Abstract:Degradation of crop straw in natural environment has been a bottleneck. There has been a recent increase in the exploration of cold-adapted microorganisms as they can solve the problem of corn straw degradation under low temperatures and offer new alternatives for the sustainable development of agriculture. The study was conducted in low-temperature (10°C) and high-efficiency cellulose-degrading bacteria were screened using carboxymethyl cellulose (CMC) selection medium and subjected to genome sequencing by the third-generation Pacbio Sequl and the second-generation Illumina Novaseq platform, and their cellulase activity was detected by 3,5-dinitrosalicylic acid (DNS) method. The results showed that the low-temperature (10°C) and high-efficiency cellulose-degrading bacterium Bacillus subtilis K1 was 4,060,823 bp in genome size, containing 4,213 genes, with 3,665, 3,656, 2,755, 3,240, 1,261, 3,336 and 4,003 genes annotated in the non-redundant protein sequence database (NR), Pfam, clusters of orthologous groups of proteins (COGs), Genome Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG), and Annotation databases, respectively. In addition, a large number of lignocellulose degradation-related genes were annotated in the genome. The cellulose activity of B. subtilis K1 was higher, exhibiting the highest activity of endo-β-glucanase (24.69 U/ml), exo-β-glucanase (1.72 U/ml) and β-glucosaccharase (1.14 U/ml). It was found that through adding cold-adapted cellulose-degrading bacteriaK1 in the corn straw composting under 6°C (ambient temperature), the average temperature of straw composting was 58.7°C, and higher 86.7% as compared to control. The HA/FA was higher 94.02% than the control and the lignocellulose degradation rate was lower 18.01–41.39% than the control. The results provide a theoretical basis for clarifying the degradation potential of cold-adapted cellulose-degrading bacteria and improving the cellulose degradation efficiency.

Abstract:The metatranscriptomic recharacterization in the present study captured microbial enzymes at the unprecedented scale of 40,000 active genes belonged to 2,269 KEGG functions were identified. The novel information obtained herein revealed interesting patterns and provides an initial transcriptional insight into the thermophilic cellulose methanization process. Synergistic beta-sugar consumption by Thermotogales is crucial for cellulose hydrolysis in the thermophilic cellulose-degrading consortium because the primary cellulose degraders Clostridiales showed metabolic incompetence in subsequent beta-sugar pathways. Additionally, comparable transcription of putative Sus-like polysaccharide utilization loci (PULs) was observed in an unclassified order of Bacteroidetes suggesting the importance of PULs mechanism for polysaccharides breakdown in thermophilic systems. Despite the abundance of acetate as a fermentation product, the acetate-utilizing Methanosarcinales were less prevalent by 60% than the hydrogenotrophic Methanobacteriales. Whereas the aceticlastic methanogenesis pathway was markedly more active in terms of transcriptional activities in key genes, indicating that the less dominant Methanosarcinales are more active than their hydrogenotrophic counterparts in methane metabolism. These findings suggest that the minority of aceticlastic methanogens are not necessarily associated with repressed metabolism, in a pattern that was commonly observed in the cellulose-based methanization consortium and thus challenge the causal likelihood proposed by previous studies.

Abstract:A lignocellulolytic microbial consortium holds promise for the in situ biodegradation of crop straw and the comprehensive and effective utilization of agricultural waste. In this study, we applied metagenomics technology to comprehensively explore the metabolic functional potential and taxonomic diversity of the microbial consortia CS (cultured on corn stover) and FP (cultured on filter paper). Analyses of the data on metagenomics taxonomic affiliations revealed considerable differences in the taxonomic composition and carbohydrate-active enzymes profile of the microbial consortia CS and FP. Pseudomonas, Dysgonomonas and Sphingobacterium in CS and Cellvibrio and Pseudomonas in FP had a much wider distribution of lignocellulose degradative ability. The genes for more lignocellulose degradative enzymes were detected when the relatively simple substrate filter paper was used as the carbon source. Clusters of Orthologous Groups (COG) and Kyoto Encyclopedia of Genes and Genomes (KEGG) annotation analyses revealed considerable levels of similarity, and carbohydrate metabolic and amino acid metabolic pathways were the most enriched in CS and FP, respectively. The mechanism used by the two microbial consortia to degrade lignocellulose was similar, but the annotation of quantity of genes indicated that they are diverse and vary greatly. These data underlie the interactions between microorganisms and the synergism of enzymes during the degradative process of lignocellulose under different substrates and suggest the development of potential microbial resources.
Cellulose-Degrading Strains: their Screening and Application to Corn Straw in Low-Temperature Environments

