Abstract:Cadmium(Cd) is a widespread and one of the most toxic heavy metals, threatening human health. As a pioneer fiber crop, hemp (Cannabis sativa L.) has a great potential in Cd-polluted soil repair, but molecular mechanisams of hemp supporting the process of soil decontamination are still unknown. In this study, the key factors for differential Cd tolerance in two contrasting hemp cultivars (Cd-tolerant: Ym, Cd-sensitive: Nx) were investigated. Ym has a stronger transport and accumulation capacity than Nx. Furthermore, Cd detoxification in Ym is stronger than in Nx. Phenotype and 3,3'-diaminobenzidine(DAB) staining observation indicated that Ym was more tolerant to cadmium stress than Nx, as well as malondialdehyde(MDA) contents, superoxide dismutase (SOD) and peroxidase(POD) activities showed Ym has a stronger detoxification ability than Nx. Compared the transcriptome analysis between Ym and Nx, many genes involved in heavy metal transport and oxidation-reduction progress were identified between the two varieties. A schematic representation of main processes about differential Cd transport and detoxification was described, based on these results, which suggested that the higher Cd tolerance in Ym due to the key genes encoding some specific Cd transporter, defense system, special proteins that have a high expression in Ym.

Abstract:Excessive accumulation of cadmium (Cd) in vegetables poses a serious threat to human health; therefore, it is urgent to screen and cultivate vegetable cultivars with low Cd accumulation in the edible parts. Eggplant has a high tendency for Cd accumulation, but research on its low Cd accumulation cultivars is still rare. In this study, to screen low-Cd cultivars, 30 eggplant cultivars were screened using soils containing 0.22 mg/kg, 2.9 mg/kg (low-Cd), and 4.7 mg/kg of Cd (high-Cd). MYCQ and ZGQ were confirmed as low-Cd cultivars, BXGZ and WCCQ were confirmed as high-Cd cultivars, and a 2.52–3.88-fold difference in Cd concentration was observed in their fruits. The subcellular distribution revealed that the root cell wall and vacuole Cd concentrations of a typical low-Cd cultivar (MYCQ) were significantly higher than those of a typical high-Cd cultivar (BXGZ); however, the Cd concentrations in the cell wall and vacuole in fruits, leaves, and stems were significantly lower in MYCQ than in BXGZ. These results indicated that the low-Cd cultivars of eggplant could lessen Cd toxicity through the elevated Cd retention and sequestration levels of root cell walls and vacuoles, thus reducing Cd transport from roots to aboveground tissues, leading to low Cd accumulation. The findings of this study can provide a physiological and biochemical foundation for the screening and breeding of low-Cd cultivars of fruit vegetables and demonstrates that the application of low-Cd cultivars is necessary for food safety in humans.

Abstract:The use of Cd-excluding cultivars on the most widespread soils moderately polluted with Cd is a sustainable solution for the safe agricultural production. In order to explore the defense mechanisms and differences in the cadmium (Cd) uptake from soils and accumulation in plant tissues between low-accumulator and excluder plants, Cd concentrations/loads in plant tissues and impact on plant growth and physiological properties (photosynthetic characteristics, antioxidant enzymes and related indices) of the cultivar Solanum melongena Liaoqie No.3 (SML) and the wild species Solanum torvum (STO) grown in soils containing from 0.24 to 18.61 mg kg−1 Cd (i.e. from the natural Cd content through moderate to high pollution level) were studied in the pot experiments carried out under natural light conditions. It was found that the major protective barrier in the more sensitive to Cd toxicity excluder species (STO) was a root system accumulating 2-3-fold lesser Cd loads through the induction of dehydration/drought stress than a more Cd-tolerant low-accumulating plant (SML). A supportive defense mechanism appeared to be a reduced Cd enrichment of a xylem sap and its increased flow rate during long-distance transport from roots to leaves, occurring in the both types of plants, but with a lesser intensity in a more Cd-tolerant low-accumulating species. Due to maintaining plant-specific Cd-adaptive level, growth and biomasses of both species were basically the same, while due to the considerably lower Cd accumulation, photosynthetic parameters were higher in Cd-excluder STO species, and the amounts of lipid peroxidation (MDA) and free proline were higher in Cd low-accumulating SML cultivar. Concurrence of Cd and drought-resistance mechanism and better physiological parameters in Cd-excluder suggest promotion of this mechanism in cultivars under the climate change conditions.

