Abstract:CONTEXT: Scutellaria baicalensis Georgi (Lamiaceae) is a popular medicinal plant. Its roots are used as the famous traditional Chinese medicine Huang-Qin, which is recorded in Chinese Pharmacopoeia, European Pharmacopoeia, and British Pharmacopoeia.OBJECTIVE: This review comprehensively summarizes research progress in phytochemistry, pharmacology, and flavonoid biosynthesis of S. baicalensis.METHODS: English and Chinese literature from 1973 to March 2018 was collected from databases including Web of Science, SciFinder, PubMed, Elsevier, Baidu Scholar (Chinese), and CNKI (Chinese). Scutellaria baicalensis, chemical constituents, phytochemistry, biological activities, and biosynthesis were used as the key words.RESULTS: A total of 126 small molecules (1-126) and 6 polysaccharides have been isolated from S. baicalensis. The small molecules can be classified into four structural types, namely, free flavonoids, flavonoid glycosides, phenylethanoid glycosides, and other small molecules. Extracts of S. baicalensis and its major chemical constituents have been reported to possess anti-viral, anti-tumor, anti-bacterial, antioxidant, anti-inflammatory, hepatoprotective, and neuroprotective activities. Key steps in the biosynthetic pathways of Scutellaria flavonoids have also been summarized.CONCLUSIONS: This article could be helpful for researchers who are interested in the chemical constituents, bioactivities, biosynthesis, and clinical applications of S. baicalensis.

Abstract:Scutellaria baicalensis, commonly known as Chinese Skullcap, is a traditional herb with a long history of cultural use in Chinese medicines. This review focuses on a comprehensive summary of the morphological description, bioactive compounds, drug-herb interactions, and pharmacological activities of Scutellaria baicalensis, as well as its therapeutic applications. A wide-ranging search using databases such as PubMed, Scopus, Google Scholar, and advanced search on the Web was conducted for data collection from prior studies. The plant root contains major active constituents including flavonoids and flavo-glycosides such as wogonin, baicalein, baicalin, oroxylin A, scutellarein, and norwogonin. These compounds have demonstrated a variety of pharmacological activities, combating inflammation, oxidative stress, cancer, neuronal disorders, and liver disorders. Chinese Skullcap represents a promising avenue for potential pharmacotherapeutic formulation advancement, emphasizing the need for further scientific exploration and clinical trials.

Abstract:Scutellaria Linn., which belongs to the family Lamiaceae, is a commonly used medicinal plant for heat clearing and detoxification. In particular, the roots of S. baicalensis and the entire herb of S. barbata have been widely used in traditional medicine for thousands of years. The main active components of Scutellaria , including: baicalein, wogonin, norwogonin, scutellarein, and their glycosides have potential or existing drug usage. However, the wild resources of Scutellaria plants have been overexploited, and degenerated germplasm resources cannot fulfill the requirements of chemical extraction and clinical usage. Metabolic engineering and green production via microorganisms provide alternative strategies for greater efficiency in the production of natural products. Here, we review the progress of: pharmacological investigations, multi-omics, biosynthetic pathways, and metabolic engineering of various Scutellaria species and their active compounds. In addition, based on multi-omics data, we systematically analyze the phylogenetic relationships of Scutellaria and predict candidate transcription factors related to the regulation of active flavonoids. Finally, we propose the prospects of directed evolution of core enzymes and genome-assisted breeding to alleviate the shortage of plant resources of Scutellaria . This review provides important insights into the sustainable utilization and development of Scutellaria resources.

