Abstract:Pyrrhotite is ubiquitously found in natural environment and involved in diverse (bio)processes. However, the pyrrhotite-driven bioreduction of toxic selenate [Se(VI)] remains largely unknown. This study demonstrates that Se(VI) is successfully bioreduced under anaerobic condition with the participation of pyrrhotite for the first time. Completely removal of Se(VI) was achieved at initial concentration of 10 mg/L Se(VI) and 0.56 mL/min flow rate in continuous column experiment with indigenous microbial consortium and pyrrhotite. Variation in hydrochemistry and hydrodynamics affected Se(VI) removal performance. Se(VI) was reduced to insoluble Se(0) while elements in pyrrhotite were oxidized to Fe(III) and SO 4 2− . Breakthrough study indicated that biotic activity contributed 81.4 ± 1.07% to Se(VI) transformation. Microbial community analysis suggested that chemoautotrophic genera (e.g., Thiobacillus ) could realize pyrrhotite oxidation and Se(VI) reduction independently, while heterotrophic genera (e.g., Bacillus, Pseudomonas ) contributed to Se(VI) detoxification by utilizing metabolic intermediates generated through Fe(II) and S(-II) oxidation, which were further verified by pure culture tests. Metagenomic and qPCR analyses indicated genes encoding enzymes for Se(VI) reduction (e.g., serA, napA and srdBAC ), S oxidation (e.g., soxB ) and Fe oxidation (e.g., mtrA ) were upregulated. The elevated electron transporters (e.g., nicotinamide adenine dinucleotide, cytochrome c) promoted electron transfer from pyrrhotite to Se(VI). This study gains insights into Se biogeochemistry under the effect of Fe(II)-bearing minerals and provides a sustainable strategy for Se(VI) bioremediation in natural aquifer.

Abstract:Interstitial water profiles of SeO 4 2− , SeO 3 2− , SO 4 2− , and Cl − in anoxic sediments indicated removal of the seleno-oxyanions by a near-surface process unrelated to sulfate reduction. In sediment slurry experiments, a complete reductive removal of SeO 4 2− occurred under anaerobic conditions, was more rapid with H 2 or acetate, and was inhibited by O 2 , NO 3 − , MnO 2 , or autoclaving but not by SO 4 2− or FeOOH. Oxidation of acetate in sediments could be coupled to selenate but not to molybdate. Reduction of selenate to elemental selenium was determined to be the mechanism for loss from solution. Selenate reduction was inhibited by tungstate and chromate but not by molybdate. A small quantity of the elemental selenium precipitated into sediments from solution could be resolublized by oxidation with either nitrate or FeOOH, but not with MnO 2 . A bacterium isolated from estuarine sediments demonstrated selenate-dependent growth on acetate, forming elemental selenium and carbon dioxide as respiratory end products. These results indicate that dissimilatory selenate reduction to elemental selenium is the major sink for selenium oxyanions in anoxic sediments. In addition, they suggest application as a treatment process for removing selenium oxyanions from wastewaters and also offer an explanation for the presence of selenite in oxic waters.

Abstract:Selenate [Se(VI)] with higher content in groundwater will be harmful for human beings. Hence, effective treatment for Se(VI) in aquifer should be conducted reasonably. Microbial reduction of Se(VI) to elemental selenium with weak movability and toxicity has attracted significant attention due to its high efficiency and no secondary contamination. However, hydrodynamic and hydrochemical influences with corresponding mechanisms during Se(VI) bioreduction are still not clear. In this study, influences of flow rate, initial Se(VI) and organic concentrations, coexisting nitrate were evaluated. Se(VI) removal efficiency and capacity reached 96.42 ± 6.82% and 41.28 ± 3.41 (g/m<sup>3</sup>·d) with flow rate of 0.56 mL/min, initial Se(VI) and chemical organic demand concentrations of 10 mg/L and 400 mg/L. Dechloromonas and Pseudomonas were presumably contributed to Se(VI) reduction, with upregulated serA and tatC genes. Solid Se<sup>0</sup> was identified as the final product from Se(VI) reduction. These results will be beneficial for the further comprehending of Se(VI) remediation in aquifer.