Abstract:Soil cadmium (Cd) contamination severely threatens human health. Therefore, screening and breeding low-Cd absorption cultivars of cherry tomato (Solanum lycopersicum L.) is essential to restrict human Cd intake. In this study, a hydroponic experiment was conducted to perform a comparative transcriptome analysis of the leaves of two cherry tomato cultivars with different Cd contents under different Cd stress (0, 10, and 50 μM), for the purpose of exploring the differences in the transcriptional responses to Cd stress between the two cultivars. Our results revealed that the Cd content in the leaves of HLZ (Hanluzhe; a low-Cd accumulation cultivar) was significantly lower than that in the leaves of LFC (Lvfeicui; a high-Cd accumulation cultivar). Transcriptome analysis showed that the different expression genes (DEGs) were mainly involved in plant hormone signal transduction, antioxidant enzymes, cell wall biosynthesis, and metal transportation. In the LFC leaves, DEGs in the IAA signal transduction and antioxidant enzymes exhibited higher transcription levels. However, the DEGs in the ETH signal transduction demonstrated a lower transcription level compared to that of HLZ. Over-expressed genes in the pectin biosynthesis and pectin methylesterase (PME) of the LFC leaves might result in the trapping of Cd by increased levels of low-methylated pectin around the cell wall. Furthermore, Cd transporter genes, such as HMA5, NRAMP6, CAX3, ABCC3, and PDR1, were up-regulated in the HLZ leaves, indicating that the HLZ cultivar comprised an active Cd transport capacity from apoplast to vacuolar. This may contribute to the low Cd concentration observed in the HLZ leaves. Overall, our study provides a molecular basis for tomato screening and breeding.

Abstract:The toxic heavy metal cadmium (Cd) is easily absorbed and accumulated in crops and affects human health through the food chains. Sunflower ( Helianthus annuus L.) is a globally important oil crop. In this study, two sunflower cultivars 62\3 (high Cd) and JB231AC (low Cd), were chosen to compare physiological and transcriptomic responses at different Cd concentrations (0, 25, 50, and 100 μM). The results showed that JB231AC had better Cd tolerance than 62\3. The contents of H 2 O 2 and MDA (malondialdehyde) in 62\3 were lower than that in JB231AC under Cd stress, but the activities of SOD (superoxide dismutase) and POD (peroxidase) in JB231AC were higher than in 62\3, which indicated that JB231AC had a strong ability to remove reactive oxygen species (ROS)-induced toxic substances. Many deferentially expressed ABC ( ATP-binding cassette ) and ZIP ( Zn-regulated transporter, Iron-regulated transporter-like protein ) genes indicated that the two gene families might play important roles in different levels of Cd accumulation in the two cultivars. One up-regulated NRAMP ( Natural resistance-associated macrophage protein ) gene was identified and had a higher expression level in 62\3. These results provide valuable information to further understand the mechanism of Cd accumulation and provide insights into breeding new low Cd sunflower cultivars.

Studies on the Criteria of Cadmium Pollution in Growth Media of Vegetable Crops Based on the Hygienic Limit of Cadmium in Food

