Abstract:Using fermentable sugars from lignocelluloses to produce chemicals and fuels is a way to resolve the energy crisis and environmental problems, but nonproductive adsorption of cellulase onto lignin is big challenge for bioconversion of cellulose to fermentable sugars. As cellobiohydrolase (CBH) is the main enzyme in cellulase mixture, understanding the adsorption mechanism of CBH onto lignin is important to enhance CBH catalytic efficiency by avoiding the nonproductive adsorption. This study suggested that CBH displayed high adsorption affinity to lignin, especially for the lignin isolated from liquid-hot-water pretreated substrate, and a stable combination of lignin-CBH was formed. The CBH adsorbed to lignin still had the catalytic ability on Avicel, although the ability was decreased. The possible reason, by molecular docking, was that the catalytic domain of CBH was irreversibly adsorbed onto lignin mainly through a hydrophobic cleft located oppositely to catalytic tunnel, not influencing the catalytic process of the enzyme except for the protein movement. Electrostatic interaction played a significant role in the adsorption of carbohydrate binding domain (CBM) of CBH to lignin. Altering the charges from positive to negative of amino acid on the CBM of CBH by engineering enzyme may be a feasible way to reduce the adsorption affinity of CBH onto lignin.
Elucidating non-productive adsorption mechanism of cellulase with lignin fractions from hydrothermally pretreated poplar using multi-dimensional spectroscopic technologies

Abstract:In situ investigation of the influence of pretreatment lignin distribution and interaction with enzymes can provide insight into the mechanism of lignin limiting the enzymatic efficiency of cellulose. One of the key focuses of this paper is to elucidate the differences in cellulase hydrolysis caused by surface lignin after pretreatment (including pseudo-lignin, deposited on biomass surface lignin and soluble lignin) compared to residual lignin in the pretreated material. Particularly, the combination of different non-destructive spectroscopies (atomic force microscopy (AFM), surface plasmon resonance (SPR), and fluorescence spectrometry (FLS)) were used to examine the non-productive adsorption mechanisms of cellulase with differing lignin fractions from pretreated poplar. The results showed that surface lignin (SL) from pretreated poplar had almost no inhibitory effect on Avicel hydrolysis yield. In contrast, residual lignin (RL) in pretreated poplar significantly inhibited Avicel hydrolysis yield from 74.6% to 50.94%. Characterization results revealed that SL has lower hydrophobicity, molecular weight, and linkage content (β-O-4, β-β and β-5), which is significantly different from the structure of RL. The multi-dimensional non-destructive spectroscopy indicated that the interaction forces between SL and cellulase fractions (0.044-2.04 nN) were smaller than that of RL and cellulase fractions (0.11-3.62 nN). The SL binds to cellulase mainly through hydrogen bonds and van der Waals forces, while the RL interacts mainly through hydrophobic forces. Furthermore, SL bound to cellulase is more likely to dissociate compared with RL. This work provides a fundamental understanding of lignin-cellulase interactions for effectively converting lignocellulose into biofuels.

Abstract:The adsorption of lignin to cellulase is the major obstacle in the sugar-platform conversion of lignocellulosic bioresources. In this study, the adsorption behavior of untreated and pretreated lignin samples from corn stover to cellulase was investigated, in particular the effects of lignin adsorption on the composition and spatial conformation of free enzymes were explored. The results showed that pretreatments decreased the hydrophobic groups contents of lignin, i.e., aromatic ring, ether and carbonyl, as well as the content of ionizable group, i.e., carboxyl, which reduced its hydrophobicity and negative charge density, thus weakening the adsorption ability of lignin to cellulase. The lignin samples mainly adsorbed the CBHII component of cellulase to inhibit the synergistic effect of free enzymes. Lignin adsorption altered the spatial position of tryptophan residues in free enzymes, exposing them to the protein surface. In addition, the secondary structure of free enzymes was altered, with a decrease in the alpha-helix content and an increase in the random coil content, thus loosening the spatial conformation of free enzymes. The change degree in the spatial structure of free enzymes correlated with the adsorption capacity of the lignin, i.e., lignin with low adsorption capacity caused the least damage to free enzyme, with NaOH pretreated lignin being the best. It appears that appropriate pretreatment and chemical modification of enzymes to resist lignin adsorption is a promising long-term pathway to overcome the lignin inhibition during sugar-platform conversion of lignocellulosic bioresources.