Novel mechanisms of selenate and selenite reduction in the obligate aerobic bacterium Comamonas testosteroni S44

Abstract:Selenium oxyanion reduction is an effective detoxification or/and assimilation processes in organisms, but little is known the mechanisms in aerobic bacteria. Aerobic Comamonas testosteroni S44 reduces Se(VI)/Se(IV) to less-toxic elemental selenium nanoparticles (SeNPs). For Se(VI) reduction, sulfate and Se(VI) reduction displayed a competitive relationship. When essential sulfate reducing genes were respectively disrupted, Se(VI) was not reduced to red-colored SeNPs. Consequently, Se(VI) reduction was catalyzed by enzymes of the sulfate reducing pathway. For Se(IV) reduction, one of the potential periplasm molybdenum oxidoreductase named SerT was screened and further used to analyze Se(IV) reduction. Compared to the wild type and the complemented mutant strain, the ability of Se(IV) reduction was reduced 75% in the deletion mutant ΔserT. Moreover, the Se(IV) reduction rate was significantly enhanced when the gene serT was overexpressed in Escherichia coli W3110. In addition, Se(IV) was reduced to SeNPs by the purified SerT with the presence of NADPH as the electron donor in vitro, showing a Vmax of 61 nmol/min·mg and a Km of 180 μmol/L. A model of Se(VI)/Se(IV) reduction was generated in aerobic C. testosteroni S44. This work provides new insights into the molecular mechanisms of Se(VI)/Se(IV) reduction activities in aerobic bacteria.

Abstract:Selenite biotransformation by microorganisms is an effective detoxification and assimilation process. Bacillus subtilis is a probiotic bacterium that can reduce Se(IV) to SeNPs under aerobic conditions. However, current knowledge on the molecular mechanisms of selenite reduction by B. subtilis remains limited. Here, the reduction of Se(IV) by probiotic bacterium Bacillus subtilis 168 was systematically analysed, and the molecular mechanisms of selenium nanoparticle (SeNPs) formation were characterised in detail. B. subtilis 168 reduced 5.0 mM selenite by nearly 40% in 24 h, and the produced SeNPs were spherical and localised intracellularly or extracellularly. FTIR (Fourier transform infrared) spectroscopy suggested the presence of proteins, lipids, and carbohydrates on the surface of the isolated SeNPs. Transcriptome data analysis revealed that the expression of genes associated with the proline metabolism, glutamate metabolism, and sulfite metabolism pathways was significantly up-regulated. Gene mutation and complementation revealed the ability of PutC, GabD, and CysJI to reduce selenite in vivo. In vitro experiments demonstrated that PutC, GabD, and CysJI could catalyse the reduction of Se(IV) under optimal conditions using NADPH as a cofactor. To the best of our knowledge, our study is the first to demonstrate the involvement of PutC and GabD in selenite reduction. Particularly, our findings demonstrated that the reduction of Se(IV) was mediated by multiple pathways both in vivo and in vitro. Our findings thus provide novel insights into the molecular mechanisms of Se(VI) reduction in aerobic bacteria.

Abstract:The reduction of selenite [Se(Ⅳ)] by microorganisms is a green and efficient detoxification strategy. We found that Se(Ⅳ) inhibited exopolysaccharide and protein secretion by Lactiplantibacillus plantarum BSe and compromised cell integrity. In this study, L. plantarum BSe reduced Se(Ⅳ) by increasing related enzyme activity and electron transfer. Genomic analysis demonstrated that L. plantarum BSe should be able to reduce Se(Ⅳ). Further transcriptome analysis showed that L. plantarum BSe enhanced its tolerance to Se(Ⅳ) by upregulating the expression of surface proteins and transporters, thus reducing the extracellular Se(Ⅳ) concentration through related enzymatic reactions and siderophore-mediated pathways. Lactiplantibacillus plantarum BSe was able to regulate the expression of related genes involved in quorum sensing and a two-component system and then select appropriate strategies for Se(Ⅳ) transformation in response to varying environmental Se(Ⅳ) concentrations. In addition, azo reductase was linked to the reduction of Se(Ⅳ) for the first time. The present study established a multipath model for the reduction of Se(Ⅳ) by L. plantarum, providing new insights into the biological reduction of Se(Ⅳ) and the biogeochemical cycle of selenium.

