Abstract:Aquatic wildlife health assessment is critically important for aquatic wildlife conservation. However, the health assessment of aquatic wildlife (especially aquatic wild animals) is difficult and often accompanied by invasive survey activities and delayed observability. As there is growing evidence that aquatic environmental microbiota could impact the health status of aquatic animals by influencing their symbiotic microbiota, we propose a non-invasive method to monitor the health status of wild aquatic animals using the environmental microbiota health index (microHI). However, it is unknown whether this method is effective for different ecotype groups of aquatic wild animals. To answer this question, we took a case study in the middle Yangtze River and studied the water environmental microbiota and fish gut microbiota at the fish community level, population level, and ecotype level. The results showed that the gut microHI of the healthy group was higher than that of the unhealthy group at the community and population levels, and the overall gut microHI was positively correlated with the water environmental microHI, whereas the baseline gut microHI was species-specific. Integrating these variations in four ecotype groups (filter-feeding, scraper-feeding, omnivorous, and carnivorous), only the gut microHI of the carnivorous group positively correlated with water environmental microHI. Alcaligenaceae, Enterobacteriaceae, and Achromobacter were the most abundant groups with health-negative-impacting phenotypes, had high positive correlations between gut sample group and environment sample group, and had significantly higher abundance in unhealthy groups than in healthy groups of carnivorous, filter-feeding, and scraper-feeding ecotypes. Therefore, using water environmental microHI to indicate the health status of wild fish is effective at the community level, is effective just for carnivorous fish at the ecotype level. In the middle Yangtze River, Alcaligenaceae, Enterobacteriaceae (family level), and Achromobacter (genus level) were the key water environmental microbial groups that potentially impacted wild fish health status. Of course, more data and research that test the current hypothesis and conclusion are encouraged.

Effects of Citrobacter freundii on sturgeon: Insights from skin mucosal immunology and microbiota

Abstract:Skin mucus analysis has recently been used as a non-invasive method to evaluate for fish welfare. The present research study was conducted to examine the skin mucosal immunity and skin microbiota profiles of sturgeons infected with Citrobacter freundii. Our histology results showed that the thickness of the epidermal layer of skin remained thinner, and the number of mucous cells was significantly decreased in sturgeons after infection (p < 0.05). Total protein, alanine aminotransferase, aspartate aminotransferase, superoxide dismutase, and creatine kinase levels in the mucus showed biphasic pattern (decrease and then increase). Lactate dehydrogenase, lysozyme, and acid phosphatase activities in the mucus showed an increasing trend after infection. Furthermore, 16S rRNA sequencing also revealed that C. freundii infection also affected the diversity and community structure of the skin mucus microbiota. An increase in microbial diversity (p > 0.05) and a decrease in microbial abundance (p < 0.05) after infection were noted. The predominant bacterial phyla in the skin mucus were Proteobacteria, Fusobacteria, Bacteroidetes, Firmicutes, and Actinobacteria. Specifically, the relative abundance of Fusobacteria increased after infection. The predominant bacterial genera in the skin mucus were Cetobacterium, Pelomonas, Bradyrhizobium, Flavobacterium, and Pseudomonas. The relative abundance of Cetobacterium, Pseudomonas, and Flavobacterium increased after infection. Our current research findings will provide new insights into the theoretical basis for future research studies exploring the mechanism of sturgeon infection with C. freundii.

Abstract:The anadromous Chinese sturgeon (Acipenser sinensis) is endangered and listed among the first class of protected animals in China. The possible causes for the decline of this species are the effects of synthetic chemicals, and loss of critical habitat. Chinese sturgeon in the Yangtze River have accumulated triphenyltin (TPT) to 31-128 ng/g wet weigh (ww) in liver, which is greater than the concentrations of tributyltin (<1.0 ng/g ww). Maternal transfer of TPT has resulted in concentrations of 25.5 +/- 13.0 ng/g ww in eggs of wild Chinese sturgeon, which poses a significant risk to the larvae naturally fertilized or hatched in the Yangtze River. The incidence of deformities in fry was 7.5%, with 1.2% of individuals exhibiting ocular abnormal development, and 6.3% exhibited skeletal/morphological deformations. The incidences of both ocular and skeletal/morphological deformations were directly proportional to the TPT concentration in the eggs of both the Chinese sturgeon and the Siberian sturgeon (Acipenser baerii) in controlled laboratory studies. The rates of deformities in the controlled studies were consistent with the rates caused at the similar concentrations in eggs collected from the field. Thus, TPT is the causal agent to induce the malformation of larvae of Chinese sturgeon. The incidence of deformed larvae of Chinese sturgeon is an indicator of overall population-level effects of TPT on Chinese sturgeon, because TPT at environmentally relevant concentrations can result in significantly decrease both quality and quantity of eggs and spawning frequency of fish.

