Abstract:Photothermal release of cargo molecules has been extensively studied for bioapplications. For instance, microcapsules decorated with plasmonic nanoparticles have been widely used in in vitro assays. However, some concerns about their suitability for some in vivo applications cannot be easily overcome, in particular the limited penetration depth of light (even infrared). Magnetic nanoparticles are alternative heat-mediators for local heating, which can be triggered by applying an alternating magnetic field (AMF). AMFs are much less absorbed by tissue than light and thus can penetrate deeper overcoming the above mentioned limitations. Here we present iron oxide nanocube-modified microcapsules as a platform for magnetically triggered molecular release. Layer-by-layer assembled polyelectrolyte microcapsules with 4.6 μm diameter, which had 18 nm diameter iron oxide nanocubes integrated in their walls, were synthesized. The microcapsules were further loaded with an organic fluorescent polymer (Cascade Blue-labelled dextran), which was used as a model of molecular cargo. Through an AMF the magnetic nanoparticles were able to heat their surroundings and destroy the microcapsule walls, leading to a final release of the embedded cargo to the surrounding solution. The cargo release was monitored in solution by measuring the increase in both absorbance and fluorescence signal after the exposure to an AMF. Our results demonstrate that magnetothermal release of the encapsulated material is possible using magnetic nanoparticles with a high heating performance.

Abstract:All‐dielectric nanophotonics becomes a versatile tool for various optical applications, including nanothermometry and optical heating. Its general concept includes excitation of Mie resonances in nonplasmonic nanoparticles. However, the potential of resonant dielectric nanoparticles in drug delivery applications still has not been fully realized. Here, optically resonant dielectric iron oxide nanoparticles (α‐Fe2O3 NPs) are employed for remote rupture of microcontainers used as a drug delivery platform. It is theoretically and experimentally demonstrated that α‐Fe2O3 NPs have several advantages in light‐to‐heat energy conversion comparing to previously used materials, such as noble metals and silicon, due to the broader spectral range of efficient optical heating, and in enhancement of thermally sensitive Raman signal. The α‐Fe2O3 NPs embedded into the wall of universal drug carriers, polymer capsules, are used to experimentally determine the local temperature of the capsule rupture upon laser irradiation (170 °C). As a proof of principle, the delivery and remote release of anticancer drug vincristine upon lowered laser irradiation (4.0× 104 W cm−2) using polymer capsules modified with the α‐Fe2O3 NPs is shown. The biological tests are performed on two primary cell types: i) carcinoma cells, as an example of malignant tumor, and ii) human stem cells, as a model of healthy cells.

Abstract:Biomolecule-mediated assembly of novel nanoconjugates has been subjected to numerous investigations nowadays, which provides a new insight into the material science and engineering. Via the molecular biology technology, the genetically engineered polypeptide for inorganics (GEPI) can be designed as a molecular binder into the bio-scaffold to assemble hybrid functional nanoarchitectures. In the present work, we constructed a multi-functional virus-like particle (VLP) scaffold based on the Tobacco mosaic virus (TMV) (wild-type strain U1) in a facile way. The S123C site-mutated coat protein (CP) of TMV recombinated with a Ti–GEPI and 6-histidine tag (His tag) was successfully cloned, expressed and purified. The as-produced r-CP products retained both binding affinity for inorganic nanoparticles and self-assembly capability. Analysis of r-CP self-assembly during condensation polymerization was conducted. The r-CP-assembled aggregates including disc-like structures and rod-like VLPs were recovered by preparative size-exclusion chromatography (SEC) and then examined by TEM analysis. Notably, r-CP protein displayed a thermo-triggered reversibly switchable transition between bistable states of transparency and opaqueness. Western blot analysis, Immunogold labelling and TEM analysis were employed to test the existence and binding function of the His tag group in r-CP VLPs. Furthermore, dispersion of TiO 2 NPs in r-CP solution and recyclable application in bio-benefication was introduced. Here we demonstrated the possibilities of combining peptide-mediated immobilization with VLP-based biotemplate bearing multi-binding moieties for prospectful functional applications.
Genetically engineered magnetic nanocages for cancer magneto-catalytic theranostics

