Abstract:This study was to investigate the antifatigue, prebiotic effects and their relationships to the structure properties of three ethanol precipitated polysaccharides from Dendrobium officinale (EPDO), as EPDO-40, EPDO-60 and EPDO-80. EPDOs with anti-fatigue activity were screened out by forced swimming test, and blood lactic acid (BLA), blood urea nitrogen (BUN), superoxide dismutase (SOD), liver glycogen, muscle glycogen, and intestinal microflora were investigated. Results showed that purified EPDO-60, 277.3 kDa, with a backbone consisted of 4-Man p and 4-Glc p . EPDO-60 had the best anti-fatigue activity, because it could significantly prolong the forced swimming time, as well as down-regulating the levels of BLA and BUN, increasing SOD. Proportions of Bacteroidetes and Firmicutes and abundance of Lactobacillus and Bifidobacterium in gut microflora increased after treated with EPDO-60. Accordingly, EPDO-60 could affect the community structure of gut microflora, leading to promote the balance of oxidation and antioxidation, and accelerated the fatigue metabolism in vivo .

Abstract:Dendrobium chrysotoxum has long been used as a health-promoting and therapeutic tonic in China. The polysaccharides from D. chrysotoxum (DCP) are considered to play an important role in its medical effects. The present study aimed at optimizing the technology for enzyme-assisted extraction (EAE) of DCP and at investigating the physicochemical characteristics and functional properties of DCP-E obtained by EAE and DCP-H obtained by hot water extraction (HVVE). Based on single factor tests and orthogonal experiments of extraction pH, extraction temperature, cellulase amount and extraction time, the optimum conditions for CP-E were extraction pH value of 5.5, extraction temperature of 40 degrees C, cellulase amount of 10 g/L, extraction time of 3.0 h and solid/liquid ratio of 1:25. Under these conditions, the yield of DCP-E was 8.41 g/100 g dry weight, which was 1.25 times of that of DCP-H. As compared to DCP-H, DCP-E displayed an increased purity, a decreased molecular weight and relative viscosity as well as a changed monosaccharide compositions. Foam stabilization and splenic cell proliferation experiments showed that DCP-E had higher cell proliferation rate while DCP-H had better foam stabilization activity. These results indicate that different extraction methods would influence the physicochemical characteristics and subsequently functional properties of polysaccharides, suggesting that the suitable extraction method should be selected according to the application intention of polysaccharides. Abstract:Dietary fiber is crucial for human health mainly due to its impact on gut microbiota structure and metabolites. This study aimed to investigate the impact of Dendrobium officinale polysaccharides (DOP) and two common fibers (β-glucan and inulin) on the gut microbiome structure and metabolic profile in vitro . Fecal samples were obtained from 30 healthy volunteers, which were then individually subjected to fermentation with each type of fiber. The results revealed that all fibers were efficiently degraded by gut microbiota, with DOP exhibiting a slower fermentation rate compared to β-glucan and inulin. The fermentation of all fibers led to a significant increase in the production of short-chain fatty acids (SCFAs) and a reduction in branched-chain fatty acids (BCFAs), sulfides, phenols, and indole. Moreover, the abundance of unclassified Enterobacteriaceae , which was positively correlated with sulfide, phenols, and indole levels, was significantly reduced by all fibers. Additionally, DOP specifically promoted the growth of Parabacteroides , while β-glucan and inulin promoted the growth of Bifidobacterium and Faecalibacterium . Taken together, these findings enhance our understanding of the role of DOP, β-glucan, and inulin in modulating gut microbiota and metabolites, where the fermentation with fecal bacteria from different volunteers could provide valuable insights for personalized therapeutic approaches.

Abstract:Oligosaccharides from Dendrobium officinale (DOO) is a kind of new potential prebiotic for health. In this study, structural characteristics, digestion properties and regulatory function on intestinal flora of DOO were investigated. An oligosaccharide, DOO 1-1, was purified by DEAE-Sepharose Fast Flow and Sephadex G-25, and its physicochemical properties were characterized as a glucomannan oligosaccharide with a molecular weight of 1560 Da (DP = 9). In vitro simulated digestion, it proved that the structure of DOO 1-1 was degraded hardly in the simulated gastric and small intestinal fluid. By evaluating the gas, short-chain fatty acids and intestinal microbiota in vitro fermentation, DOO has an excellent regulatory effect on intestinal microbiota, especially promoting the proliferation of Bacteroidetes and Actinobacteria. Therefore, DOO can be used as a potential prebiotic in functional foods.

