Abstract:Objective: This phase Ia study was designed to assess the pharmacokinetic (PK) characters of free vincristine (F-VCR, refer to as non-liposomal VCR and VCR released from liposome) and total vincristine (T-VCR, the sum of both liposomal VCR and F-VCR), urinary excretion and safety of intravenous administration of vincristine sulfate liposomes injection (VSLI) in Chinese patients with malignant lymphoma and compare the results with those for conventional vincristine sulfate injection (VSI). Methods: In the phase Ia, randomized, open-label, two sequence cross-over study, patients from one group were exposed to treatment 1 including cytoxan (cyclophosphamide power injection), hydroxyrubicin (adriamycin power injection), oncovin (VSI), and prednisone tablets (standard CHOP scheme) before crossed over to treatment 2 (modified CHOP scheme in which VSI was replaced with VSLI). Patients from another group received treatments in reverse order. Results: In this phase Ia study, a total of eight subjects participated. VCR elimination from the circulation after injection of VSLI was characterized by a significantly increased maximum concentration (Cmax, 86.6 ng/mL) and plasma area under the plasma concentration-time curve from zero to infinity (AUC0-Inf, 222.1 ng/mL h), markedly decreased distribution volume (Vz, 224.1 L) and plasma clearance (CL, 8.9 L/h) compared to lower Cmax (26.6 ng/mL) and AUC0-Inf (95.1 ng/mL h), larger Vz (688.8 L) and CL (22.1 L/h) for VSI. The small proportion of F-VCR following infusion of VSLI in circulation was reflected by very low Cmax (1.8 ng/mL) and AUC0-Inf (50.5 ng/mL h). Less than 3% of the administered dose of VSLI was excreted in urine and the extent was similar to that for VSI. The elimination percentage of 40-21-14% for VSI changed to 6.2-24-39% for VSLI at intervals of 0-5, 5-13 and 13-25 h, respectively. Significant difference of toxicity between VSLI and VSI was not observed. Conclusion: VSLI exhibits higher AUC0-Inf of T-VCR, lower CL and Vz compared with VSI. VSLI was well tolerated, maybe due to the markedly decreasing AUC0-Inf of F-VCR. The majority of VCR was enveloped in liposome and VCR was released gradually from liposome following injection of VSLI. Liposomal encapsulation of VCR does not alter the route and extent of VCR excretion in urine.

Preparation of Functional Vincristine Liposomes for Treatment of Invasive Breast Cancer

Abstract:Clinical treatment of cancers usually adopts a comprehensive strategy, which consists of surgical treatment, radiotherapy, chemotherapy, and biotherapy. Among which, chemotherapy plays a crucial role. However, conventional chemotherapy fails to obtain a satisfactory efficacy, including unfavorable pharmacokinetic properties and systemic toxicities, which are caused by the direct delivery of free drugs into the circulatory system. This chapter focuses on the liposomal formulations of encapsulated vinca alkaloid and mainly introduces the preparation and characterization of functional vincristine liposomes, which were designed to treat the invasive breast cancer along with eliminating the vasculogenic mimicry channels. In this liposomal drug delivery system, vincristine was used as an anticancer agent, dasatinib was used as a promising regulator, and the targeting molecular c(RGDyK) was modified on the surface of the liposomes. The functional liposomes were round in shape and displayed a smooth surface with approximately 100 nm in diameter. The investigations were performed on invasive breast cancer MDA-MB-231 cells and MDA-MB-231 xenografts in nude mice. The functional vincristine plus dasatinib liposomes displayed a prolonged circulation time in blood system and an increased accumulation in tumor tissue, resulting in a robust overall anticancer efficacy.
Formulation and Characterization of Spray-Dried Powders Containing Vincristine-Liposomes for Pulmonary Delivery and Its Pharmacokinetic Evaluation From In Vitro and In Vivo

Abstract:Vincristine (VCR) has been used in the treatment of lung cancer. To improve its efficacy, the designs of elevating lung exposure to drug and decreasing the clearance with extended time were brought out. Pulmonary delivery is regarded as a good choice in pulmonary diseases treatment. Spray-drying is a technology for the preparation of drugs that can be delivered to lung via a dry powder inhaler. The results showed an appropriate particle size and shape for the pulmonary delivery. The aerosol behaved a sustained-release profile while VCR solution released rapidly within 10 h. The antitumor activity was characterized by 3-(4, 5-dimethylthiazol-2-yl) -2, 5-diphenyltetrazolium bromide assay, and half maximal inhibitory concentration values of VCR-liposomes spray-dried powder were 24.42 ± 1.88 nM and 55.28 ± 4.76 nM in MCF-7 and A549 cells, respectively. Compared with the free VCR, the aerosol performed better pharmacokinetic behavior: increased maximum concentration (630.8%) and systemic exposure (429.6%) and decreased elimination half-life (81.1%). The clearance was decreased by 83.2%. Comprehensively, the pulmonary delivery seemed to be a recommendable way to effectively treat the pulmonary disease.

