Abstract:Purpose To develop a new technique that enables simultaneous quantification of whole-brain T-1, T-2, T2*, as well as susceptibility and synthesis of six contrast-weighted images in a single 9.1-minute scan. Methods The technique uses hybrid T-2-prepared inversion-recovery pulse modules and multi-echo gradient-echo readouts to collect k-space data with various T1, T2, and T2* weightings. The underlying image is represented as a six-dimensional low-rank tensor consisting of three spatial dimensions and three temporal dimensions corresponding to T-1 recovery, T-2 decay, and multi-echo behaviors, respectively. Multiparametric maps were fitted from reconstructed image series. The proposed method was validated on phantoms and healthy volunteers, by comparing quantitative measurements against corresponding reference methods. The feasibility of generating six contrast-weighted images was also examined. Results High quality, co-registered T-1, T-2, and T2* susceptibility maps were generated that closely resembled the reference maps. Phantom measurements showed substantial consistency (R-2 > 0.98) with the reference measurements. Despite the significant differences of T-1 (p < .001), T-2 (p = .002), and T2* (p = 0.008) between our method and the references for in vivo studies, excellent agreement was achieved with all intraclass correlation coefficients greater than 0.75. No significant difference was found for susceptibility (p = .900). The framework is also capable of synthesizing six contrast-weighted images. Conclusion The MR Multitasking-based 3D brain mapping of T-1, T-2, T2*, and susceptibility agrees well with the reference and is a promising technique for multicontrast and quantitative imaging.

Abstract:Quantitative imaging has been very useful in neuroscientific and clinical applications, including glioma, tumor diagnosis and prognosis, brain maturation, and Alzheimer's disease. EPI is a powerful tool for quantitative imaging owing to its extremely fast acquisition. This work aims to develop a distortion-free, blip-up/down acquisition (BUDA) 3D-EPI with controlled aliasing in parallel imaging (CAIPI) sampling and joint Hankel low-rank image reconstruction for fast and robust multi-contrast high-resolution whole-brain imaging. The developed technique could generate distortion-free high-resolution whole-brain T2* mapping and quantitative susceptibility mapping in 47s at 1.1×1.1×1 mm 3 resolution.
3D-EPI Blip-Up/Down Acquisition (BUDA) with CAIPI and Joint Hankel Structured Low-Rank Reconstruction for Rapid Distortion-Free High-Resolution T2* Mapping

Abstract:PURPOSE:This work aims to develop a novel distortion-free 3D-EPI acquisition and image reconstruction technique for fast and robust, high-resolution, whole-brain imaging as well as quantitative T 2 * $$ {\mathrm{T}}_2^{\ast } $$ mapping.METHODS:3D Blip-up and -down acquisition (3D-BUDA) sequence is designed for both single- and multi-echo 3D gradient recalled echo (GRE)-EPI imaging using multiple shots with blip-up and -down readouts to encode B0 field map information. Complementary k-space coverage is achieved using controlled aliasing in parallel imaging (CAIPI) sampling across the shots. For image reconstruction, an iterative hard-thresholding algorithm is employed to minimize the cost function that combines field map information informed parallel imaging with the structured low-rank constraint for multi-shot 3D-BUDA data. Extending 3D-BUDA to multi-echo imaging permits T 2 * $$ {\mathrm{T}}_2^{\ast } $$ mapping. For this, we propose constructing a joint Hankel matrix along both echo and shot dimensions to improve the reconstruction.RESULTS:Experimental results on in vivo multi-echo data demonstrate that, by performing joint reconstruction along with both echo and shot dimensions, reconstruction accuracy is improved compared to standard 3D-BUDA reconstruction. CAIPI sampling is further shown to enhance image quality. For T 2 * $$ {\mathrm{T}}_2^{\ast } $$ mapping, parameter values from 3D-Joint-CAIPI-BUDA and reference multi-echo GRE are within limits of agreement as quantified by Bland-Altman analysis.CONCLUSIONS:The proposed technique enables rapid 3D distortion-free high-resolution imaging and T 2 * $$ {\mathrm{T}}_2^{\ast } $$ mapping. Specifically, 3D-BUDA enables 1-mm isotropic whole-brain imaging in 22 s at 3T and 9 s on a 7T scanner. The combination of multi-echo 3D-BUDA with CAIPI acquisition and joint reconstruction enables distortion-free whole-brain T 2 * $$ {\mathrm{T}}_2^{\ast } $$ mapping in 47 s at 1.1 × 1.1 × 1.0 mm3 resolution.

