Abstract:Simulation of an electron gun with a high compression ratio for a Ka-band klystron is presented in this abstract. The electron gun operates under the voltage of 14kV with a beam current of 1A with a p shaped permanent-magnet focusing structure. The beam filling factor is less than 0.7, and the beam ripple of the average beam radius is about 10%. It is helpful for the subsequent engineering practices.

Abstract:The newest knowledge on the osteoclast allows us to consider bone resorption in a global perspective, as the resultant of three successive steps that may each be individually regulated by physiopathologic or pharmacologic agents. The first involves the formation of osteoclast progenitors in hematopoietic tissues followed by their vascular dissemination and the generation of resting preosteoclasts and osteoclasts in bone. The second consists in the activation of osteoclasts at the contact of mineralized bone. Osteoblasts appear to control this step by exposing the mineral to osteoclasts and preosteoclasts and/or by releasing a soluble factor that activates these cells. In a third step, activated osteoclasts resorb both the mineral and the organic of mineralized bone through the action of agents that they secrete in the segregated zone underlying their ruffled border. The mineral appears to be solubilized by hydrogen ions secreted by an ATP-driven proton pump located at that border and fed by protons generated from CO2 by carbonic anhydrase. The removal of organic matrix, which could be prepared by osteoblast collagenase at the level of nonmineralized bone surfaces, appears dependent on acid proteinases, particularly cysteine-proteinases, secreted, together with other lysosomal enzymes, in the acid microenvironment of the resorption zone.

Abstract:Prostate specific antigen (PSA), as a biomarker, plays important roles in early diagnosis of male prostate diseases, especially prostate cancer. In this study, a novel electrochemical probe polymer of poly(ferriporphyrin-co-acrylamide) was synthesized, by further combined with reduced graphene oxide and gold nanoparticles, and oriented immobilization of PSA antibody, a label-free electrochemical immunosensor was obtained, and then successfully applied in the determination of PSA in 10 % non-antigen serum system. The prepared biosensor expressed high selectivity toward PSA, with a LOD of 0.001 ng mL−1, a linear range of 0.01–110 ng mL−1, and a sensitivity of 15.78 µA mL ng−1. What’s more, it showed good stability, well repeatability and high selectivity in the real PSA sample detection. This work provided certain references for the design of the new electrochemical probes and the detection of tumor markers.

Abstract:This work describes a novel affinity peptide–antibody sandwich electrochemical strategy for the ultrasensitive detection of prostate-specific antigen (PSA). Herein, polydopamine-coated boron-doped carbon nitride (@BCN) was synthesized and used as a sensing platform to anchor gold nanoparticles and immobilize primary antibody. Meanwhile, AuPt metallic nanoparticle and manganese dioxide (MnO2)-functionalized covalent organic frameworks (2@COF) was facilely synthesized to serve as a nanocatalyst and ordered nanopore for the enrichment and amplification of signal molecules (methylene blue, MB). PSA affinity peptide was bound to 2@COF to form Pep/MB/2@COF nanocomposites (probe). The peptide–PSA–antibody sandwich biosensor was constructed, and the redox signal of MB was measured with the existence of PSA. The fabricated sensor exhibited a linear response (0.00005–10 ng mL−1) with a low detection limit of 16.7 fg mL−1 under the optimum condition. Additionally, the sensor showed an excellent selectivity, ideal repeatability, and good stability for PSA detection in real samples. Furthermore, the porous structure of COF can enrich more MB molecules and increase the sensitivity of the biosensor. This study provides an efficient and ultrasensitive strategy for PSA detection and broadens the use of organic/inorganic porous nanocomposite in biosensing.

