Abstract:In recent years, cardiac patches have been developed for the treatment of myocardial infarction. However, the fixation approaches onto the tissue through suture or phototriggered reaction inevitably cause new tissue damage. Herein, a paintable hydrogel is constructed based on Fe3+-triggered simultaneous polymerization of covalently linked pyrrole and dopamine in the hyperbranched chains where the in situ formed conductive polypyrrole also uniquely serves to crosslink network. This conductive and adhesive hydrogel can be conveniently painted as a patch onto the heart surface without adverse liquid leakage. The functional patch whose conductivity is equivalent to that of normal myocardium is strongly bonded to the beating heart within 4 weeks, accordingly efficiently boosting the transmission of electrophysiological signals. Eventually, the reconstruction of cardiac function and revascularization of the infarct myocardium are remarkably improved. The translatable suture-free strategy reported in this work is promising to address the human clinical challenges in cardiac tissue engineering.

Abstract:Engineering cardiac implants for treating myocardial infarction (MI) has advanced, but challenges persist in mimicking the structural properties and variability of cardiac tissues using traditional bioconstructs and conventional engineering methods. This study introduces a synthetic patch with a bioactive surface designed to swiftly restore functionality to the damaged myocardium. The patch combines a composite, soft, and conductive hydrogel‐based on (3,4‐ethylenedioxythiophene):polystyrene‐sulfonate (PEDOT:PSS) and polyvinyl alcohol (PVA). This cardiac patch exhibits a reasonably high electrical conductivity (40 S/cm) and a stretchability up to 50% of its original length. Our findings reveal its resilience to 10% cyclic stretching at 1 Hz with no loss of conductivity over time. To mediate a strong cell–scaffold adhesion, we biofunctionalize the hydrogel with a N‐cadherin mimic peptide, providing the cardiac patch with a bioactive surface. This modification promote increased adherence and proliferation of cardiac fibroblasts (CFbs) while effectively mitigating the formation of bacterial biofilm, particularly against Staphylococcus aureus, a common pathogen responsible for surgical site infections (SSIs). Our study demonstrates the successful development of a structurally validated cardiac patch possessing the desired mechanical, electrical, and biofunctional attributes for effective cardiac recovery. Consequently, this research holds significant promise in alleviating the burden imposed by myocardial infarctions.

Abstract:Conductive hydrogel is a potential therapeutic tool to treat damaged heart muscles in myocardial infarction (MI). However, it is still a quite challenge to optimize the fabrication of a therapeutic hydrogel patch that sustains favorable biocompatibility, electronic and mechanical stability under a complicated MI microenvironment. Herein, a tunable self-healing ionic hydrogel (POG1) was developed through the introduction of a biocompatible polyacrylic acid (PAA, FDA-approved) into the hydrogel matrix. The fabricated POG1 hydrogel possessed suitable stretchable (>500% strain) and compressive (>85% strain) properties, comparable modulus with mammalian heart (30∼500kPa, Young's modulus), self-healable, and highly stable conductivity during large deformations (∼50% compress strain, ∼150% tensile strain). Specifically, the established PAA nano-channels inside of POG1 endowed the hydrogel with microscopic ultra-homogeneous conductivity. Compared to those seeded in the electronic conductors-embedded (PPy, CNT, rGO) hydrogels, the cardiomyocytes (CMs) seeded in the POG1 hydrogel exhibited more significantly oriented sarcomeres. This POG1 engineered cardiac patch (ECP) also exerted robust benefits in attenuating left ventricular remodeling and restoring heart function after implantation in vivo. This paper highlighted a previously unexplored strategy for a biocompatible ionic conductive hydrogel ECP with an excellent MI repair function.

Abstract:In recent years, engineered cardiac patch (ECP) with electrical conductivity has been developed for cardiac repair after injury or infarction. However, the application of traditional conductive polymers in ECP is still limited due to their toxicity and difficulty in dispersing and degradation. Herein, a self-association crosslinked conductive zwitterionic hydrogel was developed by simple copolymerization of zwitterionic monomer carboxybetaine methacrylate (CBMA) and a biocompatible monomer hydroxyethyl methacrylate (HEMA). The fabricated p(CBMA-co-HEMA) hydrogel possessed excellent ionic conductivity, suitable compressive and stretchable properties, self-healing and mechanical sensory capability. In vitro, the zwitterionic hydrogel significantly enhanced the functionalization of the cardiomyocytes, resulting in excellent synchronous contraction by increasing the expression of cardiac-related factor α-actinin and transmission electrical signal-relevant Connexin-43 (CX-43). In vivo, the hydrogel-derived functional patch effectively promoted the transmission of electrophysiological signals, improved revascularization in infarcted area and then remarkably restored the cardiac function. These data indicated that our developed conductive zwitterionic hydrogel has good promise in repairing myocardial tissue injury.

