Abstract:The Chinese mitten crab (Eriocheir Sinensis) is a popular aquatic product in China. However, these crabs accumulate heavy metals from the ecosystem. The occurrence, source, and risk assessment of heavy metals in crabs from the main production area in China were investigated. The abundance of the metals in crabs was Mn >> As > Se > Ni > Cr > Cd > Pb> Hg. Each metal was present at a level less than the national standard (A standard for the limits of contaminants in food from China). Agricultural activities, industrial discharges, and urban sewage were the three main sources of heavy metals accumulation in Chinese mitten crab. Based on an assessment of the target hazard quotient (THQ), arsenic is the most harmful heavy metal in the crabs. In addition, mercury and lead also pose risks. The results indicate that the estimated daily intake (EDI) of mitten crabs in China would not pose a high dietary risk. (THQ < 1). The brown crab meat of females has a higher dietary risk than comparable tissues in males.

Abstract:In this study, a total of 135 Chinese mitten crab (Eriocheir sinensis) samples were collected from four major crab-producing provinces (Jiangsu, Hubei, Anhui, and Liaoning) in China. The content of 24 trace elements in the edible parts of mitten crabs was determined by inductively coupled plasma mass spectrometry (ICP-MS). In addition, the content of trace elements in crab samples collected from different provinces was compared, and the heavy metal levels exceeding the standard were evaluated. Twenty four trace metallic elements were determined in Chinese mitten crab samples. The single factor pollution index (Pi), percentage of provisionally tolerable weekly intake (PTWI), and target hazard quotients (THQ) were used to evaluate the pollution level and food safety of Chinese mitten crabs. The final assessment results of dietary risks led to the health risk assessment accompanying food safety recommendations on Chinese mitten crabs. The overall finding of this study established that Chinese mitten crab aquatic products can be eaten for nutrition; however, attention should be paid on the potential food safety risks of some heavy metals. Moreover, efforts on developing the industrial economy by the local government and peoples’ safety from food risks may also increase the awareness of environmental protection.

Abstract:Seventy Chinese mitten crab samples, encompassing a total of 2100 individuals, were collected from the main production areas in China. The objective was to assess the occurrences and patterns of 23 selected organochlorine pesticides (OCPs) in the edible tissues and assess the associated dietary risk. Concentrations of total residual OCPs in the mitten crabs ranged from 0.72 to 51.51 μg kg−1, which was comparable to other global aquatic species. Dichlorodiphenyl trichloroethanes (DDTs) and hexachlorocyclohexanes (HCHs) were the two main contributors of total OCPs, with the detected values ranging from 0.14 to 30.89 μg kg−1 and 0.23 to 4.04 μg kg−1, respectively. Source analysis indicated the coexistence of both residual and recent DDT inputs, while there was no indication of HCH usage in the main production area of mitten crab. In terms of dietary risk, at least eight individual mitten crabs per day are permissible for consumption by local residents, indicating low risk from consumption. The results presented herein should guide the production and consumption of mitten crab, as well as promote the sustainable development of aquaculture in China.

Abstract:We used a risk-benefit model to evaluate the effects of different farming methods and consumption habits on dietary risk of Chinese mitten crab. In this study, we found that cadmium levels in the total edible parts and brown meat were 0.029-0.462 mg/kg and 0.017-1.694 mg/kg, respectively. Cadmium exposure experiment showed that brown meat was the main enrichment site of cadmium. Fatty acid test found that the EPA + DHA content in total edible parts fed with formulated feed and iced trash fish was 19.64 +/- 3.62 mg/g and 28.72 +/- 2.30 mg/g (p > 0.05), respectively, and that in brown meat was 27.66 +/- 5.16 mg/g and 40.15 +/- 3.50 mg/g (p > 0.05), respectively. According to the risk-benefit model analysis, the dietary risk of brown meat under the two breeding methods is relatively high, but the risk quotient is less than 1.

