Abstract:Breast cancer is the most prevalent type of malignancy in women worldwide. Taxanes (paclitaxel and docetaxel) are widely applied as first-line chemotherapeutic agents, while the therapeutic effect is seriously limited by the development of drug resistance. In the present study, we screened out several miRNAs dysregulated in taxanes-resistant breast cancer samples and confirmed that two miRNAs (miR-335-5p and let-7c-5p) played a major role in cell proliferation, apoptosis, and chemo-resistance. In addition, the weighted gene co-expression network analysis (WGCNA) for potential target genes of miR-335-5p and let-7c-5p identified three hub genes (CXCL9, CCR7, and SOCS1) with a positive relationship to taxanes-sensitivity. Further, target relationships between miR-335-5p and CXCL9, let-7c-5p and CCR7/SOCS1 were confirmed by dual-luciferase reporter assays. Importantly, the regulatory functions of CXCL9, CCR7, and SOCS1 on proliferation and chemoresistance were validated. In conclusion, our study shed light on clinical theragnostic relationships between miR-335-5p/CXCL9, let-7c-5p/CCR7/SOCS1 axes, and taxanes-resistance in breast cancer.

Abstract:Objective:To establish a new paclitaxel resistant cell line of human ovarian carcinoma for the study on mechanism of drug resistance.Methods:A paclitaxel resistant human ovarian carcinoma cell line(SKOV3-TR30) was induced by moderate concentration and intermittent action combined with low concentration and continuously exposure to paclitaxel.The resistance index was determined by MTT assay,the cell cycle was detected by flow cytometry.Cell morphology was observed using HE staining and electron microscopy.Results:The SKOV3-TR30 cells were 27.5-fold resistant to paclitaxel compared with the parental cell line.Various cross-resistance to docetaxel,topotecan,adriamycin,and nocodazole,but not carboplatin was shown.The doubling time,cell cycle and cell morphology of SKOV3-TR30 were inequable to those of the parental cell line.Conclusion:Paclitaxel resistant SKOV3 subline is a good experimental model in vitro for the study on mechanism of drug resistance and the screening of anti-tumor drug.

Abstract:Objective To explore the biological characteristics and mechanisms of drug resistance in Taxol-induced drug resistant human lung adenocarcinoma cell line(A549/Taxol).Methods The cell line A549/Taxol was established by intermittent-inducing method of gradually increasing the concentration of Taxol in vitro.Its drug resistance,cell cycle and cell invasion were detected by MTT assay,flow cytometry and transwell chamber,respectively.The expressions of BRCA1,RAP80,Tau and MDR1 mRNA were measured by quantitative RT-PCR.Results The cell line A549/Taxol was resistant to both Taxol and Docetaxel,which was more resistant to Taxol than that to Docetaxel(RI=48.36 vs.27.21)(P<0.01).Compared with cell line A549,in cell line A549/Taxol,G1 phase cell fraction increased [(50.56±0.25)% vs.(57.75±0.16)%],S phase cell fraction decreased[(37.85±1.48)% vs.(30.21±1.87)%],the increase in apoptosis rate was smaller[(56.43±1.12)% vs.(9.23±1.18)%],the ability of cell invasion increased[(38.6±9.97) pieces vs.(116.8±21.73) pieces],the levels of BRCA1 and RAP80 declined while the levels of MDR1 and Tau significantly increased(P<0.01).Conclusion The cell line A549/Taxol has been established successfully,which is helpful in the further study on the mechanism for drug resistance.

Abstract:Cancer multidrug resistance (MDR) is a common cause of treatment failure in cancer patients. Increased expression of permeability glycoprotein (P-gp), which is also known as MDR-1, is the main cause of multidrug resistance. Podophyllotoxin derivatives hold great promise in the battle to overcome multidrug resistance, as they can induce cytotoxicity through multiple mechanisms. Here, we synthesized sixteen novel podophyllotoxin derivatives and evaluated their cytotoxicities in human cancer cell lines, HeLa, K562 and K562/A02. Some of these compounds were more potent than etoposide, a clinically relevant inhibitor of DNA repair enzymes. In particular, compound 5p exhibited the most potent activity toward drug-resistant K562/A02 cells, as it robustly inhibited tumor cell proliferation and induced apoptosis. Furthermore, preliminary investigation suggested that 5p inhibited the expression of MDR-1 in K562/A02 cells more effectively than etoposide.

