Abstract:d -allulose is a rare low-calorie sugar that has many fundamental biological functions. d -allulose 3-epimerase from Agrobacterium tumefaciens (AT-DAEase) catalyzes the conversion of d -fructose to d -allulose. The enzyme has attracted considerable attention because of its mild catalytic properties. However, the bioconversion efficiency and reusability of AT-DAEase limit its industrial application. Magnetic metal–organic frameworks (MOFs) have uniform pore sizes and large surface areas and can facilitate mass transport and enhance the capacity for enzyme immobilization. Here, we successfully encapsulated cobalt-type AT-DAEase into the cobalt-based magnetic MOF ZIF-67@Fe 3 O 4 using a self-assembly strategy. We confirmed the immobilization of enzyme AT-DAEase and characterized the enzymatic properties of the MOF-immobilized AT-DAEase@ZIF-67@Fe 3 O 4 . The AT-DAEase@ZIF-67@Fe 3 O 4 nanoparticles had higher catalytic activity (65.1 U mg −1 ) and bioconversion ratio (38.1%) than the free AT-DAEase. The optimal conditions for maximum enzyme activity of the AT-DAEase@ZIF-67@Fe 3 O 4 nanoparticles were 55 °C and pH 8.0, which were significantly higher than those of the free AT-DAEase (50 °C and pH 7.5). The AT-DAEase@ZIF-67@Fe 3 O 4 nanoparticles displayed significantly improved thermal stability and excellent recycling performance, with 80% retention of enzyme activity at a temperature range of 45–70 °C and > 45% of its initial activity after eight cycles of enzyme use. The AT-DAEase@ZIF-67@Fe 3 O 4 nanoparticles have great potential for large-scale industrial preparation of d -allulose by immobilizing cobalt-type AT-DAEase into magnetic MOF ZIF-67@Fe 3 O 4 .

Enhancing enzyme immobilization: Fabrication of biosilica-based organic-inorganic composite carriers for efficient covalent binding of D-allulose-3-epimerase

Abstract:D-allulose, an ideal low-calorie sweetener, is primarily produced through the isomerization of d -fructose using D-allulose 3-epimerase (DAE; EC 5.1.3.30). Addressing the gap in available immobilized DAE enzymes for scalable commercial D-allulose production, three core-shell structured organic-inorganic composite silica-based carriers were designed for efficient covalent immobilization of DAE. Natural inorganic diatomite was used as the core, while 3-aminopropyltriethoxysilane (APTES), polyethyleneimine (PEI), and chitosan organic layers were coated as the shells, respectively. These tailored carriers successfully formed robust covalent bonds with DAE enzyme conjugates, cross-linked via glutaraldehyde, and demonstrated enzyme activities of 372 U/g, 1198 U/g, and 381 U/g, respectively. These immobilized enzymes exhibited an expanded pH tolerance and improved thermal stability compared to free DAE. Particularly, the modified diatomite with PEI exhibited a higher density of binding sites than the other carriers and the PEI-coated immobilized DAE enzyme retained 70.4 % of its relative enzyme activity after ten cycles of reuse. This study provides a promising method for DAE immobilization, underscoring the potential of using biosilica-based organic-inorganic composite carriers for the development of robust enzyme systems, thereby advancing the production of value-added food ingredients like D-allulose.

Abstract:Enzyme reaction has been accepted widely in numerous applications owing to the high efficiency and stereo-selectivity, as well as simple preparation by gene engineering. However, the fragility and complex purification process of the enzyme are long-standing problems which limit the large-scale application. One possible solution may be the enzyme immobilization. As one type of porous material with high loading capacity and designable functionality, Metal-Organic Frameworks (MOFs) are ideal choices for the immobilization of enzyme with a considerable interest in recent years. In this study, D-amino acid transaminase (DAT), an important enzyme for industrial synthesis of D-Ala, was covalently immobilized on the surface of a star MOFs material, UiO-66-NH2. Interestingly, we found that the nanoscale hybrid enzyme UiO-66-NH2-Gd-DAT not only maintained the high catalytic efficiency but also got rid of the interference of polluting enzymes, which meant that we could obtain efficient and stereo-selective immobilized enzyme without complex purification process. In general, our findings demonstrated that using UiO-66-NH2 might be a promising strategy to immobilize enzyme and produce effective biocatalyst with high activity and stereo-selectivity. Abstract:Metal‐organic frameworks (MOFs), with attractive properties such as high surface area, tunable porosity, designable functionality and excellent stability, have aroused tremendous interests of researchers as the matrices for enzyme immobilization. Recently, several efficient strategies including surface immobilization, post‐synthetic infiltration and in situ encapsulation have been explored. The MOFs‐immobilized enzymes, named enzymes@MOFs, show remarkably enhanced stability and recyclability, accelerating the cell‐free biocatalysis in diverse applications. This concept will impart the typical strategies for enzyme immobilization with MOFs, and their potential applications.

