Abstract:Humans rely on predictive mechanisms during visual processing to efficiently resolve incomplete or ambiguous sensory signals. While initial low-level sensory data are conveyed by feedforward connections, feedback connections are believed to shape sensory processing through conveyance of statistical predictions based on prior exposure to stimulus configurations. Individuals with autism spectrum disorder (ASD) show biases in stimulus processing toward parts rather than wholes, suggesting their sensory processing may be less shaped by statistical predictions acquired through prior exposure to global stimulus properties. Investigations of illusory contour (IC) processing in neurotypical (NT) adults have established a well-tested marker of contour integration characterized by a robust modulation of the visually evoked potential (VEP) – the – that occurs over lateral occipital scalp during the timeframe of the N1 component. Converging evidence strongly supports the notion that this indexes a signal with significant feedback contributions. Using high-density VEPs, we compared the in 6–17-year-old children with ASD (n=32) or NT development (n=53). Both groups of children generated an that was equivalent in amplitude. However, the notably onset 21ms later in ASD, even though timing of initial VEP afference was identical across groups. This suggests that feedforward information predominated during perceptual processing for 15% longer in ASD compared to NT children. This delay in the feedback dependent , in the context of known developmental differences between feedforward and feedback fibers, suggests a potential pathophysiological mechanism of visual processing in ASD, whereby ongoing stimulus processing is less shaped by statistical prediction mechanisms.

Abstract:Autism, Ahead of Print. Modulation in sensory-perceptual processing is a known characteristic of autism, although the underlying mechanism is debated. A prevailing account is formulated in Bayesian terms, where either a reduced prior or reduced noise in the measurement (sensory input) may account for the modulated perception as expressed by the posterior distribution. However, research has shown that individuals with autism use priors in some conditions, and to the same extent as neurotypicals, while other studies fail to show enhanced sensory sensitivity in these individuals. We asked whether the modulated prior effects on perception may arise from non-adaptive relative weighting of priors to sensory reliability. We employed a Two-alternative forced choice (2AFC) width discrimination task, using the width–height illusion, which is based on a long-term acquired bias, where a taller rectangle is typically perceived as thinner than a shorter one. The measurement was manipulated by adding Gaussian blur on the vertical edges of the rectangles. Typically developed individuals displayed the expected increase in bias as a function of noise in the measurement. High-functioning individuals with autism exhibited typical perceptual resolutions and similar susceptibility to the illusion. However, the relative weighting of the perceptual bias and the sensory input differed in their effect on the two groups. Individuals with autism showed a non-adaptive, consistent bias across the different degrees of sensory noise, while typically developed individuals displayed monotonically increasing biases. Cluster analyses showed that this difference in the relative weighting between the groups was preserved regardless of the overall illusion magnitude displayed by individuals in each cluster.Lay abstractUnique perceptual skills and abnormalities in perception have been extensively demonstrated in those with autism for many perceptual domains, accounting, at least in part, for some of the main symptoms. Several new hypotheses suggest that perceptual representations in autism are unrefined, appear less constrained by exposure and regularities of the environment, and rely more on actual concrete input. Consistent with these emerging views, a bottom-up, data-driven fashion of processing has been suggested to account for the atypical perception in autism. It is yet unclear, however, whether reduced effects of prior knowledge and top-down information, or rather reduced noise in the sensory input, account for the often-reported bottom-up mode of processing in autism. We show that neither is sufficiently supported. Instead, we demonstrate clear differences between autistics and neurotypicals in how incoming input is weighted against prior knowledge and experience in determining the final percept. Importantly, the findings tap central differences in perception between those with and without autism that are consistent across identified sub-clusters within each group.

Abstract:Autism spectrum disorder (ASD) is a neurodevelopmental disorder characterized by impaired social interaction, atypical communication and a restricted repertoire of interests and activities. Altered sensory and perceptual experiences are also common, and a notable perceptual difference between individuals with ASD and controls is their superior performance in visual tasks where it may be beneficial to ignore global context. This superiority may be the result of atypical integrative processing. To explore this claim we investigated visual integration in adults with ASD (diagnosed with Asperger's Syndrome) using two psychophysical tasks thought to rely on integrative processing-collinear facilitation and contour integration. We measured collinear facilitation at different flanker orientation offsets and contour integration for both open and closed contours. Our results indicate that compared to matched controls, ASD participants show (i) reduced collinear facilitation, despite equivalent performance without flankers; and (ii) less benefit from closed contours in contour integration. These results indicate weaker visuospatial integration in adults with ASD and suggest that further studies using these types of paradigms would provide knowledge on how contextual processing is altered in ASD.

