determination of the phenolic content of the samples was conducted using the Folin-Ciocalteu reagent, while the quantification of flavonoid content was conducted via an aluminum chloride assay, and the measurement of the plant's total antioxidant, total oxidant, and oxidative stress index was conducted using Rel Assay kits. The antimicrobial properties of the substances were examined via the agar dilution technique. The MTT assay was employed to determine the antiproliferative activity against A549 lung cancer cells, and the LC-MS/MS instrument was utilised to screen for the presence of standard compounds. The following compounds were identified in the study: acetohydroxamic acid, catechin hydrate, vanillic acid, resveratrol, kaempferol, fumaric acid, gallic acid, protocatechuic acid, 4-hydroxybenzoic acid, caffeic acid, salicylic acid, 2-hydroxycinnamic acid, naringenin, quercetin, luteolin, and alizarin. Plant extracts were found to be effective against microorganisms in the range of 25 to 200 μg/mL; furthermore, the water extract exhibited the highest total antioxidant status value of 8.201 ± 0.113 μmol/L. Similarly, the ethanol extract demonstrated the highest total oxidant status value of 11.247 ± 0.178 μmol/L, while, lastly, the methanol extract displayed the highest oxidative stress index value of 0.151 ± 0.001. Strong antiproliferative activity was observed in all the plant extracts, and the ethanol extract with the highest concentration exhibited a total phenolic content of 61.78 ± 4.38 mg/g. The ethanol extract exhibited the highest recorded total flavonoid content, measuring 66.84 ± 3.13 mg/g. In the present context, it has been observed that the plant demonstrates significant biological activities; consequently, A. cotula is believed to possess significant potential as a valuable resource in the development of pharmacological designs.

Abstract:The present study was designed to evaluate the chemical composition, antioxidant, enzyme inhibition and cytotoxic properties of different extracts from aerial parts of V. diversifolium (family Scrophulariaceae), a plant that is native to Lebanon, Syria and Turkey. Six extracts, namely, hexane, dichloromethane (DCM), ethyl acetate (EtOAc), ethanol (EtOH), 70% EtOH, and water (aqueous) were prepared by maceration. The EtOH extract was predominated by the presence of rutin (4280.20 μg g −1 ) and p -coumaric acid (3044.01 μg g −1 ) while the highest accumulation of kaempferol-3-glucoside (1537.38 μg g −1 ), caffeic acid (130.13 μg g −1 ) and 4-hydroxy benzoic acid (465.93 μg g −1 ) was recorded in the 70% EtOH, aqueous, and EtOAc extracts, respectively. The EtOH (46.86 mg TE/g) and 70% EtOH (46.33 mg TE/g) extracts displayed the highest DPPH radical scavenging result. Both these extracts, along with the aqueous one, exerted the highest ABTS radical scavenging result (73.03–73.56 mg TE/g). The EtOH and 70% EtOH extracts revealed the most potent anti-AChE (2.66 and 2.64 mg GALAE/g) and anti-glucosidase (1.07 and 1.09 mmol ACAE/g) activities. The aqueous extract was the most efficacious in inhibiting the proliferation of prostate cancer (DU-145) cells with an IC 50 of 8.71 μg/mL and a Selectivity Index of 3.7. In conclusion, this study appraised the use of V. diversifolium aerial parts as a potential therapeutic source for future development of phytopharmaceuticals that target specific oxidative stress-linked diseases including diabetes, cancer, cardiovascular disease, and Alzheimer's disease among others.

Abstract:Bryophytes are rich sources of diverse secondary metabolites with a wide range of biological activities, including anti-inflammatory, antitumor and antimicrobial effects. The aim of this study was to investigate the chemical composition of extracts from two different genotypes (Serbian and Hungarian) of the axenic moss Atrichum undulatum and evaluate the immunomodulatory potential of the prepared extracts in vitro. Both genotypes of moss samples were cultivated in vitro and subsequently extracted in a Soxhlet apparatus with methanol or ethyl acetate. The highest concentration of total phenolic compounds was found in the methanolic extract of the Serbian genotype (54.25 mg GAE/g extract), while the ethyl acetate extract of the Hungarian genotype showed the highest concentration of phenolic acids (163.20 mg CAE/extract), flavonoids (35.57 mg QE/extract), and flavonols (2.25 mg QE/extract). The extracts showed anti-neuroinflammatory properties by reducing the production of reactive oxygen species, nitric oxide, and tumor necrosis factor alpha by lipopolysaccharide-stimulated microglial cells. Moreover, they mitigated the cytotoxic effects of the pro-inflammatory mediators produced by activated microglia on neurons. The data obtained suggest that extracts from A. undulatum moss have promising anti-neuroinflammatory and neuroprotective properties, making them interesting candidates for further research to combat neuroinflammation.

Abstract:In recent decades, natural products of plant origin have predominated among the sources of new therapeutic agents. Plants, which are intensively used in folk medicine, are the focus of particular interest, and being subjected to in-depth analyses for the assessment of their pharmacological properties. The objective of the present work was to evaluate the in vitro anticancer, antiviral, and antioxidant properties of a crude aqueous methanolic extract from the leaves of Rhus typhina L., which is a plant species with broad applications in traditional phytotherapy. The anticancer capacity of the extract was assessed on two human breast cancer cell lines (MCF7 and MDA-MB-231) using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) cell proliferation assay, and its cell growth inhibitory effect on cancer cells was compared with its action on a non-cancerous breast epithelial cell line (MCF10A). Moreover, the in vitro antiviral activity of the plant extract was studied against viral strains of three taxonomic groups: herpes simplex virus type 1 (HSV-1), coxsackievirus B1 (CVB1), and influenza A virus (IAV/H3N2) using a cytopathic effect inhibition test, extract cytotoxicity evaluation through the neutral red uptake assay, a virucidal assay, a viral attachment assay, and cell pre-treatment with the extract. In turn, a radical scavenging assay with 2,2-diphenyl-1-picrylhydrazyl (DPPH) was applied to explore the antioxidant potential of R. typhina leaf extract. Results indicated that the plant extract possessed antiproliferative activity against the tested cell lines which was stronger toward cancer cells. A modest inhibitory effect on IAV/H3N2 replication was detected; as well as on an effect on herpes virions and on the adsorption of the HSV-1 and IAV/H3N2 strains; and a protective effect on uninfected cells before HSV-1 contact. The leaf extract of R. typhina also exhibited a strong free radical scavenging activity. The results of the present work demonstrated the pharmacological potentials of the plant extract, which warrants further and more detailed study in the future.
