Abstract:Quaternary ammonium amphiphiles are a class of compounds with a wide range of commercial and industrial uses. In the pharmaceutical field, the most common quaternary ammonium surfactant is benzalkonium chloride (BAC), which is employed as a preservative in several topical formulations for ocular, skin, or nasal application. Despite the broad antimicrobial activity against Gram-positive and Gram-negative bacteria, as well as fungi and small enveloped viruses, safety concerns regarding its irritant and cytotoxic effect on epithelial cells still remain. In this work, quaternary ammonium derivatives of leucine esters (C10, C12 and C14) were synthesised as BAC analogues. These cationic surfactants were characterised in terms of critical micelle concentration (CMC, by tensiometry), cytotoxicity (MTS and LDH assays on the Caco-2 and Calu-3 cell lines) and antimicrobial activity on the bacterial species Staphylococcus aureus and Enterococcus faecalis among the Gram-positives, Escherichia coli and Pseudomonas aeruginosa among the Gram-negatives and the yeast Candida albicans. They showed satisfactory surface-active properties, and a cytotoxic effect that was dependent on the length of the hydrophobic chain. Lower minimum inhibiting concentration (MIC) values were calculated for C14-derivatives, which were comparable to those calculated for BAC toward Gram-positive bacteria and slightly higher for Gram-negative bacteria and C. albicans. Thus, the synthesised leucine-based quaternary ammonium cationic surfactants can potentially find application as promising surface-active compounds with antimicrobial activity.

Abstract:The antimicrobial and antibiofilm properties of arginine-based surfactants have been evaluated. These two biological properties depend on both the alkyl chain length and the spacer chain nature. These gemini surfactants exhibit good activity against a wide range of bacteria, including some problematic resistant microorganisms such us methicillin-resistant Staphylococcus aureus (MRSA) and Pseudomonas aeruginosa. Moreover, surfactants with a C10 alkyl chain and C3 spacer inhibit the (MRSA) and Pseudomonas aeruginosa biofilm formation at concentrations as low as 8 μg/mL and are able to eradicate established biofilms of these two bacteria at 32 μg/mL. The inhibitory activities of the surfactants over key enzymes enrolled in the skin repairing processes (collagenase, elastase and hyaluronidase) were evaluated. They exhibited moderate anti-collagenase activity while the activity of hyaluronidase was boosted by the presence of these surfactants. These biological properties render these gemini arginine-based surfactants as perfect promising candidates for pharmaceutical and biological properties.

Abstract:Glycolipid biosurfactants are natural amphiphiles and have gained particular interest recently in their biodegradability, diversity, and bioactivity. Microbial infection has caused severe morbidity and mortality and threatened public health security worldwide. Glycolipids have played an important role in combating many diseases as therapeutic agents depending on the self-assembly property, the anticancer and anti-inflammatory properties, and the antimicrobial properties, including antibacterial, antifungal, and antiviral effects. Besides, their role has been highlighted as scavengers in impeding the biofilm formation and rupturing mature biofilm, indicating their utility as suitable anti-adhesive coating agents for medical insertional materials leading to a reduction in vast hospital infections. Notably, glycolipids have been widely applied to the synthesis of novel antimicrobial materials due to their excellent amphipathicity, such as nanoparticles and liposomes. Accordingly, this review will provide various antimicrobial applications of glycolipids as functional ingredients in medical therapy.
Amelioration of lipopeptide biosurfactants for enhanced antibacterial and biocompatibility through molecular antioxidant property by methoxy and carboxyl moieties

