Abstract:Plastic nanoparticles formed from both daily use of plastics and their wastes have emerged as a potential health and environmental hazard. It is necessary to study the biological process of nanoplastics in ecological risk assessment. To address this concern, we quantitatively investigated the accumulation and depuration of polystyrene nanoplastics (PSNs) in the tissues of zebrafish after the aquatic exposure using a quantitative method based on matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS). Via the PSNs-spiked freshwater, zebrafish were exposed to three different concentrations of PSNs for 30 days, followed by 16 days of depuration. The results showed that the amounts of PSNs accumulated in zebrafish tissues were in the following order: intestine > liver > gill > muscle > brain. The uptake and depuration of PSNs in zebrafish both followed pseudo-firstorder kinetics. It was revealed that the bioaccumulation was concentration, tissue and time dependent. When the PSNs concentration is low, the steady state might take longer time (or not occur) than that of a high concentration. After 16 days of depuration, there were still some PSNs present in the tissues particularly in the brain, where it might take 70 days or more to remove 75 % of PSNs. Overall, this work offers important knowledge on the bioaccumulation of PSNs, which may be useful for future studies into the health hazards of PSNs in aquatic environments.
Interactive effects of polystyrene nanoplastics and 6:2 chlorinated polyfluorinated ether sulfonates on the histomorphology, oxidative stress and gut microbiota in Hainan Medaka ( Oryzias curvinotus )

Abstract:Nanoplastics adsorb surrounding organic contaminants in the environment, which alters the physicochemical properties of contaminants and affects associated ecotoxicological effects on aquatic life. The current work aims to explore the individual and combined toxicological implications of polystyrene nanoplastics (80 nm) and 6:2 chlorinated polyfluorinated ether sulfonate (Cl-PFAES, trade name: F-53B) in an emerging freshwater fish model Hainan Medaka ( Oryzias curvinotus ). Therefore, O. curvinotus were exposed to 200 μg/L of PS-NPs or 500 μg/L of F-53B in the single or mixture exposure for 7 days to investigate the effects on fluorescence accumulation, tissue damage, antioxidant capacity and intestinal flora. The PS-NPs fluorescence intensity was significantly higher in the single exposure treatment than it in combined exposure treatment ( p < 0.01). Histopathological results showed that exposure to PS-NPs or F-53B inflicted varying degree of damages to the gill, liver, and intestine, and these damage were also present in the corresponding tissues of the combined treatment group, illustrating a stronger extent of destruction of these tissues by the combined treatment. Compared to the control group, combined exposure group elevated the malondialdehyde (MDA) content, superoxide dismutase (SOD) and catalase (CAT) activities except in the gill. In addition, the adverse contribution of PS-NPs and F-53B on the enteric flora in the single and combined exposure groups was mainly characterised in the form of reductions in the number of probiotic bacteria (Firmicutes) and this reduction was aggravated by the combined exposure group. Collectively, our results indicated that the toxicological effects of PS-NPs and F-53B on pathology, antioxidant capacity and microbiomics of medaka may be modulated by the interaction of two contaminants with mutually interactive effects. And our work offers fresh information on the combined toxicity of PS-NPs and F-53B to aquatic creatures along with a molecular foundation for the environmental toxicological mechanism.