Abstract:Immobilization of D-amino acid dehydrogenase (DAADH) by the assembly of peptide linker was studied for the biosynthesis of D-phenylalanine. Hybrid material of zeolitic imidazolate framework-8 (ZIF-8) combined with reduced graphene oxide (RGO) was applied for the immobilization of DAADH from Ureibacillus thermosphaericus . The recovery rate of DAADH/ZIF-8/RGO was 165.6%. DAADH/ZIF-8/RGO remained 53.4% of its initial activity at 50 °C for 10 h while the free enzyme was inactivated. DAADH/ZIF-8/RGO maintained 70.5% activity in hyperalkaline solution with pH 12. Kinetic parameters indicated that DAADH/ZIF-8/RGO had greater affinity of phenylpyruvate as V max / K m of DAADH/ZIF-8/RGO was 1.27-fold than free enzyme. After seven recycles, the activity of DAADH/ZIF-8/RGO remained 64.3%. Furthermore, one-step separation and in situ immobilization of DAADH by ZIF-8/RGO/Ni was carried out with 1.5-fold activity enhancement. Combining peptide linker and metal-organic framework (MOF) immobilization, thermostability and activity of the immobilized DAADH were significantly improved.

Abstract:The strategies based on enzyme immobilization into/on metal-organic frameworks (MOFs) have been widely developed, but they still face some challenges in practical applications, such as limited recycle times, low loading efficiency. In this work, aspartate aminotransferase (AspAT), branched-chain aminotransferase (BCAT), polyvi-nyl pyrrolidone/aspartate (PA), cobalt ions and sodium 2-amino terephthalate were one-pot mixed in water at room temperature, and self-assembled to encapsulate the enzymes into cobalt-amino terephthalate framework (CAF) in 15 min. Compared with free enzymes, the biocomposite (A&B&PA@CAF) exhibited superior perfor-mances such as high encapsulation efficiency (55%), reusability (>= 12), and resistance capacity to harsh con-ditions. Meanwhile, the activity recovery (AR) value (59.4%) of the biocomposite without adding polyvinyl pyrrolidone and aspartate was almost half of that (108.2%) of A&B&PA@CAF, and this enhancement was further demonstrated to be from Asp action via 1H-NMR and UV-Vis. To investigate the general applicability of this approach, the other aminases were also de novo immobilized with all their AR values reached more than 117%, even 180% for omega-transaminase. This study provided a sustainable and green approach for clean production of the significant chiral amine compounds.
Boosting the activity of enzymes in metal-organic frameworks by a one-stone-two-bird enzymatic surface functionalization strategy

Abstract:Herein, we present a facile strategy for simultaneously solving two significant difficulties in metal-organic framework (MOF)-enzyme complexes: limited substrate accessibility and unfavorable interfacial interactions between MOFs and enzymes. We exploited the profound effect of the protein surface on the MOF structure and grafted short-chain polyacrylic acid (PAA) onto the enzyme surface to improve the microenvironment of the enzyme confined inside the MOF. PAA not only created mesopores inside the MOF via competitive coordination but also served as a protective layer to reduce the influence of interfacial interactions on enzyme conformation. Benefitting from this “one-stone-two-bird” approach, the activity of the enzyme-PAA complexes encapsulated in a zeolitic imidazolate framework (ZIF-L) was 2–23 times higher than that of the unconjugated enzyme in ZIF-L. Additionally, mesoporous enzyme-PAA@ZIF-L (∼15 nm) was able to endure extreme conditions and was reusable because of the shielding and confinement effects. This enzyme immobilization strategy was applied to several enzymes, and the obtained MOF-enzyme complexes showed great promise in the selective biodegradation of harmful dye molecules and point-of-care glucose detection.