Abstract:Biosurfactants having surface-active biomolecules have been the cynosure in environment research due to their vast application. However, the lack of information about their low-cost production and detailed mechanistic biocompatibility limits the applicability. The study explores techniques for the production and design of low-cost, biodegradable, and non-toxic biosurfactants from Brevibacterium casei strain LS14 and excavates the mechanistic details of their biomedical properties like antibacterial effects and biocompatibility. Taguchi's design of experiment was used to optimize for enhancing biosurfactant production by optimal factor combinations like Waste glycerol (1%v/v), peptone (1%w/v), NaCl 0.4% (w/v), and pH 6. Under optimal conditions, the purified biosurfactant reduced the surface tension to 35 mN/m from 72.8 mN/m (MSM) and a critical micelle concentration of 25 mg/ml was achieved. Spectroscopic analyses of the purified biosurfactant using Nuclear Magnetic Resonance suggested it as a lipopeptide biosurfactant. The evaluation of mechanistic antibacterial, antiradical, antiproliferative, and cellular effects indicated the efficient antibacterial activity (against Pseudomonas aeruginosa) of biosurfactants due to free radical scavenging activity and oxidative stress. Moreover, the cellular cytotoxicity was estimated by MTT and other cellular assays revealing the phenomenon as the dose-dependent induction of apoptosis due to free radical scavenging with an LC50 of 55.6 ± 2.3 mg/ml.

Abstract:New hexadecylpiperidinium surfactants, containing one or two butylcarbamate fragments, were synthesized. The antimicrobial activity, toxicity, aggregation behavior in aqueous solutions, and solubilization capacity of these surfactants towards the hydrophobic drug ibuprofen were characterized. These surfactants demonstrated a high antimicrobial activity against a wide range of pathogenic bacteria, including both Gram-positive and Gram-negative strains, as well as fungi. By forming mixed-micellar compositions of the cationic surfactant 1-CB(Bu)-P-16 and the nonionic surfactant Brij®35, highly functional and low-toxic formulations were obtained. Furthermore, the transition from mixed micelles to niosomes was accomplished, enhancing their potential as drug delivery systems. Niosomes were found to be less toxic compared to mixed micelles, while also increasing the solubility of ibuprofen in water. The modification of niosomes with cationic surfactants made it possible to increase the stability of the system and improve the solubility of the drug. The data obtained indicate that these new carbamate-containing hexadecylpiperidinium surfactants have significant potential in biomedical applications, particularly in the formulation of advanced drug delivery systems.

Abstract:Background: This work proposes the development of new vesicular systems based on anesthetic compounds (lidocaine (LID) and capsaicin (CA)) and antimicrobial agents (amino acid-based surfactants from phenylalanine), with a focus on physicochemical characterization and the evaluation of antimicrobial and cytotoxic properties. Method: Phenylalanine surfactants were characterized via high-performance liquid chromatography (HPLC) and nuclear magnetic resonance (NMR). Different niosomal systems based on capsaicin, lidocaine, cationic phenylalanine surfactants, and dipalmitoyl phosphatidylcholine (DPPC) were characterized in terms of size, polydispersion index (PI), zeta potential, and encapsulation efficiency using dynamic light scattering (DLS), transmitted light microscopy (TEM), and small-angle X-ray scattering (SAXS). Furthermore, the interaction of the pure compounds used to prepare the niosomal formulations with DPPC monolayers was determined using a Langmuir balance. The antibacterial activity of the vesicular systems and their biocompatibility were evaluated, and molecular docking studies were carried out to obtain information about the mechanism by which these compounds interact with bacteria. Results: The stability and reduced size of the analyzed niosomal formulations demonstrate their potential in pharmaceutical applications. The nanosystems exhibit promising antimicrobial activity, marking a significant advancement in pharmaceutical delivery systems with dual therapeutic properties. The biocompatibility of some formulations underscores their viability. Conclusions: The proposed niosomal formulations could constitute an important advance in the pharmaceutical field, offering delivery systems for combined therapies thanks to the pharmacological properties of the individual components.