A separate real-time low-rank model incorporating time-resolved phase correction was also used to compensate for both inter-shot and intra-shot phase inconsistency induced by physiological motion. The proposed method was validated on both phantom and 16 healthy subjects. The quantification of T-1/T-2/ADC was evaluated for each case. Three post-surgery brain tumor patients were scanned for demonstration of clinical feasibility. Results Multitasking T-1/T-2/ADC maps were perfectly co-registered and free from image distortion. Phantom studies showed substantial quantitative agreement (R2=0.999) with reference protocols for T-1/T-2/ADC. In vivo studies showed nonsignificant T-1 (P = .248), T-2 (P = .97), ADC (P = .328) differences among the frontal, parietal, and occipital regions. Although Multitasking showed significant differences of T-1 (P = .03), T-2 (P < .001), and ADC (P = .001) biases against the references, the mean bias estimates were small (Delta T-1% < 5%, Delta T-2% < 7%, Delta ADC% < 5%), with all intraclass correlation coefficients greater than 0.82 indicating "excellent" agreement. Patient studies showed that Multitasking T-1/T-2/ADC maps were consistent with the clinical qualitative images. Conclusion The Multitasking approach simultaneously quantifies T-1/T-2/ADC with substantial agreement with the references and is promising for clinical applications.

Abstract:PurposeTo develop a free‐breathing, non‐electrocardiogram technique for simultaneous myocardial T1, T2, T2*, and fat‐fraction (FF) mapping in a single scan.MethodsThe MR Multitasking framework is adapted to quantify T1, T2, T2*, and FF simultaneously. A variable TR scheme is developed to preserve temporal resolution and imaging efficiency. The underlying high‐dimensional image is modeled as a low‐rank tensor, which allows accelerated acquisition and efficient reconstruction. The accuracy and/or repeatability of the technique were evaluated on static and motion phantoms, 12 healthy volunteers, and 3 patients by comparing to the reference techniques.ResultsIn static and motion phantoms, T1/T2/T2*/FF measurements showed substantial consistency (R > 0.98) and excellent agreement (intraclass correlation coefficient > 0.93) with reference measurements. In human subjects, the proposed technique yielded repeatable T1, T2, T2*, and FF measurements that agreed with those from references.ConclusionsThe proposed free‐breathing, non‐electrocardiogram, motion‐resolved Multitasking technique allows simultaneous quantification of myocardial T1, T2, T2*, and FF in a single 2.5‐min scan.

Abstract:Purpose: To develop a 3D multitasking multi-echo (MT-ME) technique for the comprehensive characterization of liver tissues with 5-min free-breathing acquisition; whole-liver coverage; a spatial resolution of 1.5 x 1.5 x 6 mm(3); and simultaneous quantification of T-1, water-specific T-1 (T-1w), proton density fat fraction (PDFF), and R-2*. Methods: Six-echo bipolar spoiled gradient echo readouts following inversion recovery preparation was performed to generate T-1, water/fat, and R-2* contrast. MR multitasking was used to reconstruct the MT-ME images with 3 spatial dimensions: 1 T-1 recovery dimension, 1 multi-echo dimension, and 1 respiratory dimension. A basis function-based approach was developed for T-1w quantification, followed by the estimation of R-2* and T-1-corrected PDFF. The intrasession repeatability and agreement against references of MT-ME measurements were tested on a phantom and 15 clinically healthy subjects. In addition, 4 patients with confirmed liver diseases were recruited, and the agreement between MT-ME measurements and references was assessed. Results: MT-ME produced high-quality, coregistered T-1, T-1w, PDFF, and R-2* maps with good intrasession repeatability and substantial agreement with references on phantom and human studies. The intra-class coefficients of T-1, T-1w, PDFF, and R-2* from the repeat MT-ME measurements on clinically healthy subjects were 0.989, 0.990, 0.999, and 0.988, respectively. The intra-class coefficients of T-1, PDFF, and in healthy subjects and 0.980, 0.999, and 0.998 in patients. The T-1w was independent to PDFF (R = -0.029, P = .904). Conclusion: The proposed MT-ME technique quantifies T-1, T-1w, PDFF, and R-2* simultaneously and is clinically promising for the comprehensive characterization of liver tissue properties.

