Abstract:Plant viruses transmitted by arthropods,as an important biotic factor,may not only directly affect the yield and quality of host plants,and development,physiological characteristics and ecological performances of their vector arthropods,but also directly or indirectly affect the non-vector herbivorous arthropods and their natural enemies in the same ecosystem,thereby causing influences to the whole agro-ecosystem. This paper reviewed the progress on the effects of plant viruses on herbivorous arthropods,including vector and non-vector,and their natural enemies,and on their ecological mechanisms to provide a reference for optimizing the management of vector and non-vector arthropod populations and sustainable control of plant viruses in agro-ecosystem.

Abstract:In order to clarify the impacts of southern rice black-streaked dwarf virus (SRBSDV) infection on rice plants, rice planthoppers and natural enemies, differences in nutrients and volatile secondary metabolites between infected and healthy rice plants were examined. Furthermore, the impacts of virus-mediated changes in plants on the population growth of non-vector brown planthopper (BPH), Nilaparvata lugens, and the selectivity and parasitic capability of planthopper egg parasitoid Anagrus nilaparvatae were studied. The results showed that rice plants had no significant changes in amino acid and soluble sugar contents after SRBSDV infection, and SRBSDV-infected plants had no significant effect on population growth of non-vector BPH. A. nilaparvatae preferred BPH eggs both in infected and healthy rice plants, and tended to parasitize eggs on infected plants, but it had no significant preference for infected plants or healthy plants. GC-MS analysis showed that tridecylic aldehyde occurred only in rice plants infected with SRBSDV, whereas octanal, undecane, methyl salicylate and hexadecane occurred only in healthy rice plants. However, in tests of behavioral responses to these five volatile substances using a Y-tube olfactometer, A. nilaparvatae did not show obvious selectivity between single volatile substances at different concentrations and liquid paraffin in the control group. The parasitic capability of A. nilaparvatae did not differ between SRBSDV-infected plants and healthy plant seedlings. The results suggested that SRBSDV-infected plants have no significant impacts on the non-vector planthopper and its egg parasitoid, A. nilaparvatae.

Abstract:BACKGROUND:Southern rice black-streaked dwarf virus (SRBSDV) is a recently discovered member of the genus Fijivirus and it is transmitted by the rice whitebacked planthopper (WBPH), Sogatella furcifera (Horváth). It was found that SRBSDV infected vectors might contribute negatively to the WBPH population, although the longer nymphal period might benefit viral acquisition, transmission and increase infection rate. The interaction between SRBSDV and its vector need to be further explored to gain better understanding of the dispersal of WBPH and the spread of virus disease, in particular the feeding and reproduction behavior of viruliferous WBPH.METHODS:Newly hatched nymphs of WBPH were fed on healthy rice plant after feeding on SRBSDV-infected rice plants for 2 h, and newly emerged adults were numbered and tested. Feeding behaviors of WBPH adults were monitored electronically within a Faraday cage using a Giga-4 DC EPG amplifier. The newly emerged adults were paired, and the fecundity and egg hatchability were investigated. WBPH was molecularly identified for SRBSDV when they dead. According to the identification results, data on viruliferous and non-viruliferous WBPH were collected and analyzed.RESULTS:Feeding behavior of viruliferous WBPH was different from those of non-viruliferous WBPH. Frequency of phloem sap ingestion of viruliferous WBPH increased significantly, however the total feeding duration did not increase markedly. When both WBPH parents were infected with SRBSDV, their fecundity and hatchability of the eggs produced were significant lower than those of normal WBPH parents. However, if only one of the parents was viruliferous, fecundity and egg hatchability were only slightly affected.CONCLUSIONS:Viruliferous WBPH fed on the phloem more frequently than non-viruliferous WBPH and can thus contribute to virus transmission. When both vector parents are viruliferous fecundity and hatchability of the eggs were significantly reduced. However when only one of the parents WBPH was viruliferous, there were no significant effects.
Aphid endosymbiont facilitates virus transmission by modulating the volatile profile of host plants