Abstract:Magnetic covalent organic frameworks (COFs) are useful mesoporous materials for the enrichment and separation of analytes, and are utilized in the pretreatment of samples. However, the use of magnetic COFs in electrochemical immunosensors has rarely reported. Herein, a novel electrochemical assay for the determination of prostate specific antigen (PSA) was developed using black phosphorene (BPene) as a platform and magnetic COFs for signal amplification. BPene was prepared via water-phase exfoliation. BPene nanocomposite () was prepared by depositing Au nanoparticles (Au NPs) onto BPene. This nanocomposite was utilized as an immunsensing platform to bind primary antibodies and improve electron transfer. Subsequently, an Au NP-loaded magnetic COF was used to immobilize the secondary antibodies and abundant electronic signals of methylene blue (MB). The fabricated sensor exhibited linearity ranging from 0.0001 ng mL-1 to 10 ng mL−1 with the detection limit of 30 fg mL−1. The sensor could determine the PSA in a real sample with excellent specificity, good stability, and desirable reproducibility. The effective signal amplification of the proposed sensor is attributed to the good electron transfer of , excellent enrichment capacity of signal molecules (MB) of the COF, and efficient catalytic activity of Fe3O4. This work not only provides an effective electrochemical assay to detect PSA in real sample, but also broadens the utilization scope of magnetic COFs in immunosensing.

Abstract:The development of biosensors capable of averting biofouling and detecting biomarkers in complex biological media remains a challenge. Herein, an ultralow fouling and highly sensitive biosensor based on specifically designed antifouling peptides and a signal amplification strategy was designed for prostate specific antigen (PSA) detection in human serum. A low fouling layer of poly(ethylene glycol) (PEG) doped the conducting polymer poly(3,4-ethylenedioxythiophene) (PEDOT) was electrodeposited on the electrode surface, followed by the immobilization of streptavidin and further attachment of biotin-labelled peptides. The peptide was designed to include PSA specific recognition domain (HSSKLQK) and antifouling domain (PPPPEKEKEKE), and the terminal of the peptide was functionalized with -SH group. DNA functionalized gold nanorods (DNA/AuNRs) were then attached to the electrode, and methylene blue (MB) molecules were adsorbed to the DNA to form the signal amplifier. In the presence of PSA, the peptide was specifically cleaved and resulted in the loss of AuNRs together with DNA and MB, and thus significant decrease of the current signal. The biosensor exhibited a low limit of detection (LOD) of 0.035 pg mL-1 (S/N = 3), with a wide linear range from 0.10 pg mL-1 to 10.0 ng mL-1, and it was able to detect PSA in real human serum owing to the presence of the antifouling peptides, indicating great potential of the constructed biosensor for practical application.

Abstract:Prostatic cancer threatens the lives and health of numerous male patients all over the world. As a single-chain glycoprotein, prostate specific antigen (PSA) can be used to perform prostate cancer screening. At present, there are still problems such as time-consuming and low sensitivity in hospital detection. At the same time, electrochemical immunosensor technology has received a lot of attention due to its benefits, including high sensitivity, good selectivity, low usage and cost, and so on. Electrocatalysts are typically used to build electrochemical immunosensors for signal amplification in the recent years to realize sensitive detection of cancer biomarkers such as PSA. In recent years, three-dimensional nanomaterials have been widely used in the study of electrocatalysts, but the study of the sensitivity for PSA detection is still very scarce. The present work reported the application of a 3D graphene aerogel(GA)/AuNPs/Nafion electrochemical platform for highly sensitive PSA quantification. The electrochemical immunosensor has been designed by immobilizing aptamers on the screen-printed electrode (SPE) surface modified by GA/AuNPs/Nafion. Scanning electron microscopy (SEM), Raman spectra, and electrochemical characterization demonstrated the successful combination of GA/AuNPs/Nafion/SPE. Based on electrochemical impedance spectroscopy (EIS), the proposed immunosensor has demonstrated excellent electrochemical performance, exhibiting sensitive and stable responses for electrochemical detection of PSA ranging from 0.05 to 50 ng∙mL −1 with a limit of detection (LOD) of 0.0306 ng∙mL −1 . This study has great potential application in PSA detection and provides a valuable reference for point-of-care testing (POCT) of prostate cancer.

Abstract:An efficient quenching electrochemiluminescence (ECL) immunosensor based on ECL resonance energy transfer (ECL-RET) was studied for sensitive analysis of prostate specific antigen (PSA). In this protocol, nitrogen-doped graphene quantum dots (NGQDs) which could produce excellent ECL emission, were loaded onto Ni(OH)2 with the three-dimensional (3D) hierarchical and stacked lamellar structure. Taking advantages of high conductivity and large specific surface area of Ni(OH)2, the better electrochemical and ECL behavior of Ni(OH)2/NGQDs were presented. As another part of immunosensor, the Fe3O4@MnO2 composites with wider ultraviolet absorption range were successfully assembled. The absorption spectra of Fe3O4@MnO2 precisely overlapped with the ECL spectra of NGQDs, leading to the significant decrement of ECL signal. Herein, the sandwich-type immunosensor was developed based on the quenching mechanism between NGQDs (ECL donor) and Fe3O4@MnO2 (ECL acceptor). The system was optimized to realize reliable determination of PSA concentration within the linear range of 10-5-10ng/mL and a detection limit of 5 fg/mL (S/N = 3). Eminently, the developed method was successfully applied in real serum samples with good recoveries in a range from 94.0% to 102%, indicating the good accuracy of the proposed method for PSA detection.