Abstract:Cellulose-degrading strains play an important role in cellulose degradation at low temperatures. In this study, three cellulose-degrading strains with high carboxymethyl cellulase (CMCase) activity were isolated from the soil in a low temperature environment of 10oC. ZS-7 was Cladosporium sp, ZS-8 was Alternaria porri, and ZS-10 was Trichoderma harzianum, based on 16s rDNA gene sequence analysis. They were mixed, marked as ZS-M, and used for corn straw degradation at 15oC in order to determine their application properties. ZS-7, ZS-8, ZS-10, and ZS-M efficiently degraded corn straw by 45.98%, 47.96%, 34.215%, and 33.56%, respectively, after 30 days of stationary culture. The dynamics of CMC, β-Glucosidase, and FPA during corn straw degradation were determined. The CMCase activity of ZS-7 and ZS-8 was 31.732 U/ml and 31.111 U/ml, respectively, but the enzyme activities and degradation of ZS-M were lower than the single strain because of metabolic effects that influenced the degradation process. The pH values of ZS-7, ZS-10, and ZS-M were particularly stable, changing from 7.08 to 7.77 during the full degradation cycle. ZS-7, ZS-8, and ZS-10 could be applied more broadly to degrade corn straw in cold regions.

Abstract:Low temperature is one of the bottleneck factors that limits the degradation of straw during rice straw incorporation. Determining strategies to promote the efficient degradation of straw in cold regions has become a highly active research area. This study was to investigate the effect of rice straw incorporation by adding exogenous lignocellulose decomposition microbial consortiums at different soil depths in cold regions. The results showed that the lignocellulose was degraded the most efficiently during straw incorporation, which was in deep soil with the full addition of a high-temperature bacterial system. The composite bacterial systems changed the indigenous soil microbial community structure and diminished the effect of straw incorporation on soil pH, it also significantly increased rice yield and effectively enhanced the functional abundance of soil microorganisms. The predominant bacteria SJA-15, Gemmatimonadaceae , and Bradyrhizobium promoted straw degradation. The concentration of bacterial system and the depth of soil had significantly positive correlations on lignocellulose degradation. These results provide new insights and a theoretical basis for the changes in the soil microbial community and the application of lignocellulose-degrading composite microbial systems with straw incorporation in cold regions.

Abstract:Abstract Purpose The highly efficient degradation bacteria were selected from the humus from the very cold straw in China for many years to construct the in situ degradation bacteria, and the degradation efficiency of corn straw was determined by process optimization. Methods According to the main components of corn straw, through morphological, physiological, and biochemical screening, three highly efficient complementary degradation strains were selected to construct the compound flora, and the degradation efficiency was analyzed by Fourier transform infrared spectrometer, field emission scanning electron microscope, and X-ray diffractometer. Result The corn straw selected in this paper is mainly composed of cellulose (31.99%), hemicellulose (25.33%), and lignin (14.67%). Through the determination of enzyme activity, strain Streptomyces sp. G1 T has high decomposition ability to cellulose and hemicellulose but weak utilization ability to lignin; strain Streptomyces sp. G2 T has the strongest decomposition ability to cellulose and hemicellulose among the three strains. The decomposition ability of strain Streptomyces sp. G3 T to lignin was the strongest among the three strains. Therefore, by compounding the three strains, the decomposition ability has been greatly improved. The optimal process conditions obtained by single factor and response surface method are as follows: pH is 7, temperature is 30 °C, inoculation amount is 5%, rotational speed is 210 rpm, and the weight loss rate of straw is 60.55% after decomposing for 7 days. A large amount of degradation of corn straw can be seen by Fourier transform infrared spectrometer, field emission scanning electron microscope, and X-ray diffractometer. Conclusion Streptomyces sp. G1 T , Streptomyces sp. G2 T , and Streptomyces sp. G3 T screened from straw humus in very cold areas were used to construct in situ degradation bacteria, which had good straw degradation activity and had the potential to be used for straw treatment in cold areas after harvest. This characteristic makes the complex bacteria become a strong competitive candidate for industrial production, and it is also an effective biotechnology in line with the current recycling of resources.