Abstract:Abstract Background Most plant viruses rely on vectors for their transmission and spread. One of the outstanding biological questions concerning the vector-pathogen-symbiont multi-trophic interactions is the potential involvement of vector symbionts in the virus transmission process. Here, we used a multi-factorial system containing a non-persistent plant virus, cucumber mosaic virus (CMV), its primary vector, green peach aphid, Myzus persicae , and the obligate endosymbiont, Buchnera aphidicola to explore this uncharted territory. Results Based on our preliminary research, we hypothesized that aphid endosymbiont B. aphidicola can facilitate CMV transmission by modulating plant volatile profiles. Gene expression analyses demonstrated that CMV infection reduced B. aphidicola abundance in M. persicae , in which lower abundance of B. aphidicola was associated with a preference shift in aphids from infected to healthy plants. Volatile profile analyses confirmed that feeding by aphids with lower B. aphidicola titers reduced the production of attractants, while increased the emission of deterrents. As a result, M. persicae changed their feeding preference from infected to healthy plants. Conclusions We conclude that CMV infection reduces the B. aphidicola abundance in M. persicae . When viruliferous aphids feed on host plants, dynamic changes in obligate symbionts lead to a shift in plant volatiles from attraction to avoidance, thereby switching insect vector’s feeding preference from infected to healthy plants.

Abstract:Plant viruses interact with their insect vectors directly and indirectly via host plants and this tripartite interaction may produce fitness benefits to both the vectors and the viruses. Our previous studies show that the Middle East-Asia Minor 1 (MEAM1) species of the whitefly Bemisia tabaci complex improved its performance on tobacco plants infected by the Tomato yellow leaf curl China virus (TYLCCNV), which it transmits, although virus infection of the whitefly per se reduced its performance. Here, we use electrical penetration graph recording to investigate the direct and indirect effects of TYLCCNV on the feeding behaviour of MEAM1. When feeding on either cotton, a non-host of TYLCCNV, or uninfected tobacco, a host of TYLCCNV, virus-infection of the whiteflies impeded their feeding. Interestingly, when viruliferous whiteflies fed on virus-infected tobacco, their feeding activities were no longer negatively affected; instead, the virus promoted whitefly behaviour related to rapid and effective sap ingestion. Our findings show differential profiles of direct and indirect modification of vector feeding behaviour by a plant virus and help to unravel the behavioural mechanisms underlying a mutualistic relationship between an insect vector and a plant virus that also has features reminiscent of an insect pathogen.

Abstract:The interactions of vector-virus-plant have important ecological and evolutionary implications. While the tripartite interactions have received some attention, little is known about whether vector infestation affects subsequent viral transmission and infection. Working with the whitefly Bemisia tabaci, begomovirus and tobacco/tomato, we demonstrate that pre-infestation of plants by the whitefly vector reduced subsequent plant susceptibility to viral transmission. Pre-infestation by the cotton bollworm, a nonvector of the virus, likewise repressed subsequent viral transmission. The two types of insects, with piercing and chewing mouthparts, respectively, activated different plant signaling pathways in the interactions. Whitefly pre-infestation activated the salicylic acid (SA) signaling pathway, leading to deposition of callose that inhibited begomovirus replication/movement. Although cotton bollworm infestation elicited the jasmonic acid (JA) defense pathway and was beneficial to virus replication, the pre-infested plants repelled whiteflies from feeding and so decreased virus transmission. Experiments using a pharmaceutical approach with plant hormones or a genetic approach using hormone transgenic or mutant plants further showed that SA played a negative but JA played a positive role in begomovirus infection. These novel findings indicate that both vector and nonvector insect feeding of a plant may have substantial negative consequences for ensuing viral transmission and infection.