Abstract:Biosurfactants (BSs) are microbial compounds that have emerged as potential alternatives to chemical surfactants due to their multifunctional properties, sustainability and biodegradability. Owing to their amphipathic nature and distinctive structural arrangement, biosurfactants exhibit a range of physicochemical properties, including excellent surface activity, efficient critical micelle concentration, humectant properties, foaming and cleaning abilities and the capacity to form microemulsions. Furthermore, numerous biosurfactants display additional biological characteristics, such as antibacterial, antifungal and antiviral effects, and antioxidant, anticancer and immunomodulatory activities. Over the past two decades, numerous studies have explored their potential applications, including pharmaceuticals, cosmetics, antimicrobial and antibiofilm agents, wound healing, anticancer treatments, immune system modulators and drug/gene carriers. These applications are particularly important in addressing challenges such as antimicrobial resistance and biofilm formations in clinical, hygiene and therapeutic settings. They can also serve as coating agents for surfaces, enabling antiadhesive, suppression, or eradication strategies. Not least importantly, biosurfactants have shown compatibility with various drug formulations, including nanoparticles, liposomes, micro- and nanoemulsions and hydrogels, improving drug solubility, stability and bioavailability, and enabling a targeted and controlled drug release. These qualities make biosurfactants promising candidates for the development of next-generation antimicrobial, antibiofilm, anticancer, wound-healing, immunomodulating, drug or gene delivery agents, as well as adjuvants to other antibiotics. Analysing the most recent literature, this review aims to update the present understanding, highlight emerging trends, and identify promising directions and advancements in the utilization of biosurfactants within the pharmaceutical and biomedical fields.

Abstract:Hexadecylpyridinium chloride (CTPy), hexadecyltrimethylammonium chloride (CTAC), and thymol, typically found in mouthwash, exhibit inherent antimicrobial properties. When combined, these cationic surfactants and thymol can synergistically enhance these properties. Moreover, these molecules possess the necessary structure to form wormlike micelles (WLM) when thymol interacts with the cationic surfactants. This occurs because the aromatic rings spontaneously interpose themselves between the heads of the cationic surfactants, maintaining the hydroxyl group at the micelle interface. This arrangement minimizes repulsion between the surfactant heads, reducing aggregate curvature and leading to the formation of elongated aggregates. This preferential insertion of aromatic molecules into the micellar environment markedly increases thymol's solubility in aqueous solutions. Depending on the concentration of the components, WLM chains entangle, resulting in highly viscous systems. Consequently, these supramolecular gel-like structures enable prolonged disinfection of inclined or even vertical surfaces due to their slow sliding properties. The rheological properties of WLM, characterized using Cryo-TEM, and the phase diagram depicting thymol interactions with the two cationic surfactants, have been investigated. Additionally, the antimicrobial efficacy of a WLM formulation has been tested against various microbes.

Abstract:The spread of antimicrobial-resistant pathogens typically existing in biofilm formation and the recent COVID-19 pandemic, although unrelated phenomena, have demonstrated the urgent need for methods to combat such increasing threats. New avenues of research for natural molecules with desirable properties to alleviate this situation have, therefore, been expanding. Biosurfactants comprise a group of unique and varied amphiphilic molecules of microbial origin capable of interacting with lipidic membranes/components of microorganisms and altering their physicochemical properties. These features have encouraged closer investigations of these microbial metabolites as new pharmaceutics with potential applications in clinical, hygiene and therapeutic fields. Mounting evidence has indicated that biosurfactants have antimicrobial, antibiofilm, antiviral, immunomodulatory and antiproliferative activities that are exploitable in new anticancer treatments and wound healing applications. Some biosurfactants have already been approved for use in clinical, food and environmental fields, while others are currently under investigation and development as antimicrobials or adjuvants to antibiotics for microbial suppression and biofilm eradication strategies. Moreover, due to the COVID-19 pandemic, biosurfactants are now being explored as an alternative to current products or procedures for effective cleaning and handwash formulations, antiviral plastic and fabric surface coating agents for shields and masks. In addition, biosurfactants have shown promise as drug delivery systems and in the medicinal relief of symptoms associated with SARS-CoV-2 acute respiratory distress syndrome.

Abstract:Chlorhexidine dodecyl sulfate (CHX-DS) was synthesized and characterized via single-crystal X-ray diffraction (SC-XRD), 1H nuclear magnetic resonance (NMR), 1H nuclear Overhauser effect spectroscopy (NOESY), and attenuated total reflectance Fourier-transform infrared spectroscopy (ATR-FTIR). The solid-state structure, comprising a 1:2 stoichiometric ratio of chlorhexidine cation [C22H30Cl2N10]2+ to dodecyl sulfate anion [C12H25SO4]-, is the first report of chlorhexidine isolated with a surfactant. CHX-DS exhibits broad-spectrum antibacterial activity and demonstrates superior efficacy to reduce bacteria-generated volatile sulfur compounds (VSCs) as compared to chlorhexidine gluconate (CHG). The minimum inhibitory concentrations (MICs) of CHX-DS is 7.5, 2.5, 2.5, and 10 μM for S. enterica, E. coli, S. aureus, and S. mutans, respectively Furthermore, MIC assays for E. coli and S. mutans demonstrate that CHX-DS and CHX exhibit a statistically significant efficacy enhancement in the 2.5 μM treatment as compared to CHG. CHX-DS was incorporated into SBA-15, a mesoporous silica nanoparticle (MSN) framework, and its release was qualitatively measured via UV-Vis in aqueous media which suggests its potential as an advanced functional material for drug delivery applications.

