Abstract:Polyurethane (PU) synthetic leathers possess an intricate plastic composition, including polyester (PET) base fabrics and upper PU resin, but the release of fragments from the complexes is unclear. Therefore, we investigated the photodegradation trends of PET base fabrics with PU coating (PET-U) as a representative of composite plastics. Attention was paid to the comparison of the photoaging process of PET-U with that of pure PET base fabric (PET-P). To reveal the potential for chain scission, physical and chemical changes (e.g., surface morphology, molecular weight, and crystallinity) of the two fabrics were explored. The generation of microplastic fibers (MPFs) and microplastic particles (MPPs) was distinguished. Compared with PET-P, PET-U showed a similar but delayed trend in various characteristics and debris release rate as the photoaging time prolonged. Even so, after 360 h of illumination, the generated number of MPs (including MPFs and MPPs) rose considerably to 9.32 × 107 MPs/g, and the amount of released nanoplastics (NPs) increased to 2.70 × 1011 NPs/g from PET-U. The suppression of MP formation from PET-U was potentially directed by the physical shielding of the upper PU layer and the dropped MPs, which resisted the photochemical radical effect. The components of dissolved organic matter derived from plastics (P-DOM) were separated by molecular weight using a size-exclusion chromatography-diode array detector-organic carbon detector/organic nitrogen detector (SEC-DAD-OCD/OND), and the results showed that a larger amount of carbon- and nitrogen-containing chemical substances were generated in PET-U, accompanied by more aromatic and fluorescent compounds. The results provided theoretical bases and insights for future research on the risks of plastic debris from PU synthetic leathers on aquatic organisms and indicated feasible directions for exploring combined pollution studies of plastics.
Biodegradation of Polyester Polyurethane by Cladosporium Sp. P7: Evaluating Its Degradation Capacity and Metabolic Pathways.

Abstract:Microorganisms capable of decomposing polyurethane (PU) and other plastics have the potential to be used in bio-recycling processes. In this study, 20 PU-degrading strains were isolated, including 11 bacteria and 9 fungi, using a synthesized poly(1,4-butylene adipate)-based PU (PBA-PU) as the screening substrate. Three PU substrates with increasing structure complexities were used for a thorough evaluation of microbial degradation capacity: Impranil® DLN-SD, PBA-PU film and PU foam waste. After 4 days, the best fungal PBA-PU degrader, Cladosporium sp. P7, could degrade 94.5% of Impranil® DLN-SD. After 28 days of cultivation, 32.42% and 43.91% of solid PBA-PU film was converted into soluble small molecules when used as the sole carbon source or in a medium with other co-carbon sources, respectively. Accordingly, the weight loss of PU foam waste after 15 days was 15.3% for the sole carbon condition and 83.83% for the co-carbon conditions. Furthermore, PBA-PU was used for metabolic pathway analysis because of its known composition and chemical structure. Six metabolites were identified during the degradation process of PBA-PU, including adipic acid (AA), 1,4-butanediol (BDO), and 4,4'-methylenedianiline (MDA), which can also be used as the sole carbon source to grow the fungal strain P7, resulting in the discovery of two MDA metabolites during the cultivation processes. Based on the presence of these eight metabolites, we hypothesized that PBA-PU is first depolymerized by the fungal strain P7 via ester and urethane bond hydrolysis, followed by intracellular metabolism and mineralization of the three monomers to CO2 and H2O.

