Abstract:Lately, a great deal of attention is being paid to capsule coating, since the coat protects active pharmaceutical ingredients (APIs) from damage, as is in the case of tablet and pellet. However, moisture and heat sensitivity of gelatin shells make it challenging to coat capsules using the conventional aqueous coating techniques. In an effort to overcome this challenge, the present study aims to coat capsules using two different coating techniques: electrostatic dry powder coating (EDPC) and dip coating (DC). Both capsule coatings and free films were prepared by these two coating techniques, and the effects of coating formulations and processing conditions on the film quality were investigated. The corresponding drug in vitro release and mechanisms were characterized and compared. The results of dissolution tests demonstrated that the drug release behavior of both EDPC and DC coated capsules could be optimized to a sustained release of 24 h, following the Fick's diffusion law. The results of this study suggest that EDPC method is better than DC method for coating capsules, with respect to the higher production efficiency and better stability, indicating that this dry coating technology has promised in gelatin capsule coating applications.
Multiporous Microstructure for Enhancing the Water Absorption and Swelling Rate in Poly(sodium Acrylic Acid) Superabsorbent Hydrogels Based on a Novel Physical and Chemical Composite Foaming System

Abstract:ABSTRACTIn this article, we report a novel physical and chemical composite foaming system, which was used to successfully prepare high‐performance and low‐cost composite superabsorbent [poly(sodium acrylic acid) (PAA–Na)] hydrogels based on acrylic acid by free‐radical polymerization in a water bath under a nitrogen atmosphere without the use of any organic solvents. The prepared hydrogels showed superabsorbent properties, high water‐absorption abilities and swelling rates, a lighter packing density, and a multiporous microstructure. Fourier transform infrared spectroscopy and scanning electron microscopy revealed that the sodium dodecyl sulfate surfactant, sodium bicarbonate chemical foaming agent, and 1,1,2‐trifluorotrichloroethane physical foaming agent were evenly distributed and grafted onto the PAA–Na matrix. Water‐absorption, swelling rate, and packing density testing confirmed that the superabsorbent had a high water‐absorption ability and swelling rate and a lighter packing density. Furthermore, we investigated the effects of different foaming agents, including chemical and physical foaming agents, on the swelling and water‐retention capabilities of the superabsorbent hydrogels (SAHs).The results show that the combination of these foaming agents significantly improved the water‐absorbing capacity. With the help of these foaming agents, we obtained PAA–Na hydrogels without any organic solvents for posttreatment or special porogens; this is an environmentally beneficial way to prepare SAHs for hygiene and biomedical products. © 2016 Wiley Periodicals, Inc. Abstract:The experiment is successfully obtained a new processing procedure of effervescent tablet by using technology combined non-water granulating process with effervescent technology. The main factors that influence tablet hardness and disintegration time are wetting agent ethanol, agglutinating agent polyvinylpyrrolidone(PVP) and tablet pressure. Reducing auxiliary ethanol, increasing auxiliary PVP and tablet pressure will result higher tablet hardness and longer disintegration time. Proved by F test and t test, the relationship between tablet hardness and disintegration time shows good linearity, and the regression equation is y=10.482x+135.68. Taking all factors into consideration, the formula for optimal tablet hardness and disintegration time is auxiliary ethanol 120mg, auxiliary PVP180mg and tablet pressure 4.9×103kgf.

