Abstract:In this research, the effect of selenium (Se) enrichment on dough fermentation characteristics of yeast and the possible mechanisms was investigated. Then, the Se-enriched yeast was used as starter to make Se-enriched bread, and the difference between Se-enriched bread and common bread was investigated. It was found Se enrichment increased CO2 production and sugar consumption rate of Saccharomyces cerevisiae (S. cerevisiae) in dough fermentation, and had positive impacts on final volume and rheological index of dough. The mechanism is possibly related to higher activity and protein expression of hexokinase (HK), phosphofructokinase (PFK), pyruvate kinase (PK), citrate synthase (CS), isocitrate dehydrogenase (ICD), and α-ketoglutarate dehydrogenase (α-KGDHC) in Se-enriched yeast. Moreover, Se-enriched bread (Se content: 11.29 μg/g) prepared by using Se-enriched yeast as starter exhibited higher overall acceptability on sensory, cell density in stomatal morphology, and better elasticity and cohesiveness on texture properties than common bread, which may be due to effect of higher CO2 production on dough quality. These results indicate Se-enriched yeast could be used as both Se-supplements and starter in baked-foods making.

Abstract:The present study has comparatively investigated the variation in yeast leachates in frozen suspension and dough systems and assessed the effects of leachate released from frozen yeast cells on dough rheology, protein molecular weight distribution, gluten conformation, and water state. The results revealed that yeast in the dough was more susceptible to freezing than that in the suspension and released more low-molecular-weight metabolites (e.g., trehalose, glutathione, and amino acids) into the dough matrix. Dynamic rheological measurements showed that frozen yeasted dough had weaker viscoelastic properties than non-yeasted dough. Fourier transform infrared spectroscopy revealed a high fraction of β-turns and random coils at the expense of α-helices and β-sheets, demonstrating a flexible protein secondary structure and increase in weakened hydrogen bonds in the gluten network. The presence of yeast in the frozen dough facilitated dough depolymerization via the cleavage of interchain disulfide bonds of gluten protein and softened the gluten network structure. Yeast leachates released from frozen yeast cells caused water redistribution in the dough and increased water release from the weakened gluten network, increasing freezable water content. This study has revealed the essential role of yeast freezing in gluten depolymerization and the deterioration of dough quality.

Abstract:Summary In this experiment, the effects of gluten protein levels of 6.83% (low gluten), 10.24 (medium gluten), and 13.65% (high gluten), on the physicochemical and fermentation properties of corn dough were studied. The water absorption and weakening of corn dough increased significantly with gluten protein addition, but the development time was gradually shortened. The gas retention capacity of the dough was improved by the addition of gluten protein, which delayed the gas leakage time. The G′ and G" modulus decreased, while Tan δ increased with the increase of gluten protein content. After fermentation, corn dough with 13.65% gluten protein content had the least amount of free water. Total sulfhydryl bond content and disulfide bond content were highest in the dough with a gluten protein content of 13.65%, but irregular curling was no observed. Doughs with gluten protein content of 6.83% were less viscoelastic and elastic compared to doughs with gluten protein content of 10.24%. The gluten protein content of over 10.24% greatly improved the rheological properties of corn dough to a greater extent by strengthening the network structure of the dough.

Abstract:To clarify the correlations between dough rheology and gluten aggregation, the rheological properties of low-sodium salt dough, the molecular chain morphology, molecular weight, subunit distribution and molecular conformation of dough gluten were investigated in this paper, and these results were compared with those of NaCl and KCl alone. Results showed that hardness, extensibility and compression resistance of low-sodium salt dough were enhanced, and the doughs were more like viscoelastic solid material. Atomic force microscopy images showed that low-sodium salt induced the combination of gluten peaks to form the larger protein clusters. Size exclusion/reversed phase-high performance liquid chromatography profiles suggested that low-sodium salt increased the content of B/C-low molecular weight glutenin subunits and high molecular weight glutenin subunits by reducing the content of α- and γ-gliadin, and mediated gluten aggregation to a certain degree. Besides, fluorescence spectra demonstrated low-sodium salt changed the conformation of gluten molecules. In summary, the improvement of rheological properties of low-sodium salt dough could be caused by the aggregation of low-sodium salt and dough gluten. Correlation analysis revealed that rheological properties of low-sodium salt doughs were highly related to the content of gliadin and glutenin fractions. NaCl and KCl also presented similar effects. Among the three salts, NaCl had the most prominent effect, followed by low-sodium salt. This study provides new insights and more comprehensive theoretical basis for sodium reduction.