Abstract:Streptomyces sp. PU10 degrades Impranil and funnels degradation intermediates into polyketide biosynthesis via the fatty acid degradation pathway. Global plastic waste accumulation has become omnipresent in public discourse and the focus of scientific research. Ranking as the sixth most produced polymer globally, polyurethanes (PU) significantly contribute to plastic waste and environmental pollution due to the toxicity of their building blocks, such as diisocyanates. In this study, the effects of PU on soil microbial communities over 18 months were monitored revealing that it had marginal effects on microbial diversity. However, Streptomyces sp. PU10, isolated from this PU‐contaminated soil, proved exceptional in the degradation of a soluble polyester‐PU (Impranil) across a range of temperatures with over 96% degradation of 10 g/L in 48 h. Proteins involved in PU degradation and metabolic changes occurring in this strain with Impranil as the sole carbon source were further investigated employing quantitative proteomics. The proposed degradation mechanism implicated the action of three enzymes: a polyester‐degrading esterase, a urethane bond‐degrading amidase and an oxidoreductase. Furthermore, proteome data revealed that PU degradation intermediates were incorporated into Streptomyces sp. PU10 metabolism via the fatty acid degradation pathway and subsequently channelled to polyketide biosynthesis. Most notably, the production of the tri‐pyrrole undecylprodigiosin was confirmed paving the way for establishing PU upcycling strategies to bioactive metabolites using Streptomyces strains.

Abstract:Microorganisms have the potential to be applied for the degradation or depolymerization of polyurethane (PU) and other plastic waste, which have attracted global attention. The appropriate strain or enzyme that can effectively degrade PU is the key to treat PU plastic wastes by biological methods. Here, a polyester PU-degrading bacterium Bacillus sp. YXP1 was isolated and identified from a plastic landfill. Three PU substrates with increasing structure complexities, including Impranil DLN, poly (1,4-butylene adipate)-based PU (PBA-PU), and polyester PU foam, were used to evaluate the degradation capacity of Bacillus sp. YXP1. Under optimal conditions, strain YXP1 could completely degrade 0.5% Impranil DLN within 7 days. After 30 days, the weight loss of polyester PU foam by strain YXP1 was as high as 42.1%. In addition, PBA-PU was applied for degradation pathway analysis due to its clear composition and chemical structure. Five degradation intermediates of PBA-PU were identified, including 4,4'-methylenedianiline (MDA), 1,4-butanediol, adipic acid, and two MDA derivates, indicating that strain YXP1 could depolymerize PBA-PU by the hydrolysis of ester and urethane bonds. Furthermore, the extracellular enzymes produced by strain YXP1 could hydrolyze PBA-PU to generate MDA. Together, this study provides a potential bacterium for the biological treatment of PU plastic wastes and for the mining of functional enzymes.

Abstract:The presence of microplastics (MPs) products and particles in the environment can significantly impact the human body. Most MPs that enter the environment also enter the water cycle. During sunlight light irradiation (especially ultraviolet (UV) part) or UV disinfection, many of these MPs, particularly those rich in surface functional groups like thermoplastic polyurethanes (TPU), undergo physicochemical changes that can affect the formation of disinfection byproducts (DBPs). This study investigates the physicochemical changes of TPU in water after exposure to UV irradiation and incubation in the dark, as well as the formation of DBPs after chlorination. The results show that TPU undergo chain breakage, oxidation, and cross-linking when exposed to UV irradiation in an aqueous system. This leads to fragmentation into smaller particles, which facilitates the synthesis of DBPs. Subsequent research has demonstrated that the TPU leaching solution produces a significantly higher DBP content than the chlorination of TPU MPs, particularly at high concentrations of CHCl3. Therefore, it is important to give greater consideration to the soluble DBP precursors released by TPU.

Abstract:Bisphenol A polycarbonate (BPA-PC) is a kind of widely used engineering plastics. However, excessive usage causes the production of plastic wastes, following property changes of polymers and high risks of released chemicals during outdoor weathering. In this study, we systematically investigated the photoaging behavior of PC microplastics (MPs) in aquatic environment and evaluated the potential risk of released intermediates. Light irradiation along with mechanical abrasion facilitated the fragmentation of PC MPs and stimulated photooxidative modification during 640 h of ultraviolet (UV) exposure. Continuous degradation of the polymer was accompanied with dramatic decline of molecular weight. Also, BPA was released from irradiated PC MPs with a trend of an initial rapid increase followed by a decrease versus the irradiation time, and the maximum concentration of dropped BPA was detected up to 652.80 ± 72.89 μg/g (43.39% and 56.61% respectively in particles and leachates). However, the releasing amount of BPA in the leachate merely occupied 2.7% of the total organic carbon (TOC) leached out, suggesting that a great number of unknown organic products were produced other than BPA. Liquid chromatography-time-of-flight-mass spectrometry (LC-TOF-MS) analysis showed that these organic compounds forming MPs-derived dissolved organic matter (MPs DOM) were partly composed of 4,4'-dihydroxybenzophenone (DHB), p-hydroxybenzoic acid (p-HBA) and methyparaben (MeP), which would also contribute to the estrogenic activity. The degradation pathway of PC MPs was elaborated with the photolysis process of PC dimer and BPA, and the remarkable photoaging of PC MPs was mainly dominated by the generated reactive oxygen species (ROS). The findings of this study indicated that understanding the photoaging process of PC MPs was vital to evaluate their integral cumulative estrogenic activity in aquatic environment, and further highlighted the notable possible risks of plastic leachates to exposed biota.