Abstract:Nanoplastics (NPs, < 1000 nm) interact with chemicals and biomolecules to produce chemical-/eco-corona, altering the environmental destiny, bioavailability, and toxicity of plastic particles and co-occurring chemicals. This study employs exogenous (humic acid, HA) and endogenous (bovine serum albumin, BSA) natural organic matter (NOM) to investigate the eco-corona formation on NPs and explore the interfacial effects of eco-corona and 6:2 chlorinated polyfluorinated ether sulfonate (Cl-PFESA, commonly named as F-53B) on zebrafish (Danio rerio) after 7 days of exposure. Our results indicated significant changes in growth and developmental indices of zebrafish embryos among all eco-corona groups (p < 0.05). Additionally, NFB (BSA-corona, 1 mg/L NPs + 200 μg/L F-53B + 10 mg/L BSA), NFH (HA-corona, 1 mg/L NPs + 200 μg/L F-53B + 10 mg/L HA) and NFHB (BSA-HA-corona, 1 mg/L NPs + 200 μg/L F-53B + 10 mg/L BSA + 10 mg/L HA) showed elevated bioaccumulation of NPs, ROS generation and induction of apoptosis. Transcriptomic analysis showed the number of differentially expressed genes (DEGs) in the following order: BSA-HA-corona (NFHB (2953) > HA-corona (NFH (2797) > NH (2721) > F-53B (2292) > NF (2033) > BSA-corona (NFB (687) > NB (450)), and no DEGs were detected in the single NP compared to the control. Further, the PI3K-AKT, immune system, endocrine system, digestive system, infectious diseases, and neurovegetative disease pathways showed sensitive responses in the NFH/NFHB groups compared to those in the NFB group. Therefore, the interactive effects of NPs and F-53B on zebrafish embryos were lower in the presence of BSA-corona than those in HA- or HA-BSA-coronas, indicating a relationship between the formation of diverse eco-coronas on NPs by multiple NOM and an associated increase in the interfacial toxicological effects of plastic particles and F-35B in freshwater organisms.
[Long-term development of carotid stenosis treated by endovascular means: a prospective study in a neurovascular unit].

Abstract:AIMS Even today, unlike endarterectomy, long-term development in carotid angioplasty has still not been well defined. The aim of this study is to describe both the short- and long-term development and the rate of restenosis in a consecutive series of patients with carotid stenosis who underwent endovascular treatment at the Hospital Universitario La Fe; a distinction is made between patients with and without symptoms. PATIENTS AND METHODS From 1999 to 2010, 426 patients were submitted to a carotid angioplasty (25.5% were asymptomatic patients). Clinical follow-ups were conducted in the case of 374 patients. They had annual check-ups and every year a neurosonological study was performed. The aim was to determine what complications occurred during the first 30 days, as well as the long-term complications, to establish the rate of restenosis and to evaluate possible differences between patients with and without symptoms. RESULTS The morbidity and mortality rate in the first month was 4.2% (4.4% in symptomatic patients and 3.8% in asymptomatic patients). The mean follow-up time was 55 months: 8% of patients presented strokes, 11% suffered from myocardial infarct and 24.3% died, with no significant differences between patients with or without symptoms. The rate of restenosis was 17.3% at five years, 7.5% equal to or above 50% and, of those, 1.1% were symptomatic. Five patients with significant restenosis were successfully treated with endovascular techniques. CONCLUSIONS Carotid angioplasty is an effective, safe technique in the hands of an experienced professional; our findings are good, both in the short and the long term, with a low rate of restenosis. If this latter condition occurs, vascular intervention techniques are safe and efficient.
Toxic effect of chronic exposure to polyethylene nano/microplastics on oxidative stress, neurotoxicity and gut microbiota of adult zebrafish (Danio rerio)

Abstract:The rapid development of aquaculture industry has provided a large amount of high-quality animal protein, while the food safety caused by microplastics and nanoplastics (MP/NPs) has become a major concern. In addition, recent evidence has shown the potential toxic effect of PE-MP/NPs, highlighting the need for further research into their environmental and health impacts. Chronic exposure of polyethylene microplastics (PE-MPs) and nanoplastics (PE-NPs) on adult zebrafish were conducted in the present study for 21 d. Organ-dependent oxidative damage induced by MP/NPs was observed. Insignificant differences in neurotoxicity and dysbiosis of gut microbiota were found between MPs and NPs. Changes in glutathione S-transferase (GST), glutathione (GSH), catalase (CAT), lipid peroxidation (LPO), and superoxide dismutase (SOD) showed that MP/NPs induced oxidative damage in gill and intestinal cells of zebrafish. The inhibited AChE activity suggested the potential neurotoxicity of microplastics and nanoplastics (MP/NPs). In addition, chronic exposure increased the alpha-diversity of intestinal microbiota. At the phylum level, the average relative abundance of Proteobacteria increased from 29.73% (control group) to 66.10% (microplastics), 54.84% (nanoplastics) and 60.03% (combined exposure), respectively. Tenericutes decreased from 55.43% (control group) to 20.02% (microplastics), 22.44% (nanoplastics) and 31.77% (combined exposure), respectively. Overall, this study provides new insights and objective evidence for the toxicity assessment of PE-MPs.
Polystyrene Nanoplastics Toxicity to Zebrafish: Dysregulation of the Brain–Intestine–Microbiota Axis

