Abstract:Gold nanoclusters (Au NCs) are a type of emerging ECL emitter with molecule-like properties and low toxicity that hold great potential for sensing application. However, the application of Au NCs in ECL sensing is still limited due to their low ECL efficiency. In this work, we provided an effective way to enhance the ECL efficiency of Au NCs. By doping Ag on GSH-protected Au NCs to form bimetallic clusters (GSH-Ag/Au NCs), the ECL efficiency of Au NCs was greatly improved. Based on the enhanced ECL signal of Au NCs, an ultrasensitive ECL sensor was constructed for the detection of dopamine (DA). DA exhibited a prominent ECL quenching effect towards the formed bimetallic GSH-Ag/Au NCs which can be used for DA detection. The proposed ECL sensor exhibited excellent sensitivity, selectivity and stability and had a wide linear range from 10 nM to 1 mM with a low detection limit of 2.3 nM (S/N = 3). More importantly, this work provided a potential method to improve the ECL properties of Au NCs and widen their analytical application.
High-Efficient Electrochemiluminescence of DNA-Au Ag Nanoclusters with Au NPs@Ti3C2 as a Novel Coreaction Accelerator for Ultrasensitive Biosensing.

Abstract:In this work, an ultrasensitive electrochemiluminescence (ECL) biosensor was constructed based on DNA-stabilized Au Ag nanoclusters (DNA-Au Ag NCs) as the efficient luminophore and Au NPs@Ti3C2 as a new coreaction accelerator for determining microRNA-221 (miRNA-221) related to liver cancer. Impressively, DNA-Au Ag NCs were stabilized by the high affinity of the periodic 3C sequence, exhibiting an excellent ECL efficiency of 27% compared with classical BSA-Au Ag NCs (16%). Moreover, the Au NPs@Ti3C2 nanocomposites, as a new coreaction accelerator, were first introduced to accelerate the production of abundant sulfate free radicals (SO4•-) for promoting the ECL efficiency of DNA-Au Ag NCs in the DNA-Au Ag NCs/Au NPs@Ti3C2/S2O82- ternary system due to the energy band of Au NPs@Ti3C2 being well-matched with the frontier orbital of S2O82-. Furthermore, the trace target (miRNA-221) could drive the rolling circle amplification to generate an amount of output DNA with periodic 3C and 10A sequences. Through covalent bonds on the surface of poly A and Au NPs, the distance between the luminophor and the coreaction accelerator could be narrowed to further enhance the detection sensitivity. As a result, the constructed sensor has been applied for the ultrasensitive detection of miRNA-221 with a low detection limit of 50 aM and successfully monitored miRNA-221 in MHCC-97L and HeLa cell lysates. This strategy could be utilized for guiding the synthesis of light-emitting DNA-metal NCs, which has great potential in the construction of ultrasensitive biosensors for the early diagnosis of diseases.

Abstract:Electrogenerated chemiluminescence (ECL) of semiconductor quantum dots (QDs) has been considered as a powerful technique in the fabrication of biosensor, however, high-toxicity of heavy metal ion containing in QDs severely limits their further applications, and the search for the alternative benign nanomaterials with high ECL efficiency is urgent. Herein, ECL behavior of eco-friendly silicon quantum dots (SiQDs) was reported in neutral aqueous condition. Stable and intense cathodic ECL emission was obtained in phosphate buffer solution (PBS) with K2S2O8 as coreactant. ECL resonance energy transfer (ECL-RET) system was established with SiQDs ECL as energy donor and gold nanoparticles (AuNPs) as energy acceptor, based on which a novel ECL biosensor was fabricated. AuNPs was connected at the terminal of hairpin DNA to form a signal probe. When the probe was modified on SiQDs, ECL-RET occurred due to the short distance between AuNPs and SiQDs, resulting in the apparent decrease of ECL signal. Target DNA can open the loop of hairpin DNA, and move AuNPs away from the electrode surface. As a result, the ECL-RET process was hampered, and the ECL emission resumed. The increased ECL signals varied linearly with the target DNA concentrations in the range of 0.1fM to 1pM with the detection limit of 0.016fM (3σ). The proposed ECL sensor exhibited highly sensitivity and good selectivity in the detection of target DNA.