Abstract:OBJECTIVETo evaluate the effects of water body environments on the microbial community of Oncomelania hupensis snails in marshlands of the eastern Dongting Lake where natural extinction of O. hupensis snails are found, so as to explore the correlation between the natural extinction of O. hupensis snails and the microbial community in snails.METHODSSnails were caged water bodies in the Qianliang Lake marshland (Qianliang Lake regions) where natural extinction of snails was found and in the Junshan Park marshland (Junshan Park regions) in the eastern Dongting Lake for 30 days, and then all snails were collected and identified for survival or death. DNA sequencing of the fungi and bacteria was performed in snails before and after immersion in waters, and the biodiversity and abundance were analyzed.RESULTSThe survival rates of O. hupensis snails were 28.0% (70/250) and 64.8% (162/250) in Qianliang Lake regions and Junshan Park regions 30 days after immersion in waters, respectively (χ2 = 81.365, P < 0.01). The number of the fungal community and the biodiversity of the bacterial community were both greater in snails caged in Qianliang Lake regions post-immersion than pre-immersion, and there was a significant difference in the structure of the fungal and bacterial communities. The microbial community with a significant difference included Flavobacteriaceae,which was harmful to O. hupensis snails.CONCLUSIONSThe water body environment affects the composition of the microbial community in O. hupensis snails in marshlands with natural snail distinction around the eastern Dongting Lake; however, further studies are required to investigate whether the natural distinction of snails is caused by water body environments-induced changes of the microbial spectrum in O. hupensis snails.

Abstract:Abstract Background Siberian sturgeon ( Acipenser baeri Brandt) and Beluga sturgeon ( Huso huso ) are two important commercial fish in China, and the feeding habits of them are very different. Diets and feeding habits are two significant factors to affect the gastrointestinal microbiota in fish. The intestinal microbiota has been reported to play a key role in nutrition and immunity. However, it is rarely reported about the relationship between the intestinal microbiota and feeding habits/diets on different Acipenseridae fish. This study is to comparative analysis of gut microbial community in Siberian sturgeon and Beluga sturgeon fed with the same diet/Beluga sturgeon fed with different diets in order to determine the effects of different feeding habits/diets on the fish intestinal microbiota. Results According to the experimental objectives, BL and BH groups were Beluga sturgeon ( Huso huso ) fed with low fishmeal diet and high fishmeal diet, respectively. SH group represented Siberian sturgeon ( Acipenser baeri Brandt) fed with the same diet as BH group. After 16 weeks feeding trial, the intestinal microbiota was examined by 16S rRNA high-throughput sequencing technology. On the phylum level, Proteobacteria and Bacteroidetes were significantly higher in BL group than BH group, and Cyanobacteria showed the opposite trend. Compared with BH group, Proteobacteria and Firmicutes were significantly increased in SH group, whereas Cyanobacteria were clearly decreased. At the genus level, Pseudomonas and Citrobacter in BL group were significantly higher comparing with BH group, while Bacillus , Luteibacter , Staphylococcus and Oceanobacillus was lower in BH group than SH group. Conclusions Alpha and beta diversities indicated that the intestinal microflora were significant difference between Siberian sturgeon and Beluga sturgeon when they fed with the same diet. Meanwhile, Beluga sturgeon fed with low fishmeal diet can increase the species diversity of intestinal microbiota than it fed high fishmeal diet. Therefore, feeding habits clearly affected the gastrointestinal microbiota of sturgeons. Moreover, the impact of changes in food on the gut microbiota of sturgeons should be taken into consideration during the process of sturgeon aquaculture.