Abstract:In animal species, the brain-gut axis is a complex bidirectional network between the gastrointestinal (GI) tract and the central nervous system (CNS) consisting of numerous microbial, immune, neuronal, and hormonal pathways that profoundly impact organism development and health. Although nanoplastics (NPs) have been shown to cause intestinal and neural toxicity in fish, the role of the neurotransmitter and intestinal microbiota interactions in the underlying mechanism of toxicity, particularly at environmentally relevant contaminant concentrations, remains unknown. Here, the effect of 44 nm polystyrene nanoplastics (PS-NPs) on the brain-intestine-microbe axis and embryo-larval development in zebrafish (Danio rerio) was investigated. Exposure to 1, 10, and 100 μg/L PS-NPs for 30 days inhibited growth and adversely affected inflammatory responses and intestinal permeability. Targeted metabolomics analysis revealed an alteration of 42 metabolites involved in neurotransmission. The content of 3,4-dihydroxyphenylacetic acid (DOPAC; dopamine metabolite formed by monoamine oxidase activity) was significantly decreased in a dose-dependent manner after PS-NP exposure. Changes in the 14 metabolites correlated with changes to 3 microbial groups, including Proteobacteria, Firmicutes, and Bacteroidetes, as compared to the control group. A significant relationship between Firmicutes and homovanillic acid (0.466, Pearson correlation coefficient) was evident. Eight altered metabolites (l-glutamine (Gln), 5-hydroxyindoleacetic acid (5-HIAA), serotonin, 5-hydroxytryptophan (5-HTP), l-cysteine (Cys), l-glutamic acid (Glu), norepinephrine (NE), and l-tryptophan (l-Trp)) had a negative relationship with Proteobacteria although histamine (His) and acetylcholine chloride (ACh chloride) levels were positively correlated with Proteobacteria. An Associated Network analysis showed that Firmicutes and Bacteroidetes were highly correlated (0.969). Furthermore, PS-NPs accumulated in the gastrointestinal tract of offspring and impaired development of F1 (2 h post-fertilization) embryos, including reduced spontaneous movements, hatching rate, and length. This demonstration of transgenerational deficits is of particular concern. These findings suggest that PS-NPs cause intestinal inflammation, growth inhibition, and restricted development of zebrafish, which are strongly linked to the disrupted regulation within the brain-intestine-microbiota axis. Our study provides insights into how xenobiotics can disrupt the regulation of brain-intestine-microbiota and suggests that these end points should be taken into account when assessing environmental health risks of PS-NPs to aquatic organisms.

Abstract:In view of the accumulation of nanoplastics (NPs) in the food chain of environment and animals, and the good adsorption properties of nano-plastics to toxic substances, it is necessary to explore the influence of NPs in living organisms. In this study, single and joint toxicological effects of polystyrene nanoplastics (PS-NPs, size 80 nm) and polychlorinated biphenyls (PCBs), were explored in freshwater aquatic animal model zebrafish (Danio rerio). Our study found that exposure to single PS-NPs induced mild acute toxicity, albeit the combined exposure of PS-NPs and polychlorinated biphenyls aggravated the toxicity of PCBs in a dose-dependent manner. Results from gene expression profiling showed that NPs exposure could activate detoxification process, resulting in a slight up-regulation of antioxidant genes (sod1, gstp1), bone development genes (bmp2, bmp4) and cardiac gene (tbx20); while PCBs suppressed the detoxification through down-regulation of these genes, and the addition of NPs will exacerbate the impact of PCBs on gene suppression. Importantly, the results of in vivo purification experiments found that NPs showed prolonged retention in liver, intestine and gills of zebrafish and they might have crossed biological barrier and accumulate in lipid-rich tissues and excretion does not appear as the significant pathway for their elimination. In conclusion, the toxic effects of polychlorinated biphenyls on chorionic protected embryos were not significant as zebrafish chorion plays an important role in resisting the invasion of pollutants; PCBs can seriously damage the bone and heart development of zebrafish, while the presence of NPs significantly enhanced the toxicity of PCBs in zebrafish, which is an alarming concern for growing NPs levels and ecological safety in aquatic environment.