Abstract:Objective To establish the paclitaxel-resistant gastric cancer cell(HGC-27/PTX) and to investigate the changes of characteristics before and after resistance,as well as the possible resistant mechanisms.Methods The paclitaxel-resistant gastric cancer cell HGC-27/PTX was established by increasing paclitaxel dose gradually and intermittently.The IC50 (50% inhibitory concentration) and cell cycle were determined by CCK-8 assay and flow cytometry,respectively.The differentially expressed genes (DEGs) and signaling pathways were analyzed using RNAseq.Results The establishment of HGC-27/PTX cells lasted 9 months,and the sensitivity of paclitaxel of HGC-27/PTX cells was significantly lower than parental cells (P<0.05).Compared to parental cells,the morphology of HGC-27/PTX cells was slightly different,and the proportion of S and G2/M phase was obviously increased (P<0.01).A total of 274 DEGs were identified between the resistant and parental cells with 130 genes up-regulated and 144 genes down-regulated.DEGs were significantly enriched in extracellular matrix (ECM)-receptor interaction(P<0.001) and PI3K-Akt signaling pathways (P<0.05),which could provide evidences for reversing paclitaxel resistance.Conclusions The paclitaxel-resistant gastric cancer cells HGC-27/PTX was established with stable culturein vitro,which provides an ideal model for future study on the mechanism of drug resistance.

Abstract:Taxol (paclitaxel) response is profiled against a panel of clinical missence mutations in β‐tubulin at molecular and cellular levels, aiming to understanding of molecular mechanism and biological implication underlying drug resistance and sensitivity conferring from tubulin mutations. The Paclitaxel‐resistant mutations can be classified into three types, namely nonbonded interaction broken due to mutation, steric hindrance caused by mutation, and conformational change upon mutation. Taxol (paclitaxel) is the first approved microtubule‐stabilizing agent (MSA) by binding stoichiometrically to tubulin, which is considered to be one of the most significant advances in first‐line chemotherapy against diverse tumors. However, a large number of residue missence mutations harboring in the tubulin have been observed to cause acquired drug resistance, largely limiting the clinical application of Taxol and its analogs in chemotherapy. A systematic investigation of the intermolecular interactions between the Taxol and various tubulin mutants would help to establish a comprehensive picture of drug response to tubulin mutations in clinical treatment of cancer, and to design new MSA agents with high potency and selectivity to overcome drug resistance. In this study, we described an integration of in silico analysis and in vitro assay (iSiV) to profile Taxol against a panel of 149 clinically observed, cancer‐associated missence mutations in β‐tubulin at molecular and cellular levels, aiming to a systematic understanding of molecular mechanism and biological implication underlying drug resistance and sensitivity conferring from tubulin mutations. It is revealed that the Taxol‐resistant mutations can be classified into three types: (I) nonbonded interaction broken due to mutation, (II) steric hindrance caused by mutation, and (III) conformational change upon mutation. In addition, we identified three new Taxol‐resistant mutations (C239Y, T274I, and R320P) that can largely reduce the binding affinity of Taxol to tubulin at molecular level, in which the T274I and R320P were observed to considerably impair the antitumor activity of Taxol at cellular level. Moreover, a novel drug‐susceptible mutation (M363T) was also identified, which improves Taxol affinity by 2.6‐fold and decreases Taxol antitumor EC50 values from 29.4 to 18.7 μM.