Abstract:In fish conservation, it is important to simulate fish habitat suitability and determine the potential impact on habitat quality and species behavior. This study analyzed the impact of several hydraulic projects on fish habitat quality and spawning time by numerical modeling. The case study involved the only Chinese sturgeon spawning site in the Yangtze River, China. Four habitat assessment indexes reflected both habitat quality and spatial characteristics, including fragmentation and connectivity. Previous studies did not simulate the water temperature impact on the fish habitat; spawning time delay was attributed to water temperature increases and discharge decreases. The results of the present study showed that the water temperature increase from the Three Gorges Reservoir (TGR) impoundment primarily caused the habitat degradation and spawning time delay. The effect of the water temperature increase on the degradation of average habitat suitability was eight times that of a decrease in discharge. A 25-day delay in the water temperature trend line was significantly correlated with a 26-day delay in the first spawning date. A decrease in discharge after TGR impoundment did not cause the spawning date delay. The Dike Project mainly impacted spawning site connectivity. Controlling the water temperature of the released water discharge is the most important means of conserving the wild Chinese sturgeon. This research can benefit other regions considering hydraulic projects by identifying potential factors impacting fish habitat quality and overall ecosystem health.

Identification of the regulatory roles of water qualities on the spatio-temporal dynamics of microbiota communities in the water and fish guts in the Heilongjiang River

Abstract:The Heilongjiang River is one of the largest rivers in the cool temperate zone and has an abundant fish source. To date, the microbiota community in water samples and fish guts from the Heilongjiang River is still unclear. In the present study, water samples and fish guts were collected from four locations of the Heilongjiang River during both the dry season and the wet season to analyze the spatio-temporal dynamics of microbiota communities in the water environment and fish guts through 16s ribosome RNA sequencing. The water qualities showed seasonal changes in which the pH value, dissolved oxygen, and total dissolved solids were generally higher during the dry season, and the water temperature was higher during the wet season. RDA indicated that higher pH values, dissolved oxygen, and total dissolved solids promoted the formation of microbiota communities in the water samples of the dry season, while higher water temperature positively regulated the formation of microbiota communities in the water samples of the wet season. LEFSe identified five biomarkers with the most abundant difference at the genus level, of which TM7a was upregulated in the water samples of the dry season, and SM1A02, Rheinheimera, Gemmatimonas, and Vogesella were upregulated in the water samples of the wet season. Pearson analysis revealed that higher pH values and dissolved oxygen positively regulated the formation of TM7a and negatively regulated the formation of SM1A02, Rheinheimera, Gemmatimonas, and Vogesella (p < 0.05), while higher water temperature had the opposite regulatory roles in the formation of these biomarkers. The relative abundance of microbiota diversity in fish guts varies greatly between different fish species, even if the fishes were collected from the same water source, indicating that dietary habits and fish species may be key factors, affecting the formation and construction of microbiome community in fish gut. P. glenii, P. lagowskii, G. cynocephalus, and L. waleckii were the main fish resources, which were collected and identified from at least six sample points. RDA indicated that the microbiota in the water environment regulated the formation of microbiota community in the guts of G. cynocephalus and L. waleckii and had limited regulated effects on P. glenii and P. lagowskii. The present study identified the regulatory effects of water qualities on the formation of microbiota communities in the water samples and fish guts, providing valuable evidence for the protection of fish resources in the Heilongjiang River.

Abstract:Despite the rapid development of research on aquatic environment microbiota, limited attention has been paid to exploring the complex interactions between microbial communities and aquatic environments. Particularly, the mechanisms underlying fish diseases based on such dynamic interactions remain unknown. This study aimed to address the gap by conducting microbiome and co-occurrence network analyses on the typical freshwater aquaculture systems. High-throughput 16S rRNA gene sequencing results revealed significant differences in the microbiota between the disease and healthy groups. Notably, disease mortality varied consistently with the gradient of relative abundance of Proteobacteria (intestine, R 2 = 0.46, p < 0.05) and Cyanobacteria (gill, R 2 = 0.67, p < 0.01), indicating their potential use as diagnostic criteria. Furthermore, the elevated hepatosomatic index, NO 3 -N, COD and TC (sediment) were directly correlated with diseases (r > 0.54, p < 0.01). Mean concentrations of NO 3 -N, COD and TC were elevated by 78.87%, 25.63% and 44.2%, respectively, in ponds where diseases occurred. Quantitative analysis (qPCR) revealed that Aeromonas sobria infected hosts through a potential pathway of "sediment (4.4 × 10 5 copy number/g)-water (1.1 × 10 3 copy number/mL)-intestine (1.2 × 10 6 copy number/g)". Similarly, the potential route for Aeromonas veronii was sediment (4.9 × 10 6 copy number/g) to gill (5.1 × 10 5 copy number/g). Additionally, the complexity of microbial networks in the intestine, water, and sediment was significantly lower in the disease group, although no similar phenomenon was observed in the gill microbial network. In summary, these findings reveal that elevated concentrations of crucial environmental factors disrupt the linkages within microbiota, fostering the growth of opportunistic bacteria capable of colonizing fish gut or gills. This offers new insights into potential mechanisms by which environmental factors cause disease in fish.