Abstract:Aimed at the facile detection of lengthy DNA molecules, an easily operated sandwich-type electrochemiluminescence (ECL) DNA biosensor was constructed on a glassy carbon electrode (GCE) based on CdTe quantum dots coated hollow ZnO nanoparticles (CdTe-ZnO NPs)-S2O82- ECL system in this work. To fabricate a high-performance protocol, the GCE surface was successively modified by graphene nanosheet (GS), carbon nanotube (CNT) and gold nanoparticles (AuNPs) to form AuNPs dotted CNT-GS composites (Au@CNT-GS) platform, which improved the electronic transmission rate as well as increased the amount of immobilized capture probe CMV-F (S-1). For further ultrasensitive, stable and low-potential ECL detection, CdTe-ZnO NPs were synthesized, and employed to label signal probe T7 promoter (S-3). Based on the hybridization effect, the immobilized capture probe S-1, target DNA and labeled signal probe S3 formed a sandwich-type DNA complex, which produced the ECL emission in the presence with S2O82- coreactant. Under optimal conditions, the DNA ECL biosensor showed a good linear range over 10(-14) M to 10(-19) M with a low detection limit of 0.61 x 10(-19) M. The proposed strategy demonstrates a reproducible, stable, and potent method that can be expanded to detect the genome which exists in living cells.

Abstract:The detection of specific DNA sequences provides a powerful tool for early diagnosis and prevention of disease. Herein, an enzyme-free electrochemiluminescence (ECL) DNA biosensor was developed by coupling inorganic-organic nanocomposite as the luminophore and ZnCo2O4 as the efficient accelerator. ZnCo2O4 nanoparticles (ZCO NPs), derived from Co and Zn based bimetallic zeolite imidazole framework, was used as the ECL substrate due to their large surface area and excellent catalytic effect. AgInS2-ZnS quantum dots (ZAIS) were covalently linked with meso-tetra(4-carboxyphenyl)-porphine (TCPP) to form ZAIS/TCPP inorganic-organic nanocomposites, which were used for the first time as the cathodic ECL luminophore with the coreactant of K2S2O8. Detection of target DNA (tDNA) about chronic myeloid leukemia (CML) was achieved based on dual-signal amplification via cascade toehold-mediated strand displacement reaction (TMSDR). On the basis of experimental results, a limit of detection (LOD) of 200 aM was achieved in the range of 0.5 fM to 1 nM, which showed the excellent performance of this biosensor. This work not only indicates that inorganic-organic nanocomposites as the ECL luminophore is a feasible strategy for efficient emission, but also expands the application of nanomaterials derived from metal organic frameworks.
Highly Sensitive Non-Stripping Gold Nanoparticles-Based Chemiluminescent Detection of Dna Hybridization Coupled to Magnetic Beads

Abstract:We have developed a new nanoparticle (NP)-based chemiluminescent (CL) method for the sensitive detection of DNA hybridization. The "sandwich-type'' DNA technique comprises NH2-functionalized capture DNA immobilized on magnetic beads that can hybridize with one end of the target DNA. The other end of the target is recognized by a reporter DNA probe labeled with Au NPs on its 3-terminus. The hybridization events were monitored by a simple and direct luminol-AgNO3 CL reaction with a detection limit of 0.5 pM through a non-stripping Au NPs catalysis. Moreover, the CL reaction of non-stripping Au NPs-luminol-AgNO3 offers the advantages of being simpler and faster as compared to previous stripping Ag NPs and CuS methods. This simple and easy-to-use technique holds great promise for clinical applications of DNA hybridization.

Abstract:Since the electrogenerated chemiluminescence (ECL) of silicon nanoparticles (NPs) was reported in 2002, miscellaneous nanomaterials with various sizes and shapes have been employed as ECL nanoemitters for bioanalysis. Elucidation of the ECL derivation from these nanoemitters and pertinent biofunctionalization with multitudinous biomolecules can offer excellent ECL signal-transduction platforms for fabricating novel biosensing devices. In this review, we comprehensively describe retrospective and recent advances in NPs-based ECL and related biosensing methodologies, and review their analytical applications in the detection of small biological molecules, enzymatic sensing, immunoassay, DNA analysis and cytosensing.
A pharmacologic analysis of chronotropic and inotropic responses to direct electrical stimulation of the isolated, blood-perfused canine atrium.