Abstract:Four novel nonionic 1-alkylaminoglycerol Gemini surfactants, represented by m-s-m ( m = C 8 and C 12 , s = 3 and 4), were synthesized through a two-step reaction in high yields, and confirmed by 1 H NMR, 13 C NMR, and HRMS. The synthesized Gemini surfactants exhibited low CMC (1.41-5.25 mmol/L), excellent surface-tension-reducing ability (25.45-26.83 mN/m), and wetting ability on hydrophobic interface. Gemini surfactants in aqueous solutions were in stable nanoscale dispersion, and exhibited strong and long-lasting foaming ability and emulsifying abilities for benzene/water and triacylglyceride/water systems. Especially, 12-4-12 solution displayed excellent tolerance to wide pH range and high salinity. Biological evaluation demonstrated that these Gemini surfactants, except for 8-4-8 , showed significant antibacterial activities against Staphylococcus aureus and Escherichia coli at 25 and 50 μg/mL. Hence, these Gemini surfactants have the potential to be used in a varieties of fields as highly efficient agents.

Thiacalixarene based quaternary ammonium salts as promising antibacterial agents

Abstract:The search for new antibacterial and antiseptic drugs is an urgent problem due to the resistance of microorganisms to existing drugs. In this work, for the first time, the design of antibacterial and bactericidal agents based on quaternary ammonium compounds on thiacalixarene macrocyclic platform was proposed and implemented. A series of tetrasubstituted quaternary ammonium salts with different nature and length of the substituent (-N+(CH3)2R, R = CH2Ph, CnH2n+1, n = 1, 4, 8, 10) based on p-tert-butylthiacalix[4]arene in cone and 1,3-alternate conformations was obtained with excellent yields. The obtained compounds have a high antibacterial effect against Gram-positive (S. aureus, S. epidermidis, B. subtilis) bacteria comparable with commercial antiseptics chlorhexidine, miramistin and benzalkonium chloride. It was found that quaternary ammonium derivatives of thiacalix[4]arene in 1,3-alternate conformation more effectively inhibit the growth of the tested bacterial strains in comparison with compounds in cone conformation. Cytotoxicity studies on human skin fibroblast (HSF) cells demonstrated that all compounds were less toxic compared to reference drugs. The different type of interaction of the studied compounds with model DPPC lipid membranes explains different antibacterial activity and cytotoxicity of compounds. The compounds in cone conformation are adsorbed on the DPPC vesicles membrane surface, while the incorporation of lipophilic alkyl fragments of macrocycles in 1,3-alternate conformation into the membrane leads to "clumping" of DPPC vesicles. It was shown the saving of antibacterial activity of thiacalixarene derivatives in 1,3-alternate conformation on Gram-positive clinical strains. The obtained results allow viewing the described thiacalixarene based quaternary ammonium compounds as promising molecules in the development of the new antibacterial agents.
Enzymatic Synthesis and Biological Evaluation of Glycolipids as Potential Antibacterial, Antibiofilm and Antiquorum Sensing Agents

