Abstract:Infected wounds pose a serious threat to public health and pose a significant challenge and financial burden worldwide. The treatment of infected wounds is now an urgent problem to be solved. Herein, mild hyperthermia-assisted hydrogels composed of carboxymethyl chitosan (CMCs), oxidized dextran (Odex), epigallocatechin gallate (EGCG) and PtNPs@PVP (CAT-like nanoenzymes) were proposed for the repair of infected wounds. The incorporation of PtNPs@PVP nanoenzymes give the hydrogels excellent photothermal property and CAT-like activity. When the temperature is maintained at 42-45 degrees C under 808 nm near infrared (NIR) exposure, the CMCs/Odex/EGCG/Nanoenzymes (COEN2) hydrogel demonstrated highly enhanced antibacterial ability (95.9 % in vivo), hydrogen peroxide (H2O2) scavenging ratio (85.1 % in vitro) and oxygen supply (20.7 mg/L in vitro). Furthermore, this mild-heat stimulation also promoted angiogenesis in the damaged skin area. Overall, this multifunctional hydrogel with antibacterial, antioxidant, oxygen supply, hemostasis, and angiogenesis capabil-ities has shown great promise in the repair of infected wounds. This study establishes the paradigm of enhanced infected wound healing by mild hyperthermia-assisted H2O2 scavenging, oxygen supplemental, and photo -thermal antibacterial hydrogels.

Abstract:Bacterial infection of wounds is an escalating medical problem, issuing threats to both global public health and personal health. Photothermal antibacterial technology as a novel sterilization strategy has outstanding sterilization efficiency, high safety and low risk of emergence of drug-resistant bacteria. By combining inherent antibacterial activity and light-assisted antibacterial treatment, developing novel multifunctional dressings with synergistic high-efficiency antibacterial effects and also promoting wound healing possesses attractive advantages in the field of treating bacterial wound infections in clinical care. Herein, a multifunctional hydrogel formed by in situ photo-cross linking was designed and prepared by first grafting methacrylic anhydride as a photosensitizer onto chitosan, and then introducing oxidatively synthesized polydopamine (PDA). The physicochemical characterizations of the synthesized hydrogels demonstrated their tunability certainly associated with PDA concentration, including pore size, water swelling, rheological properties and in vitro degradability. In addition, the composite hydrogels exhibited good adhesion, anti-oxidation and photothermal properties due to the existence of PDA. Within 10 min upon exposure to 808 nm near-infrared (NIR) light irradiation, this hydrogel system displayed outstanding antibacterial activity against Staphylococcus aureus with almost 100% killing efficiency, of which rapid efficient sterilization plays a significant role in wound healing. Moreover, the hydrogel is capable of cytocompatibility and has low toxicity to murine fibroblasts (L929 and NIH/3T3). In the full-thickness wound defect infection model in mice, the wound closure ratio, inflammatory response, fibroblasts, neovascularization and epithelialization were measured. Animal experiments also reveal that the hydrogel assisted with NIR laser irradiation can inhibit effectively infection at an early stage and accelerate the wound healing process. In , this novel multifunctional injectable hydrogel exhibits excellent swelling capacity, bio-adhesion, antioxidant property, photothermal activity, efficient antibacterial property and facilitates skin healing, which has great promising application as a medical dressing biomaterial in infected wound care fields.