Abstract:How to fabricate scale low-cost microfluidic device for detection of biomarkers owns a great requirement. Herein, it is for the first time reported that a new microfluidic device based on bonding polydimethylsiloxane microfluidic channels onto the substrate of a screen-printed electrode with coating glass solution was fabricated for electrochemical sensing of prostate-specific antigen (PSA). Compared to traditional microfabrication processes, this method is simple, fast, low cost, and also suitable for mass production. The prepared screen-printed electrode-based microfluidic device (CASPE-MFD) was used for the detection of the PSA in human serum. The prepared CASPE-MFD had a detection limit of 0.84 pg/mL (25.8 fM) and a good linearity with PSA concentration ranging from 0.001 to 10 ng/mL, which showed a great promise platform toward the development of miniaturized, low-cost electrochemical microfluidic device for use in human health, environmental monitoring, and other applications.

Abstract:Based on the designed inverted Y-shaped peptide and MXene nanocomposite (MXene-Au@ZIF-67), a ratiometric anti-pollution electrochemical biosensor was designed and applied to the detection of biomarkers in serum. Au@ZIF-67 inserted into the interior of MXene can not only prevent the accumulation of MXene but also provide a large amounts of binding sites for capturing biomolecules. A designed multifunctional Y-shaped peptide containing anchoring, antifouling, and recognition sequences was anchored onto MXene-Au@ZIF-67 through Au–S bonds. Electrochemical signal molecules, ferrocenecarboxylic acid (Fc) and methylene blue (MB), were modified to another end of multifunctional peptide and interior of MXene-Au@ZIF-67, respectively, to produce a ratiometric electrochemical signal. We selected prostate specific antigen (PSA) as the model compound. PSA specifically recognizes and cleaves the recognition segment in the Y-shaped peptide, and the signal of Fc is reduced, while the signal of MB remains unchanged. The ratiometric strategy endows the present biosensor high accuracy and sensitivity with a detection limit of 0.85 pg/mL. In addition, the sensing surface has good antifouling ability due to the antifouling sequence of the two branching parts of the Y-shaped peptide. More importantly, by replacing the recognition segment of peptides also other targets are accessible, indicating the potential application of the universal detection strategy to the detection of various biomarkers in clinical diagnosis.

Abstract:In the present work, a label-free prostate specific antigen (PSA) electrochemical immunosnesor was constructed based on Au nanoparticles/MoS2-graphene aerogels nanocomposite (AuNPs/MoS2-GAs) as sensing platform. MoS2-GAs composite with porous network structure provided large specific area for the immobilization of AuNPs, which further improved the loading of PSA antibodies. In addition, the AuNPs/MoS2-GAs composite has good conductivity, which could accelerate the electron transport of the electrode interface. Based on these advantages of AuNPs/MoS2-GAs material, the prepared immunosensor exhibited wide linearity in the range of 0.00001–50 ng mL−1 and a low detection limit of 0.003 pg mL−1. The immunosensor also exhibited acceptable stability, good selectivity, and reproducibility. Moreover, when applied to analyze PSA in human serum, the immunosensor obtained good recovervries, indicating the method shows a promising application for early diagnosis of PSA.

