Abstract:In this study, marine mussels (Mytilus coruscus) were exposed to three typical PAEs (dimethyl phthalate [DMP], dibutyl phthalate [DBP] and di(2-ethylhexyl) phthalate [DEHP]) at a range of doses for different times to investigate the ecotoxicological effects. The accumulation of the three PAE congeners in M. coruscus exhibited the following trend: DEHP > DBP > DMP. The antioxidant response of mussel gonadal tissue was enhanced with increasing concentrations of PAEs. For the DBP and DEHP treatment groups, glutathione (GSH) worked in concert with antioxidant enzymes to protect cells against reactive oxygen species (ROS), while GSH played a prominent antioxidant role in the DMP-treated group. The metabolomics results revealed that PAE exposure disrupted the metabolic balance of mussels. Overall, PAEs affect the amino acid metabolism, lipid metabolism, energy metabolism, osmoregulation and nerve activities of mussels. Our results provide further insight into the toxicological effects of PAEs on marine organisms.

Abstract:Both benzo(α)pyrene (BaP) and metals are frequently found in marine ecosystem and can cause detrimental effects in marine organism, especially the filter feeder-marine mussels. Although the biological responses in mussels have been well-studied upon the single metal or BaP exposure, the information about antioxidant defense, especially in different tissues of mussels, are still limited. Considering the variety of contaminants existing in the actual marine environment, single BaP (56 μg/L) and the co-exposure with Cu, Cd and Pb (50 μg/L, 50 μg/L and 3 mg/L respectively) were applied in a 6 days exposure followed by 6 days depuration experiment. The alterations of superoxide dismutase (SOD), catalase (CAT) activities and total antioxidant capacity (TAC) level were assessed in haemolymph, gills and digestive glands of marine mussels, Mytilus coruscus. An unparalleled change in antioxidant biomarkers was observed in all cells/tissues, with the SOD activity showing higher sensitivity to exposure. A tissue-specific response showing unique alteration in gill was investigated, indicating the different function of tissues during stress responses. Depressed antioxidant effects were induced by BaP-metals co-exposure, indicating the interaction may alter the intact properties of BaP. To our knowledge, this is the first research to explore the antioxidant defense induced by combined exposure of BaP-metals regarding to tissue-specific responses in marine mussels. The results and experimental model will provide valuable information and can be utilized in the investigation of stress response mechanisms, especially in relation to tissue functions in marine organism in the future.
Assessing periodontal pathogens in children with varying levels of oral hygiene.

Abstract:This study investigated the presence of organisms Actinobacillus actinomycetecomitans (A.a.), Bacteroides intermedius (B.i.), and Bacteroides gingivalis (B.g.), believed to be associated with periodontal disease etiology in children. The study included fifty children, aged six to fourteen years, selected from patients seeking routine dental care at Children's Hospital National Medical Center. A Modified Plaque Index (MPI) denoting the clinical presence and severity of the periodontal disease was obtained for each patient and ranged from 0-3. A total of 200 samples were obtained from the gingival crevice of the mesial surface of all first permanent molars and tested with DNA probes for the presence of three microorganisms. No correlations were found between race, sex, age, and the Modified Plaque Index. The results show that A. actinomycetecomitans was found only in patients with MPI greater than or equal to 1. B. gingivalis was present only in combination with another organism.
Identification of proteomic signatures of exposure to marine pollutants in mussels (Mytilus edulis)

Abstract:Bivalves and especially mussels are very good indicators of marine and estuarine pollution, and so they have been widely used in biomonitoring programs all around the world. However, traditional single parameter biomarkers face the problem of high sensitivity to biotic and abiotic factors. In our study, digestive gland peroxisome-enriched fractions of Mytilus edulis (L., 1758) were analyzed by DIGE and MS. We identified several proteomic signatures associated with the exposure to several marine pollutants (diallyl phthalate, PBDE-47, and bisphenol-A). Animals collected from North Atlantic Sea were exposed to the contaminants independently under controlled laboratory conditions. One hundred and eleven spots showed a significant increase or decrease in protein abundance in the two-dimensional electrophoresis maps from the groups exposed to pollutants. We obtained a unique protein expression signature of exposure to each of those chemical compounds. Moreover a set of proteins composed a proteomic signature in common to the three independent exposures. It is remarkable that the principal component analysis of these spots showed a discernible separation between groups, and so did the hierarchical clustering into four classes. The 14 proteins identified by MS participate in alpha- and beta-oxidation pathways, xenobiotic and amino acid metabolism, cell signaling, oxyradical metabolism, peroxisomal assembly, respiration, and the cytoskeleton. Our results suggest that proteomic signatures could become a valuable tool to monitor the presence of pollutants in field experiments where a mixture of pollutants is often present. Further studies on the identified proteins could provide crucial information to understand possible mechanisms of toxicity of single xenobiotics or mixtures of them in marine ecosystems.