Abstract:The objectives of the present study were to (1) assess the shear bond strengths of resin-reinforced glass ionomer Fuji Ortho LC and GC Fuji Ortho cements under differing conditions and (2) compare their bonding performance with that of conventional resin composite bonding systems. A sample of 264 bovine incisors was divided into 22 groups of 12 teeth each and bonded with SPEED central incisor brackets. Enamel surfaces of the teeth in the two experimental groups were conditioned according to the manufacturer's instructions; moreover, groups unconditioned before bonding were also included under both wet and dry conditions. A self-cure composite resin (Phase II) and a light-cure composite resin (Transbond XT) served as controls and were etched with 37% phosphoric acid and bonded in a dry field. After incubation at 37 degrees C for 24 hours and for seven days, the specimens were tested to failure with a shear force in an Instron machine. The Adhesive Remnant Index (ARI) was used to assess the amount of resin left on the enamel surfaces after debonding. Selected specimens were examined using scanning electron microscopy. Statistical analyses included analysis of variance tests, t-tests, and correlation coefficient calculations and showed that no significant difference existed between the glass ionomer cements under wet or dry conditions, provided the enamel was conditioned with 10% polyacrylic acid before bonding. Both glass ionomer cements were thus acceptable for bonding. Transbond XT had the highest mean shear bond strength irrespective of the incubation period. A positive correlation was obtained between the ARI scores and bond strength.

Abstract:Biosurfactants have been put into applications in breadmaking industry, while the effects of mannosylerythritol lipid-A (MEL-A) on gluten network of frozen dough, bread quality and microbial spoilage were firstly investigated in this study. Rheology and differential scanning calorimetry (DSC) analysis showed that MEL-A significantly improved the rheological properties of frozen dough and reduced the content of frozen water. Further experiments showed that MEL-A promoted the formation of aggregates by interacting with gluten protein, and strengthened the gluten network through molecular weight distribution measurement and microstructure observation, effectively avoiding the destruction of ice crystals. A series of bread assessments illustrated MEL-A improved the loaf volume, gas retention ability and textural property. In addition, MEL-A (1.5%) killed 99.97% of the vegetative cells of Bacillus cereus and 75.54% of the spores, and at the same time had a slight inactivation effect on yeast. These results indicate that MEL-A has broad application prospects in the baking industry and the storage stage of flour products.

Abstract:PURPOSE The aim of this study was to use clinical, radiographic, and histologic examinations to compare the relative success of gray mineral trioxide aggregate (MTA), white MTA, and formocresol as pulp dressings in pulpotomized primary teeth. METHODS Twenty-four children, each with at least 3 primary molars requiring pulpotomy, were selected for this study's clinical and radiographic portion. An additional 15 carious primary teeth planned for serial extraction were selected for this study's histologic portion. All selected teeth were evenly divided into 3 test groups and treated with pulpotomies. Gray MTA was used as the pulp dressing for one third of the teeth, white MTA was the dressing for one third, and the remaining one third were treated with formocresol. The treated teeth selected for the clinical and radiographic evaluations were monitored periodically for 12 months. The treated teeth selected for histologic study were monitored periodically and extracted 6 months postoperatively. RESULTS Four children with 12 pulpotomized teeth failed to return for any follow-up evaluations in the clinical and radiographic study. Of the remaining 60 teeth in 20 patients, 1 tooth (gray MTA) exfoliated normally and 6 teeth (4 white MTA and 2 formocresol) failed due to abscesses. The remaining 53 teeth appeared to be clinically and radiographically successful 12 months postoperatively. Pulp canal obliteration was a radiographic finding in 11 teeth treated with gray MTA and 1 tooth treated with white MTA. In the histologic study, both types of MTA successfully induced thick dentin bridge formation at the amputation sites, while formocresol induced thin, poorly calcified dentin. Teeth treated with gray MTA demonstrated pulp architecture nearest to normal pulp by preserving the odontoblastic layer and delicate fibrocellular matrix, yet few inflammatory cells or isolated calcified bodies were seen. Teeth treated with white MTA showed a denser fibrotic pattern, with more isolated calcifications in the pulp tissue along with secondary dentin formation. CONCLUSIONS Gray MTA appears to be superior to white MTA and formocresol as a pulp dressing for pulpotomized primary teeth.