Abstract:Prostate-specific antigen (PSA) is the most widely used biomarker for the early diagnosis of prostate cancer. Existing methods for PSA detection are burdened with some limitations and require improvement. Herein, we developed a novel microfluidic-electrochemical (μFEC) detection system for PSA detection. First, we constructed an electrochemical biosensor based on screen-printed electrodes (SPEs) with modification of gold nanoflowers (Au NFs) and DNA tetrahedron structural probes (TSPs), which showed great detection performance. Second, we fabricated microfluidic chips by DNA TSP-Au NF-modified SPEs and a PDMS layer with designed dense meandering microchannels. Finally, the μFEC detection system was achieved based on microfluidic chips integrated with the liquid automatic conveying unit and electrochemical detection platform. The μFEC system we developed acquired great detection performance for PSA detection in PBS solution. For PSA assays in spiked serum samples of the μFEC system, we obtained a linear dynamic range of 1-100 ng/mL with a limit of detection of 0.2 ng/mL and a total reaction time <25 min. Real serum samples of prostate cancer patients presented a strong correlation between the "gold-standard" chemiluminescence assays and the μFEC system. In terms of operation procedure, cost, and reaction time, our method was superior to the current methods for PSA detection and shows great potential for practical clinical application in the future.

Abstract:We describe a highly sensitive electrochemical sandwich-type immunosensor for the prostate specific antigen (PSA), a biomarker related to prostate cancer. A glassy carbon electrode was covalently modified with multi-walled carbon nanotubes to which the anti-PSA was immobilized. Following addition of a sample containing PSA, a secondary antibody is added that consists of gold nanoparticles modified with secondary antibody and 6-ferrocenyl hexanethiol acting as the signal molecule. This approach represents a multiple signal amplification strategy in that the functionalized carbon nanotubes improve the electron transfer on the surface of the electrode, while the gold nanoparticles act as carriers for capturing large quantities of Ab2 and ferrocene. The modifying processes were characterized by cyclic voltammetry and electrochemical impedance spectroscopy. Under the optimal conditions, the differential pulse voltammetric signals (acquired at a typical potential of 0.31 V vs. SCE) is linearly related to the concentration of PSA in the 10 pg.mL−1 to 100 ng.mL−1 range. The detection limit is 5.4 pg.mL−1. When applied to the determination of PSA in spiked human serum, the recoveries were between 92.4 and 120.0 %. Abstract:In this paper, A label-free electrochemical immunosensor for sensitive detection of prostate antigen (PSA) was developed based on NiFe PBA/AuNPs composite. The prostate antigen antibody was immobilized and the immunosensor was constructed by using a glassy carbon electrode modified with a nanocomposite consisting of nickel-iron Prussian blue analog (NiFe PBA) and gold nanoparticles (AuNPs). The good biological affinity of AuNPs for biomolecules, as well as the porous nanostructure and regular shape of NiFe PBA, the NiFe PBA/AuNPs nanocomposites significantly improve the electron transport rate, while achieving the excellent performance of the sensor. Due to the interaction between the antibody and the antigen on the modified electrode, the current signal of the NiFe PBA itself is reduced due to the redox changes in Fe2 + and Fe3 +, which can be determined by differential pulse voltammetry (DPV). Therefore, the monitoring of prostate antigen detection is realized. Under optimal experimental conditions, the immunosensor exhibited excellent detection performance with a dynamic response range from 0.5 pg/mL to 1000 pg/mL for PSA concentration and a detection limit of 0.23 pg/mL (S/N = 3). In addition, the PSA aptasensor has good selectivity, high stability, and satisfactory reproducibility and has broad potential in clinical research and diagnostic applications.

Abstract:Path models with and without latent variables are extensively used in e-collaboration research. Both direct and moderating relationships can be included in such path models. Moderating relationships involve three latent variables, the moderating variable and a pair of variables that are connected through a direct link. This paper discusses the visualization of moderating relationships through two-dimensional and three-dimensional graphs. The software WarpPLS version 5.0 is used in this discussion, since it provides an extensive set of graphs that can be used to visualize moderating effects.