Non-paradoxical paradoxes?

Abstract:1. It is commonly said that, whatever the exact nature of Lewis' paradoxes of strict implication, the only sense in which strict implication is paradoxical is the sense in which 'implies' means 'entails'. Critics of the identity-thesis, the thesis, namely that strict implication and entailment are one and the same, have not wanted to deny that Lewis' four puzzle theorems are true of, or hold for, material and strict implication. On this view, if you interpret therein the main connective, ' -3 ' , as 'materially implies' or 'strictly implies' (as opposed to 'entails') the air of paradox vanishes (Von Wright [8], p. 172). What they resist is construing the main connective of these theorems to be the entailment-connective. Fundamentally, they take the occurrence of the paradoxes conclusively to show that strict implication (for which the paradoxes are true) is not the same relation as entailment (for which the paradoxes are false). Here is one version of what we might call the no -conflict hypothesis. On the present version of it, we are entitled to say both that the paradoxes are true (and their proofs sound) and that our intuitions (on the basis of which we reject the paradoxes) about entailment are true; the tension between paradox and intuition is only apparent. The paradoxes reveal facts about strict implication; whereas our intuitions reflect truths about entailment. It is only when the facts about strict implication revealed by the paradoxes are thought to be facts about entailment, and when the truths about entailment reflected by our intuitions are thought to be truths about strict implication, that the illusion of incompatibility is created. But once it is recognized that we have in strict implication and entailment two distinct, albeit similar, relations, and that what holds for the one does not, in all respects, hold for the other, the illusion of conflict evaporates. We may therefore, without further anxiety, continue to hold that it is a law "of any reasonable modal logic that an impossible proposition strictly implies any proposition whatever, and that a necessary proposition is strictly implied by any proposition whatever."

Abstract:Purpose: This study aims to develop a high-resolution whole-brain multi-parametric quantitative MRI approach for simultaneous mapping of myelin-water fraction (MWF), T1, T2, and proton-density (PD), all within a clinically feasible scan time. Methods: We developed 3D ViSTa-MRF, which combined Visualization of Short Transverse relaxation time component (ViSTa) technique with MR Fingerprinting (MRF), to achieve high-fidelity whole-brain MWF and T1/T2/PD mapping on a clinical 3T scanner. To achieve fast acquisition and memory-efficient reconstruction, the ViSTa-MRF sequence leverages an optimized 3D tiny-golden-angle-shuffling spiral-projection acquisition and joint spatial-temporal subspace reconstruction with optimized preconditioning algorithm. With the proposed ViSTa-MRF approach, high-fidelity direct MWF mapping was achieved without a need for multi-compartment fitting that could introduce bias and/or noise from additional assumptions or priors. Results: The in-vivo results demonstrate the effectiveness of the proposed acquisition and reconstruction framework to provide fast multi-parametric mapping with high SNR and good quality. The in-vivo results of 1mm- and 0.66mm-iso datasets indicate that the MWF values measured by the proposed method are consistent with standard ViSTa results that are 30x slower with lower SNR. Furthermore, we applied the proposed method to enable 5-minute whole-brain 1mm-iso assessment of MWF and T1/T2/PD mappings for infant brain development and for post-mortem brain samples. Conclusions: In this work, we have developed a 3D ViSTa-MRF technique that enables the acquisition of whole-brain MWF, quantitative T1, T2, and PD maps at 1mm and 0.66mm isotropic resolution in 5 and 15 minutes, respectively. This advancement allows for quantitative investigations of myelination changes in the brain.