Abstract:The quality of wheat flour is largely determined by the properties of gluten proteins. Chemical components that influence gluten proteins are used as flour improvers in the flour industry. In this study, tannins which are natural occurring polyphenols were found to improve dough mixing properties. The effects of tannins on the physicochemical and structural properties of gluten proteins were examined, and the results showed that tannins promoted the non-covalent interactions among gluten proteins, although they induced SH/SS interchange reactions in the dough. The β-turn and α-helix conformations were increased, whereas the β-sheet conformation was decreased in dough containing tannins as detected by FTIR. Moreover, the addition of tannins promoted the aggregation of gluten proteins, modified the microstructure of gluten networks, and improved the mixing properties. The positive effects of tannins on dough properties implied the potential of tannins as a new flour improver.

Abstract:This study investigated the impact of in situ-formed exopolysaccharides (EPS) in red bean (Vigna angularis) sourdough fermented by Weissella confusa QS813 on dough rheo-fermentation properties, bread-making quality and aroma characteristics of red bean sourdough bread. The EPS formed in red bean sourdough and sourdough-induced acidification improved the maximum dough fermentation height, gas retention coefficient and viscoelastic properties of dough. Doughs had a lower increase rate of total SDS-soluble gluten proteins, a low decline in GMP content and similar free sulfhydryl content to wheat dough. Resultantly, breads showed declines in baking loss and hardness, increase in specific volume and lower moisture loss and staling rate after 7 days of storage. Finally, despite a reduction in the total content of aroma compounds, new aroma compounds such as acetic acid and higher contents of 3-methyl-1-butanol and 2,3-butanediol were enriched in red bean sourdough bread. Sourdough acidification probably promoted interaction of EPS with gluten or red bean proteins through bond interactions to form structures which stabilized gluten in dough and increased water-binding ability in red bean sourdough bread. This study provided a better understanding of the role of EPS in sourdough in improving bread quality and of promising strategies to address consumer demand for nutritious and clean-label products.

Abstract:The effects and mechanisms of dietary selenium-enriched yeast (SeY, at Se 0, 0.25, and 0.50 mg/kg) on breast meat quality and oxidative stability of heat-stressed laying hens were studied. Two hundred and forty healthy Jingbai cross-bred hens at late laying period were randomly assigned to 4 experimental groups: one control group reared in a controlled thermal environment, and three treatment groups reared with heat stress (HS). The birds assigned to the controlled thermal environment were fed the basal diet, and the birds in the HS groups were fed the basal diet supplemented with 0, 0.25, or 0.50 mg Se (as SeY)/kg diet for 28 days. Chronic HS decreased pH(45)(min), increased lightness and drip loss, and impaired postmortem oxidative stability of breast muscle. The inclusion of SeY effectively reduced these stress-induced negative impacts, particularly on paleness, drip loss, and pH(45min) decline. The SeY significantly enhanced the cellular antioxidant defense system, suppressed the reactive oxygen species (ROS) overproduction, and mitigated the mitochondrial damage and apoptosis of stressed muscle. In conclusion, dietary modification with SeY reduced quality loss and oxidative instability of meat from heat-stressed hens by maintaining a balanced mitochondrial redox status and inhibiting ROS induced myocyte apoptosis.
Effects of Adding Quinoa Flour on the Composite Wheat Dough: a Comprehensive Analysis of the Pasting, Farinograph and Rheological Properties

