Abstract:Matrix metalloproteinases (MMPs) play important roles in cancer initiation and development. Several polymorphisms in the promoters of a number of AMP genes, which can affect the respective MMP production in an allele,specific manner, have been well characterized. We examined whether these functional polymorphisms were related to the risk of nasopharyingeal carcinoma (NPC) in Chinese populations. Eight polymorphisms in the promoter of MMPI, MMP2, MMP3, MMP7, MMP9, MMP12, and MMP13 were genotyped in two independent case-control populations; one is from Guangxi province (593 patients with NPC and 480 controls), and the other is from Guangdong province (239 patients and 286 controls). We observed significantly increased susceptibility to NPC for the MMP2-1306CC (rs243865:C > T) (odds ratio [OR] = 2.01, 95% confidence interval [CI] = 1.30-3.10) and -735CC (rs2285053:C > T) (OR = 1.569 95% CI = 1.17-2.09) genotype carriers compared with noncarriers in the Guangxi population. This association was confirmed in the Guangdong population (for -1306CC: OR= 2.19, 95% CI = 1.21-3.96; for -735CC: OR = 1.60, 95% CI = 1.13-2.28). The C-1306-C-735 haplotype was also significantly associated with increased susceptibility to NPC in both the Guangxi (OR = 1.64, 95% CI = 1.35-1.99) and Guangdong population (OR = 1.68, 95% CI = 1.29-2.19). Furthermore, stratified analysis indicated that the increased susceptibility to NPC related to the -1306CC and -735CC genotype and the C-1306-C-735 haplotype was more pronounced in heavier smokers. Our findings suggest that the genetic polymorphisms or haplotype in the MMP2 promoter may play a role in mediating the susceptibility to NPC in Chinese populations.

Local hemorrhagic reactions in the mouse and the rat after intralabial injection of endotoxin; their relationship to the Arthus and the Shwartzman reactions.

Abstract: Hemorrhagic reactions were elicited in mice when the lip was used as the site of injection of preparatory endotoxins. Two methods were used to demonstrate the reaction. In the first method, intralabial preparation was followed by the injection of endotoxin 19–21 hr later into the peritoneal cavity or tail vein. The results were not regular using this technique, and with only one of the endotoxin preparations did a small but significant number of mice show typical hemorrhagic reactions. The other method involved the use of mice sensitized to foreign proteins. The injection of the homologous antigen into the endotoxin-prepared lip of sensitized mice 19–21 hr after preparation regularly caused a hemorrhagic reaction in that lip. Hemorrhagic reactions were demonstrated in rats showing strong edematous reactions when the second method was used, but no Shwartzman reactions were obtained in rats prepared in the lip with endotoxin and given an eliciting injection of endotoxin intravenously or intra-abdominally. Hemorrhagic reactions were not elicited in sensitized rats showing weak or transient Arthus reactions in the prepared lip.
Association of Matrix Metalloproteinase‐2 and ‐9 Promoter Polymorphisms with Colorectal Cancer in Chinese

Abstract:Matrix metalloproteinase‐2 (MMP‐2) and ‐9 (MMP‐9) play an important role in cancer initiation, invasion, and metastasis. The aim of this study was to investigate whether common genetic variants in these two key MMPs are associated with the development and progression of colorectal cancer in a Chinese population. We detected the MMP‐2‐790T/G, ‐955A/C, ‐1575G/A, and MMP‐9‐1562 C/T polymorphisms in 126 colorectal cancer patients and matched normal controls by PCR‐denaturing high‐performance liquid chromatography (DHPLC) or PCR‐restriction fragment length polymorphism (RFLP), respectively. We found that the G/G genotype in the MMP‐2‐1575G/A polymorphism was significantly increased in colorectal cancer patients than in the controls (odds ratio (OR), 1.96; 95% CI, 1.06–3.64). Colorectal cancers with G/G genotype were more common with serosa/adventitia layer involvement than those with G/A + A/A genotypes (P < 0.05). However, no significant differences were observed in distribution of the MMP‐2‐790T/G, ‐955A/C, and MMP‐9‐1562 C/T polymorphisms or haplotype of MMP‐2 SNPs between patients and controls. Our results suggest that the presence of ‐1575G allele in the MMP‐2 promoter region may be of significance in the assessment of colorectal cancer risk and invasive potential. © 2007 Wiley‐Liss, Inc.
Impact of NOTCH1 Polymorphisms on Liver Cancer in a Chinese Han Population

