Abstract:Soil microbiome is the cornerstone of sustaining soil ecosystem services and performs an array of vital soil functions. Manipulating soil microbiome is a promising strategy to regulate soil ecosystem function, however, the effect and underlying mechanism for chlorinated organic pollution remediation are limited and the capacity to simultaneously control multiple redox processes is still unclear. Here, the dynamics of the biocathode microbial community after separately or synchronously adding conductive material and acyl-homoserine lactone (AHL) were investigated from the perspectives of electron transfer and microbial assembly in γ-hexachlorocyclohexane (γ-HCH) polluted soil-based bioelectrochemical system. The resulting responses of NO3−/Fe(III)/SO42− reduction, γ-HCH dechlorination, and CH4 cumulation were also analyzed. Two conductive materials of biochar (BC) and magnetite nanoparticle (NaFe) and two AHLs including N-butanoyl homoserine lactone (C4-HSL) and 3-oxo-hexanoyl-homoserine lactone (3OC6-HSL) were applied. Exogenous additives facilitated γ-HCH dechlorination but alleviated and even inhibited CH4 cumulation, with BC plus C4-HSL addition as the optimum. Adding BC/NaFe enriched Fe(III)-reducing bacteria and dechlorinators and selectively transferred more electrons to those electron sinks; C4-HSL/3OC6-HSL helped to increase biofilm aggregation; while BC plus C4-HSL stimulated the growth of dominant functional taxa and enhanced the compact microbial correlations. These findings broaden our insights and provide approaches to managing soil microbiomes to solve actual environmental emergencies. It shows an advanced strategy to inhibit greenhouse gas (CH4) emission when COP reduction is facilitated to achieve win-win soil remediation.
Redox Structures of Humic Acids Derived From Different Sediments and Their Effects on Microbial Reduction Reactions

Abstract:Herein, we investigated the chemical, electrochemical, and spectroscopic characteristics of humic acids (HAs) extracted from sediments of different origin [Ling Qiao river, Xi Xi wetland, Qi Zhen lake (QZ), and Hu Zhou pond in Zhejiang province, China], paying particular attention to their role in the enhancement of nitrate and FeOOH reduction. Notably, the highest C/N ratio (16.16), O/C ratio (1.89), and Fe content (11.57 g kg-1 sample) were observed for HAs extracted from QZ sediment. Cyclic voltammetry analyses confirmed that all HAs contained redox-active groups and exhibited redox potentials between -0.36 and -0.28 V vs. the standard hydrogen electrode. All HAs showed similar Fourier transform infrared spectra with variable absorption intensity, the spectra verified the presence of aromatic C=C, C-H, and C=O of quinone ketones group in HAs. Electron spin resonance suggested that quinone moieties within HAs are the redox-active centers. All HAs promoted the microbial reduction of nitrate and amorphous FeOOH by Shewanella oneidensis strain MR-1, achieving high nitrate reduction extents of 79-98.4%, compared to the biotic and abiotic control values of 29.6 and 0.006%, respectively. The corresponding extents of Fe(II) production equaled 43.25-60.5%, exceeding those of biotic and abiotic controls (28.5 and 0.005%, respectively). In addition to the highest C/N, O/C ratio, and Fe content, HA extracted from QZ sediment also exhibited the highest nitrate and FeOOH reduction performances. Although the proportion of organic redox-active carbon is small, the potential electron-mediating ability is not ignorable. HAs are redox active for enhancing microbial reduction of nitrate and amorphous FeOOH regardless of the location or texture of parent sediments, implying their great potential for acting as redox mediator in enhancing multiple microbial reduction, thereby affecting various biogeochemical processes (i.e., iron cycle, nitrogen cycle, etc.) as well as in situ remediation in anaerobic environment.
Simultaneous Regulation of Biocathodic Γ-Hch Dechlorination and CH4 Production by Tailoring the Structure and Function of Biofilms Based on Quorum Sensing.