Abstract:SummaryRefined wheat breads are popular staples worldwide. Although they have desirable texture, they generally attribute to a high glycaemic index value. Adding whole‐grain flour to produce composite breads with both slower starch digestibility and ideal texture worth investigation. The present study investigated the pasting, farinographic and viscoelastic properties of the quinoa flour (QF)‐wheat flour (WF) composite dough. QF addition reduced the peak, final, breakdown and setback viscosities of the composite flour, indicating that starch gelatinisation was inhibited and retrogradation was slowed. QF addition increased G′ and G′′, while decreased tan δ (G′′/G′), exhibiting that the elasticity prevailed over viscosity for the composite dough. Farinograph analysis showed that when QF addition exceeded 20%, the stability time decreased and weakening degree increased sharply. Although creep‐recovery tests showed that 40% of QF addition exhibited the highest resistance to deformation, it disrupted the gluten network and brought negative effects on the composite dough. Taken these together, the composite wheat dough containing 20% of QF exhibited ideal rheological properties comprehensively. These results might be useful to develop and optimise mixed‐grain breads with lower glycaemic index.

Abstract:The objective of this study was to unveil insights into the effects of Saccharomyces cerevisiae on the development of volatile compounds and metabolites during the dough fermentation in making Chinese steamed bread. Changes in gluten structure under the influence of baker's yeast were studied using scanning electron micrographs (SEM). A unique aroma profile was found comprising some previously reported aromatic compounds and some unreported aromatic aldehydes ((E)-2-Decenal and 2-Undecenal) and ketones (2-Heptanone and 2-Nonanone) in the baker's yeast fermentation. Among metabolites, the most preferred sugar for this yeast (glucose) showed a significant decrease in contents during the initial few hours of the fermentation and at last an increase was observed. However, most of the amino acids increased either slightly or decreased by the fermentation time. SEM of fermented dough showed that the yeast had a very little effect on starch stability. This study provided some fermentation features of the bakers' yeast which could be used for the tailored production of steamed bread.
Impact of gluten separation process and transglutaminase source on gluten based dough properties

Abstract:This study evaluated the effect of the wheat gluten (WG) separation process and transglutaminase (TG) microbial source on WG dough quality, and opportunities to use these factors to tailor dough quality. Two types of gluten (harshly and mildly separated), two types of TG (commercial and novel SB6), and three TG concentrations were evaluated for effects on dough mixing properties, protein structure and solubility. Mildly separated gluten improved dough development parameters, resulting into higher values of most compared with harshly separated gluten. Despite more strongly cross-linked proteins being found in the harshly separated gluten, both gluten types showed similar levels of cross-linking at optimum mixing time, although differences in the secondary protein structure were indicated. Thus, disulfide-sulfhydryl exchange reactions were found to be promoted by mixing, although restrictions on establishment of new bonds because of prior cross-links in the material were clearly indicated. Degree of polymerization in doughs made from mildly separated gluten increased to varying extents with TG addition depending on TG source and concentration. Thus, for the first time, we show that an appropriate combination of WG separation procedure and TG source can be used to tailor gluten dough end-use properties.

Abstract:Selenium-enriched yeast (selenium yeast) are one of the most popular sources of selenium supplementation used in the agriculture and human nutritional supplements industries. To enhance the production efficiency of selenium yeast, we sought to develop a method to identify, and ultimately select for, strains of yeast with enhanced selenium accumulation capabilities. Selenite resistance of four genetically diverse strains of Saccharomyces cerevisiae was assayed in various conditions, including varying carbon sources, nitrogen sources, and phosphate amounts, and they were correlated with selenium accumulation in a commercially relevant selenium-containing growth medium. Glycerol- and selenite-containing media was used to select for six yeast isolates with enhanced selenite resistance. One isolate was found to accumulate 10-fold greater selenium (0.13 to 1.4 mg Se g−1 yeast) than its parental strain. Glycerol- and selenium-containing medium can be used to select for strains of yeast with enhanced selenium accumulation capability. The methods identified can lead to isolation of industrial yeast strains with enhanced selenium accumulation capabilities that can result in greater cost efficiency of selenium yeast production. Additionally, the selection method does not involve the construction of transgenic yeast, and thus produces yeasts suitable for use in human food and nutrient supplements.