Abstract:The ubiquitous application of phthalate esters (PAEs) as plasticizers contributes to high levels of marine pollution, yet the contamination patterns of PAEs in various shellfish species remain unknown. The objective of this research is to provide the first information on the pollution characteristics of 16 PAEs in different shellfish species from the Pearl River Delta (PRD), South China, and associated health risks. Among the 16 analyzed PAEs, 13 were identified in the shellfish, with total PAE concentrations ranging from 23.07 to 3794.08 ng/g dw (mean = 514.35 ng/g dw). The PAE pollution levels in the five shellfish species were as follows: Ostreidae (mean = 1064.12 ng/g dw) > Mytilus edulis (mean = 509.88 ng/g dw) > Babylonia areolate (mean = 458.14 ng/g dw) > Mactra chinensis (mean = 378.90 ng/g dw) > Haliotis diversicolor (mean = 335.28 ng/g dw). Dimethyl phthalate (DMP, mean = 69.85 ng/g dw), diisobutyl phthalate (DIBP, mean = 41.39 ng/g dw), dibutyl phthalate (DBP, mean = 130.91 ng/g dw), and di(2-ethylhexyl) phthalate (DEHP, mean = 226.23 ng/g dw) were the most abundant congeners. Notably, DEHP constituted the most predominant fraction (43.98 %) of the 13 PAEs detected in all shellfish from the PRD. Principal component analysis indicated that industrial and domestic emissions served as main sources for the PAE pollution in shellfish from the PRD. It was estimated that the daily intake of PAEs via shellfish consumption among adults and children ranged from 0.004 to 1.27 μg/kgbw/day, without obvious non-cancer risks (< 0.034), but the cancer risks raised some alarm (2.0 × 10-9-1.4 × 10-5). These findings highlight the necessity of focusing on marine environmental pollutants and emphasize the importance of ongoing monitoring of PAE contamination in seafood.

Abstract:Due to the rising usage of plastics, plastic debris are present throughout marine ecosystems and detrimentally affects marine biota. Additionally, plastics likely result in elusive toxicity effects due to addition of plasticizers. The aim of the present study was to reveal the potential effects and mechanism of microplastics (MPs), di-(2-ethylhexyl) phthalate (DEHP) and copollution of MPs and DEHP (MPs-DEHP) on Peneaus vannamei ( P. vannamei ) juveniles regarding oxidative stress, transcriptomics and metabolomics. MPs, DEHP and MPs-DEHP significantly induced the activities of superoxide dismutase (SOD) and catalase (CAT); MPs and DEHP have an antagonistic effect for malondialdehyde (MDA); suggesting that disorders of the antioxidant defence systems. 13, 133 and 58 differentially expressed genes and 21, 82 and 39 differentially expressed metabolites were responsible for the distinction of MPs, DEHP and MPs-DEHP groups, respectively. The combination of transcriptomic and metabolomic analyses showed that MPs, DEHP and MPs-DEHP exposure disturbed amino acid and lipid metabolism, and further induced inflammatory responses and dysfunction of purine metabolism. Furthermore, the presence of MPs might alleviate the biotoxicity of DEHP in P. vannamei . These findings provide new insights into the single and combined toxicological effects of MPs and additives for marine biota.