Abstract:Viruses are obligate intracellular parasites with limited proteomes that heavily rely on the cell molecular machinery for their multiplication and spread. Plant viruses frequently cause symptoms through interference with host developmental programs. Despite the agricultural relevance of symptom development in virus-infected crops, the molecular mechanisms underlying these viral effects remain elusive. Here, we show that the symptoms triggered by tomato yellow leaf curl virus (TYLCV) depend on the physical interaction between the host-mimicking domain of a virus-encoded protein, C4, and a plant-specific family of RCC1-like domain-containing (RLD) proteins. C4 outcompetes endogenous interactors of RLDs, disrupting RLD function in the regulation of endomembrane trafficking and polar auxin transport, ultimately leading to the developmental alterations recognized as symptoms of the viral infection. Importantly, symptoms do not have a detectable effect on the performance of the virus in the plant host, but they serve as attractants for the viral insect vector, the whitefly Bemisia tabaci, hence promoting pathogen spread. Our work uncovers the molecular underpinnings of the viral manipulation that leads to symptom development in the TYLCV-tomato pathosystem, and suggests that symptoms have evolved as a strategy to promote viral transmission by the insect vector. Abstract:Attacks on plants by both viruses and their vectors is common in nature. Yet the dynamics of the plant-virus-vector tripartite system, in particular the effects of viral infection on plant-insect interactions, have only begun to emerge in the last decade. Viruses can modulate the interactions between insect vectors and plants via the jasmonate, salicylic acid and ethylene phytohormone pathways, resulting in changes in fitness and viral transmission capacity of their insect vectors. Virus infection of plants may also modulate other phytohormones, such as auxin, gibberellins, cytokinins, brassinosteroids and abscisic acid, with yet undefined consequences on plant-insect interactions. Moreover, virus infection in plants may incur changes to other plant traits, such as nutrition and secondary metabolites, that potentially contribute to virus-associated, phytohormone-mediated manipulation of plant-insect interactions. In this article, we review the research progress, discuss issues related to the complexity and variability of the viral modulation of plant interactions with insect vectors, and suggest future directions of research in this field.

Abstract:Abstract Transmission of many plant viruses relies on phloem-feeding insect vectors. However, how plant viruses directly modulate insect behavior is largely unknown. Barley yellow striate mosaic virus (BYSMV) is transmitted by the small brown planthopper (SBPH, Laodelphax striatellus ). Here, we show that BYSMV infects the central nervous system (CNS) of SBPHs, induces insect hyperactivity, and prolongs phloem feeding duration. The BYSMV accessory protein P6 interacts with the COP9 signalosome subunit 5 (LsCSN5) of SBPHs and suppresses LsCSN5-regulated de-neddylation from the Cullin 1 (CUL1), hereby inhibiting CUL1-based E3 ligases-mediated degradation of the circadian clock protein Timeless (TIM). Thus, virus infection or knockdown of LsCSN5 compromises TIM oscillation and induces high insect locomotor activity for transmission. Additionally, expression of BYSMV P6 in the CNS of transgenic Drosophila melanogaster disturbs circadian rhythm and induces high locomotor activity. Together, our results suggest the molecular mechanisms whereby BYSMV modulates locomotor activity of insect vectors for transmission.

Abstract:Many plant viruses modify the phenotype of their hosts, which may influence the behaviour of their vectors and facilitate transmission. Among them is the turnip yellows virus (TuYV), which can modify the orientation, feeding, and performance of its main aphid vector, Myzus persicae. However, the virus factors driving these mechanisms have not been elucidated. In this study, we compared the feeding behaviour and fecundity of aphids on TuYV‐infected and transgenic Arabidopsis thaliana expressing individual TuYV proteins (CP, RT and P0) to define the role of these proteins in aphid–plant interactions. Aphids on TuYV‐infected plants had shorter pathway phases and ingested phloem sap for longer times, which is expected to promote the acquisition of the phloem‐limited TuYV. No change in aphid fecundity was observed on TuYV‐infected plants. The transmission‐conducive feeding behaviour changes could be fully reproduced by phloem‐specific expression of the capsid protein (CP) in transgenic plants, whereas expression of P0 had minor and RT had no effects on aphid feeding behaviour. We then carried out a metabolomic analysis to determine plant compounds that could be involved in the modification of the aphid behaviour. A few metabolites were specific for TuYV‐infected or CP‐transgenic A. thaliana, and are good candidates for inducing behavioural changes.