Abstract:Reliability issues have been traditionally considered only for functional yield problems. Among them, the progressive gate-oxide soft breakdown (SBD) in the negative-channel field-effect transistors is foreseen to be a big issue for the deep-deep submicron technologies, where ultrathin gate oxide and low supply (stress) voltage will be massively deployed. Device-level investigation has largely confirmed that the SBD defect will not lead to an issue for the transistor functionality. In addition, simple gate-level characterization also indicates that the SBD defect in those gates only leads to performance degradation. Up to now, the impact of the random nature with such effect is not yet well understood at a complex circuit block or component level. In this paper, we have focused our analysis in providing insights on the SBD impact on sensitive static random access memory (SRAM) components as well as the complete matrix. Results have shown a significant shift in the performance figures of the blocks, which endangers the SRAM parametric yield. The drift becomes even worse when the process variability effect is taken into account. All the results indicate urgent needs to develop effective countermeasures by both the technology and design sides.
The Formation of D-Allulose 3-Epimerase Hybrid Nanoflowers and Co-Immobilization on Resins for Improved Enzyme Activity, Stability, and Processability

Abstract:As a low-calorie sugar, D-allulose is produced from D-fructose catalyzed by D-allulose 3-epimerase (DAE). Here, to improve the catalytic activity, stability, and processability of DAE, we reported a novel method by forming organic–inorganic hybrid nanoflowers (NF-DAEs) and co-immobilizing them on resins to form composites (Re-NF-DAEs). NF-DAEs were prepared by combining DAE with metal ions (Co2+, Cu2+, Zn2+, Ca2+, Ni2+, Fe2+, and Fe3+) in PBS buffer, and were analyzed by scanning electron microscopy (SEM), Fourier transform infrared spectroscopy, and X-ray diffraction. All of the NF-DAEs showed higher catalytic activities than free DAE, and the NF-DAE with Ni2+ (NF-DAE-Ni) reached the highest relative activity of 218%. The NF-DAEs improved the thermal stability of DAE, and the longest half-life reached 228 min for NF-DAE-Co compared with 105 min for the free DAE at 55 °C. To further improve the recycling performance of the NF-DAEs in practical applications, we combined resins and NF-DAEs to form Re-NF-DAEs. Resins and NF-DAEs co-effected the performance of the composites, and ReA (LXTE-606 neutral hydrophobic epoxy-based polypropylene macroreticular resins)-based composites (ReA-NF-DAEs) exhibited outstanding relative activities, thermal stabilities, storage stabilities, and processabilities. The ReA-NF-DAEs were able to be reused to catalyze the conversion from D-fructose to D-allulose, and kept more than 60% of their activities after eight cycles.

Abstract:Enzymes, as natural catalysts with remarkable catalytic activity and high region-selectivities, hold great promise in industrial catalysis. However, applications of enzymatic transformation are hampered by the fragility of enzymes in harsh conditions. Recently, metal-organic frameworks (MOFs), due to their high stability and available structural properties, have emerged as a promising platform for enzyme immobilization. Synthetic strategies of enzyme-MOF composites mainly including surface immobilization, covalent linkage, pore entrapment and in situ synthesis. Compared with free enzymes, most immobilized enzymes exhibit enhanced resistance against solvents and high temperatures. Besides, MOFs serving as matrixes for enzyme immobilization show extraordinary superiority in many aspects compared with other supporting materials. The advantages of using MOFs to support enzymes are discussed. To obtain a high enzyme loading capacity and to reduce the diffusion resistance of reactants and products during the reaction, the mesoporous MOFs have been designed and constructed. This review also covers the applications of enzyme-MOF composites in bio-sensing and detection, bio-catalysis, and cancer therapy, which is concerned with interdisciplinary nano-chemistry, material science and medical chemistry. Finally, some perspectives on reservation or enhancement of bio-catalytic activity of enzyme-MOF composites and the future of enzyme immobilization strategies are discussed.

Abstract:As a class of widely used biocatalysts, enzymes possess advantages including high catalytic efficiency, strong specificity and mild reaction condition. However, most free enzymes have high requirements on the reaction environment and are easy to deactivate. Immobilization of enzymes on nanomaterial‐based substrates is a good way to solve this problem. Metal‐organic framework (MOFs), with ultra‐high specific surface area and adjustable porosity, can provide a large space to carry enzymes. And the tightly surrounded protective layer of MOFs can stabilize the enzyme structure to a great extent. In addition, the unique porous network structure enables selective mass transfer of substrates and facilitates catalytic processes. Therefore, these enzyme‐immobilized MOFs have been widely used in various research fields, such as molecule/biomolecule sensing and imaging, disease treatment, energy and environment protection. In this review, the preparation strategies and applications of enzymes‐immobilized MOFs are illustrated and the prospects and current challenges are discussed.