Abstract:NOTCH1, a member of the Notch family, is up-expression in advanced liver cancer (LC) patients and is associated with tumor sizes, tumor stage, metastasis, and invasion. A few studies have discovered the contribution of NOTCH1 variants to LC risk. Our purpose was to assess the relationship of NOTCH1 rs10521, rs2229971, and rs4489420 to LC risk. We enrolled 709 LC patients and 708 healthy controls. Genotyping was determined through the Agena MassARRAY system. Multiple genetic models by logistic regression were useful for odds ratios (ORs) with 95% confidence intervals (CIs). Rs10521-G (p = 0.009, OR = 0.75, 95% CI: 0.61–0.93), rs2229971-A (p = 0.023, OR = 0.81, 95% CI: 0.67–0.97), and rs4489420-A (p = 0.014, OR = 0.38, 95% CI: 0.16–0.85) might be protective factors for LC occurrence in the Chinese Han population, especially rs10521 and rs2229971 (false-positive report probability (FPRP) <0.2 and statistical power >90%). Interestingly, stratified analysis displayed that the contribution of NOTCH1 polymorphisms to LC risk might be associated with gender, age, smoking, and drinking. Our data first determined that NOTCH1 rs10521-G, rs2229971-A, and rs4489420-A might be protective factors for LC susceptibility.

Abstract:OBJECTIVE:To investigate whether the polymorphisms of matrix metalloproteinase-2 (MMP-2) and -9 (MMP-9) promoters contribute to the development and progression of colorectal cancer in Chinese population.METHODS:the PCR-based denaturing high-performance liquid chromatography or PCR-restriction fragment length polymorphism technique respectively was applied to analyze the MMP-2 -1306C/T and MMP-9 -1562C/T polymorphisms in normal group (126 individuals) and colorectal cancer group (126 cases). Genotype frequencies were compared between patients and matched controls, and the association of genotypes with clinical-pathological parameters was studied.RESULTS:The frequency of the CC genotype in the MMP-2 gene polymorphism was significantly increased in colorectal cancer patients when compared with controls (P<0.05), and individuals with the CC genotype had an increased risk of developing colorectal cancer compared to those with CT+TT genotypes (OR: 1.959; 95%CI: 1.055-3.637). Significant correlation was found between the depth of tumor invasion and MMP-2 -1306C/T polymorphism in colorectal cancer patients. However, the genotype frequencies of MMP-9 -1562C/T in colorectal cancer patients were similar to those in control subjects.CONCLUSION:Our results indicate that MMP-2 -1306 C/T polymorphism may be associated with genetic susceptibility to colorectal cancer and the invasive capability of colorectal cancer in Chinese patients. And it is easier for the CC genotype cancer to invade through bowel wall.

Abstract:Glutathione S-transferases (GSTs) participate in the detoxification and elimination of electrophilic carcinogens by conjugating them to glutathione. Previous studies have reported a potential association between GSTM1 polymorphism and the risk of developing nasopharyngeal cancer (NPC). However, those findings remain controversial. In the present study, a meta-analysis was conducted by pooling the odds ratios (ORs) with the corresponding 95% confidence intervals (CIs) of all the available case-control studies on NPC. A comprehensive search of PubMed, Embase, Web of Science and China National Knowledge Infrastructure databases up to May 13th, 2014 was performed to identify eligible studies. A total of 12 separate publications, involving 1,593 cases of NPC and 2,868 controls, were included in the meta-analysis. The results demonstrated that the null genotype of GSTM1 was significantly associated with increased risk of developing NPC (OR=1.530, 95% CI=1.348-1.737, P-heterogeneity = 0.370). Subgroup analysis by ethnicity suggested that Asian carriers of the GSTM1 null genotype were more susceptible to NPC than individuals from other ethnic groups (OR=1.516, 95% CI=1.328-1.731, P-heterogeneity = 0.270). Sensitivity analysis confirmed the stability of these observations. In conclusion, the results from the present meta-analysis indicated that the GSTM1 polymorphism may be involved in the development of NPC, particularly in Asians.