Abstract:In the present study, we examined the ability of Enterobacter cloacae Z0206 to reduce toxic sodium selenite and mechanism of this process. E. cloacae Z0206 was found to completely reduce up to 10 mM selenite to elemental selenium (Se°) and form selenium nanoparticles (SeNPs) under aerobic conditions. The selenite reducing effector of E. cloacae Z0206 cell was to be a membrane-localized enzyme. iTRAQ proteomic analysis revealed that selenite induced a significant increase in the expression of fumarate reductase. Furthermore, the addition of fumarate to the broth and knockout of fumarate reductase ( frd ) both significantly decreased the selenite reduction rate, which revealed a previously unrecognized role of E. cloacae Z0206 fumarate reductase in selenite reduction. In contrast, glutathione-mediated Painter-type reactions were not the main pathway of selenite reducing. In conclusion, E. cloacae Z0206 effectively reduced selenite to Se° using fumarate reductase and formed SeNPs; this capability may be employed to develop a bioreactor for treating Se pollution and for the biosynthesis of SeNPs in the future.

Abstract:In nature, many microorganisms show resistance to toxic selenite by reducing selenite to non-soluble and low toxic elemental selenium. Extracellular polymeric substances (EPS), a high-molecular-weight biopolymers originated from microbial metabolism, contain many reducing groups and can induce reductive transformation of pollutants. However, the roles of EPS and its redox state in reductive detoxification or reduction removal of selenite, respectively, remain unknown yet. Herein, the reduction of selenite by different sources of EPS was investigated. Selenite was proved to be reduced by EPS and partly transformed to elemental selenium. The formed elemental selenium was mainly selenium nanoparticles confirmed by transmission electron microscopy coupled with energy dispersive spectroscopy. The redox state of EPS governed selenite reduction and elemental selenium formation, and the reduced state of EPS was in favor of selenite reduction. Dissolved oxygen concentration in water regulated EPS redox state and influenced selenite reduction. The thiols, aldehyde and phenolic groups in EPS were responsible for selenite reduction. Under selenite stress, EPS was capable of increasing cell survivability by enhancing microorganisms-mediated selenite reduction. This work revealed the previously undiscovered roles of EPS in selenite reduction and elemental selenium formation in aquatic environments and also suggested a possible crucial role of EPS in selenium biogeochemical cycle.

Abstract:Removal of selenate (SeO 4 2- ) from selenate-contaminated wastewater is challenging due to the commonly co-existing and competing anions of sulfate (SO 4 2- ) and nitrate (NO 3 - ). This study investigates SeO 4 2− reduction to elemental selenium (Se 0 ) in a cathode-based bioelectrochemical (BEC) reactor and a conventional biofilm reactor ( i.e. , an upflow anaerobic reactor). The simulated wastewater contained SeO 4 2− at a typical concentration of 5 mg Se/L, SO 4 2− at a typical concentration of 1000 mg S/L, and NO 3 − at concentrations that varied from 0 to 10 mg N/L. The impact of sulfate on the BEC reactor was much lower than that on the conventional reactor: The selenium removal, defined as (selenate in influent – dissolved selenium in effluent)/selenate in influent, was 99 % in the BEC reactor versus 69 % in the conventional biofilm reactor. The lower selenium removal in the conventional reactor was mainly due to the >10 times higher reduction of sulfate, which directly caused competition between sulfate and selenate for the common resources such as electrons. The more reduction of sulfate in the conventional reactor further led to 45 times higher production of selenide. Selenide is usually assumed to be minimal and therefore not measured in the literature. This simplification may significantly overestimate selenium removal when the influent sulfate concentration is very high. NO 3 - in the influent of the BEC reactor promoted selenium removal when it was less than 5.0 mg N/L but inhibited selenate removal when it was more than 7.5 mg N/L. This was supported by the microbial community analysis and intermediate (nitrite) analysis.