Abstract:Glycolipids are becoming as important class of biosurfactants with relevant antibacterial and antibiofilm properties. In this study, the enzymatic synthesis of glycolipids is reported, achieved through the regioselective transesterification of different sugars (fucose, arabinose, fructose, glucose, mannose, galactose, N -acetylglucosamine, N -acetylgalactosamine and maltose) with vinyl laurate. The biocatalyst employed for this process is lipase from Pseudomonas stutzeri . The biocatalytic reaction proved to be very effective for the regioselective synthesis of lauroyl monoesters and diesters. Docking and molecular dynamic simulations studies were employed to explain enzyme regioselectivity and its ability to accommodate sugar head, providing insights the observed behavior. The antibacterial, antibiofilm and anti-quorum sensing activities of the synthesized glycolipids were evaluated against a target panel of Gram-positive and Gram-negative bacteria. While 6- O -lauroyl-Man ( 6 ) and 4- O -lauroyl-Rha ( 1e ) exhibited the highest antibacterial activity in the monoester series, lauroyl diesters ( 4 , 1,6- O -dilauroyl-Fru and 7 , 2,6- O -dilauroyl-Gal) demonstrated increased activity compared to monoesters and displayed greater promise than the positive control. Regarding antibiofilm properties, 4- O -lauroyl-Rha ( 1e ), 1,6- O -dilauroyl-Fru ( 4 ) and 2,6- O -dilauroyl-Gal ( 7 ) presented higher activity than the positive control on Gram-positive bacteria. Compounds 6- O -lauroylmannose ( 6 ), 6- O -lauroyl-GlcNAc ( 8 ) and 6- O -lauroyl-GalNAc ( 9 ) showed high antibiofilm and anti-quorum sensing activities on Gram-negative bacteria, surpassing even the positive control. Docking studies provided a comprehensive understanding of the mechanisms behind their efficacy. In conclusions, a a sustainable strategy has been performed to prepare glycolipids, offering promising prospects as antibacterial, antibiofilm and anti-quorum sensing agents. These glycolipids could serve as a viable alternative to conventional antibiotics in the future.

Abstract:Nine novel cationic gemini surfactants containing both amide and ester groups were synthesized through a three-step reaction. These surfactants have the general formula of CnH2n+1OOCCH2N+(CH2)2-(CH2)3-NHOC-(CH2)m-CONH-(CH2)3-N+(CH2)2CH2COOCnH2n+1 (with n = 8, 10, 12 and m = 2, 3, 4), named BEQA-n, PEQA-n and HEQA-n (B, P, H correspond to surfactants of m = 2, 3, 4 and n = 8, 10, 12, respectively). Chemical structures, surface activity, cleavable properties, electrolytes tolerance, foam properties and antibacterial activity of these surfactants were successively investigated. The results show that the surfactants have lower critical micelle concentration (CMC) than traditional monomer surfactants. The values of CMC and the degree of counter ion dissociation (α) depend on the lengths of hydrophobic chain and spacer. The values of Krafft temperatures (Kt < 0 °C) and the standard Gibbs free energy (∆G0mic < 0) indicate that all surfactants have good water solubility and micelle-forming ability. The simultaneous presence of the amide and ester groups makes these surfactants have good hydrolytic ability in both basic and the acidic solutions. In addition, the surfactants exhibit greater electrolyte tolerance to NaCl than CaCl2. It is worth mentioning that EQA-8 and PEQA-12 have 200 g/L NaCl tolerance. Moreover, all of the surfactants have certain antibacterial activity against Gram-positive bacteria (S. aureus ATCC 25923) and Gram-negative bacteria (E. coli ATCC 25922), and the surfactants with hydrophobic chain length n = 10 have the strongest antibacterial activity.