Abstract:Abstract The accumulation of microplastics in various ecosystems has now been well documented and recent evidence suggests detrimental effects on various biological processes due to this pollution. Accumulation of microplastics in the natural environment is ultimately due to the chemical nature of widely used petroleum-based plastic polymers, which typically are inaccessible to biological processing. One way to mitigate this crisis is adoption of plastics that biodegrade if released into natural environments. In this work, we generated microplastic particles from a bio-based, biodegradable thermoplastic polyurethane (TPU-FC1) and demonstrated their rapid biodegradation via direct visualization and respirometry. Furthermore, we isolated multiple bacterial strains capable of using TPU-FC1 as a sole carbon source and characterized their depolymerization products. To visualize biodegradation of TPU materials as real-world products, we generated TPU-coated cotton fabric and an injection molded phone case and documented biodegradation by direct visualization and scanning electron microscopy (SEM), both of which indicated clear structural degradation of these materials and significant biofilm formation.

Abstract:China government has implemented regulations to completely ban the use of disposable plastics with about 54 million kt, resulting in tremendous controversies about non-degradable and biodegradable plastics. However, there is still a lack of direct evidence to elucidate the difference of degradation mechanisms among them, which limits the development of applications for biodegradable materials. In this work, we provide direct evidence to clarify the degradation mechanisms between bio- and non-degradable materials. Cellulose, polybutylene adipate terephthalate (PBAT), and polyethylene (PE) film were subjected to phosphate-buffered solution (PBS) degradation, ultraviolet (UV) aging, and soil degradation, and the possible degradation mechanism analysis was carried out. The results exhibited that the cellulose film possessed the shortest degradation cycles whether in PBS degradation, UV aging, or soil degradation, moreover, the cellulose film showed a much higher weight loss of 99.1% after 98 days of soil degradation compared with PBAT film (2.1%) and PE film (1.7%). Additionally, we proposed possible degradation mechanisms to pave the way for the fabrication of green and degradable packaging plastics.

Abstract:Polyurethanes (PUR) are ranked globally as the 6th most abundant synthetic polymer material. Most PUR materials are specifically designed to ensure long-term durability and high resistance to environmental factors. As the demand for diverse PUR materials is increasing annually in many industrial sectors, a large amount of PUR waste is also being generated, which requires proper disposal. In contrast to other mass-produced plastics such as PE, PP, and PET, PUR is a family of synthetic polymers, which differ considerably in their physical properties due to different building blocks (for example, polyester- or polyether-polyol) used in the synthesis. Despite its xenobiotic properties, PUR has been found to be susceptible to biodegradation by different microorganisms, albeit at very low rate under environmental and laboratory conditions. Discovery and characterization of highly efficient PUR-degrading microbes and enzymes capable of disassembling PUR polymer chains into oligo- and monomeric compounds is of fundamental importance for a circular plastic economy. In this review, the main methods used for screening PUR-degrading microbes and enzymes are summarized and compared in terms of their catalytic mechanisms. Furthermore, recycling and upcycling strategies of waste PUR polymers, including microbial conversion of PUR monomers into value added products, are presented.