Abstract:The microbial reduction of selenite to elemental selenium nanoparticles (SeNPs) is thought to be an effective detoxification process of selenite for many bacteria. In this study, Metasolibacillus sp. ES129 and Oceanobacillus sp. ES111 with high selenite reduction efficiency or tolerance were selected for systematic and comparative studies on their performance in selenite removal and valuable SeNPs recovery. The kinetic monitoring of selenite reduction showed that the highest transformation efficiency of selenite to SeNPs was achieved at a concentration of 4.24 mM for ES129 and 4.88 mM for ES111. Ultramicroscopic analysis suggested that the SeNPs produced by ES111 and ES129 had been formed in cytoplasm and subsequently released to extracellular space through cell lysis process. Furthermore, the transcriptome analysis indicated that the expression of genes involved in bacillithiol biosynthesis, selenocompound metabolism and proline metabolism were significantly up-regulated during selenite reduction, suggesting that the transformation of selenite to Se0 may involve multiple pathways. Besides, the up-regulation of genes associated with nucleotide excision repair and antioxidation-related enzymes may enhance the tolerance of bacteria to selenite. Generally, the exploration of selenite reduction and tolerance mechanisms of the highly selenite-tolerant bacteria is of great significance for the effective utilization of microorganisms for environmental remediation.

Abstract:Selenite biotransformation by microorganisms is an effective detoxification and assimilation process. However, current knowledge of the molecular mechanisms of selenite reduction remains circumscribed. Here, the reduction of Se(IV) by a highly selenite-resistant Bacillus sp. SL (up to 50 mM) was systematically analyzed, and the molecular mechanisms of selenite reduction were investigated. Remarkably, 10 mM selenite was entirely transformed by the strain SL within 20 h, demonstrating a faster conversion rate compared to other microorganisms. Furthermore, glutathione (GSH) and exopolysaccharides (EPS) changes were also monitored during the process. Transcriptomic analysis revealed that the genes of ferredoxin-sulfite oxidoreductase (6.82) and sulfate adenylyltransferase (6.32) were significantly upregulated, indicating that the sulfur assimilation pathway is the primary reducing pathway involved in selenite reduction by strain SL. Moreover, key genes associated with NAD(P)/FAD-dependent oxidoreductases and thioredoxin were significantly upregulated. The reduction of Se(IV) was mediated by multiple pathways in strain SL. To our knowledge, this is the initial report to identify the involvement of sulfur assimilation pathway in selenite reduction for bacillus , which is rare in aerobic bacteria.
Interaction of aqueous Se(IV)/Se(VI) with FeSe/FeSe2: Implication to Se redox process

Abstract:Since reductive precipitation is considered as the most effective way to immobilize 79Se, interaction of aqueous Se(IV)/Se(VI) with Fe(II)-bearing minerals has received extensive attention. In contrast to the thermodynamic calculations, as well as the prevalence of iron selenide phases observed in soil, sediments and ore deposits, most laboratory experiments have found that Se(0) was the reaction product. In this study, the interaction of Se(IV)/Se(VI) with FeSe/FeSe2 were investigated. The results demonstrate that FeSe and FeSe2 can be oxidized to Se(0) by Se(IV) with relatively fast kinetics, while reaction between Se(VI) and FeSe/FeSe2 only occurs under limited conditions (i.e. in the presence of high ferrous content and higher pH) with much slower kinetics, and there is no evident reaction in most case. Therefore, reduction of Se(IV) by Fe(II)-bearing minerals, in particular by natural occurring minerals, is envisioned to produce Se(0) at the early stage of experiments, rather than FeSe or FeSe2. Due to the formation of bulk Se(0) and its low solubility, the Fe–Se–O–H2O system will maintain redox disequilibrium in laboratory time-scale. This study also reveals that iron selenides, like iron sulfides, have strong reactivity toward Fe3+. The findings in this study give insight into possible controls on Se redox process.
Selenium-mediated Cr(VI) reduction and SeNPs synthesis accelerated Bacillus cereus SES to remediate Cr contamination

