Abstract:Specific detection of glycoproteins such as transferrin (TRF) related to neurological diseases, hepatoma and other diseases, always plays an important role in the field of disease diagnosis. We designed an antibody-free immunoassay sensing method based on molecular imprinted polymers (MIPs) formed by polymerization of multiple functional monomers for the sensitive and selective detection of TRF in human serum. In the sandwich surface-enhanced Raman spectroscopy (SERS) sensor, the TRF-oriented magnetic MIP nanoparticles (Fe 3 O 4 @SiO 2 -MIPs) served as to capture unit to specifically recognize TRF and 4-mercaptophenylboronic acid-functionalized gold Nanorods (MPBA-Au NRs) served as SERS probes to label the targets. In order to achieve stronger interaction between the recognition cavities of the prepared MIPs and the different amino acid fragments that make up TRF, Fe 3 O 4 @SiO 2 -MIPs were obtained through polycondensation reactions between more silylating reagents, enhancing the specific recognition of the entire TRF protein and achieving high IF. This sensing method exhibited a good linear response to TRF within the TRF concentration ranged from 0.01 ng/mL to 1 mg/mL (R 2 = 0.9974), and the LOD was 0.00407 ng/mL (S/N = 3). The good stability, reproducibility and specificity of the resulting MIP based SERS sensor were demonstrated. The determination of TRF in human serum confirmed the feasibility of the method in practical applications.

Synthesis of a multi-recognition site borate affinity magnetic surface imprinted polymers for the specific recognition and detection of two glycoproteins at physiological pH

Abstract:Glycoproteins are commonly used as biomarkers for diseases, and capturing multiple target glycoprotein biomarkers at the same time has better specificity than single marker detection in early screening for many diseases. In this work, we propose a method for preparing dual-template magnetic molecularly imprinted nanoparticles (D-MMIPs) with two recognition sites. Firstly, horseradish peroxidase (HRP) and ovalbumin (OVA), two commonly used glycoproteins, were used as double templates to simulate glycoprotein biomarkers. 4-Formylphenyl boric acid (FPBA) was introduced as a functional monomer on the surface of amino-modified Fe 3 O 4 @SiO 2 to fix simultaneously the two templates. Finally, the thin imprinted layer was prepared by surface-controlled polymerization of copolymer of dopamine (DA) and 3-aminophenylboronic acid (APBA), which reduced the mass transfer resistance of the template and improved the eluting efficiency of the template. The obtained D-MMIPs showed a high adsorption capacity of 65.52 mg/g for HRP and 93.61 mg/g for OVA. There is also a satisfactory selectivity in identifying two templates in the real sample under physiological conditions. In addition, a boronate affinity sandwich strategy was constructed by using D-MMIPs as a recognition element combined with surface-enhanced Raman scattering (SERS) technology to achieve rapid separation and detection of HRP and OVA, with detection limits of 2.15 pg/mL and 0.708 pg/mL, respectively. The results imply that the strategy may be suitable for simultaneous and selective capture of multiple glycoprotein biomarkers to detect multiple diseases, and the prepared magnetic imprinting materials have potential value in sensor applications.

Abstract:The design of a rapid, simple, and ultrasensitive detection platform of protein biomarkers has become an urgent need in early diagnosis and treatment. Surface-enhanced Raman scattering (SERS) technology, with the unique advantages of ultra-sensitivity and fingerprint recognition, has attracted much attention in biomarker detection. An innovative aptamer SERS biosensor for highly sensitive protein biomarker detection was presented. This biosensor contains a SERS tag (reporter-labeled Au nano-bridged nanogaps particles, Au NNPs) and novel magnetic capture substrate (Ag-coated Fe3O4-Au NPs, Ag MNPs). C-reactive protein (CRP), a representative biomarker for diagnosing, was used to assess the SERS performance of this biosensor. Aptamers against CRP were modified on both SERS tag and magnetic capture substrate for specific recognition by Au NNPs-CRP-Ag MNPs sandwich assay. This strategy achieved sensitive detection of CRP with a limit of detection (LOD) of 10 fM (1.14 pg/mL), and is much lower than reported analytical methods. Besides, this method exhibits excellent selectivity and specificity for CRP under the interference of other proteins and displays high accuracy in the detection of actual human serum samples. This proposed detection strategy can be extended to the sensitive detection of other biomarker proteins and hold great promise in the clinical diagnosis of diseases.