Abstract:Heat is a common source of stress in aquatic environments and can alter the physiological and metabolic functions of aquatic animals, especially their intestinal function. Here, the effects of heat stress on the structure and function of the intestine and the characteristics of the intestinal microbiota were studied in sturgeon ( Acipenser baerii ♀ × Acipenser schrenckii ♂ hybrid F1). Sturgeons were exposed to sub-extreme (24°C) and extreme (28°C) high water temperatures for 12 days. The heat stress caused systemic damage to the intestine of sturgeons, which displayed severe enteritis in the valve intestine. The microbial diversity analysis showed that heat stress led to the disorder in intestinal microbiota, manifesting as an explosive increase in the abundance of thermophilic intestinal pathogens such as Plesiomonas , Cetobacterium , and Aeromonas and causing physiological dysfunction in the sturgeons. The disorder was followed by significant inhibition of intestinal digestion with reduced chymotrypsin, α-amylase, and lipase activities in the valve intestine and of antioxidant function with reduced peroxidase (POD) and catalase (CAT) activities. Simultaneously, heat stress reduced the thermal tolerance of sturgeons by reducing Grp75 expression and damaged the valve intestine’s repair ability with increased Tgf- β expression. The results confirmed that heat stress damaged the sturgeon intestines obviously and disturbed the intestinal microbiota, resulting in serious physiological dysfunction. The present study investigated the mechanism of the effect of heat stress on the sturgeon intestine and will help develop strategies to improve the resistance to thermal stress for wild and cultured sturgeons.

Abstract:Background/Objectives: The effect of sodium butyrate (NaB), β-glucan (βG) and vitamins in the diet on gut microbiome, cortisol level, lysozyme activity and growth parameters of juvenile hybrid sturgeon (Acipenser gueldenstaedtii♀ × Acipenser baerii♂) was determined. Methods: Sturgeon hybrids (n = 144) were divided into three groups with enriched feeding (mg/kg of feed): FQV1 (50 NaB; 20 βG; const. vitamins), FQV2 (150 NaB; 20 βG; const. vitamins), FQV3 (50 NaB; 60 βG; const. vitamins) and control (not supplemented), each group in triplicate, 12 fish in each repetition. Rearing was carried out for 30 days in controlled conditions. Gut microbiome was characterized using Next Generation Sequencing (NGS) of DNA samples isolated from intestinal content. Cortisol level was determined using the ELISA test. Lysozyme activity was measured by turbidimetric test. Results: Based on data obtained from NGS, it was determined that the FQV1 group is characterized by the highest values of diversity indices (Shannon, Simpson and Chao-1) and the largest number of ASVs (Amplicon Sequence Variants). The highest abundance of probiotic bacteria (Lactobacillus, Lactococcus) was determined in the FQV1 group. The highest cortisol concentration was determined in the control (33.26 ng/mL), while the lowest was in FQV3 (27.75 ng/mL). The highest lysozyme activity was observed in FQV1 (154.64 U/mL), and the lowest in FQV2 (104.39 U/mL) and control (121.37 U/mL) (p < 0.05). FQV2 was characterized by significantly more favorable values of breeding indicators (p < 0.05). Conclusions: The obtained results prove that an appropriate composition of NaB, βG and vitamins can be used in the commercial breeding of juvenile hybrid sturgeons.