Abstract:Lignin was extracted from bamboo (Dendrocalamus latiforus) shoot shell by acetic acid process.The biological activities of lignin on glucose dialysis retardation index (GDRI),fermentation property and enzyme activity were investigated and compared with the same source of cellulose residues and coarse dietary fiber.The results showed that the GDRI ability of lignin was relatively low,but the fermentation properties were enhanced.The pH value in the fermentation liquid was decreased,the content of short-chain fatty acid (acetic acid,propionic acid and butynic acid) in the fermentation liquid was increased.Since lignin activated α-amylase activity,it can be used as an activator of α-amylase.However,no obvious influence on the lipase activity was found.Therefore,lignin has a positive effect on fermentation properties and α-amylase activity of bamboo shoot shell dietary fiber.

Abstract:The lignin present in lignocellulose seriously affects the efficiency of cellulose enzymatic hydrolysis. In addition, lignin adsorbs high-cost cellulase, causing greater economic losses. Lignin can also disturb the site of action of cellulase and reduce the efficiency of hydrolysis. Therefore, if lignin is removed or surface modified before cellulose enzymatic hydrolysis, the enzymatic hydrolysis efficiency of lignocellulosic biomass will be greatly improved. In this paper, the cellulose enzymatic properties of bamboo biomass being treated with dilute acid and alkaline under the intervention of biosurfactant rhamnolipid were evaluated. The effects of rhamnolipids on the adsorption characterization of cellulose on pretreated bamboo were studied. Besides, the inter-communication between rhamnolipids and cellulose was investigated by fluorescence probe. The results showed that rhamnolipids could have a positive effect on the enzymatic hydrolysis of bamboo biomass by reducing the non-productive adsorption of cellulase on the surface of lignocellulose. The outcome illustrated that cellulase could be combined with rhamnolipids micelles, participating in the formation of rhamnolipids micelles, thereby increasing the internal hydrophobicity of the micelles, but could not change the properties of rhamnolipids micelles higher than one CMC (Critical Micelle Concentration). It can be seen that the interaction between rhamnolipids and cellulase is beneficial to enhance the stability and enzymatic activity of cellulase, thereby improving the enzymatic hydrolysis efficiency of cellulose in biomass. Based on these results, a theoretical knowledge about the mechanism of enhancing the enzymatic hydrolysis efficiency of lignocellulose by biosurfactants rhamnolipids is provided.
Enzymatic hydrolysis of lignin by ligninolytic enzymes and analysis of the hydrolyzed lignin products

Abstract:The degradation of alkali lignin was studied using three types of pure enzyme, Lac, LiP, and MnP, using alkali lignin as substrate. The alkali lignin removal rate was found to be 28.98% when Lac, LiP, and MnP were cultured together for alkali lignin degradation. Changes in the structure and composition before and after degradation were characterized by scanning electron microscopy, Fourier-transform infrared spectroscopy, nitrogen adsorption, and gas chromatography-mass spectrometry. The degradation product pathways were analyzed. The enzyme was proven to degrade alkali lignin, resulting in destruction of the alkali lignin structure, ring-opening of the macromolecular benzene ring structure and groups in alkali lignin, and chemical bond cleavage. This study explains the principle of alkali lignin enzymatic hydrolysis and provides a theoretical basis for the biodegradation of lignin.

