STRUCTURE OF RHODOPSIN AND BACTERIORHODOPSIN

Abstract:Recently much progress has been made in the structural studies of visual pigments and bacteriorhodopsin (bR). In fact, information on the complete or partial amino acid sequences of 12 rhodopsins is available.-The bovine and ovine rhodopsin primary structures have been determined using protein sequencing (Ovchinnikov, 1982; Hargrave et al., 1983; Pappin et al., 1984). From the biochemical and structural characterization of bovine and ovine rhodopsins at the protein level the following general conclusions emerged. (a) The N-terminus of the protein is blocked by the acetyl group. (b) Two oligosaccharide chains are attached to the protein moiety at positions Asn-2 and Asn-15. (c) Retinal binding lysine residue (Lys 296) is located closer to the protein C-terminus. (d) The polypeptide chain consists of alternating hydrophilic and hydrophobic fragments-the characteristic, probably, determining disposition of membrane proteins in the lipid bilayer. Application of the recombinant DNA technique has permitted isolation and characterization of gene fragments encoding protein structures for rhodopsin in different species. It is through this technique that various rhodopsins, which cannot be obtained in reasonable amounts by conventional biochemical techniques, have been identified. Specific oligonucleotide probes were constructed (Baehr et al . , 1983; Nathans et al., 1983) and used to prime reverse transcriptase for synthesis of bovine opsin cDNA from total retinal mRNA. Nathans et ul. (1983) reported that the mRNA homologous sequences in the 6,4 kb gene consist of a 95 basepair 5' untranslated region, a 1044 base-pair region exactly encoding the 348 residue mature bovine opsin, and long -1400 base-pair 3' untranslated region. The 1044 coding region is divided into five exons by four introns. This review summarizes the results of structural investigations of rhodopsins and bR. The latest data are presented on the membrane disposition of these proteins as well as chemical modifications which promote further understanding of structure-functional relationships in the pigments.