Abstract:The presence and persistence of microplastics (MPs) in diverse aquatic environments are of global concern. Microplastics can impact marine organisms via direct physical interaction and the release of potentially harmful chemical additives incorporated into the plastic. These chemicals are physically bound to the plastic matrix and can leach out. The hazards associated with chemical additives to exposed organisms is not well characterized. We investigated the hazards of plastic additives leaching from plastic. We used the common plasticizer dibutyl phthalate (DBP) as a chemical additive proxy and the New Zealand green‐lipped mussel (Perna canaliculus) as a model. We used early‐adult P. canaliculus exposed to combinations of virgin and DBP‐spiked polyvinyl chloride (PVC), MPs, and DBP alone for 7 days. Whole transcriptome sequencing (RNA‐seq) was conducted to assess whether leaching of DBP from MPs poses a hazard. The differences between groups were evaluated using pairwise permutational multivariate analysis of variance (PERMANOVA), and all treatments were significantly different from controls. In addition, a significant difference was seen between DBP and PVC MP treatment. Transcriptome analysis revealed that mussels exposed to DBP alone had the most differentially expressed genes (914), followed by PVC MP + DBP (448), and PVC MP (250). Gene ontology functional analysis revealed that the most enriched pathway types were in cellular metabolism, immune response, and endocrine disruption. Microplastic treatments enriched numerous pathways related to cellular metabolism and immune response. The combined exposure of PVC MP + DBP appears to cause combined effects, suggesting that DBP is bioavailable to the exposed mussels in the PVC MP + DBP treatment. Tris(1-chloro-2-propyl) phosphate enhances the adverse effects of biodegradable polylactic acid microplastics on the mussel Mytilus coruscus

Abstract:Microplastics (MPs) and organophosphate flame retardants (OPFRs) have recently become ubiquitous and cumulative pollutants in the oceans. Since OPFRs are added to or adsorbed onto MPs as additives, it is necessary to study the composite contamination of OPFRs and MPs, with less focus on bio-based PLA. Therefore, this study focused on the ecotoxicity of the biodegradable MP polylactic acid (PLA) (5 μm, irregular fragments, 102 and 106 particles/L), and a representative OPFRs tris(1-chloro-2-propyl) phosphate (TCPP, 0.5 and 50 μg/L) at environmental and high concentrations. The mussel Mytilus coruscus was used as a standardised bioindicator for exposure experiments. The focus was on examining oxidative stress (catalase, CAT, superoxide dismutase, SOD, malondialdehyde, MDA), immune responses acid (phosphatase, ACP, alkaline phosphatase, AKP, lysozyme, LZM), neurotoxicity (acetylcholinesterase, AChE), energy metabolism (lactate dehydrogenase, LDH, succinate dehydrogenase, SDH, hexokinase, HK), and physiological indices (absorption efficiency, AE, excretion rate, ER, respiration rate, RR, condition index, CI) after 14 days exposure. The results of significantly increased oxidative stress and immune responses, and significantly disturbed energy metabolism and physiological activities, together with an integrated biomarker response (IBR) analysis, indicate that bio-based PLA MPs and TCPP could cause adverse effects on mussels. Meanwhile, TCPP interacted significantly with PLA, especially at environmental concentrations, resulting in more severe negative impacts on oxidative and immune stress, and neurotoxicity. The more severe adverse effects at environmental concentrations indicate higher ecological risks of PLA, TCPP and their combination in the real marine environment. Our study presents reliable data on the complex effects of bio-based MP PLA, TCPP and their combination on marine organisms and the environment.

Abstract:Salicylic acid reduced the in situ intestinal absorption of carbutamide, and enhanced the in situ intestinal exsorption of carbutamide. Salicylic acid, however, did not affect the in vitro intestinal absorption of carbutamide. These findings indicate that the displacement of carbutamide from its plasma protein binding sites by salicylic acid can become an important factor affecting the intestinal absorption of carbutamide in intact rabbits. Salicylic acid caused a significant increase in the apparent volume of distribution (Vαβ) and in the total body clearance (ClT) of carbutamide. In addition, salicylic acid was found to significantly increase the distribution of carbutamide into the pancreas and into the kidney. These findings may be explained on the basis of the displacement of carbutamide from its plasma protein binding sites by salicylic acid.
Oxidative stress responses caused by dimethyl phthalate (DMP) and diethyl phthalate (DEP) in a marine diatom Phaeodactylum tricornutum

