Abstract:Herein we report a highly specific electrochemical aptasenseor for AFB1 determination based on AFB1-controlled diffusion of redox probe (Ru(NH3)63+) through nanochannels of AFB1-specific aptamer functionalized VMSF. A high density of silanol groups on the inner surface confers VMSF with cationic permselectivity, enabling electrostatic preconcentration of Ru(NH3)63+ and producing amplified electrochemical signals. Upon the addition of AFB1, the specific interaction between the aptamer and AFB1 occurs and generates steric hindrance effect on the access of Ru(NH3)63+, finally resulting in the reduced electrochemical responses and allowing the quantitative determination of AFB1. The proposed electrochemical aptasensor shows excellent detection performance in the range of 3 pg/mL to 3 μg/mL with a low detection limit of 2.3 pg/mL for AFB1 detection. Practical analysis of AFB1 in peanut and corn samples is also accomplished with satisfactory results by our fabricated electrochemical aptasensor.

Abstract:A rapid and label free aflatoxin B1 (AFB1) microfluid sensor was proposed and tested. The device was fabricated with hollow-core photonics crystal fiber infiltrated with the AFB1 solution. The autofluorescence emitting from the AFB1 molecules was detected. The sensor length was optimized. The AFB1 concentration was tested with a 4 cm long sensor. The best limit of detection was achieved as low as 1.34 ng/ml, which meets the test requirement of the national standards for AFB1 in food. The effectiveness of this sensor being applied in beer solution was also verified to be a little more sensitive than in aqueous solution. Compared with traditional AFB1 detection methods, the proposed single-ended device perfectly satisfies the demand of process control in alcoholic beverages manufacture.

Abstract:In this study, we propose a novel surface-enhanced Raman scattering (SERS) method for quantifying aflatoxin B1 (AFB1). This method relies on the target-triggered release of a SERS reporter from aptamer-sealed aminated mesoporous silica nanoparticles (MSNs). These MSNs were synthesized to accommodate 4-mercaptophenylboronic acid (4-MPBA) within their well-defined micropores, which were subsequently sealed with AFB1 aptamers. Upon specific binding of AFB1 to its aptamer, the conformational change in the aptamer is regulated by the presence of the target. Consequently, a positive linear relationship between the AFB1 concentration and the 4-MPBA SERS signal was observed. Under optimal conditions, the method exhibited a good linear relationship over the range of 0.1 to 5 ng/mL AFB1, with a limit of detection (LOD) of 0.03 ng/mL. This strategy was validated using wheat samples, yielding results comparable to high performance liquid chromatography-fluorescence detector (P > 0.05), confirming its reliability for detecting AFB1 in complex food matrices.

Abstract:Aflatoxin B1 (AFB1) is a highly toxic mycotoxin that can contaminate foods. Hence, a surface-enhanced Raman scattering (SERS) and fluorescence dual signal-on biosensor was developed for AFB1 detection. Specifically, aminated mesoporous silica nanoparticles (MSNs) were used as nanocontainers to load the Rhodamine 6G (R6G) with fluorescence and Raman effects, aptamers and polydopamine (PDA) were sequentially assembled on the surface of the MSNs as dual-gated molecules, and MXenes and Au nanorods were used as SERS substrates. After adding AFB1 under acidic conditions, the degradation of PDA and specific bringing of the aptamer with AFB1 resulted in the release of R6G. Results showed that AFB1 could be quantified by Raman and fluorescence spectroscopic with detection limits of 0.133 pg mL −1 and 0.214 pg mL −1 , respectively. Moreover, the biosensor could be applied to the detection of AFB1 in real peanut, maize and badam samples, providing a possible alternative solution for the detection of other analytes.