Effect of fermentation methods on properties of dough and whole wheat bread

Abstract:Background: Whole wheat bread is high in nutritional value but poor in technological quality; therefore, research on how to improve its technological quality has attracted extensive attention. The effects of fermentation methods, including straight dough(STD), sourdough (SOD), sponge dough (SPD), and refrigerated SPD (RSD) methods, on the dough and bread quality of whole wheat bread were investigated, focusing on pasting properties, rheological properties, thermal properties, microstructure, basic quality, and starch digestibility. Results: The rapid viscosity analysis and rheological results demonstrated that SOD had the highest pasting temperature and the lowest viscosity, indicating an inhibition of starch pasting and partial protein hydrolysis, whereas the opposite trend presented by SPD and RSD indicated a greater starch hydration and a stronger gluten network. Thermal gravimetric analysis and differential scanning calorimetry results indicated reduced starch thermal degradation and increased starch pasting enthalpy in SOD and RSD. Scanning electron microscopy images revealed that the starch granules of SOD and RSD were tightly wrapped by a gluten network. SOD and RSD breads had the largest specific volume, the softest texture, and the lowest glycemic index. Conclusion: The effects of different fermentation methods on dough and bread structure can provide instructive information for future studies on their applications in whole wheat bread production. © 2023 Society of Chemical Industry.
Low-sodium salt mediated aggregation behavior of gluten in wheat dough

Abstract:Reducing sodium in foods has attracted the attention of consumers, it is therefore necessary to explore sodium alternatives (i.e., low-sodium salt). However, the mechanism of low-sodium salt on gluten in dough remains unclear. Effect of low-sodium salt on the aggregation behaviors of gluten in dough was investigated and compared with those with NaCl and KCl in this study. The results showed that low-sodium salt enhanced gluten strength and prolonged gluten aggregation time. Low-sodium salt decreased the content of SDS extractable protein under non-reducing conditions. Low-sodium salt changed the spatial conformation of gluten by reducing β-turn structure and increasing β-sheet structure. Confocal laser scanning microscopy images indicated that low-sodium salt promoted the formation of a larger and dense gluten network. In summary, this study showed that low-sodium salt promoted the aggregation of gluten in dough, and the change of gluten structure explained this aggregation mechanism. Its mode of action was similar to NaCl and KCl, which provided a theoretical basis for the study of sodium substitutes in flour products.

Abstract:Wheat flour can form dough with a three-dimensional viscoelastic structure that is responsible for gas holding during fermentation and oven-rise, creating a typical fixed, open-cell foam structure of bread after baking. As the major components of dough, the continuous reticular skeleton formed by gluten proteins and the concentrated starch granules entrapped in gluten matrix predominantly determine dough rheological behaviors and bread qualities. This review surveys the latest literatures and draws out a conclusion from a plethora of information related to the filling effects of starch granules on gluten matrix and the cross-linking mechanisms between gluten proteins and starch granules, which is of great significance to provide sufficient scientific knowledge for development of bread with satisfactory attributes and quality control of end products.

Abstract:The object of this research was to compare the influence of different soy protein products on wheat dough and its gluten characteristics, including soy protein isolate (SPI), texturized soy protein (TSP) and hydrolyzed soy proteins (SPH), all of which with similar protein content. Addition of TSP could increase dough stability and gluten content, but gluten could not be detected when flour was fortified with SPH. During mixing, SPI tended to interact with SDS soluble wheat proteins, SPH tended to interact with SDS soluble and alcohol soluble wheat proteins, and TSP tended to interact with SDS soluble wheat proteins and TSP. A new protein component was observed from TSP fortified dough by SDS-PAGE. Disulfide bonds change confirmed the new linkage formation in blend dough. CLSM micrographs revealed that effect of SPI, SPH and TSP was different, and this difference was responsible for the change of gluten and dough characteristics.
