Abstract:Land reclamation may expand the supply of usable land for food security. Soil microorganisms have been considered as an amendment in immature soil to improve its quality. However, different microbial fertilizers' effects on plant growth in immature soil have largely been unexplored. In order to evaluate the effects of different microbial fertilizers on immature soil, the soil quality and microbial community structure of corn rhizosphere soil samples under different microbial fertilizers were investigated. The results revealed a significant difference between microbial fertilizers (especially seaweed microbial fertilizer, SMF) and commercial chemical compound fertilizers in the soil properties and microbial community structure. Indeed, SMF caused a 486.21%, 23.17%, 21.08%, 38.33%, and 482.39% increase in Flavobacteriaceae, Planctomycetaceae, Chitinophagaceae, Acidobacteria_Gp3, and Mortierellaceae but a 23.82%, 18.66%, 42.36%, 29.12%, 81.97%, 42.19%, and 99.33% reduction in Cytophagales, Comamonadaceae, Rhodospirillaceae, Sinobacteaceae, Aspergillaceae, Myrmecridiaceae, and Typhulaceae, respectively; while CCF caused an 85.68% and 183.22% increase in Xanthomonadaceae and Mortierellaceae but a 31.29%, 36.02%, and 65.74% reduction in Cytophagales, Spartobacteria, and Cyphellophoraceae compared with the control based on 16S and ITS amplicon sequencing of soil microflora. Furthermore, redundancy discriminant analysis of the microbial communities and soil properties indicated that the main variables of the bacterial and fungal communities included exchangeable Ca, organic matter content, total N, and available P. Overall, the results of this study revealed significant changes under different fertilizer conditions in the microbiota and chemical properties of corn soil. Microbial fertilizers, particularly SMF and SM, can be used as a good amendment for newly reclaimed land.

Abstract:The rapid development of cities in the recent 10 years caused a reduction in the cultivated land area, which only accounts for 14% of the total land area in China. Land development and reclamation have been regarded as an effective way to compensate farmland occupation. However, most of the newly reclaimed land has poor soil fertility and suitability; in some cases, the production capacity is only 10–30% of the occupied farmland. In order to ameliorate the soil quality of the newly reclaimed land, this study evaluated the effects of commercial organic fertilizer (0.75, 1.50, and 2.25 kg/m2), mushroom residue (1.50, 2.25, and 3.00 kg/m2), biogas slurry (150, 225, and 300 kg/m2), vegetable cake (0.30 and 0.60 kg/m2), and chemical compound fertilizer (37.50 g/m2) on the pH, moisture content and organic matter content (OMC), available phosphate, total nitrogen, alkaline hydrolysis nitrogen, microbial biomass carbon and nitrogen, and number of total bacteria and phosphate-solubilizing bacteria, as well as the growth of maize seedlings. The results from this study indicate that the soil quality (OMC is an indicator) was ameliorated by chemical and organic fertilizers, in particular commercial organic fertilizers, which caused a 9.35–16.35% increase in moisture content, a 11.56–18.72% increase in pH, a 1.73–2.15 fold increase in OMC, a 338.44–491.41% increase in available P, a 36.80–48.14% increase in total N, a 95.32–128.34% increase in alkaline hydrolysis N, a 92.57–178.38% increase in total bacterial numbers, and a 7.57–20.87 fold increase in microbial biomass carbon compared with the control. The pot experiment further indicated that soil amended with commercial organic fertilizers caused a 20.35–30.55% increase in the height and a 12.50–16.67% increase in the total dry weight of maize seedlings. In addition, representative strains with the ability to dissolve phosphorus and fix nitrogen were successfully isolated using the culture method, and were then identified based on colony morphological observation and 16S rDNA sequence analysis, which help us to not only understand why organic fertilizer has great effect on soil improvement, but also provides beneficial microbial resources for further study.
Soil Bacterial Community Structure and Function under the Substitution of Chemical Fertilizer with Maize Straw