Abstract:It is challenging to detect and enric glycoproteins in complex biological samples because they are low in natural abundance, highly flexible in conformation and there are many interfering substances in the samples. To overcome these challenges, magnetic Fe3O4 nanoparticles (NPs) were prepared, in which the surface was modified by metal–organic frameworks (MOFs) and then by molecularly imprinted polymers (MIPs). Combined with the teamed boronate affinity (TBA) strategy, the selectivity and sensitivity detection of glycoprotein ovalbumin (OVA) were realized. The magnetic Fe3O4 NPs were coated with MOFs to facilitate the adsorption of AuNPs, onto which the TBA moieties composed of 2-mercaptoethylamine and 4-mercaptophenylboronic acid were pre-assembled. The final magnetic Fe3O4@TBA/MIPs NPs featured shallow imprinted cavities, resulting in rapid mass transfer of the target glycoprotein (equilibrium time 20 min). In particular, the TBA strategy relied on N-B synergy, resulting in high specificity (IF = 4.52) and high sensitivity (limit of detection 13 pM) for the detection of OVA at physiological pH (7.4). This strategy provides an extremely convenient method for detecting and quantifying low-abundance glycoprotein in complex biological samples, without destroying the structure and function of the target glycoprotein.
Highly Selective Determination of Acid Phosphatase in Biological Samples Using a Biomimetic Recognition-Based SERS Sensor

Abstract:In this paper, we introduce a synthetic biomimetic receptors-based surface enhanced Raman scattering (SERS) sensor for highly specific recognition and detection of acid phosphatase in complex biological samples. It depends on the establishment of molecular imprinted polymers (MIPs) which capture the target glycoprotein via biomimetic recognition, and SERS probes which bind to glycoproteins by boronate affinity. The MIPs layer was synthesized through self-polymerization of dopamine on the surface of glycoprotein-immobilized polymeric skeletons, which was prepared by choosing 4-vinylphenylboronic acid (VPBA) as functional monomers, pentaerythritol triacrylate (PETA) as crosslinker, ethyleneglycol (EG) and cyclohexanol as porogens. The accessible and stable recognition sites allowed the rebinding of the template and offered good reproducibility. The proposed biomimetic recognition-based SERS sensor showed superb performances for acid phosphatase detection with high sensitivity (LOD of 0.1 ng mL(-1)) and excellent selectivity.
Molecularly Imprinted Polymer-Based Plasmonic Immunosandwich Assay for Fast and Ultrasensitive Determination of Trace Glycoproteins in Complex Samples

Abstract:Glycoproteins play significant roles in many biological processes. Assays of glycoproteins have significant biological importance and clinical values, for which immunoassay has been the workhorse tool. However, immunoassay suffers from some disadvantages, such as poor availability of high-specificity antibodies and limited stability of biological reagents. Herein, we present an antibody-free and enzyme-free approach, called molecularly imprinted polymer (MIP)-based plasmonic immunosandwich assay (PISA), for fast and ultrasensitive detection of trace glycoproteins in complex samples. A gold based boronate affinity MIP array was used to specifically extract the target glycoprotein from complex samples. After washing away unwanted species, the captured glycoprotein was labeled with boronate affinity silver-based Raman nanotags. Thus, sandwich-like complexes were formed on the array. Upon being shined with a laser beam, the gold-based array generated a surface plasmon wave, which significantly enhanced the surface-enhanced Raman scattering (SERS) signal of the silver-based Raman nanotags. The MIP ensured the specificity of the assay, while the plasmonic detection provided ultrahigh sensitivity. Erythropoietin (EPO), a glycoprotein hormone that controls erythropoiesis or red blood cell production, was employed as a test glycoprotein in this study. Specific detection of EPO in solution down to 2.9 X 10(-14) M was achieved. Using a novel strategy to accommodate the method of standard addition to a logarithmic dose response relationship, EPO in human urine was quantitatively determined by this approach. The analysis time required only 30 min in total. This approach holds promising application prospects in many areas, such as biochemical research, clinical diagnosis, and antidoping analysis.