Abstract:The use of Chinese mitten crabs for human consumption is controversial due to their uptake of heavy metals, such as cadmium (Cd). We developed a formula for quantification of benefit and risk assessment (hazard quotient) for the intake of a product containing essential polyunsaturated fatty acids versus heavy metals. A total of 70 samples (2100 individuals) of crabs were collected from 6 representative basins in Jiangsu province to determine Cd levels. Cd levels ranged from 0.028 to 5.015 mg kg−1, and the levels among the basin regions were similar (p > 0.05). To determine the nutritional differences between crabs fed with iced trash fish and formulated feed, we conducted a comparative aquaculture experiment. We simulated the cultivation conditions of crabs in China and found that the nutritional content in the trash fish group (28.724 ± 2.301 mg kg−1) was significantly higher than in the formulated food group (19.278 ± 3.556 mg kg−1; p < 0.05). The differences in the hazard quotient of benefit and risk balance between these two diets were not significant (p > 0.05). The average hazard quotient of iced trash fish feed culture was 0.023 ± 0.019, compared with the formulated feed culture (0.034 ± 0.028). Formulated feed and iced trash fish produce similar results as culture food for the Chinese mitten crab. In terms of cadmium exposure risk, the appropriate daily consumption of crabs is less than eight. These data can help justify culture using formulated food and the sustainable development of the Chinese mitten crab industry.

Abstract:mu Aquaculture industries supply half of the global production, leading to an increasing concern about health risk of the aquaculture products because of widespread distribution of heavy metals in the environment. Considering high dependence of toxicity and bioaccumulation of one metal not merely upon total content but mainly upon its chemical forms, recent investigations into risk assessment of aquaculture products based on total contents of heavy metals may be unreasonable and unfair. In this study, we conducted reasonable assessment to bioaccumulation and health risk of aquaculture fish and crustacean collected from eight aquaculture ponds in Zhejiang Province of East China in terms of chemical forms of two model heavy metals (Sn(II), Pb(II), four organotin compounds, trimethyl lead -TML and triethyl lead -TEL). Although all metal compounds were presented below 0.2-4.1 mu g kg(-1) in aquatic food, they were at moderate contents in environmental matrices following an order of sediment > feedstuff > aquatic food > water for most of the target compounds (i.e. 54-376 mu g kg(-1) Pb(II) in sediment, 14-94 mu g kg(-1) Pb(II) in feedstuff and organotin compounds at pg L-1 levels in water), indicating feedstuff as an important exposure source apart from water adsorption. Bioaccumulation factors, biota-sediment accumulation factors and biomagnification factors demonstrated low bioaccumulation probability although the bioaccumulation level followed an order of organotin and organolead > inorganic forms > total metal. In addition, estimated daily intakes, target hazard quotients and cancer risk demonstrated no health risk for all tin compounds and no carcinogenic risks for all lead compounds but moderate non-carcinogenic risks for TML and TEL. Overall, the accumulation level and health risk were in a closer relationship with the chemical forms of Sn and Pb than their total contents, demonstrating a high necessity to risk assessment in terms of chemical forms.

Abstract:The increasing human demand for high-quality animal protein has provided impetus for the development of aquaculture. Chinese mitten crab ( Eriocheir sinensis ) is a catadromous species rapidly being cultured in China but scientific literature documenting its life cycle environmental and economic consequences remains scarce. This study aims to address this gap by examining the spatio-temporal evolution of crab aquaculture in China since the 2000s and evaluating the environmental and economic characteristics along its life-cycle stages: megalopa, juvenile crab, and adult crab cultivation. The geostatistical analysis shows a more dispersed pattern of crab aquaculture nationally as crab grows, with coastal provinces that have brackish water for megalopa cultivation but wider spatial coverage for juvenile and adult crab cultivation. Our findings reveal that the production of 1 ton of live-weight crab results in 7.65 ton of CO 2 equivalent of greenhouse gas emissions, surpassing previous estimates for finfish fish production by approximately 50%. Most environmental pressures occur during the adult crab cultivation stage, with significant contributions from upstream processes such as electricity and feed production. By comparing between different production systems, our study shows that crab aquaculture in lake systems performs better than pond systems in terms of most global environmental impact categories and economic considerations. This work contributes to the existing literature by elucidating the spatio-temporal changes of crab aquaculture boom in China and constructing a representative life cycle data pool that broadens the benchmark knowledge on its environmental and economic characteristics. We highlight the trade-offs between environmental and economic performance as well as the balance between global and local environmental impacts to promote sustainable growth in the aquaculture industry.