Abstract:Vincristine (VCR) is mainly used to treat acute lymphocytic leukemia, Hodgkin and non-Hodgkin lymphoma in clinic with definite therapeutic effect. But the obvious neurotoxicity and local stimulation of which limit its clinic use. In order to increase the lymph targeting to enhance the curative effect and to lower the adverse reaction of VCR, the VCR loaded transfersomes (VCR-T) were prepared with dry-film and ultrasonic dispersing methods, and the corresponding pharmaceutical properties, pharmacokinetical characteristics and the targeting ability were studied. The average particle size of VCR-T prepared was 63 nm with an entrapment ratio of 59%. The in vitro transdermal research with modified Franz cell showed that VCR-T permeated through the skin in accordance with polynomial equation, and with an accumulation permeation percentage of 67.4% up to 12 h. An HPLC method was utilized to determine the pharmacokinetics and tissue distribution of VCR. Compared with the iv injection of VCR solution, the retention time of VCR in blood was extended by 12 times with VCR-T, and the targeting index in rat lymph was increased by 2.75 times. As a result, transfersomes could penetrate the skin and enter into the systemic circulation carrying VCR with good lymph targeting ability, which makes it probably a new lymphtic targeting drug delivery system.
Characterization of highly stable liposomal and immunoliposomal formulations of vincristine and vinblastine

Abstract:Purpose: Liposome and immunoliposome formulations of two vinca alkaloids, vincristine and vinblastine, were prepared using intraliposomal triethylammonium sucroseoctasulfate and examined for their ability to stabilize the drug for targeted drug delivery in vivo. Methods: The pharmacokinetics of both the encapsulated drug (vincristine or vinblastine) and liposomal carrier were examined in Sprague Dawley rats, and the in vivo drug release rates determined. Anti-HER2 immunoliposomal vincristine was prepared from a human anti-HER2/neu scFv and studied for targeted cytotoxic activity in cell culture, and antitumor efficacy in vivo. Results: Nanoliposome formulations of vincristine and vinblastine demonstrated similar pharmacokinetic profiles for the liposomal carrier, but increased clearance for liposome encapsulated vinblastine (t (1/2) = 9.7 h) relative to vincristine (t (1/2) = 18.5 h). Immunoliposome formulations of vincristine targeted to HER2 using an anti-HER2 scFv antibody fragment displayed a marked enhancement in cytotoxicity when compared to non-targeted liposomal vincristine control; 63- or 253-fold for BT474 and SKBR3 breast cancer cells, respectively. Target-specific activity was also demonstrated in HER2-overexpressing human tumor xenografts, where the HER2-targeted formulation was significantly more efficacious than either free vincristine or non-targeted liposomal vincristine. Conclusions: These results demonstrate that active targeting of solid tumors with liposomal formulations of vincristine is possible when the resulting immunoliposomes are sufficiently stabilized.
[Effects of organic manures on CO2 and CH4 fluxes of farmland].

Abstract:This paper studied the effects of chicken manure, swine waste and cattle manure on the CO2 and CH4 fluxes of a farmland planted with summer maize. The results showed that the CO2 flux had the same trend under different organic manure application, which was influenced by soil temperature and soil water content. The flux was significantly related with air temperature, soil surface temperature and soil temperature (P < 0.05). When soil temperature was not a limiting factor, soil water content was significantly related with soil CO2 flux. At most maize growth stages, soil was a sink of CH4. The variation of CH4 source-sink was influenced by environmental factors, but the correlation was not significant. The mean seasonal flux of CO2 was from 0.5124 to 0.8518 g x m(-2) x h(-1), and that of CH4 was from - 0.0068 to - 0.0484 mg x m(-2) x h(-1). Compared with CK2, maize planting and organic manure application enhanced CO2 emission. The application of organic manures inhibited the CH4 uptake by soil, and higher application rate had a higher inhibitory effect.