Abstract:A pot experiment and a sandy culture experiment grown with three vegetable crops of Chinese cabbage (B. chinensis L. cv. Zao-Shu 5), winter greens (B. var. rosularis Tsen et Lee cv. Shang-Hai-Qing), and celery (A. graveolens L. var. dulce DC. cv. Qing-Qin) were conducted, respectively. The initial soil and four incubated soils with different extractable cadmium (Cd) levels (0.15, 0.89, 1.38, 1.84, and 2.30 mg Cd kg(-1) soil) were used for the pot experiment. Five treatments were designed (0, 0.0625, 0.125, 0.250, and 0.500 mg Cd L-1) in nutrient solution in the sandy culture experiment. Each treatment in both pot and sandy culture experiments was trireplicated. The objectives of the study were to examine Cd accumulation in edible parts of selected vegetable crops, its correlation with Cd concentrations in vegetable garden soil or in nutrient solution, and to evaluate the criteria of Cd pollution in vegetable garden soil and in nutrient solution based on the hygienic limit of Cd in vegetables. Cadmium concentrations in edible parts of the thr-ce selected vegetable crops were as follows: 0.01-0.15 mg kg(-1) fresh weight for Chinese cabbage, 0.02-0.17 mg kg(-1) fresh weight for winter greens, and 0.02-0.24 mg kg(-1) fresh weight for celery in the pot experiment, and 0.1-0.4 mg kg(-1) fresh weight for Chinese cabbage, 0.1-1.4 mg kg(-1) fresh weight for winter greens, and 0.05-0.5 mg kg(-1) fresh weight for celery in the pot experiment (except no-Cd treatment). Cadmium accumulation in edible parts or roots of the vegetable crops increased with increasing of cadmium concentration in the medium (soil or nutrient solution); and cadmium concentrations in edible parts of the test vegetable crops were significantly linearly related to the Cd levels in the growth media (soil and nutrient solution). Based on the regression equations established and the limit of cadmium concentration in vegetable products, the thresholds of Cd concentration in the growth medium evaluated was as follows: 0.5 mg kg(-1) soil of extractable Cd for soil and 0.02 mg L-1 for nutrient solution. The hi-fi capacity for cadmium accumulation in the edible parts of different vegetable crops together with the absence of visual symptoms implies a potential danger for humans.

Abstract:Excessive cadmium (Cd) accumulation in vegetables due to farmland pollution constitutes a serious threat to human health. Eggplant has a tendency to accumulate Cd. To investigate the mechanism of the differences in Cd accumulation levels between high-Cd (BXGZ) and low-Cd (MYQZ) eggplant cultivar, physiological and biochemical indicators and mRNA expression of eggplant were examined using photosynthetic apparatus, biochemical test kits, Fourier transform infrared (FTIR) spectroscopy and transcriptome sequencing, etc. The results of biochemical test kits and FTIR revealed that MYQZ enhanced pectin methylesterase (PME) activity, and lignin and pectin content in the root cell wall, which was associated with the upregulation of PME, cinnamyl-alcohol dehydrogenase and peroxidase (PODs). Higher levels of cysteine and glutathione (GSH) contents and upregulation of genes associated with sulfur metabolism, as well as higher expression of ATP-binding cassette transporters (ABCs), cation exchangers (CAX) and metal tolerance proteins (MTPs) were observed in MYQZ. In BXGZ, the higher stomatal density and stomatal aperture as well as higher levels of Ca 2+ binding protein-1 (PCaP1) and aquaporins and lower levels of A2-type cyclins (CYCA2-1) are consistent with an enhanced transpiration rate in BXGZ. Furthermore, a more developed root system was shown to be associated with higher levels of auxin response factor (ARF19), GATA transcription factors (GATA4, 5 and 11) and auxin efflux carrier component (PIN5) in BXGZ. In conclusion, highly active PME, and higher levels of lignin and pectin in MYQZ are expected to reduce Cd toxicity, while Cd translocation can be inhibited with the help of ABC and other Cd transporters. As for BXGZ, the uptake and translocation of Cd were enhanced by the developed root system and stronger transpiration.

