Abstract:L-valine is an essential branched-chain amino acid that cannot be synthesized by the human body and has a wide range of applications in food, medicine and feed. Market demand has stimulated people's interest in the industrial production of L-valine. At present, the mutagenized or engineered Corynebacterium glutamicum is an effective microbial cell factory for producing L-valine. Because the biosynthetic pathway and metabolic network of L-valine are intricate and strictly regulated by a variety of key enzymes and genes, highly targeted metabolic engineering can no longer meet the demand for efficient biosynthesis of L-valine. In recent years, the development of omics technology has promoted the upgrading of traditional metabolic engineering to systematic metabolic engineering. This whole-cell-scale transformation strategy has become a productive method for developing L-valine producing strains. This review provides an overview of the biosynthesis and regulation mechanism of L-valine, and summarizes the current metabolic engineering techniques and strategies for constructing L-valine high-producing strains. Finally, the opinion of constructing a cell factory for efficiently biosynthesizing L-valine was proposed.

High-yield production of L-valine in engineered Escherichia coli by a novel two-stage fermentation

Abstract:L-valine is an essential amino acid and an important amino acid in the food and feed industry. The relatively low titer and low fermentation yield currently limit the large-scale application of L-valine. Here, we constructed a chromosomally engineered Escherichia coli to efficiently produce L-valine. First, the synthetic pathway of L-valine was enhanced by heterologous introduction of a feedback-resistant acetolactate acid synthase from Bacillus subtilis and overexpression of other two enzymes in the L-valine synthetic pathway. For efficient efflux of L-valine, an exporter from Corynebacterium glutamicum was subsequently introduced. Next, the precursor pyruvate pool was increased by knockout of GTP pyrophosphokinase and introduction of a ppGpp 3′-pyrophosphohydrolase mutant to facilitate the glucose uptake process. Finally, in order to improve the redox cofactor balance, acetohydroxy acid isomeroreductase was replaced by a NADH-preferring mutant, and branched-chain amino acid aminotransferase was replaced by leucine dehydrogenase from Bacillus subtilis. Redox cofactor balance enabled the strain to synthesize L-valine under oxygen-limiting condition, significantly increasing the yield in the presence of glucose. Two-stage fed-batch fermentation of the final strain in a 5 L bioreactor produced 84 g/L L-valine with a yield and productivity of 0.41 g/g glucose and 2.33 g/L/h, respectively. To the best of our knowledge, this is the highest L-valine titer and yield ever reported in E. coli. The systems metabolic engineering strategy described here will be useful for future engineering of E. coli strains for the industrial production of L-valine and related products.

Abstract:Background L-(+)-tartaric acid (L-TA) is an important organic acid, which is produced from the cream of tartar or stereospecific hydrolysis of the cis -epoxysuccinate. The former method is limited by the availability of raw material and the latter is dependent on the petrochemical material. Thus, new processes for the economical preparation of L-TA from carbohydrate or renewable resource would be much more attractive. Production of 5-keto-D-gluconate (5-KGA) from glucose by Gluconobacter oxydans is the first step to produce L-TA. The aim of this work is to enhance 5-KGA accumulation using combinatorial metabolic engineering strategies in G. oxydans . The sldAB gene, encoding sorbitol dehydrogenase, was overexpressed in an industrial strain G. oxydans ZJU2 under a carefully selected promoter, P 0169 . To enhance the efficiency of the oxidation by sldAB , the coenzyme pyrroloquinoline quinone (PQQ) and respiratory chain were engineered. Besides, the role in sldAB overexpression, coenzyme and respiratory chain engineering and their subsequent effects on 5-KGA production were investigated. Results An efficient, stable recombinant strain was constructed, whereas the 5-KGA production could be enhanced. By self-overexpressing the sldAB gene in G. oxydans ZJU2 under the constitutive promoter P 0169 , the resulting strain, G. oxydans ZJU3, produced 122.48 ± 0.41 g/L of 5-KGA. Furthermore, through the coenzyme and respiratory chain engineering, the titer and productivity of 5-KGA reached 144.52 ± 2.94 g/L and 2.26 g/(L · h), respectively, in a 15 L fermenter. It could be further improved the 5-KGA titer by 12.10 % through the fed-batch fermentation under the pH shift and dissolved oxygen tension (DOT) control condition, obtained 162 ± 2.12 g/L with the productivity of 2.53 g/(L · h) within 64 h. Conclusions The 5-KGA production could be significantly enhanced with the combinatorial metabolic engineering strategy in Gluconobacter strain, including sldAB overexpression, coenzyme and respiratory chain engineering. Fed-batch fermentation could further enlarge the positive effect and increase the 5-KGA production. All of these demonstrated that the robust recombinant strain can efficiently produce 5-KGA in larger scale to fulfill the industrial production of L-TA from 5-KGA.
Escherichia coli W as a new platform strain for the enhanced production of L-valine by systems metabolic engineering