Abstract:The long-term extensive application of chemical fertilizers wreaks havoc on soil bacterial structure and function. To reduce the damage caused by chemical fertilizers, a six-year experiment was performed to study the effects of replacing 0% (CK), 25% (S25), 50% (S50), 75% (S75), and 100% (S100) of 225 kg ha−1 mineral nitrogen fertilizer with an equivalent amount of nitrogen from maize straw on the soil bacterial community structure, diversity, and function. The results showed that Proteobacteria, Acidobacteria, and Gemmatimonadetes were the dominant soil bacterial phyla after the replacement treatments. Replacing mineral nitrogen fertilizer with an equivalent amount of nitrogen from maize straw significantly reduced the number of Photobacterium and bacterial populations involved in genetic information processing in soil, but significantly increased the number of bacterial populations involved in organismal systems, human diseases, and environmental information processing. Compared with other treatments, the relative abundance of TK10 significantly increased by 33.52–76.36% in S25. The number of subgroup 6, Gram-negative, biofilm-forming, potentially pathogenic, and anaerobic bacteria significantly increased, whereas that of Chloroflexi and Blastocatellia subgroup 4 significantly decreased in S50 and S75 compared with CK. The number of TK10 and Blastocatellia subgroup 4 in S50 and S100, respectively, was significantly lower than that in CK. Bacterial species were significantly more present in S25 than in S75. The diversity of bacterial species in S75 was significantly lower than that in CK. S25 and S100 were more favorable to increasing the number of Gram-positive, aerobic, mobile-element containing, and stress-tolerant bacteria. Rhodobacteraceae, Pyrinomonadaceae, Xanthobacteraceae, Nocardioidaceae, and Vulgatibacteraceae with statistical differences in CK, S25, S50, S75, and S100, respectively, could be used as biomarkers. Chloroflexi, Acidobacteria, and Nitrospirae could be used as the main basis for the bacterial classification of soil samples in the equivalent substitution of nitrogen chemical fertilizer with maize straw. S25 is ideal for increasing soil bacterial species richness and abundance.

Abstract:Under the system of full straw returning, the relationship between soil fungal community diversity and soil physiochemical properties, and the combined application of slow-release nitrogen and urea is unclear. To evaluate its effect and provide an effective strategy for sustainable agricultural production, a 2-year field positioning trial was conducted using maize as the research object. The experiment was designed with two factors: straw treatment(S) and nitrogen fertilizer treatment(N),Six experimental treatments were set up,S1N0,S1N1,S1N2,S1N3,S1N4,S0N2,respectively.Analysis of 54 soil samples revealed 15 fungal phyla and 49 fungal classes. The composition of fungal communities in each treatment was basically the same, but there were significant differences in species abundance. Under total straw returning conditions, the combined application of slow-release nitrogen fertilizer and normal nitrogen fertilizer significantly increased the relative abundance of Ascomycota. During the jointing stage, tasseling stage and maturity stage, S1N4, S1N3 and S1N2 increased by 25.76%, 22.97%, 20.74%; 25.11%, 30.02%, 23.64% and 22.47%, 28.14%, 22.71% respectively compared with S0N2.The relative abundance of Basidiomycota was significantly reduced. Alpha diversity analysis showed that the straw returning mode significantly increased the Shannon index and decreased the Simpson index, which was obvious in the jointing stage and tasseling stage. The principal coordinate analysis analysis results showed that the fungal communities formed different clusters in the horizontal and vertical directions at the three growth stages of corn jointing, tasseling and maturity. At the jointing stage and tasseling stage, the communities of the straw return treatment and the straw removal treatment were separated, and the community distribution of each treatment was not significantly different in the mature stage. Total straw returning combined with slow-release fertilizer significantly (P＜0.05) increased the soil organic carbon, nitrate nitrogen and ammonia nitrogen content in each growth period, and increased the soil total nitrogen and hydrolyzable nitrogen content (P＞0.05).After the straw was returned to the field, the combined application of slow-release nitrogen fertilizer and common urea had a significant impact on soil urease, catalase, and sucrase activities. Among them, the three enzyme activities were the highest in the S1N3 treatment at the jointing stage and maturity stage, and the S1N4 treatment at the tasseling stage had the highest enzyme activity. Fungal community composition is closely related to environmental factors. Soil organic carbon, urease and catalase are positively correlated with Ascomycota and negatively correlated with Basidiomycota.
Effects of AMF on Maize Yield and Soil Microbial Community in Sandy and Saline Soils