Abstract:Matrix metalloproteinase-2 (MMP-2) is an enzyme with proteolytic activity against matrix proteins, particularly basement membrane constituents. A single nucleotide polymorphism C→T transition at −1306, which disrupts an Sp1-type promoter site (CCACC box), displayed a strikingly lower promoter activity with T allele. Our study investigated whether the MMP-2 −1306 C→T polymorphism contributed to the development and progression of colorectal cancer in the Chinese population. One hundred twenty-six colorectal cancer patients and 126 age- and sex-matched controls were included in this study. PCR-based denaturing high performance liquid chromatography analysis and sequencing were used to determine the MMP-2 genotypes. MMP-2 expression of each genotype was analyzed in four colorectal cancer cell lines by semi-quantitative RT-PCR. The correlation between the genotypes and clinicopathological parameters among colorectal cancer cases was investigated. The results showed that the levels of MMP-2 mRNA expression in cell lines containing CC genotype were much higher compared with cell with CT genotype. The frequency of MMP-2 CC genotype was significantly higher in colorectal cancer patients when compared with controls (OR, 1.959; 95% CI, 1.055–3.637). Colorectal cancers with CC genotype were more common with serosa/adventitia layer involvement compared with CT+TT genotypes. Our data suggest that MMP-2 −1306 C→T polymorphism may be associated with colorectal cancer development and invasion in the Chinese population.
Association of MCP‐1 Promoter Polymorphism with Susceptibility to Nasopharyngeal Carcinoma

Abstract:Nasopharyngeal carcinoma (NPC) is prevalent among populations from southern China and is influenced by both genetic and environmental risk factors. The monocyte chemoattractant protein-1 (MCP-1), a member of cysteine-cysteine chemokine family, plays critical roles in cancers. A polymorphism within the MCP-1 promoter, rs1024611, has been shown to be significantly associated with the risk of several cancers. Our purpose was to assess the role of rs1024611 in NPC susceptibility. By polymerase chain reaction-restriction fragment length polymorphism method, we genotyped rs1024611 in 593 patients with NPC (cases) and 480 cancer-free subjects (controls) among Guangxi population from southern China. We observed that the G allele of rs1024611 was significantly associated with the increased risk of NPC in an additive model and dominant model, respectively (P = 0.018 and 0.010, odds ratio = 1.25 and 1.41, respectively). No appreciable variation of the effects was found across the subgroups stratified by age, sex, nationality, smoking and drinking status, and smoking level. In addition, significantly higher messenger RNA (mRNA) expression level of MCP-1 was observed in NPC tissues than that in normal nasopharyngeal tissues, and the G allele of rs1024611 was significantly associated with elevated mRNA expression level of MCP-1 in Epstein-Barr virus-transformed lymphocytes. In conclusion, our findings suggested that rs1024611 at the MCP-1 promoter may be a risk factor for NPC. Further studies with larger sample size are necessary to confirm these findings.
Impacts of Matrix Metalloproteinase-8 Genotypes, Smoking, Alcohol Drinking, and Helicobacter Pylori Infection on Gastric Cancer