Abstract:Purpose To rapidly obtain high resolution T-2, T-2*, and quantitative susceptibility mapping (QSM) source separation maps with whole-brain coverage and high geometric fidelity. Methods We propose Blip Up-Down Acquisition for Spin And Gradient Echo imaging (BUDA-SAGE), an efficient EPI sequence for quantitative mapping. The acquisition includes multiple T-2*-, T-2 '-, and T-2-weighted contrasts. We alternate the phase-encoding polarities across the interleaved shots in this multi-shot navigator-free acquisition. A field map estimated from interim reconstructions was incorporated into the joint multi-shot EPI reconstruction with a structured low rank constraint to eliminate distortion. A self-supervised neural network (NN), MR-Self2Self (MR-S2S), was used to perform denoising to boost SNR. Using Slider encoding allowed us to reach 1 mm isotropic resolution by performing super-resolution reconstruction on volumes acquired with 2 mm slice thickness. Quantitative T-2 (=1/R-2) and T-2* (=1/R-2*) maps were obtained using Bloch dictionary matching on the reconstructed echoes. QSM was estimated using nonlinear dipole inversion on the gradient echoes. Starting from the estimated R-2/R-2* maps, R-2 ' information was derived and used in source separation QSM reconstruction, which provided additional para- and dia-magnetic susceptibility maps. Results In vivo results demonstrate the ability of BUDA-SAGE to provide whole-brain, distortion-free, high-resolution, multi-contrast images and quantitative T-2/T-2* maps, as well as yielding para- and dia-magnetic susceptibility maps. Estimated quantitative maps showed comparable values to conventional mapping methods in phantom and in vivo measurements. Conclusion BUDA-SAGE acquisition with self-supervised denoising and Slider encoding enables rapid, distortion-free, whole-brain T-2/T-2* mapping at 1 mm isotropic resolution under 90 s.

Abstract:Purpose To acquire whole brain T1-rho map under 5 minutes using single-shot acquisition and multi-band imaging. Introduction T1-rho reflects the spin-lattice relaxation in the rotation frame, it can be manipulated via the adjustment of the spin-lock time (TSL) and spin-lock field to provide varying contrast. T1 rho weighted images have shown superior contrast to conventional T1 and T2 in various applications such as cartilage, cardiac and tumor. In addition, T1 rho mapping has been shown to be a viable biomarker for detecting neurodegenerative diseases [1], which generally show little morphological changes. However, T1 rho acquisition is often constrained to single slice due to the long TSL needed, which makes the cross-examination with other measurements such as resting state fMRI difficult. In this work, we develop a rapid T1-rho mapping method that utilizes single-shot EPI acquisition and multi-band excitation that completes a 2mm isotropic whole brain T1 rho mapping within 5 minutes, which allows this acquisition to be added in a Parkinson disease related clinical study. Method Fig.1 shows the pulse sequence used in this study. A B0-B1 insensitive spin-lock module that uses a 180 refocus and a phase cycling scheme is used for the T1 rho preparation [2], followed by crusher gradients on three axes. The non-selective nature of the spin lock module allows multiple slices to be simultaneously acquired with a multiband excitation RF. A single shot spin echo EPI acquisition along with Z-axis gradient blipping scheme was used to introduce additional phase shift in k-space to generate a shifted-FOV aliasing pattern [3]. Figure 1 Slice multiplexing T1 rho weighted acquisition Experiment This method is used to be used in a Parkinson diseases studies and healthy volunteers were first scanned for verification. A total of 70 slices were needed to achieve whole brain coverage and an isotropic 2mm resolution. A slice multiplexing factor of 2 leads to 35 acquisitions in total. The acquisition matrix used was 128x128 with a TE of 24ms and a TR of 2000ms, and this gives an acquisition time of 1:10 per volume. Four TSL times of 20ms, 40ms, 60ms and 80ms were acquired at a spin-locking field of 400Hz for deriving the T1 rho map. Hence the overall acquisition time was 4:40 minutes. To investigate the effects of multiplexing of the T1rho map, multi-band acquisition and single band acquisition were compared. Results Fig.2a shows the resulting slice-multiplexed acquisition where the top half volume is shifted and overlapped with the bottom half volume. Fig.2b shows cross sections of the reconstructed T1 rho map obtained, and the numerical values agree with those published [1]. Fig.3c compares the T1-rho values of selected deep nuclei, WM and GM regions obtained from normal and slicemultiplexing acquisition, it shows that almost identical values were obtained although the acquisition time was halved. Discussion and conclusion In this work, single-shot EPI with slice-multiplexing was used to acquire whole brain T1 rho map within an acceptable time for clinical studies, which has great potential for further exploiting the values of T1-rho measurements in neurodegenerative diseases. There are two potential factors that may affect the quality of the slice multiplexing T1-rho map compared to conventional acquisitions: increased noise level attributed to g-factor and differing N/2 Nyquist ghosting correction for EPI. The former has been shown to have minimal effects due the favorable aliasing pattern received: actually only a small portion of the brain overlapped due to the geometry of human head (Fig2.a). The latter is related to the fact that the effects of eddy-current on different slices may be slightly different, for which the correction is generally performed on a per-slice basis. Currently it is performed based on the multiplexed slice, which could be sub-optimal. However no obvious residual ghost was observed thus far; and this may be improved by acquiring separate calibration data at different slice locations.
Efficient T-2 Mapping with Blip-Up/down EPI and Gslider-Sms (T-2-buda-gslider)