Abstract:The clinical applications of magnetic hyperthermia therapy (MHT) have been largely hindered by the poor magnetic-to-thermal conversion efficiency of MHT agents. Herein, we develop a facile and efficient strategy for engineering encapsulin-produced magnetic iron oxide nanocomposites (eMIONs) via a green biomineralization procedure. We demonstrate that eMIONs have excellent magnetic saturation and remnant magnetization properties, featuring superior magnetic-to-thermal conversion efficiency with an ultrahigh specific absorption rate of 2390 W/g to overcome the critical issues of MHT. We also show that eMIONs act as a nanozyme and have enhanced catalase-like activity in the presence of an alternative magnetic field, leading to tumor angiogenesis inhibition with a corresponding sharp decrease in the expression of HIF-1α. The inherent excellent magnetic-heat capability, coupled with catalysis-triggered tumor suppression, allows eMIONs to provide an MRI-guided magneto-catalytic combination therapy, which may open up a new avenue for bench-to-bed translational research of MHT.

Abstract:This article describes the fabrication of nanosized magneto-vesides (MVs) comprising tunable layers of densely packed superparamagnetic iron oxide nanopartides (SPIONs) in membranes via cooperative assembly of polymer tethered SPIONs and free poly(styrene)-b-poly(acrylic acid) (PS-b-PAA). The membrane thickness of MVs could be well controlled from 9.8 to 93.2 nm by varying the weight ratio of PS-b-PAA to SPIONs. The increase in membrane thickness was accompanied by the transition from monolayer MVs, to double layered MVs and to multilayered MVs (MuMVs). This can be attributed to the variation in the hydrophobic/hydrophilic balance of polymer-grafted SPIONs upon the insertion and binding of PS-b-PAA onto the surface of nanopartides. Therapeutic agents can be efficiently encapsulated in the hollow cavity of MVs and the release of payload can be tuned by varying the membrane thickness of nanovesicles. Due to the high packing density of SPIONs, the MuMVs showed the highest magnetization and transverse relaxivity rate (r2) in magnetic resonance imaging (MRI) among these MVs and individual SPIONs. Upon intravenous injection, doxorubicin-loaded MuMVs conjugated with RGD peptides could be effectively enriched at tumor sites due to synergetic effect of magnetic and active targeting. As a result, they exhibited drastically enhanced signal in MRI, improved tumor delivery efficiency of drugs as well as enhanced antitumor efficacy, compared with groups with only magnetic or active targeting strategy. The unique nanoplatform may find applications in effective disease control by delivering imaging and therapy to organs/tissues that are not readily accessible by conventional delivery vehicles.

Abstract:The advancement of synthetic cells as drug delivery devices hinges on the development of targeting strategies, in particular the controlled synthesis of biomolecules in-situ using a deeply penetrative stimulus. To address this, we have designed spherical nucleic acids comprising DNA promoter sequences decorating magnetic nanoparticle cores. By harnessing the heat dissipated from magnetic hyperthermia (a clinically-approved anticancer therapy) we tightly controlled cell-free protein synthesis. We then deployed a tissue phantom that is impenetrable by current activation methods to demonstrate the potential of this technology for the remote control of synthetic cells using deeply tissue-penetrating magnetic fields. This paves the way for targeting and controlling the in-situ synthesis of biomolecules deep within the body.

Abstract:Catalytic asymmetric synthesis of unnatural amino acids with vicinal tetrasubstituted chiral carbon stereocenters is described. In the first part, direct catalytic asymmetric aldol reaction of simple non-activated ketone electrophiles with α-substituted α-isothiocyanato ester donors was realized. A Mg/Schiff base catalyst promoted the aldol reaction, and α-amino-β-hydroxy esters were obtained in up to 98% ee and 98:2 d.r. In the second part, the Mg/Schiff base catalyst and a Sr/Schiff base catalyst were utilized for stereodivergent direct asymmetric Mannich-type reaction of ketimines. The Mg/Schiff base catalyst gave syn-α,β-diamino esters, while the Sr/Schiff base catalyst produced anti-α,β-diamino esters in good to high enantioselectivity, up to 97% ee.