Abstract:Scutellaria baicalensis Georgi is one of the most widely used medicinal plants, and is officially listed in the Chinese Pharmacopoeia. Its roots have been used for anti-inflammation, anticancer, decreasing blood pressures, reducing the total cholesterol level, treating bacterial and viral infections of the respiratory and the gastrointestinal tract, cleaning away heat, moistening aridity, purging fire and detoxifying toxicosis. This plant also possesses cholagogic, diuretic, and cathartic actions. Some concentrated composite herbal preparations containing Scutellaria baicalensis Georgi as a major ingredient in their prescriptions are widely used in oriental countries. Scutellaria baicalensis Georgi contains a variety of flavones, phenylethanoids, amino acids, sterols and essential oils. Its dried roots contain over 30 kinds of flavonoids, such as baicalin, baicalein, wogonin, wogonin 7-O-glucuronide and oroxylin A. The flavonoids are the main active components in Scutellaria baicalensis Georgi. This chapter provides up-to-date coverage of this class of flavonoids in regard to chemical structures, natural resources, biosyntheses, analytical methods and biological activities. Special attention is paid to both biosyntheses and biological activities including antioxidant and free radical scavenging, anti-inflammation, anticancer, antibacterium, anti-HIV, anti-hepatitis B virus, anti-respiratory syncytial virus and anti-SARS coronavirus properties. The structural diversity and the pronounced biological activities encountered in the flavonoids of Scutellaria baicalensis indicate that this class of compounds is worthy of further studies that may lead to new drug discovery. The review provides an account on our research work combined with a reference of the information obtained in both the English and Chinese literature.

Abstract:PURPOSE/OBJECTIVES To assess adherence and coping strategies in outpatients with chronic myeloid leukemia (CML) on oral tyrosine kinase inhibitors (TKIs). . DESIGN Prospective, descriptive. . SETTING An interdisciplinary oncology outpatient clinic in Germany. . SAMPLE 35 outpatients with CML on oral TKIs. . METHODS Adherence and coping strategies were assessed with questionnaires. Clinical data were extracted from medical charts. . MAIN RESEARCH VARIABLES Adherence rates, main coping strategies, and frequency and contents of single coping strategies. . FINDINGS 18 patients showed adherence according to the applied screening instrument. Main coping strategies were spirituality and search for meaning. The two single items most frequently specified were adhering to medical instructions and trusting in the medical personnel involved. . CONCLUSIONS The low adherence rate of 51% most likely resulted from using the Basel Assessment of Adherence Scale as the questionnaire of choice. The relevance of spirituality and search for meaning as main coping strategies has not been shown previously in outpatients with CML. Most patients wish to obey medical instructions accurately and put trust in their oncologists; this introduces a resource that should gain relevance considering the increasing number of oral anticancer drugs. . IMPLICATIONS FOR NURSING Nurses are encouraged to routinely assess adherence and spiritual needs in outpatients with CML. Spirituality and search for meaning represent pivotal coping strategies in this group, which has an excellent prognosis. Oncology nurses may help provide tailored support, thereby ameliorating care for these patients.

Abstract:Abstract Scutellaria baicalensis Georgi. (SB) is a common heat-clearing medicine in traditional Chinese medicine (TCM). It has been used for thousands of years in China and its neighboring countries. Clinically, it is mostly used to treat diseases such as cold and cough. SB has different harvesting periods and processed products for different clinical symptoms. Botanical researches proved that SB included in the Chinese Pharmacopoeia (1st, 2020) was consistent with the medicinal SB described in ancient books. Modern phytochemical analysis had found that SB contains hundreds of active ingredients, of which flavonoids are its major components. These chemical components are the material basis for SB to exert pharmacological effects. Pharmacological studies had shown that SB has a wide range of pharmacological activities such as antiinflammatory, antibacterial, antiviral, anticancer, liver protection, etc. The active ingredients of SB were mostly distributed in liver and kidney, and couldn't be absorbed into brain via oral absorption. SB’s toxicity was mostly manifested in liver fibrosis and allergic reactions, mainly caused by baicalin. The non-medicinal application prospects of SB were broad, such as antibacterial plastics, UV-resistant silk, animal feed, etc. In response to the Coronavirus Disease In 2019 (COVID-19), based on the network pharmacology research, SB’s active ingredients may have potential therapeutic effects, such as baicalin and baicalein. Therefore, the exact therapeutic effects are still need to be determined in clinical trials. SB has been reviewed in the past 2 years, but the content of these articles were not comprehensive and accurate. In view of the above, we made a comprehensive overview of the research progress of SB, and expect to provide ideas for the follow-up study of SB.