Abstract:A marine diatom (Phaeodactylum tricornutum) was exposed to different concentrations of dimethyl phthalate (DMP) and diethyl phthalate (DEP) for 96 h within a batch-culture system to investigate their toxicities. Results showed that P. tricornutum could remove DMP and DEP effectively with removal rates of 0.20?0.30 and 0.14?0.21 mg L-1 h-1, respectively. In addition, DMP and DEP significantly inhibited the photosynthesis and chlorophyll a biosynthesis of P. tricornutum with 96-h EC50 values of 390.5 mg L-1 and 74.0 mg L-1, respectively. Results of reactive oxygen species (ROS) level suggested that the two PAEs could induce excessive ROS production in the diatom. Moreover, activities of antioxidant enzymes (i.e., SOD and POD) in the diatom increased with the increase of DMP and DEP concentrations. The results will help to understand the toxic mechanisms of PAEs, and provide strong evidences for evaluating their ecological risks in the marine environment.
Physiological and transcriptome analysis of Mytilus coruscus in response to Prorocentrum lima and microplastics

Abstract:Nowadays, diarrheic shellfish toxicity (DSP) toxin and microplastics (MPs) are commonly found in coastal waters worldwide. Due to their widespread use, their persistence and toxicity, they may induce adverse effects on Mytilus coruscus . However, the underlying toxic mechanisms of DSP and MPs on M. coruscus remain unclear. This study explored the physiological index and transcriptome change of the digestive gland of adult M. coruscus exposed for 3 days to polystyrene (PS) MPs (0.2 mg/L, 90-100 μm) and Prorocentrum lima alone or in combination. The results showed that the CAT activity and MDA content significantly increased, respiration rate and feeding rate significantly decreased. The combination of MPs and P. lima caused more structural damage to the rough surface endoplasmic reticulum and mitochondria in the digestive glands of M. coruscus . The transcriptome analysis showed that 485 and 220 genes were up- and down-regulated, respectively, after exposure to P. lima ; 1,989 up-regulated DEGs and 1,098 down-regulated DEGs were identified after exposure to MP treatment, and 1,004 up-regulated DEGs and 664 down-regulated DEGs were identified after exposure to the combination of P. lima and MPs. The DEGs were mainly enriched in the lysosome, mRNA surveillance pathway, carbon metabolism, the mTOR signaling pathway, the complement and coagulation cascades, and the TNF signaling pathway. The MP, P. lima exposure mainly induced the expression of RNA-binding protein musashi, serine/arginine repetitive matrix protein 1, low affinity immunoglobulin epsilon Fc receptor, toll-like receptor 2, caspase 7, calmodulin, E3 ubiquitin-protein ligase, serine/threonine-protein kinase PRP4, glutathione S-transferase, and heat shock 70 kDa protein. MPs and P. lima poison mainly influence the expression of RNA transport, immune related gene, apoptosis, signal related gene, and antioxidant gene change. The combination of MPs and P. lima has a synergistic toxic effect. This study provides a new insights into its physiological and molecular responses of M. coruscus to MPs and P. lima toxic exposure.

Abstract:Pollution with complex mixtures of contaminants including micro- and nano-plastics (MNPs) and organic pollutants like polycyclic aromatic hydrocarbons (PAH) poses a major threat to coastal marine ecosystems. Toxic mechanisms of contaminant mixtures are not well understood in marine organisms. We studied the effects of single and combined exposures to polycyclic aromatic hydrocarbon phenanthrene (Phe) and MNPs mixture with sizes of 70 nm, 5 μm and 100 μm on the immune health and oxidative stress parameters in the thick-shell mussel Mytilus coruscus . Immune cells (hemocytes) were more sensitive to the pollutant-induced oxidative stress than the gills. In hemocytes of co-exposed mussels, elevated mortality, lower lysosomal content, high production of reactive oxygen species (ROS) and decrease mitochondrial were found. Disparate responses of antioxidant enzymes in the hemolymph (e.g. increased superoxide dismutase (SOD) activity without a corresponding increase in catalase (CAT) in Phe exposures and an increase in CAT without a change in SOD in MNPs exposures) suggests misbalance of the antioxidant defense in the pollutant-exposed mussels. Gill lacked pronounced oxidative stress response showing a decline in ROS and antioxidant levels. Tissue-specific single and combined effects of Phe and MNPs suggest variation in bioavailability and/or different sensitivity to these pollutants in the studied tissues. Notably, the combined effects of MNPs and Phe were additive or antagonistic, showing that MNPs do not enhance and occasionally mitigate the toxic effects of Phe on the hemocytes and the gills of the mussels. Overall, our study sheds light on the impact of long-term exposure to MNPs and Phe mixtures on mussels, showing high sensitivity of the immune system and modulation of the Phe toxicity by MNPs co-exposure. These findings that may have implications for understanding the impacts of combined PAH and MNPs pollution on the health of mussel populations from polluted coastal habitats.