Abstract:Microbial biotransformation of Cr(VI) is a sustainable approach to reduce Cr(VI) toxicity and remediate Cr(VI) contamination. In this study, Bacillus cereus SES with the capability of reducing both Cr(VI) and Se(IV) was isolated, and the effect of Se supplementation on Cr(VI) reduction by Bacillus cereus SES was investigated. Se(IV) addition enabled 2.6-fold faster Cr(VI) reduction, while B. cereus SES reduced 96.96% Se(IV) and produced more selenium nanoparticles (SeNPs) in the presence of Cr(VI). Co-reduction products of B. cereus SES on Cr(VI) and Se(IV) were SeNPs adsorbed with Cr(III). The relevant mechanisms were further revealed by proteomics. Se(IV) supplementation mediated the synthesis of Cr(VI) reductants and stress-resistant substances, thus enhancing Cr(VI) resistance and promoting Cr(VI) reduction. Meanwhile, high Se(IV) reduction rate was associated with Cr(VI)-induced electron transport processes, and Cr(VI) mediated the up-regulation of flagellar assembly, protein export and ABC transporters pathways to synthesis and export more SeNPs. Furthermore, Se combined with B. cereus SES had the potential to reduce the toxicity of Cr(VI) via reducing the bioavailability of Cr and improving the bioavailability of Se in soil. Results suggested that Se could be an efficient strategy to enhance the remediation of B. cereus SES on Cr contamination.

Abstract:Sediments are the major sink for selenium (Se) in aquatic environments. Se speciation in sediments is crucial for its bioavailability and toxicity in benthos, but this is relatively understudied. In this study, the background levels of Se in the river sediments, fish flakes, and Lumbriculus variegatus were also detected. Then, the dynamic changes of selenium speciation and concentrations in sediments were investigated after adding selenite (Se(IV)) and seleno-L-methionine (Se-Met) in the sediments for 90 and 7 days, and the accumulation and depuration of Se(IV) and Se-Met for 7 days in the oligochaete L. variegatus were also explored. Without the presence of worms, the levels of Se(IV) in the sediments were relatively stable within 7 days but showed a decreasing trend during the 90 days of aging. In contrast, Se-Met in the sediments showed a sharp decrease within 3 days of aging. The LC 50 -96 h values of Se(IV) and Se-Met in L. variegatus were 372.6 and 9.4 μg/g, respectively. Interestingly, the dominant Se species in Se(IV)- or Se-Met-treated L. variegatus was Se-Met, whose level was increased with time in 7 days of exposure. Se was barely depurated from L. variegatus during the 8 days of the depuration period. This study has provided indispensable data on the levels of total Se in the abiotic and biotic matrices and the biodynamics of Se in a representative benthos, which could better understand the ecological risk of Se to the freshwater benthic communities.

Abstract:Selenium is an essential trace element for humans, animals and microorganisms. Microbial transformations, in particular, selenium dissimilatory reduction and bioremediation applications have received increasing attention in recent years. This review focuses on multiple Se-reducing pathways under anaerobic and aerobic conditions, and the phylogenetic clustering of selenium reducing enzymes that are involved in these processes. It is emphasized that a selenium reductase may have more than one metabolic function, meanwhile, there are several Se(VI) and/or Se(IV) reduction pathways in a bacterial strain. It is noted that Se(IV)-reducing efficiency is inconsistent with Se(IV) resistance in bacteria. Moreover, we discussed the links of selenium transformations to biogeochemical cycling of other elements, roles of Se-reducing bacteria in soil, plant and digestion system, and the possibility of using functional genes involved in Se transformation as biomarker in different environments. In addition, we point out the gaps and perspectives both on Se transformation mechanisms and applications in terms of bioremediation, Se fortification or dietary supplementation.