Abstract:Abstract Objectives Scutellaria baicalensis Georgi. (Lamiaceae) is a plant of the genus Lamiaceae, and its root is the main part used as a medicine. In China, Scutellaria baicalensis is still an important traditional Chinese medicine with the functions of clearing away heat and dampness, purging fire and detoxification. This medicinal plant is widely distributed in China, Russia, Mongolia, North Korea and Japan. The purpose of this paper was to provide a systematic and comprehensive overview on the traditional usages, botany, phytochemistry, pharmacology, pharmacokinetics and toxicology of this plant. Furthermore, the possible development trends and perspectives for future research on this medicinal plant are also discussed. Key findings So far, over 40 compounds have been isolated and identified from Scutellaria baicalensis, including flavonoids, terpenoids, volatile oils and polysaccharides. The compounds and extracts isolated from Scutellaria baicalensis exhibit a wide range of pharmacological activities, including the effects on the nervous system, effects on the immune system, liver protection, antitumour effects, antibacterial and antiviral effects, antioxidant effects and other pharmacological effects. Summary As a traditional Chinese herbal medicine, Scutellaria baicalensis has shown significant effects on the treatment of various diseases, especially hepatitis, diarrhoea, vomiting and high blood pressure. Numerous traditional uses of Scutellaria baicalensis have been confirmed by current investigations. However, it is also necessary to further study the drug-forming properties and pharmacokinetics of the active constituents of Scutellaria baicalensis, as well as to establish quality control standards for different areas of Scutellaria baicalensis, and to carry out the research at the cellular and molecular levels.

Abstract:ETHNOPHARMACOLOGICAL RELEVANCE SCUTELLARIA BAICALENSIS: Georgi (Labiatae) is a well-known traditional Chinese medicine to treat inflammation, cardiovascular diseases, respiratory and gastrointestinal infections, etc. The present study was to understand the metabolism of the root of Scutellaria baicalensis (a.k.a. Huangqin in Chinese) in the gastrointestinal tract and the correlation between the metabolites and their respective pharmacological activities.The water extract of the root of Scutellaria baicalensis (WESB) was incubated with simulated gastric and intestinal juices, and human fecal microflora for 24h at 37 °C. The HPLC-DAD analysis was used to monitor the in vitro metabolic process and identify its metabolites by comparing their absorption spectrum and retention time with those of chemical references. The in vitro anticomplementary and antimicrobial activity was evaluated with hemolysis assay, agar disc-diffusion method and MIC value, respectively.Main constituents of WESB remain unchanged during the incubation with simulated gastric juice (pH = 1.5) and intestinal juice (pH = 6.8), whereas four flavones, baicalin, wogonoside, oroxyloside and norwogonoside were metabolized into their respective aglycons by human intestinal bacteria. All four metabolites were demonstrated to have higher anticomplementary and antimicrobial activity than those of WESB. The anticomplementary active metabolites were identified to be baicalein, oroxylin A and norwogonin, among them, norwogonin is the most active compound.The presence of intestinal bacteria is demonstrated to play an important role in the gastrointestinal metabolism of WESB, and the pharmacological effects of Scutellaria baicalensis may be dependent on the intestinal bacteria metabolism.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Ethnopharmacological relevance</h3>Scutellaria (Lamiaceae), which includes 536 accepted species, is widespread in Europe, North America, East Asia, and South America. Several species are used as traditional medicines for a long history to treat diarrhea, dysentery, inflammation, respiratory infections. The phytochemistry and pharmacology of the genus Scutellaria have developed dramatically in the past ten years, and the traditional uses and clinical studies of the genus have not been systematically summarized. Therefore, it is especially valuable to exhibit its current progress to provide the bases for further exploration of medicinal potential.Aim of the review: The aim of the review is to provide updated information on the ethnopharmacology and clinical studies, the ten-years research progress of phytochemistry and pharmacology of Scutellaria, and to explore the potential medicinal values and further studies of Scutellaria.<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Materials and methods</h3>This review is based on the literature studies and books from the library and electronic sources, including SciFinder, Scopus, PubMed, Web of Science, Baidu Scholar, CNKI, online ethnobotanical database, and ethnobotanical monographs. These literature are related to ethnopharmacology, clinical studies, the ten-years research progress of phytochemistry, and pharmacology of Scutellaria.<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3>A total of 52 species and 20 varieties of the genus Scutellaria are used as traditional medicine with various biological activities. In the past ten years, 208 chemical constituents have been identified from 16 species and 1 variety of the genus Scutellaria, such as neo-clerodane diterpenoids, sesterterpenoids, terpenoid, flavonoids. Currently, pharmacological researches have demonstrated that the extract and compound identified from this genus exhibited extensive biological activities, including anticancer, antioxidant, anti-inflammatory, antiviral activities, anti-bacterial, effects on cardiovascular and cerebrovascular diseases, hepatoprotective and effects on neurodegenerative disease. The species of S. baicalensis, S. barbata, and S. lateriflora and the main compounds baicalein, baicalin and wogonin are involved in clinical trials, which point the way for us to conduct further study including anticancer, lower blood pressure, anti-infective, anti-inflammatory, protect the central nervous system and other effects on Scutellaria.<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusions</h3>The species in Scutellaria can be used to treat cancer, infection, hepatic disorders, cardiovascular and cerebrovascular diseases, neurodegenerative diseases and other diseases. Some indications in traditional medicines have been confirmed by modern pharmacological activities, such as anticancer, anti-inflammatory, anti-infective, hepatoprotective, and neuroprotection. The available literature exhibited that most of the bioactivities could be attributed to flavonoids and neo-clerodane diterpenoids. Though there are some in clinical practice of Scutellaria, the existing researches on this genus is still limited. For more profound studies of Scutellaria, in-depth study of more concerned species, which possess great bioactivities to treat diseases in vitro/vivo and as well as in clinical are suggested to be carried out to complete the studies on their effective substances, pharmacology mechanism, quality control, clinical use, and new drug research. Also, in order to expand the development of medicinal resources of Scutellaria, the once used as local herbal medicine but unexploited species are needed more concern.