Abstract:Background/aim: In gastric cancer (GCa) tissues, mRNA expression of matrix metalloproteinase-8 (MMP-8) is notably reduced compared to healthy tissues. Furthermore, abnormally low or elevated serum levels of MMP-8 have been linked to a significantly poor prognosis. The involvement of MMP-8 genotypes in susceptibility to GCa remains underexplored. We aimed to assess the influence of MMP-8 genotypes on GCa susceptibility and their potential interactions with smoking, alcohol consumption, and Helicobacter pylori (H. pylori) infection. Patients and methods: The study utilized polymerase chain reaction-based restriction fragment length polymorphism (PCR-RFLP) to analyze MMP-8 rs11225395, rs34009635, and rs35866072 genotypes in 161 GCa patients and 483 controls. Results: No statistically significant difference was detected in the distribution of genotypic (p for trend=0.3635) or allelic (p=0.1954) frequencies of MMP-8 rs11225395. Under a dominant model, combined CT+TT genotypes showed no association with GCa risk [odds ratio (OR)=0.77, 95% confidence interval (95%CI)=0.54-1.10, p=0.1852]. Similarly, no association was observed for MMP-8 rs34009635 or rs35866072. Importantly, individuals with the MMP-8 rs11225395 CC genotype demonstrated a significant increase in GCa risk when exposed to smoking (OR=4.04, 95%CI=2.28-7.16, p=0.0001), alcohol consumption (OR=2.83, 95%CI=1.64-4.89, p=0.0002), and H. pylori infection (OR=3.53, 95%CI=2.12-5.90, p=0.0001). Conclusion: The findings indicate that individuals carrying the MMP-8 rs11225395 CC genotype have increased susceptibility to GCa, especially when combined with risk factors, such as smoking, alcohol consumption, and H. pylori infection. These results suggest that MMP-8 genotype-based preventive strategies, including lifestyle alterations and targeted infection treatments, may be valuable in mitigating GCa development.
Variations in Matrix Metalloproteinase-1, -3, and -9 Genes and the Risk of Acute Coronary Syndrome and Coronary Artery Disease in the Chinese Han Population.

Abstract:Objectives Matrix metalloproteinases (MMPs) are a group of endopeptidases involved in the pathogenesis of atherosclerosis, and MMP gene polymorphisms may contribute toward the risk of coronary heart disease. Within this context, our aim was to examine whether MMP1, MMP3, and MMP9 gene polymorphisms are associated with susceptibility to acute coronary syndrome (ACS) or angiographic coronary artery disease (CAD).Methods The MMP1 -519 A/G, MMP3 -1171 5A/6A, and MMP9 -1562 C/T polymorphisms were evaluated in 1574 individuals. Genotypes of patients with ACS (n = 660) and angiographically defined CAD (n = 382) were compared with ACS-free (n = 914) and non-CAD controls (n = 466).Results The MMP3 5A allele occurred at a higher frequency in patients with ACS than in ACS-free individuals (P = 0.001). Logistic regression analysis showed that the 5A/5A genotype of MMP3 was associated with a significantly increased risk of ACS [ adjusted odds ratio (OR)= 2.297, 95% confidence interval (CI) = 1.105-4.775, P = 0.026, 5A/5A vs. 6A/6A]. The CT and TT variant genotypes of MMP9 were associated with the occurrence of CAD (adjusted OR = 1.425, 95% CI = 1.045-1.943, P = 0.025, CT + TT vs. CC). None of the MMP1 -519 A/G polymorphisms was associated with ACS or CAD. Because of linkage disequilibrium, MMP1 and MMP3 polymorphisms were combined on chromosome 11q22.3, and the 5A(-1171)-G(-519) haplotype had a genetic risk factor for ACS (OR = 1.505, 95% CI = 1.219-1.857, P = 0.00013), whereas the 6A(-1171)-G(-519) haplotype had a decreased risk of ACS (OR = 0.815, 95% CI = 0.677-0.981, P = 0.03).Conclusion Taken together, the present findings indicate that genetic variations in MMP3 and MMP9 genes may be useful genetic markers for determining susceptibility to CAD in the Chinese Han population. Combined Effects of Genetic Variants of the Pten, Akt1, Mdm2 and P53 Genes on the Risk of Nasopharyngeal Carcinoma

