Abstract:The fall armyworm (FAW) Spodoptera frugiperda , is a voracious pest of cereals native to the Americas and which invaded Africa in 2016. Chemical control is the main management option, which however remains ineffective and unsustainable. Fungal endophytes are increasingly used as alternative for the management of insect pests of economic importance. This study assessed the potential of eight endophytic fungal isolates to colonize maize plant and their ability to promote seedlings growth through seed and foliar inoculations, as well as their suppressive effects on FAW. Fungal colonization rates of different plant parts by the endophytes varied as per the inoculation methods. Beauveria bassiana ICIPE 279 colonized more than 60% of all the seedling parts while B . bassiana G1LU3 only colonized stem (25%) and leaf (5%) tissues through foliar inoculation. Trichoderma atroviride F2S21, T. asperellum M2RT4, T. harzianum F2R41, Trichoderma sp. F2L41, Hypocrea lixii F3ST1 and Fusarium proliferatum F2S51 successfully colonized all the plant parts and therefore were selected and further evaluated through seed inoculation for their endophytic persistence, effect on plant growth, and pathogenicity to Spodoptera frugiperda immature and adult stages. Weekly assessment showed varied effect of the endophytes on maize plant growth parameters compared to the control. During the first week, percentage colonization of the plant parts ranges between 90%–100%, 65%–100%, and 60%–100%, in the roots, stems, and leaves, respectively for all the five tested isolates. However, the colonization pattern/rates significantly decreased over time for H. lixii F3ST1 in the stems and leaves, and for T. harzianum F2R41 in the leaves and for T. asperellum M2RT4 in the roots. In addition, T. harzianum F2R41 outperformed all the other isolates in boosting the plant height, whereas H. lixii F3ST1 and T. asperellum M2RT4 outperformed all the other isolates in increasing the wet and dry shoots weight. Furthermore, the number of egg masses laid on endophytically-colonized maize plants varied among the treatments. Trichoderma asperellum M2RT4 and H. lixii F3ST1 endophytically-colonized maize plants significantly reduced the number of egg masses and the defoliation/feeding rates of the pest compared to the control. Additionally, T. harzianum F2R41 had the highest negative impact on the pupation and adult emergence of S. frugiperda with a female-biased sex ratio. Our findings indicate that T. asperellum M2RT4, T. harzianum F2R41, and H. lixii F3ST1 hold a potential to be developed as endophytic-fungal-based biopesticides for sustainable management of S. frugiperda and as plant growth promoters.

Abstract:Spodoptera frugiperda, the fall armyworm (FAW), is a highly invasive polyphagous insect pest that is considered a source of severe economic losses to agricultural production. Currently, the majority of chemical insecticides pose tremendous threats to humans and animals besides insect resistance. Thus, there is an urgent need to develop new pest management strategies with more specificity, efficiency, and sustainability. Chitin-degrading enzymes, including chitinases, are promising agents which may contribute to FAW control. Chitinase-producing microorganisms are reported normally in bacteria and fungi. In the present study, Serratia marcescens was successfully isolated and identified from the larvae of Spodoptera frugiperda. The bacterial strain NRC408 displayed the highest chitinase enzyme activity of 250 units per milligram of protein. Subsequently, the chitinase gene was cloned and heterologously expressed in E. coli BL21 (DE3). Recombinant chitinase B was overproduced to 2.5-fold, driven by the T7 expression system. Recombinant chitinase B was evaluated for its efficacy as an insecticidal bioagent against S. frugiperda larvae, which induced significant alteration in subsequent developmental stages and conspicuous malformations. Additionally, our study highlights that in silico analyses of the anticipated protein encoded by the chitinase gene (ChiB) offered improved predictions for enzyme binding and catalytic activity. The effectiveness of (ChiB) against S. frugiperda was evaluated in laboratory and controlled field conditions. The results indicated significant mortality, disturbed development, different induced malformations, and a reduction in larval populations. Thus, the current study consequently recommends chitinase B for the first time to control FAW.