Abstract:Gastritis is a common disease characterized by gastric ulcer and more severe bleeding. Daily intake of large amounts of alcohol can cause acute gastritis, which brings inconvenience to people's lives. This study aims to explore the protective effects of different ethanol-fractional polysaccharides of Dendrobium officinale (EPDO) on acute alcohol-induced gastric injury in vivo. Results showed that EPDO-80, which proved as a β-glucan, had significant anti-inflammatory properties in pathology. It could reduce the area of gastric mucosal injury and cell infiltration. EPDO-80 had a dose-effect relationship in reducing the levels of malondialdehyde and cyclooxygenase-2, and decreasing the levels of inflammation mediators such as tumor necrosis factor α. More extensively, EPDO-80 could inhibit the activation of TNFR/IκB/NF-κB signaling pathway, reducing the production of TNF-α mRNA and cell apoptosis in organ. On the other hand, EPDO-80 could promote the change of gut microbiota structure. These findings suggest that EPDO-80 might have great potential in limiting oxidative stress and inflammation mediated by inhibiting the NF-κB signaling pathway, which is highly related to its β-glucan structure and functions in gut microbiota.

Abstract:Ethanol fractional precipitation can initially separate polysaccharides according to the structure, which exhibits strong correlation with the biological activities. This study aimed to investigate the impact of varying ethanol concentrations on the structural characteristics, and the antitumor and antioxidant activities of polysaccharides derived from Dendrobium officinale through ethanol fractional precipitation, as well as their internal relationships. The polysaccharides acquired by absolute alcohol additions at a final liquor-ethanol volume ratio of 1:1, 1:2, and 1:4 were named DOP-1, DOP-2, and DOP-4, and the supernatant was named DOP-S. The results of the structural analysis revealed that the increase in ethanol concentrations resulted in a reduction in the molecular weights and the acetylation degree of the polysaccharides, as well as a decrease in mannose content and an increase in glucose content. In vitro experiments demonstrated that DOP-S exhibited optimal antitumor and antioxidant activities. Animal experiments further confirmed that DOP-S suppressed the growth of solid tumors significantly, enhanced lymphocytes, mediated immune ability, and improved the activity of antioxidant enzymes. These findings would establish a theoretical foundation and provide technical support for further advances and applications of polysaccharides derived from D. officinale in the fields of food and medicine.

Abstract:Fermentation is an effective method for enhancing the biological activity of polysaccharides, but research on its effect on Dendrobium officinal polysaccharides is rare. In this study, the effects of mono-fermentation (Saccharomyces cerevisiae FBKL2.8022, Sc; Wickerhamomyces anomalous FBKL2.8023, Wa) and co-fermentation (Sc+Wa) on the physicochemical properties and bioactivity of Dendrobium officinal polysaccharides were investigated. Meanwhile, the polysaccharide (DOP) obtained from Dendrobium officinale was used as a control. Four homogeneous polysaccharides were obtained by isolation and purification and named DOSCP, DOWAP, DOSWP, and DOP. The results showed that DOSCP, DOWAP, DOSWP, and DOP consisted of mannose and glucose with ratios of 3.31:1, 5.56:1, 2.40:1, and 3.29:1, respectively. The molecular weights (Mws) of the four polysaccharides were 25.73 kDa, 15.01 kDa, 17.67 kDa, and 1268.21 kDa. The antioxidant activity of DOSCP, DOWAP, and DOSWP was better than that of DOP. Additionally, all four polysaccharides were able to reduce the inflammatory response of LPS-induced RAW 264.7 macrophages in the mice without a significant difference. Yeast fermentation significantly reduced the molecular weight and improved the antioxidant activity of Dendrobium officinale polysaccharides, indicating a potential way to improve its antioxidant activity.