Abstract:This study aimed to investigate the effects of applying arbuscular mycorrhizal fungi (AMF) on maize root growth and yield formation under different soil conditions. This study was conducted under sandy soil (S) and saline–alkali soil (Y), with treatments of AMF application (AM) and no AMF application (CK). The root characteristics, yield, and quality of maize were measured. High-throughput sequencing technology was employed to assess the impact of AMF on the soil microbial community structure, and the correlation between soil microbes and soil physicochemical properties was elucidated. The results show that under both sandy and saline–alkali soil conditions, AMF application significantly enhanced maize root growth, yield, grain quality, and soil available nitrogen (AN), available phosphorus (AP), and available potassium (AK) contents compared to the CK treatment. Soil microbial Alpha diversity analysis indicated that AMF application effectively increased soil microbial diversity and richness. Principal coordinate analysis (PCoA) and microbial community structure analysis revealed significant differences in bacterial communities between AM treatment in sandy soil (SAM) and CK in sandy soil (SCK), and significant differences in both bacterial and fungal communities between AM treatment in saline–alkali soil (YAM) and CK in saline–alkali soil (YCK). Furthermore, significant correlations between microbial communities and soil physicochemical properties were found, such as AN, AP, AK, soil salinity (SS), and organic matter (OM) content. AMF application had a greater impact on bacterial communities than on fungal communities. This study demonstrated that the use of AMF as a bio-fungal fertilizer was effective in improving spring maize yields, especially in terms of yield increase and quality stability in sandy and saline soils, thereby contributing to safe and sustainable cropping practices.

Abstract:In order to maintain high yields and protect the environment, the replacement of chemical fertilizers with organic ones has received increasing attention in recent years. A 2-year field experiment (2015–2016) was carried out to assess the effects of substituting equal amounts of mineral fertilizer with organic manure on the yield, dry matter (DM), and nitrogen (N) uptake of spring maize (Zea mays L.) and on the mineral N (Nmin) distribution in the soil profile. The treatments included chemical fertilizer; different amounts of maize straw, cow manure, and chicken manure; and an unfertilized control (CK). Compared with the chemical fertilizer treatments, equal amounts of substitutions with cow manure or chicken manure increased production, and a 25% nutrient substitution resulted in the best yield increase. Straw return had no effect on maize production, and 100% straw return resulted in reduced production. The N accumulation and DM content both exhibited a slow-fast-slow growth trend throughout the various growth stages, and the average N uptake and DM accumulation in response to the treatments followed the order of chicken manure > cow manure > chemical fertilizer > straw return > CK. The Nmin content in the profile not only increased as the Nmin application rate increased but also showed greater increases at certain depths than at the surface, indicating that excessive N led to leaching. These results suggest that an appropriate proportion of organic substitution not only provides enough nutrients but also improves the soil environment and leads to increased yields. This technique represents a practical method of continuously increasing production and reducing the risk of N leaching.

The Preliminary Research on Shifts in Maize Rhizosphere Soil Microbial Communities and Symbiotic Networks under Different Fertilizer Sources

Abstract:The use of chemical fertilizer along with organic fertilizer is an important agricultural practice that improves crop yield but also affects soil biogeochemical cycles. In this study, a maize field experiment was conducted to investigate the effects of NPK fertilizer (NPK), organic fertilizer (OF), and their combination (NPK+OF) on soil chemical properties, bacterial and fungal community structures, and diversity compared the control (CK, without any fertilizer). The results showed that the application of OF and NPK-combined OF increased soil organic matter (OM), total N, total P, available N, available P, and available K levels. For alpha diversity analyses, the application of fertilizers led to decreases in soil bacterial and fungal Shannon indices (except for NPK in fungi). Compared with CK, NPK, OF, and NPK+OF fertilization treatments significantly increased the abundances of Acidobacteriota, Gemmatimonadota, and Basidiomycota. Network analysis showed that fertilization produced fewer connections among microbial taxa, especially in the combination of NPK and OF. A redundancy analysis combined with Mantel test further found that the soil OM, available N and P were the main soil-fertility factors driving microbial community variations. Therefore, using organic fertilizer or biological fertilizer combined with chemical fertilizer to improve the status of soil C, N, and P is a promising method to maintain the balance of soil microorganisms in maize field.