Effectiveness of Entomopathogenic Fungi on Immature Stages and Feeding Performance of Fall Armyworm, Spodoptera frugiperda (Lepidoptera: Noctuidae) Larvae

Abstract:Maize is a major staple crop in China, and the sustainable productivity of this primary crop has been recently threatened by fall armyworm (FAW), Spodoptera frugiperda, invasion. The five fungal isolates, Aspergillus sp. BM-3 and SE-2-1, Cladosporium tenuissimum SE-10, Penicillium citrinum CTD-24, and Beauveria bassiana ZK-5 were assessed for their efficacy in causing mortality against first to sixth instar eggs and neonate larvae seven days post-treatment, and their effects on the feeding performance of sixth instar S. frugiperda larvae at 48 h post-treatment at three concentrations (1 × 106, 1 × 107, and 1 × 108 conidia mL−1) were also assessed. The six instar S. frugiperda larvae were not susceptible to the five tested fungal isolates. However, B. bassiana ZK-5 caused the highest egg mortality of 40, 70, and 85.6% at 1 × 106, 1 × 107, and 1 × 108 conidia mL−1, respectively, followed by P. citrinum CTD-24 (30.6, 50, and 75.6%) and C. tenuissimum SE-10 (25.6, 40, and 55.6%). In addition, B. bassiana ZK-5 caused the highest neonate mortality of 54.3% at 1 × 108 conidia mL−1. B. bassiana ZK-5 and P. citrinum CTD-24 caused cumulative mortality, including 93.3 and 83.3% mortality of eggs and neonates, respectively, at 1 × 108 conidia mL−1. Furthermore, B. bassiana ZK-5 reduced the feeding efficacy of first to third instar S. frugiperda larvae by 66.7 to 78.6%, while P. citrinum CTD-24 and C. tenuissimum SE-10 reduced larval feeding by 48.3 to 57.1% at 1 × 108 conidia mL−1. However, these fungal isolates were less potent in reducing the feeding activity of fourth to sixth instar S. frugiperda larvae (>46% with B. bassiana at 48 h post-treatment). The tested fungal isolates could play an essential role as microbial biopesticides in suppressing the S. frugiperda population in China after further investigations on their efficacy are obtained in the field.

Abstract:Background: 'The fall armyworm, Spodoptera frugiperda', represents a significant threat to maize production, a major staple crop in Asian countries. Methods: In pursuit of more effective control of this insect pest, our study assessed the physiological and biochemical effects of the entomopathogenic fungus Metarhizium anisopliae against the larvae of S. frugiperda. Results: Results revealed that, following nine days of treatment, a high concentration of conidia (1.5x107 conidia/mL-1) was toxic to all stages of larvae (second to fifth instar), resulting in 97% mortality of the second instar, 89% mortality of the third instar, 77% mortality of the fourth instar, and 72% mortality of fifth instar. All larval instars were found to have dose-dependent mortality effects. Treated S. frugiperda larvae further displayed significant physiological, morphological, and behavioral changes. Here, treated larvae displayed significantly lower levels of acetylcholinesterase, α-carboxylesterase, and β-carboxylesterase enzyme activity when compared to control groups. Treated larvae underwent an outward morphological change as the result of a decrease in the exterior cuticle of the anal papillae and a demelanization of the interior cuticle. Treated larvae also exhibited abnormal feeding behaviors as a consequence of the negative impact of conidia treatment on the neuromuscular system. Investigation into the effect of M. anisopliae on the non-target organism, the earthworm Eudrilus eugeniae, revealed that M. anisopliae conidia did not produce significant pathogenicity following three days of treatment. Furthermore, histological analysis revealed no significant effect of the entomopathogenic fungi on the gut tissue of the non-target organism. Conclusion: This study highlights the potential of M. anisopliae in the control of S. frugiperda.

Abstract:Maize is an essential crop of China. The recent invasion of Spodoptera frugiperda , also known as fall armyworm (FAW), poses a danger to the country's ability to maintain a sustainable level of productivity from this core crop. Entomopathogenic fungi (EPF) Metarhizium anisopliae MA, Penicillium citrinum CTD-28 and CTD-2, Cladosporium sp. BM-8, Aspergillus sp. SE-25 and SE-5, Metarhizium sp. CA-7, and Syncephalastrum racemosum SR-23 were tested to determine their effectiveness in causing mortality in second instars, eggs, and neonate larvae. Metarhizium anisopliae MA, P. citrinum CTD-28, and Cladosporium sp. BM-8 caused the highest levels of egg mortality, with 86.0, 75.3, and 70.0%, respectively, followed by Penicillium sp. CTD-2 (60.0%). Additionally, M. anisopliae MA caused the highest neonatal mortality of 57.1%, followed by P. citrinum CTD-28 (40.7%). In addition, M. anisopliae MA, P. citrinum CTD-28, and Penicillium sp. CTD-2 decreased the feeding efficacy of second instar larvae of FAW by 77.8, 75.0, and 68.1%, respectively, followed by Cladosporium sp. BM-8 (59.7%). It is possible that EPF will play an important role as microbial agents against FAW after further research is conducted on the effectiveness of these EPF in the field.