Abstract:We discuss the role played by Lyapunov exponents in the dynamics of Zhang's model of self-organized criticality. We show that a large part of the spectrum (the slowest modes) is associated with energy transport in the lattice. In particular, we give bounds on the first negative Lyapunov exponent in terms of the energy flux dissipated at the boundaries per unit of time. We then establish an explicit formula for the transport modes that appear as diffusion modes in a landscape where the metric is given by the density of active sites. We use a finite size scaling ansatz for the Lyapunov spectrum, and relate the scaling exponent to the scaling of quantities such as avalanche size, duration, density of active sites, etc.

Abstract:Scutellaria strigillosa Hemsl., known for its traditional use in Chinese herbal medicine, is valued for heat-clearing and detoxifying, promoting diuresis, reducing swelling, alleviating pain, and preventing miscarriage. Despite its historical use, comprehensive studies on pharmacophylogenetic analysis, including genetic and chemical profiles and the antimicrobial activity of S. strigillosa are still lacking. Understanding these aspects is crucial for fully realizing its therapeutic potential and ensuring sustainable use. This study aims to elucidate these aspects through comparative genomics, metabolomics, and antimicrobial assays with Scutellaria baicalensis Georgi and Scutellaria barbata D. Don. The chloroplast genome of S. strigillosa was assembled, measuring 152,533 bp, and revealing a high degree of conservation, especially in the protein-coding regions, and identified four regions trnK(UUU)-rps16, trnN(GUU)-trnR(ACG), accD-psaI, psbE-petL) of variability that could serve as phylogenetic markers. The phylogenetic analysis revealed a closer genetic relationship of S. strigillosa with S. tuberifera and S. scordifolia than traditionally classified, suggesting a need for taxonomic reevaluation within the genus. UPLC-Q-TOF-MS analysis in negative ion mode was used to explore the chemical diversity among these species, revealing distinct variations in their chemical compositions. S. strigillosa shared a closer chemical profile with S. barbata, aligning with phylogenetic findings. Metabolomic identification through Progenesis QI software resulted in the tentative identification of 112 metabolites, including a substantial number of flavonoids, diterpenoids, iridoid glycosides, phenylethanoid glycosides, and others. HPLC analysis further detailed the concentrations of 12 actives across the species, highlighting the variation in compound content. S. strigillosa shows antibacterial activity against Staphylococcus aureus and Pseudomonas aeruginosa, similar to S. baicalensis root extracts. This research enhances the understanding of the phylogenetic and phytochemical profiles and the antibacterial activity of S. strigillosa, offering new insights into its medicinal properties. The findings suggest a need for taxonomic reevaluation within the genus and underscore the potential antibacterial activity of S. strigillosa for therapeutic applications. Further studies are encouraged to explore its full medicinal potential and contribute to the sustainable development of Scutellaria species.