Abstract:The ubiquitous occurrence of microplastics (MPs) in the marine environment is raising concern for interactions with marine organisms. These particles efficiently adsorb persistent organic pollutants from surrounding environment and, due to the small size, they are easily available for ingestion at all trophic levels. Once ingested, MPs can induce mechanical damage, sub-lethal effects, and various cellular responses, further modulated by possible release of adsorbed chemicals or additives. In this study, ecotoxicological effects of MPs and their interactions with benzo(a)pyrene (BaP), chosen as a model compound for polycyclic aromatic hydrocarbons (PAHs) were investigated in Mediterranean mussels, Mytilus galloprovincialis. Organisms were exposed for 4 weeks to 10 mg/L of low-density polyethylene (LDPE) microparticles (2.34 * 107 particles/L, size range 20–25 μm), both virgin and pre-contaminated with BaP (15 μg/g). Organisms were also exposed for comparison to BaP dosed alone at 150 ng/L, corresponding to the amount adsorbed on microplastics. Tissue localization of microplastics was histologically evaluated; chemical analyses and a wide battery of biomarkers covering molecular, biochemical and cellular levels allowed to evaluate BaP bioaccumulation, alterations of immune system, antioxidant defenses, onset of oxidative stress, peroxisomal proliferation, genotoxicity, and neurotoxicity. Obtained data were elaborated within a quantitative weight of evidence (WOE) model which, using weighted criteria, provided synthetic hazard indices, for both chemical and cellular results, before their integration in a combined index. Microplastics were localized in hemolymph, gills, and especially digestive tissues where a potential transfer of BaP from MPs was also observed. Significant alterations were measured on the immune system, while more limited effects occurred on the oxidative status, neurotoxicity, and genotoxicity, with a different susceptibility of analyzed pathways, depending on tissue, time, and typology of exposure. Molecular analyses confirmed the general lack of significant transcriptional variations of antioxidant and stress genes. The overall results suggest that microplastics induce a slight cellular toxicity under short-term (28 days) exposure conditions. However, modulation of immune responses, along with bioaccumulation of BaP, pose the still unexplored risk that these particles, under conditions of more chronic exposure (months to years) or interacting with other stressors, may provoke long-term, subtle effects on organisms' health status.

Microplastics aggravate the adverse effects of BDE-47 on physiological and defense performance in mussels

Abstract:The highly hydrophobic surfaces make microplastics a potential carrier of organic pollutants in the marine environment. In order to explore the toxic effects of polybrominated diphenyl ethers (BDE-47) combined with microplastics on marine organisms, we exposed the marine mussel Mytilus coruscus to micro-PS combined with BDE-47 for 21 days to determine the immune defense, oxidative stress and energy metabolism of the mussels. The results showed that the clearance rate (CR) of mussels exposed to single micro-PS, single BDE-47 or both was lower than control group. In general, compared to single BDE-47 exposure, the combination of micro-PS and BDE-47 significantly increased respiration rate (RR), activities of acid phosphatase (ACP) and alkaline phosphatase (ALP), reactive oxygen species (ROS) production and malondialdehyde (MDA) concentrations, but significantly decreased lactate dehydrogenase (LDH) activity and the relative expression of heat shock protein (Hsp70 and 90). Overall, combined stress has more adverse effects on defense performance and energy metabolism in mussels and micro-PS seem to exacerbate the toxicological effects of BDE-47. As microplastics pollution may deteriorate in the future, the health of mussels may be threatened in organically polluted environment, which eventually change the stability of the structure and function of intertidal ecosystem.