Abstract:Nanoplastics (NPs) are good carriers of persistent organic pollutants (POPs) such as polybrominated diphenyl ethers (PBDEs) and can modify their bioavailability and toxicity to aquatic organisms. This study highlights the single and combined toxic effects of polystyrene nanoplastics (PS-NPs) and 2,2 ',4,4 '-tetrabromodiphenyl ether (BDE-47, one of the major PBDE congeners) on zebrafish embryos after an exposure of up to 120 hpf. Our results showed that PS-NPs and BDE-47 formed larger particle aggregates during co-exposure, which attached to the surface of the yolk membrane and even changed its structure, and these particles also bioaccumulated in the intestine of zebrafish larvae, compared with the PS-NPs single exposure. Further, the co-exposure significantly increased mortality, accelerated voluntary movements, enhanced hatching rate, and decreased heart rate. Hepatoxicity analyses revealed that the mixture exposure induced a darker/browner liver colour, atrophied liver and greater hepatotoxicity in zebrafish larvae. In addition to increased ROS accumulation, the reduced expression of the antioxidant gpx1a gene and increased expression of cyp1a1 were found after co-treatment. Moreover, ache and chrn7α genes associated with neurocentral development, were significantly downregulated, mainly in the co-exposure group. In conclusion, simultaneous exposure to PS-NPs and BDE-47 exacerbated oxidative stress, developmental impacts, hepatotoxicity, and neurodevelopmental toxicity in zebrafish larvae. Therefore, neurotoxic effects of complex chemical interactions between PS-NPs and persistent organic pollutants in freshwater environments should be paid more attention.

Abstract:There is growing concern that microplastics (MPs), which act as carriers of other organic contaminants, are mistakenly ingested by aquatic organisms, consequently causing unpredictable adverse effects. In this study, zebrafish larvae (6 d post fertilization) were exposed to either 6:2 chlorinated polyfluorinated ether sulfonate (F–53B), polystyrene microplastics (PS-MPs) or their combination for 7 d to evaluate the effects of the presence of PS-MPs on the bioaccumulation and immunomodulation of F–53B. PS-MPs greatly promoted the sorption of F–53B, which reduced the bioavailability and bioaccumulation of F–53B in zebrafish larvae. F–53B, PS-MPs, or their mixture significantly reduced the body weight of zebrafish larvae. Combined exposure of PS-MPs and F–53B resulted in a significant reduction in superoxide dismutase (SOD) and lysozyme activity, indicating the occurrence of oxidative stress and inflammatory response in zebrafish larvae. The content of malondialdehyde (MDA) and immunoglobulin M (IgM) was not affected by F–53B or PS-MPs, but significantly increased in their combined exposure. Furthermore, co-exposure of F–53B and PS-MPs significantly upregulated the transcripts of pro-inflammatory cxcl-clc and il-1β genes and increased the levels of iNOS protein in zebrafish larvae. In addition, enhanced protein expression of NF-κB paralleled the upregulation in the expression of most immune-related genes, suggesting NF-κB pathway was mechanistically involved in these responses. Collectively, the presence of MPs decreased F–53B bioaccumulation, but induced inflammatory stress in larval zebrafish. These findings highlight the health risks of co-contamination of MPs and F–53B in aquatic environments.