Abstract:Aflatoxin B1 (AFB1) has the strongest carcinogenesis and mutagenesis effects. Herein, a novel ratiometric surface-enhanced Raman scattering (SERS) aptasensor based on graphene oxide (GO) as a corrective internal standard (IS) and signal amplifier was constructed for reliable detection of AFB1. 4-Mercaptobenzoic acid as Raman reporter embedded in Au@Ag core-shell nanoparticles and AFB1aptamer conjugated to the Ag shell acted as the signal probes (Au-4MBA@Ag NPs-AFB1apt). AFB1apt was attached to the surface of GO with Raman signals modified Au NPs/Indium tin oxide glass (GO/Au NPs/ITO) via pi-pi stacking interaction, facilitating further amplification of the signal. With an increase of AFB1 concentration (0.0001 ng/mL-100 ng/mL), the probes were released owing to the affinity of aptamers, causing the decreased 4-MBA intensity (I4-MBA) and unchanged GO intensity (IGO). Therefore, the quantitative test was achieved based on the ratiometric value (I4MBA/IGO). A good negative correlation (R2 = 0.992) was achieved and the limit of detection (LOD) was as low as 0.1 pg/mL, proving the proposed sensor can become an efficient analytical tool and shows a bright application prospect in the food detection field.

Abstract:Abstract Recent experiments with 16 male and 16 female subjects indicated that they like to look down rather steeply at an average of −29° below horizontal, s.d. 11·6°, when sitting with trunk and head upright. This angle is steeper when the visual target is at 0·50 m distance (−33°±11·3°) but flatter when the target is at 1·00 m (−24° ± 10·4°). When head and trunk are reclined to 105° behind horizontal, the overall angle of the preferred line of sight is reduced to −20° ±11·7° below horizontal, with −23° (± 12·5°) at 0·5 m and −16° (±9·6°) below horizontal at 1·0 m target distance. These preferred visual angles did not depend on the visual task. The finding that people look down much more steeply than listed in most human engineering design guides has obvious implications for the layout of workplaces where the task has a strong visual component, e.g. at computer workstations.

Abstract:Aflatoxin B1 (AFB1) as toxic secondary metabolite significantly threatens human health. Hence, we designed a reliable “target control signal output” surface-enhanced Raman scattering (SERS) aptasensor based on catalytic reaction and signal amplification induced by bifunctional semiconductor-bimetallic plasmonic assemblies to monitor AFB1 contamination. SH-modified AFB1 aptamers were coupled to hollow Au–Ag NPs with enhanced SERS features, enabling rapid identification of AFB1. As artificial nanoenzymes, self-assembled Cu2O@Au NCs with peroxidase-like activity produced synergistic catalytic enhancement effects. Complementary chain-modified Cu2O@Au NCs promoted the construction of bifunctional composites. Besides, the preferred binding of the aptamer to AFB1 triggered the dissociation of Au–Ag NPs, causing the reduced SERS effect. Then, in the 3,3′,5,5′-tetramethylbenzidine (TMB) reaction mediated by nanoenzymes, oxidation products (ox TMB) with intrinsic Raman signals appeared, which were ideal quantitative signal. Compared with the labeled detection, the novel post-output mode of SERS signal reduced the signal fluctuation in the sensor construction and improved the accuracy of the results. Thus, the aptasensor exhibited a strong negative linear correlation (R2 = 0.991) in the AFB1 concentration ranging from 0.001 ng/mL to 100 ng/mL, and the detection limit (LOD) was 0.7 pg/mL, indicating that the integration of SERS and enzymatic reactions has potential application prospects in test technology.