Abstract:This study aimed to prepare effervescent tablets of traditional Chinese medicine Xianganfang with fresh juice using a semi-solid 3D printer with three cartridge holders to seperate acid and alkali source by drug paste through model design to avoid sticking impact and premature effervescence during the tableting in the conventional preparation process. The powder of Xianganfang including fresh juice of Phyllanthus emblica and licorice extract was obtained by vacuum freeze-drying with 50% mannitol as cryoprotectant. Then, the formulation of 3D-printed effervescent tablets was investigated. Further 5% HPMC hydroalcoholic gel was mixed with sodium bicarbonate and freeze-dried Xianganfang powder to prepare alkali source and drug paste respectively while 30% PVP ethanol solution was mixed with tartaric acid to prepare acid source paste; these three pastes had good printability. The pastes of drug, acid, and alkali were loaded into three syringe cartridges separately and numbered as "3," "5," and "7," according to cartridge holders of the 3D printer, and printed in the order of "537,353,735" for separating acid and alkali by drug to avoid premature effervescence. And the basic printing parameters were optimized. The tablets were evaluated by the appearance, tablet weight variation, hardness, disintegration time, friability, pH, and stability. The physicochemical properties all conformed to the Chinese Pharmacopoeia 2020 edition. The content of the active ingredient gallic acid was 0.769 ± 0.019 mg/g. This study provided a new method to prepare effervescent tablets of traditional Chinese medicine with fresh juice using 3D printing technology.

Abstract:The objective of this study was to develop effervescent cleansing tablets that can be dissolved and turned into liquid soap, which can be used for bathing or soaking the body. The asiatic-acid-loaded solid lipid microparticles (AASLMs) were prepared via the hot emulsification method followed by cold re-solidification and then freeze-dried to obtained dry powder. The physicochemical properties such as morphology and % entrapment efficiency (%EE) were evaluated. The results revealed that AASLMs have an irregular shape, and the %EE for the resulting AASLMs was 92.04 ± 3.43%. The tablets were manufactured via the direct compression technique. The compatibility test was conducted to ensure that the excipients are compatible with the active ingredient. The angle of repose, Carr’s index, and Hausner’s ratio were studied to evaluate the flowability of the powder blend before compression. The weight of each tablet was set to 1000 mg, and physicochemical characteristics, in vitro dissolution, ex vivo cleansing efficacy, and stability were evaluated. The results showed that the active ingredient was compatible with other excipients, as the results obtained from FTIR spectra indicated the absence of potential chemical interaction between the active ingredient and excipients used in this study. Additionally, all formulations had good flow properties. The effervescence times of selected formulations, F2 and F3, were <5 min, with favorable pH and hardness values. The friability values of all formulations exceeded 1% because the excipients used in effervescent tablets are very fragile. The release of asiatic acid (AA) from the tablets was dependent on the concentration of SLS. In an ex vivo test, it was discovered that the developed products F2 and F3 showed much more effective cleansing efficacy than water. Nevertheless, brown spots appeared in the tablets and the AA content was significantly decreased in both tested formulations after 3 months’ storage at 40 ± 2 °C/75% RH ± 5% RH. The stability study revealed that the developed products were not stable at high temperature and humidity. Therefore, it is recommended that the developed effervescent tablets are not stored at a high temperature.

Abstract:Using microcrystalline cellulose (MCC) with plastic behaviour and calcium phosphate anhydrous (CaHPO4) with brittle behaviour under compaction is very popular in the pharmaceutical industry for achieving desirable structural–mechanical properties of tablet formulations. Thus, mixtures of specific grades of MCC and CaHPO4 were tested in volume proportions of 100-0, 75-25, 50-50, 25-75, and 0-100 at a constant weight-by-weight concentration of sodium stearyl fumarate lubricant, utilizing a state-of-the-art benchtop compaction simulator (STYL'One Nano). Tablet formulations were prepared at 100, 150, 250, 350, 450, and 500 MPa, and characterized by tabletability profile, ejection force profile, proportion–tensile strength relationship, proportion–porosity relationship, pressure–displacement, and elastic recovery profiles, as well as by in-/out-of-die Heckel plots and yield pressures. Interestingly, the 25-75 formulation demonstrated a two-stage out-of-die Heckel plot and was additionally investigated with X-ray micro-computed tomography (μCT). By post-processing the μCT data, the degree of brittle CaHPO4 particles falling apart, along with the increasing compression pressure, was quantified by means of the surface area to volume (S/V) ratio. For the 25-75 formulation, the first stage (up to 150 MPa) and second stage (above the 150 MPa) of the out-of-die Heckel plot could be attributed to predominant MCC and CaHPO4 deformation, respectively.