Abstract:Cadmium (Cd) by plants from contaminated soils induces various physiological and biochemical changes resulting reduction in the yield and the quality of the plant products. A pot experiment was performed in order to study Cd induced alterations in physiological characters, yield components and fiber quality of two transgenic cotton cultivars (ZD-90 and SGK3) and an upland cotton standard genotype (TM-1). ZD-90, derived from our laboratory, is a glyphosate-resistant cultivar with the EPSPS-G6 gene, and SGK3 is an insect-resistant cultivar with the Bt gene. Cd stress significantly reduced the seed cotton yield, lint yield, boll numbers per plant, boll weight, net photosynthetic rate, and water use efficiency etc., while increased the activity of several antioxidant enzymes such as peroxidase (POD) and superoxide dismutase (SOD) in the three cotton cultivars. The Cd accumulation in different parts of cotton plant increased in the following order: fiber < seed < seed shell < root < leaf < shoot < boll shell < petiole. The least affected cultivar by Cd treatments in seed cotton yield and bolls per plant among the three cotton cultivars was SGK3, suggesting that it was less sensitive to Cd stress than the other two cultivars. From the present experiment, it can be well confirmed that the non-harvestable parts of cotton plant accumulated more Cd (BCF=1.854~2.449) than the fiber and seed (BCF=0.089~0.242), indicating that cotton is suitable for cultivation in industrially polluted regions and could be used as a potential crop for cleaning up contaminated soils by phytoremediation technology.

Abstract:The presence of müllerian-origin tissue in the lumbosacral region is extremely uncommon. The authors report two cases of müllerian-origin tissue in that region. In the first case a 33-year-old woman harbored a conus medullaris mass lesion. Spinal dysraphism, tethered cord syndrome (TCS), and diastematomyelia were also present. In the second case a 24-year-old woman presented with low-back pain and a conus medullaris lesion, which was a cause of the TCS. Pathological examination in both cases revealed a uterus-like structure with evidence of fresh and old hemorrhage. The rarity of this lesion and its association with diastematomyelia requires documentation.
Conjoint analysis of physio-biochemical, transcriptomic, and metabolomic reveals the response characteristics of solanum nigrum L. to cadmium stress

Abstract:Cadmium (Cd) is a nonessential element in plants and has adverse effects on the growth and development of plants. However, the molecular mechanisms of Cd phytotoxicity, tolerance and accumulation in hyperaccumulators Solanum nigrum L. has not been well understood. Here, physiology, transcriptome, and metabolome analyses were conducted to investigate the influence on the S. nigrum under 0, 25, 50, 75 and 100 μM Cd concentrations for 7 days. Pot experiments demonstrated that compared with the control, Cd treatment significantly inhibited the biomass, promoted the Cd accumulation and translocation, and disturbed the balance of mineral nutrient metabolism in S. nigrum , particularly at 100 μM Cd level. Moreover, the photosynthetic pigments contents were severely decreased, while the content of total protein, proline, malondialdehyde (MDA), H 2 O 2 , and antioxidant enzyme activities generally increased first and then slightly declined with increasing Cd concentrations, in both leaves and roots. Furthermore, combined with the previous transcriptomic data, numerous crucial coding-genes related to mineral nutrients and Cd ion transport, and the antioxidant enzymes biosynthesis were identified, and their expression pattern was regulated under different Cd stress. Simultaneously, metabolomic analyses revealed that Cd treatment significantly changed the expression level of many metabolites related to amino acid, lipid, carbohydrate, and nucleotide metabolism. Metabolic pathway analysis also showed that S. nigrum roots activated some differentially expressed metabolites (DEMs) involved in energy metabolism, which may enhance the energy supply for detoxification. Importantly, central common metabolism pathways of DEGs and DEMs, including the "TCA cycle", "glutathione metabolic pathway" and "glyoxylate and dicarboxylate metabolism" were screened using conjoint transcriptomics and metabolomics analysis. Our results provide some novel evidences on the physiological and molecular mechanisms of Cd tolerance in hyperaccumulator S. nigrum plants.