Abstract:Electrical stimulation on the posterior portion in the caval margin of the right atrium caused a biphasic chronotropic and inotropic response, i.e., a negative response followed by a positive response, in the isolated blood-perfused canine atrial preparation. The threshold of pulse duration for inducing negative responses was approximately 0.05 msec at 2 v and 20 Hz, but that for inducing positive responses was approximately 0.5 msec. When the voltage and pulse duration of electrical stimulation were increased, both the degree of negative and positive responses became greater. However, muscle contracture was frequently induced by increasing the voltage or pulse duration. The optimal frequency was 20 to 40 Hz for inducing marked chronotropic and inotropic responses. The negative chronotropic and inotropic response to subthreshold electrical stimulation was significantly suppressed by the treatment with tetrodotoxin, hexamethonium or atropine, while it was enhanced by physostigmine treatment. These suggest that subthreshold electrical stimulation readily caused a release of acetylcholine by excitation of parasympathetic preganglionic fibers in this atrial preparation. The positive chronotropic and inotropic response was significantly suppressed not only by treatment with propranolol but also by tetrodotoxin, while it was enhanced by imipramine, hexamethonium, or atropine. From these results, it is indicated that this blood-perfused preparation is very suitable to evaluate both cardiac mechanisms controlled by autonomic nervous system and drug actions on effects of endogenous hormone.
Electrochemiluminescence Ratiometry: a New Approach to DNA Biosensing.

Abstract:Inspired by dual-wavelength fluorescence ratiometric method which could reduce the influence from the environmental change, here, we present a novel dual-potential electrochemiluminescence (ECL) ratiometric sensing approach. CdS nanocrystal (NC) and luminol as two different ECL emitters are employed. ECL from CdS NCs coated on glassy carbon electrode at -1.25 V (vs SCE) could be quenched by closely contacted Pt nanoparticles (NPs) via a biological binding event, while ECL from luminol at +0.45 V (vs SCE) could be enhanced by the same Pt NPs, in the presence of their common coreactant of H₂O₂. Thus, the quenching of ECL from CdS NCs and the enhancement of ECL from luminol could indicate the same biological binding event. With the mp53 oncogene as a model DNA molecule, a molecular beacon (MB) containing a 20-base loop, which is complementary with the mp53 oncogene, is immobilized on CdS NCs/GCE first; Pt NPs are then captured on CdS NCs surface by DNA hybridization between the MB and mp53 oncogene labeled on Pt NPs. By measuring the ratio of ECL intensities at two excitation potentials, this approach could sensitively detect the concentration of target DNA in a wide range from 5.0 fM to 1.0 pM. The sensing scheme is general and can be utilized for many other biological binding events.

Abstract:Herein, CuS colloidal nanocrystals (NCs) with adjustable band gap and good film forming ability have been synthesized as new ECL materials. Furthermore, the band gap and oxygen vacancy (OV) content of CuS NCs are regulated by Al3+ doping, which significantly improves the ECL response of CuS NCs. First, the band gap of CuS-Al NCs decreases after doping with Al3+, which makes it easier for electrons to transition across the band gap. At the same time, the oxygen vacancy of CuS-Al NCs increases, which is conducive to improving the conductivity and promoting charge transfer, thus improving the ECL performance of CuS-Al NCs. Circulating tumor DNA (ctDNA) is an important tumor marker, and its sensitive monitoring is of great significance for tumor diagnosis, treatment, and prognosis detection. Therefore, an ECL biosensor for ultrasensitive detection of circulating tumor DNA (ctDNA) was prepared by using CuS-Al NCs as luminescent material and combining multiple antidromic hybrid chain reaction (anti-HCR) strategy mediated by the target. Compared with the process of target-induced HCR generation, this strategy first forms multiple HCR products and then destroys the already formed HCR products by target-induced destruction, which enhances the sensitivity of target response and improves the reaction efficiency. The constructed biosensor has good detection performance, and the detection limit is as low as 2.74 aM. This work puts forward the luminescence phenomenon of colloidal nanocrystals as new ECL materials, which broadens the application of ECL technology in ultrasensitive biochemical detection.