Abstract:Lignin is one of the important components of lignocellulosic cell walls, which endows plant cell walls with rigidity and strength and protects them from microbial invasion. The presence of lignin is thought to hinder the conversion of biomass to bioenergy, so understanding enzyme-lignin interactions is very important in order to reduce the inhibition of lignin and improve the hydrolysis yields. Conversion of lignocellulosic raw materials into bioethanol is divided into pretreatment, enzymatic hydrolysis, and fermentation. In this paper, both pretreatment and enzymatic hydrolysis of lignocellulose are described in detail. Finally, the reasons why lignin hinders enzymatic hydrolysis efficiency, mainly from forming spatial barriers and interacting with cellulase, are discussed, and the influencing factors and mechanisms of action of cellulase hydrolysis are explored with a view to targeted regulation of lignin structure to improve lignocellulosic saccharification.
Unveiling the role of long-range and short-range forces in the non-productive adsorption between lignin and cellulases at different temperatures

Abstract:Quantitatively understanding of interaction mechanism between lignin and cellulases is essential for the efficient improvement of lignocellulose enzymatic hydrolysis. However, the individual contribution of multiple forces between lignin and cellulases to the non-productive adsorption of enzymes still remains deeply ambiguous, especially in situations of near enzymatic hydrolysis temperatures. Herein, atomic force microscopy (AFM) and computational simulations were utilized to quantitatively analyze the intermolecular forces between lignin and enzyme at 25 °C and 40 °C. Our results unveiled that an increase in temperature obviously improved adsorption capacity and total intermolecular forces between lignin and cellulases. This positive relationship mainly comes from the increase in the decay length of hydrophobic forces for lignin-cellulases when temperature increases. Different from the hydrophobic interaction which provides long-range part of attractions, van der Waals forces dominate the intermolecular force only at approaches < 2 nm. On the other hand, electrostatic forces exhibited repulsive effects, and its intensity and distance were limited due to the low surface potential of cellulases. Short-range forces including hydrogen bonding (main) and π-π stacking (minor) stabilize the non-specific binding of enzymes to lignin, but increasing temperature reduces hydrogen bond number. Therefore, the relative contribution of long-range forces increased markedly at higher temperatures, which benefits protein capture and brings lignin and cellulase close together. Finally, the structure–activity relationships between lignin physicochemical properties and its inhibitory effect to enzymes indicated that hydrophobic interactions, hydrogen bonding, and steric effects drive the final adsorption capacity and glucose yields. This work provides quantitative and basic insights into the mechanism of lignin-cellulase interfacial interactions and guides design of saccharification enhancement approaches.

Abstract:The inhibitory action of lignin on cellulase cocktails is a major challenge to the biological saccharification of plant cell wall polysaccharides. Although the mechanism remains unclear, hydrophobic interactions between enzymes and lignin are hypothesized to drive adsorption. Here we evaluate the role of hydrophobic interactions in enzyme-lignin binding. The hydrophobicity of the enzyme surface was quantified using an estimation of the clustering of nonpolar atoms, identifying potential interaction sites. The adsorption of enzymes to lignin surfaces, measured using the quartz crystal microbalance, correlates to the hydrophobic cluster scores. Further, these results suggest a minimum hydrophobic cluster size for a protein to preferentially adsorb to lignin. The impact of electrostatic contribution was ruled out by comparing the isoelectric point (pI) values to the adsorption of proteins to lignin surfaces. These results demonstrate the ability to predict enzyme-lignin adsorption and could potentially be used to design improved cellulase cocktails, thus lowering the overall cost of biofuel production.