Abstract:Species difference in Cd accumulation is important for selection of agronomic technologies aimed at producing low-Cd vegetables. Six vegetable species (Chinese leek, pakchoi, carrot, radish, tomato and cucumber) were grown in pot and field experiments to study the accumulation of Cd under different conditions. In the field trial (Cd 2.55 mg kg(-1)), Cd concentrations in the edible parts ranged from 0.01 to 0.1 mg kg(-1) and were below the permissible limits (0.2 mg kg(-1) for pakchoi and leek; 0.1 mg kg(-1) for carrot and radish; 0.05 mg kg(-1) for cucumber and tomato), but exceeded the limit in pakchoi, Chinese leek, carrot and tomato at a Cd addition level of 2.0 mg kg(-1). Plant Cd concentrations increased linearly with the increasing concentration of Cd added to the soil, with the slope of the regression lines varying by 28-fold among the six species. The bioconcentration factor (BCF) varied substantially, and was much higher in the pot experiment than in the field trial. It is concluded that the vegetable species differed markedly in the Cd accumulation and species performed consistently under different growth conditions.

Difference in Cd2+ flux around the root tips of different soybean (Glycine max L.) cultivars and physiological response under mild cadmium stress

Abstract:The purpose of the study was to compare differences in Cd2+ flux in the vicinity of root tips of 20 soybean cultivars under mild Cd stress conditions using non-invasive micro-test technology (NMT). The results indicated that Cd2+ influx to the root tips under mild Cd treatment was higher compared to controls. Cd2+ influx showed an obvious spatial distribution, with the highest Cd2+ influx measured 300 μm from the root tips, and a gradually decrease above and below this site. The cultivar Liaodou32 had a lower Cd uptake (3.40 pmol cm-2 s-1), while Liaodou23 had a relatively higher Cd uptake (66.37 pmol cm-2 s-1). Cluster analysis showed that the order of the average Cd2+ influx of the cultivars at a distance of 300 μm from the root tips was as follows: high-uptake cultivars (61.80 pmol cm-2 s-1)>medium-high-uptake cultivars (33.92 pmol cm-2 s-1)>medium-low-uptake cultivars (19.78 pmol cm-2 s-1)>low-uptake cultivars (4.84 pmol cm-2 s-1). We also analyzed physiological responses of different soybean cultivars to mild Cd stress. The results indicated that mild Cd stress could inhibit soluble protein production and root vigor among individual soybean cultivars. Moreover, stress increased SOD, CAT and POD activities and MDA content in root tissues. It should be noted that the physio-biochemical indicators of low-uptake cultivars did not change significantly after exposure to mild Cd stress compared to controls. Pearson's correlation analyses showed that all physio-biochemical indicators were significantly positively associated with influx, except of root SP and biomass. PCA analysis demonstrated that root vigor was a dominant factor causing the differences in Cd tolerance among different soybean seedling cultivars. NMT is of great significance for safe utilization of contaminated soil to distinguish the cultivars with different enrichment capacity for heavy metals from different crop cultivars.
Identification of Barley Varieties Tolerant to Cadmium Toxicity

Abstract:Two successive hydroponic experiments were carried out to identify barley varieties tolerant to Cd toxicity via examining Soil–Plant Analyses Development (SPAD) value, plant height, leaves and tillers per plant, root number and volume, and biomass accumulation. The results showed that SPAD values (chlorophyll meter readings), plant height, leaf number, root number and volume, and biomass accumulation of shoot/root were significantly reduced in the plants grown in 20 μM Cd nutrient solution compared with control, and the uptake and translocation of Zn, Mn, and Cu was also strictly hindered. Furthermore, there was a highly significant difference in the reduction in these growth parameters among varieties, and varieties “Weisuobuzhi” and “Jipi 1” showed the least reduction both in the two experiments, suggesting their high tolerance to Cd toxicity, while “Dong 17” and “Suyinmai 2” with the greatest reduction and the toxicity symptoms appeared rapidly and severely, denoting as Cd-sensitive varieties. Significant variety difference in Cd concentration was also found, with Weisuobuzhi containing the highest and Jipi 1 the lowest Cd concentration in shoots.
Intraspecific variation in tomato: Impact on production quality and cadmium phytoremediation efficiency in intercropping systems with hyperaccumulating plant