Abstract:Conductive cardiac patches have been proven to promote angiogenesis in infarcted myocardium; however, their conductive integrity, elasticity, and vascularization potential have not yet been optimized. The prevascularization of conductive elastic cardiac patches could be an effective strategy for building a substantial connection between the patch and the infarcted heart. Here, a coronary artery casting is introduced into a holey graphene oxide/polypyrrole-incorporated polyhydroxyethyl methacrylate prefabricated gel to form a vascularized conductive elastic patch. The engineered patches are able to rebuild functional vascular anastomoses and provide strong electrical integration with infarcted hearts, resulting in effective myocardial infarction repair in vivo. RNA sequencing analyses further reveal that the conductive elastic patches under dynamic culture conditions upregulated cardiac muscle contraction- and ATP biosynthesis-related mRNA expression in vitro. Together, this study demonstrates that the fabricated patches have versatile conductivity, elasticity, and vascularization properties, and could therefore be a promising candidate for heart repair.

Abstract:A biomaterial system incorporating nanocellulose, poly(glycerol sebacate), and polypyrrole is introduced for the treatment of myocardial infarction. Direct ink writing of the multicomponent aqueous suspensions allows multifunctional lattice structures that not only feature elasticity and electrical conductivity but enable cell growth. They are proposed as cardiac patches given their biocompatibility with H9c2 cardiomyoblasts, which attach extensively at the microstructural level, and induce their proliferation for 28 days. Two model drugs (3i‐1000 and curcumin) are investigated for their integration in the patches, either by loading in the precursor suspension used for extrusion or by direct impregnation of the as‐obtained, dry lattice. In studies of drug release conducted for five months, a slow in vitro degradation of the cardiac patches is observed, which prevents drug burst release and indicates their suitability for long‐term therapy. The combination of biocompatibility, biodegradability, mechanical strength, flexibility, and electrical conductivity fulfills the requirement of the highly dynamic and functional electroresponsive cardiac tissue. Overall, the proposed cardiac patches are viable alternatives for the regeneration of myocardium after infarction through the effective integration of cardiac cells with the biomaterial.

Abstract:Myocardial infarction (MI) constitutes the first cause of morbidity and mortality in our life, so using highly conductive and elastic materials to produce an engineered cardiac patch is an effective way to improve the myocardium infarction area function. Here, shape memory polymers of the polyurethane/polyaniline/silicon oxide (PU/PANI/SiO2) electrospinning sub-micron fiber patch were precisely produced in the case of the hydrogen bonding effect and interaction between the carboxyl groups to provide compatibility, phase mixing/miscibility, and stability. The sub-micron fiber patch prepared by our group has some remarkable characteristics, such as sub-micron fibers, 3D porous structure, special thickness to simulate the extracellular matrix (ECM), elastic deformation, good properties in conducting weak electrical signals, stability to maintain the whole structure, and self-adhesion. This sub-micron fiber material has been proven to be effective, easy, and reliable. Through precise design of the material system, structure regulation, and performance optimization, the aim is to produce a sub-micron fiber cardiac patch to simulate the myocardium ECM and improve conductive signal transduction for potential MI therapy.This article has not yet been cited by other publications.

Abstract:Fabricating concurrently stretchable and conductive hydrogel scaffold is highly desired in the development of myocardial tissue engineering, and also very challenging especially with the anisotropic conductive properties. Existing anisotropic-strategies are limited to specific nanomaterials, or time-consuming. Here, we present a simple and new stretch-induced strategy to make conductive nanotubes aligned in the hydrogel. With this strategy, we fabricated a new type of hybrid hydrogel that possesses excellent stretchability while enabling outstanding anisotropic conductivity (6 vs 30 S/cm). Hybrid hydrogels are also successfully applied as a therapeutic cardiac patch which induces an elongated cell morphology of cultured cardiomyocytes and enables a stable directional transmission of electric signals. This patch gave successful mitigation of myocardium infarction in rats. It is believed that this strategy, and this nanocomposite hydrogel have a promising application in tissue engineering.