Abstract:Objective To prepare the PEG-PLGA nanoparticles loaded with vincristine sulfate(VCR-loaded PEG-PLGA-NPs) and evaluate their quality.Methods VCR-loaded PEG-PLGA-NPs were prepared by the double emulsion solvent evaporation method.The main experimental factors,which influenced the physical and chemical properties of the nanoparticles,were investigated and optimized.Results Under optimal conditions,the VCR-loaded PEG-PLGA-NPs had an average diameter of 135.9 nm with narrow size distribution.The encapsulation efficiency was 68.2%,while the drug loading capacity was 8.34%.In vitro,VCR was released from the PEG-PLGA-NPs sustainedly for more than 13 days with the total amount of 81%.Moreover,the VCR-loaded PEG-PLGA-NPs were relatively stable,which was confirmed by the stability testing.Conclusion The VCR-loaded PEG-PLGA-NPs are a promising nano drug with controlled release,which can be applied widely.

Abstract:Recently, developing drug delivery systems exhibiting controlled drug release at the tumor sites emerged as an attractive option for enhancing anticancer therapeutic efficacy. It seems, however, unlikely that single agent therapies will prove effective enough against the myriad cells present within the malignancy. Therefore, next generation nanotherapeutics must not only find their way to the solid tumor but also must effectively destroy the diverse populations of cells promoting tumor growth. Nanoliposomes offer an important advantage in the delivery of a combination of drugs acting synergistically in cancer treatment. They allow controlling the pharmacokinetics and biodistribution of the drugs by uniform time and spatial co-delivery of the agents. However, successful translation of such complex formulations into the clinic relies on understanding critical physicochemical characteristics. These include: liposomes' membrane phase and dynamics, size distribution, state of encapsulated drug, internal environment of liposome, state of grafted polyethylene glycol at the liposome surface, and in-vivo drug release rate. They determine the pharmacokinetics of the formulation and the bioavailability of the drugs. We encapsulated the combination of vincristine (VCR) and topotecan (TPT) in the same nanoliposome (LipoViTo). Our in-vitro and in-vivo characterization of LipoViTo provides an explanation for the good therapeutic efficacy that was previously reported by us. Moreover, we have described how to study these critical features for a two-drug in one nanoliposome formulation. This characterization is an important step for a rational clinical development and for how to ensure liposome product quality of LipoViTo and other liposomal formulations alike.
Selection of transdermal drug delivery system for vincristrine and research on the transdermal mechanism

Abstract:Vincristine loaded transfersomes (VCR-T) containing different surfactants and vincristine loaded chitosan-nanoparticles (VCR-CS-NPs) were prepared respectively. And their transdermal effects were compared through transdermal experiment in vitro. Besides, DSC was used to study factors affecting the transdermal effects. The entrapment efficiency of VCR-T varied from 50%~80% with an average diameter of 90 nm. And the entrapment efficiency of VCR-CS-NPs was 50% with an average diameter of 200 nm. Both of the carriers were spherical and smooth under transmission electron microscope. The results of transdermal experiment in vitro showed that VCR-T containing Brij78 was the best transdermal drug delivery system for vincristine, which is concerned with the interaction between Brij78 and carrier based on the proof of DSC scan. In contrast, VCR-CS-NPs could not penetrate skin efficiently, which probably has something to do with the larger particle sizes.
Preparation and in vitro evaluation of vincristine-loaded superparamagnetic iron oxide nanoparticles

Abstract:Vincristine loaded superparamagnetic iron oxide (VCR-SPIO) nanoparticles were prepared by self-assembly process. Phenanthroline spectrophotometry and HPLC were used respectively to determine the concentration of iron and vincristine of the pharmaceutical preparation. Encapsulation efficiency was calculated. The morphology and size of the preparation were characterized by transmission electron microscopy (TEM) and dynamic light scattering (DLS). Vibrating sample magnetometer (VSM) was used to detect saturation magnetization, and magnetization curve was drawn. Human leukemia K562 cells were cultured in the presence of vincristine sulfate and VCR-SPIO nanoparticles. Viability of cells was determined by CCK-8. It was found that encapsulation efficiency was (81.2 ± 1.5) %. The concentration of iron was (104 ± 1.4) μg/mL. The average particle diameter of the preparation was (83 ± 1.2) nm. VCR-SPIO nanoparticles had superparamagnetic effect with the saturation magnetization of 56.3 A·m 2/kg. VCR-SPIO nanoparticles showed higher cytotoxicity on K562 cells. It was shown in this study that the novel multifunctional superparamagnetic iron oxide nanoparticles as a novel drug delivery system had the potential for cancer therapy.
Exploring the mechanism of resistance to vincristine in breast cancer cells using transcriptome sequencing technology