Abstract:Abstract Ligninolytic bacteria degrading lignin were isolates and identified, and their biodegradation mechanism of alkaline-lignin was investigated. Four strains with lignin degradation capability were screened and identified from the soil, straw, and silage based on their decolorizing capacity of aniline blue and colony size on alkaline-lignin medium. The degradation ratio of Bacillus aryabhattai BY5, Acinetobacter johnsonii LN2, Acinetobacter lwoffii LN4, and Micrococcus yunnanensis CL32 have been assayed using alkaline-lignin as the unique carbon source. Further, the Lip (lignin peroxidase) and Mnp (manganese peroxidase) activities of strains were investigated. Lip activity of A. lwoffii LN4 was highest after 72 h of incubation and reached 7151.7 U · l –1 . Mnp activity of M. yunnanensis CL32 was highest after 48 h and reached 12533 U · l –1 . The analysis of alkaline-lignin degradation products by GC-MS revealed that the strains screened could utilize aromatic esters compounds such as dibutyl phthalate (DBP), and decomposite monocyclic aromatic compounds through the DBP aerobic metabolic pathway. The results indicate that B. aryabhattai BY5, A. johnsonii LN2, A. lwoffii LN4, and M. yunnanensis CL32 have high potential to degrade alkaline-lignin, and might utilize aromatic compounds by DBP aerobic metabolic pathway in the process of lignin degradation.

Abstract:Lignin can affect the enzymatic hydrolysis efficiency of lignocellulose. In this study, the lignin isolated from sugarcane bagasse (SCB) pretreated with p-toluenesulfonic acid (PL) was firstly aminated, and then the effects of PL and aminated PL (APL) on the bagasse enzymatic hydrolysis efficiency (EHE) were investigated. The results showed that the addition of PL and APL promoted the EHE, and EHE with APL (73.82%) was higher than PL (51.39%). To explore the reason, the data were further analyzed including cellulase adsorption capacity, enzyme activity, cellulase-lignin interaction, and molecular docking. It was found that APL adsorbed more cellulase (27.83 mg protein/g lignin) than PL (4.96 mg protein/g lignin), resulting from the greater interaction force and lower binding free energy between APL and cellulase. The addition of APL more remarkably enhanced the cellobiohydrolase and endoglucanase activities than PL due to more effectively inducing cellulase conformation optimization.

Abstract:Blocking the unproductive adsorption of cellulase to lignin has been popularly thought to be the chief mechanism of non-ionic surfactant improving enzymatic hydrolysis of lignocellulose. This study, however, proposes a different viewpoint. Laccase and hemicellulase-xylanase (HXase) which respectively modify lignin and degrade hemicellulose were used to treat raw and ball-milled sugarcane bagasse (SCB). The treated SCB maintained similar relative lignin and hemicellulose contents as the untreated SCB. Laccase treatment resulted in wrinkles on the SCB micro-surface, while HXase treatment led to the formation of many pores in SCB. It found that Tween20, a kind of non-ionic surfactant, could obviously facilitate the adsorption of cellulase to untreated and treated SCB. It was completely opposite to the abovementioned popular mechanism. The enzymatic hydrolysis efficiencies of raw SCB treated by laccase and HXase, ball-milled SCB treated by laccase for 72 h and HXase for 24 h or 72 h, and ball-milled SCB treated by HXase for 24 h with Tween20 addition were 26.10%, 61.65%, 61.42%, and 70.86%, respectively, which had significant differences against those without Tween20 addition. Combined with surface structure observations, it hints that the enhancing effect of a non-ionic surfactant on the enzymatic hydrolysis of lignocellulose tends to depend on the lignocellulosic structural feature rather than lignin presence.

Abstract:Abstract Enzymatic hydrolysis of lignocellulose for bioethanol production shows a great potential to remit the rapid consumption of fossil fuels, given the fact that lignocellulose feedstocks are abundant, cost-efficient, and renewable. Lignin results in low enzymatic saccharification by forming the steric hindrance, non-productive adsorption of cellulase onto lignin, and deactivating the cellulase. In general, the non-productive binding of cellulase on lignin is widely known as the major cause for inhibiting the enzymatic hydrolysis. Pretreatment is an effective way to remove lignin and improve the enzymatic digestibility of lignocellulose. Along with removing lignin, the pretreatment can modify the lignin structure, which significantly affects the non-productive adsorption of cellulase onto lignin. To relieve the inhibitory effect of lignin on enzymatic hydrolysis, enormous efforts have been made to elucidate the correlation of lignin structure with lignin–enzyme interactions but with different views. In addition, contrary to the traditional belief that lignin inhibits enzymatic hydrolysis, in recent years, the addition of water-soluble lignin such as lignosulfonate or low molecular-weight lignin exerts a positive effect on enzymatic hydrolysis, which gives a new insight into the lignin–enzyme interactions. For throwing light on their structure–interaction relationship during enzymatic hydrolysis, the effect of residual lignin in substrate and introduced lignin in hydrolysate on enzymatic hydrolysis are critically reviewed, aiming at realizing the targeted regulation of lignin structure for improving the saccharification of lignocellulose. The review is also focused on exploring the lignin–enzyme interactions to mitigate the negative impact of lignin and reducing the cost of enzymatic hydrolysis of lignocellulose.