Abstract:Increased attention is being focused on the biological control of agricultural pests using microorganisms, owing to their potential as a viable substitute for chemical control methods. Insect cadavers constitute a potential source of entomopathogenic microorganisms. We tested whether bacteria and fungi isolated from Spodoptera frugiperda (JE Smith) cadavers could affect its survival, development, egg-laying pattern, and hatchability, as well as induce mortality in Anthonomus grandis Boheman adults. We isolated the bacteria Enterobacter hormaechei and Serratia marcescens and the fungi Scopulariopsis sp. and Aspergillus nomiae from fall armyworm cadavers and the pest insects were subjected to an artificial diet enriched with bacteria cells or fungal spores to be tested, in the case of S. frugiperda , and only fungal spores in the case of A. grandis . Enterobacter hormaechei and A. nomiae were pathogenic to S. frugiperda , affecting the survival of adults and pupae. The fungus Scopulariopsis sp. does not affect the survival of S. frugiperda caterpillars and pupae; however, due to late action, moths and eggs may be affected. Aspergillus nomiae also increased mortality of A. grandis adults, as well as the development of S. frugiperda in the early stages of exposure to the diet, as indicated by the vertical spore transfer to offspring and low hatchability. Enterobacter hormaechei and A. nomiae are potential biocontrol agents for these pests, and warrant further investigation from a toxicological point of view and subsequently in field tests involving formulations that could improve agricultural sustainability practices.

Abstract:The gut microflora plays an important role in insect development and physiology. The gut bacterial microbiome of the fall armyworm (FAW), Spodoptera frugiperda, in both cornfield and laboratory-reared populations was investigated using a 16S metagenomic approach. The alpha- and beta-diversity of the cornfield FAW populations varied among sampling sites and were higher than those of the laboratory-reared FAW population, indicating that different diets and environments influence the gut bacterial composition. To better understand the interaction between the microbiome and entomopathogenic fungi (EPF), FAWs from organic and conventionally managed corn fields and from the laboratory-reared colony were inoculated with Beauveria bassiana NCHU-153 (Bb-NCHU-153). A longer median lethal time (LT50) was observed in the Bb-NCHU-153-infected cornfield FAW population than in the laboratory-reared FAWs. In terms of the microbiome, three Bb-NCHU-153-infected FAW groups showed different gut bacterial compositions compared to noninfected FAW. Further investigation of the cooccurrence network and linear discriminant analysis (LDA) of effect size (LEfSe) revealed that the enriched bacterial genera, such as Enterococcus, Serratia, Achromobacter, and Tsukamurella, in the gut might play the role of opportunistic pathogens after fungal infection; in contrast, some gut bacteria of Methylobacterium, Marinomonas, Paenochrobactrum, Pseudomonas, Acinetobacter, Delftia, Dietzia, Gordonia, Leucobacter, Paracoccus, and Stenotrophomonas might be probiotics against EPF infection. These results indicated that EPF infection can change the gut bacterial composition and lead to a pathobiome in the FAW and that some bacterial species might protect the FAW from EPF infection. These findings could be applied to the design of pathobiome-inducing biocontrol strategies.
Bio-Intensive Tactics for the Management of Invasive Fall Armyworm for Organic Maize Production