Abstract:In nature, a plant can be infected by multiple viruses simultaneously. However, the effects of coinfection on plant-vector interactions are less well studied.Two begomoviruses of the family Geminiviridae,Tomato yellow leaf curl virus (TYLCV) and Tomato yellow leaf curl China virus (TYLCCNV), occur sympatrically in China. Each of them is reported to increase the performance of whitefly vector via manipulation of plant traits. In this study, we examined the effects of coinfection by the two viruses TYLCV and TYLCCNV on plant-whitefly interactions, compared to that infected by a single virus. We found that plants infected by two viruses showed aggravated symptoms but the performance and preference of whiteflies were not altered significantly compared to singly-infected plants. Coinfection suppressed the transcription of genes involved in jasmonic acid (JA) signaling pathway in plants, but showed no significant difference to single-virus infected plants. These findings suggest that although TYLCV and TYLCCNV may synergistically induce plant symptoms, they did not manipulate synergistically plant-mediated responses to the insect vector.

Abstract:A parasite-infected host may promote performance of associated insect vectors; but possible parasite effects on nonvector insects have been largely unexplored. Here, we show that Begomovirus, the largest genus of plant viruses and transmitted exclusively by whitefly, reprogram plant immunity to promote the fitness of the vector and suppress performance of nonvector insects (i.e., cotton bollworm and aphid). Infected plants accumulated begomoviral beta C1 proteins in the phloem where they were bound to the plant transcription factor WRKY20. This viral hijacking of WRKY20 spatiotemporally redeployed plant chemical immunity within the leaf and had the asymmetrical benefiting effects on the begomoviruses and its whitefly vectors while negatively affecting two nonvector competitors. This type of interaction between a parasite and two types of herbivores, i.e., vectors and nonvectors, occurs widely in various natural and agricultural ecosystems; thus, our results have broad implications for the ecological significance of parasite-vector-host tripartite interactions.

Abstract:Summary The interactions among plant viruses, insect vectors, and host plants have been well studied; however, the roles of insect viruses in this system have largely been neglected. We investigated the effects of MpnDV infection on aphid and PVY transmission using bioassays, RNA interference (RNAi), and GC‐MS methods and green peach aphid (Myzus persicae (Sulzer)), potato virus Y (PVY), and densovirus (Myzus persicae nicotianae densovirus, MpnDV) as model systems. MpnDV increased the activities of its host, promoting population dispersal and leading to significant proliferation in tobacco plants by significantly enhancing the titer of the sesquiterpene (E)‐β‐farnesene (EβF) via up‐regulation of expression levels of the MpFPPS1 gene. The proliferation and dispersal of MpnDV‐positive individuals were faster than that of MpnDV‐negative individuals in PVY‐infected tobacco plants, which promoted the transmission of PVY. These results combined showed that an insect virus may facilitate the transmission of a plant virus by enhancing the locomotor activity and population proliferation of insect vectors. These findings provide novel opportunities for controlling insect vectors and plant viruses, which can be used in the development of novel management strategies.

Changes in phytohormone levels and gene expression of potato plants in response to potato virus Y and a nonvector insect

Abstract:Potato virus Y（PVY） and its nonvector insect of potato tuber moth（Phthorimaea operculella） are important disease and pest in global potato industry. In order to investigate whether PVY infection changed the physiological responses of potato plant to P. operculella, we studied performances of P. operculella on PVY-infected and non-infected healthy plants, and carried out the phytohormone analysis and comparative transcriptomic analysis for potato leaves which were affected by PVY infection and herbiory stimuli（dealing with the oral secretions of potato tuber moth）, or co-stressed treatment. The results showed that the resistance to P. operculella was significantly increased in potato plants infected by PVY. Moreover, PVY infection also inhibited the increase of abscisic acid induced by herbivory stimuli. Compared with the healthy control plants, herbivory stimuli and PVY infection treatments produced 3 998 and 104 differentially expressed genes（DEGs）, respectively; while 9 178 DEGs were induced by co-stressed treatment, suggesting that the co-stressed treatment may greatly affected the physiological responses in potato plants. Compared with the herbivory stimuli, the co-stressed plants had 743 DEGs, and many of them belong to the transferase activity, the protein processing in endoplasmic reticulum, and the phenylpropanoid biosynthesis pathways. And the expression levels of many genes in abscisic acid synthesis pathway were also down-regulated, which was consisted with the change of this phytohormone. In summary, this study systematically analyzed the physiological responses of potato plant confronted by the single-and co-stressed treatments of PVY infection and herbivory stimuli, which may provide scientific basis for exploring plant-virus-nonvector insect interactions.
Proteomic and Transcriptomic Analysis for Identification of Endosymbiotic Bacteria Associated with BYDV Transmission Efficiency by Sitobion miscanthi