Abstract:Although antibody has played a great role in highly specific recognition of protein biomarkers, it faces poor stability, reproducibility, high-cost and time-consuming preparation, etc. Here, aptamer and molecularly imprinted polymers (MIPs), both as promising substitutes of antibody, were integrated onto magnetic nanoparticles by Au-S bonds and SiO2 as imprinted layer for preparing a new nanoprobe. Highly specific and sensitive recognition of different protein biomarkers, such as insulin for diabetes and alpha-fetoprotein (AFP) for hepatic carcinoma, were achieved respectively by the system of combining hybrid aptamer-molecularly imprinted magnetic nanoprobe and mass spectrometry. With the double affinities offered by aptamer-MIPs, insulin can be detected at 0.5 ng mL(-1) in human serum dilution, the equlibrium dissociation constant between nanoprobe and insulin is measured as 23.61 +/- 2.27 mM. Likewise, AFP can be sufficiently detected in human saliva dilution from 1000 ng mL(-1) to 20 ng mL(-1), and two patients with hepatic carcinoma are discriminated from healthy person due to the abnormally high expression of AFP in serum. Oriented surface imprinted 96-well microplate-based fluorescent biosensor for glycoprotein detection by boronate affinity sandwich assay

Abstract:Glycoproteins perform vital functions in numerous biological processes and have important clinical implications. Many glycoproteins have been used as biomarkers and therapeutic targets for disease diagnosis. Due to low concentration of glycoprotein biomarkers and the presence of high-abundance interfering species in biological samples, a selective and sensitive detection method for glycoprotein is essential for real-world applications. In this study, we develop an oriented surface imprinted microplate-based fluorescent biosensor by boronate-affinity sandwich assay (BASA) for the specific, sensitive and high throughput determination of glycoproteins in complex samples. The structure of the BASA is based on sandwich formation between boronate affinity-oriented surface-imprinted microplates, target glycoproteins, and boronate affinity fluorescence probes. The imprinted microplates ensure the high specificity, high affinity and high throughput, while the fluorescence probes, consisting of boronic acid-modified CdTe QDs, provide high sensitivity. The proposed approach could exhibit a wide linear range of 1 ng/mL-105 ng/mL, with a low LOD of 0.528 ng/mL using horseradish peroxidase (HRP) as a model glycoprotein. As compared with traditional "turn off" fluorescent sensor, the developed "turn on" fluorescent sensor provided three orders of magnitude higher sensitivity at least. The fluorescent biosensor achieved average recoveries ranging from 96.8 % to 106.0 % in urine samples.
Ratiometric Surface Enhanced Raman Scattering Immunosorbent Assay of Allergenic Proteins via Covalent Organic Framework Composite Material Based Nanozyme Tag Triggered Raman Signal "Turn-on" and Amplification

Abstract:The exploration of nanomaterials with mimic enzyme activity (named nanozyme) has gained extensive attention in the fields of advanced analytical chemistry and materials science. Herein, the gold nanoparticles doped covalent organic frameworks (COFs) were prepared, which exhibited not only excellent mimic nitroreductase activity but also robust stability. By replacing the traditional natural enzyme tag in an enzyme-linked immunosorbent assay (ELISA), we employed the proposed nanozyme to label the detecting antibody. According to the catalytic properties of the nanozyme, 4-nitrothiophenol (4-NTP) was introduced as the substrate, which can be transformed to 4-aminothiophenol (4-ATP) in the presence of NaBH4. In a surface enhanced Raman scattering (SERS) assay, 4-ATP was capable of functioning as a powerful bridge to connect the gold nanostars (with excellent SERS performance) by both the Au-S bond and electrostatic force to further produce a Raman "hot spot". Meanwhile, the Raman signal of 4-nitrothiophenol at 1573 cm-1 was weakened, and a new signal at 1591 cm-1 generated by 4-ATP was turned on, leading to the generation of a ratiometric SERS signal. Based on this performance, a ratiometric nanozyme-linked immunosorbent assay (NELISA) strategy was developed delicately, which was applied to detect β-lactoglobulin (allergenic protein) by monitoring the ratiometric signal of I1591/I1573 with a limit of detection (LOD) of 0.01 ng/mL. The linear range is 25.65-6.2 × 104 ng/mL, covering more than 3 orders of magnitude. The developed method showed many advantages such as low-cost, higher recovery, and lower cross-reactivity, providing new insight into the application of SERS technology for trace target analysis.
Label-free SERS detection of Raman-Inactive protein biomarkers by Raman reporter indicator: Toward ultrasensitivity and universality