Abstract:A less frequently employed Escherichia coli strain W, yet possessing useful metabolic characteristics such as less acetic acid production and high L-valine tolerance, was metabolically engineered for the production of L-valine. The ilvA gene was deleted to make more pyruvate, a key precursor for L-valine, available for enhanced L-valine biosynthesis. The lacI gene was deleted to allow constitutive expression of genes under the tac or trc promoter. The ilvBN(mut) genes encoding feedback-resistant acetohydroxy acid synthase (AHAS) I and the L-valine biosynthetic ilvCED genes encoding acetohydroxy acid isomeroreductase, dihydroxy acid dehydratase, and branched chain amino acid aminotransferase, respectively, were amplified by plasmid-based overexpression. The global regulator Lrp and L-valine exporter YgaZH were also amplified by plasmid-based overexpression. The engineered E. coli W (ΔlacI ΔilvA) strain overexpressing the ilvBN(mut) , ilvCED, ygaZH, and lrp genes was able to produce an impressively high concentration of 60.7 g/L L-valine by fed-batch culture in 29.5 h, resulting in a high volumetric productivity of 2.06 g/L/h. The most notable finding is that there was no other byproduct produced during L-valine production. The results obtained in this study suggest that E. coli W can be a good alternative to Corynebacterium glutamicum and E. coli K-12, which have so far been the most efficient L-valine producer. Furthermore, it is expected that various bioproducts including other amino acids might be more efficiently produced by this revisited platform strain of E. coli.

Abstract:L-Valine, a branched-chain amino acid with diversified applications, is biosynthesized with α-acetolactate as the key precursor. In this study, the metabolic flux in Klebsiella oxytoca PDL-K5, a Risk Group 1 organism producing 2,3-butanediol as the major fermentation product, was rearranged to L-valine production by introducing exogenous L-valine biosynthesis pathway and blocking endogenous 2,3-butanediol generation at the metabolic branch point α-acetolactate. After further enhancing L-valine efflux, strengthening pyruvate polymerization and selecting of key enzymes for L-valine synthesis, a plasmid-free K. oxytoca strain VKO-9 was obtained. Fed-batch fermentation with K. oxytoca VKO-9 in a 7.5 L fermenter generated 122 g/L L-valine with a yield of 0.587 g/g in 56 h. In addition, repeated fed-batch fermentation was conducted to prevent precipitation of L-valine due to oversaturation. The average concentration, yield, and productivity of produced L-valine in three cycles of repeated fed-batch fermentation were 81.3 g/L, 0.599 g/g, and 3.39 g/L/h, respectively.

Abstract:Corynebacterium glutamicum is a versatile industrial microorganism for producing various amino acids. However, there have been no reports of a well-defined C. glutamicum strain capable of hyperproducing L-tryptophan. This study presents a comprehensive metabolic engineering approach to establish a robust C. glutamicum cell factory for L-tryptophan biosynthesis, including: (1) identification of potential targets using an enzyme-constrained genome-scale model; (2) targeted enhancement of the L-tryptophan biosynthetic pathway; (3) reconfiguration of central metabolic pathways; (4) identification of metabolic bottlenecks through comparative metabolome analysis; (5) engineering of the transport system, shikimate pathway, and precursor supply; and (6) repression of competing pathways and iterative optimization of key targets. The resulting engineered C. glutamicum strain achieved a remarkable L-tryptophan titer of 16.2 g/L with a yield of 0.16 g/g glucose in shake-flask fermentation. This study highlights the efficacy of integrating computational modeling with systems metabolic engineering for significantly enhancing the production capabilities of industrial microorganisms.