Abstract:Aflatoxin B1 (AFB1), one of the most toxic mycotoxins, has undergone a wide range of studies over the years. The development of rapid, simple, and sensitive analytical methods remains a major challenge for the accurate detection of AFB1 in foodstuffs. In this study, we designed an enhanced and stable ingenious electrochemiluminescence bioaptasensor (IEC-BA) for ultrasensitive detection of AFB1 based on the synergistic effects and enzyme-driven programmable assembled 3D DNA nanoflowers (EPDNs). This synergistic effect was comprised by the competitive impact on auxiliary probes (AP) and the cutting effect of the Hae III. Compared to the traditional aptamer direct-competition method, the synergistic effects ensured that the aptamer was more efficiently and adequately competed away by the target. Also, the redundant double-stranded probes were removed, which greatly facilitates simple, quick, and sensitive detection of AFB1. Besides, a large chunk of positively charged Ru(II) complexes (Ru(bpy)32+) was accumulated by the utilization of EPDNs, which resulted in tremendous improvement of the sensitivity of the designed method. Thus, even in the presence of trace amounts of AFB1, a sharply visual electrochemiluminescent signal was generated. The proposed method can realize the quantification of AFB1 with a good linear range from 1 ppt (pg mL–1) to 5 ppb (ng mL–1) with a detection limit of 0.27 ppt. In addition, it can also be successfully applied for the analysis of AFB1 in a peanut and wheat, with total recoveries ranging from 93.7 to 106.6%. Abstract:As a virulent metabolite, aflatoxin B1 (AFB1) presented in various cereal grain is tightly implicated in severe human diseases. In this study, 3D plasmonic nanohybirds of Raman molecule 4-mercaptobenzoic acid (4-MBA)-embedded and AFB1 aptamer-modified bimetallic nanostars as SERS probes bound to magnetic nanoflowers were fabricated and demonstrated as a high-performance SERS-active aptasensor to quantitatively analyze AFB1. Bimetallic Au@Ag SERS plasmonic nanoprobes with enhanced properties were capable of enhancing discriminative Raman peaks of 4-MBA. Then, the integration of iron tetroxide nanoparticles (Fe3O4 NPs) and molybdenum disulfide nanosheets (MoS2 NShs) with huge specific surface area constituted stable 3D Fe3O4@MoS2 plasmonic nanoflowers, facilitating the bind of numerous aptamer-based SERS probes via the non-covalent interaction between MoS2 NShs and aptamer, which were ideal candidates for SERS-active substrates. Additionally, Fe3O4 NPs as magnetic core endowed 3D nanocomposites with specific magnetic separation characteristic that caused the collected SERS hotspots to exhibit superior signal response, and further strengthening the sensitivity in a complex food matrix. Aptamer-target AFB1 specific recognition triggered linearly diminished 4-MBA signal intensity (I4-MBA) on the substrate to achieve a low detection limit of 58.9 pg/mL. Furthermore, the sensor has the potential to be a promising monitoring tool for other trace contaminants.

Abstract:Aflatoxin in food is a significant concern that threatens human health and life and causes a heavy burden on international trade and healthcare. In this work, we developed a new type of label-free electrochemical immunosensor for the high-sensitivity determination of aflatoxin B1 (AFB1) based on Au nanoparticles decorated urchin-like Bi 2 S 3 (Au/Bi 2 S 3 ) anchored on electrochemically reduced graphene oxide (ERGO) modified carbon fiber (CF) microelectrode (Au/Bi 2 S 3 /ERGO/CF). The morphology and characterization of the modified microelectrode were investigated using scanning electron microscopy (SEM), cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS), and differential pulse voltammetry (DPV). Under the optimum conditions, the label-free electrochemical immunosensor detected AFB1 within the linear range of 10 pg mL -1 to 20 ng mL -1 , with a low detection limit of 8 pg mL -1 and high sensitivity (0.48 μA ng -1 mL). The immunosensor showed high selectivity, good reproducibility, and acceptable stability. The immunosensor was applied to detecting AFB1 in cornflour samples, offering excellent reliability and accuracy compared with typical detection methods. Finally, this platform provides a superior prospect for the development of a nanohybrid immunosensor.