Abstract:It is still challenging to sensitively detect protein biomarkers via surface-enhanced Raman scattering (SERS) technique owing to their low Raman activity. SERS tag-based immunoassay is usually applied; however, it is laborious and needs specific antibodies. Herein, an ultrasensitive and universal "Raman indicator" sensing strategy is proposed for protein biomarkers, with the aid of a glass capillary-based molecularly imprinted SERS sensor. The sensor consists of an inner SERS substrate layer for signal enhancement and an outer mussel-inspired polydopamine imprinted layer as a recognition element. Imprinted cavities have two missions: first, selectively capturing the target protein, and second, the only passageway of Raman indicator to access SERS substrate. Specific protein recognition means filling imprinted cavities and blocking Raman indicator flow. Thus, the quantity of captured protein can be reflected by the signal decrease of ultra-Raman active indicator molecule. The capillary sensor exhibited specific and reproducible detection at the level down to 4.1 × 10−3 μg L−1, for trypsin enzyme in as-received biological samples without sample preparation. The generality of the mechanism is confirmed by using three different protein models. This platform provides a facile, fast and general route for sensitive SERS detection of Raman inactive biomacromolecules, which offers great promising utility for in situ and fast point-of-care practical bioassay.

Magnetic Nanoparticles with Dendrimer-Assisted Boronate Avidity for the Selective Enrichment of Trace Glycoproteins

Abstract:Boronic acid-functionalized materials have been the subject of increasing attention in recent years due to their capability in the facile selective extraction of glycoproteins. However, boronic acids are associated with weak binding affinity, and it is thereby difficult for boronate affinity materials to extract glycoproteins of low concentration. Here we present for the first time a boronate avidity material, dendrimeric boronic acid-functionalized magnetic nanoparticles, with significantly enhanced binding strength towards glycoproteins. Due to the dendrimer-assisted multivalent synergistic binding, the boronate avidity material exhibited dissociation constants of 10−5 to 10−6 M towards glycoproteins, which are 3–4 orders of magnitude higher than the affinities of single boronic acid binding. Such an avidity enabled the selective extraction of trace glycoproteins; an extractable concentration as low as 2 × 10−14 M was demonstrated. Meanwhile, the boronate avidity material was tolerant of the interference of abundant competing sugars. Moreover, the dendrimeric boronic acid-functionalized magnetic nanoparticles exhibited two additional advantages: high binding capacity and fast binding/desorption speed. Due to these favourable features, the selective enrichment of trace glycoproteins by the boronate avidity material became not only possible but also efficient. Efficient enrichment of trace glycoproteins from human saliva was demonstrated. The dendrimer-assisted synergistic binding strategy is also applicable to other types of boronic acid-functionalized materials and other types of functionalized materials.
Sandwich-type Electrochemical Biosensor for Glycoproteins Detection Based on Dual-Amplification of Boronic Acid-Gold Nanoparticles and Dopamine-Gold Nanoparticles.

Abstract:Glycoproteins play important roles in a wide variety of biological processes. The change in the concentration levels has been associated with many cancers, as well as other diseases. Thus, rapid, sensitive and selective determination of glycoproteins is much preferred. In this work, we reported a sandwich-type electrochemical biosensor based on dual-amplification of 4-mercaptophenylboronic acid (MBA)-capped gold nanoparticles (MBA-AuNPs) and dopamine (DA)-capped AuNPs (DA-AuNPs). Biological recognition elements such as synthetic receptor and aptamer immobilized onto gold electrodes were used to capture glycoproteins. The captured glycoproteins were then derivatized with MBA-AuNPs through the formation of tight covalent bonds between the boronic acids of MBA-AuNPs and diols of glycoproteins. Electroactive DA-AuNPs were attached by the anchored MBA-AuNPs via the interaction of boronic acids with DA tags, which facilities the amplified voltammetric detection of glycoproteins. With avidin and prostate specific antigen (PSA) as model analytes, we demonstrated the feasibility and sensitivity of the proposed method. The results indicated that sub-picomolar avidin/PSA can be readily measured. We believe that this strategy will be valuable for the electrochemical detection of other glycoproteins.

Abstract:Glycoproteins are difficult to be detected by imprinting strategy due to their low natural abundance, high flexible conformation and large size. Herein, a high-density boric acid modified metal–organic framework (MOF) surface molecularly imprinted polymer (SMIP) resonant light scattering sensor was constructed for the high-sensitivity detection of target glycoproteins. A MOF with large specific surface area was selected as the substrate material to support the boric acid group with high loading density (4.66 %). The introduction of the boric acid group in the SMIP provided a high-affinity binding site for the recognition and binding of glycoproteins. Shallow surface cavities with rapid mass transfer (equilibrium time 20 min) were thus formed by surface imprinting. Furthermore, high sensitivity (limit of detection 15 pM) was achieved at physiological pH (7.4), which was conducive to the detection of glycoproteins with low natural abundance in complex biological samples and maintaining physiological activity.