Abstract:Yolk-shell magnetic polymer nanostructures with large cavity, movable magnetic core are attractive carriers for bulk storage, release and transport of guest molecules in various applications. However, it remains great challenge to achieve a controllable transport of guest species into and out from the hollow space in a controlled way due to the lack of tailor-made nanostructures. Herein, a dual-template-induced emulsion interfacial assembly and polymerization method (denoted as de-IAP) was developed to construct yolk-shell magnetic mesoporous polydopamine (YS-MMP) vesicles with magnetic core, large hollow space and uniform mesopores in the soft polymer shell. The obtained YS-MMP vesicles possess tunable cavity size (10-900 nm), flexible mesoporous PDA layer, large pores (21 nm), high surface area (217.4 m(2.)g(-1)) and high magnetization saturation (31.0 emu(.)g(-1)), thus exhibiting the merits of high loading capacity (29.1%) and encapsulation rates (41.0%) of Curcumin (Cur), and fast magnetic separation speed. Thanks to their superior photothermal effect and magnetic property, the obtained YS-MMP@Cur vesicles exhibit smart on-demand release of Cur and excellent recyclability. With 808 nm laser irradiation, YS-MMP@Cur vesicles show high-efficient photothermal bactericidal effect (>99%) against S. aureus and E. coli, opening up a new avenue for smart cargo delivery, lasting sterilization, and environmental remediation.

Abstract:In this work, we present two types of microfluidic chips involving magnetic nanoparticles dispersed in cyclohexane with oleic acid. In the first case, the hydrophobically coated nanoparticles are self-assembled with an amphiphilic diblock copolymer by a double-emulsion process in order to prepare giant magnetic vesicles (polymersomes) in one step and at a high throughput. It was shown in literature that such diblock copolymer W/O/W emulsion droplets can evolve into polymersomes made of a thin (nanometric) magnetic membrane through a dewetting transition of the oil phase from the aqueous internal cores usually leading to “acorn-like” structures (polymer excess) sticking to the membranes. To address this issue and greatly speed up the process, the solvent removal by evaporation was replaced by a “shearing-off” of the vesicles in a simple poly(dimethylsiloxane) chip designed to exert a balance between a magnetic gradient and viscous shear. In the second example, a simple oil-in-oil emulsion chip is used to obtain regular trains of magnetic droplets that circulate inside an inductor coil producing a radio-frequency magnetic field. We evidence that the heat produced by magnetic hyperthermia can be converted into a temperature rise even at the scale of nL droplets. The results are compared to heat transfer models in two limiting cases: adiabatic vs. dissipative. The aim is to decipher the delicate puzzle about the minimum size required for a tumor “phantom” to be heated by radio-frequency hyperthermia in a general scope of anticancer therapy.

Abstract:Systems for "protein transduction," intracellular delivery of functional proteins, are needed to address deliverability challenges of protein therapeutics. However, in vivo protein transduction remains challenging because of instability in serum, extracellular protease digestion and rapid excretion from the bloodstream. Here, a magnetically guided in vivo protein transduction using magnetic nanogel chaperone (MC) composed of iron oxide nanoparticles and a polysaccharide nanogel, a protein carrier inspired by "catch and release" mechanisms of molecular chaperones is demonstrated. The MC system enables efficient delivery of anti‐cancer proteins, saporin and RNaseA, into cultured tumor lines and inhibits cell proliferation, mainly via apoptosis. Magnetic in vivo protein transduction via intravenous whole body administration is demonstrated in a fibrosarcoma model. By in vivo optical imaging, MC accumulated in tumor tissues under magnetic field three times more than without irradiation. With subcutaneous injection, saporin is delivered by MC to the cytoplasm in magnetically targeted tissues. In an oral cancer model, MC‐delivered magnetically targeted saporin decreased tumor volume without significant body weight changes and no regrowth of tumor at 3 months after complete regression. Protein transduction with MC shows promise for cancer therapeutics and, potentially, for regenerative medicine and other biomedical applications.