Abstract:Herein, an aptasensor with stable surface-enhanced Raman scattering (SERS) and fluorescence dual-signal output had been developed for the detection of aflatoxin B1 (AFB1). The quantum dots were encapsulated within silica to ensure the stability of fluorescence signal. The gold nanoparticles (AuNPs) were modified with SERS signal molecule DTNB and the complementary sequence (cDNA) of AFB1 aptamer and then connected on the surface of silica to obtain the signal probes. In the meantime, Fe3O4 was coated with silica which was modified with aptamer to gain capture probes. The two probes could bind specifically based on the base complementary pairing principle. Then, the mixture was separated and gathered by an external magnet. At this time, the maximum SERS signal could be detected in the precipitate, and there was no fluorescence signal in the supernatant. Upon introduction of AFB1 into the system, the signal probes would be released, resulting in fluorescence recovery in the supernatant while SERS signal weakened in the precipitate. The developed method showed a linear relationship of AFB1 for the SERS signal in the range of 10−4–103 ng/mL, with a detection limit of 0.087 pg/mL. The linear range for the fluorescence signal was from 10−4 to 102 ng/mL, with a detection limit of 0.100 pg/mL. The proposed technique was highly sensitive and accurate, which showed promising application prospects in food safety detection.
Optimum synthesis of Au@Ag nanoparticle as plasma amplifier to detect trace concentration of AFB1 via object-binder-metal SERS method

Abstract:The problem of aflatoxin contamination emerged gradually in the field of food safety. Surface-enhanced Raman spectroscopy (SERS) is an ultra-sensitive and non-destructive spectroscopy technology with extensive application prospects in the detection field. In this paper, with the detection of AFB1 as the target, Au@Ag NPs substrate with uniform morphology and strong SERS effect was prepared. Furthermore, the intermediates formed by hydrogen bonding between AFB1 and melamine facilitate the binding of toxin molecules to the substrate. Moreover, the AFB1-melamine-Au@Ag NPs detection system was established by optimizing the melamine dosage, and the limit of detection can reach 10-8 mol/L (M). In this method, AFB1 (concentration range of 10-4 M - 10-7 M) in tea oil, the Raman signal intensity of AFB1 shows an excellent linear, logarithmic relationship, and the correlation coefficient is 0.9685. Therefore, this work has achieved simple, sensitive, and stable AFB1 detection and has broad application potential in food safety detection.
Fabrication of a high-performance photoelectrochemical aptamer sensor based on Er-MOF nanoballs functionalized with ionic liquid and gold nanoparticles for aflatoxin B1 detection

Abstract:One of the most dangerous mycotoxins is Aflatoxin B1 (AFB1) that is commonly found in food. Thus, AFB1 has been considered as a serious food safety concern. Herein, using Er-MOF nanoballs assembled with gold nanoparticles (AuNPs) as photoactive elements, a photoelectrochemical aptasensor with high analytical performance was developed for sensing AFB1. Er-MOFs nanoballs were prepared by using ionic liquid as a ligand and Er 3+ as a metal center through a solvothermal method. Chloroauric acid was reduced in-situ onto Er-MOF nanoballs to form nanocomposites. AuNPs can effectively enhance the photocurrent of Er-MOF nanoballs attributed to their excellent conductivity and local surface plasma resonance effect. In addition, they can provide active sites for immobilizing more biorecognition elements that can further enhance the sensing performance. Through self-assembling reaction, thiol-functionalized hairpin DNA (hDNA) was immobilized onto an interface to construct a photoelectrochemical biosensing platform. As the AFB1 aptamers labeled with gold nanoparticles (AuNPs-Apt) were selectively recognized, the photocurrent response can be further amplified. Due to the high affinity of AFB1 and its aptamer (Apt), when Apt is reacted with AFB1, it causes AuNPs-Apt detaching from the PEC aptasensing interface, and thus a reduced photocurrent response is generated. Under optimized conditions, the changes of photocurrent before and after recognition of AFB1 exhibits a linear response from 0.005 to 10.0 ng mL -1 . In addition, the limit of detection obtained was 19.6 fg mL -1 . This PEC aptasensor provides relatively high selectivity, reproducibility and accuracy in the determination of AFB1 in real corn samples.