Abstract:Dechlorination of chlorinated organic pollutants and methanogenesis are attractive biocathode reductions in microbial electrolysis cells (MECs). Quorum sensing (QS) is applied to regulate microbial communications. However, how acyl-homoserine lactones (AHLs)-dependent QS organize the assembly of the biocathode microbial community, and then regulate multiple biocathode reductions remains unclear. By applying N-butanoyl homoserine lactone (C4-HSL), N-hexanoyl homoserine lactone (C6-HSL) and 3-oxo-hexanoyl homoserine lactone (3OC6-HSL) in γ-hexachlorocyclohexane (γ-HCH) contaminated MECs, this study investigated the changes of biofilm microbial structure and function and the mechanisms of AHLs-QS on γ-HCH dechlorination and CH4 production. Exogenous C4-HSL and 3OC6-HSL increased cytochrome c production and enriched dechlorinators, electroactive bacteria but not methanogens to accelerate γ-HCH dechlorination and inhibit CH4 production. C6-HSL facilitated dechlorination and CH4 production by enhancing biofilm electroactivity and increasing membrane transportation. Besides, exogenous C6-HSL restored the electron transfer capacity that was damaged by the concurrent addition of acylase, an endogenous AHL quencher. From the perspective of microbial assembly, this study sheds insights into and provides an efficient strategy to selectively accelerate dechlorination and CH4 production by harnessing microbial structure based on QS systems to meet various environmental demands.
Optimizing Humic Acid Breakdown and Methane Production Via Semiconductor-Assisted Photo-Anaerobic Digestion

Abstract:Humic acids (HAs), whether naturally present in anaerobic digestion (AD) substrates or HAs newly formed during fermentation, can become inhibitory to methanogenesis as their concentrations reach certain levels. This study delved extensively into the mechanism underlying the alleviation of HAs inhibition in photo-AD by Ndoped carbon quantum dots (NCQDs). These NCQDs were efficiently synthesized from straw using an environmentally friendly pretreatment method. Our proposed method harnessed the combined effect of light exposure and NCQDs, resulting in synergistic enhancement of the cumulative CH4 yield within the HA-inhibited AD system, achieving a remarkable yield of 293.7 +/- 17.7 mL/g VS. In-depth analyses were conducted on the remaining dissolved organic matter (DOM) within digesters using 3D-EEM and ESI FT-ICR MS. Simultaneously, the remaining HAs were extracted and subjected to FT-IR analysis. The findings revealed that NCQDs effectively degraded humic acid-like components in DOM into smaller, more manageable micromolecules characterized by lower carbon numbers and reduced double bond equivalent. Additionally, under the influence of light, NCQDs promoted the degradation of aromatic components within HAs. These resulting micromolecules were made readily available for utilization by microorganisms, further contributing to methanogenesis. Furthermore, photoelectrochemical analysis and specific gene qPCR analysis revealed that the photoelectrons from NCQDs and HAs were received and transferred by Ech (electron acceptors) for methanogenesis. Remarkably, this methanogenesis pathway, akin to photosynthesis, played a pivotal and transformative role in the photo-AD system. This work comprehensively revealed the remarkable potential mechanism of semiconductors within the photoAD system, offering profound insights that can catalyze the development of innovative AD reactors and semiconductor accelerators.

Abstract:Humic acids (HAs) are redox-active components that play a crucial role in catalyzing relevant redox reactions in various ecosystems. However, it is unclear what role the different compost-derived Has play in the dissimilatory Fe(III) bioreduction and which chemical structures could accelerate Fe reduction. In this study, we compared the effect of eighteen HAs from the mesophilic phase, thermophilic phase and mature phase of protein-, lignocellulose- and lignin-rich composting on catalyzing the bioreduction of Fe(III)-citrate by Shewanella oneidensis MR-1 in temporarily anoxic laboratory systems. The chemical composition and structure of different compost-derived HAs were analyzed by UV-Vis spectroscopy, excitation-emission matrices of the fluorescence spectra, and C-13-NMR. The results showed that HAs from lignocellulose- and lignin-rich composting, especially in the thermophilic phase, promoted the bioreduction of Fe(III). They also showed that HA from protein-rich materials suppressed significantly the Fe(II) production, which was mainly affected by the amount and structures of functional groups (e.g., quinone groups) and humification degree of the HAs. This study can aid in searching sustainable HA-rich composts for wide-ranging applications to catalyze redox-mediated reactions of pollutants in soils.