Biodegradation of polyurethane by the microbial consortia enriched from landfill

Abstract:Polyurethanes (PU) are one of the most used categories of plastics and have become a significant source of environmental pollutants. Degrading the refractory PU wastes using environmentally friendly strategies is in high demand. In this study, three microbial consortia from the landfill leachate were enriched using PU powder as the sole carbon source. The consortia efficiently degraded polyester PU film and accumulated high biomass within 1 week. Scanning electron microscopy, Fourier transform infrared spectroscopy, and contact angle analyses showed significant physical and chemical changes to the PU film after incubating with the consortia for 48 h. In addition, the degradation products adipic acid and butanediol were detected by high-performance liquid chromatography in the supernatant of the consortia. Microbial composition and extracellular enzyme analyses revealed that the consortia can secrete esterase and urease, which were potentially involved in the degradation of PU. The dominant microbes in the consortia changed when continuously passaged for 50 generations of growth on the PU films. This work demonstrates the potential use of microbial consortia in the biodegradation of PU wastes.

Abstract:Polyurethane (PU) is a type of polymer, which exists in various forms in the environment. Very few studies are available concerning the structure or enzymatic mechanism of the microbial community, which can degrade PU. Degradation of PU remains a difficult problem with respect to the environmental and biological disciplines. This study mainly focused on identifying the micro-organisms able to degrade polyurethane and confirming the degradation by performing a plate assay, Sturm test and scanning electron microscopy. Optimal culture conditions for maximum PU degradation were also analyzed through classical methods. A soil burial test was conducted by placing polyurethane films in the soil for one month, and the microbe growing on the surface of polyurethane films—with a maximum degradation of 55%—was isolated and identified as Aspergillus versicolor (ARF5). The culture medium was also optimized with different physical and chemical parameters for maximum PU degradation. The presence of CO2 as a by-product of PU biodegradation was confirmed through the Sturm test.

Abstract:Lack of degradability and the closing of landfill sites as well as growing water and land pollution problems have led to concern about plastics. Increasingly, raw materials such as crude oil are in short supply for the synthesis of plastics, and the recycling of waste plastics is becoming more important. As the importance of recycling increases, so do studies on elucidation of the biodegradability of polyurethanes. Polyurethanes are an important and versatile class of man-made polymers used in a wide variety of products in the medical, automotive and industrial fields. Polyurethane is a general term used for a class of polymers derived from the condensation of polyisocyanates and polyalcohols. Despite its xenobiotic origins, polyurethane has been found to be susceptible to biodegradation by naturally occurring microorganisms. Microbial degradation of polyurethanes is dependent on the many properties of the polymer such as molecular orientation, crystallinity, cross-linking and chemical groups present in the molecular chains which determine the accessibility to degrading-enzyme systems. Esterase activity (both membrane-bound and extracellular) has been noted in microbes which allow them to utilize polyurethane. Microbial degradation of polyester polyurethane is hypothosized to be mainly due to the hydrolysis of ester bonds by these esterase enzymes.

Abstract:More single-use plastics are accumulating in the environment, and likewise biodegradable plastics (BPs), which are being vigorously promoted, cannot escape the fate. Currently, studies on the actual degradation of BPs in open-air and freshwaters are underrepresented despite they are potentially headmost leakage and contamination sites for disposable BPs. Herein, we compared the degradation behavior of six BP materials and non-degradable polypropylene (PP) plastics over a 1-year in situ suspension in the high-humidity air, a eutrophic river, and an oligotrophic lake. Moreover, a 3-months laboratory incubation was performed to detect the release of dissolved organic carbon (DOC) from BPs. In both air and freshwaters, poly(p-dioxanone) (PPDO) degraded significantly while PP and polylactic acid (PLA) showed no signs of degradation. The average degradation rates of three poly(butylene adipate- co -terephthalate) (PBAT)-based films varied: 100% in river, 55% in lake, and 10% in air. In addition to PLA, surface chemical groups, hydrophilicity, and thermal stability of BPs changed, and microplastics were found on their surfaces. Correspondingly, BPs with faster degradation rates released relatively higher amounts of DOC. Environmental microbial and chemical characteristics may contribute to differences in BP degradation besides polymer specificity. Altogether, our results indicate the need for appropriate monitoring of BPs. Environmental Implication Plastic debris is widespread in freshwater ecosystems globally, and biodegradable plastics (BPs) also cannot escape this fate. Whereas, a lack of real environmental degradation data makes assessment of BPs' safety challenging. Here we present a long-term and discrepant degradation of six disposable BPs in the high-humidity air, a eutrophic river, and an oligotrophic lake. Correspondingly, it is found that BPs with faster degradation rates release relatively higher amounts of dissolved organic carbon. Our findings highlight the need for carefully selecting biodegradable materials for applications and the importance of monitoring their degradation in different freshwater ecosystems.