Abstract:A bioresorbable, highly conductive, and elastic cardiac patch (BCEP) designed as a heterogeneous material structure for myocardial infarction treatment. Finite element analysis defines geometries of filamentary serpentine mesh architectures in a bioresorbable metal for optimized elastic properties when integrated with a bioresorbable elastomer. Human‐induced pluripotent‐stem‐cell‐derived cardiomyocytes grown on the BCEP demonstrate regular and synchronous contractions. Ex vivo studies highlight the ability of the BECP to form electroconductive pathways for transmitting the bioelectrical signals. Myocardial infarction (MI) is one of the leading causes of death and disability. Recently developed cardiac patches provide mechanical support and additional conductive paths to promote electrical signal propagation in the MI area to synchronize cardiac excitation and contraction. Cardiac patches based on conductive polymers offer attractive features; however, the modest levels of elasticity and high impedance interfaces limit their mechanical and electrical performance. These structures also operate as permanent implants, even in cases where their utility is limited to the healing period of tissue damaged by the MI. The work presented here introduces a highly conductive cardiac patch that combines bioresorbable metals and polymers together in a hybrid material structure configured in a thin serpentine geometry that yields elastic mechanical properties. Finite element analysis guides optimized choices of layouts in these systems. Regular and synchronous contraction of human induced pluripotent stem cell‐derived cardiomyocytes on the cardiac patch and ex vivo studies offer insights into the essential properties and the bio‐interface. These results provide additional options in the design of cardiac patches to treat MI and other cardiac disorders.

Abstract:The biomimetic construction of a microstructural-mechanical-electrical anisotropic microenvironment adaptive to the native cardiac tissue is essential to repair myocardial infarction (MI). Inspired by the 3D anisotropic characteristic of the natural fish swim bladder (FSB), a novel flexible, anisotropic, and conductive hydrogel was developed for tissue-specific adaptation to the anisotropic structural, conductive, and mechanical features of the native cardiac extracellular matrix. The results revealed that the originally stiff, homogeneous FSB film was tailored to a highly flexible anisotropic hydrogel, enabling its potential as a functional engineered cardiac patch (ECP). In vitro and in vivo experiments demonstrated the enhanced electrophysiological activity, maturation, elongation, and orientation of cardiomyocytes (CMs), and marked MI repair performance with reduced CM apoptosis and myocardial fibrosis, thereby promoting cell retention, myogenesis, and vascularization, as well as improving electrical integration. Our findings offer a potential strategy for functional ECP and provides a novel strategy to bionically simulate the complex cardiac repair environment.

Flexible Conductive Decellularized Fish Skin Matrix as a Functional Scaffold for Myocardial Infarction Repair

Abstract:Engineering cardiac patches are proven to be effective in myocardial infarction (MI) repair, but it is still a tricky problem in tissue engineering to construct a scaffold with good biocompatibility, suitable mechanical properties, and solid structure. Herein, decellularized fish skin matrix is utilized with good biocompatibility to prepare a flexible conductive cardiac patch through polymerization of polydopamine (PDA) and polypyrrole (PPy). Compared with single modification, the double modification strategy facilitated the efficiency of pyrrole polymerization, so that the patch conductivity is improved. According to the results of experiments in vivo and in vitro, the scaffold can promote the maturation and functionalization of cardiomyocytes (CMs). It can also reduce the inflammatory response, increase local microcirculation, and reconstruct the conductive microenvironment in infarcted myocardia, thus improving the cardiac function of MI rats. In addition, the excellent flexibility of the scaffold, which enables it to be implanted in vivo through "folding-delivering-re-stretehing" pathway, provides the possibility of microoperation under endoscope, which avoids the secondary damage to myocardium by traditional thoracotomy for implantation surgery.

