Abstract:Kefir grain,as a complex system of microbial symbiosis,contains many useful microorganisms. The research focused on the main microbial composition of kefir grains and the preparation of a microbial starter culture.According to the colony forms and biological properties,the kefir grain samples were consisted of two strains of yeast,three strains of lactic acid bacteria( LAB) and two strains of acetic acid bacteria( AAB). After sequencing the 16 S r DNA sequence of the seven strains,it can be concluded that the yeasts were identified as Kluyveromyces marxianus and Pichia kudriavzevii,LAB were Lactobacillus pontis and Lactobacillus kefiri,AAB were Acetobacter lovaniensis and Acetobacter cibinongensis. Further,the isolated strains were cultivated and microencapsulated.After the microcapsules were mixed and fermented in milk,the performance of the nutrition contents and volatile components and the property of antimicrobial activity were very similar to the original kefir grains.

Abstract:Milk kefir is a traditional fermented milk product whose consumption is becoming increasingly popular. The natural starter for kefir production is kefir grain, which consists of various bacterial and yeast species. At the industrial scale, however, kefir grains are rarely used due to their slow growth, complex application, bad reproducibility and high costs. Instead, mixtures of defined lactic acid bacteria and sometimes yeasts are applied, which alter sensory and functional properties compared to natural grain-based milk kefir. In order to be able to mimic natural starter cultures for authentic kefir production, it is a prerequisite to gain deep knowledge about the nature of kefir grains, its microbial composition, morphologic structure, composition of strains on grains and the impact of environmental parameters on kefir grain characteristics. In addition, it is very important to deeply investigate the numerous multi-dimensional interactions among different species, which play important roles on the formation and the functionality of grains.

Abstract:The analysis of the literature on the microbiome composition and metabolic properties of kefir available at the RSCI and Web of Science was carried out. Kefir has been used by humans for centuries. It is a useful product of mixed lactic and alcoholic fermentation, produced using evolutionally established associative cultures, collected in an aggregated state termed kefir grains. General characterization of kefir grains from the territorial zones of different continents (Russia, Europe, Asia, and America) is provided. The methods for differentiation and identification of individual species are described, as well as their interactions within the community. The diversity of microbial composition of kefir grains depending on local cultivation conditions and storage processes is shown. The microorganisms present in kefir have a number of properties that determine their metabolism, interaction in the community, beneficial effects on human health and immune system, which is important for the prevention and control of bacterial and viral infections, especially during the COVID-19 pandemic.

Abstract:Background: In order to balance human health and environmental sustainability, plant-based diets have been attracting increasing attention. Plant-based fermented foods are produced using vegetables or fruits as the main raw materials. Thereafter, microorganisms and their metabolites convert these into the final products, which are often covered by biofilms during production and storage. The biofilms are composed of various microbial flora and extracellular metabolites produced during fermentation, which is generally considered as a shortcoming of fermentation. However, growing evidence suggests that these complex microbial ecosystems are sources of both probiotic substances and antimicrobial compounds, which can benefit health and improve food processing. Scope and approach: Advanced studies have established relationships between the representative film-forming microorganisms in biofilms and the quality and safety of fermented foods. Inhibition and elimination strategies have also been proposed by targeting biofilm control methods from the food and medical industries towards the formation mechanisms and compositional characteristics of the biofilms. Key findings and conclusions: Based on the data generated from previous control measures, this review introduces the key elements pertaining to biofilm formation as function of substrate and metabolic conditioning and summarizes the potential benefits of biofilms, especially in plant-based fermented foods. Further, this review highlights strategies surrounding the utilization and modulation of biofilms in plant-based fermented foods. The re-design and functionalization of biofilms are therefore discussed for a wide range of applications.

Abstract:Kefir is a traditional dairy beverage, usually made from cow or goat milk fermented with kefir grains, and has many health benefits. To elucidate the fermentation patterns of animal milk kefirs during the fermentation process and find the optimal milk types, cow, camel, goat, and donkey milk were fermented with kefir grains for 0, 1, 3, 5, and 7 days. Volatile and non-volatile metabolites and microbial changes were dynamically monitored. The results showed that volatile flavor substances were massively elevated in four kefirs on days 1-3. Lipids and carbohydrates gradually decreased, while amino acids, small peptides, and tryptophan derivatives accumulated during fermentation in four kefirs. Besides, four kefirs had similar alterations in Lactobacillus and Acetobacter, while some distinctions existed in low-abundance bacteria. Association analysis of microorganisms and volatile and non-volatile metabolites also revealed the underlying fermentation mechanism. This study found that appropriately extending the fermentation time contributed to the accumulation of some functional nutrients. Furthermore, goat and donkey milk could be the better matrices for kefir fermentation.