Abstract:BACKGROUND: As a kind of flavonoid, baicalin (C21 H18 O11 ) is extracted from Scutellaria baicalensis Georgi, the extract of which can be added to animal feed in China.OBJECTIVES: The present review will describe the current understanding of the pharmacological effects of baicalin in the regulation of inflammation, oxidative stress anti-virus and anti-tumour responses.METHODS: We highlight emerging literature that the application in livestock health and performance, the biological activities, the molecular mechanisms and the dosage forms of baicalin by analysing and summarising the main points of the cited literatures.RESULTS: It is found that baicalin can improve the functions of multiple physiological systems. Baicalin has a strong anti-inflammatory effect by regulating TLR4-NFκB-MAPK signalling pathway; it also can reduce oxidative stress by regulating Nrf2-Keap1 pathway; it can inhabit many kinds of virus such as influenza virus, respiratory virus, hepacivirus and others; it can also inhibit the growth of tumour cells by blocking the cell cycle or inducing apoptosis; and new dosage forms such as cationic solid lipid nanoparticles, cyclodextrin inclusion complexes or nanocrystalline can be applied to improve the deficiency of baicalin.CONCLUSIONS: In summary, these studies have elucidated a comprehensive report on the anti-inflammatory, anti-oxidant, anti-virus and anti-tumour of baicalin, these findings thus indicated that baicalin can be used effectively to the field of animal production in future when the appropriate dosage form is determined.

Abstract:Integrative Cancer Therapies, Volume 23, Issue , January-December 2024. Gastrointestinal (GI) cancer stands as one of the most prevalent forms of cancer globally, presenting a substantial medical and economic burden on cancer treatment. Despite advancements in therapies, it continues to exhibit the second highest mortality rate, primarily attributed to drug resistance and post-treatment side effects. There is an urgent need for novel therapeutic approaches to tackle this persistent challenge. Scutellaria baicalensis, widely used in Traditional Chinese Medicine (TCM), holds a profound pharmaceutical legacy. Modern pharmacological studies have unveiled its anticancer, antioxidant, and immune-enhancing properties. S. baicalensis contains hundreds of active ingredients, with flavonoids, polysaccharides, phenylethanoid glycosides, terpenoids, and sterols being the principal components. These constituents contribute to the treatment of GI cancer by inducing apoptosis in tumor cells, arresting the cell cycle, inhibiting tumor proliferation and metastasis, regulating the tumor microenvironment, modulating epigenetics, and reversing drug resistance. Furthermore, the utilization of modern drug delivery technologies can enhance the bioavailability and therapeutic efficacy of TCM. The treatment of GI cancer with S. baicalensis is characterized by its multi-component, multi-target, and multi-pathway advantages, and S. baicalensis has a broad prospect of becoming a clinical adjuvant or even the main therapy for GI cancer.