Abstract:Sophorolipids, glycolipid biosurfactants derived from microorganisms such as Starmerella bombicola , possess distinctive surface-active and bioactive properties, holding potential applications in cosmetics, pharmaceuticals and bioremediation. However, the limited structural variability in wild-type sophorolipids restricts their properties and applications. To address this, metabolic engineering efforts have allowed to create a portfolio of molecules. In this study, we went one step further by chemically modifying microbially produced sophorosides, produced by an engineered S. bombicola. Twenty-four new sophoroside derivatives were synthesized, including sophoroside amines with varying alkyl chain lengths (ethyl to octadecyl) on the nitrogen atom and their corresponding quaternary ammonium salts. Additionally, six different microbially produced glycolipid biosurfactants were hydrogenated to achieve fully saturated lipid tails. These derivatives, along with microbially produced glycolipids and three benchmark biosurfactants (di-rhamnolipids, alkyl polyglucosides, cocamidopropyl betaine), were assessed for antimicrobial activity against bacteria ( Bacillus subtilis, Staphylococcus aureus, Listeria monocytogenes, Escherichia coli, Pseudomonas aeruginosa ) and yeast ( Candida albicans ). Results indicated that microbially produced glycolipids, such as bola sophorosides, acidic sophorolipids and acidic glucolipids exhibit selective antimicrobial activity against the test organisms. Conversely, lactonic sophorolipids, sophoroside amines and quaternary ammonium salts display a broad antimicrobial activity. N-octyl, N-dodecyl and N-octadecyl derivatives exhibit the lowest minimal inhibitory concentrations, ranging from 0.014 to 20.0 mg mL −1 . This study demonstrates the potential synergy of thoughtful biotechnology and targeted chemistry to precisely tailor glycolipid biosurfactants to meet specific requirements across applications.

Abstract:Surfactants are versatile excipients that are commonly employed in diverse pharmaceutical formulations given their broad range of antimicrobial activities. Sulfur-based amino acid surfactants are promising candidates as substitutes for conventional antibacterial agents in light of the current antibiotic resistance crisis. Dodecyl esters of the free amine (SURF1), cationic ester hydrochloride (SURF2), and quaternary ammonium (QUAT3) derivatives of methionine, and the Gemini diester of cystine (GEM4) were synthesized and evaluated for antibacterial activity against Gram-positive and Gram-negative pathogens. All surfactants displayed moderate to high antibacterial activities, particularly on Gram-positive bacteria, with QUAT3 showing the highest activity. Among Gram-negative bacteria, QUAT3, GEM4, and SURF2 were mostly active towards K. pneumoniae with minimum inhibitory concentrations (MIC) ranging from 0.004 to 0.441 mM. QUAT3 and GEM4 displayed a bacteriostatic behavior similar to that of tetracycline, as assessed by broth macrodilution assays. Fluorescence binding studies with 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) revealed that the antibacterial activities could be attributed to a combination of electrostatic and hydrophobic interactions. Bovine serum albumin (BSA) fluorescence and molecular docking studies indicated that SURF1 and QUAT3 interact mainly via van der Waals' forces and hydrogen bonding while SURF2 binds through hydrophobic interactions. QUAT3, which displays broad-spectrum activity, has the potential to combat drug-resistant bacteria.

Abstract:The widespread problem of resistance development in bacteria has become a critical issue for modern medicine. To limit that phenomenon, many compounds have been extensively studied. Among them were derivatives of available drugs, but also alternative novel detergents such as Gemini surfactants. Over the last decade, they have been massively synthesized and studied to obtain the most effective antimicrobial agents, as well as the most selective aids for nanoparticles drug delivery. Various protocols and distinct bacterial strains used in Minimal Inhibitory Concentration experimental studies prevented performance benchmarking of different surfactant classes over these last years. Motivated by this limitation, we designed a theoretical methodology implemented in custom fast screening software to assess the surfactant activity on model lipid membranes. Experimentally based QSAR (quantitative structure-activity relationship) prediction delivered a set of parameters underlying the Diptool software engine for high-throughput agent-membrane interactions analysis. We validated our software by comparing score energy profiles with Gibbs free energy from the Adaptive Biasing Force approach on octenidine and chlorhexidine, popular antimicrobials. Results from Diptool can reflect the molecule behavior in the lipid membrane and correctly predict free energy of translocation much faster than classic molecular dynamics. This opens a new venue for searching novel classes of detergents with sharp biologic activity.
Surfactants' Interplay with Biofilm Development in Staphylococcus and Candida