Activation of the γ-secretase/NICD-PXR/Notch pathway induces Taxol resistance in triple-negative breast cancer

Abstract:Triple-negative breast cancer (TNBC) is currently the only subtype lacking efficient targeted therapies. Taxol is the primary chemotherapeutic agent for TNBC. However, Taxol resistance often develops in the treatment of TNBC patients, which importantly contributes to high mortality and poor prognosis in TNBC patients. Recent preclinical studies have shown that the inhibition of Notch pathway by γ-secretase inhibitors can slow down the progression of TNBC. Our studies in bioinformatic analysis of breast cancer patients and TNBC/Taxol cells in vitro showed that there was high correlation between the activation of Notch pathway and Taxol resistance in TNBC. Increased γ-secretase activity (by the overexpression of catalytic core PSEN-1) significantly reduced Taxol sensitivity of TNBC cells, and enhanced biological characteristics of malignancy in vitro, and tumour growth in vivo. Mechanistically, increased γ-secretase activity led to the accumulation of NICD in the nucleus, promoting the interaction between NICD and PXR to activate PXR, which triggered the transcription of PXR downstream associated drug resistance genes. Furthermore, we showed that pharmacological inhibition of γ-secretase with γ-secretase inhibitors (Nirogacestat and DAPT) can reverse Taxol resistance in vivo and in vitro. Our results for the first time demonstrate that the activation of γ -secretase/NCD-PXR/Notch pathway is one of important mechanisms to cause Taxol resistance in TNBC, and the blockades of this pathway may represent a new therapeutic strategy for overcoming Taxol resistance in TNBC.

Abstract:Paclitaxel is commonly used to treat breast, ovarian, lung, esophageal, gastric, pancreatic cancer, and neck cancer cells. Cancer recurrence is observed in patients treated with paclitaxel due to paclitaxel resistance emergence. Resistant mechanisms are observed in cancer cells treated with paclitaxel, docetaxel, and cabazitaxel including changes in the target molecule β-tubulin of mitosis, molecular mechanisms that activate efflux drug out of the cells, and alterations in regulatory proteins of apoptosis. This review discusses new molecular mechanisms of taxane resistance, such as overexpression of genes like the multidrug resistance genes and EDIL3 , ABCB1 , MRP1 , and TRAG-3/CSAG2 genes. Moreover, significant lncRNAs are detected in paclitaxel resistance, such as lncRNA H19 and cross-resistance between taxanes. This review contributed to discovering new treatment strategies for taxane resistance and increasing the responsiveness of cancer cells toward chemotherapeutic drugs.

Abstract:Objective To establish a docetaxel-induced human breast carcinoma drug-resistant cell line BT549/docetaxel and explore the multidrug resistance gene adenosine triphosphate binding cassette transporter G2 (ABCG2),adenosine triphosphate binding cassette subfamily B member 1 (ABCB1),multidrug resistance associated protein 1 (ABCC1),glutathione S-transferase-π (GST-π),dihydmfolate reductase (DHFR),topoisomerase Ⅱ (Topo Ⅱ) and β-tubulin Ⅲ (TUBB3) genes expression and significance.Methods The drug-resistant cell line BT549/docetaxel was established in culture by exposing BT549 parent cells to moderate concentration of docetaxel over several months,The resistant index to docetaxel was determined by the methyl thiazolyl tetrazolium (MTT) assay,Cell colony formation was used to determine the sensitivity of BT549/docetaxel to docetaxel.HPLC was used to determine the intracellular concentration of docetaxel.Reverse transcriptase-polymerase chain reaction (RT-PCR) was used m detect the expression of ABCG2,ABCB1,ABCC1,GST-π,DHFR,Topo Ⅱ and TUBB3 genes in BT549/docetaxel cells.Results The resistant index of BT549/docetaxel was 18,3,Cryopreservation had no significant effect on the characteristics of anti-tumor drug resistance of BT549/docetaxel,After being cultured in RPMI 1640 without docetaxel for a period of 1 month,BT549/docetaxel showed a decrease in the resistant index,The BT549/docetaxel cells were 18.1 times more resistant to docetaxel and also demonstrated crossresistance to cisplatin,5-fluorouracil,cyclophosphamide and other anticancer drugs to some extent,The concentration of docetaxel in BT549/doeetaxel cells was lower than that in BT549 cells.In BT549/docetaxel cells,the expression levels of ABCG2,ABCB1,ABCC1,GST-π and TUBB3 genes were 0.97 ±0.15,0.75 ±0.13,0.77 ±0.14,1.49 ±0.31 and 1.52 ±0.22,significantly higher than those in BT549 cells (0.45 ±0.19,0.37 ±0.12,0.33 ±0.11,0.76 ±0.33,and 0.33 ±0.21,P ＜0.05 for all).Conclusion A stable human breast docetaxel resistant cell line BT549/docetaxel was successfully established.Its resistance mechanism may be related with the up-regulation of ABCG2,ABCB1,ABCC1,GST-π and TUBB3 genes. Overcoming Taxol-resistance in A549 cells: A comprehensive strategy of targeting P-gp transporter, AKT/ERK pathways, and cytochrome P450 enzyme CYP1B1 by 4-hydroxyemodin