Abstract:Purpose: To rapidly obtain high isotropic-resolution T-2 maps with whole-brain coverage and high geometric fidelity. Methods: A T-2 blip-up/down EPI acquisition with generalized slice-dithered enhanced resolution (T-2-BUDA-gSlider) is proposed. A RF-encoded multi-slab spin-echo (SE) EPI acquisition with multiple TEs was developed to obtain high SNR efficiency with reduced TR. This was combined with an interleaved 2-shot EPI acquisition using blip-up/down phase encoding. An estimated field map was incorporated into the joint multi-shot EPI reconstruction with a structured low rank constraint to achieve distortion-free and robust reconstruction for each slab without navigation. A Bloch simulated subspace model was integrated into gSlider reconstruction and used for T-2 quantification. Results: In vivo results demonstrated that the T-2 values estimated by the proposed method were consistent with gold standard spin-echo acquisition. Compared to the reference 3D fast spin echo (FSE) images, distortion caused by off-resonance and eddy current effects were effectively mitigated. Conclusion: BUDA-gSlider SE-EPI acquisition and gSlider-subspace joint reconstruction enabled distortion-free whole-brain T-2 mapping in 2 min at similar to 1 mm(3) isotropic resolution, which could bring significant benefits to related clinical and neuroscience applications.

Abstract:PurposeTo develop a robust, accurate, and accelerated T1ρ quantification solution for submillimeter in vivo whole‐brain imaging.MethodsA multislice T1ρ mapping solution (MS‐T1ρ) was developed based on a two‐acquisition scheme using turbo spin echo with RF cycling to allow for whole‐brain coverage with 0.8‐mm in‐plane resolution. A compressed sensing–based fast imaging method, SCOPE, was used to accelerate the MS‐T1ρ acquisition time to a total scan time of 3 minutes 31 seconds. A phantom experiment was conducted to assess the accuracy of MS‐T1ρ by comparing the T1ρ value obtained using MS‐T1ρ with the reference value obtained using the standard single‐slice T1ρ mapping method. In vivo scans of 13 volunteers were acquired prospectively to validate the robustness of MS‐T1ρ.ResultsIn the phantom study, the T1ρ values obtained with MS‐T1ρ were in good agreement with the reference T1ρ values (R2 = 0.9991) and showed high consistency throughout all slices (coefficient of variation = 2.2 ± 2.43%). In the in vivo experiments, T1ρ maps were successfully acquired for all volunteers with no visually noticeable artifacts. There was no significant difference in T1ρ values between MS‐T1ρ acquisitions and fully sampled acquisitions for all brain tissues (p‐value > .05). In the intraclass correlation coefficient and Bland‐Altman analyses, the accelerated T1ρ measurements show moderate to good agreement to the fully sampled reference values.ConclusionThe proposed MS‐T1ρ solution allows for high‐resolution whole‐brain T1ρ mapping within 4 minutes and may provide a potential tool for investigating neural diseases.