Abstract:The changes in the structure of lignin during the pretreatment processes of biomass can affect its subsequent enzymatic hydrolysis efficiency. To explore the influence mechanism of dilute acid and hydrothermal pretreatment of corn stover lignin on cellulose saccharification, an integrated nondestructive spectral technology with fluorescence spectroscopy, surface plasmon resonance, and atomic force microscopy was performed. It showed that the surface lignins on the corn stover with dilute acid pretreatment at 190 °C (DA190-SL) and hydrothermal pretreatment at 190 °C (HP190-SL) possessed enhancement (from 76.95 to 80.09%) and inhibition (from 76.95 to 61.91%) for the enzymatic digestibility of Avicel, respectively. Nondestructive spectroscopic analysis indicated that HP190-SL adsorbed onto cellulase is mainly driven by hydrogen bonding and van der Waals forces, while the DA190-SL-enzyme system was mainly driven by hydrophobic interactions. The association affinity of DA190-SL combined with cellulase was higher than that of HP190-SL. The binding force of the HP190-SL-enzyme (0.16 nN) is lower than that of the DA190-SL-enzyme (0.75 nN), which leads to a higher propensity for dissociation of HP190-SL from cellulase after binding. This study aims to establish a theoretical basis for regulating the enzymatic performance during the hydrothermal and dilute acid pretreatment of corn stover at the molecular level.

Abstract:During the bioconversion of lignocellulose, enzymatic hydrolysis of cellulose to produce glucose is a key step. However, it also has become a bottle-neck for effective bioconversion of lignocellulosic biomass to fuels or chemicals. A large amount of works have demonstrated that addition of non-ionic surfactant during enzymatic hydrolysis of pretreated lignocelluloses can effectively improve the cellulose conversion thus reducing the enzyme loading. In this paper, the effects of non-ionic surfactant on enzymatic hydrolysis of pure cellulose and pretreated lignocellulose are reviewed comprehensively. The relationships of the structural features of substrate, hydrolysis conditions and cellulase formulation with the action of surfactant are discussed. Corresponding mechanisms are analyzed in terms of the adsorption of cellulases and the synergism of the cellulase components. However, the current research progress has not clearly elucidated the mechanisms for the effects of non-ionic surfactant on cellulose hydrolysis. It is proposed that to deeply understand the mechanism, further researches should be focused on systematically investigating the relations of substrate structure, hydrolysis conditions with the actions of non-ionic surfactant to the enzymatic hydrolysis of cellulose. Microscopically, the interactive actions and forces between surfactant and substrate, surfactant and cellulase enzymes should be investigated from the atomic and molecular levels. The thermodynamic and kinetic behaviors of enzymatic hydrolysis of cellulose with addition of surfactant should also be illustrated.