Abstract:Simultaneous delivery of therapeutic agents and energy by magnetic nanoparticles (MNPs) at targeted sites can boost cancer therapy and alleviate side effects. To achieve this goal, however, the magnetic fluid hyperthermia (MFH) usually exhibits the unsufficient thermal efficiency due to their narrow magnetization curves. Besides, an inappropriately large administration concentration also causes health deterioration as shown in an animal model. In this study, the core–shell cube that enhances the coercivity and magnetization related to single‐compositional MNPs by elaborately tuning their interface relaxation via the magnetocrystalline and surface anisotropy is developed. Néel and Brownian relaxation can be adjusted by the particles’ structures to maximize the hyperthermia efficacy upon an alternating‐magnetic‐field (AMF). Furthermore, temozolomide and lactoferrin‐coated CoFe2O4@Fe3O4 core–shell cubes are rapidly internalized by targeting cancer cells and penetrate into tumor spheroids while subjecting to AMF. The targeted cubes with the capabilities of enhanced coercivity, AMF‐induced drug penetration into tumors, and magnetothermal ablation for cancer therapy display potentials for clinical uses. The shape effects of core/shell on hard/soft magnetic materials reflected to their Brownian relaxation time (τB) and Néel relaxation time (τN), facilitating the controls of surface anisotropy (Ks), hyperthermia effects. Via conjugation of temozolomide (TMZ) and lactoferrin (Lf), the magnetic core–shell heterostructures facilitate the penetrative delivery to tumor spheroids, chemotherapy treatment efficacy, and intracellular mechanical destruction on a microfluidic chip.

Abstract:Nowadays there is a growing interest in bioscaffolded nanoarchitectures. Rapid progress in nanobiotechnology and molecular biology has allowed the engineering of inorganic-binding peptides termed as genetically engineered polypeptides for inorganics (GEPIs) into self-assembling biological structures to facilitate the design of novel biomedical or bioimaging devices. Here we introduce a novel nanocomposite comprising a self-assembled protein scaffold based on a recombinant tobacco mild green mosaic tobamovirus (TMGMV) coat protein (CP) and the photocatalytic TiO2 nanoparticles attached to it, which may provide a generic method for materials engineering. A template containing a modified TMGMV CP (mCP) gene, with the first six C-terminal amino acid residues deleted to accommodate more foreign peptides and expressing a site-directed mutation of A123C for bioconjugation utility, and two genetically engineered mutants, Escherichia coli-based P-mCP-Ti7 containing a C-terminal TiO2 GEPI sequence of seven peptides (Ti7) and Hi5 insect cells-derived E-CP-Ti7-His6 C-terminally fused with Ti7+His6 tag were created. Expression vectors and protocols for enriching of the two CP variants were established and the resultant proteins were identified by western blot analysis. Their RNA-free self-assembling structures were analyzed by transmission electron microscopy (TEM) and immuno-gold labeling TEM analysis. Adherence of nanoparticles to the P-mCP-Ti7 induced protein scaffold was visualized by TEM analysis. Also discussed is the Cysteine thiol reactivity in bioconjugation reactions with the maleimide-functionalized porphyrin photosensitizers which can function as clinical photodynamic therapy agents. This study introduced a novel approach to producing an assembly-competent recombinant TMGMV CP, examined its ability to serve as a novel platform for the multivalent display of surface ligands and demonstrated an alternative method for nanodevice synthesis for nanobiotechnological applications by combining GEPIs-mediated immobilization with the controllability of self-assembling recombinant TMGMV CP.

Abstract:Abstract Magnetic microspheres (MMS) used for magnetic drug targeting consist of magnetic nanoparticles (MNP) and a pharmaceutical agent embedded in a polymeric matrix material. The application of MNP for drug targeting enables guiding the MMS to a target area, imaging the position of the MMS with magnetic particle imaging, and finally inducing drug release. As latter takes place by degradation of the MMS or diffusion through the matrix, an increase in temperature, e.g. through magnetic hyperthermia, leads to an accelerated drug release. Here, MMS consisting of poly(lactic-coglycolic) acid (PLGA) with different monomer ratios were prepared by an oil-in-water emulsion evaporation method. The model drug Camptothecin (CPT) and magnetic multicore nanoparticles (MCNP) with a high specific heating rate were embedded into the microspheres. We obtained MMS in the preferred size range of 1 to 2 μm with a concentration of MCNP of 16wt%, a drug load of about 0.5wt% and an excellent heating performance of 161 W/g MMS . Investigations of the drug release behaviour showed an accelerated drug release when increasing the temperature from 20 °C to 37 °C or 43 °C by using a water bath. In addition, an increase in drug release of about 50% through magnetic heating of the MMS up to 44 °C compared to 37 °C was observed. By this, a magnetic hyperthermia induced CPT release from PLGA MMS is demonstrated for the very first time.