Abstract:The utilization of acid as a synthesis assistant provides an effective means to regulate the structure of hydrogels, thereby simplifying the design and preparation process of multifunctional hydrogels. However, there remains a dearth of discourse concerning the utilization of this convenient acid-mediated strategy, which possesses the potential to directly govern molecular interactions within gel networks for rational structure and property design. Herein, we describe the preparation of flexible dual-network conductive hydrogels using polyacrylamide (PAM) and sodium alginate (SA) as substrates, driven by the strategy of acid-mediated (HCI, H2SO4, and H2C2O4) in detail for the first time. Especially, the structure-activity relationship of hydrogels was elucidated through a comparative analysis of molecular dynamics (MD) simulations and empirical properties, thereby enhancing the understanding of this field. Furthermore, extensive investigations have been conducted to explore the distinct impacts of acid ions and concentrations. The acid-mediated method exhibits superior versatility and operability compared to the filler modification method, thereby enabling a more convenient acquisition of conductive and robust hydrogels suitable for flexible capacitors and wearable sensors. Consequently, this study presents a straightforward, efficient, and cost-effective universal strategy for targeted functional hydrogel design.

Abstract:Effervescent tablets are becoming increasingly popular due to their ease of administration and rapid onset of action. They typically contain acidic materials and carbonates or bicarbonates that react quickly in the presence of water, releasing carbon dioxide and improving API solubility and flavour masking. However, effervescent tablets can be bulky, and the reaction rate is difficult to control due to water's catalytic effect. This article discusses the advantages and disadvantages of effervescent tablets, common effervescent reactions, active ingredients that can be formulated, and the preparation and manufacturing process. It also evaluates effervescent granules and tablets and explores the latest advancements in effervescent technology. Overall, effervescent tablets offer a promising option for drug delivery, and ongoing research will undoubtedly yield even more advanced formulations in the future. Abstract:To solve real‐life problems—such as those related to technology, health, security, or climate change—and make suitable decisions, risk is nearly always a main issue. Different types of sciences are often supporting the work, for example, statistics, natural sciences, and social sciences. Risk analysis approaches and methods are also commonly used, but risk analysis is not broadly accepted as a science in itself. A key problem is the lack of explanatory power and large uncertainties when assessing risk. This article presents an emerging new risk analysis science based on novel ideas and theories on risk analysis developed in recent years by the risk analysis community. It builds on a fundamental change in thinking, from the search for accurate predictions and risk estimates, to knowledge generation related to concepts, theories, frameworks, approaches, principles, methods, and models to understand, assess, characterize, communicate, and (in a broad sense) manage risk. Examples are used to illustrate the importance of this distinct/separate risk analysis science for solving risk problems, supporting science in general and other disciplines in particular.

Abstract:A controlled release effervescent osmotic pump tablet (EOPT) of Traditional Chinese Medicine Compound Recipe (TCMCR), named Fuzilizhong prescription which includes acidic drugs consisted of many known and unknown effective components and has been used for several thousands years, was successfully prepared with sodium chloride, sodium hydrogen carbonate and hydroxypropylmethylcellulose(HPMC) as osmotic agents. Since the osmotic pressure in EOPT with sodium chloride and sodium hydrogen carbonate increased greatly, which was induced mostly by gas carbon dioxide generating from the reaction of sodium hydrogen carbonate and the acidic drugs in TCMCR after the fluid being imbibed into the compartment through the semipermeable membrane and the in vitro accumulative dissolution percent from prescription 3 was up to 96.6% at 14 hour, the problem that water insoluble drugs can not to be elementary osmotic pump tablet for its low dissolution rate was solved in the paper. On the basis of prescription 3, the drug in effervescent osmotic pump tablet was released controllably after HPMC was selected as retarder and has a good in-vitro-in-vivo correlation(IVIVC, r=0.9550). Threrfore, it could be concluded that the formulation of TCMCR is appropriate to being made into EOPT, which improves acidic drugs composed of soluble and poorly soluble components release more greatly and controllably. From the point of this, water insoluble drugs can be designed to elementary osmotic pump tablet for more complete dissolution release.
The impact of diluents on the compaction, dissolution, and physical stability of amorphous solid dispersion tablets