Abstract:Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae) is an invasive pest native to the American continent. The present study focused on bio-intensive tactics like intercropping, using natural enemies, botanical insecticides and biopesticides for managing S. frugiperda for the organic production of maize in Indian conditions. A total of eight different parasitoids attacking the different stages of S. frugiperda viz., eggs and larvae were found in the study area. The total parasitism rate due to all the parasitoids ranged from 28.37 to 42.44%. The egg-larval parasitoid, Chelonus formosanus Sonan (Hymenoptera: Braconidae) was the dominant parasitoid (12.55%), followed by Chelonus nr. blackburni (Hymenoptera: Braconidae) (10.98%) and Coccygydium sp. (4.85%). About 36.58 percent of the egg masses collected was parasitized by egg parasitoids, among which Telenomus remus (Nixon) (Hymenoptera: Scelionidae) was the dominant parasitoid. The botanicals insecticides such as citronella and annona extract were most effective, resulting in 100% mortality of FAW larvae (168 h after treatment). The essential oil of garlic (100%) was found highly effective in inhibiting egg hatching, followed by geraniol (90.76%). The maize intercropped with lady's finger (okra) recorded significantly the lowest pest infestation and recorded higher grain yield (6.17 q/ha) than other intercropping systems and control (5.10 q/ha). The overall bioefficacy of commercial biopesticides against the larvae of S. frugiperda was in the following order azadirachtin > Metarhizium anisopliae (Metch.) Sorokin (Hypocreales: Clavicipitaceae) > Beauveria bassiana (Balsamo) Vuillemin (Hypocreales: Clavicipitaceae) at 168 h after treatment.

Abstract:The Spodoptera frugiperda is a notorious pest with a broad host range. It severely damages crops, mainly in areas of the globewhere maize and sorghum are grown. The pest is difficult to control due to its adaptive nature and resistance to several insecticides available in the market. So, an identification of the alternative strategy is the prime important in the present context. Insecticidal activities of cyanobacterial extracts were evaluated in the laboratory as a biocomponent against S. frugiperda. The crude extracts of Nostoc muscorum and Spirulina sp. were prepared by using ethanol, methanol and petroleum ether solvents. Soxhlet apparatus was used for extraction. S. frugiperda larvae in their second instar were given access to fragments of maize leaf that had been treated with various cyanobacterial extracts. The findings displayed that the petroleum ether extract of N. muscorum had the lowest LC50 value of 155.22 ppm, followed by petroleum ether extracts of Spirulina, ethanol extract of N. Muscorum, methanol extract of N. muscorum, ethanol and methanol extract of Spirulina with an LC50 values of 456.02, 710, 780, 1050 and 1070 ppm respectively. Later, the effect of LC50 values on many biological parameters like the larval duration and pupal stages, the percentage of pupation, the weight of the pupal stage, the malformation of the pupal and adult stages, adult emergence percentage, fertility and the longevity of the male and female adult stages of S. frugiperda was examined. The gas chromatography-mass spectrometry (GC-MS) was used to analyse the crude extract to identify the bioactive components that were responsible for the insecticidal properties. The major compounds detected were diethyl phthalate (19.87 %), tetradecane (5.03%), hexadecanoic acid, ethyl ester (4.10 %), dodecane (4.03%), octadecane (3.72%), octadecanoic acid, methyl ester (3.40 %), ethyl oleate (3.11 %), methyl ester. octadecenoic acid (3.04 %), heptadecane (3.04 %) and phytol (3.02 %). The presence of several bioactive chemicals in the cyanobacterial extracts may be the reason for their insecticidal actions, thus it can be used as an alternative and new source to combat fall armyworm and other crop pests.
Assessing the Single and Combined Toxicity of Chlorantraniliprole with Bacillus thuringiensis against Maize Fall Armyworm Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae) under Laboratory Conditions

Abstract:Use of synthetic insecticides for the management of fall armyworm (FAW) Spodoptera frugiperda (J. E. Smith) for a longer period will led to development of insecticide resistance. Identification of an eco-friendly synergistic agent to enhance the toxicity potential and reduced pesticide use as well become mandatory in due process. Hence the present study was formulated to find the single and combined toxicity of chlorantraniliprole and Bacillus thuringiensis (Bt) against the 2nd and 3rd larval instars of S. frugiperda. Single toxicity of chlorantraniliprole against 2nd and 3rd larval instars were 0.87 and 1.52 ppm (LC25); 4.08 and 6.50 ppm (LC50), respectively. With respect to Bt, single toxicity against 2nd and 3rd larval instars were 474.39 and 693.48 ppm (LC25); 1008.62 and 1228.62 ppm (LC50), respectively. Combination effect of chlorantraniliprole with Bt revealed that 2nd instar of FAW showed supplemental synergism at LC50 of chlorantraniliprole + LC25 of Bt. In the case of LC50 of chlorantraniliprole + LC50 of Bt, LC25 of chlorantraniliprole + LC50 of Bt and LC25 of chlorantraniliprole + LC50 of Bt combinations, they showed sub additive synergism. In 3rd instar larvae, the combined toxicity results were similar for all the combinations of chlorantraniliprole + Bt except LC25 of chlorantraniliprole + LC50 of Bt where it showed an antagonistic synergism. Activity of Carboxyl Esterase (CarE), Mixed Function Oxidase (MFO) and Glutathione-S-Transferase (GST) were found to be lesser in chlorantraniliprole LC50 + Bt LC25 combinations than single toxicity treatments. Therefore, combined use of chlorantraniliprole with Bt at LC50 of chlorantraniliprole + LC25 of Bt had supplemental synergism on fall armyworm under laboratory condition.
Metagenome-Sourced Microbial Chitinases as Potential Insecticide Proteins