Abstract:Remote control of cellular functions via magnetic forces offers unique opportunities in funda-mental research and biomedical application. Intracellular delivery of functionalized magnetic nanoparticles (MNP) provides versatile opportunities to assemble signalling platforms for spa-tiotemporal control by magnetic forces. Such magnetogenetic application, however, has re-mained highly challenging due to a lack of MNP providing suitable biological, physicochemical and magnetic properties. Here, we achieved single-step surface coating of synthetic ma-ghemite core nanoparticles with green fluorescent protein fused to the iron binding site of Mms6 from magnetotactic bacteria. We yielded MNP with intracellular stealth properties (syMagIcS), which could be readily biofunctionalized in situ and translocated within cells via magnetic field gradients. Nanoplatforms for Magnetic‐Photo‐Heating of Thermo‐Resistant Tumor Cells: Singular Synergic Therapeutic Effects at Mild Temperature

Abstract:Here, magnetic nanostructures made of chain‐like iron oxide nanocubes and photothermal dyes enwrapped in amphiphilic polymers, are developed. The obtained nanostructures are eradicating, in vitro, thermo‐resistant glioblastoma cells at temperature as low as 41 °C only when this therapeutic temperature is reached by a combination of MH and PTT of clinical conditions, thus offering new alternative approaches to treat thermo‐resistant cancer. A self‐assemble amphiphilic diblock copolymer that can incorporate iron oxide nanocubes (IONCs) in chain‐like assemblies as heat mediators for magnetic hyperthermia (MHT) and tuneable amounts of IR780 dye as agent for photothermal therapy (PTT) is developed. MHT‐heating performance of photobeads in viscous media have the same heat performances in water at magnetic field conditions of clinical use. Thanks to IR780, the photobeads are activated by infrared laser light within the first biological window (808 nm) with a significant enhancement of photo‐stability of IR780 enabling the raise of the temperature at therapeutic values during multiple PTT cycles and showing unchanged optical features up to 8 days. Moreover, the photobeads fluorescent signal is preserved once internalized by glioblastoma multiforme (GBM) cells. Peculiarly, the photobeads are used as toxic agents to eradicate thermo‐resistant GBM cells at mild heat, as low as 41 °C, with MHT and PTT both of clinical use. Indeed, a high U87 GBM cell mortality percentage is obtained only with dual MHT/PTT while each single treatment dose not provide the same cytotoxic effects. Only for the combined treatment, the cell death mechanism is assigned to clear sign of apoptosis as observed by structural/morphological cell studies and enhanced lysosome permeability.

Engineering dual-stimuli responsive poly(vinyl alcohol) nanofibrous membranes for cancer treatment by magnetic hyperthermia