Abstract:Microplastics (MPs) are currently one of the major environmental problems due to the large amount released in aquatic and terrestrial ecosystems as well as their potential impacts on organisms and human health. However, the molecular mechanisms underlying MP toxicity remain elusive and insufficiently understood. In this study, oysters were exposed to irregular MPs composed of polyethylene and polyethylene terephthalate at concentrations of 10 and 1000 μg L−1 for 21 days under static conditions, and physiological responses and changes in antioxidant enzymatic activity were investigated. Moreover, the protein expression profiles in the digestive glands were analyzed by using the isobaric tags for relative and absolute quantitation (iTRAQ) method. And the expression of ten potentially regulated genes was analyzed by qRT-PCR in different tissues under MP exposure. It was demonstrated that PE and PET MP exposure had no distinct adverse effects on physiological rates, while inducing oxidative stresses in the oyster. Kyoto encyclopedia of genes and genomes (KEGG) enrichment analysis of DEPs revealed that MP exposure mainly disturbed the "arachidonic acid metabolism", "linoleic acid metabolism" and "glycerophospholipid metabolism" processes in oysters. Gene Ontology enrichment analysis revealed that MPs affected the oxidation-reduction process, lipid metabolism process and pentose phosphate pathway. Moreover, a significant increase in the mRNA expression of genes related to lipid and aerobic metabolism and apoptosis pathways was observed after MP exposure. In summary, our results indicated that MPs could induce oxidative stress and alter lipid and glucose metabolism processes in oysters. Additionally, the weight of evidence (WOE) model analysis suggested that high concentrations of MPs were more hazardous than low concentrations and typically higher for PET than PE. This study provided the effects of PE and PET MP toxicity on oysters at the molecular level, which will contribute to better understanding the defense mechanisms of oysters against MP exposure.

Abstract:Phthalate esters (PAEs), a family of emerging environmental contaminants, have been frequently detected in soils and water. However, intensive studies on the toxicity of PAEs have focused on growth response of terrestrial and aquatic animals, while only limited attention has been paid to aquatic plants, especially phytoplankton, the primary producer in aquatic ecosystems. Therefore, the acute toxic effects and underlying mechanisms of dibutyl phthalate (DBP) at different concentrations (0-20mgL-1) on two typical freshwater algae (Scenedesmus obliquus and Chlorella pyrenoidosa) were investigated. The growth of S. obliquus and C. pyrenoidosa was conspicuously inhibited by DBP exposure at 2-20mgL-1. The 96-h median effective concentration values (96h-EC50) were 15.3mgL-1 and 3.14mgL-1 for S. obliquus and C. pyrenoidosa, respectively, implying that the spherical C. pyrenoidosa is more sensitive to DBP than the spindle-shaped S. obliquus. As expected from the damage done to cell organelles (i.e. cell membranes, chloroplasts, and protein rings), cell densities and chlorophyll content conspicuously decreased under DBP treatments. Moreover, the algal growth inhibition was closely linked to the increased production of intracellular reactive oxygen species and malondialdehyde content, indicating oxidative stress and lipid peroxidation in both algae. This was proved by the increased activity of antioxidant enzymes such as superoxide dismutase and catalase. Our findings will contribute to the understanding of toxic mechanisms in PAEs and the evaluation of environmental risks for primary producers in aquatic ecosystems.
Hazards of phthalates (PAEs) exposure: A review of aquatic animal toxicology studies