Abstract:Microbial fertilizers have the characteristics of high efficiency and environmental protection in improving saline soils, and the application of functional microbial fertilizers is of great significance for the green abatement of saline barriers and the improvement of soil quality in coastal areas. The experiment was based on moderately saline soil in the coastal area of Hebei Province, with corn as the indicator crop, on the basis of conventional chemical fertilizer application. Different microbial fertilizer treatments, namely, T1 （conventional chemical fertilizer 750 kg·hm-2 + compound microbial agent 75 kg·hm-2）, T2 （conventional chemical fertilizer 750 kg·hm-2 + Bacillus megaterium 300 kg·hm-2）, T3 （conventional chemical fertilizer 750 kg·hm-2 + B. mucilaginosus 300 kg·hm-2）, T4 （conventional chemical fertilizer 750 kg·hm-2 + organic silicon fertilizer 600 kg·hm-2）, T5 （conventional chemical fertilizer 750 kg·hm-2 + bio-organic fertilizer 600 kg·hm-2）, T6 （conventional fertilizer 750 kg·hm-2 + active microalgae 15 kg·hm-2）, and CK （only fertilizer 750 kg·hm-2）, were used for these seven treatments, to study the effects of different microbial fertilizers on soil nutrients, salinity, bacterial community, and corn yield and economic efficiency during two critical periods （V12 stage and maturity stage） of corn. The results showed that compared with that in CK, T1 significantly increased soil total nitrogen （TN） and available phosphorus （AP） contents during the whole growth period. Over the whole reproductive period, soil organic matter （OM） at maturity increased by 10.35% over the V12 stage compared to that in CK, but there was no significant difference between treatments. Compared with that in CK, T5 and T6 significantly reduced soil total salinity and Ca2+ content during the whole growth period by an average of 14.51%-18.48% and 24.25%-25.51%. T1 significantly increased the bacterial diversity index over the whole growth period by 45.16% compared to that in CK. The dominant soil phyla were Actinobacteria, Proteobacteria, Acidobacteria, and Chloroflexi, and the dominant genera were Bacillus and Geminicoccaceae. The most abundant functions of the bacterial community in the study area were chemoheterotrophy and aerobic chemoheterotrophy, with average relative abundances of 28.89% and 27.11%, and T3 and T6 significantly improved soil N cycling function. The results of redundancy analysis （RDA） indicated that Na+, SO42-, pH, and EC were important factors driving the structure of the bacterial community, and correlation heatmaps showed that Na+, SO42-, pH, and EC were significantly and positively correlated mainly with the phylum Planctomycetota, whereas soil OM and TN were significantly and positively correlated with Cyanobacteria. Compared with that in CK, T6 increased the relative abundance of Cyanobacteria and optimized the bacterial community structure during the whole growth period. Using recommended dosages of bacterial fertilizers T1 and T6 increased maize yield by 7.31%-24.83% and economic efficiency by 9.05%-23.23%, respectively. The preliminary results of soil chemical properties and yield correlation analysis revealed that EC, AP, HCO3-, and Mg2+ were the obstacle factors limiting soil productivity in coastal areas. In conclusion, the use of the compound bacterial agent （T1） and active microalgae （T6） at the recommended dosage can significantly enhance soil nutrients, reduce salinity, and improve the structural diversity of soil bacterial communities, which not only ensures the increase in maize yield and efficiency but also realizes the efficient use of microbial fertilizers and the improvement of soil quality.

Abstract:Controlled-release urea (CRU) fertilizer application has been shown to improve crop yield and nitrogen (N) use efficiency. However, its effects when mixed with conventional urea fertilizer on soil aggregate stability, humic acid (HA) molecular composition and crop N uptake remain unclear. Soil and plant samples were collected from a long-term (2008–2019) experiment on field maize (Zea mays L., 'Zhengdan 958') which included two types of fertilizers [conventional urea fertilizer (CUF), blended CUF with CRU fertilizer (CRF)], four N application rates (0, 150, 300 and 450 kg ha−1), each in three replicates. The results showed that at 300 kg N ha−1, compared to CUF treatment, the CRF treatment significantly improved soil aggregate characteristics [aggregate content with particle size larger than 0.25 mm (R0.25) by 9.6%, mean weight diameter by 19.8%, and geometric mean diameter by 21.7%]. CRF treatment also increased HA content by 5.5%, fulvic acid (FA) by 5.5%, lignin-like molecules by 0.94 times, and protein-like molecules by 3.69 times. At grain-filling stage, CRF treatments significantly increased the sum of soil NH4+-N and NO3−-N content by 23.3–24.5%, sap bleeding rate by 12.8–18.2% and N delivery rate through bleeding sap by 60.6–87.7% compared to CUF treatments at the same N application rate. At the same rate of N application, the CRF treatments significantly improved the average yield during three growing seasons by 9.4–14.0% in contrast with CUF treatments. The regression equations showed that the maximum yield was 8294 kg ha−1 for CUF at the application rate of 312 kg N ha−1 while it was 9890 kg ha−1 for CRF at the application rate of 286 kg N ha−1. We conclude that the long-term application of CRF changed the HA molecular structure, enhanced the water stable aggregates, improved crop N uptake, and increased economically viable maize yield.