Abstract:Low ambient temperature become the limiting factor of composting in cold regions, thus hindering the recycle of agricultural and forestry wastes. In this study, the composting of mushroom residue and wood chips (MRWC) under low temperature was successfully implemented with inoculation of psychrotolerant cellulolytic microbial agent. Composting entered thermophilic stage on third day and the peak temperature reached to 66.25 °C. After 84 days of composting, the degradation rate of cellulose and hemicellulose was 40.85% and 100%, respectively and the compost product was completely mature and met the requirements of organic fertilizer. Metagenomic and transcriptome sequencing were applied to reveal the microbial composition and their substrates conversion functions and adaptation mechanisms through low to high temperatures. Streptomyces , Mesorhizobium , Devosia , Aspergillus and Mucor were dominant genera in the microbial community that were rich in genes of lignocellulose degradation. Various genes related to low temperature adaptation (fatty acid, trehalose, mannitol, betaine metabolism and cold shock mechanism) and high temperature tolerance (heat shock and antioxidant) were detected during MRWC composting. These results indicated that microbes during composting constituted a high-efficiency lignocellulosic ultilization system in cold conditions. Besides, the microbes of microbial agent, especially Streptomyces and Aspergillus , possessed numerous genes involving in lignocellulose degradation and temperature adaptation and quite different temperature response patterns were found to perform in bacteria and fungi. The transcription levels of most these genes in Aspergillus exhibited significant differences under different substrates and temperature conditions, suggesting that the inoculum was crucial to the composting process and beneficial to maintain the temperature of piles. This study demonstrated that the application of psychrotolerant microbes was a promising strategy to increase the efficiency of composting in cold regions and these results could also provided the guidance for optimizing microbial agent.

Abstract:Corn straw is a type of high-yield agricultural waste in China, but it has not been fully utilized owing to the diffi culty of degrading it and its low utilization rate. To improve the rate of degradation of corn straw and ensure that the straw resources are utilized more fully, we obtained an actinomycete that could simultaneously degrade cellulose and pectin from the cold region and black glebe of northeast China. The strain was named DPA-3-18, and its biological morphology and the sequence of its 16S rRNA were identifi ed. These analyses determined that it is a species of Streptomyces. The strain was combined with the lab-stored GS-30 and Bacillus subtilis to construct composite fl ora and conduct a single factor experiment. Signifi cantly higher factors were selected by Plackett Burman, and its fermentation conditions were optimized using response surface methodology. The optimal cultivation conditions were 4% of inoculum, a culture temperature of 28.51°C, an initial pH value of the medium of 7.43, and the oscillation during fermentation took place at 199.62 rpm. Under these conditions, 40.38% of the corn straw was degraded. F rom the results of SEM, we found that the structure of corn straw before degradation was complete, and the internal structure of corn straw after degradation with c omposite fl ora was destructed. The FT-IR spectra shown that the absorption peaks in all places of corn straw were reduced after degradation by the composite fl ora, the absorption peaks at 1640 cm-1 of corn straw after degradation were lower than that of before degrad ation, which confi rmed the decrease in cellulose. It indicated that the corn straw was signifi cantly degraded by the composite fl ora. This study contributed to understanding the change in corn straw after fermentation by the composite fl ora, and providing a theoretical basis for comprehensive utilization of corn straw.