Abstract:Intercropping with hyperaccumulators can facilitate the safe utilization of cadmium-contaminated soil. However, the effectiveness of this approach is influenced by plant species and varieties, which necessitates research on optimal plant consortia. In this study, 8 tomato varieties (3 cherry tomatoes and 5 common large-fruit tomatoes) were intercropped with Sedum alfredii in a moderately Cd-contaminated vegetable field. The results showed that the Cd concentration in the fruits of common large-fruit tomato varieties under monoculture was 1.03-1.50 mg/kg, while that in the fruits of cherry tomato varieties was 0.67-0.71 mg/kg. After intercropping with S. alfredii, the fruit Cd concentrations of Hangza 501, Hangza 503, and Hangza 108 decreased by 16.42 %, 19.72 %, and 6.76 %, respectively, while those of the other varieties significantly increased, except for those of Hangza 8. In contrast, the shoot Cd concentration of cherry tomatoes was greater than that of large-fruit tomatoes under monoculture. Furthermore, a significant increase in the shoot Cd concentration was noted in the Hangza 501, Hangza 503 and Hangza 603 plants following intercropping. Additionally, intercropping with S. alfredii increased the concentration of soluble sugars in the fruits of Hangza 8, Hangza 501, Hangza 503 and Hangza 603 by 4.66 %, 17.91 %, 10.60 % and 17.88 %, respectively. Intercropping with tomatoes resulted in a decrease in both the biomass and Cd uptake of S. alfredii. Interestingly, the inhibitory effect on S. alfredii was less pronounced when intercropped with cherry tomatoes than when intercropped with large-fruit tomatoes. Among the intercropping treatments, S. alfredii exhibited the greatest total Cd accumulation (0.06 mg/plant) when intercropped with Hangza 503. In conclusion, the cherry tomato variety Hangza 503 was the most suitable for intercropping with S. alfredii and can be used safely for vegetable production and simultaneous phytoremediation of polluted soil. Our findings suggest that strategic selection of tomato varieties can optimize the effectiveness of "phytoextraction coupled with agro-safe production" technology for managing soil Cd concentrations.
Cataloging of Cd Allocation in Late Rice Cultivars Grown in Polluted Gleysol: Implications for Selection of Cultivars with Minimal Risk to Human Health

Abstract:Cadmium (Cd) is a toxic trace metal that has polluted 20% of agricultural land in China where its concentration exceeds the standards for Chinese farmland. Plants are capable of accumulating Cd and other trace metals, but this capacity varies with species and cultivars within a species. Rice is a staple food consumed by half of the global population. In order to select safe late rice cultivars that are suitable late rice cultivars that can be cultivated in for growing in slightly contaminated soil, a two-year field experiment was conducted with 27 in the first year and 9 late rice cultivars in the second year. The results showed that plant Cd concentrations varied among the cultivars, with high magnitudes of variation occurred in straw and grains. Five genotypes including LR-12, LR-17, LR-24, LR-25 and LR-26 were identified as low accumulators for the first year while LR-15 and LR-17 were identified as promising cultivars based on Cd concentration in the polished rice grains (<0.02 mg kg(-1) DW). In addition, these cultivars had favorable traits, including mineral nutrition and grain yield. Therefore, these genotypes should be considered for cultivation in slightly or moderately Cd contaminated soils.
Differential responses of chili varieties grown under cadmium stress