Investigation on the function tropism of Tiaoqin and Kuqin (different specification of Scutellaria baicalensis) by comparing their curative effect on different febrile disease model

Abstract:Ethnopharmacological relevance: Scutellaria baicalensis (S. baicalensis) is the root of S. baicalensis Georgi. In traditional Chinese medicine it is divided into Tiaoqin (TQ, 1-3 years old) and Kuqin (KQ, more than 3 years old). However, the differences in TQ and KQ efficacy and their exact mechanisms are still unclear. Aim of the study: This study aimed to clarify the difference in the efficacy of TQ and KQ in relation to different fever types (damp heat and hyperpyrexia) by using rat models, as well as to determine the primary molecular mechanism. Materials and methods: This study compared the compositional content of TQ and KQ by UPLC-MS/MS. Then, rat models of hyperpyrexia (HP, LPS) and damp heat (DH, high-fat and high-sugar diet feeding + fumigation in artificial climate chamber + E. coli injection) were established and their clinical symptoms, blood biochemistry, histopathological sections, cell cytokines and protein expression were compared following treatment with TQ or KQ. Finally, the mechanisms underpinning the differences observed for TQ and KQ were determined by measuring the components of these treatments in different target organs. Results: This study identified 31 compounds in the water extracts of both TQ and KQ, which differed significantly in their relative content. TQ and KQ showed different functional tropism in HP and DH model rats. Baicalin, wogonoside, oroxin A, baicalein, wogonin and oroxylin A appeared to be the basic functional components responsible for the functional tropism hypothesis, while the remaining compounds appeared to be the efficacy-oriented components. In addition, the difference in pharmacodynamics between TQ and KQ may be related to their absorption in vivo, which was consistent with the hypothesis of functional tropism proposed in this work. Conclusion: In this study we adopted TQ and KQ-different specifications of Scutellaria baicalensis with similar chemical components-as a case study to systematically reveal the functional tropism of Chinese herbal medicine (CHM). The results showed that TQ and KQ contain the basic functional components to enable the basic function of 'clearing heat', while the variation in compositional content may result in their different therapeutic effects. A greater understanding and utilisation of the functional tropism of CHM would enormously improve the accuracy and scientific basis for the application of CHM medication, as well as in promoting the multi-function mechanism of CHM and guiding new drug development of CHM.

Abstract:This article will review selected herbal products used in traditional Chinese medicine, including medicinal mushrooms ( bā xī mó gū; Agaricus blazei, yún zhī; Coriolus versicolor, líng zhī; Ganoderma lucidum, xiāng xùn; shiitake, Lentinus edodes, niú zhāng zhī; Taiwanofungus camphoratus), Cordyceps ( dōng chóng xià cǎo), pomegranate ( shí liú; Granati Fructus), green tea ( lǜ chá; Theae Folium Non Fermentatum), garlic ( dà suàn; Allii Sativi Bulbus), turmeric ( jiāng huáng; Curcumae Longae Rhizoma), and Artemisiae Annuae Herba ( qīng hāo; sweet wormwood). Many of the discussed herbal products have gained popularity in their uses as dietary supplements for health benefits. The review will focus on the active constituents of the herbs and their bioactivities, with emphasis on the most recent progress in research for the period of 2003 to 2011.

Abstract:Scutellaria baicalensis (Huangqin in Chinese) is a major traditional Chinese medicine (TCM) herb, which has a long history of use in the treatment of a variety of symptoms correlated with cancer. In the past decade, the potential of S. baicalensis and single compounds derived from it as anticancer agents targeting various pathways has received extensive research attention. Specifically, the proliferation and metastases inhibiting properties of the single compounds in cancer have been studied; however, the underlying mechanisms remain unclear. This review summarizes the various mechanisms, pathways and molecular targets involved in the anticancer activity of S. baicalensis and its single compounds. However, the aim of this review is to provide a more thorough view of the last 10 years to link traditional use with modern research and to highlight recently discovered molecular mechanisms. Extracts and major flavonoids derived from S. baicalensis have been found to possess anticancer effects in multiple cancer cell lines both in vitro and in vivo. Further investigation is warranted to better understand the underlying mechanisms and to discover novel targets and cancer therapeutic drugs that may improve both the survival and quality of life of cancer patients.