Abstract:Myocardial infarction is one of the more common cardiovascular diseases, and remains the leading cause of death, globally. Hydrogels (namely, those using natural polymers) provide a reliable tool for regenerative medicine and have become a promising option for cardiac tissue regeneration due to their hydrophilic character and their structural similarity to the extracellular matrix. Herein, a functional ink based on the natural polysaccharides Gellan gum and Konjac glucomannan has, for the first time, been applied in the production of a 3D printed hydrogel with therapeutic potential, with the goal of being locally implanted in the infarcted area of the heart. Overall, results revealed the excellent printability of the bioink for the development of a stable, porous, biocompatible, and bioactive 3D hydrogel, combining the specific advantages of Gellan gum and Konjac glucomannan with proper mechanical properties, which supports the simplification of the implantation process. In addition, the structure have positive effects on endothelial cells' proliferation and migration that can promote the repair of injured cardiac tissue. The results presented will pave the way for simple, low-cost, and efficient cardiac tissue regeneration using a 3D printed hydrogel cardiac patch with potential for clinical application for myocardial infarction treatment in the near future.

Abstract:Myocardial infarction (MI) is a major cause of death worldwide. After the occurrence of MI, the heart frequently undergoes serious pathological remodeling, leading to excessive dilation, electrical disconnection between cardiac cells, and fatal functional damage. Hence, extensive efforts have been made to suppress pathological remodeling and promote the repair of the infarcted heart. In this study, we developed a hydrogel cardiac patch that can provide mechanical support, electrical conduction, and tissue adhesiveness to aid in the recovery of an infarcted heart function. Specifically, we developed a conductive and adhesive hydrogel (CAH) by combining the two-dimensional titanium carbide (Ti3C2Tx) MXene with natural biocompatible polymers [i.e., gelatin and dextran aldehyde (dex-ald)]. The CAH was formed within 250 s of mixing the precursor solution and could be painted. The hydrogel containing 3.0 mg/mL MXene, 10% gelatin, and 5% dex-ald exhibited appropriate material characteristics for cardiac patch applications, including a uniform distribution of MXene, a high electrical conductivity (18.3 mS/cm), cardiac tissue-like elasticity (30.4 kPa), strong tissue adhesion (6.8 kPa), and resistance to various mechanical deformations. The CAH was cytocompatible and induced cardiomyocyte (CM) maturation in vitro, as indicated by the upregulation of connexin 43 expression and a faster beating rate. Furthermore, CAH could be painted onto the heart tissue and remained stably adhered to the beating epicardium. In vivo animal studies revealed that CAH cardiac patch treatment significantly improved cardiac function and alleviated the pathological remodeling of an infarcted heart. Thus, we believe that our MXene-based CAH can potentially serve as a promising platform for the effective repair of various electroactive tissues including the heart, muscle, and nerve tissues.

Abstract:Engineered hydrogel patches have shown promising therapeutic effects in the treatment of myocardial infarction (MI), especially anisotropic patches that mimic the characteristics of native myocardium have attracted widespread attention. However, it remains a great challenge to develop cardiac patches with long-range and orderly electrical conduction based on an effective, mild, and rapid strategy. Here, a multifunctional anisotropic cardiac patch is presented based on microfluidic manipulation. The anisotropic alginate-gelatin methacrylate hydrogel patches are easily and rapidly prepared through microfluidic focusing, ion-photocrosslinking, and parallel packing processes. The fluid-based anisotropic realization process does not involve complex machining and strong field stimulation and is compatible with the loading of macromolecular biological agents. The anisotropic hydrogel patch can mimic the anisotropy of the myocardium and guide the directional polarization of cardiomyocytes. In animal model experiments, it also exhibits significant effects in inhibiting ventricular remodeling, fibrosis, and enhancing cardiac function recovery after MI. These comprehensive features make the multifunctional hydrogel patch a promising candidate for cardiac tissue repair and future provide a new paradigm for expanding microfluidic technology to solve tissue engineering challenges.