Abstract:A growing area of interest and relevance in the study of autism spectrum disorder (ASD) focuses on the relationship between multisensory temporal function and the behavioral, perceptual, and cognitive impairments observed in ASD. Atypical sensory processing is becoming increasingly recognized as a core component of autism, with evidence of atypical processing across a number of sensory modalities. These deviations from typical processing underscore the value of interpreting ASD within a multisensory framework. Furthermore, converging evidence illustrates that these differences in audiovisual processing may be specifically related to temporal processing. This review seeks to bridge the connection between temporal processing and audiovisual perception, and to elaborate on emerging data showing differences in audiovisual temporal function in autism. We also discuss the consequence of such changes, the specific impact on the processing of different classes of audiovisual stimuli (e.g. speech vs. nonspeech, etc.), and the presumptive brain processes and networks underlying audiovisual temporal integration. Finally, possible downstream behavioral implications, and possible remediation strategies are outlined. Abstract:Perceptual anomalies in individuals with autism spectrum disorder (ASD) have been attributed to an imbalance in weighting incoming sensory evidence with prior knowledge when interpreting sensory information. Here, we show that sensory encoding and how it adapts to changing stimulus statistics during feedback also characteristically differs between neurotypical and ASD groups. In a visual orientation estimation task, we extracted the accuracy of sensory encoding from psychophysical data by using an information theoretic measure. Initially, sensory representations in both groups reflected the statistics of visual orientations in natural scenes, but encoding capacity was overall lower in the ASD group. Exposure to an artificial (i.e., uniform) distribution of visual orientations coupled with performance feedback altered the sensory representations of the neurotypical group toward the novel experimental statistics, while also increasing their total encoding capacity. In contrast, neither total encoding capacity nor its allocation significantly changed in the ASD group. Across both groups, the degree of adaptation was correlated with participants’ initial encoding capacity. These findings highlight substantial deficits in sensory encoding—independent from and potentially in addition to deficits in decoding—in individuals with ASD.

Abstract:Atypical integration is a topic of debate in the autism literature. Some theories suggest that altered perception in autism spectrum disorder (ASD) is due to a failure to integrate information from meaningful context into the final percept, whereas others suggest that integration of low-level features is impaired. Empirical research which forms the basis for these theories has failed to account for higher-level influences not inherent in the stimuli (i.e., instructions and goals) and assess integration at both lower and higher perceptual levels within the same task. Here, we describe how perceived expectations and goals of a task can modulate the processing of low-level visual input via the medial prefrontal cortex (mPFC). We then go on to illustrate how future research might assess the relative contribution of both low and high-level processes using the same paradigm. We conclude by recommending that when results appear conflicting, consideration of the relative strength of low-level input vs. feedback or high-level processes may prove helpful. Importantly, research in this area needs to more broadly consider the various influences on perception, and find better ways to assess the contributions of early and later visual processes.

Abstract:Abstract Perceptual expectations influence perception, attention and the perceptual decision bias during visuospatial orienting, which is impaired in individuals with Autism Spectrum Disorder (ASD). In this study, we investigated whether during visuospatial orienting, perceptual expectations in ASD differentially influence perception, attention and the perceptual decision bias relative to neurotypical controls (NT). Twenty-three children and adolescents with ASD and 23 NT completed a visuospatial orienting task, which compared the effect of a valid relative to an invalid perceptual expectation on target detection (cue validity effect). Group differences were calculated regarding the cue validity effect on neural correlates of processing gain (N1a amplitude) and attention (N1pc amplitude), the perceptual decision bias and mean reaction time (RT). In ASD relative to NT, findings showed a reduced processing gain for validly relative to invalidly cued targets and increased attentional response following invalidly relative to validly cued targets. Increased attention correlated with faster performance across groups. Increased processing correlated with a higher perceptual decision bias and faster mean RT in NT, but not in ASD. Results suggest that during visuospatial orienting, perceptual expectations in ASD may drive changes in sensory processing and stimulus-driven attention, which may differentially guide behavioural responses.

