Effect of different doses of cationic bovine serum albumin on producing rat models of membranous nephropathy

Abstract:Background: Studies on suitable dosage of cationic bovine serum albumin (C-BSA) and models comparison of different dosages of C-BSA in creating animal models of membranous nephropathy are few. Objective: To investigate the effect of different doses of C-BSA on producing rats model of membranous nephropathy, and to discuss a suitable dose of C-BSA. Design, time and setting: The randomized control animal experiment was performed at the Pathology Laboratory and Infection Immunity Laboratory of Affiliated Hospital of Luzhou Medical College from February 2007 to January 2008. Materials: Forty healthy male Sprague-Dawley (SD) rats were obtained from Animal Center of Luzhou Medical College. Mr66 000 natural bovine serum albumin was supplied by Sinopharm Chemcial Reagent Co.Ltd (Chengdu branch), China. Interventions: Forty rats were randomized into a normal control group, C-BSA 3, 10, 16 mg/kg groups, with 10 rats in each group. After 1 week of pre-immunity, rats in the C-BSA 3, 10, 16 mg/kg groups were injected with C-BSA 3, 10, 16 mg/kg in the tail vein on other day. Rats in the normal control group were treated with the same volume of saline, for successively 4 weeks. Main outcome measures: Rats were sacrificed at the end of the fourth week of formal immunity. Kidney histological changes were observed under a light microscope, an electron microscope and immunofluorescence. Urine protein, serum creatinine, urea nitrogen, albumin, cholesterin, triglyceride and creatinine clearance were detected 24 hours before sacrificing rats from each group. Results: Three rats died in the C-BSA 10 and 16 mg/kg groups each, and randomly supplemented. Under the light microscope, glomerular capillary wall uniformly thickened in each model group, especially in the C-BSA 16 mg/kg group. Under the electron microscope, bulk electron-dense deposit under glomerular basement membrane endepidermis in the C-BSA 16 mg/kg group, with obvious rete pegs. By immunofluorescence in the C-BSA 10 and 16 mg/kg groups, immunoglobulin G (IgG) and complement (C3) were along glomerular capillary wall, showing dazzling diffuse granulo-deposit, which was obvious compared with the C-BSA 3 mg/kg group. Cholesterin and urine protein concentrations were significantly higher in the C-BSA 10 and 16 mg/kg groups compared with the normal control group and C-BSA 3 mg/kg group (P < 0.05). Plasma urea nitrogen levels were significantly higher in the C-BSA 16 mg/kg group compared with the normal control group, C-BSA 3, and 10 mg/kg groups (P < 0.05). Conclusion: Thickness of glomerular basement membrane is various in rat models with different doses of C-BSA. C-BSA 16 mg/kg induces an optimal outcome, with stable models, typical clinical manifestation and high repeatability.