Abstract:Microbial chitinases are gaining interest as promising candidates for controlling plant pests. These enzymes can be used directly as biocontrol agents as well as in combination with chemical pesticides or other biopesticides, reducing their environmental impact and/or enhancing their efficacy. Chitinolytic enzymes can target two different structures in insects: the cuticle and the peritrophic matrix (PM). PM, formed by chitin fibrils connected to glycoproteins and proteoglycans, represents a physical barrier that plays an essential role in midgut physiology and insect digestion, and protects the absorptive midgut epithelium from food abrasion or pathogen infections. In this paper, we investigate how two recently discovered metagenome-sourced chitinases (Chi18H8 and 53D1) affect, in vitro and in vivo, the PM integrity of Bombyx mori, a model system among Lepidoptera. The two chitinases were produced in Escherichia coli or, alternatively, in the unconventional - but more environmentally acceptable - Streptomyces coelicolor. Although both the proteins dramatically altered the structure of B. mori PM in vitro, when administered orally only 53D1 caused adverse and marked effects on larval growth and development, inducing mortality and reducing pupal weight. These in vivo results demonstrate that 53D1 is a promising candidate as insecticide protein.

Abstract:The fall armyworm, Spodoptera frugiperda (J.E. Smith), is a destructive transboundary lepidopterous pest affecting more than 78 countries worldwide by infesting a wide array of agronomic and horticultural crops such as maize and cabbage. In a previous study, we demonstrated the synergized toxicity of three local plant extracts (Rhazya stricta Decne., Sophora mollis (Royle) Baker and Withania somnifera (L.) Dunal) with synthetic insecticide chlorantraniliprole against 3rd instar larvae of S. frugiperda. Here we evaluated the combined efficacy of indigenous entomopathogenic fungi (EPF) (Beauveria bassiana (Balsam) Vuillemin and Metarhizium anisopliae (Metschn.) Sorokin) along with above-mentioned plant extracts and chlorantraniliprole against 3rd instar larvae of S. frugiperda under laboratory and semi-field conditions. Toxicity bioassays with two EPF revealed that the highest concentration (1.0 × 109 conidia per mL) of B. bassiana caused maximum larval mortality (76%) at 10th day post-treatment, followed by M. anisopliae, causing 70% larval mortality at the same concentration. In vitro evaluation of six binary combinations of LC33s of EPF (B. bassiana and M. anisopliae) and plant extracts revealed a synergized toxicity (83–87%) of four combinations comprising S. mollis and W. somnifera with B. bassiana and M. anisopliae against 3rd instar S. frugiperda larvae. Furthermore, these insecticidal combinations including chlorantraniliprole (@ half of the label-recommended dose) exhibited as well a significant synergized toxicity (i.e. 37.75% at 1st day to 99.25% at 7th day post-treatment) under semi-field conditions, suggesting their further investigation under field against S. frugiperda and on its natural enemies.