Abstract:Humic acids are one of the main organic matters in sediments and contribute importantly to the marine biogeochemical cycles. Extracellular electron transfer is a ubiquitous natural process and has potentials to change the macrostructure of humic acids which can act as an electron shuttle. By setting up marine sediment microbial fuel cells, the present study revealed that enhanced extracellular electron transfer process could increase the content of C and H, but decrease the O content in humic acids, which could result in an increased aromaticity and decreased polarity of humic acids, whereas no significant changes occurred to the humification degree of the humic acids. Specific bacterial groups as potential exoelectrogens including Proteobacteria (especially Pseudomonas strains) and Firmicutes were enriched under enhanced extracellular electron transfer process, indicating that they were active to exchange electrons and might play important roles during the changes of humic acids, while the relative abundance of Verrucomicrobia and Bacteroidetes was reduced during these processes. The results of the present research shed lights on the relation between exoelectrogens and the transformation of humic acids in coastal sediment, while the microbial process and mechanisms behind it require further study.

Abstract:Solid-phase humic acid (HAsolid) plays a critical role in global carbon cycle and redox biogeochemistry of topsoil, because of its unique physicochemical properties, including electrochemical property. In this study, topsoil HAsolid along a typical forest-river-paddy landscape section in eastern China was investigated in the aspects of electrochemical property, chemical structure and nanomechanics, and their relationship. Nano-size HAsolid particles were extracted from topsoil of paddy soil (PS-HAsolid), forest soil (FS-HAsolid) and riverside sediment (RS-HAsolid). Results showed that all the HAsolid were conductive and played an important role in conductivity of topsoil suspension. HAsolid contained both reversible and irreversible redox peaks, with redox activity of PS-HAsolid > RS-HAsolid > FS-HAsolid. Owing to limited humification, electron exchange capacity (EEC) values of topsoil HAsolid suspensions were modest (3.18-4.45 μmol e-g-1 HAsolid). Compared with RS-HAsolid and FS-HAsolid, PS-HAsolid showed higher aromaticity and higher degree of humification with simple and even nanomechanical property. Long-term cultivation (human activities) as well as high content of polyvalent metals in paddy soil were supposed to facilitate formation of aromatic carbon and improve humification of HAsolid. The results suggested that aromatic carbon and high humification degree of PS-HAsolid contributed to simple and even nanomechanics, which further optimized its electrochemical property. This study not only provides novel insight into the mechanism of HAsolid mediating electron transfer, but also inspires ideas for soil and environmental management with different purposes based on regulation of HAsolid.
The promotion of the polycyclic aromatic hydrocarbons degradation mechanism by humic acid as electron mediator in a sediment microbial electrochemical system

Abstract:To enhance the removal efficiencies of polycyclic aromatic hydrocarbons (PAHs) in sediments and to elucidate the mechanisms by which microbial electrochemical action aids in the degradation of PAHs, humic acid was used as an electron mediator in the microbial electrochemical system in this study. The results revealed that the addition of humic acids led to increases in the removal efficiencies of naphthalene, phenanthrene, and pyrene by 45.91%, 97.83%, and 85.56%, respectively, in areas remote from the anode, when compared to the control group. The investigation into the microbial community structure and functional attributes showed that the presence of humic acid did not significantly modify the microbial community composition or its functional expression at the anode. However, an examination of humic acid transformations demonstrated that humic acid extended the electron transfer range in sediment via the redox reactions of quinone and semiquinone groups, thereby facilitating the PAHs degradation within the sediment.
Electrochemical properties of humic acid and its novel applications: A tip of the iceberg

Abstract:The widely existed humic acid (HA) with abundant redox-active groups has been considered to play an important role in biogeochemistry in sediments and soils. Recent studies reported that HA showed great performance in terms of electron transfer capacity (up to HAEDC = 94 mmol e-/mol C, HAEAC = 42 mmol e-/mol C). Since HA is widely available, inexpensive and environmentally friendly, the electrochemistry of HA has been explored to apply in many fields, such as environmental remediation, detection sensor and energy storage. Whereas, these prospective applications of HA and their electrochemical principles were lack of a comprehensive summary. In this review, the electrochemical properties and the prospective electrochemical applications of HA were summarized. Simultaneously, the existing problems like shortages of traditional electrochemical characterization of HA, and future research directions about HA electrochemistry were prospected. This review provides a deeper understanding of HA electrochemistry, and also inspires ideas for environmental remediation, detection sensor and energy storage by exploring the potential application values of HA.
Modified humic acids mediate efficient mineralization in a photo-bio-electro-Fenton process