Abstract:Incomplete contact between a pre‐formed hydrogel and irregular wound limits the therapeutic effect of the dressing and increases the risk of infection; while great concerns have remained regarding the potential toxicity of the residual additives of chemical crosslinking for in‐situ forming hydrogels. Therefore, it is desirable to develop a self‐adaptive hydrogel in response to skin temperature with shape adaptability and efficient antibacterial property to prevent microbial invasion. Herein, a dually‐thermoresponsive hydrogel composed of poly(N‐isopropylacrylamide) (PNIPAm) and methacrylated κ‐carrageenan (MA‐κ‐CA) is designed with compliance at physiological temperature to realize shape adaptability for completely covering irregular wounds. Furthermore, the hydrogel with near‐infrared (NIR)‐responsive polypyrrole‐polydopamine nanoparticles (PPy‐PDA NPs) and Zn2+‐derived zeolitic imidazolate framework (ZIF‐8) can generate localized heat and gradually release Zn2+ to realize safe, effective synergetic photothermal‐chemical bactericidal capability. In addition, the release rate of Zn2+ can be accelerated by NIR‐induced heating, and thus a more efficient sterilization can be provided to severely infected wounds. Therefore, the proposed hydrogel would serve as a promising wound dressing for the full course of wound healing, with the abilities of perfectly covering the wound and adapting to regenerating tissue, and controllable photothermal‐chemical antibacterial capability to reach high bactericidal efficiency and long‐term release of antibacterial agents. Chitosan-based hydrogel incorporated with polydopamine and protoporphyrin for photothermal-oxidation sterilization of bacteria-infected wound therapy

Abstract:Phototherapy has emerged as a potential treatment strategy for bacteria-infected wounds, but the inadequate bacteria-capturing ability and excessive damage to normal tissues from single phototherapy are huge limitations. To solve the issues, herein we report the design of chitosan-based hydrogel with bacteria capturing and combined photothermal/photodynamic sterilization functions. Such hydrogel is prepared by mixing chitosan (CS) as matrix, protoporphyrin (PpIX) as photosensitizer and polydopamine (PDA) as photothermal agent and then chemically cross-linking CS with glutaraldehyde. The resulting CS-PpIX-PDA hydrogel possesses a porous architecture (average pore porosity = 60.9 %), excellent swelling capabilities (swelling ratio = 1855 %) and rheological property (G' > G″). The hydrogel can effectively produce reactive oxygen species (ROS) under 660 nm light irradiation due to the photodynamic effect of PpIX. Owing to the presence of PDA, the hydrogel displays a photoabsorption range between 600 and 1500 nm and can generate maximal temperature of 60 °C within 10 min under 808 nm laser illumination (0.6 W/cm2) through photothermal effect. Besides, under synergetic illumination of 808/660 nm laser, CS-PpIX-PDA hydrogel can induce the death of 99.9999 % of E. coli and 99.99999 % of S. aureus. Importantly, when coated on the wound site, the hydrogel exhibits a remarkable bacteria-trapping ability due to its porous structure and the presence of amino groups on chitosan. Under the excitation of 660/808 nm, the combined photothermal and photodynamic effects can effectively eradicate bacteria. Simultaneously, the hydrogel also demonstrates anti-inflammatory properties and upregulates Heat Shock Protein 90 (HSP90) expression, thereby promoting collagen deposition and facilitating wound healing. Therefore, the study may provide some new insights into the development of multifunctional hydrogel for photothermal-oxidation sterilization of bacteria-infected wound therapy.
Mussel-Inspired Hydrogel with Potent in Vivo Contact-Active Antimicrobial and Wound Healing Promoting Activities

Abstract:Open wounds (e.g., burns and trauma) are always challenged by various opportunistic bacteria. There is an urgent need for developing novel wound dressing that is able to prevent bacterial infection and promote the healing simultaneously. Herein, we developed a new type of antimicrobial hydrogels for the open wound healing through imitating a facile mussel-inspired catechol/polyamine chemistry. This hydrogel was prepared using catechol (CT) and ε-poly-l-lysine (EPL) by oxidative cross-linking directly in the open air at room temperature. This nonleaching CT/EPL hydrogel not only exhibited excellent contact-active antimicrobial activities against Gram-negative bacteria Escherichia coli (E. coli) and Gram-positive "superbug" methicillin-resistant Staphylococcus aureus (MRSA) but also inhibited the biofilm formation in vitro. Moreover, the animal burn wound model study clearly validated the in vivo anti-infective property of CT/EPL hydrogel against MRSA infection. More importantly, the CT/EPL hydrogel possessed low cytotoxicity and enhanced cell migration in vitro. A full-thickness cutaneous wound model study revealed that CT/EPL hydrogel upregulated the expression of vascular endothelial growth factor (VEGF) and reduced the production of the pro-inflammatory cytokines, thus promoted the wound healing. These findings suggested that the CT/EPL hydrogel have great potential as a wound dressing for preventing bacterial infection and accelerating healing of open wounds.
Bioinspired Multifunctional Injectable Hydrogel for Hemostasis and Infected Wound Management