Abstract:The capacity of micro-organisms to form biofilms is a pervasive trait in the microbial realm. For pathogens, biofilm formation serves as a virulence factor facilitating successful host colonization. Simultaneously, infections stemming from biofilm-forming micro-organisms pose significant treatment challenges due to their heightened resistance to antimicrobial agents. Hence, the quest for active compounds capable of impeding microbial biofilm development stands as a pivotal pursuit in biomedical research. This study presents findings concerning the impact of three surfactants, namely, polysorbate 20 (T20), polysorbate 80 (T80), and sodium dodecyl sulfate (SDS), on the initial stage of biofilm development in both Staphylococcus aureus and Candida dubliniensis. In contrast to previous investigations, we conducted a comparative assessment of the biofilm development capacity of these two taxonomically distant groups, predicated on their shared ability to reduce TTC. The common metabolic trait shared by S. aureus and C. dubliniensis in reducing TTC to formazan facilitated a simultaneous evaluation of biofilm development under the influence of surfactants across both groups. Our results revealed that surfactants could impede the development of biofilms in both species by disrupting the initial cell attachment step. The observed effect was contingent upon the concentration and type of compound, with a higher inhibition observed in culture media supplemented with SDS. At maximum concentrations (5%), T20 and T80 significantly curtailed the formation and viability of S. aureus and C. dubliniensis biofilms. Specifically, T20 inhibited biofilm development by 75.36% in S. aureus and 71.18% in C. dubliniensis, while T80 exhibited a slightly lower inhibitory effect, with values ranging between 66.68% (C. dubliniensis) and 65.54% (S. aureus) compared to the controls. Incorporating these two non-toxic surfactants into pharmaceutical formulations could potentially enhance the inhibitory efficacy of selected antimicrobial agents, particularly in external topical applications.

Abstract:Polysiloxane containing tertiary amino group (P(SiO)m(SiOTA)n) with different molecular weight was synthesized through equilibrium polymerization of octamethylcyclotetrasiloxane (D4) and hydrolysis of N,N-diethyl-aminopropyl-methyldimethoxysilane (TAMDESi) in the presence of potassium hydroxide (KOH) as catalyst. After that, polysiloxane quaternary ammonium salt containing epoxy group (P(SiO)m(SiOQAEp)n) with different molecular weight and cationic concentration was obtained from P(SiO)m(SiOTA)n and epichlorohydrin by quaternization. It was shown that P(SiO)m(SiOQAEp)n surfactant exhibited low surface tension (γCMC < 30.00 mN/m) at the critical micellization concentration. The thermodynamic parameters of micellization (ΔG0m, ΔH0m, ΔS0m) indicated that the micellization of P(SiO)m(SiOQAEp)n surfactant was enthalpy-driven. In particular, it was found that the glass transition temperature (Tg) of P(SiO)m(SiOQAEp)n varied in the range of −15 ∼ –70 °C according to the molecular weights, and the thermal decomposition temperature was higher than 220 °C. This type of surfactant had excellent low temperature flexibility as well as good chemical and thermal stability. The antimicrobial activity was evaluated against Gram-negative (Escherichia coli) and Gram-positive bacteria (Staphylococcus aureus), respectively. The minimum bactericidal concentration (MBC) values against S. aureus and E. coli were below 1.5–2.8 × 10−5 mol/L, which indicated that P(SiO)m(SiOQAEp)n had strong antibacterial activity.

Abstract:The emergence of multi-drug resistant pathogens is a major public health problem, leading us to rethink and innovate our bacterial control strategies. Here, we explore the antibiofilm and antivirulence activities of nineteen 6-polyaminosterol derivatives (squalamine-based), presenting a modulation of their polyamine side chain on four major pathogens, i.e., carbapenem-resistant A. baumannii (CRAB) and P. aeruginosa (CRPA), methicillin-resistant S. aureus (MRSA), and vancomycin-resistant E. faecium (VRE) strains. We screened the effect of these derivatives on biofilm formation and eradication. Derivatives 4e (for CRAB, VRE, and MRSA) and 4f (for all the strains) were the most potent ones and displayed activities as good as those of conventional antibiotics. We also identified 11 compounds able to decrease by more than 40% the production of pyocyanin, a major virulence factor of P. aeruginosa. We demonstrated that 4f treatment acts against bacterial infections in Galleria mellonella and significantly prolonged larvae survival (from 50% to 80%) after 24 h of CRAB, VRE, and MRSA infections. As shown by proteomic studies, 4f triggered distinct cellular responses depending on the bacterial species but essentially linked to cell envelope. Its interesting antibiofilm and antivirulence properties make it a promising a candidate for use in therapeutics.