Abstract:Microplastics (MPs) (< 5 mm) and nanoplastics (NPs) (< 100 nm) are emerging environmental pollutants and have been proved could cause a series of toxicity in aquatic organisms. In this study, the effects on gut microbiota of adult zebrafish exposed for 21 days to 10 μg/L and 1 mg/L of MPs (8 μm) and NPs (80 nm) were evaluated. We analyzed the intestinal microbial community of zebrafish using high throughput sequencing of the 16S rRNA gene V3–V4 region and also performed transcriptional profiling of the inflammation pathway related genes in the intestinal tissues. Our results showed that both spherical polystyrene MPs and NPs could induce microbiota dysbiosis in the gut of zebrafish. The flora diversity of gut microbiota significantly increased under a high concentration of NPs. At the phylum level, the abundance of Proteobacteria increased significantly and the abundance of Fusobacteria, Firmicutes and Verrucomicrobiota decreased significantly in the gut after 21-day exposure to 1 mg/L of both MPs and NPs. Furthermore, interestingly, the abundance of Actinobacteria decreased in the MPs treatment groups but increased in the NPs treatment groups. At the genus level, revealed that the relative abundance of Aeromonas significantly increased both in the MPs and NPs treatment groups. Moreover, it was observed that NPs increased mRNA levels of il8, il10, il1β and tnfα in the gut, but not in MPs exposure group, indicating that the NPs may have a more serious effect on the gut of zebrafish than MPs to induce microbiota dysbiosis and inflammation in the gut.

Enhanced neurotoxic effect of PCB-153 when co-exposed with polystyrene nanoplastics in zebrafish larvae

Abstract:Nanoplastics (NPs) and persistent organic pollutants such as polychlorinated biphenyls (PCBs) are ubiquitous aquatic pollutants. The coexistence of these pollutants in the environment emphasises the need to study their combined toxicity. NPs can cross biological membranes and act as vectors for other pollutants, whereas PCBs are known for their ability to bioaccumulate and biomagnify. The present work aimed to study the combined toxicity of polystyrene NPs and PCB-153 using physiological (development, heart rate, respiration), behavioural (swimming behaviour) and molecular (transcriptome) endpoints in zebrafish larvae. The results show that exposure to NPs, PCB and their mixture significantly affected the development and respiration in zebrafish larvae. Larvae co-exposed to NPs and PCB exhibited significant hyperlocomotion, whereas no such effect was observed after exposure to NPs or PCB alone. The transcriptomic results revealed that NPs exposure significantly affected several pathways associated with DNA compaction and nucleosome assembly, whereas PCB exposure significantly affected critical neurogenic pathways. In contrast, co-exposure to NPs and PCB generated multi-faceted toxicity and suppressed neurobehavioural, immune-related and detoxification pathways. The study highlights the complex interplay between NPs and PCBs, and documents how the two toxicants in combination give a stronger effect than the single toxicants alone. Understanding the mixture toxicity of these two pollutants is important to assess the environmental risks and developing effective management strategies, ultimately safeguarding ecosystems and human health.

Abstract:Microplastics (MPs) are a ubiquitous pollutant detected not only in marine and freshwater bodies, but also in tap and bottled water worldwide. While MPs have been extensively studied, the toxicity of their smaller counterpart, nanoplastics (NPs), is not well documented. Despite likely large-scale human and animal exposure to NPs, the associated health risks remain unclear, especially during early developmental stages. To address this, we investigated the health impacts of exposures to both 50 and 200 nm polystyrene NPs in larval zebrafish. From 6 to 120 h post-fertilization (hpf), developing zebrafish were exposed to a range of fluorescent NPs (10-10,000 parts per billion). Dose-dependent increases in accumulation were identified in exposed larval fish, potentially coinciding with an altered behavioral response as evidenced through swimming hyperactivity. Notably, exposures did not impact mortality, hatching rate, or deformities; however, transcriptomic analysis suggests neurodegeneration and motor dysfunction at both high and low concentrations. Furthermore, results of this study suggest that NPs can accumulate in the tissues of larval zebrafish, alter their transcriptome, and affect behavior and physiology, potentially decreasing organismal fitness in contaminated ecosystems. The uniquely broad scale of this study during a critical window of development provides crucial multidimensional characterization of NP impacts on human and animal health.