Abstract:minal Gln amide transfer (GAT) domain and is promoted by the allosteric effector GTP. 3] The resulting nascent NH3 is subsequently transferred to the N-terminal synthase domain via an NH3 tunnel, [4–6] where it reacts with UTP that has been activated by ATP-dependent phosphorylation at the 4-position (i.e. , UTP-4-P). The enzyme exhibits positive cooperativity for ATP and UTP, and these nucleotides act synergistically to promote tetramerization of the enzyme to its active form. The product CTP is a feedback inhibitor of the enzyme and plays an important role in the biosynthesis of nucleic acids, phospholipids, 12] and sialic acid. Consequently, CTPS is a recognized target for the development of antineoplastic, antiviral, and antiprotozoal agents. Although many studies have focused on delineating the regulatory properties of CTPS, very few have focused on the development of nucleotide analogues as inhibitors of this enzyme. 24] 3-Deazauridine 5’-triphosphate (IC50 ~18 mm) and cyclopentenyl cytosine (CPEC) 5’-triphosphate (IC50 ~6 mm) are substrate and product analogues, respectively, that are the most well-studied inhibitors of CTPS. In addition, the 5’-triphosphate of 2’,2’-difluorodeoxycytidine (gemcitabine) is also believed to exert its chemotherapeutic effects, in part, through the inhibition of CTPS. 28] However, mutations in CTPS lead to the loss of feedback inhibition by CTP and resistance to the cytotoxic effects of these and other chemotherapy drugs. Therefore, the development of new, potent, and selective CTPS inhibitors such as transition state/intermediate analogues or multivalent inhibitors is required. Enzymes often exhibit high affinity for structural and electronic mimics of transition states or reactive intermediates generated during catalysis. We rationalized that CTPS inhibitors might be developed by employing methylene or difluoromethylene phosphonate groups, or a phosphoramidate group as isosteric and/or isoelectronic replacements for the 4-phosphate group of the UTP-4-P intermediate (Scheme 1). Alternatively, the use of multivalent ligands (e.g. , substrate– product analogues) is often an effective strategy to develop potent and specific enzyme inhibitors, as such inhibitors may interact with multiple binding determinants on the enzyme to afford an entropic advantage in binding. Abstract:Corynebacterium glutamicum , a well-studied industrial model microorganism, has garnered widespread attention due to its ability for producing amino acids with a long history. In recent years, research efforts have been increasingly focused on exploring its potential for producing various organic acids beyond amino acids. Organic acids, which are characterized by their acidic functional groups, have diverse applications across industries such as food, agriculture, pharmaceuticals, and biobased materials. Leveraging advancements in metabolic engineering and synthetic biology, the metabolic pathways of C. glutamicum have been broadened to facilitate the production of numerous high-value organic acids. This review summarizes the recent progress in metabolic engineering for the production of both amino acids and other organic acids by C. glutamicum . Notably, these acids include, amino acids (lysine, isoleucine, and phenylalanine), TCA cycle-derived organic acids (succinic acid, α-ketoglutaric acid), aromatic organic acids (protocatechuate, 4-amino-3-hydroxybenzoic acid, anthranilate, and para-coumaric acid), and other organic acids (itaconic acid and cis, cis-muconic acid).

Abstract:The experiments presented here were based on the conclusions of our previous results. In order to avoid introduction of expression plasmid and to balance the NADH/NAD ratio, the NADH biosynthetic enzyme, i.e., NAD-dependent glyceraldehyde-3-phosphate dehydrogenase (GADPH), was replaced by NADP-dependent GADPH, which was used to biosynthesize NADPH rather than NADH. The results indicated that the NADH/NAD ratio significantly decreased, and glucose consumption and l-lysine production drastically improved. Moreover, increasing the flux through l-lysine biosynthetic pathway and disruption of ilvN and hom, which involve in the branched amino acid and l-methionine biosynthesis, further improved l-lysine production by Corynebacterium glutamicum. Compared to the original strain C. glutamicum Lys5, the l-lysine production and glucose conversion efficiency (α) were enhanced to 81.0 ± 6.59 mM and 36.45 % by the resulting strain C. glutamicum Lys5-8 in shake flask. In addition, the by-products (i.e., l-threonine, l-methionine and l-valine) were significantly decreased as results of genetic modification in homoserine dehydrogenase (HSD) and acetohydroxyacid synthase (AHAS). In fed-batch fermentation, C. glutamicum Lys5-8 began to produce l-lysine at post-exponential growth phase and continuously increased over 36 h to a final titer of 896 ± 33.41 mM. The l-lysine productivity was 2.73 g l−1 h−1 and the α was 47.06 % after 48 h. However, the attenuation of MurE was not beneficial to increase the l-lysine production because of decreasing the cell growth. Based on the above-mentioned results, we get the following conclusions: cofactor NADPH, precursor, the flux through l-lysine biosynthetic pathway and DCW are beneficial to improve l-lysine production in C. glutamicum.