Abstract:Amorphous solid dispersion (ASD) is an effective approach for enhancing the solubility, dissolution, and bioavailability of poorly water-soluble drugs. However, these metastable forms can transform into more thermodynamically stable, less soluble crystalline forms. Despite this challenge, research on processing ASDs into solid dosage forms, such as tablets, is lacking. This research paper aims to fill this gap by investigating the impact of common diluents on the tableting behavior, dissolution, and physical stability of ASDs composed of itraconazole and hypromellose acetate succinate. Four widely used diluents found in commercially available ASD tablets were selected for the study: microcrystalline cellulose (MCC), anhydrous lactose, starch, and mannitol. The performance of ASD tablets varied significantly depending on the diluent used. Notably, tablets prepared with MCC exhibited higher mechanical strength than those formulated using other diluents. ASD tablets containing mannitol and lactose revealed a faster release rate than those composed of MCC or starch. Furthermore, the study highlighted that the physical stability of ASDs within a tablet is not solely dependent on the amount of sorbed water; crystalline diluents like lactose and mannitol were found to facilitate ASD recrystallization within a tablet. In summary, the study underscores the importance of excipient selection, considering factors such as mechanical strength, dissolution rate, and physical stability of ASD tablets. These findings offer valuable insights into the selection of excipients for downstream ASD tablet development, leading to improved manufacturability, physical stability, and the overall quality of ASD drug products.

Abstract:Tablet coating with aqueous solvents represents a well-established environmentally sustainable alternative to coatings based on organic solvents. However, this practice requires a careful evaluation of potential quality issues, such as increased porosity and crack formation stemming from possible tablet components interaction with water. In this work, a preliminary study on the compression of quaternary mixtures of one model drug and three common excipients was performed to investigate the behaviour of the mixtures and the relevant components during compression. Obtained tablets were coated with a hypromellose aqueous solution followed by an enteric coating with a poly(methyl acrylate-co-methyl methacrylate-co-methacrylic acid) dispersion. At each step, tablet porosity was measured by X-Ray microcomputed tomography (XμCT). XμCT evidenced significant changes in the structure of some tablets upon coating. The mechanical characteristics of the mixture components play a crucial role in the observed porosity increase (up to 12%) after the coating. Materials with remarkable plastic densification upon compression afforded higher structural changes with swelling and cracking formation. XμCT has proven to be an essential technique for identifying morphological changes that could compromise the quality of the tablets.

Abstract:The production of effervescent tablets by using effervescent agents in pharmaceutical products has increased due to the convenient way of swallowing the tablets. The aim of this study was to formulate polyherbal extract of Phyllanthus niruri, Strobilanthes crispus and Orthosiphon aristatus in effervescent tablets using different agents as well as to evaluate the phytochemical contents and antioxidant properties of those formulated tablets. Eight different types of formulated tablets were prepared with different effervescent agents. Total phenolic content (TPC), total flavonoid content (TFC), 2,2-diphenyl-1- picrylhydrazyl (DPPH) radical scavenging activity and ferric-reducing antioxidant power (FRAP) assays were used to evaluate the antioxidant properties of the formulated tablets. The effervescent tablet formulated with 20% acidic agent especially with citric acid (T1) displayed significant properties (p < 0.05) on phytochemical contents (TPC: 36.76 ± 0.50 mg GAE/g DW; TFC: 55.54 ± 3.60 mg RE/g DW) and antioxidant activities (DPPH: IC50: 61.60 ± 0.31 mg/mL; FRAP: 57.05 ± 0.98 mg Fe2+/g DW). In conclusion, the high content of acidic agents resulted in high phytochemical contents and antioxidant activities among the formulated polyherbal effervescent tablets. The application of effervescent agents in formulating polyherbal extract into fast-disintegrating tablets could offer opportunities in producing nutraceutical and oral drugs.
Seven sour substances enhancing characteristics and stability of whey protein isolate emulsion and its heat-induced emulsion gel under the non-acid condition