Abstract:Cyanobacterial blooms are one of the most common stressors aquatic plants and animals encounter in freshwater ecosystems such as rivers and lakes. Following such outbreaks, some cyanobacteria release toxins, notably microcystins, which are highly toxic. Although numerous studies have explored the effects of microcystins on fish, their in-situ effects on the fish gut microbiome remain unexamined. Our objectives were to examine the fish gut microbiome before (Ju) and after (Au) cyanobacterial blooms and to monitor water quality. We collected Ju and Au fish gut and water samples from Hulun Lake. Using 16S rRNA full-length sequencing, we analyzed the composition, structure, and function of the fish gut microbiome. Results revealed significant disparities in microcystin concentrations between the Ju and Au water samples. In addition, the microcystin concentration in the carp gut was significantly higher than that in its muscle after the cyanobacterial bloom outbreak. Notably, the Au group demonstrated an increase in the microcystin level in water and a marked reduction in fish gut microbiota diversity compared with the Ju group. The cyanobacterial bloom decreased gut microbiome diversity in fish, with the Au group exhibiting a significantly reduced abundance of bacteria related to gut stability and microcystin degradation compared to the Ju group. Furthermore, we observed an upregulation of disease-associated bacterial metabolic functions in the Au group. In conclusion, these findings suggest that microcystins influence the composition and function of fish gut microbiota, improving our understanding of the interaction between fish gut microbiome and their environment. This study offers new perspectives on the adaptive mechanisms of aquatic organisms to cyanobacterial blooms.

Abstract:Chinese sturgeon (Acipenser sinensis) was listed as a Grade I protected animal in China in 1989, and the observed intersexual phenomenon and sex ratio deviation have suggested that chemicals have posed a risk as environment pollutants. This study analyzed seven musk fragrances in liver, muscle, heart, gonad, stomach, intestines, adipose, gill, pancreas, kidney, gallbladder, and roe from 13 female Chinese sturgeons, and the toxicokinetic behavior of musks were studied and compared with some organochlorines. Of the seven musks, HHCB, AHTN, and musk xylene were detected, and the highest concentrations were observed in adipose tissue: from 33.7 to 62.1 ng/g wet weight (ww), from 1.0 to 5.4 ng/g ww, and from 1.1 to 13 ng/g ww, respectively. Similar to the tissue distribution of DDTs and HCB, musks were detected frequently in high lipid content tissues such as roe, adipose, and liver, suggesting that tissue distribution of musks is controlled by the affinity to lipids. The concentration ratios based on lipid weight between roe and adipose were estimated to be 0.47 for HHCB, 0.58 for AHTN, and 0.51 for musk xylene, and those for the total DDTs and HCB were 0.27 and 0.61, which were relatively low compared with mammals. Relatively high concentrations of p,p'-DDE (68.4-449 ng/g ww) were detected in 10 of total 11 samples, which would cause the feminization of Chinese sturgeons during embryonic development. It was found that lipid-corrected concentrations of HHCB, AHTN, p,p'-DDE, and p,p'-DDD increased with age in female sturgeon, of which the trends were similar to those in fishes and different from those in mammals.
Biochemical, transcriptomic and metabolomic responses to total dissolved gas supersaturation and their underlying molecular mechanisms in Yangtze sturgeon (Acipenser dabryanus)

Abstract:With the rapid development of hydropower facility construction, the total dissolved gas (TDG) generated by dam discharge is seriously threatening the survival of fish and has become an ecological environmental issue of global concern. However, how TDG affects fish physiology and the underlying molecular mechanism remain poorly known. In this study, Acipenser dabryanus, an ancient living fossil that is a flagship species of the Yangtze River, was exposed to water supersaturated with TDG at a level of 116% for 48 h. A comprehensive analysis was performed to study the effect of TDG supersaturation stress on A. dabryanus, including histopathological, biochemical, transcriptomic and metabolomic analyses. The histopathological results showed that mucosal-associated lymphoid tissues were seriously damaged after TDG supersaturation stress. Plasma catalase levels increased significantly under TDG supersaturation stress, while superoxide dismutase levels decreased significantly. Transcriptomic analysis revealed 289 upregulated genes and 162 downregulated genes in gill tissue and 535 upregulated and 104 downregulated genes in liver tissue. Metabolomic analysis revealed 63 and 164 differentially abundant metabolites between the control group and TDG group in gill and liver, respectively. The majority of heat shock proteins and genes related to ubiquitin and various immune-related pathways were significantly upregulated by TDG supersaturation stress. Integrated transcriptomic and metabolomic analyses revealed the upregulation of amino acid metabolism and glycometabolism pathways under TDG supersaturation stress. Glycerophospholipid metabolism was increased which might be associated with maintaining cell membrane integrity. This is the first study revealing the underlying molecular mechanisms of effects of TDG supersaturation on fish. Our results suggested that acute TDG supersaturation stress could enhance immune and antioxidative functions and activate energy metabolic pathways as an adaptive mechanism in A. dabryanus.
Effects of Alkalinity Stress on Amino Acid Metabolism Profiles and Oxidative-Stress-Mediated Apoptosis/Ferroptosis in Hybrid Sturgeon (Huso dauricus ♀ × Acipenser schrenckii ♂) Livers