Abstract:Phthalates (PAEs) are of wide concern because they are commonly used in various plastic products as plasticizers, and can found their way into the environment. However, their interaction with the environment and their toxicity in aquatic animals is still a matter of intense debate. In this review on PAEs in aquatic environments (lakes, rivers and seas), it is found that there is a large variety and abundance of PAEs in developing countries, and the total concentration of PAEs even exceeds 200 mu g / L. The interaction between metabolic processes involved in the toxicity induced by various PAEs is summarized for the first time in the article. Exposure of PAEs can lead to activation of the detoxification system CYP450 and endocrine system receptors of aquatic animals, which in turn causes oxidative stress, metabolic disorders, endocrine disorders, and immunosuppression. Meanwhile, each system can activate / inhibit each other, causing genotoxicity and cell apoptosis, resulting in the growth and development of organisms being blocked. The mixed PAEs shows no cumulative toxicity changes to aquatic animals. For the combined pollution of other chemicals and PAEs, PAE can act as an agonist or antagonist, leading to combined toxicity in different directions. Phthalate monoesters (MPEs), the metabolites of PAEs, are also toxic to aquatic animals, however, the toxicity is weaker than the corresponding parent compounds. This review summarizes and analyzes the current ecotoxicological effects of PAEs on aquatic animals, and provides guidance for future research. (C) 2021 Elsevier B.V. All rights reserved.
Polystyrene microplastics alleviate adverse effects of benzo[a]pyrene on tissues and cells of the marine mussel, Mytilus galloprovincialis

Abstract:As two major ubiquitous pollutants, microplastics (MPs) and polycyclic aromatic hydrocarbons (PAHs) coexist in the marine environment. However, the role of MPs in altering the toxicity of PAHs to marine organisms is poorly understood. We therefore investigated the accumulation and toxicity of benzo[a]pyrene (B[a]P, 0.4 nM), in the marine mussel Mytilus galloprovincialis over a 4-day of exposure with or without the presence of 10 μm polystyrene microplastics (PS MPs) (10 particles/mL). The presence of PS MPs significantly decreased B[a]P accumulation in soft tissues of M. galloprovincialis by approximately 6.7%. Single exposure of PS MPs or B[a]P decreased the mean epithelial thickness (MET) of digestive tubules and enhanced reactive oxygen species (ROS) levels in haemolymph, while upon co-exposure the adverse impacts were alleviated. Real-time q-PCR results showed that most selected genes involved in stress response ( FKBP, HSP90 ), immune ( MyD88a, NF-κB ) and detoxification ( CYP4Y1 ) were induced for both single exposure and co-exposure. The co-presence of PS MPs down-regulated the mRNA expression of NF-κB in gills compared with of B[a]P alone. The uptake and toxicity reductions of B[a]P might result from the decrease of its bioavailable concentrations caused by the adsorption of B[a]P by PS MPs and the strong affinity of B[a]P to PS MPs. Adverse outcomes for the co-existence of marine emerging pollutants under long-term conditions remain to be further validated.

Insights into the combined toxicity and mechanisms of BDE-47 and PFOA in marine blue mussel: An integrated study at the physiochemical and molecular levels

Abstract:The coexistence of multiple emerging contaminants imposes a substantial burden on the ecophysiological functions in organisms. The combined toxicity and underlying mechanism requires in-depth understanding. Here, marine blue mussel (Mytilus galloprovincialis L.) was selected and exposed to 2,2',4,4'-tetrabromodiphenyl ether (BDE-47) and perfluorooctanoic acid (PFOA) individually and in combination at environmental related concentrations to elucidate differences in stress responses and potential toxicological mechanisms. Characterization and comparison of accumulation, biomarkers, histopathology, transcriptomics and metabolomics were performed. Co-exposure resulted in differential accumulation patterns, exacerbated histopathological alterations, and different responses in oxidative stress and biomarkers for xenobiotic transportation. Moreover, the identified differentially expressed genes (DEGs) and differential metabolites (DEMs) in mussels were found to be annotated to different metabolic pathways. Correlation analyses further indicated that DEGs and DEMs were significantly correlated with the above biomarkers. BDE-47 and PFOA altered the genes and metabolites related to amino acid metabolism, energy and purine metabolism, ABC transporters, and glutathione metabolism to varying degrees, subsequently inducing accumulation differences and combined toxicity. Furthermore, the present work highlighted the pivotal role of Nrf2-keap1 detoxification pathway in the acclimation of M. galloprovincialis to reactive oxygen species (ROS) stress induced by BDE-47 and PFOA. This study enabled more comprehensive understanding of combined toxic mechanism of multi emerging contaminants pollution.