Abstract:Structural modification of polysaccharides using microbial fermentation is of great interest due to its specificity and environmental friendliness. The aim of this research was to compare structure, physicochemical properties and in vitro bioactivities of the Dendrobium officinale polysaccharides before and after fermentation using Lactobacillus paracasei and Lactobacillus acidophilus . The main fractions of crude polysaccharides from unfermented Dendrobium officinale (DOP-1A) and fermented Dendrobium officinale (FDOP-1A) were obtained by column chromatography purification. Compare to DOP-1A, the molecular weight of FDOP-1A was significantly reduced (from 412.566 to 25.161 kDa), smaller particle size (from 21.05 to 19.35 μm) and lower apparent viscosity. Although the microscopic surface morphologies of DOP-1A and FDOP-1A were significantly different, neither formed a triple-helix structure. Additionally, FDOP-1A contained more t-Man p and less 4-Man p, and interestingly the (1 → 4)- β -D-Glc p glycosidic bond site between the repeat units →4)-2- O -acetyl- β -D-Man p -(1 → 4)- β -D-Glc p -(1→ and →4)- β -D-Man p -(1 → 4)- β -D-Glc p -(1→ was cleaved. Moreover, FDOP-1A exhibited excellent scavenging capacity for ABTS + , DPPH, hydroxyl radicals and inhibited Nitric Oxide production by RAW 264.7 macrophages, thereby demonstrating effective antioxidant and anti-inflammatory effects. In conclusion, Lactobacillus fermentation could improve physicochemical properties and enhance biological activities of Dendrobium officinale polysaccharide, which has potential prospects for development and application in dietary supplements.

Abstract:Dendrobium officinale polysaccharides (DOPs) are important active polysaccharides found in Dendrobium officinale, which is commonly used as a conventional food or herbal medicine and is well known in China. DOPs can influence the composition of the gut microbiota and the degradation capacity of these symbiotic bacteria, which in turn may determine the efficacy of dietary interventions. However, the necessary analysis of the relationship between DOPs and the gut microbiota is lacking. In this review, we summarize the extraction, structure, health benefits, and related mechanisms of DOPs, construct the DOPs-host axis, and propose that DOPs are potential prebiotics, mainly composed of 1,4-β-D-mannose, 1,4-β-D-glucose, and O-acetate groups, which induce an increase in the abundance of gut microbiota such as Lactobacillus, Bifidobacterium, Akkermansia, Bacteroides, and Prevotella. In addition, we found that when exposed to DOPs with different structural properties, the gut microbiota may exhibit different diversity and composition and provide health benefits, such as metabolism regulations, inflammation modulation, immunity moderation, and cancer intervention. This may contribute to facilitating the development of functional foods and health products to improve human health.

Abstract:Peach gum polysaccharides have multiple biological functions, such as anti-oxidation, blood lipids reduction, and immune protection. However, their impact on the gut microbiota was still unclear. Here, high throughput sequencing was used to study the regulatory effects of peach gum polysaccharide on gut microbiota using an in vitro fermentation model of human gut microbiota. The results showed that gut microbiota species increased significantly and α-diversity index (Sobs, Ace, Chao1, Shannon) decreased significantly after polysaccharide fermentation compared with control group (distilled water). At phylum level, the relative abundance of Firmicutes and Actinomyces increased (61.76% vs. 56.64%, 28.24% vs. 23.11%). In contrast, relative abundance of Bacteroides decreased significantly (8.10% vs. 19.21%). At genus level, the relative abundance of Bifidobacteria, Colinella and Parazobacteria increased significantly (21.43% vs. 19.08%, 6.32% vs. 3.73%, 6.37% vs. 0.00%), while the relative abundance of Bacteroides and Prevotella decreased significantly (3.83% vs. 5.52%, 3.49% vs. 12.15%). Precisely, peach gum polysaccharide can effectively regulate the structure of gut microbiota and has potential probiotic effects including anti-obesity, anti-inflammatory, maintenance of gut epithelial barrier and so on.