Abstract:Phosphatase and tensin homolog (PTEN), v-akt murine thymoma viral oncogene homolog 1 (AKT1), mouse double minute 2 (MDM2) and p53 play important roles in the development of cancer. We examined whether the single nucleotide polymorphisms (SNPs) in the PTEN, AKT1, MDM2 and p53 genes were related to the risk and severity of nasopharyngeal carcinoma (NPC) in the Chinese population. Seven SNPs [p53 rs1042522, PTEN rs11202592, AKT1 SNP1-5 (rs3803300, rs1130214, rs3730358, rs1130233 and rs2494732)] were genotyped in 593 NPC cases and 480 controls by PCR direct sequencing or PCR-RFLP analysis. Multivariate logistic regression analysis was used to calculate adjusted odds ratios (ORs) and 95% confidence intervals (CIs). None of the polymorphisms alone was associated with the risk or severity of NPC. However, haplotype analyses indicated that a two-SNP core haplotype (SNP4-5, AA) in AKT1 was associated with a significantly increased susceptibility to NPC risk (adjusted OR = 3.87, 95% CI = 1.96-7.65; P<0.001). Furthermore, there was a significantly increased risk of NPC associated with the combined risk genotypes (i.e., p53 rs1042522 Arg/Pro + Pro/Pro, MDM2 rs2279244 G/T + G/G, PTEN rs11202592 C/C, AKT1 rs1130233 A/A). Compared with the low-risk group (0-2 combined risk genotypes), the high-risk group (3-4 combined risk genotypes) was associated with a significantly increased susceptibility to NPC risk (adjusted OR = 1.67, 95% CI = 1.12-2.50; P = 0.012). Our results suggest that genetic variants in the PTEN, AKT1, MDM2 and p53 tumor suppressor-oncoprotein network may play roles in mediating the susceptibility to NPC in Chinese populations.

Abstract:Expression of matrix metalloproteinase-1 (MMP1), an interstitial collagenase regulating the extracellular matrix, plays a major role in carcinogenesis of gastric cancer, a leading cause of death worldwide. In literature, the single-nucleotide polymorphism (SNP) promoter -1607 1G/2G (rs1799750) at the MMP1 gene promoter has been reported to alter its own transcription level. While the importance's of the genotype of MMP1 promoter -1607 has not yet been studied in gastric cancer in Taiwan, our aim was to investigate MMP1 promoter -1607 genotypes and gastric cancer (GC) susceptibility in central Taiwan population. In the current hospital-based case-control study, the contribution of MMP1 promoter -1607 genotypes to GC risk was investigated among 121 GC patients and 363 gender- and age-matched healthy controls recruited and genotyped by the polymerase chain reaction-based restriction fragment length polymorphism (PCR-RFLP) methodology. We found that the genotypic and allelic frequencies were not differentially distributed between GC patient and control groups. The variant 1G containing genotypes have interactions with cigarrete smoking behaviors and Helicobacter pylori infection status, but not alcoholism on GC susceptibility determination. Our findings suggest that the variant 1G allele on MMP1 promoter -1607 may contribute to GC carcinogenesis and may be useful for GC early detection and prevention when combined with cigarrete smoking behaviors and Helicobacter pylori infection status.

Abstract:Purpose: Mouse double minute 2 (MDM2) is a key negative regulator of the p53 activity. Recently, a polymorphism in the MDM2 intronic promoter, SNP309, was shown to influence MDM2 expression and p53 activity. We examined whether the SNP309 was related to the risk of developing nasopharyngeal carcinoma (NPC) among Chinese populations.Experimental Design: We genotyped the SNP309 in two independent case-control populations in southern China, one is from Guangxi province (including 593 NPC patients and 480 controls) and the other is from Guangdong province (including 239 patients and 286 controls), by PCR direct sequencing. Multivariate logistic regression analysis was used to calculate adjusted odds ratio (OR) and 95% confidence interval (95% CI).Results: We observed that compared with the TT genotype, the genotypes containing G allele (GT + GG genotype) were associated with significant increased susceptibility to NPC in both Guangxi (OR, 1.43; 95% Cl, 1.04-1.91) and Guangdong population (OR, 1.53; 95% Cl, 1.00-2.36). When these two sample sets were combined, the OR of the GT + GG genotype developing NPC was 1.45 (95% Cl, 1.12-1.85) compared with the TT genotype. Furthermore, compared with the TT genotype, the GT + GG genotype was also significantly associated with the advanced lymph node metastasis (OR, 1.84; 95% Cl, 1.09-3.05).Conclusions: Our findings suggest that the MDM2 SNP309 may be a risk factor for the occurrence and advanced neck lymph node metastasis of NPC in Chinese population.