Abstract:Humic acids (HA) are common mediators in redox reactions in the aquatic environment. The structures and properties of HA are greatly influenced by environmental factors such as external electrons. In this study, qualitative changes in electron-modified HA and the underlying mechanisms were reported, which not only contribute to understanding the fate of HA and their impact on organic pollutants, but could facilitate their potential use for water purification. The photochemical activity and electron-donating capacity of HA were improved due to the increase of phenolic and carboxyl components via the reduction modification by electrons, creating a novel and efficient photo-bio-electro-Fenton system mediated by HA under neutral conditions without the use of hydrogen peroxide (H2O2). The in-situ continuous production of H2O2 ensured an adequate supply of hydroxyl radicals in this coupled system, achieving mineralization (92%) of HA and 17α-ethinylestradiol (EE2), a common synthetic estrogen with high estrogenic potency. Two degradation pathways with five degradation intermediates of EE2 were identified in our study. Effluents from the coupled system showed decreased endocrine-disrupting activity. Our findings demonstrated a new approach for the in-situ modification and potential use of HA for water treatment and particularly the concurrent degradation of HA and organic pollutants through a photo-bioelectrochemical system mediated by HA.

Effect of Long-Term Fertilization on Humic Redox Mediators in Multiple Microbial Redox Reactions

Abstract:This study investigated the effects of different long-term fertilizations on humic substances (HSs), humic acids (HAs) and humins, functioning as redox mediators for various microbial redox biotransformations, including 2,2',4,4',5,5'- hexachlorobiphenyl (PCB153) dechlorination, dissimilatory iron reduction, and nitrate reduction, and their electron-mediating natures. The redox activity of HSs for various microbial redox metabolisms was substantially enhanced by long-term application of organic fertilizer (pig manure). As a redox mediator, only humin extracted from soils with organic fertilizer amendment (OF-HM) maintained microbial PCB153 dechlorination activity (1.03 mu M PCB153 removal), and corresponding HA (OF-HA) most effectively enhanced iron reduction and nitrate reduction by Shewanella putrefaciens. Electrochemical analysis confirmed the enhancement of their electron transfer capacity and redox properties. Fourier transform infrared analysis showed that C=C and C=O bonds, and carboxylic or phenolic groups in HSs might be the redox functional groups affected by fertilization. This research enhances our understanding of the influence of anthropogenic fertility on the biogeochemical cycling of elements and in situ remediation ability in agroecosystems through microorganisms' metabolisms. Abstract:The hyperaccumulating mechanism concerning heavy metal activation or passivation and plant response triggered by fulvic acid (FA) and humic acid (HA) recruitments are investigated herein. We carefully examine the Cd activation effect by various FA and HA, tracing from pig, goat, and duck manure composts to straw compost and commercial materials (i.e., PC, GC, DC, SC, and CM), as well as their roles in plant growth promotion and Cd uptake. Our results indicate that due to the decrease of soil pH and their multiple functional groups, the contents of available Cd (AE-Cd) increased by 4.3-4.8% and 3.6-6.3% when all FA and HA sources were applied for 30 days. A 13.1-19.9% increase in AE-Cd was observed when CFA, DFA, and PFA were applied for five days, and a 9.5% increment was found when PHA was applied for 10 days. In the pot experiment, the Cd accumulation in plants increased by 2.78 and 2.17 folds with PFA and PHA applications, respectively, compared to the blank control group. This result can be attributed to the stimulative effects of the simultaneous Sedum alfredii growth and Cd phytoavailability. Notably, the Cd accumulation increased by 2.26 times with the SFA amendment due to the predominant stimulation effect to the phytoavailable Cd rather than plant growth. However, slight inhibitory effects were observed upon plant growth or Cd uptake, which led to the reduction of the Cd accumulation with DHA, SHA, and CHA employments. Consistently, the corresponding soil Cd removal efficiencies were 43.5% and 34.6% with PFA and PHA, respectively, which hold abundant O- and N-containing groups. Our research aims to gain insights into the ternary interaction in the presence of heavy metal, humic substances, and S. alfredii to simultaneously accelerate Cd activation and hyperaccumulation.
The promotion of the atrazine degradation mechanism by humic acid in a soil microbial electrochemical system