Abstract:Introduction Lyophyllum decastes (Fr.) Singer polysaccharides (LDSPs) have been verified to possess strong biological properties. However, the effects of LDSPs on intestinal microbes and their metabolites have rarely been addressed. Methods The in vitro -simulated saliva-gastrointestinal digestion and human fecal fermentation were used to evaluate the effects of LDSPs on non-digestibility and intestinal microflora regulation in the present study. Results The results showed a slight increase in the content of the reducing end of the polysaccharide chain and no obvious change in the molecular weight during in vitro digestion. After 24 h in vitro fermentation, LDSPs were degraded and utilized by human gut microbiota, and LDSPs could be transformed into short-chain fatty acids leading to significant ( p < 0.05) decrease in the pH of the fermentation solution. The digestion did not remarkably affect the overall structure of LDSPs and 16S rRNA analysis revealed distinct shifts in the gut microbial composition and community diversity of the LDSPs-treated cultures, compared with the control group. Notably, the LDSPs group directed a targeted promotion of the abundance of butyrogenic bacteria, including Blautia , Roseburia , and Bacteroides , and an increase in the n-butyrate level. Discussion These findings suggest that LDSPs might be a potential prebiotic to provide a health benefit.

Abstract:Ethanol fractional precipitation is an important technology to classify polysaccharides at different molecular weights. In this study, polysaccharides of Dendrobium officinale were step-precipitated by final ethanol concentrations of 40%, 60% and 80%, named as EPDO-40, EPDO-60 and EPDO-80, respectively. Chemical composition, molecule weights and monosaccharides were identified. Effects of concentration, temperature, pH and metal ions on rheological properties were investigated. Results showed that all EPDOs behaved as non-Newtonian fluid in a certain shear rate range of 0.1–1000 s −1 which is affected by their different chemical characteristics. Viscosities of all polysaccharides showed a concentration dependent manner, and they decreased with the increased temperatures from 10-90 °C. Viscosities of EPDO-40 and EPDO-80 increased at a range of lower pH significantly, while that of EPDO-60 decreased. Na + , K + , Mg 2+ and Ca 2+ could all affect the viscosity of EPDOs but in different patterns. Accordingly, it demonstrated that EPDOs with different chemical characteristics had different rheological properties, which could be applied more accurately in foods industry.

Distinct prebiotic effects of polysaccharide fractions from Polygonatum kingianum on gut microbiota

Abstract:This study investigated the physicochemical properties, digestive stability, and in vitro fermentation behavior of Polygonatum kingianum polysaccharide (PKP) fractions (PKP60, PKP70, PKP80) obtained through graded ethanol precipitation. High-performance gel permeation chromatography revealed significant molecular weight differences among the fractions, while reverse-phase high-performance liquid chromatography indicated consistent monosaccharide types with variations in their proportions. Uronic acid analysis confirmed that all polysaccharide fractions met the criteria for neutral polysaccharides. Congo red staining confirmed the presence of a triple-helix structure in all PKP fractions. Comprehensive analysis demonstrated that these fractions remained stable during in vitro digestion, as evidenced by consistent molecular weights and total carbohydrate content, with no significant production of free monosaccharides or reducing sugars. All PKP fractions were fermented by gut microbiota, resulting in the production of short-chain fatty acids. Beta diversity and structural analyses of gut microbiota revealed distinct modulatory effects associated with each PKP fraction. The PKP fractions promoted probiotic growth, especially PKP70, which significantly enhanced Bifidobacterium proliferation, indicating strong prebiotic potential. These findings underscore the importance of isolation and purification methods in determining the functionality and gut microbiota-modulating effects of plant-derived polysaccharides, emphasizing the need for in-depth research that extends beyond merely evaluating their source.

Abstract:Recently, Dendrobium officinale polysaccharide (DOP), a typical acetylated glucomannan, has been widely applied in functional foods owing to its excellent bioactivity. However, the insufficiency of studies on in vivo process severely limits the further utilization of DOP. The aim of this study was to systematically investigate the gastrointestinal digestive behaviour of DOP after oral administration by labelling it with two fluorescein aminopyrene-1,3,6-trisulfonic acids, trisodium salt (APTS) and cyanine 7.5 (Cy7.5). Combining the results of NIR imaging and HPGPC, we found that DOP was poorly absorbed directly in the prototype form; instead, DOP moved with the intestinal contents to the distal part of the intestine, where Bacteroides aggregated for a prolonged time and was metabolized to oligosaccharide-like substances. In contrast, the digestive degradation of DOP in pseudo-sterile mice with a targeted clearance of Bacteroides significantly weakened, which provided the basis and direction for the subsequent search for more specific metabolic pathways of DOP in vivo.