Abstract:This long-term study used a lysimeter platform to monitor the NH3 and N2O emissions of summer maize resulting from various fertilization treatments in the Huanghuaihai area, with the goal to assess the efficiency of fertilization measures aimed at reducing NH3 and N2O losses during the production of summer maize; the results provide a theoretical basis for synergistically improving maize yield and fertilizer utilization efficiency. A 2-year field trial was conducted. The trial included a no-N-fertilizer treatment as a control to study the following three fertilizer treatments: the exclusive application of urea, the exclusive application of cattle manure, and the combined application of organic and inorganic fertilizers. The results show that during the two maize growth seasons included in the trial, the average N2O losses associated with exclusively applying urea, exclusively applying cattle manure, and the combined application of organic and inorganic fertilizers were 273%, 542%, and 376% higher than those associated with the control treatment, respectively. During the same period, the average accumulative ammonia volatilization losses were 311%, 542% and 376% higher than those for the control treatment, respectively. The average nitrogen accumulation resulting from the combined application of organic and inorganic fertilizers was 82% higher than that for the control treatment, 12% higher than that for the exclusive application of cattle manure, and 6% higher than that for the exclusive application of urea. The average grain yield for the combined application of organic and inorganic fertilizers, the exclusive application of urea and the exclusive application of cattle manure were 76%, 68% and 61% higher than that for the control treatment, respectively. Overall, the combined application of organic and inorganic fertilizers showed a lower ammonia volatilization loss than the exclusive application of urea, which resulted in a higher ammonium nitrogen and nitrate nitrogen content in the soil, an increased nitrogen uptake, an increased dry matter accumulation of maize, and a high grain yield and nitrogen recovery efficiency.

Abstract:Due to the development of urbanization and industrialization, a large amount of cultivated land resources has been occupied, while new reclamation land could expand the supply of usable land for food security. Organic fertilizers, such as crop residues, biosolids, sheep manure, mushroom residue, and biogas liquid, have been considered as an effective amendment in immature soil to improve its quality. Recently, two kinds of commercial organic fertilizers, pig manure and mushroom residue organic fertilizer (PMMR-OF), and sheep manure organic fertilizer (SM-OF), have been more regularly applied in agriculture production. However, the information available on effect of the two kinds of fertilizers on plant growth and rhizosphere soil properties in immature field is very limited. In order to evaluate PMMR-OF and SM-OF on immature soil, the soil quality and microbial community structure of corn rhizosphere soil samples under the two kinds of organic fertilizers at different concentrations was investigated. The results revealed a significant difference between commercial organic fertilizers (especially SM-OF) and chemical compound fertilizers (CCF) in soil properties and microbial community structure. Indeed, compared with the control based on16S and ITS amplicon sequencing of soil microflora, SM-OF caused a 10.79-19.52%, 4.33-4.39%,and 14.58-29.29% increase in Proteobacteria, Actinobacteria, and Ascomycota, but a 5.82-20.58%, 0.53-24.06%, 10.87-16.79%, 2.69-10.50%, 44.90-59.24%, 8.88-10.98%, and 2.31-21.98% reduction in Acidobacteria, Gemmatimonadetes, Bacteroidetes, Verrucomicrobia, Basidiomycota, Mortierellomycota, and Chytridiomycota, respectively. CCF caused a 24.11%, 23.28%, 38.87%, 19.88%, 18.28%, and 13.89% reduction in Acidobacteria, Gemmatimonadetes, Bacteroidetes, Verrucomicrobia, Basidiomycota, Chytridiomycota, but a 22.77%, 41.28%, 7.88%, and 19.39% increase in Proteobacteria, Actinobacteria, Ascomycota, and Mortierellomycota, respectively. Furthermore, redundancy discriminant analysis of microbial communities and soil properties of PMMR-OF, SM-OF, CCF, and the control treatments indicated that the main variables of bacterial and fungal communities included organic matter content, available P, and available K. Overall, the results of this study revealed significant changes under different fertilizer conditions (PMMR-OF, SM-OF, CCF, under different concentrations) in microbiota and chemical properties of corn soil. Commercial organic fertilizers, particularly SM-OF, can be used as a good amendment for the new reclamation land.