Abstract:Ultrasensitive detection of protein biomarkers is essential for early diagnosis and therapy of many diseases. Glycoproteins, differing from other types of proteins, contain carbohydrate moieties in the oligosaccharide chains. Boronic acid can form boronate ester covalent bonds with diol-containing species. Herein, we present a sensitive and cost-effective electrochemical method for glycoprotein detection using 4-mercaptophenylboronic acid (MBA)/biotin-modified gold nanoparticles (AuNPs) (MBA-biotin-AuNPs) as labels. To demonstrate the feasibility and sensitivity of this method, recombinant human erythropoietin (rHuEPO) was tested as a model analyte. Specifically, rHuEPO was captured by the anti-rHuEPO aptamer-covered electrode and then derivatized with MBA-biotin-AuNPs through the boronic acid-carbohydrate interaction. The MBA-biotin-AuNPs facilitated the attachment of streptavidin-conjugated alkaline phosphatase for the production of electroactive p-aminophenol from p-aminophenyl phosphate substrate. A detection limit of 8 fmol L(-1) for rHuEPO detection was achieved. Other glycosylated and non-glycosylated proteins, such as horseradish peroxidase, prostate specific antigen, metallothionein, streptavidin, and thrombin showed no interference in the detection assay.
Direct Glycan Analysis of Biological Samples and Intact Glycoproteins by Integrating Machine Learning Driven- Surface-Enhanced Raman Scattering (SERS) and Boronic Acid Array

Abstract:Frequent monitoring of glycan patterns is a critical step for studying glycan-mediated cellular processes. However, the current glycan analysis tools are resource-intensive and less suitable for routine use in standard laboratories. We developed a novel glycan detection platform by integrating surface-enhanced Raman spectroscopy (SERS), boronic acid (BA) receptors, and machine learning (ML) tools. This sensor monitors the molecular fingerprint spectra of BA binding to cis-diol-containing glycans. Different types of BA could yield different stereoselective reactions toward different glycans and exhibit unique vibrational spectra. By integrating the Raman spectra collected from different BA receptors, the structural information can be enriched, eventually improving the accuracy of glycan classification and quantification. Here, we established a SERS-based sensor incorporating multiple different BA receptors. This sensing platform could directly analyze the biological samples, including the whole milk and intact glycoproteins (fetuin and asialofetuin), without tedious glycan release and purification steps. The results demonstrate the platform's ability to classify milk oligosaccharides with remarkable classification accuracy, despite the presence of other non-glycan constituents in the background. This sensor could also directly quantify sialylation levels of fetuin/asialofetuin mixture without glycan release procedures. Moreover, by selecting appropriate BA receptors, the sensor exhibits an excellent performance of differentiating between α2,3 and α2,6 linkages of sialic acids. This low-cost, rapid, and highly accessible sensor will provide the scientific community with an invaluable tool for routine glycan screening in standard laboratories.

SERS-based immunocapture and detection of pathogenic bacteria using a boronic acid-functionalized polydopamine-coated Au@Ag nanoprobe

Abstract:A surface-enhanced Raman scattering (SERS)-based immunocapture nanoprobe is described for the detection of pathogenic bacteria. The probe uses boronic acid-functionalized polydopamine-coated Au@Ag nanoparticles as an advanced SERS nanotag. Modified magnetic IgG@Fe 3 O 4 nanoparticles are used for magnetic separation. Au@Ag@PDA nanoparticles, where PDA stands for polydopamine, were functionalized with boronic acid to bind to pathogenic bacteria and induce signal amplification. The Raman signal is amplified 10 8 times when the SERS tag binds the surface of bacteria. The SERS spectra exhibit fingerprint-like patterns that enable bacterial classification. The results of principal component analysis (PCA) and hierarchical cluster analysis (HCA) of the spectral regions were compared. The bacterial surface protein and glycan signals (1300–1450 cm −1 ) were the best regions for bacterial classification. Staphylococcus aureus , Escherichia coli , Shigella dysenteriae , Pseudomonas aeruginosa , and Klebsiella pneumonia were successfully classified by this method. The lowest detection limit was 10 colonies/mL (CFU·mL). The assay can be completed within 30 min. Conceivably, this method may be extended to the quantitative detection or classification of bacteria under various other conditions. Abstract:Protein assays with fingerprints and high sensitivity are essential for biomedical research and applications. However, the prevailing methods mainly rely on indirect or labeled immunoassays, failing to provide fingerprint information. Herein, we report a dual biomimetic recognition-driven plasmonic nanogap-enhanced Raman scattering (DBR-PNERS) strategy for ultrasensitive protein fingerprinting and quantitation. A pair of molecularly imprinted nanoantennas were rationally engineered for specifically trapping a target protein into well-defined plasmonic nanogaps through dual-terminal recognition for ultrahigh Raman signal amplification. Meanwhile, a Raman-active small molecule was embedded into the nanoantenna as an internal standard to provide a ratiometric assay for robust quantitation. DBR-PNERS exhibited several significant merits over existing approaches, including fingerprinting, ultrahigh sensitivity, quantitation robustness, speed, sample consumption, and so on. Therefore, it can be a promising tool for a protein assay and holds a great perspective in important applications.