Abstract:The development of new cancer treatment options, such as multifunctional devices, allows for a more personalized treatment, avoiding the known severe side effects of conventional options. In this context, on-demand drug delivery systems can actively control the rate of drug release offering a precise control of treatment. Magnetically and thermally controlled drug delivery systems have been explored as on-demand devices to treat chronic diseases and cancer tumors. In the present work, dual-stimuli responsive systems were developed by incorporating Fe3O4 magnetic nanoparticles (NPs) and poly(N-isopropylacrylamide) (PNIPAAm) microgels into electrospun polymeric fibers for application in cancer treatment. First, Fe3O4 NPs with an average diameter of 8 nm were synthesized by chemical precipitation technique and stabilized with dimercaptosuccinic acid (DMSA) or oleic acid (OA). PNIPAAm microgels were synthesized by surfactant-free emulsion polymerization (SFEP). Poly(vinyl alcohol) (PVA) was used as a fiber template originating fibers with an average diameter of 179 ± 14 nm. Stress tests of the membranes showed that incorporating both microgels and Fe3O4 NPs in electrospun fibers increases their Young's modulus. Swelling assays indicate that PVA membranes have a swelling ratio of around 3.4 (g/g) and that the presence of microgels does not affect its swelling ability. However, with the incorporation of Fe3O4 NPs, the swelling ratio of the membranes decreases. Magnetic hyperthermia assays show that a higher concentration of NPs leads to a higher heating ability. The composite membrane with the most promising results is the one incorporated with DMSA-coated NPs, since it shows the highest temperature variation, 5.1 °C. To assess the membranes biocompatibility and ability to promote cell proliferation, indirect and direct contact cell viability assays were performed, as well as cell adhesion assays. Following an extract method viability assay, all membrane designs did not reveal cytotoxic effects on dermal fibroblasts and melanoma cancer cells, after 48 h exposure and support long-term viability. The present work demonstrates the potential of dual-stimuli composite membranes for magnetic hyperthermia and may in the future be used as an alternative cancer treatment particularly in anatomically reachable solid tumors.
A Mitochondria-Targeting Magnetothermogenic Nanozyme for Magnet-Induced Synergistic Cancer Therapy

Abstract:Magnetic hyperthermia therapy (MHT) and chemodynamic therapy (CDT) are non-invasive in situ treatments without depth limitations and with minimum adverse effects on surrounding healthy tissue. We herein report a mitochondria-targeting magnetothermogenic nanozyme (Ir@MnFe2O4 NPs) for highly efficient cancer therapy. An iridium(III) complex (Ir) acts as a mitochondria-targeting agent on the surface of MnFe2O4 NPs. On exposure to an alternating magnetic field (AMF), the Ir@MnFe2O4 NPs induce a localized increase in temperature causing mitochondrial damage (MHT effect). Meanwhile glutathione (GSH) reduces Fe(III) to Fe(II) on the NPs surface, which in turn catalyzes the conversion of H2O2 to cytotoxic •OH (CDT effect). The depletion of GSH (a •OH scavenger) increases CDT efficacy, while the localized increase in temperature increases the rate of conversion of both Fe(III) to Fe(II) and H2O2 to •OH further enhancing the CDT effect. In addition, the disruption of cellular redox homeostasis due to CDT, leads to greater sensitivity of the cell towards MHT. This nanoplatform integrates these excellent therapeutic properties, with two-photon microscopy (TPM) (demonstrated in vitro) and magnetic resonance imaging (MRI) (demonstrated in vivo) to enable the precise and effective treatment of cancer.
Low Dimensional Assemblies of Magnetic MnFe$_2$O$_4$ Nanoparticles and Direct In Vitro Measurements of Enhanced Heating Driven by Dipolar Interactions: Implications for Magnetic Hyperthermia

Abstract:Magnetic fluid hyperthermia (MFH), the procedure of raising the temperature of tumor cells using magnetic nanoparticles (MNPs) as heating agents, has proven successful in treating some types of cancer. However, the low heating power generated under physiological conditions makes necessary a high local concentration of MNPs at tumor sites. Here, we report how the in vitro heating power of magnetically soft MnFe$_2$O$_4$ nanoparticles can be enhanced by intracellular low-dimensional clusters through a strategy that includes: a) the design of the MNPs to retain N\'eel magnetic relaxation in high viscosity media, and b) culturing MNP-loaded cells under magnetic fields to produce elongated intracellular agglomerates. Our direct in vitro measurements demonstrated that the specific loss power (SLP) of elongated agglomerates ($SLP=576\pm33$ W/g) induced by culturing BV2 cells in situ under a dc magnetic field was increased by a factor of 2 compared to the $SLP=305\pm25$ W/g measured in aggregates freely formed within cells. A numerical mean-field model that included dipolar interactions quantitatively reproduced the SLPs of these clusters both in phantoms and in vitro, suggesting that it captures the relevant mechanisms behind power losses under high-viscosity conditions. These results indicate that in situ assembling of MNPs into low-dimensional structures is a sound possible way to improve the heating performance in MFH.