Abstract:Kefir grains are symbiotic microbiota starters in the production of dairy products such as fermented milk, kefir. In recent decades, microbial symbiosis in kefir grains and its role in fermentation have attracted a lot of attention. This study aimed to obtain novel kefirs with different characteristics by "stimulating" kefir grains with juices of Actinidia chinensis , Solanumly copersicum , Daucus carrot , and Lactuca sativa , respectively, before being used as starter cultures for kefir fermentation. Then, compositions of the predominant microbiota in novel kefirs were determined. These novel kefirs showed changes in their physiochemical characteristics and enhanced antimicrobial activities against Shigella dysenteriae , Candida albicans , Staphylococcus aureus , Serratia marcescens , and Escherichia coli . Therefore, preprocessing of kefir grains with natural substrates could be a feasible strategy in the production of novel kefirs with different flavors and various health benefits.

Abstract:A comprehensive metagenomics-based investigation of the microorganisms present within milk kefir communities from across the globe was carried out with a view to defining the milk kefir pan-metagenome, including details relating to core and non-core components. Milk kefir samples, generated by inoculating full fat, pasteurized cow's milk with 64 kefir grains sourced from 25 different countries, were analyzed. We identified core features, including a consistent pattern of domination by representatives from the species Lactobacillus helveticus or the sub-species Lactobacillus kefiranofaciens subsp. kefiranofaciens, Lactococcus lactis subsp. lactis or Lla. cremoris subsp. cremoris in each kefir. Notably, even in kefirs where the lactococci did not dominate, they and 51 associated metabolic pathways were identified across all metagenomes. These insights can contribute to future efforts to create tailored kefir-based microbial communities for different applications and assist regulators and producers to ensure that kefir products have a microbial composition that reflects the artisanal beverage.
Microbiological profiles, physiochemical properties and volatile compounds of goat milk kefir fermented by reconstituted kefir grains

Abstract:Kefir is a health-promoting fermented dairy products, consisting in a variety of bacterial and fungal species. The physical and chemical properties and the correlation between the microbiota and volatile compounds of goat milk kefir fermented by reconstituted kefir grains were investigated. Cluster analysis showed that a strong symbiotic homeostasis system existed in goat milk kefir, indicating that the composition of microorganism in reconstituted kefir grains was still restricted by the original one. Compared with other kefir samples, goat milk kefir 80DE20CN (ratios of kefir grains CN/DE = 80/20) showed significant improvement in physiochemical properties and "Fruity" flavor. Positive correlations among Lactobacillus helveticus and ethyl acetate, amyl acetate and ethanol, Kazachstania unispora and octyl formate, 1-heptanol and hexanoic acid were obtained by Partial least squares regression analysis (PLSR) model. The results showed that changes in microbial succession patterns may be useful to optimize the physiochemistry and flavor of goat milk kefir or related products.

Abstract:Water kefir is a sparkling, slightly acidic fermented beverage produced by fermenting a solution of sucrose, to which dried fruits have been added, with water kefir grains. These gelatinous grains are a symbiotic culture of bacteria and yeast embedded in a polysaccharide matrix. Lactic acid bacteria, yeast and acetic acid bacteria are the primary microbial members of the sugary kefir grain. Amongst other contributions, species of lactic acid bacteria produce the exopolysaccharide matrix from which the kefir grain is formed, while yeast assists the bacteria by a nitrogen source that can be assimilated. Exactly which species predominate within the grain microbiota, however, appears to be dependent on the geographical origin of the grains and the fermentation substrate and conditions. These factors ultimately affect the characteristics of the beverage produced in terms of aroma, flavour, and acidity, for example, but can also be controlled and exploited in the production of a beverage of desired characteristics. The production of water kefir has traditionally occurred on a small scale and the use of defined starter cultures is not commonly practiced. However, as water kefir increases in popularity as a beverage - in part because of consumer lifestyle trends and in part due to water kefir being viewed as a health drink with its purported health benefits - the need for a thorough understanding of the biology and dynamics of water kefir, and for defined and controlled production processes, will ultimately increase. The aim of this review is to provide an update into the current knowledge of water kefir.