Abstract:The impact of heavy metals (HMs) on the quality of aquaculture products has attracted worldwide attention. Since Litopenaeus vannamei is a popular aquaculture product among consumers worldwide, it is of great importance to guarantee its dietary safety. An in-situ monitoring program lasting for three months in a typical Litopenaeus vannamei farm found that Pb (100 %) and Cr (86 %) in the adult shrimp were higher than the safety guidelines. In the meantime, Cu (100 %), Cd (100 %) in the water and Cr (40 %) in the feed exceeded the corresponding thresholds. Therefore, quantification of different exposure pathways of shrimp and contamination origins in pond is valuable to improve the dietary safety of the shrimp. Based on Optimal Modeling for Ecotoxicological Applications (OMEGA), Cu was primarily from the ingestion of feed, accounting for 67 % of bioaccumulation, while Cd, Pb and Cr primarily entered shrimp through the adsorption from overlying water (53 % for Cd and 78 % for Pb) and porewater (66 % for Cr), respectively. The HMs in the pond water were further tracked based on a mass balance analysis. The main source of Cu in the aquaculture environment was feed, being responsible for 37 % of the total input. Pb, Cd and Cr were primarily from the inlet water with contributions of 84 %, 54 % and 52 %, respectively. In summary, the proportions of different exposure pathways and origins of HMs in pond-cultured shrimp and its living environment varied widely. To keep end-consumers eating healthily, species specific treatment is required. Feed should be regulated more for Cu. Aimed pretreatments for Pb and Cd in influent water are needed and an additional immobilization for Cr in sediment porewater should be investigated. After implementation of these treatments, the food quality improvement could be further quantified based on our prediction model.

Abstract:Ion-specific stress effects of Na(+) and Cl(-) on photosynthesis of seedlings of two soybean (Glycine max) cultivars (the salt-tolerant 'Lee68' and the salt-sensitive 'N23674') were studied and compared under isoosmotic (-0.53 MPa) solutions [PEG-6000, NaCl, Na(+) (without Cl(-)) and Cl(-) (without Na(+))] for 6 d. The results showed that the chlorophyll contents and the ribulose bisphosphate carboxylase/oxygenase (Rubisco) activities of seedlings of both cultivars were inhibited to a less degree by PEG-6000 than by NaCl, Na(+) (without Cl(-)) or Cl(-) (without Na(+)) solutions. The maximum photochemical efficiency of photosystem II (PSII) (F(v)/F(m)), electron transfer rate (ETR) and effective quantum yield of PSII photochemistry (F(v)'/F(m)') were lowered significantly by PEG-6000 treatment for 2 d and 6 d. But when treated with the three isoosmotic salt stresses, most of the above three indexes significantly declined. Leaf stomatal conductance (G(s)) and net photosynthetic rate (P(n)) in both cultivars were significantly decreased under the four isoosmotic treatments, and much more drops were observed in the three salt stresses, but the intercellular CO(2) concentration (C(i)) decreased by PEG-6000 treatment and increased by the three salt stresses. Higher decreases including chlorophyll content, Rubisco activity, F(v)/F(m), ETR, F(v)'/F(m)', P(n) and G(s), and higher contents of Cl(-) as well as the content of Cl(-) plus Na(+) in chloroplasts were all observed under Cl(-) (without Na(+)) treatment than those under Na(+) (without Cl(-)) treatment, especially for the salt-sensitive cultivar 'N23674'. The results indicate that, among the adverse effects of NaCl stress on photosynthesis of G. max seedlings, the ionic toxicity was stronger than osmotic stress, and the toxicity of Cl(-) was more severe than that of Na(+).
A large geographic-scale characterization of organochlorine pesticides (OCPs) in surface sediments and multiple aquatic foods of inland freshwater aquaculture ponds in China: Co-occurrence, source and risk assessment