Abstract:Taxol-based chemotherapy is widely used as the first-line treatment for non-small cell lung cancer (NSCLC), however, the subsequent development of taxol-resistance is a major concern and challenge, resulting in tumor relapse and poor prognosis. Given the complex nature of taxol-resistance, we further delved into its mechanisms and demonstrated that CYP1B1 was associated to taxol response in taxol-resistant A549/Taxol cells. Compared to its parent A549 counterpart, A549/Taxol presented much higher level of CYP1B1, which was paralleled by increased aryl hydrocarbon receptor (AhR) expressions likely due to the long term taxol exposure and thereby allowed a subsequent up-regulation of CYP1B1. Inhibition of CYP1B1 by TMS [(E)-2,3',4,5'-tetramethoxystilbene], the specific CYP1B1 inhibitor, remarkably enhanced the sensitivity of A549/Taxol to taxol. Moreover, pre-incubation of taxol with human recombinant CYP1B1 did not affect drug toxicity in A549 cells, precluding the possibility of drug resistance ascribed to CYP1B1 due to directly inactivating taxol. Indeed, CYP1B1 is responsible for bio-transforming estrogen (E2) into the carcinogenetic metabolite that would inhibit microtubule stabilization induced by taxol and thereby compromising treatment efficacy. Remarkably, our data revealed potent CYP1B1 inhibition efficacy of 4-hydroxyemodin (HEM) as reflected by both molecular docking simulations and EROD assay, which posed HEM the advantage of breaking the vicious circle between E2 and CYP1B1, not only favoring to overcome taxol-resistance, but also offering long term benefit via circumventing carcinogenesis and tumor progression induced by E2. In addition to CYP1B1 inhibition, HEM notably inhibited P-gp activity and expression, a common feature of drug resistance, as well as significantly inactivated AKT/ERK pathways that contributed to the cell proliferation, migration, and drug resistance. Thus, HEM may act in concert to overcome taxol-resistance through comprehensive targeting three considered arms of drug-resistance mechanisms. Moreover, HEM profoundly resisted E2-stimulated cell migration in both A549 and A549/Taxol cells, a primary reason for tumor patients' mortality, as well as inflicted selective injury to A549/Taxol cells rather than normal lung cells, supporting HEM to be a promising agent for overcoming taxol-resistance in A549 cells.
Study on Biological Characteristics and Mechanism of Paclitaxel Induced Drug Resistance in Endometrial Carcinoma Cells