Abstract:Purpose: Matrix metalloproteinase 9 (MMP-9) plays critical roles in cancer development and aggression. Nonsynonymous single-nucleotide polymorphisms (SNP) in the functional domain of the MMP-9 gene may influence substrate and inhibitor binding and contribute to cancer predisposition and aggression. Patients and Methods: To test our hypothesis that common nonsynonymous SNPs, R279Q, P574R, and R668Q, in MMP-9 are associated with lung cancer development and metastasis, we conducted a case-control study of 744 patients with incident lung cancer and 747 cancer-free controls in Southeast China. Multivariate logistic regression analysis was used to calculate adjusted odds ratio (OR) and 95% confidence interval (95% CI). Results: We found that compared with the 279QQ genotype, the 279RR genotype was associated with significant elevated risk of lung cancer with metastasis (adjusted OR, 1.79; 95% CI, 1.03-3.08), whereas the 574PR heterozygote and 574PP homozygote had 1.46-fold (95% CI, 0.94-2.26) and 1.69-fold elevated risk (95% CI, 1.10-2.60), respectively, compared with the 574RR genotype. When we examined the combined effect of R279Q and P574R and used the 279R and 574P as the risk alleles, a significantly increased risk of lung cancer was associated with both the genotypes containing “1 to 2 risk alleles” (adjusted OR, 2.16; 95% CI, 1.30-3.59) and containing “>2 risk alleles” (adjusted OR, 2.44; 95% CI, 1.48-4.03), and it was more pronounced in 290 lung cancer cases with metastasis [adjusted OR, 2.30 (95% CI, 1.09-4.85) for the 1 to 2 risk alleles subgroup and adjusted OR, 2.82 (95% CI, 1.35-5.88) for the >2 risk alleles subgroup], compared with those without any risk alleles. However, no overall significant associations were observed between R668Q and lung cancer risk in this study population. Conclusion: These findings indicate that the potentially functional polymorphisms, MMP-9 P574R and R279Q, may confer the biomarker in the occurrence and metastasis of primary lung cancer. Further functional studies including these two genetic variants are warranted to confirm our findings.

Abstract:Matrix metalloproteinases; (MMPs) play an important role in several steps of cancer development. A single nucleotide polymorphism (-1306C-->T) in the MMP2 promoter sequence disrupts an Sp1 site and thus results in strikingly lower promoter activity. We examined the relationship between this polymorphism and risk for lung cancer in 781 cases and 852 age- and sex-matched controls in a Chinese population. We found that the allele frequency of MMP2-1306C was significantly higher among cases than among controls (0.91 versus 0.83). Subjects with the CC genotype had an overall 2-fold increased risk for developing lung cancer [adjusted odds ratio (OR) 2.18; 95% confidence interval (0), 1.70-2.79] compared with those with the CT or TT genotype. The elevated risk was observed evenly among different subtypes of this cancer. Stratified analysis indicated an additive interaction between the CC genotype and smoking on the elevated risk. The ORs of lung cancer for the CC genotype, smoking, and both factors combined were 2.38 (95% CI 1.64-3.45), 4.26 (95% CI 2.57-8.44), and 7.64 (95% CI 4.74-12.33), respectively. Furthermore, when the data were stratified by the pack-years smoked, this joint effect was more evident and stronger in heavy smokers (OR 10.25, 95% CI 5.80-18.09) than in light smokers (OR 5.55, 95% CI 3.34-9.22). These results demonstrate a significant association between the MMP2 -1306C/T polymorphism and risk of developing lung cancer solely or in a manner of interaction with carcinogen exposure.
Detection of nasopharyngeal carcinoma susceptibility with single nucleotide polymorphism analysis using next-generation sequencing technology