Abstract:Combined application of synthetic nitrogen (N) fertilizers and organic materials can enhance soil quality, but little is known about the distribution of fertilizer N among different soil fractions after crop harvest. A pot experiment using 15 N tracer was employed to address this question with three treatments, i.e., labeled urea-only ( 15 NU), labeled urea + rice straw ( 15 NU-S) and labeled rice straw + urea ( 15 NS-U) applied to a Ferallic Cambisol (1:1 type soil clay mineral) and a Calcaric Fluvisol (2:1 clay mineral). Soil microbial biomass N, fixed ammonium (fixed NH 4 + ), exchangeable ammonium and soil organic N fractions by hydrolysis (6 N HCl) and their isotope abundance were determined after the rice harvest. Soil newly formed N in urea + straw (U-S) treatments ( 15 NU-S, 15 NS-U) was the sum of labeled urea-N in 15 NU-S and labeled straw-N in 15 NS-U. Compared with 15 NU, U-S significantly ( P < 0.05) increased the content and percentage of newly formed total soil N, acid insoluble N, amino acid N, and hydrolysable unknown N in both soils. In U-S treatment, straw amendment significantly ( P < 0.05) reduced the content and percentage of newly formed fixed-NH 4 + -N in Fluvisol as compared with 15 NU treatments. Soil microbes contributed to the larger percentage of newly formed amino acid N ( P < 0.01) in Cambisol as compared with Fluvisol. Fertilizer N in various soil fractions was therefore strongly affected by clay mineral type and microbes after the combined application of organic materials and synthetic N fertilizer.

Abstract:Under a full straw returning system, the relationship between soil bacterial community diversity and straw decomposition, yield, and the combined application of slow-release nitrogen and urea remains unclear. To evaluate these effects and provide an effective strategy for sustainable agricultural production, a 2-year field positioning trial was conducted using maize as the research object. Six experimental treatments were set up: straw returning + no nitrogen fertilizer (S1N0), straw returning + slow-release nitrogen fertilizer:urea = 0:100% (S1N1), straw returning + slow-release nitrogen fertilizer:urea = 30%:70% (S1N2), straw returning + slow-release nitrogen fertilizer:urea = 60%:40% (S1N3), straw returning + slow-release nitrogen fertilizer:urea = 90%:10% (S1N4), and straw removal + slow-release nitrogen fertilizer:urea = 30%:70% (S0N2). Significant differences (p < 0.05) were observed between treatments for Proteobacteria, Acidobacteriota, Myxococcota, and Actinobacteriota at the jointing stage; Proteobacteria, Acidobacteriota, Myxococcota, Bacteroidota, and Gemmatimonadota at the tasseling stage; and Bacteroidota, Firmicutes, Myxococcota, Methylomirabilota, and Proteobacteria at the maturity stage. The alpha diversity analysis of the soil bacterial community showed that the number of operational taxonomic units (OTUs) and the Chao1 index were higher in S1N2, S1N3, and S1N4 compared with S0N2 at each growth stage. Additionally, the alpha diversity measures were higher in S1N3 and S1N4 compared with S1N2. The beta diversity analysis of the soil bacterial community showed that the bacterial communities in S1N3 and S1N4 were more similar or closely clustered together, while S0N2 was further from all treatments across the three growth stages. The cumulative straw decomposition rate was tested for each treatment, and data showed that S1N3 (90.58%) had the highest decomposition rate. At the phylum level, straw decomposition was positively correlated with Proteobacteria, Actinobacteriota, Myxococcota, and Bacteroidota but significantly negatively correlated with Acidobacteriota. PICRUSt2 function prediction results show that the relative abundance of bacteria in soil samples from each treatment differed significantly. The maize yield of S1N3 was 15597.85 ± 1477.17 kg/hm2, which was 12.80 and 4.18% higher than that of S1N1 and S0N2, respectively. In conclusion, a combination of slow-release nitrogen fertilizer and urea can enhance the straw decomposition rate and maize yield by improving the soil bacterial community and structure within a full straw returning system.