Abstract:Routine wound management faces significant challenges including rebleeding and bacterial infection that affect millions of people each year. However, conventional wound dressings (e.g., gauze, bandage) are limited to simply cover the injured surfaces and rarely show special functionality to promote the wound recovery. Currently, injectable hydrogels have been widely designed as multifunctional wound dressings to manage the hemostatic and wound healing process. Nevertheless, the integration of multiple functions through simple composition and easy construction is still difficult and hardly achieved. Herein, we reported a bioinspired multifunctional injectable hydrogel (CQCS@gel) consisted of only two components, catechol-functionalized quaternized chitosan (CQCS) and dibenzaldehyde-terminated poly(ethylene glycol) (DB-PEG2000). The building blocks endowed CQCS@gel with tissue-adhesive, antibacterial, antioxidant, self-healing and pH-responsive properties. Based on the in vivo hemostatic study, quick hemostasis for acute tissue injuries such as liver and carotid wounds was realized owing to the rapid gelation rate and strong tissue-adhesiveness of CQCS@gel. Moreover, CQCS@gel remarkably boosted the chronic recovery process of MRSA-infected cutaneous wounds by promoting collagen deposition, hair follicles regeneration and angiogenesis. Overall, this multifunctional injectable hydrogel shows potentials as a universal wound dressing in clinical applications, enabling both hemostasis and infected wound management. STATEMENT OF SIGNIFICANCE: This is the first report showing the multifunctional injectable hydrogel (CQCS@gel) consisted of catechol-functionalized quaternized chitosan and dibenzaldehyde-terminated poly(ethylene glycol). The incorporation of quaternary ammonium groups imparted the CQCS@gel with outstanding contact-active bacterial killing efficiency and the catechol moieties enhanced its tissue adhesive and antioxidant properties. Moreover, the reversible imine crosslinks endowed the CQCS@gel with self-healing and pH-responsive drug release capabilities. These multiple functions were integrated into a single injectable hydrogel system with easy availability and low cost. In vitro and in vivo results showed that the newly designed hydrogel was biocompatible, realized successful sealing hemostasis under multiple bleeding scenarios and enabled accelerated healing of infected skin wounds.

Abstract:The skin, the primary barrier of the body, is inevitably broken. However, the development of materials that facilitate wound healing with sustained antimicrobial, hemostatic, and biocompatible properties remains a formidable challenge. In this article, we prepared a photopolymerizable composite hydrogel consisting of a hydrogel matrix, a hemostatic/antibacterial agent, and a photothermal therapy agent. The photopolymerizable hydrogel matrix was prepared by grafting the photoinitiator and polymerizable active monomer onto the chitosan chain segment, which exhibits excellent biocompatibility. Furthermore, linalool is adsorbed on the surface of halloysite nanotubes (HNTs) to form a hemostatic and antibacterial. Meanwhile, dopamine is employed as a coating material for hollow glass microsphere (HGM), which enables them to function as photothermal therapy agents. Upon exposure to near-infrared radiation, the PHA hydrogel releases linalool molecules from the surface of the HNTs, which diffuse into the hydrogel matrix, resulting in a sustained antimicrobial effect. At the same time, rapid curing of the photopolymerizable hydrogel under UV light forms a physical barrier that synergistically enhances the hemostatic properties of the HNTs. From the above, the results pave the way to develop a potential hemostatic antimicrobial dressing for clinical use in wound healing.