Abstract:A novel confinement-induced ECL enhancement (CIECL) sensing system has been constructed based on copper clusters (Cu NCs) and nano-vesicles in this work. The CIECL strategy resolved the shortcoming of poor stability and low luminous intensity of Cu NCs. Firstly, nano-vesicles were prepared through liquid-liquid self-assembly with amyl gemini quaternary ammonium salt and anionic surfactant. Subsequently, Cu NCs were confined in the nano-vesicles by electrostatic interaction. Due to the restriction on the intramolecular vibration and rotation of the ligands of Cu NCs, the Cu NC/nano-vesicle-based CIECL system showed the significant luminescence enhancement, good stability and high biocompatibility. Additionally, the Au nanoparticles-array with significant surface plasmon resonance effect has been prepared to further amplify the ECL intensity. Finally, the CIECL-based biosensor has been successfully used to detect miRNA-145 in clinic samples in the linear range of 100 fM to 100 nM, which displayed the potential value in the vascular calcification analysis.

Abstract:A new electrochemiluminescence (ECL) DNA assay is developed using quantum dots (QDs) as DNA labels. When nanoporous gold leaf (NPGL) electrodes are used, sensitivity of the ECL assay is remarkably increased due to ultra-thin nanopores. In this assay, target DNA (t-DNA) is hybridized with capture DNA (c-DNA) bound on the NPGL electrode, which is fabricated by conjugating amino-modified c-DNA to thioglycolic acid (TGA) modified at the activated NPGL electrode. Following that, amino-modified probe DNA is hybridized with the t-DNA, yielding sandwich hybrids on the NPGL electrode. Then, mercaptopropionic acid-capped CdTe QDs are labeled to the amino group end of the sandwich hybrids. Finally, in the presence of S2O82− as coreactant, ECL emission of the QD-labeled DNA hybrids on the NPGL electrode is measured by scanning the potential from 0 to −2V to record the curve of ECL intensity versus potential. The maximum ECL intensity (Im,ECL) on the curve is proportional to t-DNA concentration with a linear range of 5×10−15 to 1×10−11mol/L. The ECL DNA assay can be used to determine DNA corresponding to mRNA in cell extracts in this study.
Electrochemiluminescence Energy Resonance Transfer System Between RuSi Nanoparticles and Hollow Au Nanocages for Nucleic Acid Detection

Abstract:This paper describes an electrochemiluminescence resonance energy transfer (ECL-RET) system using Ru(bpy)(3)(2+)-doped silica nanoparticles (RuSi NPs) as the ECL donor and hollow Au nanocages as the ECL acceptor. Tetrahedron DNA (TD) was used to construct the biosensing interface and control the distance (4.8 nm) between the ECL donor-acceptor pairs. The surface plasmon resonance (SPR) nanostructures, Au nanocages were assembled via the hairpin based sandwich assay. Due to the well overlap between the plasmon absorption spectrum of Au nanocages (628 nm) and the ECL emission spectrum of RuSi NPs (620 nm), high efficient energy transfer could occur. Subsequent cyclic DNA amplification further increased the binding amount of Au nanocages. Since the ECL inhibition is closely related with the binding amount of Au nanocages, a general "signal-off" ECL bioassay could thus be tailored with high sensitivity. At the optimized conditions, this ECL-RET system performed well with great stability and repeatability for nucleic acid detection in the range from 1.0 fM to 10 pM. This work manifested the great promise of hollow Au nanocages for an ECL-RET biosensor that to the best of our knowledge has not been reported. We believe that it could inspire more interest in the design and development of numerous other SPR nanostructures for advanced ECL-RET biosensors.