Abstract:Clinical EEG and Neuroscience, Ahead of Print. Aim: Deviant visual processing has been observed in autism spectrum disorder (ASD), manifesting as decreased P1 and P2 components of visual event-related potentials (ERPs). Alterations have been attributed to a failure of Bayesian inference, characterized by hypo-activation of top-down predictive abilities. To test this hypothesis, we measured the visual negativity (vN) as an ERP index of visual preparation hypothesized to mirror predictive brain activity. Method: ERPs in a cued visual GO/NOGO task in 63 adolescents with ASD (IQ > 70, attention-deficit hyperactivity disorder excluded) were compared with ERPs in a sex- and age-matched group of 60 typically developing (TD) controls. Results: The behavioral variables (omissions, commissions, reaction time, and reaction time variability), as well as ERP components reflecting, among other processes, cognitive control (contingent negative variation, P3 GO, P3 NOGO, N2 NOGO) did not differ between the groups. There were group differences in visually based ERPs. Besides P1 and P2 differences, the vN component differentiated the 2 groups with the highest effect size (d = 0.74).Conclusion: This ERP study lends support to the hypothesis suggesting that a Bayesian hypo-prediction could underlie unique perceptual experiences in individuals with ASD. This could lead to a predisposition to perceive the world with reduced influence and modulation from contextual cues, prior experiences, and pre-existing expectations.

Abstract:We show in this work that the inhibition of Cdk4 (6) in Rb −/− 3T3 cells enhances the accumulation of the p27 kip1 cyclin-dependent kinase inhibitor when these cells are induced into quiescence. Two different forms of inhibition of Cdk4 (6), namely overexpression of the Cdk4 (6) inhibitor p16 and overexpression of a dominant negative mutant of Cdk4 (Cdk4N158), result in this effect. This suggests that the relevant activity of Cdk4 (6) that has to be inactivated in this setting is its kinase activity. The accumulation of p27 kip1 is due to enhanced translation of the protein, mediated by the 3′-untranslated region of the p27 kip1 mRNA. Moreover, the cells that overexpress p16 ink4a or Cdk4N158 show a delay in G1 when made quiescent and restimulated to proliferate. This delay is overcome by transfection of a plasmid expressing antisense p27 kip1 , which shows that the accumulation of p27 kip1 in these cells is related to their G1delay. In summary, we report a new functional link between two important cell cycle regulators, Cdk4 and p27 kip1 , and provide a mechanistic explanation to the previously reported epistatic relations between these two proteins.
Reduced monitoring of task performance is an effective biomarker of autism

Abstract:People continuously track and adjust their behavior using external and internal signals. Autistic individuals manifest reduced sensorimotor error correction and slower updating of perceptual priors, yielding reduced behavioral flexibility. A potential reason for this reduced flexibility is a lessened sensitivity to both external and internal feedback. A key brain region involved in online monitoring of behavior and updating world models, known to be atypically activated in autism spectrum disorder (ASD), is the anterior cingulate cortex (ACC). During a two-tone pitch discrimination task, we used EEG to track ASD participants' ACC response dynamics, as manifested in their feedback-related negativity (FRN) component produced following "incorrect" feedback. We found that the FRN is missing in more than half of the ASD (17/30) participants, while clearly present in almost all non-autistics (33/35). Moreover, only in non-autistics, feedback affected performance in the following trial. Their responses were slower following an erred trial, their accuracy was improved following hard and correct trials, and they were more affected by the perceptual priors following easy and correct trials. Non-autistics' EEG response to feedback was also sensitive to trial difficulty, while the EEG response of ASD participants was not. Overall, we observed EEG and behavioral correlates of reduced sensitivity to both external and internal feedback in ASD. These observations suggest that increasing the salience of the external feedback could improve ASDs' error monitoring and perhaps increase their behavioral flexibility.

Abstract:The neural underpinnings of enhanced locally oriented visual processing that are specific to autistics with a Wechsler's Block Design (BD) peak are largely unknown. Here, we investigated the brain correlates underlying visual segmentation associated with the well-established autistic superior visuospatial abilities in distinct subgroups using functional magnetic resonance imaging. This study included 31 male autistic adults (15 with (AUTp) and 16 without (AUTnp) a BD peak) and 28 male adults with typical development (TYP). Participants completed a computerized adapted BD task with models having low and high perceptual cohesiveness (PC). Despite similar behavioral performances, AUTp and AUTnp showed generally higher occipital activation compared with TYP participants. Compared with both AUTnp and TYP participants, the AUTp group showed enhanced task-related functional connectivity within posterior visuoperceptual regions and decreased functional connectivity between frontal and occipital-temporal regions. A diminished modulation in frontal and parietal regions in response to increased PC was also found in AUTp participants, suggesting heavier reliance on low-level processing of global figures. This study demonstrates that enhanced visual functioning is specific to a cognitive phenotypic subgroup of autistics with superior visuospatial abilities and reinforces the need to address autistic heterogeneity by good cognitive characterization of samples in future studies.