Abstract:Plastic pollution, including micro- and nanoplastics, is a growing concern. Tyre-wear particles (TWPs) are the second largest source of microplastics in the ocean following abrasion of synthetic fibres. In addition to the particles themselves, TWPs contain many harmful chemicals, including 6PPD. This chemical reacts with atmospheric ozone and forms the toxic compound 6PPD-quinone (6PPDq), which poses a danger to aquatic life. There is a knowledge gap in understanding risks associated with the combined toxicity of nanoplastics (NPs) and 6PPDq. The present study aimed to investigate the toxicity of NPs and 6PPDq on adult zebrafish using phenotypic (behaviour, histology) and transcriptomic endpoints. Zebrafish were exposed to four treatments: control (contaminant-free), 50 μg/L 6PPDq, 3 mg/L polystyrene (PS)-NPs, and a combination of 50 μg/L 6PPDq and 3 mg/L PS-NPs. We did not observe locomotory dysregulation in zebrafish exposed to NPs. However, we found significant hyperlocomotion in zebrafish exposed to 6PPDq and this effect was even more substantial after co-exposure with PS-NPs. This study explores the molecular mechanisms behind these effects, identifying genes associated with neurotransmitters and fatty acid metabolism that were dysregulated by the co-exposure. Transcriptomic analysis further showed that both 6PPDq and PS-NPs impacted cellular processes associated with sterol biosynthesis, cholesterol metabolism, and muscle tissue development. The effects on these mechanisms were stronger in co-exposed zebrafish, indicating a heightened risk to cellular integrity and mitochondrial dysfunction. These results highlight the significance of mixture toxicity when studying the effects of NPs and associated chemicals like 6PPDq.

Abstract:The environmental occurrence of nanoplastics (NPs) is now evident but their long-term impacts on organisms are unclear, limiting ecological and health risk assessment. We hypothesized that chronic exposure to low particle concentrations of NPs can result in gut-associated toxicity, and subsequently affect survival of fish. Japanese medaka Oryzias latipes were exposed to polystyrene NPs (diameter 100 nm; 0, 10, 104, and 106 items/L) for 3 months, and histopathology, digestive and antioxidant enzymes, immunity, intestinal permeability, gut microbiota, and mortality were assessed. NP exposures caused intestinal lesions, and increased intestinal permeability of the gut. The trypsin, lipase, and chymotrypsin activities were increased, but the amylase activity was decreased. Oxidative damage was reflected by the decreased superoxide dismutase and alkaline phosphatase and increased malondialdehyde, catalase, and lysozyme. The integrated biomarkers response index values of all NP-exposed medaka were significantly increased compared to the control group. Moreover, NP exposures resulted in a decrease of diversity and changed the intestinal microbiota composition. Our results provide new evidence that long-term NPs exposure impaired the health of fish at extremely low particle concentrations, suggesting the need for long-term toxicological studies resembling environmental particle concentrations when assessing the risk of NPs.
Combined effects of polyethylene and organic contaminant on zebrafish (Danio rerio): Accumulation of 9-Nitroanthracene, biomarkers and intestinal microbiota