Abstract:The production and use of plastic blends have been gradually increasing owing to their versatility and low cost. However, the photodegradation of plastic blends in seawater and the potential risk to the marine environment are still not well understood. In this study, plastic blends including polypropylene/thermoplastic starch blends(PP/TPS) and polylactic acid/poly(butylene adipate-co-terephthalate)/thermoplastic starch blends(PLA/PBAT/TPS) were investigated. The corresponding neat polymers, namely polypropylene(PP) and polylactic acid(PLA), were set as control groups. We investigated the formation of MPs and the changes in the physicochemical properties of plastic blends after photodegradation in seawater. The size distribution of MPs indicated that PP/TPS and PLA/PBAT/TPS were more likely to produce small-sized particles after photodegradation than PP and PLA owing to their poorer mechanical properties and lower resistance to UV irradiation. Noticeable surface morphology alterations, including cracks and wrinkles, were observed for plastic blends following photodegradation, whereas PP and PLA were relatively resistant. After photodegradation, the ATR-FTIR spectrum of PP/TPS and PLA/PBAT/TPS showed a significant decrease in the characteristic bands of thermoplastic starch(TPS), indicating the degradation of their starch fractions. The C 1s spectra demonstrated that aged plastic blends contained fewer -OH groups than the pristine MPs did, further confirming the photodegradation of TPS. These results indicate that PP/TPS and PLA/PBAT/TPS had a higher degree of photodegradation than PP and PLA and thereby generated more small-sized MPs. In summary, plastic blends may pose a higher risk to the marine environment than neat polymers, and caution should be taken in the production and use of plastic blends.

Abstract:This report shows by light microscopy the appearance of secondary neurulation separated from primary neurulation and its developmental fate in the spinal cord of mice exposed to retinoic acid in utero. The embryos and fetuses were derived from pregnant mice (ICR strain) given 60, 40, or 0 mg/kg of retinoic acid in olive oil on day 8 of gestation orally and killed 1, 2, or 10 days later. Separation of the primary neural fold from the secondary neural tube was seen in 9- and 10-day-old embryos: the caudal part of the neuroepithelium of the primary neural fold was disarranged with non-closed posterior neuropore, and underneath it the secondary neural tissue extended caudally with abnormal notochord. At term, fetuses showed spina bifida, including myeloschisis, myelocele, and diplomyelia (diastematomyelia) with abnormal distribution of ganglionic cells. These cord lesions were located between the third lumbar and second coccygeal levels. The former two cord anomalies were associated with diplomyelia and split the dorsal and ventral portions of the spinal cord with an overlapping zone between the third lumbar and third sacral levels. These findings suggest that the separation from primary neurulation is due to the lesions in both primary neural folds and notochord induced by retinoic acid and that the spinal cord caudal to the third lumbar level originates from both neuroectoderm and mesenchyme-like cells while that caudal to the third sacral level originates from mesenchyme-like cells only.