Abstract:Ethnopharmacological relevance Gastrodia elata Blume is a traditional Chinese medicine with the effects of improving the deficiency of the body and maintaining health, and polysaccharide (GEP) is one of the effective ingredients to play these activities of G. elata . Traditionally, G. elata is orally administered, so the activities of GEP are associated with digestive and intestinal metabolism. However, the digestive behavior of GEP and its effects on the human gut microbiota are unclear and need to be fully studied. Aim of the study This study aimed to investigate the changes in structural characteristics of GEP during digestion and the related impacts of its digestive product on gut microbiota in human fecal fermentation, and to explain the beneficial mechanism of GEP on human health from the perspective of digestive characteristics and "gut" axis. Materials and methods The changes of reducing sugars, free monosaccharides and physicochemical properties of GEP during digestion were investigated by GPC, HPLC, FT-IR, CD, NMR, SEM, and TGA. Moreover, polysaccharide consumption, pH value changes, SCFAs production, and changes in gut microbiota during fermentation were also discussed. Results During digestion of GEP, glucose was partially released causing a decrease in molecular weight, and a change in monosaccharide composition. In addition, the characteristics of GEP before and after digestion, including configuration, morphology, and stability, were different. The digestive product of GEP was polysaccharide (GEP-I), which actively participated in the fecal fermentation process. As the fermentation time increased, the utilization of GEP-I by the microbiota gradually increased. The abundance of probiotics such as Bifidobacterium , Collinsella , Prevotella , and Faecalibacterium was significantly increased, and the abundance of pathogenic Shigella , Dorea , Desulfovibrio , and Blautia was significantly inhibited, thereby suggesting that GEP has the potential to maintain human health through the "gut" axis. In addition, the beneficial health effects of GEP-I have also been observed in the influence of microbial metabolites. During the fermentation of GEP-I, the pH value gradually decreased, and the contents of beneficial metabolites such as acetic acid, propionic acid, and caproic acid significantly increased. Conclusion The structure of GEP changed significantly during digestion, and its digestive product had the potential to maintain human health by regulating gut microbiota, which may be one of the active mechanisms of GEP.

Abstract:Polyonatum cyrtonema polysaccharides (PCPs) have been illustrated to have pharmacological and biological activity. However, the impact of PCPs on gut microorganisms and their metabolic pathways has been scarcely addressed. In order to investigate the antidigestion potential of PCPs, an in vitro simulation of digestion was developed. Moreover, an in vitro fermentation model was used to explore the influence of PCPs on the microbial community. The findings revealed that the concentration of the remaining carbohydrate and reducing end of the polysaccharide chain grew slightly, while the molecular weight was unaltered after being digested. Notably, the gut microbiota degraded the PCPs during the subsequent fermentation, resulting in the production of short-chain fatty acids and a significant decrease in the pH level ( p < 0.05 ). Compared with the control group, the result of 16S rRNA sequencing revealed that PCPs could affect the diversity of the community and the composition of the gut microbes significantly ( p < 0.05 ), having similar regulatory effects to inulin on the structure of the microbial community. The PCPs group notably promoted the relative abundance of acetic-acid bacteria, such as Bifidobacterium. In addition, PICRUSt2 analysis demonstrated that PCPs mainly enhanced phenotypic KEGG pathways such as amino acid metabolism and carbohydrate metabolism. Overall, PCPs may have prebiotic properties that support human health by controlling the make-up and metabolic activity of intestinal flora.