Abstract:Wound healing is a prolonged physiological process involving a series of complicated biological responses influenced by various factors, such as bacterial infection and oxidative damage. Existing treatments, while effective, have certain limitations in terms of biocompatibility, responsiveness to the wound environment, and efficacy in promoting tissue regeneration. To address these shortcomings, bioinspired by hydration lubrication mechanism of articular cartilage, we successfully designed and developed an injectable in situ photo-cross-linkable hydrogel, which was synthesized via modifying aminated hyaluronic acid with a polymer obtained by free radical polymerization of o-nitrobenzyl alcohol derivative (NB) and 2-methoxyethyl phosphocholine (MPC). The hydrophilic hydration layer formed around the zwitterionic charges in MPC endowed the hydrogel with enhanced lubrication and antimicrobial effects, whereas the hydroxymethyl group in NB enabled the hydrogel to process ultraviolet light-induced cross-linking property. The hydrogel, doped with polydopamine nanoparticles, facilitated multifaceted wound healing by the mechanism of photothermally responsive antibacterial, anti-inflammation, and angiogenesis. Specifically, upon injection of the hydrogel precursor solution into the wound site and exposure to ultraviolet light, the hydrogel cross-linked and adhered closely to the skin tissues, achieving hemostasis and exudate absorption. Additionally, the hydrogel showed excellent reactive oxygen species scavenging property, reducing cellular oxidative stress and meanwhile promoting the bactericidal effect with near-infrared light irradiation. Furthermore, the hydrogel, with its networked porous structure, was conducive to cell adhesion and proliferation, remarkably enhancing the cellular activity and tissue regeneration. The wound-healing performances of the hydrogel were investigated in both in vitro and in vivo experiments, showcasing its potential for wound treatment applications, offering a promising improvement over the current methods by providing a photothermal responsive, biocompatible, and effective healing process.
Versatile Hydrogel Dressing with Skin Adaptiveness and Mild Photothermal Antibacterial Activity for Methicillin‐Resistant Staphylococcus Aureus‐Infected Dynamic Wound Healing

Abstract:A versatile hydrogel conferred skin adaptiveness on dynamic wound healing, including flexible mechanical properties, strong tissue adhesion and excellent self‐healing ability. Moreover, the design of specific bacterial capture, membrane puncture and enhanced near‐infrared permeability, produced a safe and efficient mild PTT antibacteria against methicillin‐resistant Staphylococcus aureus and accelerated the tissue regeneration of MRSA‐infected wounds. Bacterial infection often induces chronic repair of wound healing owing to aggravated inflammation. Hydrogel dressing exhibiting intrinsic antibacterial activity may substantially reduce the use of antibiotics for infected wound management. Hence, a versatile hydrogel dressing (rGB/QCS/PDA–PAM) exhibiting skin adaptiveness on dynamic wounds and mild photothermal antibacterial activity is developed for safe and efficient infected wound treatment. Phenylboronic acid‐functionalized graphene (rGB) and oxadiazole‐decorated quaternary carboxymethyl chitosan (QCS) are incorporated into a polydopamine–polyacrylamide (PDA–PAM) network with multiple covalent and noncovalent bonds, which conferred the hydrogel with flexible mechanical properties, strong tissue adhesion and excellent self‐healing ability on the dynamic wounds. Moreover, the glycocalyx‐mimicking phenylboronic acid on the surface of rGB enables the hydrogel to specifically capture bacteria. The enhanced membrane permeability of QCS enhanced bacterial vulnerability to photothermal therapy(PTT), which is demonstrated by efficient mild PTT antibacteria against methicillin‐resistant Staphylococcus aureus in vitro and in vivo at temperatures of <49.6 °C. Consequently, the hydrogel demonstrate accelerated tissue regeneration on MRSA‐infected wound in vivo, with an intact epidermis, abundant collagen deposition and prominent angiogenesis. Therefore, rGB/QCS/PDA–PAM is a versatile hydrogel dressing exhibiting inherent antibacterial activity and has considerable potential in treating wounds infected with drug‐resistant bacteria.