Abstract:The rapid development of agriculture has led to the production of a large amount of crop straw, necessitating effective strategies for its management. Microbial degradation offers a promising method. In this study, a novel microbial consortium composed of Phanerochaete chrysosporium, Aspergillus niger, and Streptomyces griseorubens, known for their robust lignocellulose degradation capabilities, was constructed for rice straw degradation. The establishment of this microbial consortium was based on the growth curve and antagonistic tests. Orthogonal optimization revealed that Streptomyces griseorubens played a predominant role in the degradation of rice straw. The optimal degradation conditions were determined as follows: nitrogen source concentration of 2.5 gL- 1, material-liquid ratio of 40 g L- 1, inoculum size of 3%, and pH value of 9. Under these conditions, the degradation efficiency reached 42% within 15 days. The decomposition of lignocellulosic components in the straw was confirmed through various characterization methods. Additionally, as the degradation process progressed, there was a noticeable decrease in protein-like substances and an increase in humic acid-like substances in the degradation solution.

Abstract:Degradation of most of the lignocellulose-rich agricultural residue in the cold regions of China is limited due to the cold climate. Lignin is the main component of lignocellulose, and the effective degradation of lignin is one of the most crucial processes in degrading lignocellulose. Psychrotrophic lignin-degrading bacteria and cold adapted ligninolytic enzymes have promising potential for the degradation and transformation of lignin, which are conducive to the resource utilization of lignocelluloses and energy-saving production under cold conditions. In this study, a newly psychrotrophic bacterial strain, Arthrobacter sp. C2, was isolated. The optimal enzyme activity conditions and lignin degradation pathways of C2 were investigated using sodium lignin sulfonate as substrate. The optimal conditions for enzyme activity included an initial pH of 6.74, a temperature of 14.9 °C, an incubation time of 6.87 days, and an inoculum size of 2.24%. Under the optimal conditions, the lignin peroxidase and manganese peroxidase activities and the degradation rate reached 29.8 U/L, 56.4 U/L and 40.1%, respectively. The biodegradation products including acids, phenols, aldehydes and alcohols were analyzed by gas chromatography–mass spectrometry and Fourier transform infrared spectroscopy. Further, the potential degradation pathways were proposed according to the results obtained in this study and those presented in the relevant literature. This study not only provides valuable psychrotrophic strain resources for the sustainable utilization of lignocellulose in cold regions, but also supplies potential application options for energy-saving production of useful chemicals using cold adapted enzymes.Graphic abstract

Abstract:A consortium-APcT2 composed of 92% of Trichoderma sp. T-1, 6.7% of P. chrysosporium and 1.3% of A. oryzae A-4 that produces cellulolytic enzyme system with strong wheat straw saccharification was constructed using Taguchi design combined with variance analysis (ANOVA). Among 32 consortia constructed from 6 fungi using Taguchi design, consortium 20 with the strongest composition was selected. The inhibitive fungal constituent in consortium 20 was subsequently removed according to the ANOVA results. The finally optimized consortium-APcT2 yielded 805.12 mg gds(-1) sugars, 26.98% higher than the pure Trichoderma sp. T-1. Protein profile analysis of the cellulolytic enzyme systems, sugar composition analysis of the hydrolysates and compatibility evaluation of the fungal constituents showed that the enhanced straw saccharification of the consortium-APcT2 could be mainly attributed to the enhancement of the co-cultivation for enzyme production and the synergistic action of different types of enzymes in the hydrolysis process.
Degradation of lignocelluloses in straw using AC-1, a thermophilic composite microbial system