Abstract:Protein emulsion gels, as potential novel application ingredients in the food industry, are very unstable in their formation. However, the incorporation of sour substances (phosphoric acid, lactic acid, acetic acid, malic acid, glutamic acid, tartaric acid and citric acid) would potentially contribute to the stable formation of whey protein isolate (WPI) emulsion as well as its gel. Thus, in this work, physical stability of seven acid-treated WPI emulsions, and microstructures, rheological properties, water distribution of its emulsion gels were characterized and compared. Initially, the absolute zeta-potential, interfacial protein adsorption, and emulsifying characteristics of acid-induced WPI emulsions were higher in contrast to acid-untreated WPI emulsions. Moreover, acid-induced WPI emulsions were thermally induced (95 ℃, 30 min) to form its emulsion gel networks via disulfide bonds as the main force (acid-untreated WPI emulsions were unable to form gels). High-resolution microscopic observation revealed that acid-induced WPI in emulsion gel network showed the morphology of aggregates. Dynamic oscillatory rheology results indicated that acid-induced emulsion gel exhibited highly elastic behavior and its viscoelasticity was associated with the generation of protein gel networks and aggregates. In addition, PCA and heatmap results further illustrated that malic acid-induced WPI emulsion gels had the best water holding capacity and gel characteristics. Therefore, this study could provide an effective way for the foodstuffs industry to open up new texture and healthy emulsion gels as fat replaces and loading systems of bioactive substances.
Impact of incorporated drugs on material properties of amorphous solid dispersions

Abstract:Formulation development of amorphous solid dispersions (ASD) still is challenging although several poorly water-soluble drugs have been marketed using this technique. During development of novel drugs, the selection of the preparation technique and polymer matrix is commonly performed for the certain drug via screening tools. However, if general trends regarding material properties are to be investigated, this approach is not beneficial, although often utilized in literature. The main component of the ASD usually is the polymer and thus it predominantly determines the material properties of the system. Therefore, to study the impact of different drugs and their drug loads on mechanical properties and wettability, three poorly soluble model drugs with drug loads ranging from 10% to 40% were incorporated into copovidone via hot-melt extrusion. The obtained extrudates were subsequently characterized regarding mechanical properties by applying diametral compression test and nanoindentation and the results were compared to the performance during tablet compression. Incorporation of all tested drugs resulted in a similar increase in brittleness of the ASDs, whereas the Young's modulus and hardness changed differently in dependence of the incorporated drug. These observations correlated well with the performance during tablet compression and it was concluded, that the brittleness seemed to be the predominant factor influencing the compression behavior of copovidone-based ASDs. Furthermore, the degree of water absorption and wettability was assessed by applying dynamic vapor sorption experiments and contact angle measurements. Here, the incorporated drugs impacted the contact angle to different degrees and a strong correlation between the contact angle and disintegration time was observable. These results highlight the importance of thorough characterization of the ASDs as it helps to predict their performance during tablet compression and thus facilitates the optimal selection of excipients.