Abstract:Although Ru(bpy)3 2+ -doped silica nanoparticles have been widely explored as the labelling tags for electrochemiluminescence (ECL) sensing different targets, the poor electrical conductive properties of the silica nano-matrix greatly limit their ECL sensitivity. Therefore, a novel scheme to overcome this drawback on Ru(bpy)3 2+ -doped silica nanoparticles ECL is desirable. Here, a new scheme for this purpose was developed based on electrochemically depositing a nanoscale chitosan hydrogel layer on the carbon nanotube (CNT) surface to form chitosan hydrogel shell@CNT core nanocomposites. In this case, the nanoscale chitosan hydrogel layer only formed on the CNT surface due to the superior electrocatalytic effect of CNT on H+ reduction compared with the basic glass carbon electrode. Due to both the superhydrophilic properties and polyelectrolyte features of nanoscale chitosan hydrogel on the CNT surface, chemical affinity as well as the electric conductivity between Ru(bpy)3 2+ -doped silica nanoparticles and CNT were obviously enhanced, and then the ECL effectivity of Ru(bpy)3 2+ inside silica nanoparticles was improved. Furthermore, based on the discriminative interaction of these Ru(bpy)3 2+ -doped silica nanoparticles towards both the ssDNA probes and the ssDNA probe/miRNA complex, as well as the specific adsorption effect of these nanoparticles on the nanoscale chitosan shell@Nafion/CNT core-modified glass carbon electrode, a highly sensitive ECL method for miRNA determination was developed and successfully used to detect miRNA in human serum samples.
Cu Single-Atom Nanozyme-Mediated Electrochemiluminescence Biosensor for Highly Sensitive Detection of MicroRNA-622

Abstract:MicroRNA (miRNA) detection is a critical aspect of disease diagnosis, and recent studies indicate that miRNA-622 could be a potential target for lung cancer. Herein, Cu single atoms were anchored on graphitic carbon nitride (Cu SAs@CN) as a coreaction accelerator applied in luminol-H2O2 system, thereby establishing an efficient and sensitive electrochemiluminescence (ECL) biosensor for miRNA-622 detection. Cu SAs@CN was explored to possess excellent enzyme-like activities that promote the generation of abundant reactive oxygen species, which amplified ECL emission. Meanwhile, in order to improve the accuracy and sensitivity for miRNA-622 detection, the highly specific trans-cleavage ability of CRISPR/Cas12a was combined with a catalytic hairpin assembly strategy. Therefore, an ECL biosensor for miRNA-622 detection was systematically constructed as a proof of concept, achieving an ultralow limit of detection of 1.09 fM, and the feasibility was demonstrated in human serum samples. The findings of this research provide a promising strategy to enhance the ECL response using versatile single-atom catalysts, thus advancing the development of ECL biosensing applications.
Amplified Electrochemiluminescence Detection of DNA-binding Protein Based on the Synergy Effect of Electron and Energy Transfer Between CdS Nanocrystals and Gold Nanoparticles

Abstract:In this work, we firstly enunciated the presence of synergy effect between the electron and energy transfer in an electrochemiluminescence (ECL) system involving the CdS NCs and Au NPs, based on which an amplified ECL biosensor was constructed for sensitive detection of DNA–protein interactions. Specifically, Au NPs modified on electrode, following by assembled probe DNA. Then the modified GCE was hybridized with SiO2@CdS/DNA conjugates. With DNA duplex as a rigid spacer, Au NPs could not only accelerate the electron transfer but also produce the surface plasma resonance upon excited ECL of CdS NCs. However, the enhanced ECL could be greatly suppressed by the binding of target protein. The prepared biosensor possesses excellent analytical performance with the linear range of target protein from 0.015 to 150nM.

Abstract:Development of sensitive detection methods for microRNAs has realistic application value for early clinical analysis and accurate diagnosis. In this study, a triple-helix molecular switch electrochemiluminescence resonance energy transfer (ECL-RET) nanoamplifier was designed to construct an electrochemiluminescence (ECL) biosensor for microRNA determination. The newly synthesized S-doped Lu2O3, which shows a 3 times better ECL performance than Lu2O3, was chosen as the donor in this ECL-RET system. Accordingly, Ag2S quantum dots (QDs) were used as the matched acceptor. They exhibited overlapping spectra and efficient energy transfer between each other. Furthermore, using a triple-helix switch structure with an improved quenching effect for signal amplification and a nano-DNA walker transformational system as a powerful method for target amplification, a supernanoamplifier was achieved. As a consequence, the proposed ECL biosensor for microRNA-141 detection exhibited good analytical performance with a low detection limit (16 aM). The sensor not only led to the development of a novel ECL-RET pair but also provided a strong nanoamplifier for biosensing platform construction, which may offer some new considerations regarding material design, signal amplification, and show promising application value in biomedical and clinical diagnosis.