Abstract:Combined pollution consisted of various types of microplastics (MPs) was extensively detected in the environment; however, little is known about their interaction on degradation behavior during exposure in sunlight. This study investigated the effects of polystyrene (PS) MPs and mechanisms on photodegradation of pure and commercial polypropylene (PP) MPs co-existed in aquatic environment. Results showed that PS MPs significantly accelerated photodegradation of co-existed PP, including faster oxidation and fragmentation. Photodegradation route of PP MPs such as the reaction priority of partial chemical bindings was even altered with the presence of PS MPs, highlighting the important role of PS in photodegradation process of PP. Analysis of leachate and free radical indicated that the critical effects were derived from photosensitization of PS polymer itself and its released dissolved organic matter (PS-DOM); here, more important role of PS itself in initial period and that of PS-DOM in later period. Among generated ROS, OH· was the key species for accelerating photodegradation of PP by PS itself and its released DOM, which were generated from the reaction of polymer radical with dissolved oxygen. The findings firstly reveal the important role of PS in photodegradation of co-existed MPs and suggested the shorter duration of (micro)plastics in combined system than that in the single, which provide useful information to assess environmental behavior and fate of MPs more holistically.

Abstract:The extensive utilization and inadequate handling of plastics have resulted in severe environmental ramifications. In particular, plastics composed solely of a carbon-carbon (C-C) backbone exhibit limited degradation due to the absence of hydrolyzable functional groups. Plastics with enduring longevity in the natural environment are susceptible to environmental factors and their intrinsic properties, subsequently undergoing a series of aging processes that culminate in biodegradation. This article focuses on polystyrene (PS), which constitutes 20% of total plastic waste, as a case study. Initially, the application of PS in life and the impacts it poses are introduced. Following that, the key factors influencing the aging of PS are discussed, primarily encompassing its properties (e.g., surface characteristics, additives) and environmental factors (e.g., water matrices, biofilms). Lastly, an overview of microbial degradation of PS is provided, including potential microorganisms involved in PS degradation (bacteria, fungi, algae, and insects), four processes of microbial degradation (colonization, bio-fragmentation, assimilation, and mineralization), and potential mechanisms of microbial degradation. This study provides a comprehensive understanding of the multifaceted influences affecting the aging and biodegradation mechanisms of PS, thereby contributing valuable insights for the future management of plastic pollution.
Photodegradation of biobased polymer blends in seawater: A major source of microplastics in the marine environment

Abstract:Introduction: Biobased polymer blends have been recommended as an eco-friendly solution to abate plastic pollution in the environment. However, the formation of microplastics (MPs) by photodegradation of biobased polymer blends in the marine environment is still not well understood. In this study, we investigated the formation of MPs and the changes in the physicochemical properties of three types of biobased polymer blends after photodegradation in seawater. Methods: The investigated materials included non-biodegradable polyethylene/ thermoplastic starch blends (PE/TPS) and polypropylene/thermoplastic starch blends (PP/TPS), as well as biodegradable polylactic acid/poly (butylene adipate-co-terephthalate)/thermoplastic starch blends (PLA/PBAT/TPS). The control groups were the corresponding neat polymers, including polyethylene (PE), polypropylene (PP), and polylactic acid (PLA). Results: The size distribution of the pristine and aged MPs indicated that the polymer blends were more likely to produce small-sized particles after photodegradation due to their poorer mechanical properties and lower resistance to UV irradiation than the neat polymers. Noticeable surface morphology alterations, including cracks, holes, and pits, were observed for polymer blends after photodegradation, while neat polymers were relatively resistant. After photodegradation, the attenuated total reflection Fourier transformed infrared spectroscopy (ATR-FTIR) spectrum of the polymer blends showed a significant decrease in the characteristic bands of thermoplastic starch (TPS), indicating depletion of their starch fractions. The C1s spectra of the polymer blends demonstrated that the aged MPs contained fewer -OH groups than the pristine MPs, further confirming the photodegradation of TPS. The molecular weight distribution curve of the polymer blends shifted significantly towards low molecular weight, suggesting the occurrence of chain scission during photodegradation. These results indicate that the polymer blends have a higher degree of photodegradation than neat polymers, and thereby generate more small-sized MPs than neat polymers. Photodegradation caused changes in the contact angle and surface charge of MPs derived from biobased polymer blends, which may affect the vector effects of MPs on any coexisting pollutants. Discussion: In summary, polymer blends may pose a higher risk to the marine environment than neat polymers, and caution should be taken in promoting biobased polymer blends.