Abstract:In composting, the degradation of lignocellulose in straw is problematic due to its complex structures such as lignin. A common solution to this problem is the addition of exogenous inoculants. AC-1, a stable thermophilic microbial composite, was isolated from high temperature compost samples that can decompose lignocellulose at 50–70 °C. AC-1 had a best degradation efficiency of rice straw at 60 °C (78.92%), of hemicellulose, cellulose and lignin were 82.49%, 97.20% and 20.12%, respectively. It showed degrad-ability on both simple (filter paper, absorbent cotton) and complex (rice straw) cellulose materials. It produced acetic and formic acid during decomposition process and the pH had a trend of first downward then upward. High throughput sequencing revealed the main bacterial components of AC-1 were Tepidimicrobium , Haloplasma, norank-f-Limnochordaceae, Ruminiclostridium and Rhodothermus which provides major theoretical basis for further application of AC-1.

Abstract:In order to solve the problem of low and poor straw degradation in typical black soil areas of Northeast China, the present study was carried out to screen the potential of in situ strains with cellulose degradation ability from black soils of Northeast China to play a role in the resourceful utilization of straw and the development of sustainable agriculture. The straw degradation potential of the strains was evaluated by combining sodium carboxymethyl cellulose plate screening and cellulase viability assay; the species identification of the strains was carried out by morphology, physiology, biochemistry, and molecular biology; and the basic medium formulation of the strains was optimized by Box–Behnken response surface methodology. Ten cellulose-degrading strains were identified: ZL-5, ZL-69, ZL-88, ZL-95, ZL-111, ZL-137, ZL-139, ZL-140, ZL-187, and ZL-216, of which ZL-139 had the highest cellulase production capacity, with a cellulase secretion of 7.8781 U/mL in the enzyme-producing medium. ZL-139 was identified as Bacillus cereus; the optimized best formulation was glucose—4.284 g/L, yeast extract—1.454 g/L, MgSO4—0.417 g/L, KH2PO4—0.5 g/L, KH2PO4—0.5 g/L, K2HPO4—1.5 g/L, and NaCl—1.0 g/L. In conclusion, strain ZL-139 has good potential for crop straw degradation and can be a candidate strain for a straw-rotting agent in northeast China, with promising prospects for development and utilization.

Characterization of a Thermophilic Lignocellulose-Degrading Microbial Consortium with High Extracellular Xylanase Activity.

Abstract:A microbial consortium, TMC7, was enriched for the degradation of natural lignocellulosic materials under high temperature. TMC7 degraded 79.7% of rice straw during 15 days of incubation at 65°C. Extracellular xylanase was effectively secreted and hemicellulose was mainly degraded in the early stage (first 3 days), whereas primary decomposition of cellulose was observed as of day 3. The optimal temperature and initial pH for extracellular xylanase activity and lignocellulose degradation were 65°C and between 7.0 and 9.0, respectively. Extracellular xylanase activity was maintained above 80% and 85% over a wide range of temperature (50-75°C) and pH values (6.0-11.0), respectively. Clostridium likely had the largest contribution to lignocellulose conversion in TMC7 initially, and Geobacillus, Aeribacillus, and Thermoanaerobacterium might have also been involved in the later phase. These results demonstrate the potential practical application of TMC7 for lignocellulosic biomass utilization in the biotechnological industry under hot and alkaline conditions.
Screening and characterization of a low-temperature-resistant cellulose-degrading strain, Trichoderma harzianum L-8, from a primitive forest

Abstract:Low temperature is a major factor limiting the bio-sustainable and efficient conversion of cellulose-based resources in cold regions. In this study, a low-temperature resistant cellulose-degrading fungus with high cellulase production was screened from samples found in a primitive forest in Daqing by straw using the enrichment-restricted culture technique. The fungus was identified as genus Trichoderma harzianum, strain L-8 by morphological and molecular biological analysis. The enzyme production conditions were optimized via response surface methodology, and the optimal conditions for the enzyme production of Trichoderma harzianum L-8 were as follows: a CMC-Na addition of 10.63 g·L-1, an ammonium sulfate addition of 2.22 g·L-1, an initial pH of 5.29, and a lecithin addition of 5.18 g·L-1 when the CMCase reached 53.40 IU·mL-1. The leading enzyme families of Trichoderma harzianum L-8 were identified via proteomic analysis. Proteases including glycosyl hydrolase family 3-4 and cellobiohydrolase play important roles in cellulose degradation. The strain Trichoderma harzianum L-8 showed a strong cellulose degradation ability under low temperatures, providing strain resources for cellulose resource biotransformation technology in cold regions.