Abstract:Abstract Heavy metal cadmium (Cd) naturally occurs in soil and is a hazardous trace contaminant for humans, animals, and plants. The main sources of Cd pollution in soil include overuse of phosphatic fertilizers, manure, sewage sludge, and aerial deposition. That’s why an experiment was conducted to analyze the effect of Cd toxicity in Capsicum annuum L. by selecting its seven varieties: Hybrid, Desi, Sathra, G-916, BR-763, BG-912, and F1-9226. Cadmium was spiked in soil with four levels, i.e., (0, 3, 4, and 5 mg Cd kg − 1 of soil) for a week for homogeneous dispersion of heavy metal. Chili seeds were sown in compost-filled loamy soil, and 25-day-old seedlings were transplanted into Cd-spiked soil. Cadmium increasing concentration in soil decreased chili growth characteristics, total soluble sugars, total proteins, and amino acids. On the other hand, the activities of antioxidant enzymes were increased with the increasing concentration of Cd in almost all the varieties. Treatment 5 mg Cd/kg application caused − 197.39%, -138.78%, -60.77%, -17.84%, -16.34%, -11.82% and − 10.37% decrease of carotenoids level in chili V2 (Desi) followed by V4 (G-916), V1 (Hy7brid), V7 (F1-9226), V6 (BG-912), V5 (BR-763) and V3 (Sathra) as compared to their controls. The maximum flavonoids among varieties were in V5 (BR-763), followed by V6 (BG-912), V7 (F1-9226), V3 (Sathra) and V1 (Hybrid). Flavonoids content was decreased with − 37.63% (Sathra), -34.78% (Hybrid), -33.85% (G-916), -31.96% (F1-9226), -31.44% (Desi), -30.58% (BR-763), -22.88% (BG-912) as compared to their control at 5 mg Cd/kg soil stress. The maximum decrease in POD, SOD, and CAT was − 31.81%, -25.98%, -16.39% in chili variety V7 (F1-9226) at 5 mg Cd/kg stress compared to its control. At the same time, maximum APX content decrease was − 82.91%, followed by -80.16%, -65.19%, -40.31%, -30.14%, -10.34% and − 6.45% in V4 (G-916), V2 (Desi), V3 (Sathra), V6 (BG-912), V1 (Hybrid), V7 (F1-9226) and V5 (BR-763) at 5 mg Cd/kg treatment as compared to control chili plants. The highest CAT was found in 5 chili varieties except Desi and G-916. Desi and G-916 varieties. V5 (BR-763) and V6 (BG-912) were susceptible, while V1 (Hybrid), V3 (Sathra), and V7 (F1-9226) were with intermediate growth attributes against Cd stress. Our results suggest that Desi and G-916 chili varieties are Cd tolerant and can be grown on a large scale to mitigate Cd stress naturally.

Abstract:A pot experiment and a sandy culture experiment grown with three vegetable crops of Chinese cabbage ( B. chinensis L., cv. Zao-Shu 5), winter greens ( B. var. rosularis Tsen et Lee, cv. Shang-Hai-Qing) and celery (A. graveolens L. var. dulce DC., cv. Qing-Qin) were conducted, respectively. The initial soil and four incubated soils with different extractable Cd (0.15, 0.89, 1.38, 1.84 and 2.30 mg Cd/kg soil) were used for the pot experiment. Five treatments were designed (0, 0.0625, 0.125, 0.250 and 0.500 mg Cd/L) in nutrient solution in the sandy culture experiment. Each treatment in pot and sandy culture experiments was trireplicated. The objectives of the study were to examine Cd accumulation in edible parts of selected vegetable crops, its correlation with Cd concentrations in vegetable garden soil or in nutrient solution, and evaluate the criteria of Cd pollution in vegetable garden soil and in nutrient solution based on the hygienic limit of Cd in vegetables. Cadmium concentrations in edible parts of the three selected vegetable crops were as follows: 0.01-0.15 mg/kg fresh weight for Chinese cabbage, 0.02-0.17 mg/kg fresh weight for winter greens, and 0.02-0.24 mg/kg fresh weight for celery in the pot experiment, and 0.1-0.4 mg/kg fresh weight for Chinese cabbage, 0.1-1.4 mg/kg fresh weight for winter greens, and 0.05-0.5 mg/kg fresh weight for celery in the pot experiment( except no-Cd treatment). The order of the three test vegetable crops for cadmium accumulation in the edible parts was celery > winter greens > Chinese cabbage in both the pot experiment and the sandy culture experiment. Cadmium accumulation in edible parts or roots of the vegetable crops increased with increasing of cadmium concentration in the medium (soil or nutrient solution). And cadmium concentrations in edible parts of the test vegetable crops were significantly linearly related to the Cd levels in the growth media (soil and nutrient solution). Based on the regression equations established and the limit of cadmium concentration in vegetable products, the thresholds of Cd concentration in the growth medium evaluated was as follows: 0.5 mg/kg soil of extractable Cd for soil and 0.02 mg/L for nutrient solution. The high capacity for cadmium accumulation in the edible parts of different vegetable crops together with the absence of visual symptoms implies a potential danger for humans.