Abstract:Traditional enzyme-linked immunosorbent assay (ELISA) suffers from the limitations of relatively low sensitivity and stability, and enzyme-labelled antibodies are hard to be prepared and purified. Based on a nanozyme, an aptamer and Fe3O4 magnetic nanoparticles (MNP), a nanozyme and aptamer-based immunosorbent assay (NAISA) was developed for aflatoxin B1 (AFB1) detection with simpler operation and separation. In this work, mesoporous SiO2/Au-Pt (m-SAP) were prepared to act as signal labels, which showed high catalase-like activity and was denoted as nanozyme. Aptamer was adopted to specifically recognize with AFB1, and MNP facilitated to realize magnetic separation. To verify the performance of NAISA, traditional ELISA (t-ELISA) and enhanced ELISA (e-ELISA) using MNP and m-SAP nanozyme were applied in AFB1 detection. The NAISA method showed the lowest limit of detection (LOD) with 5 pg mL−1 (n = 3, ±4.2 %), 600 and 12-fold lower than that of t-ELISA (3 ng mL−1) and e-ELISA (0.06 ng mL−1), respectively. In the interference tests, AFB1 can be identified among six different interfering substances. The NAISA method, thus, can be of great importance as it allows selective and sensitive AFB1 detection, while providing the simplicity of use and need for screening hazardous materials.

Abstract:Aflatoxin B 1 (AFB 1 ) is a virulent metabolite secreted by Aspergillus fungi, impacting crop quality and posing health risks to human. Herein, a dual -mode Raman/fluorescence aptasensor was constructed to detect AFB 1 . The aptasensor was assembled by gold nanoparticles (AuNPs) and magnetic nanoparticles (MNPs), while the surfaceenhanced Raman scattering (SERS) and fluorescence resonance energy transfer (FRET) effects were both realized. AuNPs were modified with the Raman signal molecule 4 -MBA and the complementary chain of AFB 1 aptamer (cDNA). MNPs were modified with the fluorescence signal molecule Cy5 and the AFB 1 aptamer (AFB 1 apt). Through base pairing, AuNPs aggregated on the surface of MNPs, forming a satellite -like nanocomposite, boosting SERS signal via increased "hot spots " but reducing fluorescence signal due to the proximity of AuNPs to Cy5. Upon exposure to AFB 1 , AFB 1 apt specifically bound to AFB 1 , causing AuNPs detachment from MNPs, weakening the SERS signal while restoring the fluorescence signal. AFB 1 concentration displayed a good linear relationship with SERS/fluorescence signal in the range of 0.01 ng/mL -100 ng/mL, with a detection limit as low as 5.81 pg/mL. The use of aptamer assured the high selectivity toward AFB 1 . Furthermore, the spiked recovery in peanut samples ranged from 91.4 % to 95.6 %, indicating the applicability of real sample detection. Compared to single -signal sensor, this dual -signal sensor exhibited enhanced accuracy, robust anti -interference capability, and increased flexibility, promising for toxin detection in food safety applications.
High-efficiency enzyme-free catalyzed hairpin assembly-mediated homogeneous SERS and naked-eyes dual-mode assay for ultrasensitive and portable detection of mycotoxin

Abstract:We developed an aptamer recognition-trigged enzyme-free catalyzed hairpin assembly (CHA) assisted signal amplification homogeneous naked-eyes and surface-enhanced Raman scattering (SERS) dual-mode sensor for highly sensitive and portable detection of Aflatoxin B1 (AFB1) in food samples. The recognition of AFB1 by aptamer induced the generation of HP1-AFB1 complexes, which hybridized with Ag+-labeled hairpin DNA (HP2) and released Ag+, subsequently initiating the enzyme-free CHA reaction by a designed helper DNA (HP3) to form double-stranded DNA (HP2-HP3) and accompanied by the release of HP1-AFB1 complexes. The released HP1-AFB1 complexes were recognized by HP2 and HP3 again to trigger cascade recycling amplification and resulted in the generation of a larger number of free Ag+ and dsDNA. Then, methylene blue as Raman tag to intercalate into dsDNA and generating strong SERS signal with the assistance of Fe3O4@Au. Meanwhile, the free Ag+ induced the AuNPs aggregation and resulted in naked-eye distinguishable color transitions from red to black blue. Benefitting from the efficiently enzyme-free CHA assisted signal amplification and portably dual-mode detection system, this work successfully proposed a novel homogeneous biosensing strategy for highly sensitive and portable detection of AFB1. The SERS intensity and visualization signals were linearly correlated with the concentration of AFB1 ranging from 0.0156 to 31.2 ng mL−1 and 0.61–39 ng mL−1, and the limit of detections were 1.6 pg mL−1 and 152 pg mL−1, respectively. This strategy was successfully applied to real samples and provided an alternative approach for the highly sensitive detection of mycotoxins.