Abstract:Sitobion miscanthi, an important viral vector of barley yellow dwarf virus (BYDV), is also symbiotically associated with endosymbionts, but little is known about the interactions between endosymbionts, aphid and BYDV. Therefore, two aphids' geographic populations, differing in their BYDV transmission efficiency, after characterizing their endosymbionts, were treated with antibiotics to investigate how changes in the composition of their endosymbiont population affected BYDV transmission efficiency. After antibiotic treatment, Rickettsia was eliminated from two geographic populations. BYDV transmission efficiency by STY geographic population dropped significantly, by −44.2% with ampicillin and −25.01% with rifampicin, but HDZ geographic population decreased by only 14.19% with ampicillin and 23.88% with rifampicin. Transcriptomic analysis showed that the number of DEGs related to the immune system, carbohydrate metabolism and lipid metabolism did increase in the STY rifampicin treatment, while replication and repair, glycan biosynthesis and metabolism increased in the STY ampicillin treatment. Proteomic analysis showed that the abundance of symbionin symL, nascent polypeptide−associated complex subunit alpha and proteasome differed significantly between the two geographic populations. We found that the endosymbionts can mediate vector viral transmission. They should therefore be included in investigations into aphid–virus interactions and plant disease epidemiology. Our findings should also help with the development of strategies to prevent virus transmission.

Abstract:Aboveground and belowground herbivory can alter host quality and trophic interactions. However, little research has explored the impacts of belowground herbivory on plant virus epidemiology. To understand this interaction in sweetpotato, Ipomoea batatas L. (Lam), we studied the impact of herbivory by sweetpotato weevil, Cylas formicarius elegantulus (Summers), to storage roots on vector herbivore, Myzus persicae (Sulzer), the green peach aphid, and Aphis gossypii Glover, the cotton aphid, feeding behavior (M. persicae and A. gossypii) and population dynamics (M. persicae only as A. gossypii does not colonize sweetpotato). In addition, a nonvector herbivore, Chrysodeixis includens (Walker), the soybean looper, weight gain was included for comparison. Infestations of sweetpotato weevil in storage roots reduced C. includens weight gain and M. persicae intrinsic rate of increase (rm), net reproductive rate (Ro), and finite rate of increase (λ) on aboveground portions of the plant. In probing behaviors related to nonpersistent virus spread (30 min trials), M. persicae probed less often, took longer to reach the first pd (potential drop), had shorter II-3 (intracellular subphase sap ingestion) durations, and had fewer archlets on plants infested with C. formicarius elegantulus. Total phloem duration did not differ between treatments, indicating that the lowered performance of M. persicae on infested plants was not due to discrepancies in probing duration. Results were less clear for A. gossypii. Thus, in sweetpotato, belowground herbivory can alter plant virus vector abundance and behavior. Understanding the mechanisms underlying these changes in behavior could improve plant virus vector integrated pest management.
Edaravone diminishes free radicals from circulating neutrophils in patients with ischemic brain attack.

Abstract:OBJECTIVE Treatment with a free radical scavenger could be a new option for ischemic brain attack, however, little is known about the alteration of oxidative stress markers induced by edaravone, a novel free radical scavenger, in human ischemic brain attack. METHODS We investigated the effects of edaravone on the oxidative stress markers in patients with ischemic brain attack. Twenty-one patients with ischemic brain attack and 19 controls were enrolled in this study. Blood samples were obtained just before and soon after the first administration of edaravone (30 mg) or ozagrel (40 mg). Intracellular reactive oxygen species of neutrophils were measured using 6-carboxy-2', 7'-dichlorodihydrofluorescin diacetate and a fluorescence-activated cell sorter. Superoxide from neutrophils, induced by phorbol myristate acetate (PMA), was determined by luminol-amplified chemiluminescence assay. RESULTS Treatment with 30 mg of edaravone significantly decreased the intracellular reactive oxygen species (ROS) of neutrophils (Wilcoxon test, p=0.0001) and PMA-induced superoxide produced by neutrophils (Wilcoxon test, p=0.001). Ozagrel did not alter the intracellular ROS or superoxide production of neutrophils. CONCLUSION Reduction of intracellular ROS and suppression of superoxide production in neutrophils provide a potential explanation for the clinical efficacy of edaravone in patients with ischemic brain attack.