Abstract:Cadmium (Cd) is present in all soils, usually as a trace constituent, but it can reach higher levels in agricultural soils. Cd can then be absorbed by plants and become a potential risk to human health. Once taken up by a plant, there are mechanisms for heavy metal detoxification in the plant. Here, a cadmium-tolerant and a cadmium-sensitive rice cultivars were grown hydroponically to investigate the effects of cadmium (Cd) applied at low levels on uptake and transport, subcellular distribution and binding forms of Cd in rice plants. Our results showed that increasing the Cd treatment from 1.0 μM to 5.0 μM Cd increased the shoot Cd content by 55% in the cadmium-tolerant cultivar, and by 108% in the cadmium-sensitive cultivar. For the cadmium-tolerant cultivar, increasing Cd treatment from 1.0 μM to 5.0 μM increased the root Cd content by 116%, whereas for the cadmium-sensitive cultivar, increasing Cd treatment from 1.0 μM to 5.0 μM increased the root Cd content by 80%. Further, the ratio of Cd accumulation in shoots over roots decreased from 0.19 to 0.14 in the cadmium-tolerant cultivar, while it increased from 0.20 to 0.26 in the cadmium-sensitive cultivar, showing that the transportation ability for Cd was different between the two tested rice cultivars. At the higher Cd level of 5.0 μM, most of the Cd in the plants was localized in cell walls and vacuoles in both cultivars, whereas small portions of Cd were distributed in the cytoplasm, suggesting that the important metabolic and physiological processes were not impaired under Cd stress. Furthermore, the major portions of Cd in the cells were combined with organic acids, proteins and polysaccharide, and were consequently detoxified. The difference in the distribution of cadmium in rice plants resulted in the difference in Cd tolerance between the two rice cultivars used. It can be concluded that the retention of Cd in root cell walls, compartmentation of Cd into vacuoles and the suppressed transportation of Cd from roots to shoots are the most important mechanisms involved in the detoxification of Cd in rice plants.

Abstract:Solanum nigrum is a well-documented cadmium (Cd) hyperaccumulator; however, its Cd-induced tolerance capability and detoxification mechanism remain elusive. Hence, a short-term hydroponic experiment was performed in a multiplane glasshouse to determine the influence of Cd toxicity on subcellular distribution, chemical forms, and the physiological responses of cell wall towards Cd stress in a 4-week-old plant. The experiment was conducted following completely randomized design (CRD) with five treatments (n = 4 replicates). The results showed that Cd stress showed dose-dependent response towards growth inhibition. The subcellular distribution of Cd in S. nigrum was in the order of cell wall > soluble fractions > organelles, and Cd was predominantly extracted by 1 M NaCl (29.87~43.66%). The Cd contents in different plant tissues and cell wall components including pectin, hemicellulose 1 (HC1), hemicellulose 2 (HC2), and cellulose were increased with the increase in Cd concentrations; however, the percentage of Cd concentration decreased in pectin and cellulose. Results of the polysaccharide components such as uronic acid, total sugar contents, and pectin methylesterase (PME) activity showed Cd-induced dose-dependent increase relative to exposure Cd stress. The pectin methylesterase (PME) activity was significantly (p < 0.05) enhanced by 125.78% at 75 μM Cd in root, 105.78% and 73.63% at 100 μM Cd in stem and leaf, respectively. In addition, the esterification, amidation, and pectinase treatment of cell wall and Fourier transform infrared spectroscopy (FTIR) assay exhibited many functional groups that were involved in cell wall retention Cd, especially on carboxyl and hydroxyl groups of cell wall components that indicated that the –OH and –COOH groups of S. nigrum cell wall play a crucial role in Cd fixation. In summary, results of the current study will add a novel insight to understand mobilization/immobilization as well as detoxification mechanism of cadmium in S. nigrum.