Abstract:Aflatoxin B1 (AFB1) is one of the most toxic mycotoxins and poses a high risk to human health. Highly sensitive and rapid detection is one of the most effective preventive measures to avoid potential hazards. Herein, an electrochemical aptasensor based on DNA nanotetrahedron and zeolitic imidazolate framework-67 loading gold nanoparticles, horseradish peroxidase, and aptamers was designed for the ultrasensitive detection of AFB1. The high specific surface area and large pore volume of zeolitic imidazolate framework-67 can increase the loading capacity and further improve the detection sensitivity of electrochemical aptasensors. DNA nanotetrahedron can enhance the capture ability of AFB1 with steady immobilization. The developed aptasensor showed good analytical performance for AFB1 detection, with a detection limit of 3.9 pg mL-1 and a wide linear range of 0.01-100 ng mL-1. The aptasensor detected AFB1 in corn samples with recovery rates ranging from 94.19%-105.77% and has potential for use in food safety monitoring.

Abstract:Facile detection methods for mycotoxins with high sensitivity are of great significance to prevent potential harm to humans. Herein, a label-free amperometric immunosensor based on a 3-D interconnected carbon nanofibers (CNFs) network coupled with well-dispersed Au nanoparticles (AuNPs) is proposed for the quantitative determination of aflatoxin B1 (AFB1) in wheat samples. In comparison to common carbon nanotubes (CNTs), the CNFs network derived from bacterial cellulose biomass possesses a unique hierarchically porous structure for fast electrolyte diffusion and a larger electrochemical active area, which increases the peak current of differential pulse voltammetry curves for an immunosensor. Combined with AuNPs that are incorporated into CNFs by using linear polyethyleneimine (PEI) as a soft template, the developed Au@PEI@CNFs-based immunosensor showed a good linear response to AFB1 concentrations in a wide range from 0.05 to 25 ng mL-1. The limit of detection was 0.027 ng mL-1 (S/N = 3), more than three-fold lower than that of an Au@PEI@CNTs-based sensor. The reproducibility, storage stability and selectivity of the immunosensor were proved to be satisfactory. The developed immunosensor with appropriate sensitivity and reliable accuracy can be used for the analysis of wheat samples.

Abstract:Aflatoxin B1 (AFB1) is one of the most toxic mycotoxins, which is frequently detected in agricultural products. Herein, a novel functional DNA -linked immunosorbent assay (DLISA) with dual-modality based on hybrid chain reaction (HCR) has been successfully developed for ultrasensitive detection of AFB1. The strategy relies on AFB1 immune-bridged occurrence of HCR and the salt-induced aggregation of gold nanoparticles (AuNPs). An aptamer-initiator stand (Apt-Ini stand) is designed for the AFB1 recognition and the activation of HCR, which can recognize the matched hairpins and cause the crossing-opening of H1 and H2, producing a long double-stranded DNA polymer. The addition of SYBR Green I achieves the fluorescent signal output. Remaining less DNA hairpins were added and stuck on the surface of AuNPs, which were insufficient to protect the AuNPs, resulting in the salt-induced aggregation with the color change from red to blue. The dual-modality provides limits of detections of 1.333 × 10-14 g/mL and 2.471 × 10-15 g/mL, respectively. This DLISA with dual-modality provides not only a colorimetry that can meet the needs of on-the-spot preliminary inspection, but also a fluorescence assay that can acquire the precise results.