Injectable Photothermal PDA/Chitosan/β‐Glycerophosphate Thermosensitive Hydrogels for Antibacterial and Wound Healing Promotion

Abstract:Controlling infections while reducing the use of antibiotics is what doctors as well as researchers are looking for. As innovative smart materials, photothermal materials can achieve localized heating under light excitation for broad‐spectrum bacterial inhibition. A polydopamine/chitosan/β‐glycerophosphate (PDA/CS/β‐GP) temperature‐sensitive hydrogel with excellent antibacterial ability was synthesized in the study. Initially, the hydrogel has good biocompatibility. In vitro experiments reveal its non‐cytotoxic property when co‐cultured with gingival fibroblasts and non‐hemolytic capability. Concurrently, the in vivo biocompatibility is confirmed through liver and kidney blood markers and staining of key organs. Crucially, the hydrogel has excellent photothermal conversion performance, which can realize the photothermal conversion of hydrogel up to 3 mm thickness. When excited by near‐infrared light, localized heating is attainable, resulting in clear inhibition impacts on both Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli), with the inhibition rates of 91.22% and 96.69%, respectively. During studies on mice's infected wounds, it is observed that the hydrogel can decrease S. aureus' presence in the affected area when exposed to near‐infrared light, and also lessen initial inflammation and apoptosis, hastening tissue healing. These findings provide valuable insights into the design of antibiotic‐free novel biomaterials with good potential for clinical applications. This article is protected by copyright. Biocompatible gellan gum/sericin hydrogels containing halloysite@polydopamine nanotubes with hemostasis and photothermal antibacterial properties for promoting infectious wound repair

Abstract:Skin wounds accompanied by massive bleeding and bacterial infection can be life-threatening and pose a significant challenge to clinical wound management. Herein, we prepared gellan gum/sericin hydrogels containing PDA-encapsulated halloysite nanotubes () to achieve fast hemostasis and photothermally antibacterial effects. The morphology and compressive properties of the composite hydrogels were characterized, and the biocompatibility was investigated by hemocompatibility and cytocompatibility tests. The hemostatic properties were further evaluated in the rat liver hemorrhage and tail amputation model. The mechanical performance (compressive strength 353.2 KPa) and the hemostatic effect (30.33±3.68 s in the liver injury model and 28.33±2.35 s in the caudal amputation model) of the composite hydrogel were improved due to the introduction of In addition, the composite hydrogel rapidly heated under 808 nm near-infrared (NIR) light irradiation, making the hydrogel extremely effective against E. coli and S. aureus . Furthermore, the wound closure rate, granulation tissue thickness, collagen deposition, and expression of inflammation-associated cytokines (CD68) and angiogenesis-associated cytokines (CD31) were investigated in a rat full-thickness skin defect model infected with S. aureus. All results showed that this multifunctional hydrogel provided better healing effects, suggesting that the composite hydrogel has become a promising alternative for infected wound repair.

Abstract:Cur@AIE@MnO2/gel may open new avenues for the development of novel dressings for bacterial‐infected wounds. Cur@AIE@MnO2/gel can interfere with the metabolic process and genetic material repair process to impact bacteria proliferation and biofilm dispersal. Moreover, Cur@AIE@MnO2/gel can also up‐regulate anti‐inflammatory genes, down‐regulate pro‐inflammatory genes in macrophages, and facilitate the promotion of macrophage polarization from M1 to M2 to relieve inflammation. Bacterial infection at wound sites can easily induce robust inflammation. M1 macrophages at wound sites can cause cascade inflammatory reactions to mediate a vicious inflammation cycle. Meanwhile, the hypoxia situation further aggravates the destruction and contraction of blood vessels in the wound microenvironment, therefore, hindering the infected wound healing process. Here, a thermosensitive hydrogel is reported to solve the above challenges by eliminating bacteria based on the photothermal effect, regulating the inflammatory immune microenvironment, and providing oxygen (O2). In vitro and in vivo experiments confirmed that the hydrogel has great O2‐producing properties. Based on the excellent photothermal properties of AIEgens, the hydrogel has a good bactericidal effect against S. aureus (100%) and E. coli (98.85%). The RNA‐sequencing revealed that Cur@AIE@MnO2/gel can interfere with the metabolic process and genetic material repair process to impact bacteria proliferation and biofilm dispersal, thus achieving excellent anti‐bacterial efficacy. Moreover, Cur@AIE@MnO2/gel can also up‐regulate anti‐inflammatory genes, down‐regulate pro‐inflammatory genes in macrophages, and facilitate the promotion of macrophage polarization from M1 to M2 to relieve inflammation. The therapeutic experiments show that this hydrogel accelerated the wound‐healing process. Nanocomposite Multifunctional Hyaluronic Acid Hydrogel with Photothermal Antibacterial and Antioxidant Properties for Infected Wound Healing