Abstract:In this research, we proposed a novel fluorescence imprinted sensor (FIS@MIP) for rapid and sensitive detection of transferrin (TrF) based on the boronic acid functionalized Zr-MOF (Zr-UiO-66-B(OH) 2 ) and high fluorescence quantum yield of Mg/N co-doped carbon dots (Mg/N-CDs). The boronic acid moieties of Zr-UiO-66-B(OH) 2 could significantly covalently bind TrF and load more Mg/N-CDs to avoid fluorescence quenching caused by self-aggregation of Mg/N-CDs. Additionally, Zr-UiO-66-B(OH) 2 was prepared though linker fragmentation approach, which eliminated the complex post modification process and could provide sufficient boric acid groups. High luminescent and fluorescence quantum yield endowed Mg/N-CDs remarkable fluorescence response to TrF. Under the optimal conditions, the fluorescence intensity of FIS@MIP at 438 nm was quenched by Trf within 1 min. Based on specific recognition sites, a good linear relationship was obtained in the linear range of 0.1 μM–20 μM, and the low detection limit was determined as 0.067 μM. In order to explore portable detection application, a home-made device based on smartphone was designed for on-site detection of TrF with a detection limit of 0.38 μM. Significantly, this smartphone platform was successfully applied for detection of TrF in simulated actual samples (i.e., human urine and human serum) with satisfactory results, which provided a new promising strategy for on-site and sensitive discrimination of TrF in actual samples. This research will promote the development of the application of boronic-acid functionalized Zr-MOF in molecularly imprinted sensing of glycoproteins..

Abstract:Surface-enhanced Raman scattering (SERS) is a powerful method for detecting breast cancer-specific biomarkers due to its extraordinary enhancement effects obtained by localized surface plasmon resonance (LSPR) in metallic nanostructures at hotspots. In this research, gold nanostars (AuNSs) were used as SERS probes to detect a cancer biomarker at very low concentrations. To this end, we combined molecularly imprinted polymers (MIPs) as a detection layer with SERS for the detection of the biomarker CA 15-3 in point-of-care (PoC) analysis. This required two main steps: (i) the deposition of MIPs on a gold electrode, followed by a second step (ii) antibody binding with AuNSs containing a suitable Raman reporter to enhance Raman signaling (SERS). The MPan sensor was prepared by electropolymerization of the monomer aniline in the presence of CA 15-3. The template molecule was then extracted from the polymer using sodium dodecyl sulfate (SDS). In parallel, a control material was prepared in the absence of the protein (NPan). Surface modification for the control was performed using electrochemical techniques such as cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The performance of the sensor was evaluated using the SERS technique, in which the MPan sensor is first incubated with the protein and then exposed to the SERS probe. Under optimized conditions, the device showed a linear response to CA 15-3 concentrations from 0.016 to 248.51 U mL-1 in a PBS buffer at pH 7.4 in 1000-fold diluted serum. Overall, this approach demonstrates the potential of SERS as an optical reader and opens a new avenue for biosensing applications.

Abstract:A surface-enhanced Raman scattering(SERS) optical fiber sensor was prepared by the laser-induced deposition of Ag nanoparticle membrane on a silica optical fiber tip, which was applied to the real time SERS spectral monitoring on the biorecognition of biotin/avidin. The bioidentification of biotin/avidin was carried out through a indirect method, in which the bioidentification is based on the SERS response signal of a labeled dye(Atto610) after its fluorescence has been quenched totally by the deposited Ag nanoparticle membrane. By SERS monitoring the bioidentification process of biotin/avidin, it has been found that this recognition process is finished in 40 min. The lowest detection concentration of biotin is 1.0×10−7 mg/mL. This research is promising in the application of immunoassays on line and in vivo.