Abstract:Background Maize is an economical crop of China, and its production has been severely affected by the invasive Spodoptera frugiperda in recent year. Application of synthetic pesticides are one of the most effective practices against FAW as an emergency control. This pest causes serious damage to the maize crop worldwide in recent decade, especially in China. Methods To find an alternative to synthetic insecticides there were total 16 different chemicals were used including ten botanical insecticides comprising of seven botanicals, azadirachtin, pyrethrin, nicotine, osthole, rotenone, Celastrus angulatus , matrine, and three insect growth regulators, diflubenzuron, lufenuron and buprofezin. Six synthetic insecticides, including emamectin benzoate, indoxacarb, imidacloprid and thiamethoxan, chlorantraniliprole, chlorfenapyr, were evaluated against 2nd instar of S. frugiperda larvae using leaf-dip method. Results: The results revealed that osthole, azadirachtin, buprofezin and pyrethrin were showed the significant larval mortality of 98.0, 96.7, 94.0 and 90.7%, in 120 h observation and exhibiting minimum LC 50 (39.04, 35.58, 61.45 and 48.46 mg/L, respectively) and LT 50 (48.91, 68.85, 58.67 and 58.57 hours, respectively) values. Among tested synthetic insecticides, emamectin benzoate, chlorantraniliprole and chlorfenapyr were showed significant higher mortality against larvae of S. frugiperda (99.3, 96.0 and 89.3%, respectively) in 72 h observation by exhibiting minimum LC 50 (0.26, 0.39 and 0.72 mg/L, respectively) and and LT 50 (10.18, 10.57 and 13.42 hours, respectively) values. More study is needed to test the laboratory findings in the field, although the efficient biorational pesticides might be utilized as part of integrated pest management against S. frugiperda . Conclusion The effective chemicals could be used in the management for S. frugiperda . The highest discriminating concentrations of tested botanical insecticides, insect growth regulators and insecticides caused significant mortality of S . frugiperda larvae.

Abstract:The fall armyworm is an exotic pest which destroys a wide variety of crops Querywhereas the brinjal ash weevil is a serious pest of eggplant and other solanaceous vegetables. The goal of this research is to find a sustainable and ecologically friendly bio-control agent for managing FAW and brinjal ash weevils. Twelve natural Bacillus thuringiensis strains were isolated from cowpea root nodules, and the Gram-positive cells with characteristic Bt crystal structures were discovered using phase contrast and scanning electron microscopy. There were bipyramidal, cuboidal, rhombus, and spherical crystals. The Bt cry gene content was characterized by PCR analysis, which revealed the presence of cry1 , cry1I , cry3 , cry7 , cry7 , 8 , cry14 , cry26 , and cry55 genes. The identity of Bt was confirmed by cloning and sequencing the cry genes. In the nucleotide sequences, no pseudo genes or indels were found in cry sequences. SDS-PAGE examination indicated the presence of bands ranging in size from 13 to 130 kDa, with 50–60 kDa being the most common. When compared to the control, the new native Bt strains were lethal, with pathogenicity ranging from 93 to 100% against S. frugiperda larvae and M. subfasciatus adults. The studies revealed that the native strains with conserved regions of 16S rRNA genes were compared to NCBI database sequences and classified as native Bt strains with 99–100% similarity to known Bt strains. In conclusion, native Bt strains from cowpea root nodules were shown to have bio-insecticidal activity against fall armyworm larvae and brinjal ash weevil adults.

Abstract:The fall armyworm (FAW), Spodoptera frugiperda (Lepidoptera: Noctuidae) is a recent invasive pest species that has successfully established across sub-Saharan Africa where it continues to disrupt agriculture, particularly smallholder cereal production. Management of FAW in its native range in the Americas has led to the development of resistance to many commercial pesticides before its arrival in Africa. Pesticide use may therefore be ineffective for FAW control in Africa, so new and more sustainable approaches to pest management are required that can help reduce the impact of this exotic pest. Pesticidal plants provide an effective and established approach to pest management in African smallholder farming and recent research has shown that their use can be cost-beneficial and sustainable. In order to optimize the use of botanical extracts for FAW control, we initially screened ten commonly used plant species. In laboratory trials, contact toxicity and feeding bioassays showed differential effects. Some plant species had little to no effect when compared to untreated controls; thus, only the five most promising plant species were selected for more detailed study. In contact toxicity tests, the highest larval mortality was obtained from Nicotiana tabacum (66%) and Lippia javanica (66%). Similarly, in a feeding bioassay L. javanica (62%) and N. tabacum (60%) exhibited high larval mortality at the highest concentration evaluated (10% w/v). Feeding deterrence was evaluated using glass-fibre discs treated with plant extracts, which showed that Cymbopogon citratus (36%) and Azadirachta indica (20%) were the most potent feeding deterrents among the pesticidal plants evaluated. In a screenhouse experiment where living maize plants infested with fall armyworm larvae were treated with plant extracts, N. tabacum and L. javanica were the most potent species at reducing foliar damage compared to the untreated control whilst the synthetic pesticide chlorpyrifos was the most effective in reducing fall armyworm foliar damage. Further field trial evaluation is recommended, particularly involving smallholder maize fields to assess effectiveness across a range of contexts.
Virulence of Beauveria sp. and Metarhizium sp. fungi towards fall armyworm (Spodoptera frugiperda)