Abstract:The experiments in this paper employed the strains of Bacillus HBS4 isolated from oil reservoir. By growing in the facultative aerobic condition the cells acted on selenite. After 36 hours,all the samples with sodium selenite radical from 1mM up to 11mM produced many red deposits. Under biomicroscope the authors found one or more red round intracellular deposits in the cell with free floating deposits outside the cell. Intracellular organization of deposits includes one or two regular chains, curve or irregular in form. The XPS patterns of the deposits show that the sodium selenite has been exclusively reduced to elemental selenium. Furthermore,glucose in the culture media can impact greatly the quantity of the deposits of the reduced elemental selenium.

Abstract:Antimony (Sb), a toxic metalloid, has serious negative effects on human health and its pollution has become a global environmental problem. Bio-reduction of Sb(V) is an effective Sb-removal approach. This work, for the first time, demonstrates the feasibility of autotrophic Sb(V) bio-reduction and removal coupled to anaerobic oxidation of elemental sulfur (S0). In the S0-based biological system, Sb(V) was reduced to Sb(III) via autotrophic bacteria by using S0 as electron donor. Meanwhile, S0 disproportionation reaction occurred under anaerobic condition, generating sulfide and SO42- in the bio-systems. Subsequently, Sb(III) reacted with sulfide and formed Sb(III)-S precipitate, achieving an effective total Sb removal. The precipitate was identified as Sb2S3 by SEM-EDS, XPS, XRD and Raman spectrum analyses. In addition, it was found that co-existing nitrate inhibited the Sb removal, as nitrate is the favored electron acceptor over Sb(V). In contrast, the bio-reduction of co-existing SO42- enhanced sulfide generation, followed by promoting Sb(V) reduction and precipitation. Illumina high-throughput sequencing analysis revealed that Metallibacterium, Citrobacter and Thiobacillus might be responsible for Sb(V) reduction and S0 disproportionation. This study provides a promising approach for the remediation of Sb(V)-contaminated water.

Abstract:Arsenate [As(V)] reduction is a major cause of arsenic (As) release from soils, which threatens more than 200 million people worldwide. While heterotrophic As(V) reduction has been investigated extensively, the mechanism of chemolithotrophic As(V) reduction is less studied. Since As is frequently found as a sulfidic mineral in the environment, microbial mediated sulfur oxidation coupled to As(V) reduction (SOAsR), a chemolithotrophic process, may be more favorable in sites impacted by oligotrophic mining (e.g. As-contaminated mine tailings). While SOAsR is thermodynamically favorable, knowledge regarding this biogeochemical process is still limited. The current study suggested that SOAsR was a more prevalent process than heterotrophic As(V) reduction in oligotrophic sites, such as mine tailings. The water-soluble reduced sulfur concentration was predicted to be one of the major geochemical parameters that had a substantial impact on SOAsR potentials. A combination of DNA stable isotope probing and metagenome binning revealed members of the genera Sulfuricella, Ramlibacter, and Sulfuritalea as sulfur oxidizing As(V)-reducing bacteria (SOAsRB) in mine tailings. Genome mining further expanded the list of potential SOAsRB to diverse phylogenetic lineages such as members associated with Burkholderiaceae and Rhodocyclaceae. Metagenome analysis using multiple tailing samples across southern China confirmed that the putative SOAsRB were the dominant As(V) reducers in these sites. Together, the current findings expand our knowledge regarding the chemolithotrophic As(V) reduction process, which may be harnessed to facilitate future remediation practices in mine tailings.