Abstract:Objective . To study the biological characteristics of paclitaxel resistant endometrial carcinoma cells and its mechanism of drug resistance. Method . The paclitaxel resistant cell lines were established by high-dose paclitaxel (TAX) injection. The IC50 of paclitaxel was determined by CCK-8 assay in Ishikawa and Ishikawa-TAX. The cell cycle and apoptosis rate were detected by flow cytometry. Western blot was used to detect the expression of p-AKT and p-p70S6K. The expression of drug resistance-related genes Pgp and MDR1 was determined by RT-PCR. Cell viability was determined by soft agarose assay and invasive ability in vitro by transwell assay. Results . Paclitaxel and NVP-BEZ235 cotreatment group can further inhibit the clonogenicity and invasion of Ishikawa and Ishikawa-TAX cells compared with paclitaxel alone and NVP-BEZ235 treatment group. Paclitaxel and NVP-BEZ235 cotreated groups increased the apoptosis rate of Ishikawa and increased G0/G1 phase arrest in both cells. Paclitaxel alone significantly inhibited p-AK and p-p70 S6K protein expression in Ishikawa and Ishikawa-TAX cells and the inhibition was enhanced by NVP-BEZ235 when cotreated with paclitaxel. Conclusion . Paclitaxel can inhibit Ishikawa and Ishikawa-TAX cell via PI3K/Akt/mTOR signaling pathway. Paclitaxel and NVP-BEZ235 cotreatment can enhance the inhibitory effect.

Abstract:Objective To establish paclitaxel-resistant cell line of human ovarian cancer(Skov-3/PTX)and to investigate its biological features.Methods Paclitaxel-resistant cell line of human ovarian cancer(Skov-3/PTX)was established by continuous stepwise selection in increasing concentration of paclitaxel.Cell morphology,growth curve and population doubling time,cell uptake of Rh-123 and paclitaxel,mRNA level of MDR1,MRP and GST-π were investigated to determine the biological features of Skov-3/PTX cell line.Results The resistance index(RI)of Skov-3/PTX cells was up to 45.90.They showed an obvious cross-resistance to gefitinib,9-nitrocamptothecin and adriamycin.The results of uptake of Rh-123 and PTX further proved that Skov-3/PTX cells had significant resistance to Rh-123 and PTX,and P-glycoprotein substrate,CsA could increase the uptake of drug in resistant cells.PT-PCR studies showed that the levels of MDR1,MRP and GST-π mRNA expression in the Skov-3/PTX cells were higher than the sensitive cells.Conclusions Skov-3/PTX cell line shows typical multidrug resistance phenotypes and might serve as an ideal model for exploring the new route of reversing multidrug resistance.

Abstract:Resveratrol, which is highly concentrated in the skin of grapes and is abundant in red wine, has been demonstrated to account for several beneficial properties, including antioxidant, anticoagulant, anti-inflammatory and anticancer effects. Taxol is a microtubule-stabilizing drug that has been extensively used as effective chemotherapeutic agents in the treatment of solid tumors. Here, we investigated whether the combination of the two compounds would yield increased antitumor efficacy in human cancer cells. Unexpectedly, resveratrol effectively prevented tumor cell death induced by taxol in 5637 bladder cancer cells. This pronounced antagonistic function of resveratrol against taxol was associated with changes in multiple signal transduction pathways, but not with tubulin polymerization. Importantly, cell cycle analysis showed that resveratrol prevented the cells from entering into mitosis, the phase in which taxol exerts its action. Furthermore, resveratrol blocked the cytotoxic effects of vinblastine but not cisplatin in 5637 cells. Interestingly, resveratrol pre-treatment followed by taxol resulted in synergistic cytotoxicity. Finally, we extended our studies to various human cancer cell lines. Taken together, our results indicate that resveratrol may have the potential to negate the therapeutic efficacy of taxol and suggest that consumption of resveratrol-related products may be contraindicated during cancer therapy with taxol.