Abstract:Breast cancer has replaced lung cancer as the leading cancer globally, but various chemotherapy drugs for breast cancer are prone to resistance, especially in patients with distant metastases who are susceptible to multiple chemotherapy drug resistance often leading to treatment failure. Vincristine (VCR) is an alkaloid extracted from Catharanthus roseus, and is often used in combination with other chemotherapy drugs to treat various types of cancer, including breast cancer. Research on the development of resistance to VCR has been carried out using transcriptome sequencing technology. Firstly, gradient increase of VCR concentration was used to produce a VCR-resistant breast cancer cell line. Mechanistically, RNA was extracted from the VCR-resistant breast cancer cell line, and the transcriptome was sequenced. Further analysis showed changes in the expression levels of various genes in the aforementioned VCR-resistant breast cancer cell line. Meanwhile, the analysis of splicing events also indicated a change in variable splicing events. Further validation showed that the expression levels of multiple genes, including interleukin-1β, were altered in the VCR-resistant breast cancer cell line, and these gene expression changes were related to VCR resistance. The results of the present study provide a theoretical basis for exploring the mechanism of VCR resistance clinically.
Biodegradable Self-Assembled Micelles Based on MPEG-PTMC Copolymers: An Ideal Drug Delivery System for Vincristine

Abstract:Despite advantageous properties, micelles using methoxy poly(ethylene glycol)-poly(trimethylene carbonate) (MPEGPTMC) have not been widely studied. In this work, we aim to develop a novel vehicle for vincristine (VCR) based on a MPEG-PTMC micelle system. MPEG-PTMC with a series of molecular weights were synthesized and screened for the appropriate range for forming stable VCR micelles. The prepared micelles were then characterized in vitro and in vivo . VCR micelles presented high stability and ideal sustained release profile. The passive targeting effect was also enhanced compared with liposomal VCR. These results provide critical data to give the first clues regarding novel VCR micelles which exhibit potential for clinical application.
Batch Normalization in the final layer of generative networks

Abstract:Generative Networks have shown great promise in generating photo-realistic images. Despite this, the theory surrounding them is still an active research area. Much of the useful work with Generative networks rely on heuristics that tend to produce good results. One of these heuristics is the advice not to use Batch Normalization in the final layer of the generator network. Many of the state-of-the-art generative network architectures use this heuristic, but the reasons for doing so are inconsistent. This paper will show that this is not necessarily a good heuristic and that Batch Normalization can be beneficial in the final layer of the generator network either by placing it before the final non-linear activation, usually a $tanh$ or replacing the final $tanh$ activation altogether with Batch Normalization and clipping. We show that this can lead to the faster training of Generator networks by matching the generator to the mean and standard deviation of the target distribution's image colour values.

Abstract:The use of vincristine (VCR) to treat cancer has been limited by its dose-dependent toxicity and development of drug resistance after repeated administrations. In this study, we investigated the mechanism by which quinine hydrochloride (QN) acts as a sensitizer for VCR. Our experiments used three kinds of multidrug-resistant cancer cells and demonstrated that QN worked by inducing intracellular depletion of adenosine triphosphate, increasing adenosine triphosphatase activity, and decreasing P-glycoprotein expression. Based on these results, we designed and prepared a VCR and QN codelivery liposome (VQL) and investigated the effect of coencapsulated QN on the in vitro cytotoxicity of VCR in cells and three-dimensional multicellular tumor spheroids. The antitumor effects of the formulation were also evaluated in multidrug-resistant tumor-bearing mice. The results of this in vivo study indicated that VQL could reverse VCR resistance. In addition, it reduced tumor volume 5.4-fold when compared with other test groups. The data suggest that VQL could be a promising nanoscaled therapeutic agent to overcome multidrug resistance, and may have important clinical implications for the treatment of cancer.