Abstract:Introduction: The partial substitution of chemical fertilizer with organic manure takes on a critical significance to enhancing soil quality and boosting sustainable agricultural development. However, rare research has studied the effects of partial substitution of chemical fertilizer with organic manure on soil bacterial community diversity and enzyme activity in maize field in the mountain red soil region of Yunnan. Methods: In this study, four treatments were set up in which chemical fertilizer (the application rates of N, P 2 O 5 and K 2 O were 240, 75 and 75 kg·ha −1 , respectively) was substituted by 10% (M10), 20% (M20), 30% (M30) and 40% (M40) of organic manure with equal nitrogen, as well as two control treatments of single application of chemical fertilizer (M0) and no fertilization (CK). The maize ( Zea mays L.) crop was sown as a test crop in May 2018. The effects of partial substitution of chemical fertilizer with organic manure on soil physicochemical properties, soil bacterial community diversity and enzyme activity were studied. Results: The activities of Cellulase (CBH), Invertase (INV) and β-glucosidase (BG) increased with the increase of organic manure substitution ratio. The activities of β-1,4-N-acetylglucosaminidase (NAG), Urease (URE), and leucine aminopeptidase (LAP) also had the same trend, but the highest activities were 159.92 mg·g −1 ·h −1 , 66.82 mg·g −1 ·h −1 and 143.90 mg·g −1 ·h −1 at 30% substitution ratio. Compared with CK and M0 treatments, Shannon index increased notably by 82.91%–116.74% and 92.42%–128.01%, respectively, at the organic manure substitution ratio ranging from 10% to 40%. Chao1 and ACE index increased significantly at the organic manure substitution ratio ranging from 10% to 30%. Proteobacteria was the dominant phylum in all treatments, the relative abundance of Proteobacteria decreased as the organic manure substitution ratio increased. Redundancy analysis showed that microbial biomass C was the main factor affecting the bacterial community composition under partial replacement of chemical fertilizer treatment, while Actinobacteria was the main factor affecting the enzyme activity. In addition, the maize yield of M30 and M40 treatments was significantly higher than that of CK and M0-M20 treatments, and the yield of M30 treatment was the highest, reaching 7652.89 kg·ha −1 . Conclusion: Therefore, the partial substitution of chemical fertilizer with organic manure can improve soil biological characteristics, while increasing bacterial community diversity and soil enzyme activity. Therefore, a thirty percent organic manure substitution was determined as the optimal substitution ratio for maize farmland in the mountain red soil area of Yunnan, China.

Abstract:Management of nitrogen (N) fertilizer is a critical factor that can improve maize ( Zea mays L. ) production. On the other hand, high volatilization losses of N also pollute the air. A field experiment was established using a silt clay soil to examine the effect of sulfur-coated urea and sulfur from gypsum on ammonia (NH 3 ) emission, N use efficiency (NUE), and the productivity of maize crop under alkaline calcareous soil. The experimental design was a randomized complete block (RCBD) with seven treatments in three replicates: control with no N, urea 150 alone (150 kg N ha −1 ), urea 200 alone (200 kg N ha −1 ), urea 150 + S (60 kg ha −1 S from gypsum), urea 200 + S, SCU 150 (sulfur-coated urea) and SCU 200 . The results showed that the urea 150 + S and urea 200 + S significantly reduced the total NH 3 by (58 and 42%) as compared with the sole application urea 200 . The NH 3 emission reduced further in the treatment with SCU 150 and SCU 200 by 74 and 65%, respectively, compared to the treatment with urea 200 . The maize plant biomass, grain yield, and total N uptake enhanced by 5–14%, 4–17%, and 7–13, respectively, in the treatments with urea 150 + s and urea 200 + S, relative to the treatment with urea 200 alone. Biomass, grain yield, and total N uptake further increased significantly by 22–30%, 25–28%, and 26–31%, respectively, in the treatments with SCU 150 and SCU 200 , relative to the treatment with urea 200 alone. The applications of SCU 150 enhanced the nitrogen use efficiency (NUE) by (72%) and SCU 200 by (62%) respectively, compared with the sole application of urea 200 alone. In conclusion, applying S-coated urea at a lower rate of 150 kg N ha −1 compared with a higher rate of 200 kg N ha −1 may be an effective way to reduce N fertilizer application rate and mitigate NH 3 emission, improve NUE, and increase maize yield. More investigations are suggested under different soil textures and climatic conditions to declare S-coated urea at 150 kg N ha −1 as the best application rate for maize to enhance maize growth and yield.