Abstract:The enhancing effects of anodes on the degradation of the organochlorine pesticide atrazine (ATR) in soil within microbial electrochemical systems (MES) have been extensively researched. However, the impact and underlying mechanisms of soil microbial electrochemical systems (MES) on ATR degradation, particularly under conditions involving the addition of humic acids (HAs), remain elusive. In this investigation, a soil MES supplemented with humic acids (HAs) was established to assess the promotional effects and mechanisms of HAs on ATR degradation, utilizing EEM-PARAFAC and SEM analyses. Results revealed that the maximum power density of the MES in soil increased by 150%, and the degradation efficiency of ATR improved by over 50% following the addition of HAs. Furthermore, HAs were found to facilitate efficient ATR degradation in the far-anode region by mediating extracellular electron transfer. The components identified as critical in promoting ATR degradation were Like-Protein and Like-Humic acid substances. Analysis of the microbial community structure indicated that the addition of HAs favored the evolution of the soil MES microbial community and the enrichment of electroactive microorganisms. In the ATR degradation process, the swift accumulation of Hydrocarbyl ATR (HYA) was identified as the primary cause for the rapid degradation of ATR in electron-rich conditions. Essentially, HA facilitates the reduction of ATR to HYA through mediated bonded electron transfer, thereby markedly enhancing the efficiency of ATR degradation.

Abstract:Electricity-stimulated anaerobic system (ESAS) has shown great potential for halogenated organic pollutants removal. Exogenous redox mediators can improve electron transfer efficiency to enhance pollutants removal in ESAS. In this study, humic acid (HA), a low-cost electron mediator, was added into ESAS to enhance the simultaneous reductive debromination and mineralization of 4-bromophenol (4-BP). Results showed that the highest 4-BP removal efficiency at 48 h was 95.43 % with HA dosage of 30 mg/L at − 700 mV, which was 34.67 % higher than that without HA. The addition of HA decreased the requirement for electron donors and enriched Petrimonas and Rhodococcus for humus respiratory. HA addition regulated microbial interactions, and enhanced species cooperation between Petrimonas and dehalogenation species ( Thauera and Desulfovibrio ), phenol degradation-related species ( Rhodococcus ) as well as fermentative species ( Desulfobulbus ). Functional genes related to 4-BP degradation ( dhaA/hemE/xylC/chnB/dmpN ) and electron transfer ( etfB/nuoA/qor/ccoN/coxA ) were increased in abundance by HA addition. The enhanced microbial functions, as well as species cooperation and facilitation, all contributed to the improved 4-BP biodegradation in HA-added ESAS. This study provided a deep insight into microbial mechanism driven by HA and offered a promising strategy for improving halogenated organic pollutants removal from wastewater.

Abstract:The role of Fe(II) on the humic acid (HA) transformation and the effects of humic acid Fe (HA-Fe) on simultaneous partial nitrification, anammox and denitrification (SNAD) system were investigated. After adding Fe(II), the HA content decreased and the HA inhibition on the SNAD system was released. Results showed that Fe(II) and HA formed the lower water-soluble HA-Fe, promoting the HA removal. HA-Fe with stronger electron transfer capacity constituted the interface with microorganisms to forming the biogeobattery effect. This accelerated the microbial electron transfer, as well as improved the key enzymes and ATP, indicating that HA-Fe stimulated the microbial activity of the SNAD system. Microbial community and quorum sensing analysis further demonstrated that HA-Fe enhanced the mutual symbiosis between electroactive and nitrogen removal bacteria, to ensure the stability of the SNAD system. The study provided references for efficient HA removal and revealed the biogeobattery effect of HA-Fe in the SNAD system.