Abstract:Quantitative assessment of brain myelination has gained attention for both research and diagnosis of neurological diseases. However, conventional pulse sequences cannot directly acquire the myelin-proton signals due to its extremely short T2 and T2* values. To obtain the myelin-proton signals, dedicated short T2 acquisition techniques, such as ultrashort echo time (UTE) imaging, have been introduced. However, it remains challenging to isolate the myelin-proton signals from tissues with longer T2. In this article, we extended our previous two-dimensional ultrashort echo time magnetic resonance fingerprinting (UTE-MRF) with dual-echo acquisition to three dimensional (3D). Given a relatively low proton density (PD) of myelin-proton, we utilized Cramér-Rao Lower Bound to encode myelin-proton with the maximal SNR efficiency for optimizing the MR fingerprinting design, in order to improve the sensitivity of the sequence to myelin-proton. In addition, with a second echo of approximately 3 ms, myelin-water component can be also captured. A myelin-tissue (myelin-proton and myelin-water) fraction mapping can be thus calculated. The optimized 3D UTE-MRF with dual-echo acquisition is tested in simulations, physical phantom and in vivo studies of both healthy subjects and multiple sclerosis patients. The results suggest that the rapidly decayed myelin-proton and myelin-water signal can be depicted with UTE signals of our method at clinically relevant resolution (1.8 mm isotropic) in 15 min. With its good sensitivity to myelin loss in multiple sclerosis patients demonstrated, our method for the whole brain myelin-tissue fraction mapping in clinical friendly scan time has the potential for routine clinical imaging.
Phase-sensitive deep reconstruction method for rapid multiparametric MR fingerprinting and quantitative susceptibility mapping in the brain

Abstract:Introduction: This study explores the potential of Magnetic Resonance Fingerprinting (MRF) with a novel Phase-Sensitivity Deep Reconstruction Network (PS-DRONE) for simultaneous quantification of T1, T2, Proton Density, B1+, phase and quantitative susceptibility mapping (QSM). Methods: Data were acquired at 3 T in vitro and in vivo using an optimized EPI-based MRF sequence. Phantom experiments were conducted using a standardized phantom for T1 and T2 maps and a custom-made agar-based gadolinium phantom for B1 and QSM maps. In vivo experiments included five healthy volunteers and one patient diagnosed with brain metastasis. PSDRONE maps were compared to reference maps obtained through standard imaging sequences. Results: Total scan time was 2 min for 32 slices and a resolution of [1 mm, 1 mm, 4.5 mm]. The reconstruction of T1, T2, Proton Density, B1+ and phase maps were reconstructed within 1 s. In the phantoms, PS-DRONE analysis presented accurate and strongly correlated T1 and T2 maps (r = 0.99) compared to the reference maps. B1 maps from PS-DRONE showed slightly higher values, though still correlated (r = 0.6) with the reference. QSM values showed a small bias but were strongly correlated (r = 0.99) with reference data. In the in vivo analysis, PS-DRONE-derived T1 and T2 values for gray and white matter matched reference values in healthy volunteers. PS-DRONE B1 and QSM maps showed strong correlations with reference values. Conclusion: The PS-DRONE network enables concurrent acquisition of T1, T2, PD, B1+, phase and QSM maps, within 2 min of acquisition time and 1 s of reconstruction time.
Fast Whole-Brain MR Multi-Parametric Mapping with Scan-Specific Self-Supervised Networks

Abstract:Quantification of tissue parameters using MRI is emerging as a powerful tool in clinical diagnosis and research studies. The need for multiple long scans with different acquisition parameters prohibits quantitative MRI from reaching widespread adoption in routine clinical and research exams. Accelerated parameter mapping techniques leverage parallel imaging, signal modelling and deep learning to offer more practical quantitative MRI acquisitions. However, the achievable acceleration and the quality of maps are often limited. Joint MAPLE is a recent state-of-the-art multi-parametric and scan-specific parameter mapping technique with promising performance at high acceleration rates. It synergistically combines parallel imaging, model-based and machine learning approaches for joint mapping of T1, T2*, proton density and the field inhomogeneity. However, Joint MAPLE suffers from prohibitively long reconstruction time to estimate the maps from a multi-echo, multi-flip angle (MEMFA) dataset at high resolution in a scan-specific manner. In this work, we propose a faster version of Joint MAPLE which retains the mapping performance of the original version. Coil compression, random slice selection, parameter-specific learning rates and transfer learning are synergistically combined in the proposed framework. It speeds-up the reconstruction time up to 700 times than the original version and processes a whole-brain MEMFA dataset in 21 minutes on average, which originally requires ~260 hours for Joint MAPLE. The mapping performance of the proposed framework is ~2-fold better than the standard and the state-of-the-art evaluated reconstruction techniques on average in terms of the root mean squared error.