Abstract:Inland freshwater aquaculture ponds (IFAPs) represent the key component of the global lentic freshwater environment and are increasingly important for global aquaculture production, yet the occurrence of organochlorine pesticides (OCPs) in these pond systems remains largely unknown. Here, we characterized the residual concentrations of 19 individual OCPs in sediments and in cultured fish and crustacean species (crabs, shrimp, crayfish and lobster), which were on-spot sampled from the IFAPs at a large region-scale in China. The total OCP levels in sediments varied dramatically between regions. Crabs presented the greatest OCP contamination among the studied species. Dichlorodiphenyltrichloroethanes (DDTs) was the dominating contaminant in sediments and crabs and its stable degradation products 4,4′-DDE and 4,4′-DDD were co-occurrent between these two compartments. The diagnostic ratio analysis indicated fresh inputs of DDTs, lindane and aldrin in multiple regions, which may be resulted from agricultural soil erosion, surface runoff and local anthropogenic activities. Ecological impacts of these pesticides could be expected at some sites due to their levels in sediments above the risk level. Risk assessment based on the OCP levels corrected by the cooking loss revealed that daily consumption of the IFAPs-derived aquatic foods may pose carcinogenic risks in humans.

Abstract:The rice-crab co-culture system has become the dominant mode of farming due to its advantageous production of crabs. To investigate the effects of different culture modes on trace element accumulation and health risks though consumption, we determined the concentrations of 16 trace elements (Ag, Al, As, Ba, Cd, Co, Cr, Cu, Fe, Mn, Ni, Pb, Se, Tl, V, Zn) in Chinese mitten crabs (Eriocheir sinensis) from conventional monoculture (CM) and rice-crab co-culture system (RCCS) in two primary producing areas of China. Significant variations were found between CM and RCCS concerning trace element accumulation, with most trace elements showing higher levels in CM than in RCCS, except for Mn and Al. Correlation and PCA analysis indicate that RCCS has less environmental impact than CM. Moreover, the metal pollution index was also higher in CM than in RCCS. Based on the target hazard quotient (THQ), RCCS reduces the health risk associated with trace elements intake compared to CM. THQ values for both modes of culture are less than 1, indicating no or low health risks posed by trace elements in crabs. Further studies are needed in comparing the trace element accumulation in crabs from CM and RCCS within the same geographic area.

Abstract:Pyrethroid pesticides are widely used in agriculture, aquaculture, and daily life in China, leading to concerns about their environmental residues. However, there is limited information available regarding the compositional distribution, bioaccumulation, and associated health and environmental risks of pyrethroids in the aquaculture areas of Eastern China. This study investigated the residues, distribution, and risk assessment of pyrethroids including deltamethrin, permethrin, fenvalerate, cypermethrin and bifenthrin in Yangcheng Lake using solid-phase extraction and gas chromatography-tandem mass spectrometry. The risk assessment of water, sediment, and aquatic products of Yangcheng Lake was conducted using various evaluation methods. The results revealed that pyrethroids were found at higher levels in both water and sediment in Yangcheng Lake with some seasonal variations observed in their distribution patterns. Compared with other watersheds in China, typical aquaculture areas of the Yangtze River Delta showed higher detection rates and quantities of pyrethroid pesticides. Ecological risk assessments indicated high risks to the environment from all five tested pyrethroids at Yangcheng Lake; deltamethrin posed the highest risk among them. Dietary assessment results showed no risks associated with aquatic products from Yangcheng Lake but drinking water at certain sites presented unacceptable health risks.