Abstract:Corynebacterium glutamicum has a long and successful history in the biotechnological production of l-lysine. Besides the adjustment of metabolic pathways, intracellular and extracellular transport systems are critical for the cellular metabolism of l-lysine or its by-products. Here, three amino acid transmembrane transporters, namely, GluE, BrnE/BrnF, and LysP, which are widely present in C. glutamicum strains, were each investigated by gene knockout. In comparison with that in the wild-type strain, the yield of l-lysine increased by 9.0%, 12.3%, and 10.0% after the deletion of the gluE, brnE/brnF, and lysP genes, respectively, in C. glutamicum 23,604. Moreover, the amount of by-product amino acids decreased significantly when the gluE and brnE/brnF genes were deleted. It was also demonstrated that there was no effect on the growth of the strain when the gluE or lysP gene was deleted, whereas the biomass of C. glutamicum WL1702 (ΔbrnE/ΔbrnF) in the fermentation medium was significantly reduced in comparison with that of the wild type. These results also provide useful information for enhancing the production of l-lysine or other amino acids by C. glutamicum.

ADULT STEM CELLS FOR TREATMENT OF OA IN THE DOG : DATA TO AND FROM THE CLINIC

Abstract:Adult stem cells in veterinary medicine can be derived from adipose tissue or from bone marrow. In small animal veterinary medicine stem cells are more commonly collected from adipose tissue but my opinion is that bone marrow-derived cells remain a perfectly reasonable option. There are key species differences and this is one difference between the canine and equine world. In horses, adult stem cells are usually collected from bone marrow because it can be performed in the standing horse and the BM provides a better source of cells than fat. In dogs, adipose-derived stem cells (ASC) are usually collected from the falciform ligament but can also be collected from any fat deposit such as just caudal to the shoulder or from the inguinal region. Although the data is not definitive; recent investigations have suggested that cell harvest from the falciform ligament generally yields the largest, most predictable amount of cells. Adiposederived cells appear to have a similar potential for differentiation into bone, cartilage, adipose, and fibrous tissues as bone marrow derived cells. This multipotency is a key component to calling a cell a “stem cell” and is something we, and others, have been able to demonstrate from fat and BM derived cells in the dog and horse. Adipose-derived stem cells hold great potential for the treatment of many conditions in the dog because of their trophic and anti-inflammatory properties and because they can differentiate into several other cell lines. The most notable tissues that adult stem cells can form is bone and cartilage. Because of this the main focus in the veterinary literature has been the treatment of osteoarthritis (OA). In two, prospective, randomized, placebo-controlled studies, dogs with lameness from osteoarthritis improved as compared to dogs treated with placebo. However, the strength of this evidence should be considered as the number of dogs evaluated was small and only subjective outcome measures (owner and veterinarian opinion) were used. Data in other species would suggest that ASCs could be used for bone and ligament injuries, spinal cord injury, cardiomyopathies, and inflammatory conditions of the gastrointestinal tract and skin. Certainly, many empirical stories are available for these types of conditions. In addition, the potential to gain valuable information from naturally occurring animal diseases in order to translate it to the human field is immeasurable. Currently, for veterinary use, ASCs are isolated by collagenase digestion of adipose tissue that yields an initial stromal vascular fraction (SVF) that can be further processed by cell culture. Historically, stem cell therapy in dogs has been limited to commercial operations (e.g. Vet Stem) that will process fat obtained from a patient, and return a stromal vascular fraction (SVF) preparation of stem cells to the veterinarian for injection. Stem cells are not specifically isolated or cultured. This process is time intensive, requiring a veterinarian specifically trained (this is an online training process via Vet Stem) in proper handling of the care and acquirement of tissue. However, systems are now available that allow this processing to be performed directly in the veterinary clinic with little training. While this adds convenience and reduces time and cost it introduces error and variability in the product (SVF). Thus the patient, with systemic illness, must undergo a general anesthetic episode for surgical collection of fat. Significant cost can be incurred by the owner for the surgical procedure and the process of preparing the SVF from the fat. Abstract:OBJECTIVE To explore the preventive effect of lanthanum chloride on enteral bacterial translocation in scalded rats. METHODS Ninety Sprague-Dawley (SD) rats were employed in the study and randomly divided into three groups, i.e. normal control (A), burn control (B) and treatment (C) groups. Plasmid PUC19 labelled by JM109 was transfected to Escherichia coli (E. coli), so that restriction endonuclease finger - print image spectrum analysis could be applied to the tracing and quantification of the translocation of E. coli from intestine to mesenteric lymph nodes (MLNs) and blood. The intestinal tissue contents of endotoxin (ET), nitric oxide (NO), nitric oxide synthase (NOS), malondialdehyde (MDA) and superoxide dismutase (SOD) were determined. RESULTS It was identified that the bacteria in MLNs and blood exhibited the same gene map with those from gastric gavage in B and C groups. But the bacterial quantity in MLNs in C group on 3 postburn day (PBD) was much lower than that in B group (P < 0.05). The intestinal MDA content in C group on 1 and 3 PBDs was obviously higher than that in B group (P < 0.05). CONCLUSION Bacteria (E. coli) could be translocated from gut to MLNs and blood, which could be evidently alleviated by lanthanum chloride by means of its bactericidal property, inhibition of NOS activity, so that NO production decreased, and its ability to increase SOD activity leading to less production of MDA.