Abstract:Baicalin is one of the major bioactive components of Scutellaria radix, a Chinese herb that has been used since ancient times. Baicalin has various pharmacological activities, including antitumor, antimicrobial, and antioxidant, and has wide clinical applications. Baicalin displays a distinct pharmacokinetic profile including gastrointestinal hydrolysis, enterohepatic recycling, carrier-mediated transport, and complicated metabolism. The in vivo disposition of baicalin is affected by combinations of other herbs and baicalin can interact with other co-administered drugs due to competition between metabolic enzymes and protein binding. Furthermore, baicalin exhibits altered pharmacokinetic properties under different pathological conditions. Due to its low bioavailability, emerging novel baicalin preparations including nano/micro-scale baicalin delivery systems show better absorption and higher bioavailability in preclinical studies, and show promise for future clinical applications. Thus, this current review offers a comprehensive report on the pharmacokinetic behavior of baicalin and strategies to improve its bioavailability.

Effects of elicitors from culture filtrate of Fusarium solani CL105 on flavonoid production of Scutellaria baicalensis calli

Abstract:Introduction: Endophytic fungi can promote secondary metabolite accumulation in medicinal plants. Previously, we observed that the culture filtrate of Fusarium solani CL105 promoted flavonoid production in Scutellaria baicalensis calli. However, the active ingredients and mechanisms associated with this secondary metabolite accumulation remain unclear. Methods: This study evaluates the effects of different elicitors from the culture filtrate of F. solani CL105 namely, exopolysaccharide (EPS), exoprotein (EP), and other parts (OP), on the flavonoid production in S. baicalensis calli by HPLC. Subsequently, the underlying mechanism of EPS induced flavonoid production in S. baicalensis calli was revealed by transcriptomics and RT-PCR. Results and discussion: The results indicated a significant increase in flavonoid production in S. baicalensis calli following treatment with EPS. Baicalin (1.40 fold), wogonoside (1.91 fold), and wogonin (2.76 fold) were most significantly up-regulated compared with the control. Transcriptome analysis further revealed up-regulation of key enzyme genes ( CHS, CHI, FNS , and F6H ) involved in flavonoid synthesis after 5 days of EPS treatment. Moreover, the expression of GA2ox and CYP707A —genes involved in gibberellin acid (GA) and abscisic acid biosynthesis (ABA), respectively—were significantly up-regulated. The expression levels of certain transcription factors, including MYB3, MYB8 , and MYB13 , were also significantly higher than in controls. Our results indicated that EPS was a main active elicitor involved in promoting flavonoid production in S. baicalensis calli. We postulated that EPS might stimulate the expression of MYB3, MYB8, MYB13, GA2ox , and CYP707A , leading to markedly upregulated CHS, CHI, FNS , and F6H expression levels, ultimately promoting flavonoid synthesis. This study provides a novel avenue for large-scale in vitro production of flavonoids in S. baicalensis .

Abstract:Scutellaria baicalensis was collected from four wild and four cultivated populations from different locations in China. Forty-two samples were analyzed using random amplification of polymorphic DNA (RAPD) techniques for genetic profiling, and high performance liquid chromatography (HPLC) techniques to determine the flavonoid content. The selected 23 RAPD primers yielded a total of 838 clear and reproducible bands of which 237 were found to be polymorphic. The wild population exhibited higher polymorphism than that of the cultivated population. The dendrogram generated by the UPCMA method via Nei's genetic distance revealed three distinct genotypes from the cultivated populations and several branches from the wild populations. The contents of baicalin and wogonoside in dried roots of the samples ranged from (w/w) 8.63 to 17.84%, and from 1.99 to 4.21%, respectively, whereas their aglycones, baicalein and wogonin, were within the range of only 0.04-0.23%. The total content of the four flavonoids varied front 9.45 to 26.24%. Comparatively, the cultivated populations contained much higher levels of baicalin and total flavonoids than those in the wild populations. The results from genetic characterization and phytochemical analysis demonstrated that the quality variation of this drug was mainly determined by extrinsic environmental or agricultural factors, rather than by genetic differences. Our findings can be used for the commercial production and germplasm management of this medicinal plant.