Abstract:Objective To establish a docetaxel-induced human breast carcinoma drug-resistant cell line BT549/docetaxel and explore the multidrug resistance gene adenosine triphosphate binding cassette transporter G2 (ABCG2),adenosine triphosphate binding cassette subfamily B member 1 (ABCB1),multidrug resistance associated protein 1 (ABCC1),glutathione S-transferase-π (GST-π),dihydmfolate reductase (DHFR),topoisomerase Ⅱ (Topo Ⅱ) and β-tubulin Ⅲ (TUBB3) genes expression and significance.Methods The drug-resistant cell line BT549/docetaxel was established in culture by exposing BT549 parent cells to moderate concentration of docetaxel over several months,The resistant index to docetaxel was determined by the methyl thiazolyl tetrazolium (MTT) assay,Cell colony formation was used to determine the sensitivity of BT549/docetaxel to docetaxel.HPLC was used to determine the intracellular concentration of docetaxel.Reverse transcriptase-polymerase chain reaction (RT-PCR) was used m detect the expression of ABCG2,ABCB1,ABCC1,GST-π,DHFR,Topo Ⅱ and TUBB3 genes in BT549/docetaxel cells.Results The resistant index of BT549/docetaxel was 18,3,Cryopreservation had no significant effect on the characteristics of anti-tumor drug resistance of BT549/docetaxel,After being cultured in RPMI 1640 without docetaxel for a period of 1 month,BT549/docetaxel showed a decrease in the resistant index,The BT549/docetaxel cells were 18.1 times more resistant to docetaxel and also demonstrated crossresistance to cisplatin,5-fluorouracil,cyclophosphamide and other anticancer drugs to some extent,The concentration of docetaxel in BT549/doeetaxel cells was lower than that in BT549 cells.In BT549/docetaxel cells,the expression levels of ABCG2,ABCB1,ABCC1,GST-π and TUBB3 genes were 0.97 ±0.15,0.75 ±0.13,0.77 ±0.14,1.49 ±0.31 and 1.52 ±0.22,significantly higher than those in BT549 cells (0.45 ±0.19,0.37 ±0.12,0.33 ±0.11,0.76 ±0.33,and 0.33 ±0.21,P ＜0.05 for all).Conclusion A stable human breast docetaxel resistant cell line BT549/docetaxel was successfully established.Its resistance mechanism may be related with the up-regulation of ABCG2,ABCB1,ABCC1,GST-π and TUBB3 genes.
Establishment and Characterization of a Docetaxel-Resistant Variant of Human Lung Adenocarcinoma Cell Line SPC-A1

Abstract:Background and qurpose:Docetaxel is one of the most effective chemotherapeutic agents developed in the past few years and has been used for the treatment of various cancers including lung cancer. Drug resistance to docetaxel is one of the main factors accounting for the failure of chemotherapy. This study was done to establish a human lung adenocarcinoma cell line SPC-A1 with the characterization of docetaxel resistance, to investigate its biological mechanism of drug resistance and how to reverse the resistance. Methods:A docetaxel resistant human lung adenocarcinoma cell line SPC-A1/Docetaxel was induced by continuously exposing human lung adenocarcinoma cell line SPC-A1 to gradually increasing doses of docetaxel. The multidrug resistance of SPC-A1/Docetaxel was evaluated by MTT assay. The distribution of cell cycle and rhodamine 123 accumulation in the two cell lines were detected by flow cytometry. Growth fraction was calculated by cytometry, and differentiation of genetics between drug resistance cell line and its parent cell line was analyzed by karyotype analysis.Results:Index number of drug resistance (IR) of SPC-A1/Docetaxel were 13.20, 2.18,1.12,1.39,1.38,0.93, 10.14 and 2.12 to docetaxel, paclitaxel, pharmorubicin, cisplatin,carboplatin, gemzar, vinorelbine and etoposide , respectively. Flow cytometry analysis showed G1/G0 arrest in SPC-A1/Docetaxel cell lines and there was no difference in terms of G2 cell proportion between SPC-A1/Docetaxel and SPC-A1 cell lines. Rhodamine concentration in SPC-A1/Docetaxel cells were markedly lower than that in SPC-A1 cells(806.34±0.49 Vs. 1382.26±0.32). Cell doubling time of SPC-A1/Docetaxel cells and SPC-A1 cells were 35.1 and 27.4 hours, respectively, with no difference in terms of genetics between the two cell lines.Conclusions:The SPC-A1/Docetaxel cell line showed high resistance to docetaxel, and had the characteristic of multidrug resistance. The cell line could be useful for the study of drug resistant mechanism and its reversal of docetaxel.