Abstract:Microplastics, as emerging pollutant, are predicted to act as carriers for organic pollutants, but the carrier role and bio-toxic effects with other pollutants in environments are poorly acknowledged. In this study, both the single and combined effects of polyethylene (PE, 10 and 40 mg/L) with the particle size of 100-150 μm and 9-Nitroanthracene (9-NAnt, 5 and 500 μg/L) on zebrafish (Danio rerio) had been investigated. The results illustrated that PE could be as 9-NAnt carrier to enter into zebrafish body, but significantly reduced the bioaccumulation of 9-NAnt, due to the occurrence of adsorption interactions between the simultaneous presence of both PE and 9-NAnt. After 4 days, the enzymes activity of cytochrome P4501A, acetylcholinesterase (AChE), superoxide dismutase (SOD), catalase (CAT), lactate dehydrogenase (LDH), and the abundance of malondialdehyde (MDA), lipid peroxide (LPO) responded strongly to low-dose PE exposure (10 mg/L). After 7 days exposure to PE-9-NAnt (40 mg/L), the P4501A activity increased significantly, but the activities of AChE and LDH were inhibited clearly, causing certain neurotoxicity and disorders of energy metabolism to zebrafish. The analysis of integrated biomarker response index (IBR) suggested that PE had greater bio-toxicity to zebrafish in all exposure groups after short-term exposure, but the PE-9-NAnt complex showed greater bio-toxicity after 7 days, which indicated that complex exposure of PE-9-NAnt had a delayed effect on the bio-toxicity of zebrafish. Furthermore, analysis of the intestinal microbiota exhibited that under the conditions of the exposure group with 9-NAnt, the relative abundance of the five dominant bacterial phyla (Proteobacteria, Firmicutes, Fusobacteriota, Bacteroidota and Verrucomicrobiota) changed greatly. Overall, this study confirmed that PE could carry 9-NAnt into fish causing bioaccumulation, but in the case of coexisting exposures, PE reduced 9-NAnt bioaccumulation, suggesting that microplastics with other emerging pollutants in chronic toxicity are probably next objects in future works.

Abstract:Nanoplastics, as emerging environmental pollutants, can transport contaminants across marine environments, polluting pristine ecosystems and being ingested by marine organisms. This transfer poses a severe threat to global aquatic ecosystems and potentially impacts human health through the food chain. Neurobehavioral and reproductive toxicity are critical areas of concern because they directly affect the survival, health, and population dynamics of aquatic species, which can have cascading effects on the entire ecosystem. Using zebrafish as a model organism, we investigated the toxic effects of environmental concentrations of polystyrene nanoplastics (PS-NPs). Behavioral assessments, including the novel tank test and open field test, demonstrated significant neurobehavioral changes, indicating increased anxiety and depressive behaviors. A pathological analysis of brain and gonadal tissues, along with evaluations of neurobehavioral and reproductive toxicity biomarkers, revealed that exposure to PS-NPs leads to brain tissue lesions, inflammatory responses, oxidative stress activation, hormone level disruptions, and gonadal damage. Real-time quantitative PCR studies of reproductive gene expression further showed that PS-NPs disrupt the endocrine regulation pathways of the brain-pituitary-gonadal (BPG) axis, causing reproductive toxicity with sex-specific differences. These findings provide crucial insights into the impacts of nanoplastics on aquatic organisms and their ecological risks, offering theoretical support for future environmental protection and pollutant management efforts.

Exercise intolerance in obese children--is it asthma?

Abstract:This is a pilot study designed to examine the frequency of asthma in obese children who have exertional dyspnea. Obese children who complained of breathlessness with exercise and who denied asthma were invited to enroll. If there was evidence of airflow limitation on spirometry, nebulized albuterol was administered and spirometry was repeated. If there was no significant improvement or if the baseline spirometry was normal, exercise testing was performed. A total of 20 patients (ages from 8 to 16 years) with BMI from 22 to 61 were enrolled. Of the 19 who completed the study, 9 (47.3%) met criteria for asthma. Recognizing and treating asthma may lead to improved exercise tolerance and improved weight status in these obese individuals.

Abstract:The widespread consumption of nanoplastics (NPs) and bisphenol A (BPA) affected the aquatic ecosystem and imposed risks to the safety of aquatic organisms. This study was aimed at assessing the ecotoxicological effects of single and combined exposure to BPA and polystyrene nanoplastics (PSNPs) on the channel catfish ( Ictalurus punctatus ). A total of 120 channel catfish were separated into four groups with triplicate (each contains 10 fish) and exposed to chlorinated tap water (control group), PSNP single exposure (0.3 mg/L), BPA single exposure (500 μg/L) and PSNPs (0.3 mg/L) + BPA (500 μg/L) co-exposure for 7 days. Our results showed a relatively higher intestinal accumulation of PSNPs in co-exposure group, compared to PSNP single exposure group. Histopathological analysis showed that single exposure to PSNPs and BPA caused breakage of intestinal villi and swelling of hepatocytes in channel catfish, while the co-exposure exacerbated the histopathological damage. In addition, co-exposure significantly increased SOD, CAT activities and MDA contents in the intestine and liver, inducing oxidative stress. In terms of immune function, the activities of ACP and AKP were significantly decreased. The expressions of immune-related genes such as IL-1β , TLR3 , TLR5 , hepcidin and β-defensin were significantly up-regulated, and the expression of IL-10 was down-regulated. Additionally, the co-exposure significantly altered the composition of the intestinal microbiota, leading to an increase in the Shannon index and a decrease in the Simpson index. In summary, this study revealed that mixture exposure to PSNPs and BPA exacerbated toxic effects on histopathology, oxidative stress, immune function and intestinal microbiota in channel catfish. It emphasized the threat of NPs and BPA to the health of aquatic organisms and human food safety, with a call for effective ways to regulate the consumption of these anthropogenic chemicals.