Abstract:Abstract Preparing a micropatterned elastomer film with characteristics that can simulate the mechanical properties, anisotropy, and electroactivity of natural myocardial tissues is crucial in cardiac tissue engineering after myocardial infarction (MI). Therefore, in this study, we developed several elastomeric films with a surface micropattern based on poly (glycerol sebacate) (PGS) and graphene (Gr). These films have sufficient mechanical strength (0.6 ± 0.1–3.2 ± 0.08 MPa) to withstand heartbeats, and the micropatterned structure also satisfies the natural myocardium anisotropy in the transverse and vertical. Moreover, Gr makes these films conductive (up to 5.80 × 10 −7 S m −1 ), which is necessary for the conduction of electrical signals between cardiomyocytes and the cardiac tissue. Furthermore, they have good cytocompatibility and can promote cell proliferation in H9c2 rat cardiomyocyte cell lines. In vivo test results indicate that these films have good biocompatibility. Notably, a film with 1 wt% Gr content (PGS–Gr1) significantly affects the recovery of myocardial function in rats after MI. This film effectively decreased the infarct size and degree of myocardial fibrosis and reduced collagen deposition. Echocardiographic evaluation showed that after treatment with this film, the left ventricular internal dimension (LVID) in systole and LVID in diastole of rats exhibited a significant downward trend, whereas the fractional shortening and ejection fraction were significantly increased compared with the control group. Abstract:Heart failure caused by acute myocardial infarction still remains the main cause of death worldwide. Development of conductive hydrogels provided a promising approach for the treatment of myocardial infarction (MI). However, the therapeutic potential of these hydrogels is still limited by material toxicity or low conductivity. The latter directly affects the coupling and the propagation of electrical signals between cells. Here, we prepared a functional conductive hydrogel by combining hydrophilic and biocompatible poly(vinyl alcohol) (PVA) with conductive melanin nanoparticles under physical crosslinking conditions. The composite hydrogels prepared by a facile fabrication process of five freeze/thaw cycles possessed satisfying mechanical properties and conductivity close to those of natural heart. The physical properties and biocompatibility were evaluated in vitro experiments, showing that the introduction of melanin particles successfully improved the elasticity, conductivity and cell adhesion of PVA hydrogel. In vivo, the composite hydrogels could enhance cardiac repair effect by reducing myocardial infarction area, slowing down ventricular wall thinning, and promoting the vascularization of infarct area in MI rat model. We believe that the PVA/melanin composite hydrogel might be a suitable candidate material for MI repair. This article is protected by copyright. All rights reserved.

A Conductive Bioengineered Cardiac Patch for Myocardial Infarction Treatment by Improving Tissue Electrical Integrity

Abstract:Conductive scaffolds are of great value for constructing functional myocardial tissues and promoting tissue reconstruction in the treatment of myocardial infarction (MI). Here, a novel scaffold composed of silk fibroin and polypyrrole (SP50) with a typical sponge‐like porous structure and electrical conductivity similar to the native myocardium is developed. An electroactive engineered cardiac patch (SP50 ECP) with a certain thickness is constructed by applying electrical stimulation (ES) to the cardiomyocytes (CMs) on the scaffold. SP50 ECP can significantly express cardiac marker protein (α‐actinin, Cx‐43, and cTnT) and has better contractility and electrical coupling performance. Following in vivo transplantation, SP50 ECP shows a notable therapeutic effect in repairing infarcted myocardium. Not only can SP50 ECP effectively improve left ventricular remodeling and restore cardiac functions, such as ejection function (EF), but more importantly, improve the propagation of electrical pulses and promote the synchronous contraction of cardiomyocytes in the scar area with normal myocardium, effectively reducing the susceptibility of MI rats to arrhythmias. In conclusion, this study demonstrates a facile approach to constructing electroactive ECPs based on porous conductive scaffolds and proves the therapeutic effects of ECPs in repairing the infarcted heart, which may represent a promising strategy for MI treatment. Novel Biomimetic Sandwich-Structured Electrospun Cardiac Patches with Moderate Adhesiveness and Excellent Electrical Conductivity