Abstract:The development of effective pest management strategies for Spodoptera frugiperda is a high priority for crop protection across its invasive ranges. Here, we examined six Beauveria and five Metarhizium fungal isolates against this pest. Two Beauveria isolates (B-0571, B-1311) induced high mortality toward 3rd and 6th instar caterpillars and adults. For B-0571 mortality was 82.81 ± 5.75%, 61.46 ± 6.83%, and 93.75 ± 3.61%, and 73.72 ± 2.51%, 71.88 ± 5.41%, and 97.92 ± 2.08% for B-1311, with deaths in caterpillars largely occurring under 24 h (3rd instar control 0.74 ± 0.33%, B-0571 73.96 ± 7.85% and B-1311 62.08 ± 3.67%; 6th instar control 0%, B-0571 66.67% ± 11.02% and B-1311 62.5% ± 9.55%). Infection from both Beauveria isolates fully prevented reproduction in surviving S. frugiperda females. In contrast, all five Metarhizium isolates tested and the remaining four Beauveria isolates exhibited lower virulence. The discovery of two highly virulent Beauveria fungal isolates to S. frugiperda opens avenues to develop novel biological control tools against this highly invasive pest.
Chitin-enriched insect frass fertilizer as a biorational alternative for root-knot nematode (Meloidogyne incognita) management

Abstract:Root-knot nematodes ( Meloidogyne spp.) are serious pests of most food crops, causing up to 100% yield loss. Nevertheless, commercial nematicides are costly and harmful to the environment. While the nematicidal potential of crustacean and synthetic chitin has been demonstrated globally, research on the potential of insect-derived chitin for nematode control has received limited attention. Here, seven chitin-fortified black soldier fly frass fertilizer extracts (chFE) were assessed for their suppressiveness of Meloidogyne incognita and impacts on spinach growth in comparison with a commercial nematicide using in vitro and in vivo bioassays. The performance of chFE and control treatments was assessed by determining their effects on nematode egg hatchability; infective juvenile (J2) mortality and paralysis; number of galls, egg masses, and J2s per plant; and spinach root and shoot biomass. In vitro results showed that chFE and commercial nematicide suppressed nematode egg hatchability by 42% and 52%, respectively, relative to the control (sterile distilled water). Up to 100% paralysis was achieved when M. incognita J2s were exposed to either chFE or commercial nematicide. Further, the J2 mortality achieved using chFE (95%) was comparable to the value achieved using commercial nematicide (96%); in all treatments, mortality increased with exposure time. Similarly, up to 85% suppression of gall development was achieved when spinach plants were grown in soil drenched with chFE; up to 79% reduction in egg mass formation and 68% suppression of J2 development in the root system were achieved using chFE. Also, chFE application significantly increased spinach root and shoot biomass by 54%–74% and 39%–58%, respectively, compared to commercial nematicide. Our findings demonstrate the nematicidal potential of chFE and its benefits on crop production. Thus, chFE could be considered as a promising multipurpose, regenerative, and cost-effective input for sustainable management of plant-parasitic nematodes and enhancement of crop yield.

Abstract:The invasive fall armyworm, Spodoptera frugiperda (J.E. Smith; Lepidoptera: Noctuidae), is considered one of the most harmful pests in corn, rice and cotton crops and has developed resistance to the most widespread methods used to control it, chemical insecticides and transgenic crops based on Bt toxins. On the other hand, gut microbiota plays an important role in the insect's fitness, and metatranscriptomic analyses of the insect host are an invaluable tool for identifying new pest control targets. Notwithstanding, there are extremely few reports on the functionally active profile of the gut microbiota in insects, especially in lepidopterans. Moreover, most studies have only described the bacterial community even though all the components of the microbiota, Bacteria, Archaea, fungi, protozoa and viruses, influence different physiological aspects of the insect host. This is the first time an unbiased and comprehensive metatranscriptomic approach has been used to describe the taxonomic and functional profile of the gut microbiota of S. frugiperda larvae, both bacterial and non‐bacterial. We identified novel and very active components which were putatively playing an important nutritional role, such as ammonia‐oxidizing Archaea and Bacteria, and a xylan‐ and hemicellulose‐degrading Actinobacteria. This analysis also allowed us to identify potential biocontrol agents.