Abstract:Highly efficient cell-free enzymatic systems are typically difficult to achieve in traditional immobilized enzyme systems because of the lack of optimal spatial control of enzyme localization, substrate and product diffusion, and enzyme and coenzyme accessibility. Here, we report a strategy for expanding the pore apertures (from 3.3 to 6.7 nm) of a series of Zr-based metallic-organic frameworks (MOFs) (termed NU-100x, x = 3, 4, 5, 6, 7) with interconnected hierarchical pores by maintaining precise control of torsional angles associated with the linkers. As a proof of concept, we use the expanded NU-100x MOF structures to encapsulate lactate dehydrogenase (LDH) and demonstrate the use of the captured protein in a cell-free biosynthetic catalytic system. Remarkably, LDH immobilized in the large pores of the MOF is accessible to nicotinamide adenine dinucleotide coenzymes (NAD and NADH), allowing for in situ coenzyme regeneration leading to higher activity than that of the free enzyme.
Construction of multiple enzyme metal–organic frameworks biocatalyst via DNA scaffold: A promising strategy for enzyme encapsulation

Abstract:The encapsulated enzyme system by metal–organic frameworks exhibited great potential in pharmaceuticals, enzyme fuel cells, and biocatalysis. However, the enzymatic activity and overall catalytic efficiency are severely hampered due to their enzyme leaching and causal enzyme location. Herein, for the first time, horseradish peroxidase (HRP) and glucose oxidase (GOx) used as model enzymes were cross-linked by rationally designed DNA scaffold network, and the multienzyme system has been successfully encapsulated into zeolite imidazolate framework-8 (ZIF-8). The prepared encapsulation multienzyme can efficiently suppress enzyme leaching from the MOFs skeleton, and the leaching amount was more than 10-times lower than the traditional MOFs encapsulation strategy. The well-tailored proximity and colocalization of cascade enzymes by the DNA scaffold network obviously improved the overall catalytic efficiency, enzymatic activity, and kinetic performance. The K m and k cat/K m for encapsulation multienzyme was 13.8 mM and 3.47 s−1 mM−1, which was 0.27-fold lower and 9.9-fold higher than those of free enzymes, respectively, indicating a better substrate affinity. Furthermore, the multienzyme system exhibited excellent stability and reusability compared with free enzymes and control groups, which could preserve 95% initial activity at 50 °C after 75 min incubation and more than 87% of the original activity after 14 cycles. Finally, the immobilized multienzyme exhibited enhanced performance to detect low concentration of glucose (0.4 µM) with excellent selectivity. The outcome indicated that the encapsulated multienzyme could be extended for broad practical applications in biotechnology, biocatalysis, and biomedical engineering.

Abstract:As a typical green methodology, enzymatic catalysis has been extensively employed in multitudinous chemical and biological transformation procedures. However, intrinsic fragile nature of enzymes makes them prone to denaturation or destabilization in harsh practical conditions, leading to unavoidably shortened lifespan and extremely high cost. It was proven that enzyme immobilization is an efficient strategy for enhancing their catalytic performance in continuous industrial practices. Metal-Organic Frameworks (MOFs) with extremely high specific surface area, abundant porosity, extraordinary multifunctionality, and relatively high stability, in recent years, have attracted remarkable research interests as novel supporting matrices for efficient enzyme immobilization and protection. Many reported MOFs-enzyme composites exhibit unprecedented catalytic performances than those of free enzymes, including improved enzyme efficiency, stability, selectivity, and recyclability, due to the protection of enzymes by highly ordered frameworks. To present a systematic overview of this emerging and developing field, herein, we summarize an update review about the most recent advances in MOFs immobilizing enzymes from the aspects of general synthetic approaches, critical impact factors, enhanced catalytic performances, and the practical applications. Subsequently, the emerging theories, methodologies and technologies in this thriving area are briefly introduced. Finally, barriers and future perspectives about MOFs for enzyme immobilization are also discussed.

Abstract:Metal-organic frameworks (MOFs) have attracted tremendous interests of many researchers as a new kind of porous materials. Compared to conventional matrices, MOFs possess high surface area and porosity, thousands of various kinds and structures, abundant designable organic ligands and metal nodes. In recent years, more and more attention has been paid to using MOFs as the matrices for enzyme immobilization and different strategies have been explored extensively. To present a clear outline of this new field, this review systematically summarizes the progress of enzyme immobilization on MOFs (enzyme/MOFs) from different points, including various synthetic approaches, main characteristics, mechanism, improved performance and application of immobilized enzyme/MOFs. Finally, the prospect of enzyme/ MOFs is also discussed.