Abstract:<p>Enzymatic digestibility of an acid-pretreated poplar (AP, 42.9%) was superior to that of a similarly acid-pretreated larch (AL, 12.5%). Effects of lignin-related recalcitrance on enzymatic hydrolysis were comprehensively investigated by disrupting the two predominant lignin fractions present in acid-pretreated material (extractable lignin and bulk lignin). Lignin removal and bovine serum albumin (BSA) addition were performed to estimate the relative contributions of lignin towards physical blocking and enzyme binding on enzymatic hydrolysis. The lignin physical blocking played a more significant role in limiting the enzymatic hydrolysis of AL. BSA addition improved enzymatic hydrolysis of AP more significantly than AL. Moreover, the effects of lignin embedded in the lignocellulosic matrix on enzyme non-productive binding were compared with the isolated lignin. It indicated that the lignin distribution would influence the lignin effects on enzyme non-productive binding during enzymatic hydrolysis. Results will give insights towards improvement of enzymatic hydrolysis on acid-pretreated woody biomass.</p>

Abstract:The objective of this research was to investigate cellulase adsorption and recycling during enzymatic hydrolysis of two differently pretreated wheat straws (WS). Dilute acid treated WS showed lower hydrolysis yield of polysaccharides fraction and adsorbed more cellulase with hydrolyzed residue than dilute alkali treated sample. Four methods capable of recovering and recycling the enzyme bound to the residual substrate and the enzyme free in solution were used for three consecutive rounds of hydrolysis to compare their recycling efficiencies. Compared to the absorption recycling method, ultrafiltration recycling method possessed the capacity to retain p-glucosidase, thereby avoiding the supplementation of fresh p-glucosidase in subsequent rounds of hydrolysis. It was found that whatever recycling method was used, better recycling results were obtained for dilute alkali treated substrate than for dilute acid treated substrate. These results suggested that the great difference in the lignin content between acid treated WS and alkali treated WS would significantly affect enzymatic hydrolysis, cellulase adsorption and cellulase recycling efficiencies. Abstract:The hydrolysis of lignocellulose into fermentable monosaccharides using cellulases represents a critical stage in lignocellulosic bioconversion. However, the inactivation of cellulase in the presence of lignin is attributed to the high cost of biofinery. To address this challenge, a comprehensive investigation into the structure-function relationship underlying lignin-driven cellulase inactivation is essential. In this study, molecular docking and molecular dynamics (MD) simulations were employed to explore the impacts of lignin fragments on the catalytic efficiency of cellulase at the atomic level. The findings revealed that soluble lignin fragments and cellulose could spontaneously form stable complexes with cellulase, indicating a competitive binding scenario. The enzyme's structure remained unchanged upon binding to lignin. Furthermore, specific amino acid residues have been identified as involved in interactions with lignin and cellulose. Hydrophobic interactions were found to dominate the binding of lignin to cellulase. Based on the mechanisms underlying the interactions between lignin fragments and cellulase, decreased hydrophobicity and change in the charge of lignin may mitigate the inhibition of cellulase. Furthermore, site mutations and chemical modification are also feasible to improve the efficiency of cellulase. This study may contribute valuable insights into the design of more lignin-resistant enzymes and the optimization of lignocellulosic pretreatment technologies.Communicated by Ramaswamy H. Sarma.

Abstract:In this study, 15 plant species representing plant evolution were selected, and distinct lignocellulose compositions for largely varied biomass enzymatic saccharification were detected. By comparison, the acid-pretreated lignocellulose of rice mutant was of the highest Congo-red adsorption (298 mg/g) accounting for cellulose accessibility, leading to complete cellulose hydrolysis and high bioethanol production. By conducting oxidative-catalysis with the acid-pretreated lignocellulose of moss plant, the optimal biosorbent was generated with maximum Cd/Pb adsorption (54/118 mg/g), mainly due to half-reduced cellulose polymerization degree and raised functional groups accountable for multiple physical and chemical interactions. Furthermore, the acid-pretreated lignocellulose of eucalyptus was of large and small pores for much higher adsorption capacities with direct-yellow and direct-blue than those of the previously-reported. Therefore, this study raises a mechanism model about how distinct lignocelluloses of plant evolution are selective for complete biomass saccharification and optimal biosorbents assembly, providing insights into lignocellulose biosynthesis and biomass conversion.