Abstract:Physicochemical properties and conformations of WEPG (water extracted peach gum), AEPG2.0 (alkali extracted peach gum, 2M NaOH) and HPPG8 (H2O2 extracted peach gum, 8 h), prepared from the gum of Prunus persica Batsch, were investigated. The yields of WEPG, AEPG2.0 and HPPG8 were 77.25%, 82.60% and 83.34%, respectively. All of the three peach gums were arabinogalactan-type polysaccharides with similar chemical composition. Conformational analysis revealed that WEPG, AEPG2.0 and HPPG8 were macromolecules with compacted coil structures and their molecular weights of 1.34 x 10(7) g/mol, 1.64 x 10(7) g/mol and 5.17 x 10(6) g/mol, respectively. Rheological test showed that WEPG, AEPG2.0 and HPPG8 exhibited non-Newtonian behavior, and their apparent viscosities followed the order of AEPG2.0 > WEPG > HPPG8. Results also indicated that all of the three peach gums significantly (P < 0.05) enhanced the stability of whey protein isolate oil-water emulsions, and AEPG2.0 was the optimal gum extract showing no phase separation over 5 weeks, suggesting it could be used as an effective stabilizer in food emulsions and beverage industry. Influence of citric acid concentrations on the porosity and performance of cellulose acetate-based porous membranes: A comprehensive study

Abstract:This study investigates the influence of citric acid concentration on the fabrication of porous cellulose acetate (CA) membranes using the Non-Solvent Induced Phase Separation (NIPS) method. A notable aspect is the precise control over membrane properties, particularly pore size and porosity, achieved solely through the adjustment of citric acid concentration, serving as the additive. Higher concentrations of citric acid increase pore size by rendering polymer chains more pliable, whereas lower concentrations lead to smaller, denser pores due to improved dispersion in the CA matrix and altered water interactions during phase separation. A decrease in porosity and Gurley values with reducing citric acid concentrations (from 5 × 10-2 to 1 × 10-3 M ratios) indicates less plasticization of CA chains. However, at very low concentrations (1 × 10-4 and 1 × 10-5), porosity increases, despite the presence of smaller pores, and Gurley values approach those of pure CA in terms of gas permeability. Fourier Transform Infrared (FT-IR) spectroscopy confirms the presence of citric acid and its interaction with carbonyl groups, consistent with the pore size observations from Scanning Electron Microscopy (SEM). Spectral data deconvolution reveals weakened carbonyl bonds due to the reduced presence of citric acid, correlating with the smaller pores observed in SEM. Thermal Gravimetric Analysis (TGA) demonstrates that composite membranes are more thermally stable than pure CA, attributed to the citric acid-induced crosslinking within the polymer chains. Stability increases with decreasing citric acid concentration, with some anomalies at the lowest levels. In conclusion, this study highlights the capability of adjusting citric acid concentration to tailor membrane properties, offering valuable insights for the creation of porous materials across diverse industrial applications.

Abstract:In the present study, we aimed to characterize the effect of four different alkaline stabilizers on the physicochemical properties of lansoprazole in a designed multiple coating system. These multiple-coated pellets were prepared by layer-layer film coating with a fluid bed coater. Swelling behavior, in vitro release and acid resistance studies were carried out in simulated gastric fluid and ted intestinal fluid, respectively. Scanning electron microscopy, powder X-ray diffractometry and differential scanning calorimetry were used to examine the physical state of the drug. The drug/polymer interaction was also explored by fourier transform infrared spectroscopy. After the addition of alkaline stabilizers, we find that the swelling behavior, acid resistance, drug release and moisture uptake are influenced by their solubility, pH of saturated solution, and buffering capacity. Furthermore, the gastric stability and drug release are markedly affected by humidity and temperature given the presence of magnesium carbonate and -oxide. However, the humidity and temperature have no impact on the gastric stability and release rate from the pellets with dibasic sodium phosphate. The solid state forms predominated by the stabilizers are crucial to the drug stability in formulations. Our results also suggest that dibasic sodium phosphate is the most efficient stabilizer for protecting the drug against degradation. Our investigation provides valuable insights into the stabilizer factors affecting release rate, stability and solid state form of the drug, which facilitates the rational selection of suitable alkalizers not only for stabilizing lansoprazole but also for other proton pump inhibitors.