Abstract:Bacterial infection and subsequent reactive oxygen species (ROS) damage are major factors that delay wound healing in infected skin. Recently, photothermal therapy (PTT), as a new antibacterial method, has shown great advantages in the treatment of infected skin wound. Antibacterial and antioxidant hydrogels can reduce bacterial colonization and infection, scavenge ROS, relieve inflammation, and accelerate wound healing. In this study, an enzyme-crosslinked hyaluronic acid-tyramine (HT) hydrogel loaded with antioxidant and photothermal silver nanoparticles (AgNPs), named HTA, was developed as functional wound dressing to promote the infected skin wound healing. Natural antioxidant tannic acids (TA) were used as both reducing and stabilizing agents to facilely synthesize the silver nanoparticles capped with TA (AgNPs@TA). The incorporation of AgNPs@TA significantly enhanced the antioxidant, antibacterial, photothermal antibacterial, adhesive, and hemostatic abilities of the resulted HTA hydrogel. Besides, HTA hydrogel has rapid gelation, well injection and biocompatibility. In vivo results on the Staphylococcus aureus and Escherichia coli co-infected mouse skin wound model showed that HTA0.4 (containing 0.4 mg/mL AgNPs@TA) hydrogel combined with near infrared ray radiation highly alleviated inflammation, promoted angiogenesis, and accelerated the healing process. Therefore, this nanocomposite hydrogel wound dressing with antibacterial and antioxidant capabilities has great application potential in the treatment of infected skin wounds.

Abstract:Antibacterial antioxidant adhesion hydrogel was obtained by photopolymerization.These hydrogels exhibited good hemostatic property and cell compatibility.The hydrogels showed good antibacterial property against MRSA.The hydrogels significantly enhanced wound healing of infected skin wound.With the increasing prevalence of drug-resistant bacterial infections and the slow healing of chronically infected wounds, the development of new antibacterial and accelerated wound healing dressings has become a serious challenge. In order to solve this problem, we developed photo-crosslinked multifunctional antibacterial adhesive anti-oxidant hemostatic hydrogel dressings based on polyethylene glycol monomethyl ether modified glycidyl methacrylate functionalized chitosan (CSG-PEG), methacrylamide dopamine (DMA) and zinc ion for disinfection of drug-resistant bacteria and promoting wound healing. The mechanical properties, rheological properties and morphology of hydrogels were characterized, and the biocompatibility of these hydrogels was studied through cell compatibility and blood compatibility tests. These hydrogels were tested for the in vitro blood-clotting ability of whole blood and showed good hemostatic ability in the mouse liver hemorrhage model and the mouse-tail amputation model. In addition, it has been confirmed that the multifunctional hydrogels have good inherent antibacterial properties against Methicillin-resistant Staphylococcus aureus (MRSA). In the full-thickness skin defect model infected with MRSA, the wound closure ratio, thickness of granulation tissue, number of collagen deposition, regeneration of blood vessels and hair follicles were measured. The inflammation-related cytokines (CD68) and angiogenesis-related cytokines (CD31) expressed during skin regeneration were studied. Abstract:Designing adhesive hydrogel wound dressings with inherent antibacterial and antioxidant properties is desirable to treat cutaneous full-thickness injuries in clinical care. Herein, a series of photo-induced Schiff base crosslinking-based adhesive hydrogels with promising traits are designed and prepared through Diels-Alder (DA) reactions between functional groups-grafted carboxymethyl chitosan (CMCS) and a photo-responsive polyethylene glycol (PEG) crosslinker. The quaternary ammonium and phenol groups in modified CMCS endows hydrogels excellent antibacterial and antioxidant properties. Upon UV (365 nm) irradiation, the generated o-nitrosobenzaldehyde from the photo-isomerization of o-nitrobenzyl in PEG derivative can subsequently crosslink with amino groups on tissue interfaces via Schiff base, endowing the hydrogel with well adhesiveness. Additionally, the hydrogel exhibits good BSA adsorption capacity, cytocompatibility and hemostatic property. The in vivo full-thickness skin defect study on mice indicates that the multi-functional hydrogel with considerable collagen deposition and vascularization capacities can be an effective and promising adhesive dressing for improving wound healing.