Abstract:Clinical cardiac patches exhibit unsatisfied biocompatibility, low adhesion, and inadequate compliance and suboptimal mechanical properties for cardiac disorders repair. To address these challenges, herein we have innovatively proposed a biomimetic nanofiber electrospun membrane with a sandwich structure strategy. The composite patch comprises a stretchable polyurethane (PU) as basic material, then infiltrated with biocompatible silk fibroin methacryloyl (Silk-MA) as the middle layer via electrospinning and finally covered with Bio-ILs (chemically modified biocompatible ionic liquids) to impart electrical conductivity. Results indicated that the incorporation of Bio-ILs significantly enhances the conductivity reaching 2877 mS/m; particularly due to the positive charges of Bio-ILs, the composite film exhibits mild adhesive properties, inducing minimal damage to the substrate tissue. Furthermore, the basic PU of bilayer nanofiber membrane increased the film's stretching strain to approximately 250%, the Silk-MA hydrogel coating changed the film from hydrophobic to hydrophilic, creating a favorable and biocompatible microenvironment. Finally, in vitro experiments on cardiomyocytes confirmed that the material exhibits low cytotoxicity and excellent biocompatibility. Overall, the biomimetic sandwich electrospun membrane could restore electrical conduction and synchronized contraction function, providing a promising strategy for the treatment of cardiac tissue engineering.
3D Printed Multifunctional Bioadhesive Patch with Intrinsic Bioelectronic Properties for Decoding Electromechanical and Anisotropic Cardiac Microenvironment

Abstract:Fabricating anisotropic multifunctional bioadhesive patches with tunable mechanical stiffness, electrical conductivity, antimicrobial activity, and modulating cellular behavior is crucial for the successful management of cardiac tissue injury and boosting immunogenic microenvironments. Direct ink writing (DIW)-based 3D printing holds tremendous potential for developing electroactive cardiac patches (ECPs) with anisotropic microarchitecture. Inspired by the native myocardium, we developed a multifunctional and anisotropic ECP with tunable stiffness by incorporating a highly conductive graphene oxide/nanodiamond (GO@ND) complex into a biocompatible carboxymethyl chitosan/polyvinyl alcohol (CSA) matrix for regulating the immunogenic and cardiomyogenic cues. The incorporation of GO@ND enhanced the electrical conductivity (~22.6 S mm-1) with high interfacial toughness (>250 MJ m-1) and improved the printability (n = 0.5) with concentration-dependent self-assembly into the CSA matrix. We observed that electrical stimulation (EFs; 250 mV/20 min/day) through nanoengineered CSA resulted in broad-spectrum antibacterial activity against E. coli and S. aureus by 99.29% and 98.74% via sustained release of curcumin (Cur). Moreover, the electromechanical study revealed that CSA with higher stiffness (~6.2 kPa) activated cytoplasmic YAPs during macrophage polarization. Besides, the stiffness and EFs regulated the human cardiomyocyte differentiation through anisotropic force-driven early activation of Vinculin, triggering the phosphorylation of NFATc3 and activated Lamin A/C in a YAP-dependent manner. Based on these findings, we anticipated that the fabricated nanoengineered patch had tremendous potential for regulating the electro-cardiomyogenic microenvironment with multifunctional abilities.

Abstract:Reactions of [Co(N(SiMe3)2)2thf] with 2.1 equiv. of MesSH (Mes = C6H2-2,4,6-(CH3)3) yield dark brown crystals of the one dimensional chain compound [Co(SMes)2]. In contrast reactions of [Co(N(SiMe3)2)2thf] with 2.1 equiv. of PhSH result in the formation of a dark brown almost X-ray amorphous powder of 'Co(SPh)2'. Addition of aliquots of CH3OH to the latter reaction resulted in the almost quantitative formation of crystalline ammonia thiolato complexes either [Co(SPh)2(NH3)2] or [Co(SPh)2NH3]. Single crystal XRD reveals that [Co(SPh)2NH3] forms one-dimensional chains in the crystal via μ2-SPh bridges whereas [Co(SPh)2(NH3)2] consists at a first glance of isolated distorted tetrahedral units. Magnetic measurements suggest strong antiferromagnetic coupling for the two chain compounds [Co(SMes)2] (J = -38.6 cm-1) and [Co(SPh)2NH3] (J = -27.1 cm-1). Interestingly, also the temperature dependence of the susceptibility of tetrahedral [Co(SPh)2(NH3)2] shows an antiferromagnetic transition at around 6 K. UV-Vis-NIR spectra display d-d bands in the NIR region between 500 and 2250 nm. Thermal gravimetric analysis of [Co(SPh)2(NH3)2] and [Co(SPh)2NH3] reveals two well separated cleavage processes for NH3 and SPh2 upon heating accompanied by the stepwise formation of 'Co(SPh)2' and cobalt sulfide.