Abstract:Background: Reduced social attention - looking at faces - is one of the most common manifestations of social difficulty in autism central to social development. Although reduced social attention is well-characterized in autism, qualitative differences in how social attention unfolds across time remains unknown. Methods: We used a computational modeling (i.e., hidden Markov modeling) approach to assess and compare the spatiotemporal dynamics of social attention in a large, well-characterized sample of autistic (n = 280) and neurotypical (n = 120) children (ages 6-11) that completed three social eye-tracking assays across three longitudinal time points (Baseline, 6 weeks, 24 weeks). Results: Our analysis supported the existence of two common eye movement patterns that emerged across three ET assays. A focused pattern was characterized by small face regions of interest, which had high probability of capturing fixations early in visual processing. In contrast, an exploratory pattern was characterized by larger face regions of interest, with lower initial probability of fixation, and more non-social regions of interest. In the context of social perception, autistic children showed significantly more exploratory eye movement patterns than neurotypical children across all social perception assays and all three longitudinal time points. Eye movement patterns were associated with clinical features of autism, including adaptive function, face recognition, and autism symptom severity. Conclusions: Decreased likelihood of precisely looking to faces early in social visual processing may be an important feature of autism that was associated with autism-related symptomology and may reflect less visual sensitivity to face information.

Abstract:In addition to core symptoms, i.e., social interaction and communication difficulties and restricted and repetitive behaviors, autism is also characterized by aspects of superior perception. One well-replicated finding is that of superior performance in visual search tasks, in which participants have to indicate the presence of an odd-one-out element among a number of foils. Whether these aspects of superior perception contribute to the emergence of core autism symptoms remains debated. Perceptual and social interaction atypicalities could reflect co-expressed but biologically independent pathologies, as suggested by a "fractionable" phenotype model of autism. A developmental test of this hypothesis is now made possible by longitudinal cohorts of infants at high risk, such as of younger siblings of children with autism spectrum disorder (ASD). Around 20% of younger siblings are diagnosed with autism themselves, and up to another 30% manifest elevated levels of autism symptoms. We used eye tracking to measure spontaneous orienting to letter targets (O, S, V, and +) presented among distractors (the letter X; Figure 1). At 9 and 15 months, emerging autism symptoms were assessed using the Autism Observation Scale for Infants (AOSI;), and at 2 years of age, they were assessed using the Autism Diagnostic Observation Schedule (ADOS;). Enhanced visual search performance at 9 months predicted a higher level of autism symptoms at 15 months and at 2 years. Infant perceptual atypicalities are thus intrinsically linked to the emerging autism phenotype.

Abstract:Significance Descriptions of impaired coherent motion perception in autism spectrum disorder (ASD) underlie theories that individuals with ASD have difficulty integrating local “parts” into a global percept. This notion maintains widespread influence and motivates recent theories of defective multisensory integration in ASD. However, heightened sensitivity to sensory noise, used to manipulate task difficulty in predominant visual motion stimuli, may provide an alternative explanation for impaired performance. Here we indeed found increased sensitivity in ASD to sensory noise. Noiseless motion perception and multisensory integration (even with noise) were unimpaired. These findings question prevalent theories of global and multisensory integration deficits in ASD. Rather, they suggest increased reliance on—and sensitivity to—incoming sensory information and less use of prior knowledge in ASD.

Abstract:Research has shown that children on the autism spectrum and adults with high levels of autistic traits are less sensitive to audiovisual asynchrony compared to their neurotypical peers. However, this evidence has been limited to simultaneity judgments (SJ) which require participants to consider the timing of two cues together. Given evidence of partly divergent perceptual and neural mechanisms involved in making temporal order judgments (TOJ) and SJ, and given that SJ require a more global type of processing which may be impaired in autistic individuals, here we ask whether the observed differences in audiovisual temporal processing are task and stimulus specific. We examined the ability to detect audiovisual asynchrony in a group of 26 autistic adult males and a group of age and IQ‐matched neurotypical males. Participants were presented with beep‐flash, point‐light drumming, and face‐voice displays with varying degrees of asynchrony and asked to make SJ and TOJ. The results indicated that autistic participants were less able to detect audiovisual asynchrony compared to the control group, but this effect was specific to SJ and more complex social stimuli (e.g., face‐voice) with stronger semantic correspondence between the cues, requiring a more global type of processing. This indicates that audiovisual temporal processing is not generally different in autistic individuals and that a similar level of performance could be achieved by using a more local type of processing, thus informing multisensory integration theory as well as multisensory training aimed to aid perceptual abilities in this population.