Abstract:Alkaline water is toxic to cultured aquatic animals that frequently live in pH-neutral freshwater. Overfishing and habitat destruction have contributed to the decline in the wild sturgeon population; consequently, the domestic hybrid sturgeon has become an increasingly important commercial species in China. Hybrid sturgeons are widely cultured in alkaline water, but little is known about the effects of alkalinity stress on hybrid sturgeon liver tissues. We exposed hybrid sturgeons to four alkaline concentrations (3.14 ± 0.02 mmol/L, 7.57 ± 0.08 mmol/L, 11.78 ± 0.24 mmol/L and 15.46 ± 0.48 mmol/L). Histopathology, biochemical index assessment, gene expression level detection and metabolomics analysis were used to investigate the negative effects on liver functions following exposure to NaHCO3. Livers exposed to alkaline stress exhibited severe tissue injury and clear apoptotic characteristics. With increased exposure concentrations, the hepatic superoxide dismutase, catalase, glutathione peroxidase and alkaline phosphatase activities significantly decreased in a dose-dependent manner. NaHCO3 exposure up-regulated the transcriptional levels of apoptosis/ferroptosis-related genes in livers. Similarly, the expression trends of interleukin-1β and heat shock protein genes also increased in high-alkalinity environments. However, the expression levels of complement protein 3 significantly decreased (p < 0.05). Hepatic untargeted metabolomics revealed the alteration conditions of various metabolites associated with the antioxidant response, the ferroptosis process and amino acid metabolism (such as beta-alanine metabolism; alanine, aspartate and glutamate metabolism; and glycine, serine and threonine metabolism). These data provided evidence that NaHCO3 impaired immune functions and the integrity of hybrid sturgeon liver tissues by mediating oxidative-stress-mediated apoptosis and ferroptosis. Our results shed light on the breeding welfare of domestic hybrid sturgeons and promote the economic development of fisheries in China.

Abstract:The mechanism by which an organism can adapt to subtle environmental changes is predicated on modifications to biochemical processes within the cellular metabolism in response to such changes. Changes in these processes have the potential to induce alterations in cellular structures and tissue organization, as well as establish a causal link between fluctuations in these parameters and stressors exposure. This investigation's main goal and innovation is to evaluate the environmental stress indicators in the aquatic ecosystem of Lake Qarun. Pesticide residues in freshwater fish should be the primary focus of evaluation of environmental stressor concentrations, since they serve as bioindicators at different times and places on a spatiotemporal scale. A thorough analysis of suggestive biochemical biomarker reactions should also be conducted. The effects of environmental stressors, specifically pesticide contamination in Qarun Lake, have been observed and investigated in relation to two fish species: Solea aejabtiaca and Oreochronis niloticus . The results of a hazard assessment conducted at six sampling sites using spatio-temporal data revealed elevated mean values for the pesticides, persistent organic pollutants (POPs), organochlorines, organophosphates, and pyrethroids that were detected. A multi biomarker approach facilitates a more comprehensive understanding of stress responses induced by exposure to pollutants. As a result, the activities of the biochemical biomarkers CYP-450, GST, GSH, and LDH in the blood and liver of fish samples were found to be notably elevated. The suitability of the identified variables for biomonitoring of aquatic pollution was validated, and the data unveiled variations in sensitivity among species, implying that Nile tilapia could potentially function as a bioindicator with high sensitivity. The findings were correlated with the concentrations of detrimental organochlorines, organophosphorus, and pyrethroids in the muscles and gills. The data indicates that pollutants linked to agricultural wastes, runoff, and municipal effluent may be discharged into the lake ecosystem. Consequently, to safeguard the environment, it is essential to enforce and implement policies, acts, and regulations that already exist. Assessing the effects of additional environmental stressors on aquatic ecosystems is another way in which biomarker screening with an integrative approach improves our comprehension of how toxicants impact various levels of biological organization and is particularly useful in realistic environmental exposure scenarios.