Abstract:Ectoine is a compatible solute that functions as a cell protector from various stresses, protecting cells and stabilizing biomolecules, and is widely used in medicine, cosmetics, and biotechnology. Microbial fermentation has been widely used for the large-scale production of ectoine, and a number of fermentation strategies have been developed to increase the ectoine yield, reduce production costs, and simplify the production process. Here, Corynebacterium glutamicum was engineered for ectoine production by heterologous expression of the ectoine biosynthesis operon ectBAC gene from Halomonas elongata, and a series of genetic modifications were implemented. This included introducing the de3 gene from Escherichia coli BL21 (DE3) to express the T7 promoter, eliminating the lysine transporter protein lysE to limit lysine production, and performing a targeted mutation lysCS301Y on aspartate kinase to alleviate feedback inhibition of lysine. The new engineered strain Ect10 obtained an ectoine titer of 115.87 g/L in an optimized fed-batch fermentation, representing the highest ectoine production level in C. glutamicum and achieving the efficient production of ectoine in a low-salt environment.
Williams ’ Test Comparing Multiple Dose Groups to a Control

Abstract:In dose-ranging pharmaceutical trials, several groups of subjects who receive active doses are compared to a group receiving a placebo or conventional therapy. When the effect of the dose on the mean level of the response is expected to be monotonic, Williams (1971, 1972) proposed a test to compare multiple groups to a control. He also computed tables of the critical values for the simple design that corresponds to a one-way analysis of variance (ANOVA). Guo and Brown (1996) compared the power of Williams’ test with those of the step-down procedure and Dunnett’s multiple comparison method. Williams’ test had acceptable power over all orderings of the means, but was substantially more powerful than its two competitors for some orderings of the observed effects. In this paper we consider the implementation of Williams’ test in a more general setting, that of the general linear model. Even when the primary comparison is between groups, it is necessary to adjust for potential confounding effects, such as centre, age or gender. In section 2 we show that the 5% critical values of Williams’ test correspond to the 4 th percentile of Student’s t-distribution. More extreme critical values correspond to a percentile of Student’s t that is less than 0.8 of the desired size. Based on this we propose that the usual p-values obtained by contrasts that are used in Williams’ test be multiplied by 1.25 to approximate the p-values for Williams’ test. Using simulation, we evaluate the empirical sizes of the proposed approximation to show that the approximation is conservative.
Systems engineering of Escherichia coli for high-level glutarate production from glucose

Abstract:Abstract Glutarate is a key monomer in polyester and polyamide production. The low efficiency of the current biosynthetic pathways hampers its production by microbial cell factories. Herein, through metabolic simulation, a lysine-overproducing E. coli strain Lys5 is engineered, achieving titer, yield, and productivity of 195.9 g/L, 0.67 g/g glucose, and 5.4 g/L·h, respectively. Subsequently, the pathway involving aromatic aldehyde synthase, monoamine oxidase, and aldehyde dehydrogenase (AMA pathway) is introduced into E. coli Lys5 to produce glutarate from glucose. To enhance the pathway’s efficiency, rational mutagenesis on the aldehyde dehydrogenase is performed, resulting in the development of variant Mu5 with a 50-fold increase in catalytic efficiency. Finally, a glutarate tolerance gene cbpA is identified and genomically overexpressed to enhance glutarate productivity. With enzyme expression optimization, the glutarate titer, yield, and productivity of E. coli AMA06 reach 88.4 g/L, 0.42 g/g glucose, and 1.8 g/L·h, respectively. These findings hold implications for improving glutarate biosynthesis efficiency in microbial cell factories.