Abstract:The occurrence and increased dispersion of plant viruses and insect vectors are serious global threat to the production of agricultural crops. Facing novel pathogenic plant viruses, the ability to accurately identify plant virus species, and understand the interaction between plant viruses, host plants and their insect vectors would provide an important basis for formulating effective plant virus control measures. In this study, we explored the transmission mechanism, pathogenic symptoms, host range and the interactions between virus and aphid vectors of a novel polerovirus from Nicotiana tabacum, named Tobacco yellow virus (TYV). The results indicate that TYV can be transmitted by Myzus persicae in a persistent manner, and cause yellowing and shrinking of tobacco leaves. TYV can successfully infect a total of 9 plant species belonging to 3 families. The effect of TYV-infected tobacco plants on M. persicae behavior and life characteristics was found to be stage-dependent. TYV can directly and indirectly manipulate the performance and life history traits of M. persicae vectors to promote their own transmission. These results provide a certain theoretical basis for the possibility of control strategies of the virus, and the in-depth exploration of the interaction among plant virus, vector aphid and host plants.

Abstract:Plant viruses threaten food security and are often transmitted by insect vectors. Non-persistently transmitted (NPT) plant viruses are transmitted almost exclusively by aphids. Because virions attach to the aphid's stylet (mouthparts) and are acquired and inoculated via brief epidermal probes, the aphid–virus interaction is highly transient, with a very short aphid virus retention time. Many NPT viruses manipulate their host plant's phenotype to change aphid behaviour to optimise virus transmission. Epidemiological models of this have overlooked a key feature of aphid NPT virus retention: probing or feeding on a plant causes aphids to lose the virus. Furthermore, experimental studies suggest aphids could possibly inoculate multiple healthy plants within one infective period if they do not feed. Consequences of this for virus manipulation of host plant phenotype have not been explored. Our new compartmental epidemiological model includes both behaviour-based aphid dispersal and infectivity loss rates, and the ability of infective aphids to probe multiple plants before virus loss. We use our model to explore how NPT virus-induced host phenotypes affect epidemic outcomes, comparing these results to representative previous models. We find that previous models behave fundamentally differently and underestimate the benefit of an 'attract-and-deter' phenotype, where the virus induces increased aphid attraction to infected plants but deters them from prolonged feeding. Our results also highlight the importance of characterising NPT virus retention upon the aphid during probing. Allowing for multiple infective probes increases disease incidence and the effectiveness of virus manipulation, with implications for epidemic prediction and control. Plant viruses can cause devastating disease epidemics. Non-persistently transmitted viruses are almost always vectored (transmitted between plants) by aphids. Experiments show virus infection can affect whether aphids are attracted to plants (by altering how infected plants 'smell'), as well as whether aphids settle for an extended feed after a brief initial probe (by altering how infected plants 'taste'). Since virus transmission requires an individual aphid to briefly probe an infected plant followed by one or more healthy plant(s), this strongly affects disease transmission. However, most studies exploring virus epidemics do not account for how aphid feeding behaviour affects how long an aphid holds the virus for, or that an aphid could infect multiple healthy plants before losing the virus. We use mathematical modelling to dissect how these aspects of aphid feeding behaviour affect virus transmission, particularly when viruses manipulate the 'smell' and 'taste' of plants. We show how previous studies, by omitting crucial aspects of aphid feeding behaviour, underestimate how viruses can promote their own transmission. We also highlight that there are very few experimental studies exploring the number of plants an aphid with the virus can consecutively infect, which is a key parameter affecting the severity of epidemics.