Abstract:Skin damage is one of the most prevalent human injuries, which affects the health of human beings. However, skin damage is often accompanied by bacterial infection and wound microenvironment changes, causing damage to normal cells and inhibiting wound healing. Herein, we designed a thermal-responsive antibacterial hydrogel (GAG hydrogel) loaded with catalase (CAT)-like Au@Pt@MgSiO3 nanoparticles (APM NPs) and gentamicin (GM) to promote wound healing. The GAG hydrogel was used in a photothermal therapy (PTT)/antibiotic combination to kill bacteria, reduce the use of antibiotics, improve the wound microenvironment, promote cell proliferation, and accelerate wound healing. Under near-infrared laser irradiation, APM NPs in the hydrogel generated local hyperthermia to kill bacteria. Meanwhile, the generated heat led to a change in the hydrogel's morphology, enabling it to release GM and APM NPs to prevent the overuse of antibiotics. Subsequently, the CAT-like ability of the APM NPs decreased the oxidative stress caused by hydrogen peroxide (H2O2), thus remodeling the wound microenvironment. Then, the weakly acidic microenvironment of the wound caused the decomposition of the APM NPs and the release of magnesium ions (Mg2+), promoting the growth and migration of cells for wound healing. Therefore, the studied thermal-responsive antibacterial (GAG) hydrogel has potential in the field of wound healing.

Abstract:Bacterial infection in the most mobile area usually leads to delayed healing and functional restriction, which has been a long-term challenge in clinic. Developing hydrogel-based dressings with mechanical flexibly, high adhesive and anti-bacterial properties, will contribute to the healing and therapeutic effects especially for this typical skin wound. In this work, composite hydrogel named PBOF through multi-reversible bonds between polyvinyl alcohol, borax, oligomeric procyanidin and ferric ion demonstrated a 100 times ultra-stretch ability, 24 kPa of highly tissue-adhesive, rapid shape-adaptability within 2 min and self-healing feature within 40 s, was designed as the multifunctional wound dressing for the Staphylococcus aureus -infected skin wound in the mice nape model. Besides, this hydrogel dressing could be easily removed on-demand within 10 min by water. The rapid disassembly of this hydrogel is related to the formation of hydrogen bonds between polyvinyl alcohol and water. Moreover, the multifunctional properties of this hydrogel include strong anti-oxidative, anti-bacteria and hemostasis derived from oligomeric procyanidin and photothermal effect of ferric ion/polyphenol chelate. The killing ratio of the hydrogel on Staphylococcus aureus in infected skin wound reached 90.6% when exposed to 808 nm irradiation for 10 min. Simultaneously, reduced oxidative stress, suppressed inflammation, and promoted angiogenesis all together accelerated wound healing. Therefore, this well-designed multifunctional PBOF hydrogel holds great promise as skin wound dressing especially in the high mobile regions of the body. Statement of Significance : An ultra-stretchable, highly tissue-adhesive, and rapidly shape-adaptive, self-healing and on-demand removable hydrogel based on multi-reversible bonds among polyvinyl alcohol, borax, oligomeric procyanidin and ferric ion is designed as dressing material for infected wound healing in the movable nape. The rapid on-demand removal of the hydrogel relates to the formation of hydrogen bonds between polyvinyl alcohol and water. This hydrogel dressing shows strong antioxidant capacity, rapid hemostasis and photothermal antibacterial ability. This is derived from oligomeric procyanidin and thephotothermal effect of ferric ion/polyphenol chelate, which eliminates bacterial infection, reduces oxidative stress, regulates inflammation, promotes angiogenesis, and finally accelerates the infected wound healing in movable part.