Abstract:The fall armyworm (Spodoptera frugiperda, J.E. Smith) is one of the most important agricultural pests in the world and causes serious damage to many significant crops. Insect gut microbiota plays a vital role in host immunity, digestion, and development, helping the higher organism colonize in a new environment. However, the effects of different diets on midgut microbial composition and host immunity in S. frugiperda remain unclear. So far, no reports have compared the gut microbiota of fall armyworm reared using an artificial diet compared to corn leaf in Guangzhou, China. High-throughput 16S rRNA sequencing technology was applied to gain insight into the composition of the gut microbiota of S. frugiperda feeding on corn leaf (field diet) and on a starch-rich artificial diet (lab diet). The fall armyworm gut microbiota was dominated by the bacterial phyla Firmicutes and Proteobacteria. Despite the difference in diet, the core bacterial community was represented by the genus Enterococcus. However, the bacterial community is dominated by a few phylotypes, namely operational taxonomical units 1 (OTU1) (Enterococcus casseliflavus), OTU3 (Enterobacteriaceae), OTU2 (Weissella), and OTU4 (Clostridium), accounting for 97.43% of the total OTUs in the complete dataset. A significant difference was identified in the bacterial communities between the "lab diet" and the "field diet" groups. OTU1 and OTU2 were significantly higher in the "field diet" group, whereas OTU3 and OTU4 were higher in the "lab diet" group. A phylogenetic investigation of the communities by reconstruction of unobserved states (PICRUSt) predicted functional analysis indicates the presence of several genes associated with plant biomass degradation. Importantly, antibiotic-mediated perturbation of the midgut microbial community significantly impacts the expression profile of the important immune genes of the host. Furthermore, the oral reintroduction of gut bacterial isolates (E. mundtii and E. gallinarum) significantly enhances host resistance to AcMNPV infection. Taken together, our results indicate that diet composition is an important driver in shaping insect gut microbiome and immune gene expression, ultimately playing an important role in the pest defense system.
Combining insect pathogenic fungi and a pheromone trap for sustainable management of the fall armyworm, Spodoptera frugiperda (Lepidoptera: Noctuidae)

Abstract:Fall armyworm, Spodoptera frugiperda (Lepidoptera: Noctuidae), is a key invasive pest of maize and other crops in Africa. Entomopathogenic fungi play an important role in regulating the immature stages of this invasive pest as opposed to synthetic pesticides that are hazardous to human, environment and biodiversity. To tackle the adult stage of the pest (the moth) and to improve on the application strategy of the fungal-based biopesticides, this study evaluated the effect of various entomopathogenic fungi isolates on S. frugiperda moths. Twenty-two isolates (16 Metarhizium anisopliae and 6 Beauveria bassiana) were screened in the laboratory to assess their pathogenicity and virulence against S. frugiperda moths. The compatibility of the most pathogenic isolates with S. frugiperda pheromone FALLTRACT lure, the horizontal transmission of the inoculum among S. frugiperda moths, and the effect on oviposition were also determined under laboratory conditions. All 22 fungal isolates screened were pathogenic to the moths, but the mortality varied significantly among the isolates (P < 0.0001) seven days post-treatment. Beauveria bassiana ICIPE 621 and M. anisopliae ICIPE 7 outperformed all the other isolates by causing 100% mortality of the moths with the lowest LT50 values of 3.6 ± 0.1 and 3.9 ± 0.0 days, respectively. Both isolates were also found compatible with FALLTRACT lure, as the lure had no effect on the conidial germination in the laboratory. Male and female moths were able to horizontally transmit conidia of both fungal isolates to untreated moths, causing high mortality of S. frugiperda in 'donor' and 'recipient' groups. In addition, the oviposition, hatchability of eggs and longevity of larvae were significantly affected on the fungal infected females. Although single moths still retained high conidial numbers 72 h post-inoculation, the number of conidia decreased with time. These results suggest that ICIPE 7 and ICIPE 621 could be used in combination with S. frugiperda pheromone in an autodissemination approach to suppress S. frugiperda population.