Abstract:In many respects, enzymes offer advantages over traditional chemical processes due to their decreased energy requirements for function and inherent greener processing. However, significant barriers exist for the utilization of enzymes in industrial processes due to their limited stabilities and inability to operate over larger temperature and pH ranges. Immobilization of enzymes onto solid supports has gained attention as an alternative to traditional chemical processes due to enhanced enzymatic performance and stability. This study demonstrates the co-immobilization of glucose oxidase (GOx) and horseradish peroxidase (HRP) as an enzyme system on Metal-Organic Frameworks (MOFs), UiO-66 and UiO-66-NH2, that produces a more effective biocatalyst as shown by the oxidation of pyrogallol. The two MOFs utilized as solid supports for immobilization were chosen to investigate how modifications of the MOF linker affect stability at the enzyme/MOF interface and subsequent activity of the enzyme system. The enzymes work in concert with activation of HRP through the addition of glucose as a substrate for GOx. Enzyme immobilization and leaching studies showed HRP/GOx@UiO-66-NH2 immobilized 6% more than HRP/GOx@UiO-66, and leached only 36% of the immobilized enzymes over three days in the solution. The enzyme/MOF composites also showed increased enzyme activity in comparison with the free enzyme system: the composite HRP/GOx@UiO-66-NH2 displayed 189 U/mg activity and HRP/GOx@UiO-66 showed 143 U/mg while the free enzyme showed 100 U/mg enzyme activity. This increase in stability and activity is due to the amine group of the MOF linker in HRP/GOx@UiO-66-NH2 enhancing electrostatic interactions at the enzyme/MOF interface, thereby producing the most stable biocatalyst material in solution. The HRP/GOx@UiO-66-NH2 also showed long-term stability in the solid state for over a month at room temperature.

Abstract:The porous material has been considered as a potential candidate for immobilizing enzymes. Recently, metal organic framework (MOF) has been emerged as a hybrid organic inorganic material with unique intrinsic properties such as well-defined pore structure, excellent chemico-thermal stability, and extremely high surface areas which make them as a suitable scaffold for enzyme immobilization. The outstanding improvement in catalytic performance, high enzyme loading capacity, remarkable interaction between enzyme and MOF are the key features of the novel enzyme–MOF biocomposites. Amongst different immobilization approaches of enzyme–MOF composite development, de novo strategy received immense attention due to rapid, facile, mild immobilization procedure which exhibits potentially superior catalytic activity and extraordinary operational stability. This review presents a holistic insight of two different de novo strategies i.e. co-precipitation and biomineralization with state-of-art examples. Further, the recent developments in enzyme–MOF composites along with their potential features and characteristics are exploited in terms of catalytic activity, thermal/chemical stability, Michaelis–Menten kinetics, recyclability and storage stability. The advanced de novo strategies such as multi-enzyme catalytic system and magnetic enzyme–MOF are explored in the latter part of highlights.

Abstract:Metal-organic frameworks (MOFs) for enzyme encapsulation-induced biomimetic mineralization under mild reaction conditions are commonly microporous and hydrophobic, which result in a rather high mass transfer resistance of the reactants and restrain the enzyme catalytic activity. Herein, we prepared a type of hierarchical porous and hydrophilic MOF through the biomimetic mineralization of enzymes, zinc ions, 2-methylimidazole, and lithocholic acid. The hierarchical porous structure accelerated the diffusion process of the reactants and the increased hydrophilicity conferred interfacial activity and increased the enzyme catalytic activity. The immobilized enzyme retained higher catalytic activity than the free enzyme and exhibited enhanced resistance to alkaline, organic, and high-temperature conditions. The nanobiocatalyst was reusable and showed long-term storage stability.

Abstract:Enzyme immobilization has been demonstrated to be a favorable protocol to promote industrialization of biomacromolecules. Despite tremendous efforts to develop new strategies and materials to realize this process, maintaining enzyme activity is still a formidable challenge. Herein we created a sacrificial templating method, using metal–organic frameworks (MOFs) as sacrificial templates to construct hollow covalent organic framework (COF) capsules for enzyme encapsulation. This strategy can provide a capacious microenvironment to unleash enzyme molecules. The improved conformational freedom of enzymes, enhanced mass transfer, and protective effect against the external environment ultimately boosted the enzymatic activities. We also found that this strategy possesses high versatility that is suitable for diverse biomacromolecules, MOF templates, and COF capsules. Moreover, the dimensions, pore sizes, and shell thickness of COF capsules can be conveniently tuned, allowing for customizing bioreactors for specific functions. For example, coencapsulation of different enzymes with synergistic functions were successfully demonstrated using this bioreactor platform. This study not only opens up a new avenue to overcome the present limitations of enzymatic immobilization in porous matrixes but also provides new opportunities for construction of biomicrodevices or artificial organelles based on crystalline porous materials.