Abstract:Infection caused by pathogenic microorganisms is an important obstacle to wound healing in clinical practice. Antibacterial hydrogel dressing is an ideal treatment strategy for wound infection. Here, we develop a highly stretchable, skin-adhesive and absorbent hydrogel dressing by simple one-step synthesis of composite polyacrylamide (PAM) hydrogel containing phenylboronic acid (BA) and PEDOT:PSS polymer. The resulting PAM/BA/PEDOT (PBP) hydrogel not only shows outstanding photothermal heating property but also possesses good peroxidase-like activity due to the existence of PEDOT:PSS. The addition of BA improves the water absorption of the network structure of the PBP hydrogel, which provides convenience for the absorption of wound exudates. Moreover, the PBP hydrogel has remarkable skin adhesion, toughness and tensile properties. In vitro assays demonstrate the significant photothermal/chemodynamic (PTT/CDT) antibacterial ability of the PBP hydrogel to both Gram-positive and Gram-negative bacteria. Furthermore, in vivo antibacterial results indicate that the PBP hydrogel can effectively treat the skin wound infection and promote the wound healing. Taken together, the PBP hydrogel displays great potential as an antibacterial dressing for synergistic anti-infective therapy in vivo.

Abstract:Massive bleeding and infectious complication remain the leading cause of worldwide trauma deaths. To tackle this issue, a platelet-inspired degradable antibacterial hydrogel (Gel/PP-TA-Ag) is synthesized based on gelatin methacrylate (GelMA), tannic acid (TA), polyphosphate (PolyP) and gallic acid functionalized silver nanoparticles (Ag NPs) via a facile photopolymerization process. Accompanied by the degradation of the hydrogel, the released PolyP can activate the coagulation pathway, resulting in a better hemostatic effect than commercial gauze in the mice-bleeding model. Simultaneously, benefiting from hyperthermia originated from Ag NPs and photothermal accelerated release of TA and Ag+, 97.57% of methicillin-resistant Staphylococcus aureus (MRSA) and 95.99% of E. coli are eliminated in vitro. Moreover, in vivo experiments reveal that 91.76% of MRSA in wounds is removed, and the wound healing is effectively improved with more angiogenesis and collagen deposition. The level of inflammation in the wounds is also significantly reduced. As a result, Gel/PP-TA-Ag hydrogel possesses great potential in achieving satisfactory efficacy in infected wound healing.

Abstract:Hydrogel dressings that can fit irregular wounds, promote wound healing, and detach from wounds without damage represent the development trend of modern medical dressings. Herein, a novel composite hydrogel with excellent wound shape matching and painless removability via a gel-sol phase transition is constructed through dynamic borate ester bonds between phenylboronic acid-grafted F127 (PF127) and polydopamine-coated reduced graphene oxide/silver nanoparticles (rGO@PDA/Ag NPs). After contact with the skin tissues, the administered liquid-like sols gradually transform into solid-like gels, robustly adhering to the wound. The hydrogel dressings containing near-infrared (NIR)-responsive rGO@PDA and in situ formed Ag NPs can generate localized heat and gradually release Ag+ to realize safe, effective, and durable photothermal-chemical combined sterilization. In addition, catechol-rich PDA endows the hydrogel dressings with good antioxidant activity and adhesiveness. In vivo study results indicate that the hydrogel dressings can significantly accelerate full-thickness skin infected wound healing by eliminating bacteria, promoting collagen deposition and angiogenesis, as well as reducing inflammation. Collectively, the thermoreversible rGO@PDA/Ag-PF127 hydrogel dressings with an improved self-adapting ability, superior antimicrobial activity, and tunable adhesion appear to be a promising candidate for the treatment of infected wounds.