Abstract:Purpose Humins, as solid-phase redox mediators, are important for enhancing anaerobic biotransformation processes. Although humins are broadly distributed in estuarine and marine sediments, to date, their electron transfer capacity (ETC) from marine sediments has been poorly characterized. This study determined the potential of humins as redox mediators for microbial redox metabolic reactions in sediments along a transect from the Yangtze Estuary to the adjacent East China Sea and analyzed their electron-mediating characteristics. Materials and methods Comparative analyses of humins in sediments along this transect were conducted to investigate their ETC as redox mediators for microbial redox metabolic reactions, namely dissimilatory iron reduction (DIR) and dissimilatory nitrate reduction to ammonium (DNRA), using Shewanella oneidensis MR-1. Elemental composition, electrochemical, and Fourier transform infrared analyses were performed to better understand the functional redox groups in humins. Results and discussion All the humins functioned as solid-phase redox mediators and enhanced DIR and DNRA by Shewanella oneidensis , although with varying efficacy. The humins extracted from sampling site P1 (P1-HM), located close to the Yangtze Estuary, most effectively enhanced iron and nitrate reduction without nitrite accumulation. Electrochemical analysis confirmed the highest redox activity and ETC in P1-HM and a weakening trend of ETC with increasing distance from the estuary. Fourier transform infrared analysis suggested that quinone moieties might serve as key redox functional groups in humins. Conclusions Considering their high yield and ETC, P1-HM could be important redox mediators that influence microbial redox metabolic reactions in sedimentary ecosystems of the Yangtze Estuary. Thus, this study provides insights into the distribution of humins, their ECT, and their potential role as redox mediators for microbial redox reactions, which could improve understanding of elemental biogeochemical cycling processes involving microorganisms in the Yangtze Estuary and East China Sea sedimentary ecosystems and provide important reference information for researches on other estuary areas.

Abstract:Abstract The artificial humic acids (AHA) approach contributes to achieving the carbon (C) emission peaking and neutrality goal through efficient recycling of waste biomasses and promotion of plant photosynthesis. However, the dependence of their production processes and photosynthetic promotion mechanisms on feedstocks remains unclear. In this study, waste biomasses including camphor leaves (CL), corn stalks (CS), peanut shells (PS), and mixed cyanobacteria (MC) have been respectively converted into artificial humic acids through an environmentally friendly hydrothermal humification approach. The dynamic humification process of different feedstocks and the composition, structural properties, and electron transfer capacity of AHA products were determined. Moreover, the different AHA products were applied to corn to explore their respective photosynthetic promotion mechanisms. High relative contents of lignin and C/N in feedstocks are not conducive to the formation of photodegradable substances and the redox property in AHA. The application of AHA increased the net photosynthetic rate and biomass C of corn by 70–118% and 22–39%, respectively. The AHA produced from higher H/C (0.19) and hemicellulose content (17.09%) in feedstocks (e.g., MC) increased corn photosynthesis by improving light energy capture and conversion efficiency in the PSII process. In contrast, the AHA produced from a higher content of lignin (19.81%) and C/N (7.67) in feedstocks (e.g., CS) increased corn photosynthesis by providing functional enzymes (proteins) and nutrients for leaves. This work provides new insights into the utilization of renewable resources, and the artificial humic acids approach sheds light on environmental sustainability by constructing a closed loop of C in environments. Abstract:Humins (HMs), the insoluble faction of humic substances (HSs), play a pivotal role in the bioremediation of pollutants by acting as electron shuttles that modulate the interactions between microorganisms and pollutants. This crucial function is intricately linked to their structural composition and electron transfer capabilities. However, the dynamics of the electron transfer capacity (ETC) of HM extracted during the composting process and its determinants have yet to be fully elucidated. This study undertakes a comprehensive analysis of the ETC of HM derived from composting, employing electrochemical techniques alongside spectroscopic methods and elemental analysis to explore the influencing factors, including the electron accepting capacity (EAC), electron donating capacity (EDC), and electron reversible rate (ERR). Our findings reveal substantial variations in the EAC and EDC of HM throughout the composting process, with EAC values ranging from 133.03–220.98 μmol e − gC −1 and EDC values from 111.17–229.33 μmol e − gC −1 . Notably, the composting process enhances the ERR and EDC of HM while diminishing their EAC. This shift is accompanied by an augmented presence of aromatic structures, polar functional groups, quinones, and nitrogen – and sulfur-containing moieties, thereby boosting the HM's EDC. Conversely, the reduction in EAC is associated with a decline in lignin carbon content and the abundance of oxygen-containing moieties, as well as the diminishment of visible fulvic-like and protein-like substances within HM. Importantly, humic-like substances and nitrogen-containing moieties within HM demonstrated the capacity for repeated electron transfer, underscoring their significance in the context of environmental remediation.