Abstract:Genistein [4',5,7-trihydroxyisoflavone or 5,7-dihydroxy-3-(4-hydroxyphenyl)-4H-1-benzopyran-4-one] is a natural isoflavone occurring in many plants known to possess various biological activities, ranging from phyto-oestrogenic to antioxidative actions. Recent studies indicated that this isoflavone can also be considered as a drug for as yet untreatable genetic diseases. In the present review, we discuss a plausible use of genistein in treatment of two genetic disorders: CF (cystic fibrosis) and MPS (mucopolysaccharidosis). Although various biological actions of genistein are employed in these two cases, in vitro studies, tests on animal models and pilot clinical trials suggest that this plant-derived compound might be a real hope for patients suffering from severe inherited disorders with relatively complicated pathomechanisms, including those affecting the central nervous system.

Abstract:Purpose Phosphorus MRS (31P MRS) enables noninvasive assessment of energy metabolism, yet its application is hindered by sensitivity limitations. To overcome this, often high magnetic fields are used, leading to challenges such as spatial B1+ inhomogeneity and therefore the need for accurate flip‐angle determination in accelerated acquisitions with short TRs. In response to these challenges, we propose a novel short TR and look‐up table–based double‐angle method for fast 3D 31P B1+ mapping (fDAM). Methods Our method incorporates 3D weighted stack‐of‐spiral gradient‐echo acquisitions and a frequency‐selective pulse to enable efficient B1+ mapping based on the phosphocreatine signal at 7 T. Protocols were optimized using simulations and validated through phantom experiments. The method was validated in the human brain using a 31P 1Ch‐trasmit/32Ch‐receive coil and skeletal muscle using a birdcage 1H/31P volume coil. Results The results of fDAM were compared with the classical DAM. A good correlation (r = 0.95) was obtained between the two B1+ maps. A 3D 31P B1+ mapping in the human calf muscle was achieved in about 10:50 min using a birdcage volume coil, with a 20% extended coverage (number of voxels with SNR > 3) relative to that of the classical DAM (24 min). fDAM also enabled the first full‐brain coverage 31P 3D B1+ mapping in approximately 10:15 min using a 1Ch‐transmit/32Ch‐receive coil. Conclusion fDAM is an efficient method for 31P 3D B1+ mapping, showing promise for future applications in rapid 31P MRSI.

Highly Accelerated Multi-Shot Intravoxel Incoherent Motion Diffusion-Weighted Imaging in Brain Enabled by Parametric POCS-based Multiplexed Sensitivity Encoding

Abstract:Recently, intravoxel incoherent motion (IVIM) diffusion-weighted imaging (DWI) has also been demonstrated as an imaging tool for applications in neurological and neurovascular diseases. However, the use of single-shot diffusion-weighted echo-planar imaging for IVIM DWI acquisition leads to suboptimal data quality: for instance, geometric distortion and deteriorated image quality at high spatial resolution. Although the recently commercialized multi-shot acquisition methods, such as multiplexed sensitivity encoding (MUSE), can attain high-resolution and high-quality DWI with signal-to-noise ratio (SNR) performance superior to that of the conventional parallel imaging method, the prolonged scan time associated with multi-shot acquisition is impractical for routine IVIM DWI. This study proposes an acquisition and reconstruction framework based on parametric-POCSMUSE to accelerate the four-shot IVIM DWI with 70% reduction of total scan time (13 min 8 s versus 4 min 8 s). First, the four-shot IVIM DWI scan with 17 b values was accelerated by acquiring only one segment per b value except for b values of 0 and 600 s/mm2. Second, an IVIM-estimation scheme was integrated into the parametric-POCSMUSE to enable joint reconstruction of multi-b images from under-sampled four-shot IVIM DWI data. In vivo experiments on both healthy subjects and patients show that the proposed framework successfully produced multi-b DW images with significantly higher SNRs and lower reconstruction errors than did the conventional acceleration method based on parallel imaging. In addition, the IVIM quantitative maps estimated from the data produced by the proposed framework showed quality comparable to that of fully sampled MUSE-reconstructed images, suggesting that the proposed framework can enable highly accelerated multi-shot IVIM DWI without sacrificing data quality. In summary, the proposed framework can make multi-shot IVIM DWI feasible in a routine MRI examination, with reasonable scan time and improved geometric fidelity. The proposed reconstruction framework based on parametric-POCSMUSE can enable 3.2-fold scan acceleration compared with four-shot IVIM DWI with MUSE. In addition to being able to produce multi-b DW images with less distortion, it can also derive the IVIM quantitative maps with quality comparable to that of fully sampled four-shot IVIM DWI data.image