Abstract:In the present study, 27 bacterial strains were isolated from environmental samples and screened for higher lignocellulose-degrading efficiency. The best degrader was combined in pairs with 14 strains with high β-glucosidase activity to formulate a consortium. Microbial consortium 625 showed high lignocellulose degradation efficiency. ZJW-6 with low β-glucosidase activity and the best lignocellulose decomposer was identified as a member of Cellulomonas. Consortium 625 composed of ZJW-6 and DA-25, an Acinetobacter, showed the highest degradation rate (57.62%) under optimized conditions. The DA-25 filtrate promoted ZJW-6 growth, upregulating the activity of key lignocellulose-degrading enzymes, including β-glucosidase, endoglucanase, xylanase, laccase, and lignin peroxidase. ZJW-6 and DA-25 worked in a subordination manner when co-cultivated. ZJW-6 acted as the major decomposer whose growth and enzymatic activities were promoted by DA-25. This study proposes a novel microbial consortium with improved lignocellulose degradation efficiency and reduce the C:N ratio of lignocellulose materials, which can enhance bioenergy production.

Abstract:[1] One of the challenges for model prediction and validation is providing them with data of appropriate spatial and temporal resolution. The maturation and increased application of autonomous underwater vehicles (AUVs) in aquatic environments allows systematic data collection on these model-relevant scales. The goal of this study was to apply a fine-scale circulation and transport model (Delft3D) to improve AUV mission planning and use data collected by the AUV to evaluate and improve model performance. A dye release was conducted in a tidally dominated embayment, and a planning phase model based on the best available data was used as a baseline for evaluation and for AUV mission planning (forecast). The planning phase model correctly predicted the general shape and direction of the dye plume and allowed for successful mission planning. Subsequently, bathymetry data collected by the AUV was incorporated into the model (hindcast), with temperature and salinity collected before the experiment. Comparisons with fluorometer measurements from the AUV indicate that the model effectively predicted the edges of the plume and centerline location. The location was also confirmed by remote sensing from an aircraft. Thermal stratification was found to be an important fate mechanism in the final model, and the results demonstrate the integration of observational data sets for small, short-duration surface-contaminant releases. This study highlights the strength of a phased, iterative approach with observation platforms and may serve as a guide toward improving the performance and evaluation of future coastal hydrodynamic and transport modeling efforts.

Abstract:The biodegradation of straw by cellulose-degrading strains plays an important role in the disposal of agricultural waste. In this study, a Penicillium sp. strain JiTF01 that could degrade cellulose was isolated at the low temperature (10°C), and its fermentation conditions were optimized by response surface method. Strain JiTF01 was applied to degrade rice straw at 10°C. It was found that the straw degradation ratio could reach to 46.53% on the 25th day of culture with the enzyme activity of 44.30 U/mL under optimized fermentation conditions. In addition, given that strain JiTF01 showed excellent acid resistance and the cellulase produced by strain JiTF01 exhibited higher enzymatic activity and stability under acidic conditions, the uninterrupted integration process of acid pretreatment and acid fermentation of straw was then performed. The results showed that the degradation ratio of 53.61% was observed after 25 days of the fermentation at pH 3, which increased by 7.08% than that under optimized conditions. Besides, the effect of straw fermentation broth on the growth of rice seed was also investigated, and the 10 −5 and 10 −6 diluted fermentation broth was found that could significantly promote the growth of rice seedlings and enhance the germination rate of rice seeds under the salt stress of 100 and 200 mmol/L. The conclusions found in this article might have important implications for straw recycling in cold regions.