Abstract:Nasopharyngeal carcinoma (NPC) is a head and neck cancer with high incidence in South China and East Asia. To provide a theoretical basis for NPC risk screening and early prevention, we conducted a meta-analysis of relevant literature on the association of single nucleotide polymorphisms (SNP)s with NPC susceptibility. Further, expression of 15 candidate SNPs identified in the meta-analysis was evaluated in a cohort of NPC patients and healthy volunteers using next-generation sequencing technology. Among the 15 SNPs detected in the meta-analysis, miR-146a (rs2910164, C>G), HCG9 (rs3869062, A>G), HCG9 (rs16896923, T>C), MMP2 (rs243865, C>T), GABBR1 (rs2076483, T>C), and TP53 (rs1042522, C>G) were associated with decreased susceptibility to NPC, while GSTM1 (+/DEL), IL-10 (rs1800896, A>G), MDM2 (rs2279744, T>G), MDS1-EVI1 (rs6774494, G>A), XPC (rs2228000, C>T), HLA-F (rs3129055, T>C), SPLUNC1 (rs2752903, T>C; and rs750064, A>G), and GABBR1 (rs29232, G>A) were associated with increased susceptibility to NPC. In our case-control study, an association with increased risk for NPC was found for the AG vs AA genotype in HCG9 (rs3869062, A>G). In addition, heterozygous deletion of the GSTM1 allele was associated with increased susceptibility to NPC, while an SNP in GABBR1 (rs29232, G>A) was associated with decreased risk, and might thus have a protective role on NPC carcinogenesis. This work provides the first comprehensive assessment of SNP expression and its relationship to NPC risk. It suggests the need for well-designed, larger confirmatory studies to validate its findings.

Abstract:Background: Genetic variations in microRNAs may alter their processing, expression, and binding to target mRNAs, consequently affecting many cancer-related biological processes. Recently, a polymorphism rs11614913 in MIR196A2 was shown to affect the processing of the precursor microRNA into its mature forms and the repertoire of target mRNAs with which it interacts. We examined whether this polymorphism was relevant to the risk of occurrence or progression of nasopharyngeal carcinoma (NPC) in the Chinese population. Methods: We genotyped the MIR196A2 rs11614913 in a case-control study of 1084 patients with NPC and 1036 cancer-free controls using the TaqMan assay. The genetic associations with the risk of occurrence and progression of NPC were analyzed by logistic regression. Results: We observed a significantly increased occurrence of NPC associated with the rs11614913 T allele (odds ratio [OR]=1.15, 95% confidence interval [CI]=1.02-1.32, p=0.026) compared with the C allele. The T allele was also significantly associated with the advanced local tumor invasion (T-3+T-4 vs. T-1+T-2; OR=1.27, 95% CI=1.04-1.54, p=0.015) and advanced lymph node metastasis (N-2+N-3 vs. N-0+N-1; OR=1.23, 95% CI=1.02-1.49, p=0.031) of NPC compared with the C allele. Furthermore, stratified analysis indicated that the increased susceptibility to advanced lymph node metastasis of NPC related to the T allele was more pronounced in patients with a positive family history (N-2+N-3 vs. N-0+N-1; p=0.016, test for homogeneity). Conclusions: Our study suggests that the functional polymorphism rs11614913 in the MIR196A2 gene may contribute to the risk of occurrence and progression of NPC in the Chinese population.

Abstract:Background The tumor suppressor TP53 and its negative regulator MDM2 play crucial roles in carcinogenesis. Previous case-control studies also revealed TP53 72Arg>Pro and MDM2 309T>G polymorphisms contribute to the risk of common cancers. However, the relationship between these two functional polymorphisms and nasopharyngeal carcinoma (NPC) susceptibility has not been explored. Methods In this study, we performed a case-control study between 522 NPC patients and 722 healthy controls in a Chinese population by using PCR-RFLP. Results We found an increased NPC risk associated with the MDM2 GG (odds ratio [OR] = 2.83, 95% confidence interval [CI] = 2.08-3.96) and TG (OR = 1.49, 95% CI = 1.16-2.06) genotypes. An increased risk was also associated with the TP53 Pro/Pro genotype (OR = 2.22, 95% CI = 1.58-3.10) compared to the Arg/Arg genotype. The gene-gene interaction of MDM2 and TP53 polymorphisms increased adult NPC risk in a more than multiplicative manner (OR for the presence of both MDM2 GG and TP53 Pro/Pro genotypes = 7.75, 95% CI = 3.53-17.58). Conclusion The findings suggest that polymorphisms of MDM2 and TP53 genes may be genetic modifier for developing NPC.