Abstract:Kefir is a popular and indigenous fermented product that possess various health claims. Kefir grains with varied bacteria are utilized as starters for kefir production of popular genra including Lactobacillus, Leuconostoc, Lactococcus, and Acetobacter species. The complex microbial community in kefir produce various bioactive compounds owing to their metabolic activities. Kefir has various functional and therapeutic properties, which include immune system stimulation, lactose intolerance symptoms, cholesterol reduction, anti-carcinogenic and anti-mutagenic qualities. Several studies have shown anticarcinogenic, anti-microbial and synbiotic potential of kefir. Current review highlights the production technology, nutritional, functional and therapeutic potential of kefir. Furthermore, production strategies, kefir characteristics and probiotic potential are the limelight of the current review.

Abstract:A global epidemiological shift has been observed in recent decades, characterized by an increase in age-related disorders, notably non-communicable chronic diseases, such as type 2 diabetes mellitus, cardiovascular and neurodegenerative diseases, and cancer. An appreciable causal link between changes in the gut microbiota and the onset of these maladies has been recognized, offering an avenue for effective management. Kefir, a probiotic-enriched fermented food, has gained significance in this setting due to its promising resource for the development of functional or value-added food formulations and its ability to reshape gut microbial composition. This has led to increasing commercial interest worldwide as it presents a natural beverage replete with health-promoting microbes and several bioactive compounds. Given the substantial role of the gut microbiota in human health and the etiology of several diseases, we conducted a comprehensive synthesis covering a total of 33 investigations involving experimental animal models, aimed to elucidate the regulatory influence of bioactive compounds present in kefir on gut microbiota and their potential in promoting optimal health. This review underscores the outstanding nutritional properties of kefir as a central repository of bioactive compounds encompassing micronutrients and amino acids and delineates their regulatory effects at deficient, adequate, and supra-nutritional intakes on the gut microbiota and their broader physiological consequences. Furthermore, an investigation of putative mechanisms that govern the regulatory effects of kefir on the gut microbiota and its connections with various human diseases was discussed, along with potential applications in the food industry.

Abstract:The microbial community diversity of Tibetan kefir grains (TKGs) contributes to intricate metabolite changes and metabolic interactions during milk fermentation. In present research, metagenomic sequencing analysis was used to reveal microflora composition of TKGs, indicating the presence of 522 species, with Lactobacillus kefiranofaciens as the most abundant species. Lactobacillus and Bifidobacterium were the most diverse probiotics. Subsequently, the metabolites changes of kefir were investigated using metabolomics with UHPLC - MS/MS. Datasets at the 8 h, 24 h and 36 h of fermentation (group A, B, C, respectively) were analyzed to reveal the apparent differences during fermentation. A total 192 differential metabolites were identified in the A vs B group, and 96 differential metabolites were identified in the C vs B group. In addition, 7 metabolic pathways were enriched based on the differential metabolites in group A vs B, and mainly related to lipid metabolism, amino acid biosynthesis, carbohydrate metabolism and phenylpropanoid biosynthesis. The differential metabolites in C vs B group covered 11 pathways, mainly involving amino acid metabolism, citrate cycle (TCA cycle), translation, membrane transport and biosynthesis of secondary metabolites. The study of metabolomics in kefir fermentation provides a theoretical foundation for visualization of the fermentation process in future studies.

Biodiversitas 24: 5302-5308. Kefir is a type of traditional fermented milk made from kefir grains. The quality of kefir grain depends on the milk medium, which impacts microbial diversity and population. This study uses metagenomic analysis to assess the microbial populations of kefir grains prepared from cow's milk and goat milk using the 16s rRNA method. The results revealed that >99% Relative Abundance (RA) of the sample's total bacteria belonged to the phylum Firmicutes and the phylum Bacteroidota, respectively. However, most of the phylum Firmicutes was found in the sample of goat milk kefir. Further investigation revealed that goat milk samples had Lactobacillus, Streptococcus, and Staphylococcus genera, while the cow milk samples contained >99% RA of Lactobacillus, Bacteroides, and Muribaculaceae. Regarding yeast, the family Saccharomycetaceae predominated in cow's milk, while goat milk had 2% of the family Saccharomycetaceae and 97% of unassigned microorganisms. The functional analysis revealed that kefir grains from the two sources had a variety of amino acid metabolisms, secondary metabolite metabolisms, and vitamin production. This metagenomic analysis shows that the type of milk affects the diversity of the population of both bacteria and yeast. This study opens up opportunities to further investigate the potential of kefir as a functional food beverage, a source of probiotics, and an antibacterial agent property.