Abstract:4-methylbenzylidene camphor (4-MBC) and micro/nanoplastics (MNPs) are common in personal care and cosmetic products (PCCPs) and consumer goods; however, they have become pervasive environmental contaminants. MNPs serve as carriers of 4-MBC in both PCCPs and the environment. Our previous study demonstrated that 4-MBC induces estrogenic effects in zebrafish larvae. However, knowledge gaps remain regarding the sex- and tissue-specific accumulation and potential toxicities of chronic coexposure to 4-MBC and MNPs. Herein, adult zebrafish were exposed to environmentally realistic concentrations of 4-MBC (0, 0.4832, and 4832 μg/L), with or without polystyrene nanoplastics (PS-NPs; 50 nm, 1.0 mg/L) for 21 days. Sex-specific accumulation was observed, with higher concentrations in female brains, while males exhibited comparable accumulation in the liver, testes, and brain. Coexposure to PS-NPs intensified the 4-MBC burden in all tested tissues. Dual-omics analysis (transcriptomics and proteomics) revealed dysfunctions in neuronal differentiation, death, and reproduction. 4-MBC-co-PS-NP exposure disrupted the brain histopathology more severely than exposure to 4-MBC alone, inducing sex-specific neurotoxicity and reproductive disruptions. Female zebrafish exhibited autism spectrum disorder-like behavior and disruption of vitellogenesis and oocyte maturation, while male zebrafish showed Parkinson's-like behavior and spermatogenesis disruption. Our findings highlight that PS-NPs enhance tissue accumulation of 4-MBC, leading to sex-specific impairments in the nervous and reproductive systems of zebrafish.

Abstract:Aquatic environments serve as critical repositories for pollutants and have significantly accumulated micro- and nanoplastics (MNPs) due to the extensive production and application of plastic products. While the disease resistance and immunity of fish are closely linked to the condition of their aquatic habitats, the specific effects of nanoplastics (NPs) and microplastics (MPs) within these environments on fish immune functions are still not fully understood. The present study utilized zebrafish (Danio rerio) embryos and larvae as model organisms to examine the impacts of polystyrene NPs (100 nm) and MPs (5 μm) on fish immune responses. Our findings reveal that NPs and MPs tend to accumulate on the surfaces of embryos and within the intestines of larvae, triggering oxidative stress and significantly increasing susceptibility to Edwardsiella piscicida infection in zebrafish larvae. Transmission electron microscopy examined that both NPs and MPs inflicted damage to the kidney, an essential immune organ, with NPs predominantly inducing endoplasmic reticulum stress and MPs causing lipid accumulation. Transcriptomic analysis further demonstrated that both NPs and MPs significantly suppress the expression of key innate immune pathways, notably the C-type lectin receptor signaling pathway and the cytosolic DNA-sensing pathway. Within these pathways, the immune factor interleukin-1 beta (il1b) was consistently downregulated in both exposure groups. Furthermore, exposure to E. piscicida resulted in restricted upregulation of il1b mRNA and protein levels, likely contributing to diminished disease resistance in zebrafish larvae exposed to MNPs. Our findings suggest that NPs and MPs similarly impair the innate immune function of zebrafish larvae and weaken their disease resistance, highlighting the significant environmental threat posed by these pollutants.