Abstract:Autism spectrum disorder (ASD) is a common neurodevelopmental condition with approximately 1-2 per cent prevalence in the population. The condition has lifelong effects for the individual and family, and early intervention and management helps maximise quality of life and outcomes. Many studies of vision in ASD have attempted to link the behavioural and sensory deficits in ASD with underlying visual processing. From this work, it is clear that individuals with ASD 'see' and process the world differently, but there remain gaps in our understanding. This review will summarise our current knowledge of key aspects of visual functions and the optometric profile of ASD. This includes findings regarding visual acuity and contrast sensitivity, refractive error, eye movements, binocular vision, near visual functions and retinal structure in ASD. From this, a pattern of knowledge emerges for children with ASD: we should expect normal visual acuity; there will likely be atypical eye movements and susceptibility for subtle visuo-motor deficits, there is an increased prevalence of strabismus; an increased likelihood of astigmatism and possibly other refractive errors; attention, crowding and task complexity will likely be problematic; and retinal structure and function may be compromised. Bringing this together, these findings highlight that further work is necessary, not only to understand how higher-level functions link to behaviours, but also to ensure there is a sound understanding of the building-blocks of vision to fully grasp the profile of visual processing as a whole in ASD. This review will give a translational viewpoint for clinicians, and underline the benefits of comprehensive vision care in ASD.

Visual Perception in Autism Spectrum Disorder: A Review of Neuroimaging Studies

Abstract:Although autism spectrum disorder (ASD) is a neurodevelopmental disorder characterized by social impairments, patients with ASD frequently manifest atypical sensory behaviors. Recently, atypical sensory perception in ASD has received much attention, yet little is known about its cause or neurobiology. Herein, we review the findings from neuroimaging studies related to visual perception in ASD. Specifically, we examined the neural underpinnings of visual detection, motion perception, and face processing in ASD. Results from neuroimaging studies indicate that atypical visual perception in ASD may be influenced by attention or higher order cognitive mechanisms, and atypical face perception may be affected by disrupted social brain network. However, there is considerable evidence for atypical early visual processing in ASD. It is likely that visual perceptual abnormalities are independent of deficits of social functions or cognition. Importantly, atypical visual perception in ASD may enhance difficulties in dealing with complex and subtle social stimuli, or improve outstanding abilities in certain fields in individuals with Savant syndrome. Thus, future research is required to elucidate the characteristics and neurobiology of autistic visual perception to effectively apply these findings in the interventions of ASD.
Elevated and accelerated: Locus coeruleus activity and visual search abilities in autistic children

Abstract:Background: Autistic individuals excel at visual search, however, the neural mechanism(s) underlying this advantage remain unclear. The locus coeruleus-norepinephrine (LC-NE) system, which plays a critical role in sensory perception and selective attention, has been shown to function in a persistently elevated state in individuals on the spectrum. However, the relationship between elevated tonic LC-NE activity and accelerated search in autism has not been explored. Objective: To examine the relationship between visual search abilities and resting pupil diameter (an indirect measure of tonic LC-NE activation) in autistic and neurotypical children. Methods: Participants were 24 school-aged autistic children and 24 age- and IQ-matched neurotypical children aged 8-15 years. Children completed two tasks: a resting eye-tracking task and a visual search paradigm. For the resting eye-tracking task, pupil diameter was monitored while participants fixated a central crosshair. For the visual search paradigm, participants were instructed to find the target (vertical line) embedded within an array of tilted (10°) distractor lines. The target was present on 50% of trials, and displayed within set sizes of 18, 24, and 36 items. Results: Consistent with previous studies, autistic children had significantly larger resting pupil size and searched faster and more efficiently compared to their neurotypical peers. Eye-tracking findings revealed that accelerated search was associated with fewer, not shorter, fixations in the autism group. Autistic children also showed reduced leftward search bias. Larger resting pupil size, indicative of increased tonic activation of the LC-NE system, was associated with greater search efficiency, longer fixation durations, and reduced leftward bias. Finally, within both groups reduced leftward bias was associated with increased autism symptomatology. Discussion: Together, these findings add to the existing body of research highlighting superior search in autism, suggest that elevated tonic LC-NE activity may contribute to more efficient search, and link non-social visual-spatial processing strengths to autism symptoms.