Yeasts like P. kudriavzevii and P. guilliermondii isolated in this study were the first time reported in Tibetan kefir grains. For TK-ZJUJ 01-03, lactic acid bacteria were the major microorganisms, which accounted for more than 50% of all the microbial population, while for TK-ZJUJ04, the largest microbial group was yeasts which accounted for more than 50%. In a word, study of diversity and composition of microflora provided us theoretical foundation for further investigation and application of Tibetan kefir grains. PRACTICAL APPLICATION This is the basic research in order to develop and industrialize a new kind of yogurt starter which is naturally formed microbiota with both lactic acid bacteria and yeasts in it.

Abstract:Gluconacetobacter sp. A4 ( G. sp. A4), which had strong ability to produce d-saccharic acid 1, 4 lactone (DSL), was the key functional bacteria isolated from the kombucha preserved. This paper investigated the interaction between G. sp. A4 and ten different strains of lactic acid bacteria (LAB) obtained from kefir. The result suggested that the LAB promoted DSL production of G. sp. A4 to different extents, ranging from 4.86% to 86.70%. Symbiosis between G. sp. A4 and LAB was studied. LAB’s metabolites, xylitol, and acetic acid, were utilized by G. sp. A4, and it promoted the growth of G. sp. A4 and yield of DSL. Therefore, in developing starter cultures for kombucha fermentation process, a mixed flora of LAB and G. sp. A4 would be the optimal combination.

Abstract:Kefir is a high nutritional fermented dairy beverage associated with a wide range of health benefits. It constitutes a unique symbiotic association, comprising mainly lactic acid bacteria, yeasts, and occasionally acetic acid bacteria, which is strongly influenced by the geographical origin of the grains, the type of milk used, and the manufacture technology applied. Until recently, kefir microbiota has been almost exclusively studied by culture-dependent techniques. However, high-throughput sequencing, alongside omics approaches, has revolutionized the study of food microbial communities. In the present study, the bacterial, and yeast/fungal microbiota of four home-made samples (both grains and drinks), deriving from well spread geographical regions of Greece, and four industrial beverages, was elucidated by culture-dependent and -independent analyses. In all samples, classical microbiological analysis revealed varying populations of LAB and yeasts, ranging from 5.32 to 9.60 log CFU mL –1 or g –1 , and 2.49 to 7.80 log CFU mL –1 or g –1 , respectively, while in two industrial samples no yeasts were detected. Listeria monocytogenes , Salmonella spp. and Staphylococcus spp. were absent from all the samples analyzed, whereas Enterobacteriaceae were detected in one of them. From a total of 123 isolates, including 91 bacteria and 32 yeasts, Lentilactobacillus kefiri , Leuconostoc mesenteroides , and Lactococcus lactis as well as Kluvyeromyces marxianus and Saccharomyces cerevisiae were the mostly identified bacterial and yeast species, respectively, in the home-made samples. On the contrary, Streptococcus thermophilus, Lactobacillus delbrueckii subsp. bulgaricus , and Lacticaseibacillus rhamnosus along with Debaryomyces hansenii and K. marxianus were the main bacterial and yeast species, respectively, isolated from the industrial beverages. In agreement with the identification results obtained from the culture-dependent approaches, amplicon-based metagenomics analysis revealed that the most abundant bacterial genera in almost all home-made samples (both grains and drinks) were Lactobacillus and Lactococcus , while Saccharomyces, Kazachstania , and Kluvyeromyces were the predominant yeasts/fungi. On the other hand, Streptococcus, Lactobacillus , and Lactococcus as well as Kluvyeromyces and Debaryomyces dominated the bacterial and yeast/fungal microbiota, respectively, in the industrial beverages. This is the first report on the microbiota of kefir produced in Greece by a holistic approach combining classical microbiological, molecular, and amplicon-based metagenomics analyses.