Abstract:Arsenic (As) is a major hazardous element in natural environments, and arsenic pollution is becoming an issue of concern worldwide. The more toxic inorganic arsenic, such as trivalent arsenite As(Ⅲ) and pentavalent arsenate As(Ⅴ), which normally can transform to organic arsenic compounds, such as arsenobetaine (AsB), arsenocholine (AsC), monomethyl arsenic acid (MMA), dimethyl arsenic acid (DMA), trimethyl arsenic acid (TMA), arsenosugars and arsenolipids. In this study, a total of 2130 individuals of Chinese mitten crabs were collected from seven locations and mixed to 71 samples. Total arsenic and six major species (AsC, AsB, DMA, MMA, As (Ⅲ), and As (Ⅴ)) were determined by inductively coupled plasma mass spectrometry (ICP-MS) and high-performance liquid chromatography-inductively coupled plasma mass spectrometry (HPLC-ICP-MS), respectively. The target hazard quotient (THQ) was utilized to evaluate the human health risk. The total arsenic concentration and totals for the six arsenic forms are 0.25–1.66 mg/kg and 0.05–1.19 mg/kg wet mass, respectively. Except for HH and LH, the six arsenic species concentrations accounted for more than 50.0 % of the total arsenic. The less toxic AsB is the most predominant in crabs, comprising 50.0 %–90.0 % of the sum of six types of arsenic. The more toxic inorganic arsenic only range from 0.01 to 0.21 mg/kg wet mass, much less than the limit content of 0.50 mg/kg inorganic As in crustacea. The THQ values of inorganic arsenic through the consumption of Chinese mitten crabs are estimated, and the values are all less than 1, indicating that intaking Chinese mitten crabs collected from China will not cause an appreciable hazard risk to human health.

Abstract:Introduction: This single blind study was conducted to address safety concerns regarding co-administration of diphtheria/ tetanus (DT,Td) boosters and meningococcal C conjugate (MCC) vaccines containing diphtheria or tetanus conjugate proteins. Methods: 1754 subjects (835 preschoolers and 919 school leavers) were randomised to receive one of three MCC products a month before, after or at the same as routine DT or Td boosters. They (or their parents) completed a health diary for 10 days to record local reactions and systemic symptoms after each vaccination. The effect of antibody levels pre- and post-vaccination, pre-existing allergies and medication taken before and for up to ten days after vaccination on reactogenicity was assessed. Results: No relationship between pre-vaccination antibody levels and local reactions or systemic symptoms was found. Local reactions were more common after DT/Td than MCC vaccination and were related to post-vaccination diphtheria and tetanus antibody levels in younger children and to the post-vaccination tetanus only antibody levels in older children. Pre-existing allergies were not related to reactogenicity. Use of analgesics/ antipyretic medication significantly reduced the incidence of local reactions and of some systemic symptoms. In the first three days after vaccination there was an excess of some systemic symptoms including fatigue/ malaise and headache for all ages, and crying/ irritability for younger children and nausea/ vomiting and dizziness/ faintness for older subjects. Eleven serious adverse events were reported, none of which was deemed related to vaccination. Conclusion: MCC vaccines can be given under the above schedules and safely in the presence of raised diphtheria and tetanus antibody levels without materially altering reactogenicity profiles of either vaccine.

Abstract:In recent years,there has been a steady increase in the demand of aquatic products and more attention has been given to the safety of the water environment.Heavy metal pollution may pose risk to human because of the toxicity and persistence of metals.In order to evaluate the ecological risk of heavy metals in the shrimp ponds,we measured the contents of Cr,Cu,As,Cd,and Pb in water,aquatic plants,shrimps,and sediments.The potential ecological risk of heavy metals in the shrimp ponds was assessed using potential ecological risk factors.And then,we studied the chemical speciation of five metals in sediments using sequential extraction.Our study showed that Cu had the highest ecological risk in water and Cd had the highest ecological risk in sediments while the potential ecological risk of the other heavy metals was low.The level of As in shrimps was higher,while other metals were lower than safety limits.The concentrations of Cr,As,Cd,Pb in different parts of the shrimp decreased following the order of exoskeleton>muscle>head,while Cu were concentrated in shrimp heads.In the sediments,Cr,Cu,Cd,and Pb mainly existed in the form of complexes phase and residuals phase,its bioavailability was low.About 30% of As existed in exchangeable phase and the bioavailability was high.
Transcriptomics Analysis of the Toxicological Impact of Enrofloxacin in an Aquatic Environment on the Chinese Mitten Crab (eriocheir Sinensis).