Abstract:Ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB) are key drivers of nitrification in rainfed soil ecosystems. However, within a semi-arid region, the influence of different soil amendments on the composition of soil AOA and AOB communities and soil properties of rainfed maize is still unclear. Therefore, in this study, the abundance, diversity, and composition of AOA and AOB communities and the potential nitrification activity (PNA) was investigated across five soil treatments: no fertilization (NA), urea fertilizer (CF), cow manure (SM), corn stalk (MS), and cow manure + urea fertilizer (SC). The AOB amoA gene copy number was influenced significantly by fertilization treatments. The AOB community was dominated by Nitrosospira cluster 3b under the CF and SC treatments, and the AOA community was dominated by Nitrososphaera Group I.1b under the CF and NA amendments; however, manure treatments (SM, MS, and SC) did not exhibit such influence. Network analysis revealed the positive impact of some hub taxonomy on the abundance of ammonia oxidizers. Soil pH, NO3−-N, Module 3, biomass, and AOB abundance were the major variables that influenced the potential nitrification activity (PNA) within structural equation modeling. PNA increased by 142.98–226.5% under the treatments CF, SC, SM, and MS compared to NA. In contrast to AOA, AOB contributed dominantly to PNA. Our study highlights the crucial role of bacterial communities in promoting sustainable agricultural production in calcareous soils in semi-arid loess plateau environments.

Abstract:Ammonia (NH3) emitted from synthetic nitrogen (N) fertiliser applications to farmland leads to air pollution and terrestrial acidification. Previous studies have indicated that replacing urea-based N fertilisers by manure may reduce NH3 emissions and enhance crop yield. However, the long-term effects of replacing urea N fertiliser by manure on crop yield and NH3 emissions have not been sufficiently quantified. The objective of the field study presented here was to examine the effects of three treatments (i.e., 100% synthetic fertiliser N application (NPK); 50% synthetic fertiliser N +50% manure N application (50%MNS); and 100% manure N application (100%MNS)) on NH3 emissions and maize grain yields over a 3-year period in a long term (13 years) study site in the North China Plain. Results showed that the NH3 emissions in the NPK treatment ranged from 9.7 to 11.7 kg ha− 1 during the three maize growing seasons. Replacement of urea fertiliser by manure significantly decreased the NH3 emissions by 22–54% in the 50% MNS treatment and by 47–71% in the 100%MNS treatment. Maize grain yields were 14–30% higher in the 50%MNS treatment and 17–45% higher in the 100% MNS treatment, compared to the NPK treatment (8.1–8.8 t ha− 1). The NH3 emission factor for the NPK treatment ranged from 4.1 to 4.8%. Additional 15 N labelling work established that 83% of NH3 emissions originated from the urea fertiliser in the NPK treatment. Manure treatments had a higher maize N uptake and soil organic matter content, and a lower soil pH than the NPK treatment. This study highlights that replacing synthetic N fertiliser with manure on farmland in the North China Plain has long-term beneficial effects on maize yield and N uptake, NH3 emission mitigation and soil carbon storage.

Abstract:Soil microorganisms play an important role in agricultural ecosystems, but their response to organic fertilizer application has not been thoroughly elucidated. Thus, high-throughput sequencing was used to investigate the responses of soil bacterial to organic fertilizer amendment (composted from pig manure) in the field during the entire growth cycle of maize plants. Four treatments were studied: treatment with 2 kg·m−2 organic fertilizer application (OF_2), treatment with 4 kg·m−2 organic fertilizer application (OF_4), treatment with 6 kg·m−2 organic fertilizer application (OF_6), and a controlled treatment (CK) without fertilization. The results revealed that the bacterial richness in OF_2 was significantly lower than that of CK (p < 0.05). Soil eutrophication bacteria Bacteroidetes increased effectively in all fertilized soils, relative abundance in OF_2, OF_4, and OF_6 for the entire maize growth cycle was 68.00%, 71.40%, and 77.93% higher than that in CK, respectively. In addition, soil nitrobacteria (Nitrospirae, Nitrospira), were markedly decreased (p < 0.05) with fertilization amount. The relative abundance of the nitrogen-fixing genus Adhaeribacter in OF_6 was 209.28%, 72.8% and 35.66% higher than that in CK, OF_2 and OF_4 at mature stage. The pathogenic genus Flavolibacterium was significantly increased (p < 0.05) in fertilized soil at the seeding stage. The driving factor governing the variations of bacterial community in CK, OF_2, OF_4 and OF_6 were pH value, available phosphorus, available phosphorus, and chromium, respectively. The findings highlight that part of the soil functional or pathogenic bacteria population was susceptible to organic fertilizer application; and the driving factor of bacterial composition change was associated with the rate of fertilization. More targeted experiments are needed to enhance the understanding of functional bacteria and the synergistic effect of soil physicochemical property physical on soil bacteria.