Effects of acclimation temperature on the thermal tolerance, hypoxia tolerance and swimming performance of two endangered fish species in China

Abstract:Temperature is one of the most important environmental factors affecting the physiological activities and, thus, the fitness of fish, and physiological studies can help predict the effects of climate change on fish species in the field. The aim of this study was to investigate the effect of acclimation temperature on the thermal tolerance, hypoxia tolerance and swimming ability of two endangered fish species in the upper reach of the Yangtze River, namely, the Chinese sucker (Myxocyprinus asiaticus) and rock carp (Procypris rabaudi). The fish were acclimated at either 15 °C or 25 °C for a 3-week period. Then, thermal tolerance as indicated by the critical thermal maximum (CTmax) and critical thermal minimum (CTmin), hypoxia tolerance as indicated by the aquatic surface respiration (ASR50) and loss of equilibrium (LOE50), swimming performance as indicated by the critical swimming speed (Ucrit), aerobic capacity as indicated by the maximum metabolic rate and aerobic scope were measured. As expected, the thermal indicators of both species increased with temperature, and their values at both acclimation temperatures were similar to those of fish living in the Yangtze River. However, both species showed poor hypoxia tolerance compared to most fish species in the Yangtze River, according to previous studies. In particular, Chinese sucker acclimated at a low temperature exhibited an unusually strong decrease in hypoxia tolerance with decreasing temperature (fish usually showed high hypoxia tolerance due to decreased oxygen demand and high environmental oxygen tension at low temperature). Furthermore, Chinese sucker exhibited poorer swimming performance than rock carp (which is also a relatively poor swimmer among the fish species in the Yangtze River) when maintained at a high temperature due to low swimming efficiency, possibly as a consequence of its deep body shape. The difference in Ucrit was magnified at low temperature due to the more profound decrease in metabolic scope in Chinese sucker than in rock carp (55% vs 20%), but Chinese sucker showed a higher resting metabolic rate than rock carp at a low temperature, which is difficult to explain. This result suggested that low hypoxia tolerance and poor swimming performance due to the low cardiorespiratory capacity and (or) non-streamlined body shape of both fish species, especially Chinese sucker, reared at low temperature might be two of the reasons why they are not well adjusted to the change in their natural habitat and have thus declined in recent decades. The underlying physiological and biochemical mechanisms involved in the unusual adjustment of the physiological function of Chinese sucker and its ecological relevance must be investigated further. The present study provides a good example of a physiological investigation yielding very interesting and useful data for species conservation in a changing world.

Abstract:Unusually deep water due to dam construction has the potential to negatively effect endangered sturgeons, which lack a physiological mechanism to inflate their swimbladder and may be unable to remain buoyant under high pressure at depth. In a previous study, some juvenile sturgeons released in a deep (>100 m) reservoir lost buoyancy and stayed nearly motionless on the bottom. However, it is not clear whether this behavior represents a negative effect of the dam, because natural sturgeon swimming behavior is unknown. In this study, we attached multi-sensor data loggers to nine wild adult Chinese sturgeons Acipenser sinensis in an unimpounded reach of the Yangtze River, China. The depth utilization, tail beating activity, swim speed, and body inclination of these fish were monitored for 1–3 days. Fish swam up and down successively in the water column (mean depth, 9.9 m) with a cycle of 100–1,000 s during 64 % of the time on average, and stayed at depth, presumably on the riverbed, during rest periods. Tail beats were continuous (mean frequency, 0.77 Hz) throughout the records, indicating that their buoyancy was maintained. These results contrast with the previous study, suggesting that the behavior observed in the reservoir is unusual and that deep water poses a risk of losing buoyancy for sturgeons. Furthermore, all fish intensively swam (maximum speed, 3.0 m s−1) to the river surface at a mean frequency of 0.35 times per hour—a behavior that could explain why ship strikes are a serious cause of mortality in sturgeons.