Abstract:γ-Aminobutyric acid (GABA) is a non-proteinogenic amino acid with important physiological functions, which has been widely used in food, pharmaceuticals, and polyamides production. The fermentative GABA production by Corynebacterium glutamicum was recognized as one of the most promising methods. However, the problems of low catalytic activity of the heterologously expressed glutamate decarboxylase (GAD) and the imbalanced carbon flux between cell growth and GABA synthesis severely limited the GABA production by C. glutamicum . This study applied combinational metabolic engineering and catalytic condition optimization strategies to solve these two major obstacles. The secretory expression of GAD was enhanced using a bicistronic-designed expression cassette. This bicistronic expression cassette was further triply inserted into the genome by substituting the ldhA , pqo , and ack genes, thus stabilizing the expression of GAD and reducing the accumulation of by-products of lactate and acetate. A growth-regulated promoter P CP_2836 was applied to dynamically control the expression of odhA , thus controlling the α-oxoglutarate dehydrogenase complex activity for balanced cell growth and GABA production. The glutamate precursor synthesis and pyridoxal 5′-phosphate supply were also strengthened by promoter substitution. Finally, through a two-stage pH-controlled fed-batch fermentation under optimized conditions, the engineered strain reached GABA titer of 81.31 ± 1.31 g/L with a yield and productivity of 0.50 ± 0.01 g/g and 1.36 ± 0.23 g L −1 h −1 , which was 4.8%, 13.6%, and 11.2% higher than that of the original strain. This study laid a solid foundation for industrial fermentative GABA production by engineered C. glutamicum .

Abstract:There is increasing industrial demand for five-carbon platform chemicals, particularly 5-hydroxyvaleric acid (5-HV), a widely used building block for the synthesis of polyesters and polyurethanes. Here, we developed an efficient 5-HV microbial producer by systems metabolic engineering of Escherichia coli W3110. To derive 5-HV, Pseudomonas putida l -lysine monooxygenase ( davB ) and 5-aminovaleramide amidohydrolase ( davA ), E. coli 4-aminobutyrate aminotransferase ( gabT ), and different aldehyde reductase genes were introduced to E. coli W3110 to convert l -lysine hydrochloride to the target product. Three aldehyde reductases, including YajO from P. putida and YahK and YqhD from E. coli, were examined, revealing that the overexpression of aldehyde reductases was critical for 5-HV production and YajO was first reported for efficient production of 5-HV. The E. coli 5-aminovalerate exporter encoded by gabP and E. coli lysine-specific permease encoded by lysP were expressed to further enhance 5-HV production. In addition, a hok/sok toxin/antitoxin system based on the E. coli XL1-Blue genome was inserted to the dual-vector system to decrease plasmid loss. Finally, 5.59 g/L of 5-HV with a yield of 0.96 mol/mol (5-HV/ l -lysine hydrochloride) by shake flask cultivation of the engineered E. coli W3110 and 35.6 g/L of 5-HV with a yield of 0.92 mol/mol (5-HV/ l -lysine hydrochloride) by fed fermentation were achieved respectively. The developed metabolic engineering strategies provide a basis for the efficient and sustainable bio-based production of 5-HV.

Abstract:The acetoin was synthesized using metabolic engineering method fromCorynebacteriumglutamicum ATCC 13032. ThealsSD operon derived fromBacillus subtilis 168 was expressed inC. glutamicum ATCC 13032 to yield acetoin of 2.14,g/L in the engineering strain. Additional inactivation ofaceE gene encoding the E1p enzyme of the pyruvate dehydrogenase complex andldhA gene encoding NAD+-dependent L-lactate dehydrogenase increased the supply of acetoin precursor. The acetoin production increased to 5.09,g/L with the newly constructed strain. Subse-quently,formation of 2,3-butanediol was inhibited by deletingbutA gene encoding for 2,3-butanediol dehydro-genase. Total yield of acetoin reached 8.33,g/L(51.1% of theoretical yield)under the optimum fermentation condition in the strain CGL3. The experimental results show that the strain ofC. glutamicum ATCC 13032,engineered metaboli-cally can provide a potential application for acetoin synthesis.