Abstract:Enrofloxacin is an important antimicrobial drug that is widely used in aquaculture. Enrofloxacin residues can have negative effects on aquatic environments and animals. The toxicological effects of different concentrations of enrofloxacin residues in cultured water on Chinese mitten crabs (Eriocheir sinensis) were compared. A histological analysis of the E. sinensis hepatopancreas demonstrated that the hepatopancreas was damaged by the different enrofloxacin residue concentrations. The hepatopancreas transcriptome results revealed that 1245 genes were upregulated and that 1298 genes were downregulated in the low-concentration enrofloxacin residue group. In the high-concentration enrofloxacin residue group, 380 genes were upregulated, and 529 genes were downregulated. The enrofloxacin residues led to differentially expressed genes related to the immune system and metabolic processes in the hepatopancreas of the Chinese mitten crab, such as the genes for alkaline phosphatase, NF-kappa B inhibitor alpha, alpha-amylase, and beta-galactosidase-like. The gene ontology terms "biological process" and "molecular function" were enriched in the carboxylic acid metabolic process, DNA replication, the synthesis of RNA primers, the transmembrane transporter activity, the hydrolase activity, and the oxidoreductase activity. A Kyoto Encyclopedia of Genes and Genomes pathway analysis determined that the immune and metabolic signal transduction pathways were significantly enriched. Furthermore, the nonspecific immune enzyme (alkaline phosphatase) and the metabolic enzyme system played a role in the enrofloxacin metabolism in the E. sinensis hepatopancreas. These findings helped us to further understand the basis of the toxicological effects of enrofloxacin residues on river crabs and provided valuable information for the better utilization of enrofloxacin in aquatic water environments.
Occurrence and dietary exposure risk assessment of phthalate esters in Chinese mitten crabs (Eriocheir sinensis) from Hubei, central China

Abstract:Recently, the wide existence of phthalate esters (PAEs) in aquatic environment has drawn great public attention, due to their negative health impacts on human and aquatic organisms. Chinese mitten crab is a major aquaculture economic resource in Hubei, feeding a world population of more than 10 million. Due to their wide consumption and great economic value, it's imperative to study the dietary risks of PAEs in mitten crabs ensuring food safety. In this study, a simple and selective method based on matrix matched calibration strategy coupled with gas chromatography-tandem mass spectrometry (GC-MS/MS) was developed to quantify 17 PAEs in Chinese mitten crabs (Eriocheir sinensis) from Hubei, central China and their dietary exposure risks were investigated. Recoveries were in the range of 56.9–131.2%. The limits of detection and quantification were 0.01–0.2 μg kg−1 and 0.5–7.5 μg kg−1, respectively. The PAEs were found to be present in both juvenile and adult mitten crabs in the study area. The mean concentration of total PAEs (∑17 PAEs) was 495.73–2033.39 μg kg−1 in crab muscles, and 159.69–651.69 μg kg−1 in hepatopancreas and gonad, respectively. The highest detection frequency of PAEs in all samples was di(2-ethylhexyl) phthalate (DEHP), and followed by dibutyl phthalate (DBP) and diisobutyl phthalate (DiBP). The estimated daily intakes of DEHP, DBP and DiBP, were more than ten times greater than any other PAEs. Calculated hazard ratio of PAEs were all below 0.1, suggesting that intake of PAEs by dietary of mitten crabs had non-cancer risk for both adults and children. Overall, this study provides a reliable method to evaluate PAEs pollution in aquatic food and critical data on the occurrence of PAEs in Chinese mitten crabs (Eriocheir sinensis) from Hubei, central China, which is a key part of PAEs pollution prevention in food source.