Structure and Fermentation Characteristics of Five Polysaccharides Sequentially Extracted from Sugar Beet Pulp by Different Methods

Abstract:Pectic polysaccharides can serve as the best alternative for prebiotics, as they can be fermented in the gut by the microbiota. Many reports suggest that pectic polysaccharides' fermentation characteristics are superior to commercially available prebiotics as Inulin. However, these researches have focused on commercial pectin commonly extracted from apples or citrus, while pectins can exhibit complex and diverse structures. In this study, we extracted and characterized five structurally different polysaccharides from sugar beet pulp by water, 1,2-Cyclohexanediaminetetraacetic acid (CDTA), sodium carbonate, and sodium hydroxide, and then assessed their impacts on the human fecal microbiome. CDTA-soluble fraction and sodium carbonate-soluble fraction were identified as pectic polysaccharides, while water-soluble fraction and sodium hydroxide-soluble fractions were rich in the arabinose side chain. All polysaccharides were fermented more easily than inulin. Some neutral sugars were more easily fermented than acid sugar, and arabinose may be the first monosaccharide to be utilized by the microbiota, followed by glucose, fucose and galacturonic acid. After 48h of fermentation, some health-promoting bacteria like Faecalibacterium, Bifidobacterium, Lactobacillus, Blautia, and Prevotella increased, while some pathogenic bacteria like Fusobacterium, Sutterella, and Dorea decreased in all polysaccharides and Inulin. Hereby, through the study of the correlation between monosaccharide composition and microbiota, we showed that monosaccharide composition might be a critical structural feature linked to the polysaccharide-microbiota modulating effect. This could be helpful to have a criterion to choose the polysaccharides to better modulate the microbiota.

Abstract:This study aimed to understand the in vitro simulated digestion and fecal fermentation behaviors of a pectic polysaccharide from okra (OPP-D) and its impacts on human gut microbiota. Results showed that the content of reducing sugars increased from 0.20 to 0.35 mg/mL during gastric digestion. The molecular weight of OPP-D slightly decreased from 2.242 × 105 to 1.932 × 105 Da during in vitro digestion, and then significantly decreased to 1.269 × 105 Da after in vitro fecal fermentation for 48 h. The apparent viscosity of OPP-D was overall stable during in vitro digestion, but significantly decreased during in vitro fecal fermentation. The constituent monosaccharides of OPP-D, including rhamnose, galacturonic acid, and galactose, were stable during in vitro digestion and fecal fermentation, but their molar ratios were changed from 1: 0.76: 1.82 to 1: 0.74: 2.08 during fecal fermentation for 48 h. The results indicate that OPP-D was partially degraded during in vitro digestion, and was significantly degraded and utilized by human gut microbiota. In addition, some bacteria, such as Bacteroides, Phascolarctobacterium, Megasphaera, and Desulfovibrio, significantly increased after fecal fermentation, suggesting that OPP-D could alter the composition and abundance of human gut microbiota. Moreover, OPP-D could promote the production of short-chain fatty acids during fermentation for 48 h. Especially, acetic acid increased from 11.45 to 19.71 mmol/L, and propionic acid increased from 2.59 to 8.02 mmol/L. In general, this study can be helpful to better understand the potential digestion and fermentation mechanism of OPP-D.

Abstract:Dictyophora indusiata is one of the most famous edible mushrooms in China. D. indusiata polysaccharide (DP) has attracted increasing attention because of its multiple beneficial effects. In this study, the in vitro simulated digestion and microbial fermentation were designed to reveal the potential catabolic property of DP and its impacts on the modulation of gut microbial composition. The results showed that the reducing sugar content, total polysaccharides content, molecular weight, and rheological property of DP were not significantly altered under in vitro simulated digestive conditions. However, the molecular weight, apparent viscosity, and total polysaccharides content of indigestible DP (DPI) significantly decreased during in vitro fecal fermentation, and the reducing sugar content and the release of free monosaccharides notably increased, suggesting that DP could be degraded and used by gut microbiota. Additionally, the relative abundances of several beneficial bacteria, such as Bacteroides, Catenibacterium, Parabacteroides, and Megamonas, increased significantly, indicating that DP can regulate the composition and abundance of gut microbiota. Moreover, DP could also promote the production of SCFAs, thus changing the acid–base environment of the large intestine. The results of this study are beneficial for deeply clarifying the catabolic behavior of DP in the gastrointestinal tract, which can provide a theoretical basis for developing microbiota-directed products based on DP.