Abstract:Zoledronic acid (ZOL) is a potent amino-bisphosphonate used for the treatment of bone metastases with recently reported antitumor activity. However, its short plasma half-life as well as rapid uptake and accumulation within bone limits the use of ZOL as an antitumor agent. Cationic liposomes have a propensity for localizing in newly formed tumor vessels, improved anti-tumor efficacy associated with impaired function of the tumor microvasculature. At present study, we aim to proposed new delivery systems cationic liposomes to slow ZOL metabolize, so as to improve its effect on tumors. The antiproliferative effects of ZOL-loaded cationic liposomes (ZOL-CLPs), ZOL-loaded neutral liposomes (ZOL-NLPs), and free ZOL in the human lung adenocarcinoma A549 cell lines were evaluated by MTT assay. The advantage of ZOL-CLPs over ZOL-NLPs and free ZOL with regard to antitumor activity in vivo was evaluated in a lung tumor-bearing mouse model. The particle size, zeta potential, and entrapment efficiency (EE%) of the ZOL-CLPs were observed as 106.76 +/- 1.94 nm, +42.37 +/- 2.60 mV, and 38.54 +/- 0.99%, respectively. The antiproliferative effects experiment in vitro, the IC50 of ZOL-CLPs, ZOL-NLPs and free ZOL were 6.38 +/- 1.66, 66.92 +/- 3.75, and 184.65 +/- 12.31 mu g/mL, respectively. Compared with the control group, the tumor inhibition rate of ZOL-CLPs group, ZOL-NLPs group, free ZOL group were 48.18, 36.66, and 2.10%, respectively. Immunohistochemical study showed that the expression of VEGF protein and COX-2 protein were high in control group, medium in ZOL group, low in ZOL-NLPs group, and lower in ZOL-CLPs group. In conclusion, the ZOL-CLPs is a tumor-targeting liposomes which has significant antitumor activity against adenocarcinoma A549 cell lung cancer.

Abstract:Vincristine is a dimer-indo-alkaloid which is extracted from the leaves of Catharanthus roseus. It is effective to treat acute lymphocytic cell leukemia, Hodgkin disease and non-Hodgkin disease clinically. But the severe side effects, such as neurotoxic and tissue damage, limit its application. In this paper, we summarize physical, chemical, pharmacological and pharmacokinetical properties of VCR and advances in decreasing its side effects. In clinic, association with other medication is adopted. In pharmaceutics, people adopt some new methods and technology such as conjugation with the antibody, encapsulation in liposomes or controlled release films.

Abstract:A water-in-oil-in-oil double-emulsion solvent/evaporation method was used to prepare vincristine sulfate (VCR) loaded poly(lactide-co-glycolide) microspheres, and then VCR microspheres were mixed with collagen and (or) chitosan swelling solution and lyophilized to form polymeric films. The films were cross-linked by 0.3% glutaraldehyde (GA). Encapsulation efficiency and release kinetics of VCR microspheres were determined, as well as release kinetics and in vitro degradation of the film. The rate of VCR release from the film submerged in PBS (pH 6.8) and the content were measured by high-performance liquid chromatography (HPLC). The physichemical properties of the film, such as surface morphology, mechanical function, and differential scanning calorimetry, were also measured. VCR was released from the film in a prolonged period and the initial burst release of the film was less significant. In the degradation experiment, the film containing chitosan degraded more slowly than that without chitosan. The films comprising collagen and chitosan could achieve the release kinectics of a relatively constant release. It has a promising future.

Abstract:We express the partition functions of the spanning tree on finite square lattices under five different sets of boundary conditions in terms of a principal partition function with twisted-boundary conditions. Based on these expressions, we derive the exact asymptotic expansions of the logarithm of the partition function for each case. We have also established several groups of identities relating spanning-tree partition functions for the different boundary conditions. We also explain an apparent discrepancy between logarithmic correction terms in the free energy for a two-dimensional spanning-tree model with periodic and free-boundary conditions and conformal field theory predictions. We have obtained corner free energy for the spanning tree under free-boundary conditions in full agreement with conformal field theory predictions.

Abstract:Standard chemotherapy cannot eradicate triple-negative breast cancer (TNBC) while the residual cancer cells readily form the vasculogenic mimicry (VM) channels, which lead to the relapse of cancer after treatment. In this study, the functional vincristine plus dasatinib liposomes, modified by a targeting molecule DSPE-PEG(2000)-c(RGDyK), were fabricated to address this issue. The investigations were performed on TNBC MDA-MB-231 cells and MDA-MB-231 xenografts in nude mice. The liposomes exhibited the superior performances in the following aspects: the enhancement of cellular uptake via targeted action; the induction of apoptosis via activation of caspase 8, 9, and 3, increased expression of Bax, decreased expression of Mcl-1, and generation of reactive oxygen species (ROS); and the deletion of VM channels via inhibitions on the VM channel indicators, which consisted of vascular endothelial-cadherin (VE-Cad), focal adhesion kinase (FAK), phosphatidylinositide 3-kinase (PI3K), and matrix metallopeptidases (MMP-2, and MMP-9). Furthermore, the liposomes displayed the prolonged circulation time in the blood, the increased accumulation in tumor tissue, and the improved therapeutic efficacy along with deletion of VM channels in the TNBC-bearing mice. In conclusion, the nanostructured functional drug-loaded liposomes may provide a promising strategy for the treatment of invasive TNBC along with deletion of VM channels.