Abstract:Conductive poly(3,4-ethylenedioxythiophene) (PEDOT) composites have emerged as potential candidates for electrochemical biosensors, however, the sensing performance was mainly relied on their structure and composition. In this work, a novel three-dimensional (3D) structural nano-PEDOT-graphene aerogel (nano-PEDOT-GA) composite was designed via the in-situ polymerization of nano-PEDOT on porous 3D GA. SEM revealed that the introduction of GA could induce the growth of worm-like PEDOT. Subsequently, Au nanoparticles (AuNPs) were directed to nano-PEDOT-GA surface by electrodeposition method. Featuring the excellent biocompatibility, high specific surface area and good conductivity of AuNPs/nano-PEDOT-GA, a highly sensitive label-free immunosensor was developed using AuNPs/nanoPEDOT-GA as signal amplification platform for detecting prostate specific antigen (PSA). Under the optimal conditions, the fabricated immunosensor presented a good linear in the analytical range of 0.0001-50 ng mL(-1) with a low detection limit of 0.03 pg mL(-1) (S/N = 3), high selectivity against other serum interfering substance and good stability and reproducibility, which provides a promising platform for the clinical immunoassay of PSA. Unlike most interdigitated electrode biosensors, biorecognition happens in between the interdigitated electrodes, which enhances the sensitivity and limit of detection of the sensor. Using the Faradaic mode electrochemical impedance spectroscopy (EIS) technique to quantify the PSA antigen, the developed sensing platform demonstrates a logarithmic detection of PSA ranging from 0.5 ng/ml to 5000 ng/ml, an estimated LOD down to 0.51 ng/ml in the serum, and a good sensor's reproducibility. The sensor's detection range covers the clinical threshold value at 4 ng/ml and the crucial diagnosis 'grey zone' of 4-10 ng/ml of PSA in serum for an accurate cancer diagnosis. The selectivity test revealed an excellent discrimination of other competing proteins, with a recorded detection signals at 5 ng/ml PSA as high as 7-fold increase versus the human serum albumin (HSA) and 8-fold increase versus the human glandular kallikrein 2 (hK2). The stability test showed an acceptable stability of the aptasensor recorded at six (6) days before the detection signal started degrading below 10% of the peak detection value. The developed sensing scheme is proven to exhibit a great potential as a portable prostate cancer biosensor, also as a universal platform for bio-molecular sensing with the versatility to implement nanoparticles and other surface chemistry for various applications.

Abstract:For the purpose of detecting prostate specific antigen (PSA), a ratiometric electrochemical aptasensor has been developed based on target-aptamer binding triggered isothermal strand displacement polymerase reaction (ISDPR). A ferrocene (Fc)-modified hairpin probe 2 (Fc-HP2) was self-assembled on the modified electrode surface. One terminus of hairpin probe 1 (HP1) bound to PSA and formed the HP1/PSA complex, and the other hybridized with Fc-HP2. Then, the S1 strand hybridized with the stem of Fc-HP2 and was extended with the aid of phi29 DNA polymerase (phi 29) and deoxyribonucleoside triphosphates (dNTPs), and the HP1/PSA complex could be displaced to trigger a new cycle of polymerization reaction. Finally, numerous double-stranded DNAs (dsDNAs) were produced for methylene blue (MB) intercalation. With the raising of PSA concentrations, the electrochemical responses of Fc (IFc) and MB (IMB) decreased and increased, respectively. When IMB/IFc was used as the response signal for quantitative determination of PSA, the proposed strategy possessed a wide linear range (10 pg mL−1–100 ng mL−1) and a low detection limit of 3.2 pg mL−1. Moreover, the proposed aptasensor displayed good performance for PSA determination in spiked human serum samples, showing great application prospects in bioanalysis.

Abstract:In this work, a novel ratiometric electrochemical aptasensor was designed for prostate specific antigen (PSA) detection based on toehold-mediated strand displacement reaction (TMSDR). For PSA detection, PSA could bind with the aptamer sequence in hairpin probe 1 (HP1), and HP1/PSA complex was obtained. Then, in the presence of S3 DNA, HP1/PSA could trigger two TMSDR processes by hybridizing with Fc-S2/S1 double-stranded DNA, which would generate a large amount of Fc-S2 and recycle HP1/PSA. This enabled the hybridization of Fc-S2 with methylene blue labeled thiolated hairpin probe 2 (MB-HP2) immobilized on the electrode surface, causing MB molecules away from the electrode and Fc close to the electrode. Thus, with the increasing PSA concentrations, the current of MB (I MB ) decreased and the current of Fc (I Fc ) increased. The ratio of I Fc /I MB was shown to be linear with the logarithm of PSA concentration from 1 pg mL −1 to 100 ng mL −1 , with a detection limit of 0.28 pg mL −1 . In addition, the aptasensor had good specificity, reproducibility, repeatability, and stability for PSA detection, and was successfully applied to spiked diluted human serum samples.
